Annotation:

The localization of calcitonin in hypothalamic tissue and cerebrospinal fluid and calcitonin binding sites in circumventricular organs suggests involvement in neural mechanisms, possibly hydromineral balance. Therefore, we compared the effects of intracerebroventricular and intravenous infusions of porcine calcitonin (0.04-0.05 mU X kg-1 X min-1) on renal water, Ca, Mg, Na, and K excretions. The intracerebroventricular administration of calcitonin for 60 min induced a pronounced increase (P less than 0.01) in free water clearance, from a preinfusion value of -0.73 +/- 0.45 to +3.02 +/- 1.15 ml/min at the end of the infusion. In contrast, the free water clearance was unaltered during the intravenous infusion of calcitonin. Renal Ca excretion fell significantly (53%) 40 min after the end of the intracerebroventricular infusion of calcitonin, whereas the corresponding intravenous infusion of calcitonin did not change (P greater than 0.05) urinary Ca excretion. Renal Mg excretion was increased (P less than 0.01) during the intracerebroventricular infusion of calcitonin but was not altered during intravenous infusion of calcitonin. Urinary excretions of Na and K were not affected by either intracerebroventricular or intravenous administration of calcitonin. It is concluded that calcitonin can act centrally to modify renal water, Ca, and Mg excretions.

The present study was conducted to investigate local effects of endothelin-1 (ET-1) on basal adrenal catecholamine (CA) secretion and underlying mechanisms of action of ET-1 in anesthetized dogs. ET-1 was locally administered to the left adrenal gland through a catheter inserted into the left adrenolumbar artery. Plasma CA concentrations in adrenal venous and aortic blood were determined by an HPLC–electrochemical method. The local infusions (1 min, 0.5 mL/min) of ET-1, in doses of 0.01, 0.1, and 1.0 μg/mL, resulted in a dose-dependent increase in epinephrine secretion, rising from an initial basal value of 6.9 ± 1.4 ng∙min−1∙g−1 to maximum values of 7.6 ± 1.5, 81.1 ± 38.3*, and 680.9 ± 107.6* ng∙min−1∙g−1 (*p < 0.05, n = 7), respectively. Neither mean arterial pressure nor plasma CA levels in aortic blood altered following ET-1 infusions. A similar dose-dependent increase in norepinephrine output was also observed with ET-1. Following the initial peak observed during infusion with the highest dose of ET-1, the output of both epinephrine and norepinephrine remained significantly elevated over a period of 30 min. This ET-1-induced, long-lasting increase in adrenal CA output was abolished by nifedipine (100 μg/mL) similarly administered 10 min before ET-1 infusion. The results indicate that ET-1 directly enhances the basal secretion of CA from dog adrenal medulla in vivo. The study suggests that the ET-1-induced increase in adrenal CA secretion is, at least in part, mediated by the opening of dihydropyridine-sensitive L-type calcium channels at the level of adrenal medullary chromaffin cells.Key words: adrenal catecholamines, anesthetized dogs, calcium channels, endothelin-1, epinephrine, intra-adrenolumbar arterial infusion, nifedipine, norepinephrine.

To better delineate acid-base transport properties in the distal tubule (DT) of Necturus in vivo, we 1) studied the effects of peritubular (pt) isohydric increase of PCO2 and [HCO3-]pt on luminal pH (pHlu), and 2) measured the steady-state pHlu under various experimental conditions. The experiments were carried out on initial (DTi) or distal (DTd) loops of the DT in control state and then during intravenous infusion of carbonic anhydrase (CA) or CA inhibitors (CAI). In control state, isohydric increase of PCO2 and [HCO3-]pt results in transient acidification of the DTi lumen, whereas in DTd lumen the same maneuver yields sustained (plateau) acidification. Under systemic infusion of CAI, isohydric increase of PCO2 and [HCO3-]pt lowers pHlu (sustained fall of pHlu) in DTi and DTd, whereas under CA infusion both segments exhibit only transient acidification. During intravenous infusion with benzolamide DTi steady-state pHlu falls, suggesting that this maneuver inhibits a functional luminal CA, in contrast to the DTd, whose pHlu remains unaltered. Intravenous infusion of CA significantly increases steady-state DTd pHlu; by contrast, steady-state pHlu in DTi does not change. These data are consistent with the presence of functional luminal CA in the DTi, whereas the DTd segment lacks the luminal enzyme.

