Abstract Mixed populations of cardiosphere-derived stem and progenitor cells containing proliferative and cardiomyogenically committed cells were obtained from adult rat hearts. The cells were cultured in either static 2D monolayers or dynamic 3D scaffold systems with fluid flow. Cardiomyocyte lineage commitment in terms of GATA4 and Nkx2.5 expression was significantly enhanced in the dynamic 3D cultures compared with static 2D conditions. The transformation of pentachlorophenol (PCP) by Phanerochaete chrysosporium I-1512 in relation to pentachloroanisole (PCA) was studied in two different modes of culture. The degradation of PCP by immobilized cells in agitated medium was compared with that by static cultures. The development of in vitro models is critical for furthering understanding of the intervertebral disc and the development of disc regeneration/tissue engineering. An in vitro culture system targeted towards mechano-biology studies of the intervertebral disc (IVD) was built and validated using bovine coccygeal discs. Discs were maintained in culture for up to one week with and without vertebral endplates. Water content and glycosaminoglycan content were found to be stable and cells were metabolically active when cultured under a 5kg static load.
