Testo completoRocky islands in Saldanha Bay, southwestern Cape, South Africa, support aggregations of seabirds. Guano produced by the seabirds washed off the islands into intertidal and nearshore waters,where it provides a source of inorganic and organic nutrients. The concentrations of nutrients in intertidal rockpools at island and mainland sites in the Saldanha Bay area were measured,and the biological structures of intertidal communities at these sites were compared in the light of differences in intertidal nutrient status. Monthly measurements were made of algal production and of the growth, density, reproductive condition and mortality of the dominant intertidal herbivore, Patella granularis.The rate of intertidal algal production was enhanced on shores washed by nutrient-rich waters, and the growth rates and life-history patterns of populations of P. granularis were, in turn, modified by the increased availability of algal food. On unenriched shores rates of algal production and limpet growth and reproduction were slow. Seabirds thus influence trophic relationships between intertidal organisms, without themselves being components of the community. Islands in Saldanha Bay also support dense populations of African Black Oystercatchers Haematopus moqu.ini, which forage intertidally. Predation by oystercatchers on P. granular is resulted in a modification of limpet densities and population size structures, and a reduction in the intensity of grazing. The impact of predation was, however, modified by the presence, in intertidal waters, of nutrients from seabird guano, and the consequences thereof. It is proposed that differences in the structures of communities on rocky shores, and in the roles of processes influencing structure, might •be due, in part, to differences in the nutrient status of intertidal waters.

Testo completoENGLISH ABSTRACT: The privilege of utilising resources from the globe's marine ecosystems is threatened. Adequate regional and global conservation efforts are vital. Understanding species' distributions and the environmental variables influencing community structures are important for instigating optimum conservation plans. A major threat facing marine ecosystems is the introduction and proliferation of alien invasive species. The distribution and geographical expansion of marine alien invasive species, along with environmental variables driving their increase is also critical for drawing up conservation plans. In response to this point, this study addresses gaps in our knowledge regarding intertidal marine invertebrate species distributions particularly that of marine alien species in the Kogelberg Biosphere Reserve (KBR) and adjacent Betty's Bay Marine Protected Area (MPA) in the Western Cape of South Africa, a prime area for south-coast marine conservation. Species identifications were done in September 2010. Two marine alien invertebrate species were identified along the coastline. These were the invasive mytilid Mediterranean mussel Mytilus galloprovincialis and the bryozoan Watersipora subtorquata. A quantitative systematic survey of the intertidal region, consisting of seven rocky shore sites was then undertaken from October 2010 to December 2010. The abundance of all focal marine invertebrate species, within six selected habitat types was assessed. Environmental variables were also recorded. Statistical tests were done to investigate for significant interaction in mean abundance and mean species richness among intertidal zones and protection status (MPA and non-MPA), and among intertidal sampling units (SU‟s) and protection status. There was also investigated for significant difference in mean abundance and mean species richness across intertidal zones and habitat types where protection status was not taken into account, and to test for significant differences in mean abundance and mean species richness between the intertidal zones and habitat types. Correspondence analyses were used to illustrate relationships between species composition and the defined environmental variables. Tests were done to investigate for significant differences in mean abundance of the identified marine alien species between the MPA and non-MPA, study sites, SU's, and wave exposures. These results indicated that vertical and horizontal stress gradients along the shore mainly influence abundance and species richness. The significantly higher abundance of M. galloprovincialis outside the MPA is due to its preference for wave-exposed areas. The reason for the difference in the total abundance of W. subtorquata in the different habitats is not clear, although it appears that the higher abundance of this species in the MPA is because the sheltered nature of the MPA. M. galloprovincilais displaces indigenous species. W. subtorquata does not cause any significant harm along the coast of South Africa. This study indicates that the abundance of M. galloprovincialis and W. subtorquata in the MPA is relatively low. The MPA is therefore in good condition with regards to intertidal alien species. It is important that the MPA and adjacent KBR's coastline maintain its current pristine status.