The objective of the present study was to evaluate the effects of an antioxidant and anti-inflammatory compound found in rosemary plants (Salvia rosmarinus) named carnosic acid during the transition period of dairy cows. From day 1 to 3 after calving, 16 multiparous Holstein cows received a daily intravenous infusion of either 500 mL of saline (NaCl 0.9%; Saline; n = 8) or carnosic acid at a rate of 0.3 mg/kg of BW supplied in 500 mL of saline (CA; n = 8). Blood samples were taken at –7, 2, 5, 7, 14, and 21 d relative to parturition, then analyzed for metabolites related to energy metabolism, muscle mass catabolism, liver function, inflammation, and oxidative stress. CA infusion tended to improve milk performance; however, DMI was unaffected by treatment. At 2 d relative to parturition, CA cows had lower blood concentrations of haptoglobin, paraoxonase, FRAP, and NO2– than saline cows. After treatment infusions, haptoglobin remained lower in CA cows than saline at 5 d relative to parturition. Our results demonstrate that carnosic acid promoted positive responses on inflammation and oxidative stress biomarkers and may promote beneficial effects on lactation performance in peripartal dairy cows.

Parathyroid hormone (PTH) seems to affect the risk of cardiovascular disease. The aim of the present study was to investigate PTH's acute effect on endothelial vasodilatory function in forearm resistance vessels. Ten healthy subjects underwent forearm venous occlusion plethysmography. We measured forearm blood flow at baseline and at a stable, locally increased PTH level after intra-arterial infusion of metacholine and nitroprusside. The contralateral arm served as a control. Ionized calcium (Ca++) and PTH values were normal in all subjects at baseline (1.26 ± 0.02 mM/L, 3.6 ± 1.2 pM/L). After 30 minutes of PTH infusion, the PTH level increased in the active arm (13.8 ± 4.0 pM/L P < 0.01), while the Ca++ level was unchanged (1.25 ± 0.04; mM/L). Both the PTH and the Ca++ level in the contralateral arm remained unchanged, which indicates no systemic influence. The endothelial-dependent vasodilation was inversely correlated to the Ca++ level at baseline (r = −0.75, P < 0.05) and after PTH infusion (r = −0.68, P < 0.05). The vasodilatory function was not affected during PTH-infusion.

