This study was aimed at checking out a possible connection between detrusor primary lesions and clinical and urodynamic data in patients with cervico-prostatic obstruction. 40 patients were evaluated, of whom 32 had symptomatic benign prostatic hyperplasia and 8 were controls. A biopsy of the right side wall of the bladder was carried out. The lesions found (muscular hypertropy, fibroelastosis, alteration of the nerve fibres) in relation to their gravity, correlated to the presumable date when symptoms started and to three data from urodynamic evaluation (significant residual p.m., degree of obstruction according to Schafer's chart, presence of detrusor-motor instability). The interference of physiological ageing on results is not underestimated, but this correlation exists. However, the lack of fine quantitative evaluations of follow-up on the possible reversibility of the same lesions after removal of the obstruction, make it impossible at this stage to state how close the correlation is.

A steady increase in the number of young sportsmen predetermines the relevance of studying the nature and severity of cardiovascular system remodeling in young athletes, including the development of arterial hypertension (AH) and left ventricular myocardial hypertrophy (LVHM).Objective. To study the prevalence and nature of arterial hypertension (AH) and its associations with signs of cardiovascular remodeling in young athletes taking into account pathogenetic relationship and genetic determination.Characteristics of children and methods. The study included 80 young athletes (including 42 boys, average age 14,5±1,54 years), divided into 4 groups depending on the kind of sports (1 – cyclic, 2 – game, 3 – complex coordination, 4 – power). We used the following research methods: “office” (i.e. during visit to the doctor) measurement of blood pressure, electrocardiography, echocardiography, color Doppler scanning of the neck vessels, daily monitoring of blood pressure, bicycle ergometry, genetic testing using the original panel with 448 markers.Results. An increase in blood pressure at rest was detected in 2,5% of children engaged in power sports, an insufficient decrease at night – in 16,3% of children, and an excessive reaction of blood pressure to dosed physical activity – in 25% of children. These changes correlated with signs of sports remodeling, in particular with the formation of left ventricular myocardial hypertrophy, which was diagnosed in 7–12,5% of athletes. According to the results of genetic testing, pronounced polymorphism in the NO synthase gene at three loci (rs10918594, rs12143842, rs16847548) was associated with a reorganization of the cardiovascular system in 45% of athletes.Conclusion. Violation of the circadian profile of blood pressure and excessive reaction to physical activity demonstrate 7,5–25% of young athletes, especially those involved in complex coordination and power sports. These changes were associated with the severity of sports remodeling and determined by a genetic predisposition.

Aim. The aim of this report is to identify variables determining the effectiveness of small football games at subsequent stages of training. Material and Methods. The study material comprised the results of tests conducted among 42 football players who were assigned to 3 categories: youngsters, younger juniors and seniors. The scope of the study included basic somatic features, selected motor skills tests and specific coordination tests of motor skills recommended for football players. Detailed correlation matrices were calculated using Spearman’s signed-rank test between all the analysed variables and the overall game perforance index (OPI). For each training group, morpho-functional profiles were calculated taking variables from previous analyses into account. Variables forming the structure of these models were normalised on the T scale. Results. 11 statistically significant Spearman’s signed-rank correlation coefficients were obtained between game performance indices and the analysed variables. In the 3 training groups of football players, considerable diversity of the internal structure of morpho-functional models was noted. In the group of juniors and seniors, differentiation between features was 1.2 of standard deviation. Conclusions. In terms of fitness, the influence of circulatory-respiratory endurance and speed ability on the indicator of overall performance for the seniors’ game was noted. In turn, taking the neuro-functional side of the players’ motor ability into account, correlations with the effectiveness of the game were mainly found within the ability of balance, kinesthetic differentiation and motor adaptation. Such regularities mainly refer to the youngsters and younger juniors.

(1) Objective: To use optical coherence tomography angiography (OCTA) and microperimetry (MP) to evaluate the correlation between retinal structure and function in patients with idiopathic, full-thickness macular holes (FTMHs) (2) Methods: This prospective, observational study included 11 eyes of 10 patients with FTMHs evaluated before surgery using OCTA and MP. MP sensitivity maps were superimposed and registered on slabs corresponding to superficial capillary plexus (SCP) and deep capillary plexus (DCP) on OCTA, and on the outer plexiform layer (OPL) and the Henle fiber layer (HFL) complex in en face OCT. On these maps, mean retinal sensitivity was calculated at 2° and 4°, all centered on the FTMH. Cystic cavity extension was assessed on the slab corresponding to the OPL + HFL complex in en face OCT and DCP in OCTA using the Image J software (Version 1.49v; National Institutes of Health, Bethesda, MD, USA); (3) Results: Absolute scotomas were observed corresponding to the FTMH. Additionally, rings of relative scotoma in the perilesional area were detected and correlated to the cystic spaces on en face OCT and OCTA. There was a significant correlation between reduced retinal sensitivity at 2° and 4° diameters around the FTMH and the extension of cystic areas (p < 0.01). There was a significant correlation between the extension of cystic cavities and BCVA (p < 0.01). (4) Conclusions: Morpho-functional analysis of FTMH using OCTA and MP, and the correlation between vascular abnormalities and impaired retinal sensitivity, may provide new, useful information. This integrated evaluation of FTMH may be useful to determine the function–structure correlation before and after vitreoretinal surgery, in order to gain a better understanding of the functional consequences induced by the morphological alterations, assessing outcomes in a more objective way, and potentially adding new surgical prognostic factors.

A study of cross-impact degree and morpho-functional correlation characteristics of primary open angle glaucoma and age-related macular degeneration was performed at 25 clinical sites in Russia and some CIS countries from January through May 2013. The study final protocol included 87 patients (108 eyes) divided into two groups, mean age of patients was 66.6 ± 5.65 years (min 55; max 81). Intragroup and intergroup variance analysis did not establish any significant difference in the macular area between the «glaucoma» group and the «glaucoma + AMD» group depending on the disease stage. The findings suggest that macular degeneration develops subsequent to a preexisting glaucomatous state.

The aim of this study was to identify new disease-related circulating miRNAs with high diagnostic accuracy for breast cancer (BC) and to correlate their deregulation with the morpho-functional characteristics of the tumour, as assessed in vivo by positron emission tomography/magnetic resonance (PET/MR) imaging. A total of 77 untreated female BC patients underwent same-day PET/MR and blood collection, and 78 healthy donors were recruited as negative controls. The expression profile of 84 human miRNAs was screened by using miRNA PCR arrays and validated by real-time PCR. The validated miRNAs were correlated with the quantitative imaging parameters extracted from the primary BC samples. Circulating miR-125b-5p and miR-143-3p were upregulated in BC plasma and able to discriminate BC patients from healthy subjects (miR-125-5p area under the receiver operating characteristic ROC curve (AUC) = 0.85 and miR-143-3p AUC = 0.80). Circulating CA15-3, a soluble form of the transmembrane glycoprotein Mucin 1 (MUC-1) that is upregulated in epithelial cancer cells of different origins, was combined with miR-125b-5p and improved the diagnostic accuracy from 70% (CA15-3 alone) to 89% (CA15-3 plus miR-125b-5p). MiR-143-3p showed a strong and significant correlation with the stage of the disease, apparent diffusion coefficient (ADCmean), reverse efflux volume transfer constant (Kepmean) and maximum standardized uptake value (SUVmax), and it might represent a biomarker of tumour aggressiveness. Similarly, miR-125b-5p was correlated with stage and grade 2 but inversely correlated with the forward volume transfer constant (Ktransmean) and proliferation index (Ki67), suggesting a potential role as a biomarker of a relatively more favourable prognosis. In situ hybridization (ISH) experiments revealed that miR-143-3p was expressed in endothelial tumour cells, miR-125-5p in cancer-associated fibroblasts, and neither in epithelial tumour cells. Our results suggested that miR-125-5p and miR-143-3p are potential biomarkers for the risk stratification of BC, and Kaplan-Maier plots confirmed this hypothesis. In addition, the combined use of miR-125-b-5p and CA15-3 enhanced the diagnostic accuracy up to 89%. This is the first study that correlates circulating miRNAs with in vivo quantified tumour biology through PET/MR biomarkers. This integration elucidates the link between the plasmatic increase in these two potential circulating biomarkers and the biology of untreated BC. In conclusion, while miR-143-3b and miR-125b-5p provide valuable information for prognosis, a combination of miR-125b-5p with the tumour marker CA15-3 improves sensitivity for BC detection, which warrants consideration by further validation studies.

Correlation of structural and functional data might lead to better identification of hormone-dependent tumors. Sixty breast cancer specimens, sent to the biochemistry laboratory for estrogen receptor (ER) analysis, were studied here by a combined morpho-functional approach. Histological examination of needle biopsies on frozen tissue blocks showed that 12 cases (10%) were free of tumor cells; these cases mostly proved ER negative. On the other 48 cases, an immunocytochemical reaction was performed on the biopsy sections with a monoclonal antibody directed against p 29, an estrogen receptor related antigen. The staining values for p 29 and the biochemical ER findings were significantly correlated. A combined histological, immunocytochemical study seems to offer advantages in the selection of patients for hormonal therapy.

AbstractFrom ancient times, the principal medical cultures tried to classify individuals according to their morpho-functional characteristics in order to study and understand their tendencies in terms of health and sickness. In the last two centuries, in Europe and America, a number of academic schools emerged whose theoretical concepts and ideas were important theroretically but had relatively little practical application. By contrast, in France and Italy, there was the joining of biotypological schools and homoeopathy, which permitted the correlation between biotypes and their relevant homoeopathic remedies. This article outlines the evolution of biotypological thinking and the results of the above said merger.

