An evaluation of the effects of soil structural heterogeneity on maize (<I>Zea mays</I> L.) root system architecture was carried out on plants grown in boxes containing fine soil and clods. The clods were prepared at two levels of moisture (0.17 and 0.20 g/g) and bulk density (ranges 1.45–1.61 g/ml and 1.63–1.79 g/ml). Soil moisture directly affected the probability of clod penetration by maize roots. Primary roots inside the clods manifested morphological deformations in the form of bends. We observed a significant increase of bends per root length at lower soil moisture (<I>P</I> = 0.02). Root diameter and branching density increased, and lateral root length decreased considerably inside the clods. However, once emerging out of the clods and into free soil, values of all three characteristics remained low. While changes in root diameter were caused mainly by clod moisture (<I>P</I> < 0.05), length of lateral roots was related to bulk density (<I>P</I> < 0.01). Branching density was modified exclusively by an interactive effect of both factors (<I>P</I> < 0.05).

Four treatments, no sexual preparation (control), 10 min restraint, three false mounts and 10 min restraint followed by three false mounts, were replicated with eight sexually mature buffalo bulls, before the first and second ejaculates to study their effects on the seminal characteristics of buffalo bulls. Seminal characteristics of both first and second ejaculates were improved significantly by sexual stimulation. Animals allowed no sexual stimulation yielded inferior first ejaculates than second ejaculates in all seminal characteristics. First ejaculates, in this case, served as stimulus to second ejaculates and contributed to its superiority. Moderate sexual stimulation (R or FM) before ejaculation improved the first ejaculates and reduced the difference between them and second ejaculates in all seminal characteristics. However, high degrees of sexual stimulation (R or FM) caused the superiority of the first ejaculates to the second ejaculates in seminal characteristics.

The study objective is to compare the biochemical characteristics of seminal fluid in men with astheno- and teratozoospermia and in men with idiopathic infertility. Materials and methods. We examined 207 patients aged 25 to 45 years. The level of total protein, albumin, alanine aminotransferase, aspartate aminotransferase, gamma-glutamyl transferase in seminal fluid was determined on an automatic biochemical analyzer. Results. Similar changes in the biochemical profile of the seminal fluid, in particular, a decrease in the level of albumin, were revealed in infertile men and men with infertility of unclear genesis. Conclusion. Biochemical characteristics of seminal fluid (level of proteins and enzymes) can serve as an indirect marker of male infertility.

Over the past few decades, MRI of the prostate has made great strides in improving cancer detection and is being embraced by more clinicians each day. This article aims to review the imaging characteristics of common and uncommon, but consequential lesions involving the seminal vesicles (SV), as seen predominantly on MRI. Many of these findings are seen incidentally during imaging of the prostate. Anatomy and embryology of the SV will be described which will help illustrate the associations of abnormalities seen. Congenital, infectious, neoplastic, and tumor mimics will be explored in detail, with discussion on clinical presentation and treatment strategies.

Testicular degeneration is a multifactorial process increasing the concentration of prostaglandins in seminal plasma. Both increase and decrease of these hormones tend to promote loss of seminal quality. Flunixin meglumine is a potent anti-inflammatory drug capable of modulating the production of prostaglandins and is widely used in female reproduction. However, it is rarely used in males with the same objective. Therefore, this study aimed to evaluate the effects of this drug on seminal quality of male lambs and goats with spermatic characteristics unfavorable for reproduction. To this end, a total of 15 breeding animals were evaluated, of which six goats and four sheep with poor seminal quality were selected according to the criteria established by the Brazilian College of Animal Reproduction (CBRA). Three semen samples were collected from each animal. Then, the flunixin meglumine treatment was initiated and ejaculates were collected at two different periods after the drug was administered (from day 21 until day 35 and from day 49 to day 63). Macroscopic and microscopic parameters were assessed in semen samples and scrotal circumference and percentages of sperm pathologies were measured and compared between the three periods. Data with normal distribution were analyzed using ANOVA at 5% probability, and comparisons between periods within the same species were performed using the Tukey test. An improvement was observed in the analyses of mass motility, percentage motility, and sperm vigor. Scrotal circumference had no variation. Concerning sperm pathologies, an increase in the number of normal spermatozoids was observed due to a significant reduction in minor and major defects, and the latter remained low even after the treatment was finished. Therefore, flunixin meglumine presented beneficial effects on seminal parameters of male goats and lamb with unfavorable spermatic characteristics. These findings indicate this drug may be used in the treating of males with testicular injuries or of breeders with reduced seminal quality due to advanced age.