AFRIKAANSE OPSOMMING: Die voorreg om mariene hulpbronne te benut, word bedreig. Voldoende nasionale en globale mariene bewaring is krities. 'n Deeglike kennis van spesies verspreiding en omgewingsveranderlikes wat gemeenskapstrukture beïnvloed is noodsaaklik vir optimale bewaringspogings. Mariene indringerspesies en verspreiding van indringerspesies bedreig mariene ekosisteme. Dit is belangrik om basiese kennis in te win oor die geografiese verspreiding van indringerspesies saam met die omgewingsveranderlikes wat hierdie verspreiding beïnvloed. Hierdie studie fokus dus daarop om die kennis met betrekking tot intergety ongewerwelde spesiesverspreiding, veral dié wat uitheems is langs die kus van die Kogelberg Biosfeer Reservaat (KBR) en aangrensende Bettys Baai Mariene Beskermde Gebied (MBG) in die Wes-Kaap in Suid-Afrika uit te brei. Hierdie streek word beskou as gesog vir mariene bewaring in Suid-Afrika. Identifikasie van spesies is in September 2010 gedoen. Twee intergety uitheemse ongewerwelde spesies is langs die kus geïdentifiseer; die indringer 'mytilid' Mediterreense mossel - Mytilus galloprovincialis en die 'bryozoa' Watersipora subtorquata. Daar is statisties getoets vir verskille in die talrykheid en spesiesrykheid tussen die intergety zones en beskermingsstatus (MBG en nie-MBG), en tussen die intergety habitattipes en beskermingsstatus. Daar is ook getoets vir verskille in talrykheid en spesiesrykheid tussen die intergety zones en habitattipes waar beskermingsstatus nie in ag geneem is nie. Ooreenstemmende analises is gedoen om die verhoudings tussen die spesiesamestelling en die gedefinieerde omgewingsveranderlikes te illustreer. Daar is statisties getoets vir verskille in die talrykheid van die geïdentifiseerde uitheemse spesies tussen die MBG en nie-MBG, die verskillende lokaliteite, die verskillende habitattipes en tussen die verskillende golf blootstellings. Hierdie studie dui aan dat vertikale en horisontale stresgradiënte die hoofoorsaak is wat talrykheid en spesiesrykheid beïnvloed. Die aansienlik hoër talrykheid van M. galloprovincialis buite die MBG is van die gebied se hoër golfblootstelling. Die beduidende verskil in talrykheid van W. subtorquata is onduidelik. Testo completoENGLISH ABSTRACT: Various aspects of the biology and ecology of woolly apple aphid, Eriosoma lanigerum, were investigated, including initial galling damage caused by E. lanigerum to the roots of apple trees, the possible relationship between E. lanigerum and Xiphinema and Pratylenchus nematodes and the effectiveness of Biostart 2000® and Furfural® as possible control agents of E. lanigerum in the orchard. Preliminary root damage by first instar E. lanigerum feeding was characterized by the mechanical injury of endodermis and parenchyma tissues. Damage by second, third and fourth instar E. lanigerum was similar, but the symptoms were more pronounced. Damage caused by adults included a pronounced swelling at infected areas of the root. Cell walls hardened until the root was radially strengthened with sclerenchyma tissue and nonconducting xylem vessels while the cuticle expanded greatly through the growth of corklike cambium tissue. There was no direct relationship between the population dynamics of E. lanigerum and those of Xiphinema and Pratylenchus nematodes. The occurrence of E. lanigerum appeared to be seasonal while P. penetrans and Xiphinema numbers fluctuated erratically. Undamaged root nitrogen levels seemed to correspond with the normal root growth cycle. Nitrogen levels from galled roots were significantly lower than those of undamaged roots, probably due to E. lanigerum feeding. Soils rich in fine sand and clay sustained higher populations of E. lanigerum and Xiphinema than sandy soils. The number of E. lanigerum found in soil samples correlated well with the damage index allocated to the samples. The numbers of Xiphinema found in soil samples also correlated well with the damage index allocated to the samples according to suspected Xiphinema damage symptoms. Both Biostart 2000® and Furfural® were effective as control agents of woolly apple aphid. Furfural'Ï, a chemical waste product of the sugarcane industry, was however not as effective as Biostart 2000®, a product that includes an activator and three bacterial species, Bacillus laterosporus, B. chitinosporus and B. licheniformis. The bacteria in the Biostart 2000® treated pots could replicate themselves under suitable conditions while Furfural® dilutes with each watering. Biostart 2000® is also easier to prepare than Furfural® since the components of Biostart 2000® readily mix to form a paste easily thinned by water, whereas Furfural® is an oily substance that does not easily disperse in water. Root damage was initiated soon after E. lanigerum started feeding, however there was no apparent relationship between E. lanigerum and the nematode species. The most promising, environmentally friendly control measure was Biostart 2000®.
AFRIKAANSE OPSOMMING: Verskeie aspekte van biologie en die ekologie van die appel bloedluis, Eriosoma lanigerum, was ondersoek insluitende aanvanklike galvorming veroorsaak deur E. lanigerum op wortels van appelbome, die moontlike verwantskap tussen E. lanigerum en Xiphinema en Pratylenchus nematodes en die effektiwiteit van Biostart 2000® en Furfural® as moontlike beheeragente van E. lanigerum in die boord. Aanvanklike wortelskade deur eerste ins tar E. lanigerum voeding was gekenmerk deur die meganiese beskadiging van endodermale en parenchiem weefsel. Die voorkoms van E. lanigerum was seisoenaal terwyl P. penetrans en Xiphinema se getalle onvoorspelbaar gefluktueer het. Onbeskadigde wortel stikstofvlakke het ooreengestem met die normale wortel groeisiklus. Stikstof vlakke van galwortels was noemenswaardig laer as dié van onbeskadigde wortels, heel waarskynlik as gevolg van voeding deur E. lanigerum. Grond ryk aan fyn sand en klei het groter bevolkings van E. lanigerum en Xiphinema onderhou as sanderige gronde. Die aantal E. lanigerum in grondmonsters het goed ooreengestem met die skade indeks wat aan die monsters toegeken was. Die aantal Xiphinema in grondmonsters het ook goed ooreengestem met die beskadigingsindeks wat aan die monsters toegeken is weens vermoedelike Xiphinema skade simptome. Testo completoENGLISH ABSTRACT:Heavy metals are persistent environmental contaminants whose sources of inputs into the environment are both natural and anthropogenic. The levels of heavy metals (cadmium, copper, nickel, lead and zinc) in the False Bay intertidal zone were measured in the water, sediments and invertebrate species between August 2000 and August 2001. The results of the water and sediment analyses revealed that most pollution was associated with the northern shore of the bay between Strand and Muizenberg, where the most populated and industrial catchments occur. Significant spatial variations occurred, indicating the presence of localised contamination, while seasonal variations may be related to changes in precipitation and runoff at different times of the year. The concentrations of cadmium, nickel and lead were occasionally higher than the levels recommended by the South African Water Quality Standards. The possible sources of pollution at the different sites are also discussed. The concentrations of the five metals in the different invertebrate species (Oxystele tigrina, 0. sinensis, Choromytilus meridionalis, Patella oculus, Patiriella exigua and Tetraclita serrata) also revealed significant seasonal and spatial variations, with both the soft tissues and shells accumulating heavy metals. The barnacle T serrata from Rooiels had the highest cadmium concentration (70.67 J.lg/g dry weight), which may be related to historic pollution inputs from the military activities which took place at a weapons testing site at this site between 1987 and 1994, although no evidence was found to confirm this. The periwinkle 0. tigrina from Strand had the highest copper concentration (70.25 J.lg/g) while the limpet P. oculus from the same site had the highest nickel concentration (35.75 J.lg/g). The shells of the mussel C. meridionalis from Muizenberg had the highest concentration of lead (25.75 J.lg/g). Since cadmium occurs as a constituent of phosphate fertilisers used widely in the False Bay catchments, the effects of cadmium exposure on the different species were investigated during 14-day laboratory exposures to 200 and 400 J.lg/LCdCh. The results revealed a general pattern of tissue metal increase in the exposed organisms, followed by slight reductions after decontamination in clean seawater. The viscera and kidneys of C. meridionalis accumulated most of the dissolved cadmium. The shells of the mussels also accumulated cadmium, indicating the possible use of shells as a detoxification matrix.