Material and methods. 55 patients with VLC class А, B, C according to Сhild-Pugh were examined. The average age of patients 42 years, disease duration – 3,8 years. Depending on the presence of ascites, patients were divided into 2 groups: 28 patients had no ascites (group 1), 27 patients had ascites (group 2), and a control group of 22 healthy people. Echocardiography was performed by the "VIVID E 95" device. All patients were evaluated diastolic function of the LV in accordance with the АSЕ/ЕAЕ-2009 and ASE/EACVI-2016 recommendations. Holter monitoring of ECG was performed. Statistical data processing was carried out using Statistica 6.0 statistical software package and Mann-Whitney nonparametric test. The correlation analysis was performed using the Spearman coefficient.Results. In all patients there was an increase in the index of myocardial mass and relative wall thickness of LV in comparison with the control group, more severe disorders were identified in patients with VLC with ascites, p<0.001. In the study of LV remodeling, concentric hypertrophy was most common in patients with VLC – in 11 (43%) patients of the 1st group and in 19 (70%) patients of the 2nd group. Holter monitoring of ECG revealed supraventricular extrasystoles in 9 (32%) patients of the 1st group and 11 (40%) – 2nd group; paroxysmal form of atrial fibrillation – in 2 (7%) patients and in 5 (18%), respectively. It was found that the maximum index of the left atrial volume was higher in patients with VLC compared to control, and in patients with ascites this disorder was more pronounced (p<0.001). In accordance with the ASE/EACVI-2016 algorithm, normal diastolic LV function was registered almost twice as often as when using the ASE/EAE-2009 algorithm. Undefined result was recorded in 8% of cases due to tricuspid regurgitation.Conclusion. Thus, patients with VLC develop an increase in the LV myocardial mass with the formation of concentric hypertrophy, remodeling of the left atrium with the occurrence of cardiac arrhythmias, diastolic LV dysfunction, and these changes are more pronounced in patients with ascites. In accordance with the new algorithm ASE/EACVI-2016 for detection of diastolic LV dysfunction, normal LV diastolic function was almost 2 times more common in patients with VLC than in accordance with the old ASE/EAE-2009 algorithm.

The goal of the study was to assess the correlation between height and body mass deviations measured with the aid of Z-index in teens and cell component of their peripheral blood.
Materials and methods. Apical height was measured in 83 adolescent boys in the course of regular prophylactic medical examination. Z-index was calculated according to WHO Growth Reference, 2007. Cell blood composition was studied with the help of automatic hematologic analyzer. The empiric data was processed statistically using Spearmans correlation coefficient.
Results. The analysis of hematologic parameters correlation with deviations of height and bodymass by Z-index has yielded relationship as follows: moderate positive correlation of Hb concentration with bodymass index Z-index, hematocrite with bodymass and height Z-index. Besides there is a mild positive correlation of Hb concentration with bodymass Z-index; RBC number with bodymass and height Z-index, MCV with bodymass and height Z-index. There is no correlation of MCH, MCHC, WBC and Plt number with bodymass and height Z-index.
Conclusion. The study has demonstrated there is a correlation between RBC number, Hb concentration and height deviations in case of normochromicity. The relationship ascertained may be explained by an assumption that peculiarities of erythropoiezis correlate with body size. Lower RBC number and Hb concentration in children with smaller bodymass index may be due to general plastic deficiency in their organism. The results obtained in the present study may be used to reveal and analyze physical development and the organism morpho-functional peculiarities correlation markers.

The aim: To assess the vitamin D level in blood plasma of patients with chronic heart failure and to identify the effect of its deficiency on the state of peripheral blood erythrocytes during physical exertion. Materials and methods: A total of 25 patients with CHF grade IIA stages II and III functional class were examined. The control group consisted of 25 relatively healthy people. All patients were offered to complete a 6 minutes walking test. The level of 25 (OH) D total in plasma was determined by enzyme immunoanalysis. Morphological studies of erythrocytes were performed on the light-optical (Leica CME) and electron-microscopic level («JEOL-25M-T220»). Results: Patients with chronic heart failure experienced 22.9% decrease in their vitamin D level (17.2±0.04 ng/ml) compared to the control group (38.4±0.05 ng/ml). Correlation analysis showed a direct proportional relationship between vitamin D deficiency and the number of erythrocytes of a modified form (r = 0.58; p <0.05) and erythrocytes with low osmotic resistance (r = 0.87; p <0.05). During the timed physical evaluation patients who experienced chronic heart failure accompanied by vitamin D deficiency developed an increase in the number of their reversibly and irreversibly deformed erythrocytes and a decrease in the cellular osmotic tability. Conclusions: During physical exertion, patients who experienced chronic heart falure accompanied by with vitamin D deficiency experienced morpho-biochemical changes in their red blood cells. These changes indicated structural disturbances in the membranes of their erythrocytes and could potentially have negative consequences for the somatic health of these patients.

Reproduction in vertebrates is controlled by the hypothalamo-pituitary-gonadal axis, and both the sex steroid and pituitary hormones play a pivotal role in the regulation of the physiology of the oviduct and events occurring within the oviduct. Their hormonal actions are mediated through interaction with specific receptors. Our aim was to locate α and β estrogen receptors, progesterone receptors, gonadotropin and prolactin receptors in the tissues of the oviduct of Typhlonectes compressicauda (Amphibia, Gymnophiona), in order to study the correlation between the morphological changes of the genital tract and the ovarian cycle. Immunohistochemical methods were used. We observed that sex steroids and pituitary hormones were involved in the morpho-functional regulation of oviduct, and that their cellular detection was dependent on the period of the reproductive cycle.

81 patients with active pulmonary tuberculosis were investigated. The morpho-functional status of vascular endothelium was evaluated by plasma levels of stable metabolites of nitric oxide, endothelin-1 and von Willebrand factor antigen. Typical increase of endothelin-1 in positive correlation with systemic inflammatory response syndrome (SIRS) expression was established. Nitric oxide level decreased in patients with chronic and severe course of disease. Decrease of nitric oxide level was not associated with SIRS but was consequence of specific intoxication. von Willebrand factor antigen decreased in patients with recent and limited spread of tissue damage but increased progressively with intensity of SIRS. This complex of changes (contrast shifts of nitric oxide and endothelin-1 and von Willebrand factor antigen increase) manifested in endothelium metabolic dysfunction syndrome and developed pre-conditions for microcirculation disturbances.

The evaluation of morpho-functional sperm characteristics alone is not enough to explain infertility or to predict the outcome of Assisted Reproductive Technologies (ART): more sensitive diagnostic tools are needed in clinical practice. The aim of the present study was to analyze Sperm DNA Fragmentation (SDF) and sperm-borne miR-34c-5p and miR-449b-5p levels in men of couples undergoing ART, in order to investigate any correlations with fertilization rate, embryo quality and development. Male partners (n = 106) were recruited. Semen analysis, SDF evaluation and molecular profiling analysis of miR-34c-5p and miR-449b-5p (in 38 subjects) were performed. Sperm DNA Fragmentation evaluation- a positive correlation between SDF post sperm selection and the percentage of low-quality embryos and a negative correlation with viable embryo were found. SDF > 2.9% increased the risk of obtaining a non-viable embryo by almost 4-fold. Sperm miRNAs profile—we found an association with both miRNAs and sperm concentration, while miR-449b-5p is positively associated with SDF. Moreover, the two miRNAs are positively correlated. Higher levels of miR-34c-5p compared to miR-449b-5p increases by 14-fold the probability of obtaining viable embryos. This study shows that SDF, sperm miR-34c-5p, and miR-449b-5p have a promising role as biomarkers of semen quality and ART outcome.

Objective: to study the functional disorder, joint structural changes with acute phase parameters, and interleukin-1 beta (IL-1β) in patients with hand osteoarthritis (HOA). Materials and methods: the study included 52 women with HOA, the mean age was 63.4 (10.0) years old. The degree of functional impairment was evaluated according to the visual analogue scale (VAS) and the author’s questionnaire. The laboratory study included an assessment of ESR, C-reactive protein (CRP), and IL-1β levels in the blood. The instrumental diagnostic was performed by X-ray, ultrasonography (US), and magnetic resonance imaging (MRI) of the hand joints. Results: no significant data were obtained on the dependence of the severity of structural and functional disorders from ESR, CRP, and IL-1β levels (r<0.5; rs<0.5). Conclusions: there was no correlation between HOA activity and CRP and IL-1β levels but some authors propose to use highly sensitive methods to detect CRP. The application of highly sensitive methods for CRP detection could reveal the association between this indicator and the HOA activity. The absence of dependence between IL-1β level and morpho-functional parameters agrees with the data obtained by other researchers. It is possible that the evaluation of the IL-1β level in dynamics can be useful for assessing the treatment response but this requires further studies.