ABSTRACT Rat liver exhibits a reversed sexual dimorphism of its two endogenous soluble carbonic anhydrase (CA) isozymes, CA II and CA III. Normal males have hepatic CA III concentrations ten–twenty times those in the female, while female liver contains two–three times more CA II than the male. Hypophysectomy abolishes this sexual differentiation, having no effect on male liver but producing isozyme concentrations in the female liver similar to those in the male. Clinacanthus nutans L. (locally known as ‘Sabah Snake Grass’) has been used traditionally to treat chronic diseases. However, there is insufficient information regarding the nutritional quality of the herbal. This study was conducted to evaluate the nutritional properties and trace elements contents of unfermented and fermented herbal teas developed from C. nutans leaves using different drying techniques (microwave- oven dried and freeze dried) in different infusion time (1, 2, 5, 10, 15 and 20 min). The proximate analysis were conducted according to AOAC’s standard methods, while, the colorimetric color of infusions were determined using HunterLab Color Meter. The trace elements in infusions were determined using Inductively Coupled Plasma-Optical Emission Spectrophotometric (ICP-OES) analysis. Among the C. nutans herbal teas, the freeze dried of unfermented herbal tea showed high ash (12.39 ± 0.39%) and fat (2.23 ± 0.10%) content, while, protein content was showed high in freeze dried of fermented herbal tea (23.15 ± 0.51%). Carbohydrate content was showed high in microwave-oven dried of unfermented herbal tea (63.40 ± 0.53%) with 3026.24 ± 28.23 kcal/kg of Metabolizable Energy (ME). For color infusion determination, C. nutans herbal teas displayed no significant difference (P >0.05) in color darkness as compared to commercial teas (L: 3.63 to 5.77). There were no significant differences between unfermented and fermented C. nutans herbal tea for its greenish (a: -2.69 to -1.20) and yellowish (b: 3.45 to 5.59) color infusion. Sixteen elements (Al, Ba, Be, Ca, Fe, In, K, Li, Mg, Mn, Na, Zn, Se, Sr, Tl and Si) were detected in the C. nutans and commercial dried leaves. Potassium (K), calcium (Ca), magnesium (Mg) and sodium (Na) were higher in C. nutans herbal infusions as compared to commercial teas infusions. Consumption of these herbal teas as dietary intake is able to overcome nutrients and minerals deficiency.

Abstract The objective was to evaluate short-chain fatty acid (SCFA) absorption and permeability of the gastrointestinal tract (GIT) of heifers infused either with Ca-gluconate or Ca-butyrate. Thirty-two ruminally cannulated beef heifers were fed a common diet (forage-to-concentrate ratio of 50:50) for 28 d and once daily infused with water (ruminal infusion; control), Ca-gluconate embedded in a fat matrix (ruminal infusion; 0.192% BW), unprotected Ca-gluconate (abomasal infusion; 0.077% of BW), and unprotected Ca-butyrate (abomasal infusion; 0.029% BW). Treatments were designed to provide the same amount of butyrate to the small intestine. DMI was restricted to 95% of voluntary DMI on d 8 and was recorded until heifers were slaughtered on d 28. Rumen, jejunum, and colon tissues were collected to determine the rate and pathway of SCFA transport, and for measurement of permeability.14C-acetate and 3H-butyrate absorption across the ruminal and colonic epithelium was measured with no inhibition and under maximal inhibition. Permeability was assessed for the rumen, jejunum and colon using mucosal-to-serosal flux of 14C-mannitol. Initial and final BW were not different (P > 0.60) averaging 388 ± 5.5 kg and 409 ± 6.6 kg, respectively. DMI was not affected by treatment averaging 7.7 kg/d (P = 0.77). Treatment did not affect the flux of acetate or butyrate across the ruminal or colonic epithelium (P > 0.33), but flux rates of SCFA were numerically greater across the ruminal epithelium. Moreover, approximately 50% of the ruminal acetate was transported via bicarbonate-dependent mechanisms. Mannitol flux was not affected by treatment (P > 0.29) and was numerically lower in the rumen and colon than in the jejunum, supporting previous research evaluating permeability across the GIT. According to this experiment, provision of Ca-butyrate or Ca-gluconate in an attempt to increase intestinal butyrate supply does not impact SCFA absorption or permeability of the rumen, jejunum, or colon.

The objectives of the present study were to determine the effects of jugular vein infusions of glucagon like peptide-1 (GLP-1) and dietary fat inclusion on dry matter intake, nutrient digestibility and hypothalamic mRNA concentration of neuropeptide Y, agouti related peptide, and proopiomelanocortin in growing sheep. Thirty-six wethers were used (40.7±3.3 kg BW). Treatments were a control diet (n=11), dietary addition (6% of dry matter) of Ca salts of palm oil fatty acids (n=12), or 6-d jugular vein infusions of 0.155 µg kg−1 body weight/day of GLP-1 (n=11). Hormone concentrations were measured in jugular vein plasma from samples taken on days 1, 4 and 6. On day 7, the wethers were slaughtered for hypothalamus collection to measure mRNA concentration. The dietary addition of 6% of Ca salts of palm oil increased plasma GLP-1 concentration (P<0.01) and decreased dry matter intake on day 1, but not on day 6 (time×treatment interaction, P<0.05). The infusion of GLP-1 did not change dry matter intake (P>0.20), but increased neutral detergent fibre digestibility (P<0.01). In conclusion, glucagon like peptide-1 infusion or feeding fat did not decrease dry matter intake or affect hypothalamic neuropeptide mRNA concentrations of sheep.

