Testo completoThis doctoral thesis intends to explore the ‘making-of’ of fashion photography, over several decades and geographies. The aim is to operate an epistemological displacement to rethink fashion photography beyond certain interpretations that identified its scope with semiotic, gender or identarian paradigms. Rather, this study proposes to think the ‘making of’ as a result of practices, negotiations, exchanges and hierarchies that characterize – but also move beyond – the very action of producing and publishing a fashion photograph. By shedding light on the multiple and overlooked sources produced around the production and circulation of the fashion image, the research explores three moments in the life of fashion photographs: the making of a fashion photograph; the collecting and conserving practices; and its exhibition. Therefore, the ‘making-of’ is here understood in a broad sense. This term is not only used to identify the descriptions and detailed information showing creative processes behind the production of a fashion photograph or an editorial series, but it also takes into consideration the mechanisms of circulations beyond the industry, the networks and the institutional processes concurring to the formation of the cultural value of an image. In doing so, this thesis sheds light on the polysemic meanings and values of fashion photography, moving beyond an interpretational path that has restricted this practice into an “applied” art in search of artistic legitimization

O presente trabalho reflete sobre a configuração imagética dos indígenas Maká. Tal população, desde o início das relações de intercâmbio com a sociedade circundante, passa a constituir um reflexo da “imagem do índio”. Além disso, passa a aperfeiçoar seus trabalhos na elaboração de peças ornamentais como uma maneira de acompanhar as transformações culturais, econômicas e tecnológicas no cenário onde estão incluídos. Os Maká são originários do Chaco Boreal, pertencentes à família linguística dos Mataco. Por conta da influência do General russo Juan Balaiff, estes povos foram transferidos, em 1944, para uma colônia ao lado ocidental do Rio Paraguai, próxima a Assunção. Atualmente, os Maka estão em quatro comunidades, localizadas na região fronteiriça do país. A partir desses locais, eles se deslocam aos sítios turísticos da região para vender as suas produções. Nesses espaços, os Maká configuram uma encenação da fantasia: refletem a imagem esperada deles. Nos eventos oficiais e folclóricos, esses grupos costumam aparecer como uma referência indígena do Paraguai, representando um componente étnico-racial da nação. Esta investigação entende a produção material como uma reinterpretação do próprio, apropriação e transformação do outro: essa elaboração da imagem pessoal e coletiva responde, portanto, a uma demanda da “diferença”, tanto do setor turístico, como a da sociedade nacional.
Este trabajo reflexiona en torno a la configuración de la imagen del indígena Maká quienes desde el inicio de relacionamiento e intercambio con la sociedad envolvente han sacado a relucir la “imagen de indio”, también han perfeccionado el trabajo manual de elaboración de piezas ornamentales como una forma de adscribirse a las transformaciones culturales, económicas y tecnológicas de los escenarios a los cuales están insertos. Los Maká originarios del Chaco Boreal pertenecen a la familia lingüística Mataco. Mediante influencia de su protector el Gral. ruso Juan Belaieff fueron trasladados en 1944 en una colonia del lado occidental de río Paraguay, en la otra orilla de la capital, Asunción. Actualmente se encuentran asentados en cuatro comunidades, todas ellas ubicadas en fronteras territoriales y desde allí se desplazan a los lugares turísticos donde comercializan piezas propias así como de otras comunidades En estos espacios los Maká configuran la escena de la fantasía, la imagen que se demanda de ellos. Del mismo modo en los eventos oficiales y folclóricos estos aparecen como referentes de lo indígena en el Paraguay ya que son los convocados a representar el componente étnico de la nación paraguaya. Dadas estas características esta investigación entiende que la producción material como la de reinterpretación de lo propio, apropiación y transformación de lo ajeno; esta elaboración de la imagen personal y colectiva responde a una demanda de la “diferencia” tanto del sector turístico como de la sociedad nacional.