This study aimed to determine the influence of mineral supplementation on the concentration of macroelements and microelements in the seminal plasma and the semen characteristics of young buffaloes. To this end, 60 animals were kept under rotational grazing and were randomly divided into two groups: G-Bov (conventional mineral supplement recommended for bovine cattle; n = 30) and G-Buf (mineral supplement recommended for buffaloes; n = 30). Mineral supplement consumption was calculated from the difference between the amount of supplement offered and the leftover bunk, calculated every 28 days. Eight ejaculates from each animal were collected. The means of the response variables were analyzed using the Kruskal-Wallis test, with a significance level of 5%, and Pearson’s correlation was analyzed between the concentration of minerals in seminal plasma and fresh semen characteristics. Animals in the G-Buf group had lower mineral supplement consumption (G-Bov = 83.18 vs. G-Buf = 77.14 g AU-1 day-1, P < 0.05) and had higher ejaculate volume, sperm motility, sperm vigor, and sperm mass motility than did the animals in the G-Bov group. This research presented the concentration of macroelements and microelements in the seminal plasma of buffaloes receiving different mineral supplements. In addition, the study concluded that the physical and morphological characteristics of the semen of young buffaloes are influenced by the formulation of the mineral supplement, which resulted in better sperm quality in the animals receiving a species-specific supplement. Among the minerals present in buffalo seminal plasma, phosphorus is the element that shows the highest positive correlation with semen characteristics.

ABSTRAK. Penelitian ini bertujuan untuk menentukan karakteristik dan komposisi plasma semen sapi aceh. Sampel semen dikoleksi dari 2 ekor sapi aceh jantan sehat berumur 3,0-3,5 tahun menggunakan vagina buatan. Koleksi semen dilakukan satu kali dalam seminggu selama enam minggu. Evaluasi karakteristik semen meliputi volume, warna, konsistensi, pH, konsentrasi spermatozoa, motilitas, viabilitas, abnormalitas spermatozoa dan komposisi kimia plasma semen. Data kualitas dan komposisi kimia plasma semen yang diperoleh dianalisa secara diskriptif. Hasil penelitian menunjukkan semen segar sapi aceh memiliki volume 3,82 ± 0,47 ml, warna krem keputihan, konsistensi kental, pH 6,84 ± 0,17, konsentrasi 1194 ± 4,80 × 106sp/ml, persentase motilitas 77,28 ± 3,17 % dan abnormalitas 5,98 ± 1,77%. Plasma seminalis semen segar sapi aceh mengandung protein total sebanyak 1,51 ± 0,21 g/100ml, Kalium 75,80 ± 22,63 mg/100 ml dan Magnesium 32,30 ± 17,64 mg/100 ml lebih tinggi, tetapi Natrium 170,00 ± 81,60 mg/100 ml; kalsium 40,00 ± 12,76 mg/100 ml, Fosfor 16,70 ± 6,17 mg/100 ml, dan Mangan 7,08 ± 2,48 mg/100 ml lebih rendah dibandingkan dengan sapi potong. Secara umum kuantitas dan komposisi plasma seminalis semen sapi aceh yang dihasilkan cukup baik, dan memenuhi syarat untuk diproses sebagai semen beku. (The characteristics of semen and chemical composition of the seminal plasma of aceh cattle maintaned in BIBD Saree Aceh Besar) ABSTRACT. This study aim were to determine characteristics and composition of semen plasma Aceh cattle. Semen samples were collected from two healthy Aceh bulls that aged from 3.0 to 3.5 year. Samples were yielded by using an artificial vagina. Samples collecting were conducted once a week for six consecutive weeks. Data of semen quality and chemical composition of seminal plasma were analyzed descriptively. The evaluation showed that Aceh Cattle semen had 3.82 ± 0.47 ml volume, beige whitish color, thick consistency, 6.84 ± 0.17 pH, 1194 ± 4.80×106 sp/ml sperm concentration, 77.28 ± 3.17% motility and 5.98 ± 1.77% abnormal spermatozoa. Plasma seminalis fresh semen of Aceh Cattle contained 1,51 ± 0,21 g/100ml total protein; 75,80 ± 22,63 mg/100ml potassium and 32,30 ± 17,64 mg/100ml magnesium higher than the heifer, but 170,00 ± 81,60 mg/100ml sodium; 40,00 ± 12,76 mg/100ml calcium, 16,70 ± 6,17 mg/100ml fosfor, and 7,08 ± 2,48 mg/100ml mangan lower. It was concluded that quality and composition of seminal plasma Aceh cattle semen could be made as frozen semen.