Objective: To determine the presence and informative significance of the relationship between physical development, functional status and physical fitness of students 7-10 years. Material and methods: the research was conducted in rural schools of villages Kovalyn and Divychky of Pereyaslav-Khmelnitsky district of the Kiev region. 137 children participated in the research, among them 33 pupils 1 form, 29 of 2 form, 37 pupils 3 form, and 38 pupils of 4 form. It used a range of methods, theoretical analysis and synthesis of the literature data; pedagogical methods; testing; anthropometric; physiological (Rufie); methods of mathematical statistics. Results: The factor analysis of the main components of motor activity of children of 7-10 years shows that there is a correlation between physical abilities and morphofunctional state, which is manifested in the presence of the overwhelming majority of integrated factors. Some gender and age differences were identified. However, as far as the content of most factors is concerned, identical trends are observed. So, it has been found that anthropometric and functional indicators in all sex-age groups are located in the most influential factors - I and II combined with indicators of motor tests. Conclusions: the results of factor analysis allowed us to identify the physical qualities that are more related to the morpho-functional state of the body. According to the rating, both boys and girls are distributed as follows: І - speed, ІІ - coordination abilities, ІІІ - speed-power, ІV - endurance, V - strength, VI - flexibility. It gives an opportunity to determine the correlation of different exercises in different directions in physical education and extracurricular activities. Abstract We have lived in an era in sports history in which due to the intensity of the confrontations between athletes, disorders manifested by headaches, shoulder, pelvis and knee aches etc. appear more often. The performers during puberty require special attention on their morpho – functional development from the kynesiotherapist who is part of the multidisciplinary team, due to many transformations of the human body specific at this age category. 13 juniors female athletes have participated to the study, being assessed anthropometric, plantar pressures and posture with softwear FreeStep by Sensor Medica, biomechanical analysis and from technical & tactical point of view by 5 tests with DartFish 360S. Within the statistical – mathematical analysis, we have used Pearson correlation coefficient which favoured the identification of the strongest correlations resulted from the technical – tactical - anthropometric – plantar pressures and postural unbalances (r > 1.00 at test 1 and r > 0.84 at test 2). Assumption: It is assumed that by using established methods and means to assess postural deficiencies and to assess the level of consolidation of the topspin attack, we can identify the correlations between them which would facilitate the creation in the future of an efficient postural improvement program. The goal is to improve the quality of life and sports performance in junior female table tennis players. The conclusions of this study demonstrate the fact that the mobility of the spine in frontal plan, high thoracic elasticity and an increased lateral mobility on the opposite side of the active arm represent advantages intended to optimize the attack by topspin, while the supraponderability, the pain in lumber area and the shoulder’s asymmetry constitute disruptive factors of attack’s performance. The coordination of leaf hydraulic-related traits with leaf size is influenced by environmental conditions and especially by VPD. Water and gas flows are guided by leaf anatomical and physiological traits, whose plasticity is crucial for plants to face environmental changes. Only a few studies have analysed how variations in VPD levels influence stomatal and vein development and their correlation with leaf size, reporting contrasting results. Thus, we applied microscopy techniques to evaluate the effect of low and high VPDs on the development of stomata and veins, also analysing leaf functional traits. We hypothesized that leaves under high VPD with a modified balance between veins and stomata face higher transpiration. We also explored the variability of stomata and vein density across the leaf lamina. From the results, it was evident that under both VPDs, plants maintained a coordinated development of stomata and veins, with a higher density at low VPD. Moreover, more stomata but fewer veins developed in the parts of the lettuce head exposed to light, suggesting that their differentiation during leaf expansion is strictly dependent on the microclimatic conditions. Knowing the plasticity of hydraulic-related morpho-functional traits and its intra-leaf variability is timely for their impact on water and gas fluxes, thus helping to evaluate the impact of environmental-driven anatomical variations on productivity of natural ecosystems and crops, in a climate change scenario.

The biodiversity of assemblages that experience the introduction and extinction of species may lead to responses in two important facets: The taxonomic and functional diversity. The way in which these facets are associated may reveal important implications and consequences for the conservation of those assemblages. Considering the critical situation of freshwater fishes in continental Chile (30° – 56° S), we analyzed how the taxonomic (TDβ) and functional (FDβ) facets of β-diversity, and their components of turnover and nestedness, are associated. We evaluated changes in β-diversity (ΔTDβ and ΔFDβ), turnover (ΔTDtur and ΔFDtur), and nestedness (ΔTDnes and ΔFDnes) in 20 fish assemblages from their historical (pre-European) to current composition. We also simulated future trends of these changes, assuming that native species with conservation issues would become extinct. Our results show that the fish assemblages studied are in a process of loss of β-diversity, both in taxonomic and functional facets (ΔTDβ = -3.9%; ΔFDβ = -30.4%); also, that these facets are positively correlated in the assemblages studied (r = 0.617; P < 0.05). Both components showed by loss in nestedness (ΔTDnes = -36.9%; ΔFDnes = -60.9%) but gain in turnover (ΔTDtur = 9.2%; ΔFDtur = 12.3%). The functional β-diversity decreased more than the taxonomic (ΔFDβ > ΔTDβ), which was caused chiefly by six exotic species of Salmonidae, whose geographical spread was wider and that at the same time shared several morpho-functional traits. Our forecasts, assuming an intensification in the extinction of Endangered and Vulnerable native species, indicate that the process of homogenization will continue, though at a lower rate. Our study shows that the freshwater ichthyofauna of continental Chile is undergoing biotic homogenization, and that this process involves the facets of taxonomic and functional β-diversity, which are show high correlation between historical and current compositions. Both facets show that process is influenced by nestedness, and while turnover contributes to differentiation (both taxonomic and functional), its importance is overshadowed by nestedness.

Adiponectin (ADPN), a hormone produced by adipose tissue, facilitates gastric relaxation and can be a satiety signal in the network connecting peripheral organs and the central nervous system for feeding behavior control. Here, we performed preclinical research by morpho-functional analyses on murine gastric fundus smooth muscle to add insights into the molecular mechanisms underpinning ADPN action. Moreover, we conducted a clinical study to evaluate the potential use of ADPN as a biomarker for eating disorders (ED) based on the demonstrated gastric alterations and hormone level fluctuations that are often associated with ED. The clinical study recruited patients with ED and healthy controls who underwent blood draws for ADPN dosage and psychopathology evaluation tests. The findings of this basic research support the ADPN relaxant action, as indicated by the smooth muscle cell membrane pro-relaxant effects, with mild modifications of contractile apparatus and slight inhibitory effects on gap junctions. All of these actions engaged the ADPN/nitric oxide/guanylate cyclase pathway. The clinical data failed to unravel a correlation between ADPN levels and the considered ED, thus negating the potential use of ADPN as a valid biomarker for ED management for the moment. Nevertheless, this adipokine can modulate physiological eating behavior, and its effects deserve further investigation.

The permanent first molar is the first tooth in the permanent series that erupts into the archway and thus transforms the temporary dental arcade into mixed arcade, being the only stable element throughout the mixed dentition. The plurivalent importance of the permanent molar must be considered permanently in the harmonious development of the dento apparatus. This tooth has a determining role in dental arcade functionality, being the oldest witness of the permanent occlusion. The permanent first molar is a guardian of the occlusal memory and contributes to the agreement of the predetermined with the posterior one (the temporo-mandibular joint). The first permanent molar improves the relationships in the vertical plane, the early loss of it generates morphological and functional disorders which takes place cascaded across the arcade, and its extraction must be considered a personal failure of the dentist. The study consists of 80 patients divided into two groups; the group I consists of 45 children with chronological ages ranged between 5-6.6 years; the second group selected from dental care, a total of 35 children aged between 8.6-10 years. Dental caries morbidity has increased significantly over time and surprised by age the younger becoming a true social scourge. The current pediatric dentistry emphasizes new aspects of the normal and pathological growth of the dento-maxillary apparatus and underlines the special significance of the morpho-structural and functional integrity of the odontous units in close correlation with the subsequent development of the dento-maxillary apparatus in general.

Dissection of the genetic basis of the adaptive response of durum wheat to unfavourable water and temperature regimes is an important prerequisite for the selection of genotypes less vulnerable to environmental constraints. An elite durum population of 249 recombinant inbred lines was tested across 16 Mediterranean environments characterised by contrasting thermo-pluviometric conditions and, consequently, a broad range of productivity (from 0.56 to 5.88 t ha–1). Among the environmental variables investigated, soil moisture during grain filling showed the most consistent correlation with yield components and grain yield, whereas a weaker, albeit in some cases significant, association was noted with temperature at heading and thermal time during grain filling. Ear peduncle length appeared as a valid and easy-to-phenotype morphological proxy for the water available to the plant. In total, 76 quantitative trait loci (QTLs) were identified for yield components and for several morpho-physiological traits (peduncle length, the spectral reflectance index NDVI and leaf greenness at the milk-grain stage expressed in SPAD units) associated with the adaptive response of wheat to water and heat stresses. Although most of the QTLs were significant in only one or two environments, two major QTLs on chromosomes 2BL and 3BS showed consistent additive and epistatic effects on 1000-kernel weight, peduncle length, SPAD values and grain yield in half of the environments. In view of their strong phenotypic effects on kernel weight, these two QTLs are good candidates for positional cloning in order to gain a better understanding of the functional basis of their effect on the plasticity of grain weight and grain yield.

The mechanisms of hemorrhagic manifestations in patients with ANKRD26associated thrombocytopenia (ANKRD26-AT) are poorly understood. The aim of this work is to detect possible morpho-functional disorders of platelets in patients with mutations in the ANKRD26gene by flow cytometry with activation. The study was approved by the Independent Ethics Committee of the Dmitry Rogachev National Medical Research Center of Pediatric Hematology, Oncology and Immunology. 8 children aged from 1.5 to 15 years were examined. The platelet count ranged from 29 to 172 thousand/μl, with a median of 60 thousand/μl. The severity of hemorrhagic manifestations was assessed on a standardized scale (Pediatric Bleeding Questionnaire, PBQ) and it ranged from 0 to 5 points, with a median of 3.5 points. Platelet activation was performed with a CRP + TRAP mixture. Comparison was carried out with the results of examination of 26 apparently healthy children (control group, CG) aged 2 to 15 years. When compared with CG, patients showed an increase in platelet size (FSC; p= 0.018) and granularity (SSC; p< 0.001) after activation. In contrast to the CG, the correlation between FSC and SSC of platelets in patients was not significant (cor. = 0.55; p= 0.15). Patients showed a high, significant relationship between the number and FSC of platelets (cor. = –0.93; p< 0.001), as well as an increased density of CD42b (p < 0.001) and a decrease in the proportion of procoagulant platelets (p= 0.01) after activation. The revealed changes indicate violations of the mechanisms of activation and shape changes of platelets in patients with ANKRD26-AT.