Testo completoEpidermal growth factor receptor (EGFR) belongs to the family of protein-tyrosin kinase family, whose activation is associated with the proliferation of malignant cells, invasion, inhibition of apoptosis, tumor angiogenesis and metastatic spread and thus plays an important role in carcinogenesis and tumor progression. Activated mutations take place around the catalytic tyrosine kinase domain. Biopsy, cytological and surgical specimens are used for the detection of EGFR mutations at the time of diagnosis of adenocarcinoma or carcinoma with an adenocarcinoma component, most reliably using a polymerase chain reaction. The fact that the application of molecular tyrosin kinase inhibitor therapy to patients with EGFR mutated lung adenocarcinoma improves the prognosis of the disease, there is resistance in certain types of EGFR mutations and connection with histopathological and immunohistochemical characteristics of tumor, that the bronchoscopic specimen is often the only specimen in which it is necessary to determine the molecular profile of the tumor, the primary objective of this thesis is to determine the frequency and type of EGFR mutations and their connection with the characteristics of adenocarcinoma. In order to realize this objective, the following secondary objectives have been set: to execute histopathological reclassification of lung adenocarcinoma based on the criteria set by the International Association for the Study of Lung Cancer, the American Thoracic Society and European Respiratory Society; determine the expression of TTF-1 in lung adenocarcinoma and connection with EGFR mutation status; determine the frequency, type and connection of EGFR mutations with predominant type of adenocarcinoma and confirm whether bronchoscopic biopsy may be a representative specimen for the determination of EGFR mutation status. Histopathological material of lung adenocarcinoma in surgical specimens is more heterogeneous in relation to biopsy specimens and such difference is statistically significant (p<0,001). Acinar predominant type is the most common in surgical and biopsy specimens with no statistically significant differences in the distribution of predominant type among them (p=0,65883). The predominant type in the primary tumor determines the predominant type in lymphatic metastases. EGFR mutations in the type of insertions on exon 21 and L858R mutations on exon 20 have been detected in three out of 60 (5%) of patients in five out of 120 specimens (three surgical and two biopsy samples), more often in women older than 60, smokers and in a solid predominant type. There are no statistically significant differences in the concentration of isolated DNA between EGFR mutated and wt EGFR adenocarcinoma in biopsy (p=0,132) and surgical specimens (p=0,641). The percentage of invalid results in determining the EGFR mutation status is higher in biopsy specimens compared to the surgical specimens. There is a statistically significant difference between the number of TTF-1 positive and TTF-1 negative adenocarcinoma (p<0,001), but not in the distribution of these patients according to gender (p=0,1231), average age, smoking habits (p=0,6488) and average tumor size (p-0,21). There is a positive correlation between TTF-1 positive expression and EGFR mutation status and therefore TTF-1 positive expression can be a predictor of positive EGFR mutation status. Bronchoscopic biopsy is a representative sample for the determination of EGFR mutation status because: most diagnostic biopsy specimens have more than 100 preserved tumor cells, there is no difference in the distribution of predominant types in relation to surgical specimens, EGFR mutations are detected in samples with less than 100 tumor cells and less than 20% of volume density of tumor tissue, the difference between the concentration of isolated DNA in EGFR mutated and wt EGFR adenocarcinoma in biopsy and surgical specimens is not statistically significant (p=0,132 and p=0,641).

Testo completoThe interest of this PhD-thesis originates from the conjunction of two important questions, the first one being that of representation of Africans in literature - and especially in travel literature and in photos. The thesis aims to show that the representation of the "other" is not always as fixed and as clear as one might think. Parting from different theories of the perception of the "other", we aim to show that the representation of the "other" in these illustrated travel books is very often ambiguous. The second is a question of communication and deals with the combined usage of different types of media (text, photographs, drawings, layout, peritexte (Gérard Genette). It originates thus from intermediality. The work aims to clarify the role of these different types of media in respect to the representation of Africans. Does the combination of these different types of media draw a picture of Africans which is different from that depicted in the text or in the image on its own? It is interesting to look at the added value which might be achieved by the combination of these elements. Does the added value arise from newly created dimensions which could not have been achieved by any one form of media on their own?