This study aimed to evaluate donkey seminal plasma metabolites and relate this information to the main characteristics of sperm quality. Sperm kinetics from 10 donkey stallions were analyzed with a computerized system at the time of collection (T0) and after 24 h storage at 4 °C (T24). Seminal plasma was frozen at −80 °C for subsequent proton nuclear magnetic resonance (1H NMR) spectroscopy. On three stallions, semen collection was repeated monthly for three times and sperm analysis also included mitochondrial activity and oxidative status. One stallion was azoospermic and a second semen collection was performed after one month. In the seminal plasma, 17 metabolites were identified; their levels showed numerous significant variations between the azoospermic and the normospermic individuals and grouped in well-defined clusters in a multivariate analysis. Comparing individuals with high and low sperm motility, the only discriminating metabolite was phenylalanine, whose levels were lower in the latter, as in the azoospermic individual. Phenylalanine was also the only metabolite highly correlated with all sperm kinematic parameters at T24. In conclusion, the present study has provided relevant information on the chemical characteristics of donkey semen, identified relationships between seminal metabolites, semen parameters, and sperm kinetics, and offered insights for future technological applications.

Characteristics of selected seminal plasma proteins and their application in the improvement of the reproductive processes in mammals Understanding the biochemical processes associated with ovum fertilization and knowledge about the structure and function of individual substances participating in these processes is crucial for the development of biotechnological methods to improve reproduction of animals and humans. Among many components of seminal plasma, proteins and peptides play a specific role in regulation of the fertilization process, particularly through their ability to bind various types of ligands such as polysaccharides, lipids and ions. Heparin-binding proteins regulate capacitation and acrosome reaction processes. Affinity of plasma proteins to mannans of the fallopian tube epithelium facilitates formation of spermatozoa reservoirs in the female reproductive tract. Ability to bind phosphorylcholine is one of the conditions for the coating of the seminal plasma proteins on the sperm membrane and also determines the formation of oligomeric forms of certain proteins. Zinc binding by seminal plasma proteins regulates sperm chromatin condensation state. It also affects motility of these cells and acrosome reaction. The interspecies analysis indicates significant structural and functional similarities, especially for the proteins with low molecular weight. Fertility associated proteins (FAPs) have been determined in the bull, stallion, boar, ram and dog. The contents of these proteins correlate with the indicators of the fertilizing abilities of sperm. In humans, several seminal plasma proteins were found which serve as diagnostic markers of spermatogenesis, seminiferous epithelium state, and azoospermia. To determine the semen ability for preservation, measurement of some seminal plasma protein content may also be used. Addition of specific plasma proteins to a spermatozoa solution undergoing the process of preservation may be used to retain the features of the cells responsible for efficient fertilization.

Tarnished plant bugs (Heteroptera: Miridae) were dissected and photographed to show the morphological characteristics of mated and unmated individuals. The presence of what appears to be a spermatophore, and/or the enlargement of the genital pouch and the seminal depository through infusion of fluids from the male was evidence of females having mated. Loss of relative size and white material from the accessory glands of males held from females for 7 d prior to mating was evidence of males having mated. Decreased size of seminal vesicles resulting from insemination was not seen to be an indicator of males having mated. The photographs provide a simple method for determining mating, and when used in conjunction with drawings of tarnished plant bug morphology provided by early authors, may facilitate the understanding of other aspects of tarnished plant bug reproductive morphology.