Objective. To study the parameters of the morpho-functional state of the vascular wall in patients with rheumatoid arthritis (RA) with different levels of total cardiovascular risk (CVR). Materials and methods. 181 patients with active RA (mean age 40.4 [31.8; 49.6] years) were examined, and 2 groups were formed: 1) patients with the seronegative subtype (n=87); 2) those with the seropositive subtype (n=94). CVR was determined using the modified Systematic COronary Risk Evaluation (SCORE) scale; the intima-media complex thickness (IMCT) and the stiffness index (SI) of common carotid arteries (CCA) were assessed with specific methods. Results. Low CVR was determined in 86 (48%), moderate risk - in 47%, and high risk - in 5% patients with RA. 176 (97%) patients with RA had signs of subclinical atherosclerosis, with simultaneous IMCT increase above the 75th percentile and SI increase over 10 m/s in 21% patients. Mean IMCT values and SI in the 2nd group were higher than in the 1st group (р=0.018 and р=0.048, respectively). The lower risk was more commonly accompanied by the IMCT increase; as the CVR increased, the proportion of RA patients with SI over 10 m/s increased (р<0.01). A correlation was detected between the age and SI (r=0.58, p=0.035), IMCT parameters (r=0.64, p=0.002). SI correlated with the body mass index (BMI) (r=0.64, p=0.04), and IMTC correlated with the С-reactive protein level (r=0.61, p<0.01). Direct correlation was detected between SI, IMTC, and the Disease Activity Score 28 (DAS28) (r=0.51, p=0.02; r=0.45, p=0.008), RA duration (r=0.42, p=0.04; r=0.58, p=0.04), rheumatoid factor (RF) (r=0.68, p=0.04; r=0.49, р=0.04) and anti-citrullinated peptide antibody (ACPA) levels (r=0.62, p=0.018; r=0.45, р=0.003), respectively. Conclusion. It is feasible to perform an advanced examination for the diagnosis of subclinical atherosclerosis along with the routine evaluation of total CVR using the SCORE scale in patients with active RA.

Purpose. To study the functional status in the body of the common carp Cyprinus carpio (Linnaeus, 1758) after the addition of biologically active substances in feed composition as an adaptogene and corrector of metabolic processes including the antioxidant system, catalase activity in carp body. Methodology. The experimental part of the study was performed at the laboratory of the Scientific and Research Laboratory “Aquaculture Perspectives”, Scientific and Research Laboratory of “S. Pentelyuk Physiological and Biochemical Studies”, Scientific and Research Laboratory of Ecological and Chemical Analysis and Water Monitoring of the State Education Institution “Kherson State Agrarian and Economical University” and at the laboratory of the “Kherson Production and Experimental Hatchery of Ordinary Fish Breeding”. The duration of the experimental part was 30 days. During this period, visual observation of the fish was carried out, biological material was collected for the study in accordance with the current methods in fish farming. Cytological studies of blood were carried out on smears using Micromed MDC-500-5MP eyepiece camera. ThermoMix equipment was used to transport biological material to the laboratory. The hydrochemical parameters in tanks of the recirculating aquaculture system (RAS) were investigated by express methods (tests) with the corresponding recording of results in the work journal. The reaction and ethology of the common carp Cyprinus carpio to the light was carried out using digital equipment and a LDT test. Findings. The obtained results demonstrated the positive effect of the feed factor as an adaptogene and growth stimulator on the redistribution of metabolic energy in the carp body, which in turn had a stimulating effect on the development rate and improved the general functional status of the fish body. The paper presents the results of morpho-functional, biochemical parameters of carp blood in ontogenesis when grown in recirculating aquaculture systems against the background of the application of elements to improve technological aspects while improving the adaptive capabilities of the fish body. It was found that the feed factor, when introduced into the general economic ration, promoted an increase in the average carp body weight in the experimental group, improved the parameters of homeostatic equilibrium in comparison with fish in the control group. Positive results of a scientific and experimental study on the use of the additive as an adaptogene were obtained; it was studied with the aim of activating protective mechanisms, metabolic processes of aquatic organisms under constant or systematic exposure to stress factors of a technological nature. The study showed that the body weight, yield, total number of erythrocytes, hemoglobin content, total protein, and the level of catalase activity in the experimental groups of fish exceeded these parameters in the control group. The results obtained allowed noting the activation of the antioxidant system, the restructuring of physiological and biochemical processes, mobilization systems in the carp body in the experimental group. Originality. We found a correcting effect of the feed factor on metabolic processes, the adaptive capacity of the fish organism, as well as an increase in the resistance of the body as a whole. After implementing the technological scheme of cultivation of a feed factor of natural origin as an adaptogene against the background of the influence of technological stress factors, we found a correlation between the morpho-functional, biochemical parameters of carp blood and stress markers. Addition of biologically active substances of natural origin with a pronounced adaptive and correcting physiological and biochemical parameters into the general diet of carp improved the adaptive capacity. The antioxidant system was activated against the background of the action of technological factors. Practical value. The results of scientific and experimental studies of the functional status of carp body under the influence of technological factors were presented. Parameters of the development of carp in ontogenesis when growing in recirculating aquaculture systems against the background of the introduction of multilevel elements to improve technological aspects were studied. The results demonstrated the positive effect of biologically active substances invlufrf in the feed mixture, in particular, activation of the growth rate, increase in body weight, and yield. This work investigates the relationships between leaf traits and olive response to water stress through two complementary field experiments in order to screen genotypes for drought tolerance. The first experiment consisted of the phenotyping of 32 olive varieties for 11 leaf morpho-physiological traits during a fruit set phase under well-watered conditions. In the second experiment, the yield and vegetative responses of five representative varieties to the withholding of irrigation during the whole fruit growth period were assessed to identify leaf traits that are associated with olive resilience plasticity and that could be used as drought tolerance markers. The results highlighted large phenotypic variability for leaf area (LA, 2.9–9.5 cm2), petiole elasticity (8.0–36.0°), water loss in detached leaves (WLDL, 3.6–11.6%), stomatal density (222.6–470.1 no mm−2), stomatal length (11.4–18.7 µm), trichome density (120.5–204.4 no mm−2), trichome width (103.6–183.4 µm), leaf contents in cuticular wax (CWC, 44.7–606.2 µg cm−2), and soluble sugars (SSC, 15.8–536.9 mg gdw−1). Among these leaf traits, WLDL displayed a significant correlation with the yield stability index (r = −0.973) and water use efficiency (r = −0.939), suggesting its use as relevant drought tolerance phenotypic marker. Likewise, LA, SSC, and CWC were singled out as second-level drought tolerance markers, being strongly correlated with stability of leaf size and leafing intensity in response to water stress. Cluster analysis highlighted 12 distinct levels of drought tolerance within the studied olive collection. Based on the four identified phenotypic markers, “Lechin de Sevilla” alongside “Picholine Marocaine” were found to be the most drought-tolerant varieties, while Frantoio was the most sensitive. This study provides the first, unprecedented, insights into the usefulness of leaf phenotyping in olive drought tolerance screening, with a focus on structural and functional leaf traits.

Revealing peculiarities of the course of physiological and biochemical processes in the formation of parameters of ejaculate allows effective correlation of biological completeness of spermatozoids based on the use of chelate compounds of limiting microelements, particularly copper. The study was focused on determining changes in processes of spermatogenesis during correction of mineral nutrition. In the experiment, we used adult boars of the same age, live weight and quality of ejaculates. They consumed copper citrate during the 45 day experiment in doses higher than the norm by 10% (1.5 mg per 1 kg of combined feed) and 20% (3 mg per 1 kg of combined feed) compared with the control. We determined difference in the effects of different doses of copper citrate on the formation of parameters of ejaculates in sire boars. Addition of the mineral additive to the feed in the doses 10% exceeding the norm promoted increase in the parameters of functional activity of spermatozoids – mobility and survival – on day 45 of the intake, which occurred against the background of acceleration of peroxidation processes (increase in the concentration of thiobarbituric acid of active compounds) and activation of the system of antioxidant protection – increase in the activity of superoxide dismutase and decrease in the level of catalase in the sperm. At the same time, we observed increase in the morphometric parameters of spermatozoids – general length, width, length and volume of the head. On day 30, sire boars that had been consuming copper citrate in the amounts 20% above the norm were observed to have increase in concentration of spermatozoids, their mobility and survivability, though a decrease was seen in the morphometric parameters of the gametes. Further intake of this element caused increase in the amount of abnormal forms of spermatozoids. Under the action of this additive, we determined increase in the amount of metabolites of lipid peroxidation – conjugated dienes and thiobarbituric acids of active compounds, and also weakening of the system of antioxidative protection – decrease in the content of reduced glutathione and accumulation of dehydroascorbic acid. Thus, the intake of copper citrate in the amounts of 10% above the norm facilitated morpho-functional parameters of spermatozoids by activating the system of antioxidant protection. It would be promising to conduct further research to determine the effects of copper citrate on the processes of activation and capacitation of spermatozoids and fertilization of oocytes in in vivo and in vitro conditions.