AFRIKAANSE OPSOMMING:Swaarmetale is persisterende omgewingskontaminante waarvan die insetbronne beide natuurlik of van menslike oorsprong kan wees. Die kontaminasievlakke van swaarmetale (kadmium, koper, nikkel, lood en sink) in die Valsbaai tussengetysone is in die water, sedimente en invertebraatspesies bepaal vanaf Augustus 2000 tot Augustus 2001. Voorlopige resultate van die water- en sedimentontledings het getoon dat die meeste besoedeling by die noordelikke oewer van die baai voorgekom het tussen Strand and Muizenberg, waar die mees digbewoonde en ge-industrialiseerde opvangsgebiede is. Betekenisvolle ruimtelike en seisoenale variasie het in die konsentrasies van swaarmetale voorgekom, met die ruimtelike variasie wat moontlik gelokaliseerde kontaminasie aandui terwyl die seisoenale variasies weer verband mag hou met veranderings in die neerslag en afloop gedurende verskillende tye van die jaar. Die konsentrasie van kadmium, nikkel en lood was somtyds hoer as die vlakke wat deur die Suid-Afrikaanse Waterkwaliteitsstandaarde voorgestel word. Die moontlike bronne van besoedeling in die verskillende areas is ook in bespreking genoem. Die konsentrasies van die vyf swaametale in die verskillende invertebraatspesies (Oxystele tigrina, 0. sinensis, Choromytilus meridionalis, Patella oculus, Patiriella exigua and Tetraclita serrata) het ook seisoenale en ruimtelike variasies vertoon, die swaarmetale het in die sagte weefsel en skulpe van die invertebrate geakkumuleer. Die hoogste gemiddelde konsentrasie van kadmium (70.67 ).lg/g droe massa) is in die heel-liggaam monsters van die eendemossel T serrata gemeet wat by Rooiels versamel is. Testo completoENGLISH ABSTRACT: Runoff is generally regarded as one of the most important routes of nonpoint source pesticide pollution in agricultural surface waters. Of major concern is the fact that low, sub-lethal levels of pesticide exposure are responsible for negative ecotoxicological effects, stressing the need for methods capable of identifying problem areas where populations could be at risk. Predicted average losses of three pesticides in tributaries of nine sub-catchments of the Lourens River were calculated through use of a GIS-based runoff model. There was a significant (p < 0.005) positive correlation between the predicted average loss and mean measured concentrations of the insecticides both in water and suspended sediments (R2 between 0.75 and 0.9), indicating that the model could serve as a powerful tool for the risk assessment and management of surface waters in South African orchard areas. Based on field relevant exposure scenanos, the potential effects of azinphos-methyl on macroinvertebrate communities were evaluated in a combined microcosm and field approach. Microcosms were contaminated for 1 h with AZP (control, 0.2; 1,5and 20 ug/L; three replicates each) and acute effects on survival were evaluated 6 days after exposure. The sensitivity or tolerance of 12 core taxa was determined based on their response to the exposure scenarios and compared to field tolerance/sensitivity as was established by a field investigation at a control and contaminated site of the Lourens River. The sensitivity/tolerance of ten of the 12 taxa corresponded to that which was found in the field. Thus microcosm studies employing a field relevant design can be successfully linked to field studies and indicate that transient pesticide contamination affects the aquatic communities of the Lourens River.