This review provides an updated overview of the seminal plasma composition, and the role of metabolic and protein components on the sperm function of avian species. In addition, the implication of seminal plasma on assisted reproductive techniques of birds was discussed. The semen of birds usually has exceptionally high sperm concentration with relatively little seminal plasma, but this contributes to very fast changes in sperm metabolism and function. The biochemical characteristics and physiological roles of the various seminal plasma components in birds (carbohydrates, lipids, amino acids, hormones, and proteins) are poorly understood. Seminal plasma content of proteins has an action on most cellular functions: metabolism, immunity, oxido-reduction regulation, proteolysis, apoptosis, ion homeostasis, and antimicrobial defenses. The variable amount of many proteins is related to a different fertility capacity of poultry sperm. The role of seminal plasma on semen conservation (chilling and freezing) remains largely a matter of speculation, as both inhibitory and stimulating effects have been found. Whereas the presence of seminal plasma did not seem to affect the sperm survival after freezing–thawing, DNA fragmentation is lower in the absence of seminal plasma. The molecular basis of the influence of seminal plasma on sperm cryo-resistance was also discussed in the present review.

Aim: The research was aimed on analysis of the remote consequences of Chornobyl accident on the reproductive function of men adult residing in Ukraine. Materials and Methods: 232 male volunteers with mean age of 34 years (range 20–47) from 5 different regions of Ukraine (Zhytomyr, Ivano-Frankivsk, Kyiv, Poltava, and Kyiv city) were enrolled in cross-sectional studies of long term radiation effects on seminal plasma and sperm. All manipulations, analysis and classification of ejaculates were done accordingly to WHO recommendations. The content of neutral α-glucosidase, fructose, citric acid in the seminal plasma was determined by spectrophotometry, L-carnitine — by high performance liquid chromatography, zinc — by atomic absorption spectroscopy. Results: In the men residing in the regions heavily contaminated with radioonuclides, the decreased sperm quality with concurrent asthenozoospermia, oligozoospermia, teratozoospermia, asthenoteratozoospermia, oligoasthenoteratozoospermia and oligoteratozoospermia was revealed. Moreover, the concomitant shifts in seminal plasma content of neutral α-glucosidase, fructose, citric acid, L-carnitine and zinc were detected. Conclusion: The study has revealed the ample sperm and semen abnormalities amongst the inhabitants of radiation polluted territories that should have to be a subject of careful research in forthcoming years. Seminal fluid often makes up a large part of an ejaculate, yet most empirical and theoretical studies on sperm competition have focused on how sperm characteristics (number and quality) affect fertilization success. However, seminal fluid influences own sperm performance and may potentially influence the outcome of sperm competition, by also affecting that of rivals. As a consequence males may be expected to allocate their investment in both sperm and seminal fluid in relation to the potential level of competition. Grass goby ( Zosterisessor ophiocephalus ) is an external fertilizer with guard-sneaker mating tactics, where sperm competition risk varies according to the tactic adopted. Here, we experimentally manipulated grass goby ejaculates by separately combining sperm and seminal fluid from territorial and sneaker males. While sperm of sneaker and territorial males did not differ in their performance when they interacted with their own seminal fluid only, sperm of sneakers increased their velocity and fertilization rate in the presence of territorial males' seminal fluid. By contrast, sneaker males' seminal fluid had a detrimental effect on the performance of territorial males' sperm. Sperm velocity was unaffected by the seminal fluid of males employing the same tactic, suggesting that seminal fluid's effect on rival-tactic sperm is not based on a self/non-self recognition mechanism. Our findings show that cross interactions of sperm and seminal fluid may influence the fertilization success of competing ejaculates with males investing in both sperm and seminal fluid in response to sperm competition risk.

