Testo completoThe thesis analyzed the role of the arousal as a psychophysiological index of activation, and the application of non-invasive transcranial electrical stimulation (tES) technique with the aim to modulate this index. In this work we investigated if the application of tES could increase the performance of healthy young subjects during cognitive tasks. The thesis is based on 2 main studies for a total of 4 experiments with the recording of behavioural (reaction times, accuracy) and psychophysiological (skin conductance, pupil diameter) indeces. The results showed that it is possible to modulate arousal with bursts of tES, administered during the presentation of salient stimuli for the subject. This modulation resulted in a reduction of reaction times and an increase of the skin conductance response. These data support the possibility to use this protocol of stimulation with patients with attentional and other cognitive deficits in a rehabilitative context.

Testo completoThe microbiota of the large intestine plays a fundamental role in maintaining the state of health of the gastrointestinal tract and the host. Considerable evidence suggests that dietary macronutrients and prebiotic substances may affect microbial composition and activity. Knowledge about the modulation of canine and feline intestinal microbiota in response to macronutrients and prebiotic supplementation is limited. However, such information is necessary to investigate further the complex interplay between host and intestinal microbiota in response to changes of diet. The research for this PhD thesis focused upon the changes of the intestinal microbiota and bacterial metabolites of dogs and cats, in response to feeding diets differing in their protein content and digestibility, and containing or not a source of prebiotics. Additionally, the effects of tannins and Yucca schidigera extract on feline intestinal microbiota were evaluated. Furthermore, aim of the last trial was to evaluate the effect of increasing level of lactose on composition and metabolism of dog intestinal microbiota. The results from the present thesis show that different prebiotics exert different effects on the composition and activity of canine and feline intestinal microbiota and that high dietary protein levels can have negative effects on the animal intestinal environment. However, administration of oligosaccharides doesn’t seem to counteract the negative effects that can be observed when companion animals are fed high-protein diets. In the other study, Yucca extract and tannins does not affect feline fecal microbiota. Despite this, a reduction of odour components was observed due to fermentation of substrates. Finally, results from the final study showed no difference between treatments on canine microbial population. However, proteolytic compounds were decreased by lactose. Future studies must determine how diet composition and prebiotic supplementation affect the gut microbiota and how this information may be used to improve diets and the host’s health.

Testo completoBone is a dynamic tissue, with the ability to adapt to its functional demands and repair itself by bone remodelling. The major effector cells of bone remodelling are osteoclast and osteoblast, they cooperate in order to maintain the balance between bone formation and bone resorption, essential for bone homeostasis. Disruption of this balance can diminish bone mass and micro-architectural integrity of the bone resulting in an increase in bone fragility and susceptibility to fractures, as evident in osteoporosis. It is known that, in the pathophysiology of the bone, a crucial role is played by endocrine and paracrine factors. Recent data suggest that bone remodeling may be influenced by the nervous system. The hypothesis is supported by the presence, in proximity of the bone, of sensory nerve fibers responsible for the release of some neuro peptides, like substance P. Iin capsaicin-treated animal has been shown the direct involvement of the nervous system in the maintenance of bone, this animal showed bone loss and increased bone fragility. For these reasons in recent years has intensified research in this field trying to understand the role of neuropeptides in the process of differentiation of mesenchymal precursors into osteogenic lineage. The mesenchymal stromal cells are undifferentiated multipotent cells present in the bone marrow, adipose tissue, umbilical cord and dental pulp. In these districts, MSC are in a quiescent state until they are required to local repair or tissue regeneration. MSC, suitably stimulated, can differentiate into different types of connective tissue such as, bone, cartilage and adipose. The research was designed to optimize a protocol for ex vivo expansion and to evaluate the effect of substance P, neuropeptide in the sensory endings in the vicinity of the bone, in the process of picking osteogenic.