AFRIKAANSE OPSOMMING Afloop word oor die algemeen beskou as een van die belangrikste roetes van niepuntbron pestisiedbesoedeling in landbou oppervlakwaters. Die feit dat lae, sub-letale vlakke van pestisiedblootstelling negatiewe ektoksikologiese gevolge kan hê, is van groot belang. Dit beklemtoon die behoefte aan metodes om probleemgebiede te kan identifiseer waar bevolkings aan risiko onderhewig is. 'n GIS-gebaseerde afloopmodel is gebruik om die gemidddelde verlies van drie pestisiede in die sytakke van nege sub-opvangsgebiede van die Lourensrivier te voorspel. Daar was 'n beduidende (p < 0.005) positiewe korrelasie tussen die voorspelde gemiddelde verlies en gemete konsentrasies van insektisiede in beide die water en sediment (R2 between 0.75 and 0.9) fases, wat aandui dat die model as 'n kragtige hulpmiddel vir risikobestuur van oppervlakwaters in Suid Afrikaanse boord-gebiede kan dien. Testo completoChronic exposure to high levels of metals in the environment can cause severe damage to ecosystems and human health. The City of Cape Town, an immense contributor to atmospheric pollution lies beneath the Table Mountain range, one of the Seven Wonders of the World and renowned for its biodiversity and ancient, indigenous forests, yet no plan exist to monitor metal concentrations. These ecosystems are subjected to considerable metal inputs all year round, which greatly increases during brown haze episodes in winter. Soil, leaf litter, moss, lichen and millipedes are key organisms in forests and reliable indicators for determining critical loads and preventing broader impacts to forests, which leads to the main purpose of this study: to determine the health of metal-contaminated forest ecosystems in the western cape, the smallest biome in South Africa, yet of utmost importance to our environment and ultimately human health. The objectives of this study were a) to determine concentrations of prominent metals in soil, leaf litter and sentinel organisms in a pilot study b) to compare the dry and wet season with regard to: (i) seasonal fluctuations of the concentrations of the metals (ii) the oxidative stress effect of metals, using two markers indicative of induced oxidative stress (oxidative lipid damage products and glutathione (total GSH levels) in the pill millipede, Spaerotherium compressum, the moss, Hypnum cupressiforme and the lichen, Parmotrema sp. and c) to expose millipedes to metal contaminated soil for a period of six weeks: (i) to determine whether they accumulated metals and (ii) to assess the induced oxidative damage to lipids and redox status of glutathione in the pill millipede, Spaerotherium compressum. Three sampling sites in three afromontane forests in the Western Cape, Platbos, Orange Kloof and Newlands forest formed the study area. Five subsamples of soil, leaf litter and each sentinel organism were collected and chemically analysed to determine the metal concentrations, using an Inductively Coupled Plasma Mass Spectrophotometer (ICP–MS). The oxidative stress effect was determined in freeze dried samples. Millipedes were exposed in terrariums to a cocktail of metals Al, Fe and Mn for six weeks at control, low and high concentrations and analysed thereafter. Significant (P<0.05) findings in this study were the metal contamination patterns observed at the sites and forests in closest proximity of the City of Cape Town and related pollutant sources, especially vehicle volumes, and traffic behaviour. Even more significant (P<0.05) was the enhanced metal concentrations found during winter in the forest in closest proximity of the city, impacted by the brown haze phenomena. Metals may further have triggered an overproduction of ROS (reactive oxygen species) judging from the activated antioxidant, tGSH levels in an effort to scavenge ROS, as well as the MDA (malondialdehyde) levels (measured as TBARS), which indicated damage to cells. An important finding was that MDA levels were mostly higher in winter during the brown haze episodes signifying more damage in the organisms during that season. A programme to monitor metals in these forests was also suggested in view of a concern for the survival of forests and human health, as a result of the growing population, vehicle traffic and urbanization.

ENGLISH ABSTRACT: The biologically catalysed oxidation of pyrite in the outer layers of coal waste dumps leads to the formation of acid mine drainage. The oxidation of pyrite to ferric iron and sulphate is a complex process involving various abiotic and biologically catalysed reactions. Pyrite is abiotically oxidized by ferric iron, with the formation of thiosulphate and ferrous iron. Thiosulphate decomposes to form various inorganic sulphur compounds. Bacterial catalysis of pyrite oxidation is achieved by iron-oxidizing bacteria oxidizing ferrous iron to ferric iron. Bacteria that oxidize sulphur compounds assist the catalysis by oxidizing thiosulphate and its decomposition products. Heterotrophic organisms may play a role by consuming organic substances inhibitory to the lithotrophic bacteria. Abiotic ecological factors, acid formation and populations of iron-oxidizing bacterial groups were studied in 10 differently constructed pilot scale coal waste dumps, as the second phase of a study which started in September 1993. Gas samples were withdrawn weekly from coal waste through permanently buried stainless steel probes, for analysis in the field using a portable oxygen/carbon dioxide meter. Samples of coal waste were extracted by auger for analysis of moisture, pH and microbial populations. The analyses of oxygen and pH can be recommended for the routine monitoring of rehabilitated waste dumps. Covers of Avalon soil 0.3 or 0.5 m thick, were not adequate to prevent acidification. Coal waste covered with 0.7 m compacted beneath 0.3 m uncompacted Avalon soil, showed a slow pH decline, but reached approximately pH 3 in 1997. Covers of compacted Estcourt soil beneath tmcompacted Avalon soil to a cover depth of 1 m were effective in preventing acidification and generally kept the coal waste anaerobic. However, all covers developed cracks during drought conditions in 1995, allowing aeration. Low pH of some samples from these dumps during 1995/1996 may have indicated the start of acidification. Bacteria oxidizing high concentrations of ferrous iron and considered to be Thiobacillus ferrooxidans, were monitored routinely, but may not have been the dominant iron-oxidizer, as population counts using media with a lower ferrous iron concentration were higher. The majority of the latter organisms could also not oxidize sulphur, hence were not T. ferrooxidans. The populations of the high ferrous iron-oxidizing bacteria were affected by pH, tending to be high in acidified and low in non-acidified coal waste. Investigations of microbial populations forming iron-oxidizing consortia in enrichment cultures from coal waste and acid drainage samples showed the presence of T. ferrooxidans, the heterotrophic bacterial genus Acidiphilium, fungi of the genus Penicillium, unidentified filamentous fungi, including Cladophialophora-like morphological types, and a yeast of the genus Dipodascus. In interaction studies, the Penicillium isolate had an inhibitory effect on T. ferrooxidans (subjected to organic compound stress), but the Cladophialophora-like fungi reduced inhibition by organics. Fungi have not previously been studied in detail as components of iron-oxidizing consortia, but the bacterial isolations agree with those elsewhere, indicating that appropriate conclusions from acid mine drainage research in other parts of the world can be applied in KwaZulu-Natal.