Testo completoLung cancer is the leading cause of mortality among all malignancies, and despite advances in diagnostics and therapy over the past 30 years, there has been no significant improvement in the extremely low overall rate of a five-year survival with these patients. At the time of the diagnosis, more than a third of all newly discovered cases are at the IV stage of the disease. Precise and adequate monitoring of the response of the tumor to therapeutic treatment with lung cancer patients in IV stage, as well as the early detection of progression of the disease or inefficiency of therapy in this group of patients is of great importance as chemotherapy is the only therapeutic option for these patients. The existing conventional methods of one-dimensional measurement and assessment of tumor response to therapeutic treatment according to RECIST criteria do not use all the advantages of CT diagnostics and rely on the subjectivity of manual measurements. Advanced radiological techniques, such as volumetry, can contribute to the development of the image monitoring of the therapeutic response of tumors in patients with lung cancer. The aim of this study is to evaluate the application of one-dimensional and volumetric measurement in the assessment of the therapeutic response to lung cancer with multslice computerized tomography. Methodology: A study per type of prospective study included 100 patients with lung cancer at the IV stage of the disease at the time of detection, which were tested in the period between 2013 and 2016 at the Institute of Pulmonary Diseases of Vojvodina in Sremska Kamenica and met the criteria for entering the study. With all patients involved in the study, two radiologists independently assessed all CT chest exams, performed one-dimensional manual and volumetric measurements of selected target lesions, which enabled the determination of intraobserver and interobserver variability and the agreement of the target lesion measurement results using the test method. Based on the results of the performed measurements, the categorization of the therapeutic response of the tumor with conventional RECIST criteria, as well as the application of a modified categorization system for volumetric assessment of the therapeutic response with modified optimal limit values for classification (progression of the disease and positive response to the therapy) was performed, calculated on the basis of the tested sample. Comparison of manual and volumetric estimates of the therapeutic response was made using various classification systems with the determination of the degree of difference in classification and analysis of survival of patients until the progression of the disease. The influence of morphological characteristics of target lesions on the results of volumetric measurement was determined by the analysis of the deviation of the measured tumor volume relative to the arithmetic mean between the groups of lesions of the examined morphological characteristics. Results: The application of volumetric measurements on the test sample leads to a lower rate of variability in the results of measuring the dimensions of the target lesions compared to the conventional manual measurement method, and in the case of interobserver variability (0.9% versus 6.5%) and in terms of intraobserver variability (4.9% to 0.9%). The volumetric assessment of the therapeutic response of the tumor using modified boundary categorization values (3Dindividual model) results in a significantly different classification of the therapeutic response in relation to the use of conventional RECIST criteria. In the case of volumetric assessment of the therapeutic response, the classification of patients using the new “3D-individual” categorization system leads to a misclassification in 22.2% of cases compared to RECIST equivalent to volumetric criteria, reflecting the equally good predictability of PFS in these two systems. The results of the study showed that the appearance of the lesion margins and relation to the surrounding anatomical structures influenced the variability of the results of volumetric measurements. Conclusion: The application of volumetric measurements as a new aspect of the morphological evaluation of lung cancer response to applied therapies can help in making therapeutic decisions both in the treatment of individual patients and in the conduct of clinical trials.

In recent years, commercial cotton lint have been developed with better quality, presenting different characteristics, but with similar coloring. This can be a problem because these samples is identified, large-scale, performed by a visual inspection, which is a very subjective method and error prone. Another way available for classification of samples is the use of HVI system (High Volume Instruments) to determine physical quality parameters. However, this apparatus has a high cost when compared to digital imaging technique, furthermore has the need for adequate infrastructure and a trained analyst for analysis procedure. This work proposes the development of a novel analytical method based on the use of digital image and multivariate analysis to (1) naturally colored cotton plumes classification according to the type of cultivar and (2) simultaneous determination of degree of yellowness (+b), reflectance (Rd) and wax content (WAX). The acquisition of digital images of cotton lints was carried out through a webcam and histograms containing distributions in levels of colors in standard RGB (red-green-blue), grayscale and HSV system (hue-saturation-value) they were obtained. In the classification of samples, models based discriminant analysis by partial least squares (PLS-DA) and linear discriminant analysis (LDA) with variable selection by the successive projections algorithm (SPA) or stepwise (SW) were evaluated. For the determination of the parameters +b, Rd and WAX, PLS models and multiple linear regression (MLR) with variable selection by the SPA were developed and compared. The best classification results were obtained with LDA / SW model with a correct classification rate (TCC) of 96% for the test group using the HSV combination. As the calibration methods, satisfactory prediction results were obtained for both models (PLS and MLR-SPA) with values of RMSEP near repeatability of the reference method. Furthermore, no systematic error was observed and there were no significant differences between the predicted values and reference, according to a paired t-test at 95% confidence. As advantages of the method is simple, low cost, does not use reagent, does not destroy the sample and realizes analysis at short time intervals.