Testo completoMesenchymal stem cells (MSC) have been isolated from various tissues, including bone marrow, adipose tissue, and dental pulp for regenerative strategies in orthopaedics and dentistry. MSC are critical elements that may contribute to bone healing, either directly as osteoblast precursors or indirectly by producing cytokines and growth factors, and by modulating vascularization and inflammation. The infusion of MSC therefore represents a valuable therapeutic approach to enhance bone healing. However, the success of regenerative strategy may be hampered by an unfavourable microenvironment. The aim of this study was to investigate how physicochemical characteristics of extracellular microenvironment, including oxygen tension and extracellular acidity, can modulate the stemness and the osteogenic potential of MSC. We first examined whether a prolonged exposure to hypoxia affects the osteogenic differentiation of adipose derived mesenchymal stem cells (ASC). The effect of oxygen tension was evaluated on cell proliferation, surface antigens, stemness gene and bone-related genes expression, alkaline phosphatase activity, mineral matrix deposition, and growth factor release. We demonstrated that hypoxia acts dually, promoting ASC proliferation and stemness in the absence of osteogenic stimuli, but also inducing their differentiation in a bone-like milieu. We subsequently investigated the effect of different levels of extracellular pH (6.5, 6.8, 7.1 and 7.4) on stemness and osteogenic differentiation of mesenchymal stem cells from dental pulp (DPSC). We were able to provide consistent evidence that an acidic microenvironment promotes the stemness state of DPSC. Moreover, extracellular acidosis significantly reduces cells growth, and push cells to reside in the quiescent G0 phase of the cell cycle. Finally, we demonstrated the inhibitory effect of acidic pH on the osteogenic potential of DPSC. Modulation of oxygen tension and extracellular pH variations must be considered when MSC are included as a therapeutic tool in bone-related applications.

Testo completoIschemic preconditioning (IPC), the exposure of the heart to short cycles of sublethal ischemia before a prolonged ischemic damage, is a phenomenon able to provide a considerable resistance to myocardial infarct, one of the most prominent cause of disability and death in the world. Recent studies suggest that IPC can improve survival, homing, and engrafment of stem cells in ischemic areas and that it can constitute a new therapeutic strategy to enhance stem cell cardioprotective therapy, a developing research area. IPC is difficult to apply in clinical practice, but is well known that opioids and their receptors are cardioprotective and that they activate signaling pathways involved in IPC; for this reasons opioids are suitable candidate for a possible pharmacological therapy alternative to IPC. Given these assumptions, the purpose of the present study was to investigate whether conditioning cardiomyocytes with opioid receptor agonists Dynorphin B, DADLE e Met-Enkephalin could protect cells from apoptosis caused by an ischemic environment and whether it could induce the damaged cells to produce factors capable to attract stem cells. To tests this hypothesis we developed an in vitro model of “ischemic microenvironment” applied to H9c2 rat cardiomyoblasts. Preconditiong the cells in a “sustained” way (24 hours of opioids preconditioning followed by 24 hours of induction of damage, keeping on administering the opioid peptides) with Dynorphin B and DADLE leads to a direct protection from apoptosis. Thereafter, migration and adhesion assays showed that DADLE drives “ischemic” H9c2 to create a microenvironment capable to attract human Mesenchymal stem cells (FMhMSCs) and to improve FMhMSC grafting abilities. Moreover, the results obtained until now suggest that the ability of the ischaemic microenvironment conditioned with DADLE to attract FMhMSC could be ascribed to chemokines upregulation in H9c2.

Testo completoSince the beginning of the XXI century, Italian and French administrative courts have shown an increasing interest in the protection of legal certainty, which eventually culminated in the adoption of the power to modulate the temporal effects of judicial annulments, along the lines of the powers granted to the European Court of Justice by art. 264, sect. 2 T.F.E.U. An in-depth analysis of Italian and French case law revealed that several legal doctrines aimed at the preservation of unlawful administrative acts from the effects of an annulment had already been developed by domestic Courts prior to the introduction of the power to modulate their decisions, thus demonstrating a long-lasting commitment to legal certainty by the Courts. The new techniques generated different responses in the two sides of the Alps. While in France, they received general approval by legal scholars – at least initially – and their use is now widespread, in Italy they have been almost unanimously deemed in contravention of the guarantees enshrined in article 113 of the Italian Constitution. The dissertation describes and analyses the old and new techniques by which the stability of an unlawful act or of its effects are protected by Italian and French Courts, with the aim of further defining the concept of legal certainty by illustrating what is actually protected by the Court when the principle of legal certainty is acknowledged to prevail on the principle of legality. Furthermore, the dissertation focuses on the impact of these techniques on the justiciability of citizens’ rights, in France and in Italy, as a means to explain the different reception the power of modulation has had in the two Countries.

Testo completoThe aim of this study was to better understand G1 to S transition control in Saccharomyces cerevisiae, studying by a proteomic approach the exponential growth of mutants in FAR1 (involved in cell size control of Start) and nutritional shift-up from poor to rich carbon source of wild type strain. Observing correlation between cell cycle mutant phenotypes and mRNA and protein expression, we looked for possible new actors of the control of growth and cell cycle progression by nutrients in budding yeast. We found that FAR1 gene dosage affects ribosome biosynthesis and the main pathway of carbohydrate metabolism, suggesting a possible feedback control of cell size threshold on these two pathways. Moreover, we identified an unknown function protein, Gvp36, that may be involved in G1 to S transition control, taking part in a pathway which link Far1 to budding
