The lipid raft proteins reggie-1 and -2 (flotillins) are implicated in membrane protein trafficking but exactly how has been elusive. We find that reggie-1 and -2 associate with the Rab11a, SNX4, and EHD1–decorated tubulovesicular recycling compartment in HeLa cells and that reggie-1 directly interacts with Rab11a and SNX4. Short hairpin RNA–mediated down-regulation of reggie-1 (and -2) in HeLa cells reduces association of Rab11a with tubular structures and impairs recycling of the transferrin–transferrin receptor (TfR) complex to the plasma membrane. Overexpression of constitutively active Rab11a rescues TfR recycling in reggie-deficient HeLa cells. Similarly, in a Ca2+ switch assay in reggie-depleted A431 cells, internalized E-cadherin is not efficiently recycled to the plasma membrane upon Ca2+ repletion. E-cadherin recycling is rescued, however, by overexpression of constitutively active Rab11a or SNX4 in reggie-deficient A431 cells. This suggests that the function of reggie-1 in sorting and recycling occurs in association with Rab11a and SNX4. Of interest, impaired recycling in reggie-deficient cells leads to de novo E-cadherin biosynthesis and cell contact reformation, showing that cells have ways to compensate the loss of reggies. Together our results identify reggie-1 as a regulator of the Rab11a/SNX4-controlled sorting and recycling pathway, which is, like reggies, evolutionarily conserved.

The reggie protein family consists of two proteins, reggie-1 and -2, also called flotillins, which are highly ubiquitous and evolutionarily conserved. Both reggies have been shown to be associated with membrane rafts and are involved in various cellular processes such as T-cell activation, phagocytosis and insulin signalling. However, the exact molecular function of these proteins remains to be determined. In addition, the mechanism of membrane association of reggie-1, which does not contain any transmembrane domain, is not known. In this study, we have produced a fusion protein of reggie-1 with enhanced green fluorescent protein and generated targeted substitutions for the inactivation of putative palmitoylation and myristoylation sites. We were able to show that reggie-1 is myristoylated and multiply palmitoylated and that lipid modifications are necessary for membrane association of reggie-1. Overexpression of reggie-1 resulted in the induction of numerous filopodia-like protrusions in various cell lines, suggesting a role for reggie-1 as a signalling protein in actin-dependent processes.

Abstract (sommario):
Reggie-1 and reggie-2 are two evolutionarily highly conserved proteins which are up-regulated in retinal ganglion cells during regeneration of lesioned axons in the goldfish optic nerve. They are located at the cytoplasmic face of the plasma membrane and are considered to be 'lipid raft' constituents due to their insolubility in Triton X-100 and presence in the 'floating fractions'; hence they were independently named flotillins. According to our current view, the reggies subserve functions as protein scaffolds which form microdomains in neurons, lymphocytes and many other cell types across species as distant as flies and humans. These microdomains are of a surprisingly constant size of less than or equal to 0.1 mm in all cell types, whereas the distance between them is variable. The microdomains co-ordinate signal transduction of specific cell-surface proteins and especially of GPI (glycosylphosphatidylinositol)-anchored proteins into the cell, as is demonstrated for PrPc (cellular prion protein) in T-lymphocytes. These cells possess a pre-formed reggie cap scaffold consisting of densely packed reggie microdomains. PrPc is targeted to the lymphocyte reggie cap when activated by antibody cross-linking, and induces a distinct Ca2+ signal. In developing zebrafish, reggies become concentrated in neurons and axon tracts, and their absence, after morpholino antisense RNA-knockdown, results in deformed embryos with reduced brains. Likewise, defects in Drosophila eye morphogenesis occur upon reggie overexpression in mutant flies. The defects observed in the organism, as well as in single cells in culture, indicate a morphogenetic function of the reggies, with emphasis on the nervous system. This complies with their role as scaffolds for the formation of multiprotein complexes involved in signalling across the plasma membrane.