Nos últimos anos, plumas de algodão comerciais têm-se desenvolvido com melhor qualidade, apresentando características diferentes, mas com coloração similar. Isto pode ser um problema porque a identificação destas amostras é, em larga escala, realizada por meio de uma inspeção visual, que é um método subjetivo e sujeito a erros. Outra forma disponível para classificação dessas amostras consiste no uso do sistema HVI (High Volume Instruments) na determinação de parâmetros físicos de qualidade. Contudo, tal equipamento apresenta um alto custo, se comparado a técnica de imagens digitais, além do mais tem-se a necessidade de uma infraestrutura adequada e de um analista treinado para o procedimento de análise. Este trabalho propõe o desenvolvimento de uma nova metodologia analítica baseada na utilização de imagens digitais e análise multivariada para (1) classificação de plumas de algodão naturalmente colorido de acordo com o tipo de cultivar e (2) determinação simultânea de grau de amarelamento (+b), reflectância (Rd) e teor de cera (WAX). A aquisição das imagens digitais das plumas de algodão foi realizada por meio de uma webcam e foram obtidos os histogramas contendo as distribuições nos níveis de cores no padrão RGB (vermelho-verde-azul), escala de cinza e o sistema HSV (matiz-saturação-valor). Na classificação das amostras, modelos baseados na análise discriminante pelos mínimos quadrados parciais (PLS-DA) e análise discriminante linear (LDA) com seleção de variáveis pelo algoritmo das projeções sucessivas (SPA) ou pelo stepwise (SW) foram avaliados. Para a determinação dos parâmetros de +b, Rd e WAX, modelos PLS e regressão linear múltipla (MLR) com seleção de variáveis pelo SPA foram desenvolvidos e comparados. Os melhores resultados de classificação foram obtidos com o modelo LDA/SW, com uma taxa de classificação correta (TCC) de 96% para o conjunto de teste utilizando a combinação HSV. Quanto aos métodos de calibração, resultados de previsão satisfatórios foram obtidos para ambos os modelos (PLS e MLR-SPA), com valores de RMSEP próximos à repetitividade do método de referência. Além disso, nenhum erro sistemático foi observado e não foram encontradas diferenças significativas entre os valores previstos e de referência, de acordo com um teste t-pareado ao nível de confiança de 95%. Como vantagens o método é simples, de baixo custo, não utiliza reagente, não destrói a amostra e realiza análise em curtos intervalos de tempo.

Testo completoThe intensity at which soils provide ecosystem services are function of soil properties that permanently evolved according to numerous processes. Lessivage and bioturbation are of crucial importance as they imply the clay size fraction but are still poorly characterized. This study aims at i) developing a digital 2D image analysis method to quantify both processes intensity, ii) quantifying the effect of two centuries of continuous cultivation and of a decade of organic amendments spreading on their intensity, and iii) characterizing their dynamics. We succeeded in quantifying those processes by carefully considering different levels of soil organization while combining a colorimetric and a textural approach. The percentage volume of worm-worked soil since 10 000 to 15 000 years is 65% at 40 cm depth and between 20 and 30% at 150 cm depth that corresponds to a soil mass flow of 6 500 t.ha-1, i.e. 1 700 t.ha-1 of clay size fraction. Illuviation is responsible for a clay size fraction mass flow of 1 100 t.ha-1. On a time scale as short as two centuries, cultivation was found to induce i) a change of the soil poral network characteristics until 1 meter depth, ii) a modification of the structure of the worm-worked soil volume, and finally iii) an increase of the lessivage intensity. A decade of organic matter spreading tended to lower the intensity of lessivage. Finally, our study points out the fact that soils are highly reactive and that our method may be particularly helpful to predict soil evolution while facing climate change among others

Testo completoAs células estaminais embrionárias (ESC) apresentam como principais características a capacidade de autorrenovação e pluripotência. Refira-se que a manutenção da pluripotência destas células in vitro constitui um dos principais desafios desde que as ESC foram pela primeira vez isoladas em cultura. As ESC em cultura necessitam de fatores específicos responsáveis quer pela ativação de vias de sinalização de pluripotência quer pela inibição de vias de diferenciação. Numerosos moduladores bioquímicos e mecânicos foram já implicados no controlo do estado de pluripotência das células, mas o grau de influência mútua entre tais moduladores, responsável pela referida capacidade de pluripotência / diferenciação das ESC, continua por demonstrar. O trabalho desenvolvido nesta tese teve como objetivo desacoplar o efeito de moduladores bioquímicos e mecânicos na regulação das ESC. De modo a avaliar a influência que os diversos moduladores possuem na expressão genética das ESC, testaram-se os efeitos de diferentes moduladores bioquímicos – efeito de LIF (do inglês leukemia inhibitory factor) e moduladores da cadeia respiratória mitocondrial – e mecânicos – rigidez do substrato. Para tal, começou-se por realizar múltiplas combinações com os diferentes estímulos. Por fim, o efeito de cada modulador na expressão genética das ESC e a inter-relação entre combinações de moduladores foi determinado. Como esperado, a expressão