Pregnant women with intrauterine growth restriction (IUGR) have an increased risk of adverse perinatal and long-term complications compared with the birth of children with normal body weight. Thus, IUGR is one of the main challenges for the global health system, especially in poor and developing countries. Morpho-functional studies of the placentas help in determining the causes of IUGR, and therefore, timely prevent complications in pregnant women with IUGR. The objective: The purpose of this study is to investigate various morphometric and pathomorphological changes in the placenta, including inflammatory, in cases of IUGR, and to establish a correlation of these results with the etiology and complications for the fetus. Materials and methods. In the current study, 54 placentas of the fetuses with IUGR (the main group) were compared with 50 placentas of the fetuses with normal development (control group). The criteria for the inclusion of IUGR were gestational age more than 30 weeks and all fetuses with a weight less than 10th percentile for this period of pregnancy. The placenta material was studied pathomorphologically with laboratory screening for infection and inflammation. Similarly, the results were determined for placentas of the fetuses with normal development compared to placentas with IUGR. Results. The placenta study showed the presence of calcification in the case of IUGR, as well as in the case of prolonged pregnancy. However, calcification of the placenta in the case of IUGR was more progressive compared with placenta in the normal pregnancy. In addition, the presence of intrauterine infection and inflammation was observed, which could also lead to an adverse outcome for the further progression of pregnancy with IUGR. Conclusion. A comparative macro- and microscopic pathomorphological study of the placentas in the two groups has shown a significant increase in the pathological changes in all the anatomical structures of the fetuses with IUGR. Wide-field cone bipolar cells with sparse dendritic branching and proposed connectivity to blue cones were first identified in rabbit and cat. In rabbit, these were subdivided into type a (wa) and type b (wb), with axonal branching in sublamina a, and sublamina b, respectively, of the inner plexiform layer (IPL). Recent studies in rabbit support the earlier hypothesis of exclusive blue/short wavelength cone connectivity for both types. The homologues of wb cells (but not wa cells) have been identified in other mammals. The axonal branching of wa cone bipolar cells is shown to co-stratify with the dendrites of the “fiducial,” type a starburst amacrine cell, although a few branches extend into sublamina b. The axon terminal of wb cone bipolar cells is shown to be narrowly stratified in stratum 5α, deep to the dendrites of the type b starburst amacrine cell. Rabbit ganglion cells postsynaptic to wa cells are unknown, but may include class III.2a cells, similarly stratified in the IPL. The wb axon terminal is shown here to co-stratify with and to make close, likely synaptic, contacts with the dendrites of a recently described morphological subtype of class II ganglion cell in rabbit retina, IIb2. Recent morpho-physiological correlation indicates that class IIb2 cells correspond to the blue-ON-center-X or ON-brisk-sustained ganglion cells, defined physiologically in rabbit. In contrast, the wb cell in cat retina must innervate the physiologically identified blue-ON-center-sluggish-sustained ganglion cell. In monkey retina, the wb-like bipolar cells apparently innervate a small, partly bi-stratified ganglion cell. Mammals share a common pathway from short-wavelength-sensitive (S/blue) cone photoreceptors to ON-center ganglion cells in sublamina b of the IPL, in the form of wb or wb-like cone bipolar cells, but the type of ganglion cell innervated appears to be particular, and may serve different functional roles in different mammalian orders.

Objective. To investigate the relationship between platelet (Pl) adhesive activity and total cardiovascular risk (CVR) and the effect of L-arginine (Tivortin®, “Yuria-Pharm”) on morpho-functional parameters of Pl activity in patients with arterial hypertension (AH) in combination with chronic obstructive pulmonary disease (COPD).
Materials and methods. The study included 44 patients with AH stage II in combination with COPD clinical categories A-D in the stable phase of diseases (group I) and 15 healthy individuals (group II). The calculation of the CVR by SCORE, the study of the degree of adhesion (Adh) Pl (optical turbidometry, analyzer AP 2110, Belarus). 20 patients (IA group) in addition to standard therapy of AH and COPD for 4 weeks received 4 g of L-arginine (Tivortin®, “Yuria-Pharm”), 10 patients (IB group) received only standard therapy. Before and at the end of therapy, the study was performed Pl Adh, in 10 patients of IA – transmission electron microscopy (TEM) Pl (electron microscope PEM-100-01 (SELMP, Ukraine). Statistical processing of the obtained results was performed using the program Statistica 6.1.
Results and discussion. In group I the average age was 56.0±6.7, men – 35 (79.5 %), body mass index (BMI) – 26.3 [24.0; 30.3] kg/m2, active smokers – 25 (57 %). In group II the average age was 52.7±4.9, men – 10 (66.7 %), BMI – 25.4 [24.3; 26.2] kg/m2, active smokers were not detected. The degree of Pl Adh in group I was higher than in group II (42.0 [36.0; 52.0] %) vs 29.0 [26.0; 34.0] %; p=0.005). In patients of group I the degree of Pl Adh had a direct correlation with the CVR by SCORE (R=0.50; p=0.001). Against the background of therapy in the IA group, the degree of Pl Adh decreased from 42.0 [34.0; 52.0] % to 30.0 [26.0; 39.0] % (p=0.004), in the IB group – from 38.5 [36.0; 47.0] % to 36.0 [33.5; 43.0] % (p=0.18). According to the results of TEM of Pl in patients of IA group, the number of activated forms of Pl decreased from 45.0 [43.0; 48.0] % to 37.0 [33.0; 41.0] % (p=0.028), aggregated Pl – from 11.5 [10.0; 9.5] % to 9.5 [8.0; 12.0] % (p=0.043), degranulated Pl – from 16.5 [16.0; 20.0] % to 11.0 [9.0; 13.0] % (p=0.028). No adverse effects were identified during L-arginine therapy.
Conclusions. Increased adhesive activity of Pl in stable patients with AH in combination with COPD is associated with the total CVR by SCORE. The use of L-arginine (Tivortin®, “Yuria-Pharm”) effectively and safely helps to reduce the functional activity of Pl in this category of patients.

El objetivo del estudio fue realizar una comparación entre dermatoglifia dactilar y los datos morfofuncionales en jugadores de fútbol profesional. Se contó con una población de 24 jugadores del fútbol profesional colombiano con edad promedio de 21 ± 1.99 años; talla 177.3 ± 4.3 cm; peso 73.21 ± 6.42 kg, integrantes de la selección masculina profesional del equipo de fútbol Fortaleza de la ciudad de Bogotá, Colombia. Se realizó una caracterización descriptiva de las variables de estudio (dermatoglifia, composición corporal, fuerza explosiva, fuerza máxima y consumo de oxígeno); posteriormente, se realizó el contraste de normalidad de los datos mediante prueba de Shapiro–Wilk y finalmente se aplicó un análisis bivariado. Los resultados evidenciaron la presencia de correlaciones fuertes entre la fuerza y la potencia máxima pico y el consumo máximo de oxígeno (p=.737; p=.699); así como entre la altura del salto como el potencial de energía (p=.952) y una correlación negativa entre la altura del salto y el porcentaje de masa grasa (p=-.554). Los marcadores dermatoglíficos, evidenciaron que la población de futbolistas evaluados tuvo comportamientos similares comparados con estudios internacionales y nacionales. Finalmente se identificaron relaciones importantes entre variables asociadas con fuerza, potencia, VO2máx, altura de salto y porcentaje de masa grasa como elementos determinantes del rendimiento óptimo del futbolista.
Abstract. The objective of the study was to make a comparison between dermatoglyphic dactilar and morpho functional data in professional football players. It had a population of 24 players of Colombian professional football with an average age of 21 ± 1.99 years; size 177.3 ± 4.3 cm; weight 73.21 ± 6.42 kg, members of the men's team of the Fortaleza football team of the city of Bogotá, Colombia. A descriptive characterization of the study variables (dermatoglyphic, body composition, explosive force, maximum force and oxygen consumption) was performed; posteriorly, the normality contrast of the data was performed by Shapiro–Wilk test and a bivariate analysis was finally applied. The results showed the presence of strong correlations between the force and the maximum peak power and the maximum oxygen consumption (p=.737; p=.699); as well as between the height of the jump as the energy potential (p=.952) and a negative correlation between the jump height and the percentage of fat mass (p= -.554). The dermatoglyphic markers showed that the population of footballers evaluated had similar behaviors compared to international and national studies. Finally, important relationships were identified between variables associated with strength, power, VO2max, jump height and percentage of fat mass as determining the optimal performance of the footballer.

The studbook of Pura Raza Menorquina horses only permits the use of black-coated animals with a small quantity of white marks as breeding stock. Its breeding program uses linear morpho-functional traits as selection criteria. Our aim was to estimate the genetic parameters of linear morpho-functional traits, and reveal relationship of quality of black coat color (QB) and percentage of white marks (WM) with the other morphological and functional linear traits in this breed. A total of 46 linear traits were scored by four appraisers using seven classes, with a total of 772 records from 333 animals (≥4 years old). Univariate animal models using a Bayesian approach were used, with a pedigree of 757 animals. Sex (two) and appraiser-season (13) were included as fixed effects, age as a linear covariate, and permanent environmental and additive genetic as random effect. The heritabilities of the morpho-functional traits were low to medium (0.09–0.58) and matched the range in the bibliography. Heritabilities for coat color traits were 0.36 for QB and 0.23 for WM. The highest genetic correlations were obtained between QB and upper neck line (0.816) and between WM and form of the hoof (0.638). The negative signs of most of the genetic correlations between WM and the functional traits is also remarkable, contributing to the selection of functional traits against the presence of white marks in this population. A clear genetic differentiation was observed between animals with better breeding values for QB and WM, corroborated by a study on founders. In conclusion, QB and WM could show different genetic backgrounds.

Friedreich’s ataxia (FRDA) is a rare autosomal recessive neurodegenerative disorder due to the homozygous pathological expansion of guanine-adenine-adenine (GAA) triplet repeats in the first intron of the FXN gene, which encodes for the mitochondrial protein frataxin. In the visual system, the typical manifestations are ocular motility abnormality, optic neuropathy, and retinopathy. Despite the evidence of ophthalmological impairment in FRDA patients, there is a lack of information about the morpho-functional condition of the retina and of the optic pathways in healthy heterozygous carriers of Friedreich’s ataxia (C-FRDA). Ten C-FRDA subjects (providing 20 eyes) and thirty-five Controls (providing 70 eyes) underwent a complete neurological and ophthalmological examination comprehensive of functional (full-field Electroretinogram (ffERG), multifocal Electroretinogram (mfERG), Visual Evoked Potential (VEP), and Pattern Reversal Electroretinogram (PERG)) and morphological assessments (Optical Coherence Tomography, OCT) of the retina, macula, retinal ganglion cells, and visual pathways. The groups’ data were compared using a two-sample t-test. Pearson’s test was used to investigate the morpho-functional correlations. Statistically significant differences (p < 0.01) between C-FRDA and Control eyes for the values of the following parameters were found: ffERG b-wave amplitude, mfERG Response Amplitude Densities, PERG P50 implicit time and P50-N95 amplitude, VEP P100 implicit time, Retinal Nerve Fiber Layer (RNFL) Overall, and Nasal thickness. The values of the OCT macular volume were not statistically different (p > 0.01) between the two Groups. Therefore, our data suggest that, in C-FRDA, a dysfunction of retinal elements without morphological macular impairment may occur. In addition, a morphological impairment of RNFL associated with an abnormal neural conduction along the visual pathways can be also detected.