Abstract (sommario):
Reggie-1 and -2 proteins (flotillin-2 and -1 respectively) form their own type of non-caveolar membrane microdomains, which are involved in important cellular processes such as T-cell activation, phagocytosis and signalling mediated by the cellular prion protein and insulin; this is consistent with the notion that reggie microdomains promote protein assemblies and signalling. While it is generally known that membrane microdomains contain large multiprotein assemblies, the exact organization of reggie microdomains remains elusive. Using chemical cross-linking approaches, we have demonstrated that reggie complexes are composed of homo- and hetero-tetramers of reggie-1 and -2. Moreover, native reggie oligomers are indeed quite stable, since non-cross-linked tetramers are resistant to 8 M urea treatment. We also show that oligomerization requires the C-terminal but not the N-terminal halves of reggie-1 and -2. Using deletion constructs, we analysed the functional relevance of the three predicted coiled-coil stretches present in the C-terminus of reggie-1. We confirmed experimentally that reggie-1 tetramerization is dependent on the presence of coiled-coil 2 and, partially, of coiled-coil 1. Furthermore, since depletion of reggie-1 by siRNA (small interfering RNA) silencing induces proteasomal degradation of reggie-2, we conclude that the protein stability of reggie-2 depends on the presence of reggie-1. Our data indicate that the basic structural units of reggie microdomains are reggie homo- and hetero-tetramers, which are dependent on the presence of reggie-1.

Using confocal laser scanning and double immunogold electron microscopy, we demonstrate that reggie-1 and -2 are colocalized in ≤0.1-μm plasma membrane microdomains of neurons and astrocytes. In astrocytes, reggie-1 and -2 do not occur in caveolae but clearly outside these structures. Microscopy and coimmunoprecipitation show that reggie-1 and -2 are associated with fyn kinase and with the glycosylphosphatidyl inositol-anchored proteins Thy-1 and F3 that, when activated by antibody cross-linking, selectively copatch with reggie. Jurkat cells, after cross-linking of Thy-1 or GM1 (with the use of cholera toxin), exhibit substantial colocalization of reggie-1 and -2 with Thy-1, GM1, the T-cell receptor complex and fyn. This, and the accumulation of reggie proteins in detergent-resistant membrane fractions containing F3, Thy-1, and fyn imparts to reggie-1 and -2 properties of raft-associated proteins. It also suggests that reggie-1 and -2 participate in the formation of signal transduction centers. In addition, we find reggie-1 and -2 in endolysosomes. In Jurkat cells, reggie-1 and -2 together with fyn and Thy-1 increase in endolysosomes concurrent with a decrease at the plasma membrane. Thus, reggie-1 and -2 define raft-related microdomain signaling centers in neurons and T cells, and the protein complex involved in signaling becomes subject to degradation.

Fish--in contrast to mammals--regenerate retinal ganglion cell axons when the optic nerve is severed. Optic nerve injury leads to reexpression of proteins, which typically are first expressed in newly differentiated retinal ganglion cells and axons. Here we identified two new proteins of fish retinal ganglion cells, reggie-1 and reggie-2, with monoclonal antibody M802 and molecular cloning techniques. In normal fish, M802 stained the few retinal axons derived from newborn ganglion cells which in fish are added lifelong to the retinal margin. After optic nerve injury, however, M802 labeled all retinal ganglion cells and retinal axons throughout their path into tectum. Consistent with M802 staining, reggie-1 and reggie-2 mRNAs were present in lesioned retinal ganglion cells, as demonstrated by in situ hybridization, but were not detectable in their normal mature counterparts. In western blots with membrane proteins of the adult goldfish brain, M802 recognizes a 48x10(3) Mr protein band. At the amino acid level, 48x10(3) Mr reggie-1 and reggie-2 are 44% identical, lack transmembrane and membrane anchor domains, but appear membrane associated by ionic interactions. Reggie-1 and reggie-2 are homologous to 35x10(3) Mr ESA (human epidermal surface antigen) but are here identified as neuronal surface proteins, present on newly differentiated ganglion cells at the retinal margin and which are reexpressed in mature ganglion cells upon injury and during axonal regeneration.