genética das ESC foi alterada quando os moduladores foram combinados. A magnitude do efeito do LIF (- LIF vs. + LIF) mostrou-se dependente do inibidores da cadeia respiratória mitocondrial Antimicina A (AA), mas independente da ativação indireta da fosforilação oxidativa através do uso de galactose. No entanto, a magnitude do efeito dos moduladores da cadeia respiratória mitocondrial mostrou-se dependente da presença de LIF. Inesperadamente, a magnitude do efeito da rigidez do substrato (substratos rígidos vs. substratos menos rígidos) na expressão genética das ESC não se mostrou dependente de qualquer modulador bioquímico testado. Estes resultados sugerem que a rigidez do substrato e os vários moduladores bioquímicos regulam a pluripotência das ESC através de vias de sinalização independentes sendo que a rigidez do substrato se assume como um modulador chave na regulação das ESC. Em resposta à normal heterogeneidade das ESC em cultura assim como ao aumento dos ensaios de pluripotência baseados em imagem e escassez de procedimentos para a quantificação automática da pluripotência associados aos mesmos, a presente tese descreve igualmente o desenvolvimento de um software de código aberto, designado por Pluri-IQ, para quantificar a pluripotência das ESC em cultura. O software Pluri-IQ segmenta automaticamente colónias de ESC com elevada precisão, sendo capaz de utilizar imagens de contraste de fase ou de fluorescência para tal. Pluri-IQ minimiza a necessidade de intervenção humana no seu funcionamento, conseguindo quantificar a percentagem de colónias pluripotentes, mistas e diferenciadas após ensaios de fosfatase alcalina ou de imunocitoquímica com anticorpos específicos para pluripotência. Destaca-se ainda a sua capacidade de análise de uma imagem única ou de imagens múltiplas combinadas, de microscopia de baixa ampliação, com uma exatidão de cerca de 90 %. Em suma, os resultados apresentados nesta tese permitiram identificar a rigidez do substrato como um modulador chave na regulação das ESC, sendo o seu efeito independente de estímulos bioquímicos de pluripotência. Por outro lado, o desenvolvimento de um software de código aberto para quantificação de pluripotência em imagens de ESC, reúne as características necessárias para a sua utilização como uma ferramenta de análise rotineira capaz de aumentar a imparcialidade e a reprodutibilidade de dados obtidos neste tipo de estudos.
Self-renewal and pluripotency are the two major hallmarks of embryonic stem cells (ESC). Since ESC were first isolated in culture, one of the big challenges has been the maintenance of their pluripotency in vitro. ESC cultures require specific factors, which are responsible for the activation of pluripotency pathways and inhibition of differentiation pathways. A plethora of biochemical and mechanical modulators have been shown to individually affect the pluripotency state. However, how these different modulators work together to determine the pluripotency/differentiation state of ESC remains to be established. The work developed in this thesis aimed at uncoupling the biochemical and mechanical modulators effect in ESC fate. To understand how different modulators regulate pluripotency, the ESC gene expression was evaluated upon the effect of different biochemical modulators – leukemia inhibitory factor (LIF) and mitochondria respiratory chain modulators – and mechanical modulators – substrate stiffness. First, multiple combinations of the different stimuli were performed. Then, the effect of each modulator in ESC gene expression and the inter-relationship between paired modulators was determined. As expected, when two modulators were combined, ESC gene expression changed. Nevertheless, the magnitude of LIF effect (- LIF vs. + LIF) was dependent on mitochondria respiratory chain inhibitor Antimycin A (AA). However, it was independent of the indirect activation of oxidative phosphorylation through galactose. Interestingly, the magnitude of mitochondria respiratory chain modulators effect was dependent on the presence of LIF. Unexpectedly, the magnitude of stiffness effect (stiff substrates vs. soft substrates) in gene expression was not dependent on any biochemical pluripotent modulator tested. These results suggest that stiffness and biochemical modulators regulate ESC pluripotency through non-interfering pathways, where stiffness is a key modulator in the regulation of ESC fate. Challenged by the common heterogeneity of ESC culture, the increase of image-based-assays to evaluate pluripotency, and the lack of image analysis approaches available to automatically quantify pluripotency, we developed an open-source software, Pluri-IQ, to quantify ESC pluripotency in culture. Pluri-IQ was designed to automatic segment ESC colonies in both phase-contrast and fluorescence images, with high precision. With a low user input required, Pluri-IQ was able to quantify the percentage of pluripotent, mixed and differentiated colonies after alkaline-phosphatase, or pluripotent antibodies staining assays. Moreover, it was capable to analyze single or multiple low magnification images, with an accuracy around 90%. In summary, the results presented in this thesis identify the stiffness as a key role in ESC fate, working independently of biochemical pluripotent cues. In addition, a new free-software for ESC image quantification was developed, which has the potential to be routinely used, increasing unbiased data acquisition and reproducibility.