Piriet word abioties deur ferri-yster geoksideer, met die vrystelling van tiosulfaat en ferro-yster. Tiosulfaat verval om verskeie anorganiese swawelverbindings te vorm. Bakteriese katalise van pirietoksidasie word deur ysteroksiderende bakteriee wat ferro-yster na ferri-yster oksideer, bewerkstellig. Bakteriee wat swawelverbindings oksideer maak 'n bydrae tot die katalise deur tiosulfaat en vervalprodukte daarvan te oksideer. Testo completoA series of incubation studies and greenhouse experiments were conducted to evaluate the use of EM seed treatments, at different application levels, handling techniques and soil conditions on germination and seedling vigour of selected cultivars of maize, sorghum and sunflower. Two incubation studies were conducted to evaluate the germination and seedling vigour of maize, sorghum and sunflower seeds treated with M-EM from three different suppliers, multiplied at two different ratios (1% and 3%) and diluted at three different levels (0.01%, 0.1% and 1.0%) compared to a control treated with pure water. Results revealed no significant differences under optimum germination conditions, while seedlings under cold stress indicated that M-EM treatments positively affected germination and seedling vigour compared to the control treatments. Two incubation studies were also conducted to evaluate the germination and seedling vigour of maize, sorghum and sunflower seeds treated with M-EM from three different suppliers, multiplied at two different ratios (1% and 3%) and exposed to the influences of irradiation and temperature fluctuation. From the results became clear that the correct storage and handling is essential in optimizing the effect of M-EM on seeds. Even though M-EM was exposed to irradiation and temperature fluctuation, M-EM still had positive effects on germination and seedling vigour. Pot experiments were conducted to determine the effect of EM as seed treatment, at different dilutions, on germination, seedling vigour and dry mass of maize, sorghum and sunflower at different planted depths. Germination were not affected by the M-EM treatment, while shoot length results indicated that seed treated with M-EM could have significant effect on seedling survival. A greater effect was visible on the shoot length of shallow planted seeds, than on deeper planted seeds. From the results no single company, ratio or dilution could be prescribed as paramount. To further investigate the effect of M-EM subjected to the influences of irradiation and temperature fluctuation; maize, sorghum and sunflower seeds were treated with M-EM from three different suppliers, multiplied at two different ratios (1% and 3%) and exposed to the influences of irradiation and temperature fluctuation and planted in soil. M-EM treatments only benefited the germination of deeper planted sorghum seeds compared to the control treatments. The shoot lengths of deeper planted maize and sunflower seed were positively increased by the M-EM treatments while also resulting in significant results for the overall shoot length of sorghum. The third pot study was conducted to determine the influence of EM as a seed treatment on maize, sorghum and sunflower planted in three different soils, namely: sterilized soil, soil treated with M-EM and Fusarium containing soil. Germination and seedling vigour results of the sterilized and M-EM treated soil revealed to be superior to that of the Fusarium containing soil. From the results was concluded that M-EM treatments will probably improve early seedling growth of maize, sorghum and sunflower compared to untreated seed and that M-EM seed treatment and a pre-plant EM soil treatment might assist seeds in unfavourable germination and growth conditions.

Testo completoThe existence of geophagia from as early as 460 BC up to now, makes it relevant to investigate all aspects related to geophagia. Geophagia is a direct route for potential transmission of pathogens to the human host, through the ingestion of soil. Soil-borne diseases in humans are causing growing concern as sewage disposal, which involve sewage sludge and waste water drainage from these plants, is on the increase. It is estimated that approximately seven million tons of sewage sludge is produced annually and that 54% of this sewage sludge is introduced into soil. Data on enteric infection in humans caused by contamination from soil is limited and need further investigation. The aim of the study was, therefore, to collect information on the microbiological presence in geophagic soil in the Qwa-Qwa district. Objectives included the collecting of information regarding various sampling sites in the Qwa-Qwa district and also soil samples sold by vendors, investigation of the prevalence of known human pathogenic bacteria and fungi in geophagic soil, investigating the culturability of Salmonella enteritidis in geophagic soil in comparison with the viability of these organisms in soil for long periods of time, investigating potential antimicrobial activity of geophagic soil, as some of the geophagists are convinced that the geophagic soils have medicinal properties, and to determine the microbial diversity of geophagic soils, which can not be accomplished by conventional microbial culturing methods.