Podosomes are actin-based matrix contacts in a variety of cell types, most notably monocytic cells, and are characterized by their ability to lyse extracellular matrix material. Besides their dependence on actin regulation, podosomes are also influenced by microtubules and microtubule-dependent transport processes. Here we describe a novel role for KIF9, a previously little-characterized member of the kinesin motor family, in the regulation of podosomes in primary human macrophages. We find that small interfering RNA (siRNA)/short-hairpin RNA–induced knockdown of KIF9 significantly affects both numbers and matrix degradation of podosomes. Overexpression and microinjection experiments reveal that the unique C-terminal region of KIF9 is crucial for these effects, presumably through binding of specific interactors. Indeed, we further identify reggie-1/flotillin-2, a signaling mediator between intracellular vesicles and the cell periphery, as an interactor of the KIF9 C-terminus. Reggie-1 dynamically colocalizes with KIF9 in living cells, and, consistent with KIF9-mediated effects, siRNA-induced knockdown of reggies/flotillins significantly impairs matrix degradation by podosomes. In sum, we identify the kinesin KIF9 and reggie/flotillin proteins as novel regulators of macrophage podosomes and show that their interaction is critical for the matrix-degrading ability of these structures.

In different transgenic mouse models Aβ accumulates inside the cells and mice develop behavioral deficits well before visible extracellular β-amyloid accumulation. Here we show that intracellular Aβ accumulates in flotillin-1 positive endocytic vesicles. We also demonstrate that flotillin-1 is not only associated with intracellular Aβ in transgenic mice but also with extracellular β-amyloid plaques in AD patient brain sections

Abstract (sommario):
Syftet med denna studie är att få en fördjupad förståelse av hur en förskola kan arbeta med ämnena naturvetenskap och teknik på ett genomtänkt sätt. Avsikten med studien är också att bidra med mer kunskap om på vilka olika sätt pedagoger kan ta tillvara och utveckla barnens intresse för teknik och fysik. Studien är kvalitativ och har genomförts genom att studera dokumentation från sociala medier som den studerade förskolan publicerat. Dokumentationen har sedan varit en grund för de semistrukturerade intervjuer som genomfördes. För att komma fram till resultatet som gjorts har den fenomenografiska analysmodellen använts. Resultatet som framkom ifrån de tre intervjuer som gjordes, visar att pedagogerna på denna förskola inte ser några hinder med att arbeta med ämnena naturvetenskap och teknik med de allra yngsta barnen, 1–3 år, trots att de saknar en formell fördjupad utbildning inom ämnena. Pedagogerna ser bara möjligheter. Förskolan arbetar Reggio Emilia inspirerat och har arbetat fram en praktikgemenskap, där de skapar goda förutsättningar för undervisning i naturvetenskap och teknik.
The aim of this study is to gain an in-depth understanding of how a preschool can work with the subjects of science and technology in a well-thought-out way. The purpose of the study is also to provide more knowledge about which different ways pedagogues can use and develop the children's interest in technology and physics. The study is qualitative and has been carried out by studying documentation from social media published by the preschool. The documentation has since been a basis for the semi-structured interviews that were conducted. To reach the result, the phenomenographic analysis model has been used. The result shows that the three teachers interviewed at this preschool see no obstacles to working with the subjects of science and technology with the youngest children, 1-3 years, even though they lack a formal in-depth education in the subjects. The preschool teachers only see opportunities. To create the best conditions for teaching science and technology the preschool has worked out a community of practice inspired by Reggio Emilia.

Abstract (sommario):
The key role of toys and play in early years education has been highlighted by several childhood pedagogues such as Froebel, Montessori, Weikart, and Malaguzzi, among many others. It is consensual among the international educational community that children now spend far more time being instructed and tested in literacy and math than they do learning through play and exploration exercising their bodies and using their imagination. This chapter aims to reflect on the power of play for children's learning and development and to analyze how three pedagogical models—the High Scope, Reggio Emilia, and Montessori—integrate play through their curriculum development. The chapter is organized in several topics, namely (1) the role of play in early childhood education (0 to 6 years); (2) the High Scope curriculum and opportunities given to children to engage in free play and play with purposes; (3) the Reggio Emilia approach: play through 100 languages; (4) the Montessori method, from hands-on activity and self-directed activity to collaborative play; (5) final remarks.