This review focuses on spinal and central mechanisms of fatigue associated with the implementation of adaptation programs. The morpho-functional correlations of the moto-neurons activity of the spinal cord, the activities of the functional systems of the human body, neurotransmitter systems, serotonergic, dopaminergic and noradrenergic systems in the mechanisms of fatigue were considered. The phases of the process of adaptation to physical loads as well as cumulative training effect were identified. The authors have identified and described the participation of the autonomic nervous system, glucocorticoids and other hormones and mediators in long-term adaptation to physical loads, in the transition from carbohydrate to fat type of energy metabolism. The stages of disadaptation and re-adaptation to physical activities (a manifestation of stress, rate adaptation, cybernetic principles of homeostasis) have been identified and described. The two-circuit system homeostasis: the autonomic (controlling) and myocardial-hemodynamic (controlled) was demonstrated and an hierarchy of control has been defined.

Abstract Abstract 2299 Introduction Hematopoietic stem cells (HSCs) constitute a reservoir of undifferentiated cells that can be committed, upon appropriate stimuli, in the haematic lineages. Although residing in a bone-marrow hypoxic microenvironment (niche) and mainly relying on anaerobic glycolysis, HSCs are endowed with mitochondria. Recently, specific interest has been focused on HSCs mitochondria and on their role as reactive oxygen species (ROS) generators during the early phases of commitment. Indeed, consolidated evidences highlight the importance of redox signalling in the homeostasis of fundamental processes in cell adaptive biology and particularly in controlling the balance between self-renewal and differentiation of stem cells. HSCs constitutively generate low levels of ROS produced by both mitochondrial respiratory chain and NADPH oxidases (NOXs). ROS would act as secondary messengers, modulating the expression of master transcription factors leading or (pre)conditioning stem cells towards differentiation. Myelodisplastic syndrome (MDS) is characterized by disturbance of the HSC differentiation and most MDS patients are treated with iron chelators to compensate for the iron overload consequent to the blood cell transfusion-based standard therapy. Intriguingly, a robust percentage of patients, treated with the iron chelator deferasirox (DFX), recover correct HSCs differentiation whereas other chelators, like deferoxamine (DFO) did not. In the presented study we investigated the effect of DFX and DFO on the redox homeostasis of hematopoietic stem/progenitor cells (HSPCs) in order to get insights on the differential effect of iron chelators in rescuing altered hematopoiesis. Materials and Methods Human HSPCs were isolated from peripheral blood (PB) or bone marrow (BM) of G-CSF-treated or untreated healthy donors, respectively, by immuno-selection against the specific markers CD133 and CD34 and resulted >99% immunophenotypically homogeneous. Mitochondrial respiratory activity was measured in intact HSPCs by high resolution oxymetry. Morpho-functional features of HSPC-mitochondria, expression of the HSPC-surface commitment markers and ROS level were analyzed by laser scanning confocal microscopy (LSCM) and flow cytometry using specific probes or antibodies. HSPCs were treated with 100 mM DFX or DFO for 24 hrs. Results Measurement of oxygen consumption rate as well as molecular analyses in PB-HSPCs confirmed a poor mitochondrial oxidative phosphorylation phenotype. LSCM imaging of mitochondria in either PB- and BM-HSCs displayed a punctuate rather than interconnected network. However, co-staining of mitochondria and CD34/CD133 stemness-markers revealed a striking inverse correlation. Finally HSPCs produced DCF-detectable and DPI-inhibitable ROS attributable to constitutive NOX activity and related to stabilization of the hypoxia-inducibile factor (HIF1a) under normoxic condition. DFX treatment of HSPCs resulted in a significant up-regulation of ROS level whereas no significant change was observed following DFO treatment. Importantly, the DFX-mediated ROS production was insensitive to treatment with low NOX-inhibiting concentration of DPI but was abrogated by high concentration of DPI thus pointing to mitochondria as ROS source. Conclusions Our results show that HSCs in the early phase of commitment undergo a progressive increase of mitochondrial mass indicating the need of a bioenergetic up-regulation to cope with the oncoming energy-demanding proliferative/differentiative phenotype. Redox signaling, mediated by ROS production and likely triggered by changes in the environmental oxygen tension, appears to be essential in regulating HSC self-renewal and preservation of pluripotency. DFX treatment, by modulating ROS production, might lead to activation of redox-sensitive key factors able to restore the hematopoietic function in MDS patients. This effect seemingly is independent on the iron-chelating property of DFX but pertains to additional pharmacological properties that warren further investigation. The article is devoted to the study of ultrasound changes in the fetoplacental complex in the early gestational period in pregnant womenwith miscarriage. Placental insufficiency (PI) is a syndrome caused by morpho-functional changes and is the result of a complex responseof the fetus and placenta to various pathological conditions of the maternal body [1]. It is based on disorders of compensatory-adaptivemechanisms of the fetoplacental complex (FPC) at the molecular, cellular and tissue levels. Ultrasound fetometry and placentometry are very important methods for diagnosing the state of the fetoplacental complex and can be used from the early term. Many researchers identify a number of prognostically unfavourable ultrasound markers: low chorion attachment, especially with the appearance of areas of detachment, discrepancy between the size of the foetus and gestational age, lack of clear visualization of the embryo, especially its heartbeat. In pregnant women with a history of miscarriage it is important to identify early signs of placental dysfunction from the firsttrimester onwards and to provide adequate correction for this condition. For this situation it is important to create appropriate conditions for the development and growth of the placenta and its adequate functioning. Significant fetal abnormalities occur as early as the first trimester and have a significant impact on the further process of ontogenesis.Aims and objectives of the study. To assess the normal parameters of embryonic and extraembryonic formation and to evaluate theparticular features of the formation and development of the fetoplacental system during pregnancy in women with a history of miscarriage.Materials and Methods. We erformed a comprehensive ultrasonographic examination of 25 somatically healthy women with aphysiological gestational process (control group) between 5 - 40 weeks’ gestation and 25 pregnant women with a history of miscarriage (study group). The SONOACE 8800 GAI MT with a 3.5 to 7.5 MHz convex transducer was used in the study. The data were statistically processed using standard methods of mathematical analysis with the use of Student’s and Fisher’s criteria, standard and specialized computer programs. Paired correlation indices were used to study the nature and degree of correlation between different parameters. The scientific research was positively evaluated by the Bioethics Commission of the Bukovinian State Medical University (Protocol No. 4 of 6.12.2021).Results and their discussion. In the transvaginal examination, uncomplicated pregnancy is characterized by the mandatoryvisualization of the embryo in the fetal cavity with a diameter of 14 mm or more, corresponding to 6 weeks' gestation. From the time of embryo identification in our study, we measured the coccygeal-parietal size (CPS) and compared its values with gestational age. It was found that in 22 (88.0%) cases the embryo size corresponded to the calculated gestational age and the mean inner diameter of the fetal egg. In severe clinical manifestations of threatened miscarriage the most sensitive echographic sign was a decrease in the volume of the foetus, which is a marker of further adverse pregnancy course and outcome. However, when there was an isolated increase in myometrial tone in the absence of clinical manifestations of threatened abortion, there was predominantly an isolated reduction in amniotic cavityvolume, easily corrected by the administration of conventional antispasmodic therapy.The ultrasonic criteria for alterations in pregnancy were developed, including delay in embryo CTR by 2 weeks or more on ultrasound examination up to 9 weeks' gestation; bradycardia at 90 bpm or less up to 8-12 weeks' gestation; chorionic detachment withretrochorial hematoma formation (over 25 ml); tachycardia at over 200 bpm with clinical manifestations of spontaneous miscarriage; marked progressive decrease of amniotic cavity volume; severe polyhydramnios with a thick echopositive suspension in the amniotic cavity. The risk of spontaneous miscarriage and formation of placental insufficiency increases with the simultaneous detection of 2 or more echographic markers.Conclusions. On the basis of the above, it is possible to conclude on the need to conduct an ultrasound examination with an assessment of the echographic parameters of the formation and development of the embryo and extraembryonic structures in the first trimester in pregnant women with a history of miscarriage and a risk of developing placental insufficiency in the history in order toidentify markers of a complicated course of gestation and the and the subsequent choice of rational tactics of pregnancy management.

Adult male rats (Wistar lineage) were alcoholized with sugar cane liquor diluted at 30(0) GL during 300 days and sacrificed every 60 days in 5 stages. Samples of choroid plexuses of lateral ventricles were collected and examined at transmission electronic microscope to detect possible ultrastructural alterations and to raise possible pathological correlations. Gradual changes were observed in these animals during all the experiment: dilatation and enlargement of cisternae of Golgi complex, dilatation of RER, presence of digestive vacuoles and a large amount of pinocytic vesicles as well as vesicles with electronlucent content throughout cytoplasm, as well as an enlargement of intercellular space between basolateral interdigitation of the cells and of the connective tissue. The changes observed in the epithelium and connective tissue of choroid plexuses specially in 240 and 300 days of treatment are presumably due to a disturbance in hydroelectrolitic homeostasis, contributing to several morpho-functional disturbs of central nervous system. No changes were observed in the control group animals.