Testo completoENGLISH ABSTRACT: Introduced species displace native species and alter ecological communities, affect agriculture as well as human health and are economically costly to eradicate. Long term monitoring of introduced species including the documentation of levels of genetic variation is therefore of the utmost importance. This study investigated the distribution of genetic variation in two introduced species distributed across South Africa the bud gall-forming wasp Trichilogaster acaciaelongifoliae and the Argentine ant Linepithema humile. The bud gall-forming wasp was introduced into South Africa as a biological control agent to curb the spread of the invasive long-leaved wattle Acacia longifolia. In addition to the intended (target) host, the bud gall-forming wasp has also colonised A. floribunda, a noninvasive ornamental plant. Limited genetic variation was found across South Africa based on the mitochondrial DNA cytochrome oxidase subunit I (COI) gene. Only 3 haplotypes characterized 53 individuals collected from 23 localities (nucleotide diversity £k = 0.002 ¡Ó 0.001, haplotype diversity h = 0.482 ¡Ó 0.045). No significant partitioning of genetic variation was found across South Africa including between host plants (target host = A. longifolia, non target host = A. floribunda) or between the core (sites of introduction) and edge (naturally dispersed) sites (ƒ¶ST = 0.094, P = 0.288). The limited genetic variation and the absence of significant genetic structure are congruent with patterns described for many other introduced species and may suggest that propagule pressure plays only a minor role in species establishment and spread of the gall-forming wasp across South Africa. Mitochondrial and nuclear markers were used to describe the distribution of genetic variation within Argentine ants across their introduced range in South Africa. For the mitochondrial DNA, low genetic diversity was found for the COI gene with only five haplotypes, separated by single mutational changes, characterizing 101 specimens from 35 localities (nucleotide diversity π = 0.001 ± 0.001, haplotype diversity h = 0.151 ± 0.048). Notwithstanding the low levels of genetic diversity, mitochondrial variation was significantly structured (ST = 0.54, P < 0.001) across the landscape. In contrast, microsatellite analyses of 230 ants from 23 localities, employing six polymorphic microsatellite markers, revealed a relatively high amount of genetic diversity (HE = 0.51 ± 0.22). Significant population structure was similarly evident (RST = 0.14, P < 0.001) with the localities of Elim2, Porterville2 and Bloemfontein2 clustering as a distinct population from the remainder of the localities. Importantly, individuals from these localities also had a unique mitochondrial haplotype and, when taken with the nuclear results, may indicate the occurrence of more than one introduction event (and possibly more than one colony) in South Africa. This is further underscored by the presence of unique microsatellite alleles in these three populations. In an attempt to establish the source populations for the introduction of Argentine ants into South Africa, mitochondrial cytochrome b sequences were generated for a subset of ants representing the two major genetic clades across South Africa. A comparison with the published data from across the world including the native range of the Argentine ant in South America grouped Argentine ants from South Africa with three potential source populations namely Ocampo and Rosario in Argentina and Passo do Lontra in Brazil. The results of this study underscore the role of human-mediated dispersal in shaping the levels of genetic variation in both species. Human-mediated dispersal can lead to genetic homogenization across populations.

Langtermyn monitering van indringer spesies asook die dokumentasie van genetiese variasie is dus baie belangrik. Hierdie studie bestudeer die verspreiding van genetiese variasie in twee indringer spesies wat regoor Suid-Afrika voorkom, naamlik die kroongal-vormende wespe Trichilogaster acaciaelongifoliae en die Argentynse mier Linepithema humile. Die kroongal-vormende wesp is na Suid Afrika gebring as biologiese beheeragent om die verspreiding van indringer lang-blarige wattle Acacia longifolia te beveg. Die kroongalvormende wespe het sowel die teiken spesies, asook A. floribunda, ‘n nie-indringer ornamentele plant gekoloniseer. Beperkte genetiese variasie is gevind regoor Suid Afrika gebasseer op die mitochondriale DNA sitokroom-oksidasie subeenheid I (COI) geen. Slegs 3 haplotipes karakteriseer 53 indiviue van 23 bevolkings (nukleotied diversiteit π = 0.002 ± 0.001, haplotiep diversiteit h = 0.482 ± 0.045). Geen beduinde groepering van genetiese variase is gevind regoor Suid Afrika nie (ST = 0.094, P = 0.288). Hierdie bevinding geld onafhanklik van die gasheer plant (teiken gasheer = A. longifolia, nie-teiken gasheer = A. floribunda). Ook is geen beduidende genetiese groepering gevind tussen die sentrale (plek van inisiele blootstelling) en perifêre (natuurlik verspreide) lokaliteite nie. Die kleinskaalse genetiese variase en die afwesigheid van beduidende genetiese struktuur wat hier gevind is, verskil van die patrone wat voorheen vir baie ander indringer-spesies beskryf is. Dit mag daarop dui dat ‘propagule’ druk slegs ’n klein rol speel in spesies-vestiging en verspreiding van die galvormende wespe regoor