This paper presents the EVITA electric car. EVITA is the acronym of Electric Vehicle Improved by Three-phase Asynchronous cooled motor. It is a research project developed jointly by RGEngineering and University of Modena and Reggio Emilia. It aims to produce a novel electric power train with the capability of solving three fundamental problems of today commercial electric vehicles: 1. direct torque dependency of the rotation speed, and its reduction at high speed regimes; 2. electric motors performances reduction due to the overheating effects under heavy load conditions; 3. acclimatization of the car cabin interior in winter times.

The present paper deals with the estimation of the short-term extreme motions of a spar floating wind turbine in parked rotor conditions, through a 1:30 at-sea experiment, carried out at the Natural Ocean Engineering Laboratory (NOEL) of Reggio Calabria (Italy). Thanks to some favorable local environmental conditions of the site, several wind-generated sea states with relatively low significant wave height (Hs < 0.50 m) have been collected during the experiment. These sea states are scale models of ocean storms, which are relevant hydrodynamic design conditions for the spar platform. The 30-minutes extreme values of the model structure motions have been estimated for all the six degrees of freedom, using the Weibull Tail Method (WTM), and the results obtained are presented in the paper. Such estimations are 1:30 scale models of the 3-hours extreme values of the spar motions in parked rotor conditions and may be directly used for design purposes.

In the last decades, the research has directed its efforts and resources paper is to investigate towards the possibility to incorporate wave energy converters, into the traditional maritime breakwaters to combine classical use with new opportunities and developments (for example, the Green Ports). Since the nineties, the OWC (Oscillating Water Column) plants were developed at full scale to produce electrical power from ocean waves. For instance, a new plant was built in Mutriku (Spain) recently. A new kind of OWC caisson, named U-OWC or REWEC3, which has the advantage to obtain an impressive natural resonance without any device for phase control, has been patented by Boccotti [1]. This new U-OWC device gives performances better than those of a conventional OWC either with small wind waves or with high waves [2,3,4]. The properties of the REWEC3 have been verified with two small-scale field experiments carried out in the natural ocean engineering laboratory NOEL of Reggio Calabria off the eastern coast of the Sicily Channel [5–7]. The aim of the present two sites along Italian coasts for possible installations of REWEC3 devices: i) the port of Civitavecchia (Rome, Italy)in the Tyrrhenian sea; ii) the port of Pantelleria, in the Sicily Channel.

In breakwaters embodying an OWC connected to the sea through a vertical duct or through a small opening, the oscillations of the water column are due to the wave pressure acting on the outer opening of the vertical duct or on the small opening. In fact, in neither of the two cases waves can enter the plant, like it happens in conventional OWCs. The additional vertical duct extends along the wave-beaten wall, giving the device the characteristic form of a U-conduit; for this reason they were also named U-OWC. Experiments on a small-scale breakwater embodying a U-OWC were carried out in the natural laboratory of Reggio Calabria. The plant is a 1:10 scale model of a breakwater suitable for the North-East Pacific coast. The paper describes new experiments on the U-OWC device connected to a monoplane Wells turbine. During an intensive measurement campaign, more than 260 sea states, 5 min long were recorded in order to characterize the energy conversion process. From the experiments, the analysis of the energy conversion shows that: 1) the system is able to absorb a share up 80% of the incident wave energy; this result is similar to that obtained in previous experiments carried out without the turbine; 2) a large fraction of the energy entering the U-OWC is converted in pneumatic power acting on the turbine, being head losses in the water flow limited; 3) the efficiency of conversion of the pneumatic power in turbine power is relatively low (about 36% of the pneumatic power) due to the small dimensions of the turbine that lead to low Reynolds number and large influence of secondary losses.