Great consideration is placed on the choice of capping agents’ base on the proposed application, in order to cater to the particular surface, size, geometry, and functional group. Change in any of the above can influence the characteristics properties of the nanomaterials. The adoption of hexadecylamine (HDA) as a capping agent in single source precursor approach offers better quantum dots (QDs) sensitizer materials with good quantum efficiency photoluminescence and desirable particles size. Structural, morphological, and electrochemical instruments were used to evaluate the characterization and efficiency of the sensitizers. The cyclic voltammetry (CV) results display both reduction and oxidation peaks for both materials. XRD for SnS/HDA and SnS photosensitizers displays eleven peaks within the values of 27.02° to 66.05° for SnS/HDA and 26.03° to 66.04° for SnS in correlation to the orthorhombic structure. Current density–voltage (I–V) results for SnS/HDA exhibited a better performance compared to SnS sensitizers. Bode plot results indicate electrons lifetime (τ) for SnS/HDA photosensitizer have superiority to the SnS photosensitizer. The results connote that SnS/HDA exhibited a better performance compared to SnS sensitizers due to the presence of HDA capping agent.

Changes in the levels and the dynamics in the development of anaerobic capabilities are of utmost importance in basketball – a sport with intermittent and speed-strength characteristics. The objective of the study is to establish the anthropometric and anaerobic profile of elite basketball players from the national team of women up to 20 years of age. This profile will help the future selection, forecasting and the refinement of the training methodology for the preparation of elite basketball players of this age group, In order to solve the identified tasks, we have used the following methods of study: anthropometrics, BMI, cycle-ergometrics, and mathematics-statistical methods (variational and correlations analyses). As a result of the carried-out training process, between the two tests we have noticed statistically reliable changes in the AcTM (active total body mass), PRMM (percentage of the muscle mass out of the total), PP (peak power), and AnP (average anaerobic power). Based on the conducted research, a model was created for the assessment of the studied morpho-functional indicators and the anaerobic capabilities of under-20 y.o. women basketball players, for their participation in a European Championship.

Aim: to study characteristics of hemodynamics, spectral analysis of heart rate variability (HRV) and their association with the total blood concentration of stable metabolites of nitric oxide (nitrites and nitrates) in patients with hypertension living in highlands. Patients and methods: 110 natives of the highlands of Elbrus region were examined. All of them were divided into 2 groups — with 2d stage of hypertension (n =50) and conditionally healthy (n =60). All the participants were performed routine examination under the conditions of hospitals and out-patient clinics. Moreover they were performed ECG, rhythm-cardiography, echo-CG evaluation of the blood pressure, renal and thyroid ultrasound and eye-ground examination. The assessment of HRV was made. Results: in the group of patients with hypertension there was decrease in the total power of HRV — TP, low frequency range — VLF, LF and high frequency range HF. The main characteristics of the central and intracardiac hemodynamics were significantly higher in patients with hypertension in decreasing of nitric oxide production. Conclusions: reverse reliable correlations were established between low frequency values of HRV, morpho-functional characteristics of the left ventricle and blood concentration of nitric oxide in patients with hypertension.

Aim. To study the condition of the right heart and hepatolienal circulation in patients three months after COVID-19.Material and Methods. A total of 87 patients aged on average 36.2 years who were treated for COVID-19 three months before underwent echocardiography and the study of hepatolienal blood flow. Patients were divided into groups: group 1 comprised patients with CT 0; group 2 comprised patients with bilateral pneumonia CT 1–2, and group 3 comprised patients with CT 3–4. Control group comprised 22 patients who did not undergo COVID–19.Results. In patients of group 1, the diastolic velocity of the transtricuspid flow increased by 24% compared to the control p <0.001. The ratio of diastolic velocities E/A and Em/Am decreased in all groups, and the greatest abnormalities were found in group 3. In group 3, systolic pressure in the pulmonary artery increased; the right ventricle, fibrous ring, right branch of the pulmonary artery, and diameters of the inferior vena cava, portal vein, and splenic veins increased. There was a decrease in the flow rate in the splenic vein (14%), and sizes of the liver and spleen increased. Correlations were established between increased pressure in the pulmonary artery and functional parameters of the heart and hepatolienal blood flow as well as between morpho-functional parameters of the heart and indicators of the portal vein, p < 0.001.Conclusion. Three months after the coronavirus infection, patients with mild and moderate severity of the disease developed subclinical abnormalities in the diastolic function of the right ventricle. In patients with severe viral pneumonia, there was also increased pressure in pulmonary artery; the dilatations of the right ventricle, pulmonary artery, splenic vein, and portal veins developed along with a decrease in the flow rate in the splenic vein and increases in the liver and spleen.

Objective: The increase of morbidity results from both an increase of life expectancy of the population, and influence of various risk factors contributing to development and increase of chronic heart failure (CHF). The combination of several atherogenic mechanisms (abdominal obesity (AO), insulin resistance (IR), arterial hypertension (AH), hyperglycemia, dyslipidemia), combined as “metabolic syndrome” (MS), causes a more rapid development of CHF. Materials and methods: The research finding of 74 patients with class II-III of CHF, including 37 patients (50%) with MS, are presented. The age structure of the pathology, severity of clinical course, data of laboratory and instrumental examination in various groups of patients were evaluated. A special program included an echocardiographic test with an assessment of various myocardial parameters. Results and Discussion: Research materials find out a number of characteristics of CHF clinical course (its earlier development and severe course) in patients with MS. Echocardiographic tests reveal an increase of heart chambers sizes, thickness of left and right ventricle, pulmonary hypertension. Myocardium morpho-functional changes are more significant in patients with CHF and MS than in those without MS. An increase in leptin levels, a marker of obesity, fibrosis and inflammation, has been found. Leptin, C-reactive peptide (CRP) and high-sensitive troponin in patients with MS significantly exceeded those in patientswithout MS. Correlations of leptin levels, adiponectin, CRP and left ventricular mass, thickness of epicardial fat (TEF), ejection fraction were established. Conclusion: Materials of the research indicate the important role of inflammatory and dysmetabolic processes in development and progression of CHF in patients with MS. The problem of asymmetry is one of the fundamental in modern biology, theoretical and clinical medicine, it has deep evolutionary roots. Analysis of literature suggests that the coverage of various aspects of morpho-functional asymmetry of the kidneys to this time is not always unambiguous, and sometimes controversial. The purpose of the work is to establish manifestations of asymmetry of entering into regression models of sonographic sizes of kidneys anthropometric and somatotypological parameters of a body in practically healthy women and men of different somatotypes. Within the framework of the agreement on scientific cooperation from the database of the research center of the National Pirogov Memorial Medical University, Vinnytsya taken the primary sonographic parameters, anthropometric and somatotypological indicators of 121 practically healthy women and 97 practically healthy men of the first mature age (correspondingly, of the age from 21 to 35 and from 22 to 35 years old) who in the third generation live on the territory of Podillia region of Ukraine. For the construction of regression models of individual sonographic sizes of the kidneys, depending on the peculiarities of anthropo-somatotypological parameters of the body, the license package “Statistica 6.1” was used. To analyze the asymmetry of entering into regression models of sonographic sizes of anthropo-somatotypological indicators, all constructed reliable models, but not only models with a determination coefficient of greater than 0.6, were taken into account. Also, in the analysis, anthropometric and somatotypological indicators refer either to highly genetically determined (longitudinal body sizes, cephalometric indexes, width of distal epiphyses of long tubular limb bones, trunk diameters, pelvic sizes, ectomorphic component of somatotype, bone component of body weight) or to low genetically determined (mass, body surface area, body dimensions thickness of skin fat folds, endomorphic and mesomorphic components of the somatotype, muscle and fat components of the body mass). In men of various somatotypes, when analyzing the asymmetry of entering the models of sonographic sizes of the anthropometric and somatotypological indicators of kidneys, it has been established: in the mesomorphic somatotype representatives - 84.1% (among which 50.0% belong to highly genetically determined indices), in the representatives of the ecto-mesomorphic somatotype - 86.0% (among which 46.5% belong to highly genetically determined indicators), in the representatives of the endo-mesomorphic somatotype - 88.1% (among which 51.4% belong to a highly genetically determined indices). In women of various somatotypes, when analyzing the asymmetry of entering the models of sonographic sizes of the anthropometric and somatotypological indicators of kidneys, it was established:), in the representatives of the mesomorphic somatotype - 82.0% (among which 51.2% belong to highly genetically determined indices), in the representatives of the ectomorphic somatotype – 89.0% (among which 58.0% belong to highly genetically determined indicators), in the representatives of the endo-mesomorphic somatotype - 83.3% (among which 50.0% belong to highly genetically determined indicators), in the representatives of middle intermediate somatotype - 82.5% (among which 56.5% belong to highly genetically determined indicators).