Whole ejaculates were dialyzed against Tris-citrate-fructose extender for a 5 h period at room temperature (using semi-permeable cellulose tubing of 12-14 kDa molecular weight cut-off). It has been demonstrated that the long-term dialysis of dog semen causes a significant decrease in sperm quality parameters and disrupts the biochemical properties of seminal plasma. This procedure requires further improvement.

Since its identification in seminal fluid in 1971, much new information has been obtained about the biology and expression of prostate-specific antigen (PSA). PSA is a glycoprotein composed of 93% amino acids and 7% carbohydrates, with a molecular weight of about 30,000 Da. Functionally and structurally PSA is a kallikrein-like serine protease, and its physiologic role is degradation of the major proteins of seminal coagulum (semenogelin I and II, fibronectin), which leads to semen liquefaction. The PSA gene is located on the 13q region of chromosome 19, and it has a high degree of homology (more than 80%) with genes of the human glandular kallikrein (hKGK1). PSA production and expression are preferentially but not exclusively associated to the normal, benign hyperplastic and cancerous tissues of the prostate. In fact, it has been demonstrated that PSA is also present in accessory male sex glands and breast cancer. It was recently reported that PSA was also present in milk of lactating women. Many factors may influence PSA synthesis and production, and among them the most important are androgen, retinoic acid and growth factor stimulation. Significant advances have been recently made as regards the molecular isoforms of PSA. In the seminal fluid PSA seems partially bound to a serpine (protein C inhibitor), whereas in serum it is predominantly associated to α-1-antichymotrypsin and in a small quantity to α-2-macroglobulin. These new findings will have implications for the clinical application of PSA as a tumor marker for prostate cancer.

The study was carried out with the objective to evaluate the influence of season (summer and winter) on seminal characteristics of alpacas. Forty semen samples, 20 for each season, were collected from 4 adult alpaca by artificial vagina technique and evaluated immediately after collection to determine the following semen characteristics: (a) volume, (b) pH, (c) viscosity, (d) color, (e) concentration, (f) motility, (g) viability, (h) abnormalities, and (i) HOST test. Statistical analyses of data were made with least square ANOVA. The males were fed the same diet during both seasons and collection time was standardized in 10 min. The results obtained present differences in viscosity (5.9 ± 0.4 v. 3.7 ± 0.7 mm), motility (58.8 ± 22.1 v. 77.3 ± 11.6%), and concentration (135.9 ± 88.7 × 106 v. 242.4 ± 140.9 × 106; P < 0,05) but not in volume (1.9 ± 0.8 v. 2.1 ± 0.8 mL), pH (7.01 ± 0.3 v. 7.1 ± 0.3), viability (52.9 ± 18.4 v. 64.5 ± 15.9%), HOST test (56.1 ± 15.8 v. 49.3 ± 13.2%), and abnormalities (82.5 ± 7.2 v. 88.1 ± 7.0%) for the summer and winter seasons, respectively. The results suggest differences for some characteristics and possible differences in the thermoregulation scrotal of the alpacas with respect to others species.

The study has revealed the presence of fibronectin (FN) fragments and a lack of intact FN in 72 seminal plasma samples. The FN fragmentation was examined by immunoblotting with a monoclonal antibody specific to the central cellular FN domain and was confirmed with a monoclonal antibody directed to the C-terminal domain of FN. Nine FN fragments between 60 and 200 kDa and five fragments of 60-150 kDa were identified in seminal plasma samples of normozoospermic and of terato-, oligoterato-, and oligoasthenoterato-spermic groups, respectively. The relative amounts of the 60, 90 and 100 kDa FN fragments were 2-3 times higher in seminal plasmas with abnormal semen characteristics than in the normozoospermic group. The results suggest that seminal plasma FN fragments may contribute to fertilization and the analysis of FN fragmentation may have a diagnostic value in andrological investigations.