Suid-Afrika. Mitochondriale asook kern merkers is gebruik om die verspreiding van genetiese variasie in Argentynse miere in Suid Afrika te beskryf. Vir die mitochondriale DNA is lae genetiese variase gevind vir die COI geen, met slegs 5 haplotipes, gedifferensieer deur enkele mutasie veranderinge wat 101 monsters van 35 lokaliteite karakteriseer (nukleotied diversiteit π = 0.001 ± 0.001, haplotiep diversiteit h = 0.151 ± 0.048). Desondanks die lae genetiese variasie, is gevind dat mitochondriale variasie beduidend gestruktureerd is (ST = 0.54, P < 0.001) oor die landskap. Hierteenoor het mikrosatelliet analises van 230 miere van 23 lokaliteite, deur gebruik te maak van ses polimorfiese mikrosatelliet merkers, ’n relatiewe hoë hoeveelheid genetiese diversiteit aangedui. Beduidende bevolkingstruktuur was ook gevind (RST = 0.14, P < 0.001) in die areas Elim2, Porterville2 en Bloemfontein2 wat saam groepeer as eiesoortige bevolkings vergeleke met die res van die areas. Ook van belang is dat individue van die areas ’n unieke mitochondriale haplotipe besit, en in kombinasie met die kern resultate, mag dit die voorkoms van meer as een blootstellingsgeleentheid (en moontlik meer as een kolonie) in Suid Afrika aandui. Hierdie bevinding word verder beklemtoon deur die teenwoordigheid van unieke mikrosatelliet allele in die drie bevolkings. In ‘n poging om die oorsprong van die oorsprong-bevolking vir die blootstelling van Argentynse miere in Suid Afrika vas te stel, is mitochondriale sitokroom b volgordes gegenereer vir ’n substel miere wat die twee hoof genetiese klades in Suid Afrika voorstel. Vergelyking met gepubliseerde data van regoor die wêreld, insluitende die endemiese gebied van die Argentynse mier in Suid Amerika, het die Argentynse mier van Suid Afrika met drie potensiele oorsprong-bevolking verbind, naamlik Ocampo en Rosario in Argentina en Passo do Lontra in Brazil.

Testo completoThe toxicant dichlorodiphenyl-trichloroethane, commonly known as DDT, is a broad spectrum insecticide and is currently banned in most countries due to its toxic effects. However, in some countries restricted use of DDT has been authorized as an effective vector control within malarial control programmes. South Africa is one such country, where spraying of DDT occurs in three provinces including the Limpopo Province, KwaZulu Natal and Mpumalanga. Specifically in the Limpopo Province, spraying of DDT has been ongoing for almost 56 years within the eastern malaria belt of the province. Despite this long term spraying there is still a scarcity of data regarding DDT and its effects on indigenous aquatic organisms in South Africa. Any research regarding DDT will therefore be of the utmost value. It was in this context that the present study was initiated, which primarily aimed to assess the extent of contamination within DDT sprayed areas in South Africa and the associated effects on indigenous species, whilst identifying techniques that could be used in future monitoring of these areas. This assessment was done in the Luvuvhu River catchment at three reference sites and four exposure sites situated within the areas where indoor residual spraying of DDT is done annually. At these sites the extent of DDT contamination within the water, sediment and biota (using the bioindicator pecies C. gariepinus from only the lentic sites) in the Luvuvhu river was evaluated. The results showed that DDT concentrations were well above recommended levels in all three of the measured phases, with the highest concentrations predominantly observed at the Xikundu weir. This site was particularly impacted by DDT due to a combination of its close proximity to the DDT sprayed areas, concentration accumulation from upstream sources and environmental conditions that accentuated contamination. These elevated levels of DDT did, however, not induce significant quantifiable effects in the bioindicator C. gariepinus or in the fish and macro-invertebrate community structures. Specifically, the effects in the catfish, C. gariepinus, were assessed using a range of biomarkers specific to the endocrine disrupting effects of DDT, including indirect measures of vitellogenin (calcium, zinc, magnesium and alkali-labile phosphate (ALP) that are all present on the VTG molecule in high abundances), gonad-somatic index (GSI), condition factor (CF), analysis of covariance (ANCOVA) manipulated gonads, protein carbonyls (PC) and intersex. Although none of these biomarkers could be significantly correlated with the DDT contaminations, DDT was shown to induce a slight sub-organismal effect by slightly inducing the synthesis of ALP and Ca as well as reducing the gonad mass (shown by GSI and adjusted gonad mass biomarkers) and body condition. In contrast, the fish and macroinvertebrate communities showed no conclusive relationship with DDT contamination, using a variety of methodologies, including informal assessments, univariate diversity indices, multivariate statistics, abundance models, fish response assessment index (FRAI) as well as average score per taxon (ASPT) and Ephemeroptera, Plecoptera and Trichoptera (EPT) richness. In conclusion, it was shown that DDT concentrations within the Luvuvhu River only induced effects at the lower levels of complexity, which highlights the importance of the utilisation of biomarkers to measure more subtle long-term effects as compared to the usage of community level effects.