Abstract The targeted therapy ibrutinib inhibits B cell receptor signaling (BTK inhibitor) and has yielded high response rates and durable remissions in patients with chronic lymphocytic leukemia (CLL). However, it is widely believed that the addition of immune therapies to targeted drugs will be required to activate anti-tumor immunity and work towards curative therapy. Identifying effective combinations of targeted drugs and/or standard chemotherapy with immunotherapy is a priority research area and particularly relevant for CLL, as patients' T cells have been shown to exhibit profound tolerance/exhaustion and notably, no activity was reported in a recent trial of anti-PD-1 immunotherapy for relapsed disease. Ibrutinib has shown beneficial immunomodulatory activity in CLL by inhibiting IL-2-inducible T cell kinase (ITK) as well BTK that is associated with increased effector CD4+and CD8+ T cell numbers and decreased expression of inhibitory checkpoint receptors such as PD-1 on patient T cells. Here we have performed comparative immune bioassays from a randomized phase III trial comparing ibrutinib-based therapy to traditional FCR chemoimmunotherapy to assess the effects of treatments on anti-tumor T cell function. Viable peripheral blood mononuclear cell samples were collected serially (baseline, 6 months and 12 months) from CLL patients on the randomized phase III E1912 trial of ibrutinib and rituximab versus FCR for previously untreated disease to allow longitudinal batched immune analysis. Cytotoxicity assays revealed that highly purified CD3+ T cells from the FCR treated patients at 6 and 12-month time-points did not change their activated killing function against autologous baseline CD19+ CLL tumor B cells (acting as target antigen-presenting cells pulsed with superantigen, sAg) compared to pre-treatment/baseline exhausted T cells (n=22). In contrast, patients treated with ibrutinib-based therapy had a significant increase in activated anti-tumor T cell killing function (P<.01, n=22) at both 6-month (66% increase) and 12-month (89% increase) time-points. Flow cytometric analysis of circulating immune subsets revealed that the percentage of PD-1 and PD-L1 positive cells among CD8+ and CD4+ T cells (particularly effector compartments) were reduced with ibrutinib-based therapy, whereas only a partial reduction was detected following FCR treatment. However, patients' T cells from both treatment arms responded normally to T cell receptor engagement by upregulating these checkpoint molecules. This led us to explore ex vivo treatment of highly purified CD3+ T cells and CD19+ CLL B cells from both treatment arms with anti-PD-L1 or anti-PD-1 immunotherapy prior to cytotoxicity assays. Our functional data revealed that the T cells from both FCR time-points (6 and 12-months) were not sensitive to either anti-PD-L1 (n=14) or anti-PD-1 (n=14) treatment. In contrast, ibrutinib-based treatment sensitized anti-tumor T function (23% increase in killing) following anti-PD-L1 treatment (n=14) at the 6-month time-point only (P<.01) but not with anti-PD-1. To investigate the mechanism underlying these effector function differences, we compared the ability of highly purified CD3+ T cells from each treatment arm (n=45) time-point to form F-actin immunological synapses with baseline autologous CLL tumor B cells. Quantitative confocal image analysis revealed that ibrutinib-based therapy significantly (P<.01) enhanced polarization of F-actin, tyrosine-phosphorylated proteins and granzyme B at immune synapses with tumor cells at both 6 and 12-month time-points, whereas FCR treated patient T cells failed to mobilize these lytic synapse molecules. Importantly, our assays have revealed that T cells from both FCR treatment time-points formed "non-polarized" immune synapses with tumor cells, in keeping with cytotoxic dysfunction and insensitivity to additional checkpoint immunotherapy. In contrast, our functional correlative bioassays have revealed that ibrutinib-based therapy can reactivate exhausted cytolytic T cell function and suggest to us, a potential therapeutic window for anti-PD-L1 immunotherapy at the earlier 6-month time-point. We believe this data supports the concept of incorporating functional bioassays to immune-monitoring assays associated to clinical trials that should aid knowledge-led design of future combination immunotherapy. Abstract Introduction: It has become increasingly apparent that chimeric antigen receptor T (CART) cell activation and differentiation level is an important determinant of CART cell fate and response to therapy. In this study, we aimed to 1) measure levels of activation-induced surface death receptors and ligands on CART cells; 2) investigate how CART cell activation could impact their fitness and clinical responses, and 3) identify cell-based targets to modulate CART cell activation, apoptosis, and cytotoxicity to improve anti-tumor activity. Methods: We performed flow cytometric studies on ex-vivo stimulated, clinically annotated CART products of patients with large B cell lymphoma from the pivotal ZUMA-1 clinical trial that led to FDA-approved Axicabtagene ciloleucel (Axi-Cel). We investigated possible correlations of a number of surface death receptors and ligands with T cell differentiation status and post-infusion CART cell expansion, utilizing samples from ZUMA-1 patients who achieved a complete response as a best outcome ('responders') vs patients who achieved stable or progressive disease('non-responders'). CART cell effector functions in vitro were measured, and CART apoptosis was assessed using Annexin V. For in vitro and in vivo functional studies, we used CART19 generated from healthy donors (HD CART19) as indicated in the specific experiment. CRISPR/Cas9 was employed during CART cell production to disrupt specific genes. A xenograft model of lymphoma was used to investigate the in vivo antitumor activity of CART19. Results: Following an ex vivo stimulation of Axi-Cel products with CD19 + target cells, we observed upregulation of death receptors and ligands in CART19 from non-responders, compared to responders. We also observed a possible association between such upregulated surface markers with CART cell differentiation as measured by CCR7 expression. In an extended in vitro co-culture assay, where HD CART19 cells were repeatedly stimulated through the CAR, we found that tumor necrosis factor α receptor 2 (TNFR2), unlike other death receptors and ligands, was persistently elevated, suggesting a possible role for TNFR2 in long-term antigen-dependent CART19 dysfunction (Figure 1A). We further found that HD CART19 upregulate TNFR2, but not TNFR1, upon CAR stimulation (Figure 1B). While non-specific TCR activation (CD3 stimulation) of HD CART19 cells protected them from activation-induced apoptosis, antigen-specific activation through the CAR resulted in significant initiation of apoptosis within 2 hours of stimulation (Figure 1C). Having identified a possible association between TNFR2 and CART19 dysfunction, we aimed to study the impact of TNFR2 knockout on HD CART19 functions. Using CRISPR/Cas9 during CART cell manufacturing, we generated TNFR2 k/o HD CART19 cells with a knockout efficiency of around 50%, where the expression levels of TNFR2 in activated CART19 cells were reduced, compared to control CART19 cells (with non-targeting gRNA CRISPR/Cas9, Figure 1D). TNFR2 k/o CART19 cells demonstrated reduced early activation surface markers compared to control CART19, as measured by CD25 and CD69 surface expression (Figure 1E), reduced apoptosis initiation as measured by the Annexin V assay (Figure 1F), and enhanced antigen-specific proliferation and cytotoxicity (Figure 1G). Finally, in an in vivo xenograft model of CD19 + lymphoma, TNFR2 k/o CART19 resulted in enhanced CART cell expansion and anti-tumor activity (Figure 1H). Conclusions: Our results indicate that TNFR2 plays a role in early activation and apoptosis initiation of CART19 following CAR stimulation with CD19 + target cells and present TNFR2 knockout as a strategy to enhance CART19 anti-tumor activity. AbstractPurpose. Since human voice characteristics are specific to each individual, numerous anthropological studies have been oriented to find significant relationships between voice and morpho-functional features. The goal of this study was to identify the correlation between seven morpho-functional variables and six voice acoustic parameters in sportsmen. Methods. Following the protocols of the International Biological Program, seven morpho-functional variables and six voice acoustic parameters have been measured in 88 male professional athletes from Kosovo, aged 17-35 years, during the period of April-October 2013. The statistical analysis was accomplished through the SPSS program, version 20. The obtained data were analysed through descriptive parameters and with Spearman’s method of correlation analysis. Results. Spearman’s method of correlation showed significant negative correlations (R = -0.215 to -0.613; p = 0.05) between three voice acoustic variables of the fundamental frequency of the voice sample (Mean, Minimum, and Maximum Pitch) and six morpho-functional measures (Body Height, Body Weight, Margaria-Kalamen Power Test, Sargent Jump Test, Pull-up Test, and VO

Background Left ventricular (LV) diastolic dysfunction (DD) is a hallmark of hypertrophic cardiomyopathy (HCM) and its phenocopies, such as Fabry disease (FD). Together with left atrial (LA) size, LA function is emerging as a sensitive marker of the adaptive changes to backward transmission of LV cardiac filling pressure, thus implementing DD assessment. Additionally, both HCM and FD are characterized by a primitive atrial myopathy, but LA morpho-functional changes in HCM and FD have never been directly compared. More recently, LA strain by Cardiovascular Magnetic Resonance Feature Tracking (CMR-FT) has been demonstrated to be a feasible and reproducible tool to explore LA function. Purpose To compare LA morpho-functional changes in HCM and FD and to explore their correlation with tissue alterations. Methods 15 HCM and 15 sex-, age- and LV mass index-matched FD patients underwent CMR (Magnetom Aera 1.5T, Siemens) and Doppler Echocardiography for LV diastolic function assessment (E/e’ and DD grading from 0 to 3). LA phasic function was evaluated by CMR-FT strain (Qstrain Medis). The software output included passive (εe, conduit function), active (εa, booster pump function) and total strain (εs, reservoir function), along with LA volumes and ejection fraction (EF). Late gadolinium enhancement (LGE) was quantified as a percentage of LV mass using the standard deviations (SDs) method (≥ 5 SDs). Interstitial fibrosis was assessed by extracellular volume (ECV) quantification in remote myocardium. All patients were in sinus rhythm. Results In the HCM group, the proportion of patients with DD grade 2-3 was only slightly higher than in FD (p 0.26). Accordingly, no significant difference was found in E/e’ value (p 0.78). Compared to FD, HCM patients showed more severe LA morpho-functional changes, including larger LA end-systolic volume (ESV) (113 ± 35 vs 84 ± 23 ml), lower LA EF (37 ± 7 vs 44 ± 9 %) and a greater reduction of εs (-20 ± 5 vs -25 ± 6 %) and εa (-10 ± 4 vs -15 ± 4 %) (all p < 0.05). LV size and function and the burden of fibrosis (LGE quantification and ECV) were comparable between the two groups. Interestingly, in HCM population, unlike in FD, LA morpho-functional measurements significantly correlated with tissue characterization parameters (LA ESV with LGE, r 0.56, p 0.03; εs and εa with ECV, r -0.51, p 0.05 and r -0.59, p 0.02, respectively). Conclusions LA morpho-functional alterations are much more severe in HCM compared to FD with similar degree of LV hypertrophy. A more severe atrial myopathy or different mechanisms of atrial damage in the two cardiomyopathies may explain these findings. LA CMR-FT analysis may represent a sensitive tool to discriminate between HCM and FD, although larger studies are needed to confirm this finding and the possible correlation with the occurrence of atrial arrhythmias and thromboembolic risk.