Effect of seed-priming with potassium (K) sources (K-nano-chelate, KNC, and sulfate (0, 2 and 4 %)) under drought stress (DS) conditions (0, -0.3, -0.6, -0.9, -1.2 and -1.5 MPa water potential) on the corn seedling traits was studied. Drought stress decreased the germination indices and seedling vigor. The highest germination, seminal root fresh and dry mass (RFM and RDM) was obtained in KNC primed seeds at -0.3 MPa DS. Mean germination time increased under DS conditions mainly in non-primed seeds. Increasing DS to -1.2 MPa led to decrease in RFM and RDM. Influence of DS on the fresh mass of shoots was more severe than on seminal roots. The highest shoots and seminal roots length was observed in 4 % KNC without any DS. Proper priming can be suggested to increase the plant tolerance under DS.

Seminal plasma purportedly plays a critical role in reproduction, but epididymal spermatozoa are capable of fertilisation following deposition in the uterus, calling into question the biological requirement of this substance. Through a combination of direct observation of spermatozoa in utero using probe-based Confocal Laser Endomicroscopy, in vivo assessment of sperm fertility and in vitro analysis of various sperm functional parameters, this study investigated the role of seminal plasma in spermatozoa transit through the cervix of the ewe. Following deposition in the cervical os, epididymal spermatozoa previously exposed to seminal plasma displayed an enhanced ability to traverse the cervix as evidenced by both significantly higher pregnancy rates and numbers of spermatozoa observed at the utero-tubal junction when compared with epididymal spermatozoa not previously exposed to seminal plasma. The beneficial effect of seminal plasma on sperm transport was clearly localised to transit through the cervix as pregnancy rates of spermatozoa deposited directly into the uterus were unaffected by exposure to seminal plasma. This phenomenon was not explained by changes to sperm motion characteristics, as seminal plasma had no effect on the motility, kinematic parameters or mitochondrial membrane potential of spermatozoa. Rather, in vitro testing revealed that seminal plasma improved the ability of epididymal spermatozoa to penetrate cervical mucus recovered from ewes in oestrus. These results demonstrate that the survival and transport of ram spermatozoa through the cervix of the ewe is not linked to their motility or velocity but rather the presence of some cervical penetration trait conferred by exposure to seminal plasma.

The reproductive capacity of the parasitoïd of cowpea bruchids Anisopteromalus calandrae Howard (Hymenoptera: Pteromalidae) male depends on the number of spermatozoa produced and spilled in the seminal vesicle. The objective of this study is to determine the dimensional characteristics of the different parts of the male genital tract of A. calandrae depending on age. Photographs of the parts of the male genital tract of the nymph and the virgin adults of 0- 1- 5- 10- and 15-day old were taken by light microscopy. The dimensional characteristics of the organs of the genital tract of A. calandrae has shown that when the surface of the testes decreases, that of the anterior portion of the seminal vesicle increases and the ratio between the width of the light and the total width of the accessory gland increases from emergence to the fifteenth day. The dimension of the testes decreases in favor of that of the seminal vesicle and the accessory gland of A. calandrae male depending on age.

Mutual relationships between selected chemical elements (Na, K, Fe, Cu, Mg, and Zn), basic motility characteristics (motility and progressive motility), and markers of the oxidative balance (superoxide dismutase, catalase, glutathione, albumin, and malondialdehyde) were investigated in bovine seminal plasma and spermatozoa. Computer assisted sperm analysis was used to assess the motility parameters; mineral concentrations were determined by the voltammetric method and flame absorption spectrophotometry; antioxidants and malondialdehyde were evaluated by UV/VIS spectrophotometry. Concentrations of chemical elements in both seminal fractions were in the following descending order: Na > K > Zn > Mg > Fe > Cu. Higher amounts of all minerals and nonenzymatic antioxidants were detected in the seminal plasma (P<0.01; P<0.001), while higher MDA concentration and activity of enzymatic antioxidants were recorded in the cell lysates (P<0.01; P<0.001). Na, Fe, Cu, Mg, and Zn were positively correlated with the motility and antioxidant parameters (P<0.05; P<0.01; P<0.001). Inversely, K exhibited the positive associations with malondialdehyde (P<0.05). This study demonstrates that most chemical elements are integral components of bovine semen and are needed for the protection against oxidative stress development.