Testo completoThe Olifants River is one of the major river systems in South Africa, providing water for industries, mines, agricultural and domestic activities. Consequently, the river has turned out to be one of the most polluted rivers in South Africa. The Blyde River is an important tributary of the Olifants River, known for its continuous flow and good water quality. However, the recent increase in human activities such as, commercial agricultural and large-scale forestry practices in the catchment may have a negative impact on the quality of the water, which may consequently affect the aquatic biota community structure. The aim of the study was to assess the water and sediment quality of the Blyde River using aquatic macroinvertebrates as bioindicators and the South African Scoring System version 5 (SASS5) protocol was followed. Seven sampling sites were selected at Blyde River and five samples were collected per site. Collected macroinvertebrates were identified to family level and classified according to their tolerance levels to pollution at the University of Limpopo Biodiversity Laboratory. In addition, seasonal readings of the physico-chemical parameters such as, pH, dissolved oxygen (DO), temperature, salinity, total dissolved solutes (TDS) and electrical conductivity (EC) were recorded in situ using YSI Model 554 Datalogger with a 4 m multiprobe and a Mettler Toledo SevenGo™. Most of the physico-chemical parameters recorded at Blyde River were within the South African water quality guidelines for aquatic ecosystems. Metals such as, As, Al, Ba, B, Cd, Cr, Co, Cu, Fe, Pb, Mn, Ni, Se, Ag, Sr, Sn, Ti, V, and Zn were tested from the water column, sediments and macroinvertebrate tissue. Almost all the metals analysed from the water column and sediments were within the quality guidelines. Out of the twenty tested metals, As, Ag, Cu, and Zn exceeded the recommended quality guidelines in the water column at some sites. Only As, Cr and Cu exceeded the recommended quality guidelines in the sediments. The most abundant macroinvertebrates were from the orders Ephemeroptera, Diptera, and Trichoptera at all sites. There was a similar macroinvertebrate community structure across the sites, supported by the positive correlation analysis among the sites (r > 0.4). Wildlife Estate and Wildrivers Estate had the highest and lowest numbers of sensitive macroinvertebrates respectively. The highest percentage of iv sensitive macroinvertebrate taxa was recorded in winter and the highest percentage of tolerant species was recorded in summer. This was also supported by the SASS indices, the SASS score, number of taxa and average score per taxon (ASPT) were highest in winter, followed by autumn, spring and lowest in summer. The overall SASS indices showed that the water quality condition at most of the sites at Blyde River could be considered as good. All the seven sites on average fell under ecological band A and B, except for the Wildrivers Estate, thus, the water quality status of the river could be described as unmodified and largely natural with few modifications.

Testo completoMining operations significantly influence the environment due to direct and indirect discharges of waste products into the aquatic systems. The primary aim of this study was to assess the current situation in the platinum mining area and develop a management plan to ensure that existing and potential environmental impacts caused by platinum mining and processing are mitigated. To do this, an assessment was carried out to investigate changes in critical aquatic invertebrate and fish community distributions and assess how they relate to measured environmental factors. Five sites were selected, one reference site which is upstream of heavy mining activities and four sites within heavy mining and processing activities. Standard techniques for water, sediment, invertebrate and fish sampling were used. Macro-invertebrates sampled were identified to family level whereas fish were identified to species level. Multivariate analysis used was cluster analysis by non-metric multidimensional scaling (NMDS) for both macro-invertebrates and fish. Three methods of ordination were used to analyze the biotic and abiotic data namely N-MDS, Correspondence Analysis (CA) and Canonical Correspondence Analysis (CCA). Cluster analysis of macro-invertebrates data revealed three major groups based on sampling period (low flow or high flow) and the last cluster according to the locality. Multidimensional scaling ordination of high and low flow for macro-invertebrate communities confirmed the groupings detected by cluster analysis. Cluster analysis for fish communities revealed two groups at 50% similarity; the first group is the combination of reference and exposure sites for both high and low flow sampling regimes. No fish were sampled at site 4 during both low and high flow regimes. Multidimensional scaling ordination of high and low flow fish communities confirmed the groupings detected by cluster analysis. Analysis using a similarity profile (SIMPROF) test indicated that fish communities are statistically (p=5%) the same. It was found that macro-invertebrates and fish respond differently to environmental variables.

Testo completoSediment contamination occurs as a result of various anthropogenic activities; mainly through mining-, agricultural- and industrial practices. Many of the contaminants arising from these activities enter the aquatic system and precipitate from the surrounding water, binding to sediment particles. In the sediment compartment, these contaminants reach concentrations much higher than in solution with the overlying water. Even though the quality of the overlying water may prove acceptable in accordance to water quality uidelines, an aquatic system may still be at risk from the contaminated sediment. If the contaminated sediment were to be disturbed through flooding, bioturbation or changes in the water chemistry, these contaminants will desorb into the water column and as a result be detrimental to life forms in contact and dependent on that water source. Monitoring sediment has been a widespread initiative internationally and has led to the development of various sediment toxicity test methods, including different bioassays. This study focused on sediment bioassays such as the Phytotoxkit-F and Ostracodtoxkit-F, and the Diptera bioassay to assess the sediment quality of the Tweelopiespruit-Rietspruit-Bloubankspruit (TRB) river system in Gauteng, South Africa. This river is known to be impacted by acid mine drainage (AMD) since late August 2002. Exposure of the bioassays to river sediment from preselected sampling sites (Site 1, closest to the mine, to Site 6, furthest from the mine, and Site 7, the reference site) provided an eco-toxicological estimation of the acute toxicity emanating from contaminants in the sediment. Physico-chemical analyses revealed high concentrations of metals and other contaminants in the water and sediment. A general linear decrease in contaminant concentrations was observed from Site 1 to Site 6. The results from the bioassays displayed a similar trend, since there were greater sensitivities (mortalities and growth inhibition) to sediments sampled closer to the mine. Due to high levels of contamination in sediments, compared to the overlying water, and the potential impact on aquatic organisms, sediment toxicity monitoring should be a compulsory requirement for environmental studies in South Africa