La tenerezza è la caratteristica più importante per definire l'accettabilità dei prodotti da parte dei consumatori, mentre le carni ottenute da bovini allevati con sistema estensivo solitamente sono dure. Dopo una dissertazione sulle reazioni biochimiche che avvengono durante il periodo di frollatura e che provoca l’intenerimento della carne, questo studio si è proposto di applicare diverse tecniche per migliorare la tenerezza della carne derivante da animali allevati in estensivo, cercando di modulare le reazioni biochimiche. In questo studio tre differenti tecniche, infusione di calcio (Ca), contrazione da freddo indotta (VFC) e la sospensione per le pelvi (PS), anziché per il tendine di Achille (AT), sono state testate in fase di pre-rigor sulla razza Maremmana e Frisona. L’infusione di Ca ha riportato uno sforzo di taglio più basso rispetto al gruppo di controllo (5,17 vs 10,48 kg) mentre il gruppo VFC ha occupato una posizione intermedia. L’attacco per le pelvi ha dimostrato una differenza significativa sullo sforzo di taglio e sulla lunghezza dei sarcomeri sui muscoli Longissimus thoracis e Biceps femoralis (4,26 vs 5,88 kg e 3,83 vs 4,78 kg e 1,97 e 1,64 µm e 2,10 vs 1,76 µm, rispettivamente per i muscoli per WBS e lunghezza dei sarcomeri in PS e AT). In conclusione, questi metodi possono influire sul processo di intenerimento della carne.
Tenderness is the most important characteristic to define products acceptability by consumers, while meat obtained from bull rearing in extensive system is usually tough. After a wide dissertation on biochemical reactions that occur during the ageing time that cause tenderization of meat, this study propose differences techniques to improve meat tenderness acting on this important pathway. In this study three different methods, Calcium infusion (Ca), Very Fast Chilling (VFC) and pelvic suspension hanging from aitch bone (PS) or the Achilles tendon (AT) are applied in pre-rigor phase on Maremmana and Holstein meat. Ca injection has reported the lowest shear force on cooked meat compared to control group (5.17 vs. 10.48 kg) instead VFC group has intermediate values. Pelvic hanging has shown significant differences on shear force and sarcomere length on Longissimus thoracis (LD) and Biceps femoralis (BF) muscles (4.26 vs. 5.88 kg and 3.83 vs. 4.78 kg and 1.97 and 1.64 µm and 2.10 vs. 1.76 µm respectively for LD and BF per WBS and sarcomere length in PS and AT). In conclusion these methods could influence tenderization processes even if the action of some processes are still unclear.

This work presents Leibniz’s view of infinity and the central role it plays in his theory of living beings. Chapter 1 introduces Leibniz’s approach to infinity by presenting the central concepts he employs; chapter 2 presents the historical background through Leibniz’s encounters with Galileo and Descartes, exposing a tension between the notions of an infinite number and an infinite being; chapter 3 argues that Leibniz’s solution to this tension, developed through his encounter with Spinoza (ca. 1676), consists of distinguishing between a quantitative and a nonquantitative use of infinity, and an intermediate degree of infinity—a maximum in its kind, which sheds light on Leibniz’s use of infinity as a defining mark of living beings; chapter 4 examines the connection between infinity and unity; chapter 5 presents the development of Leibniz’s views on infinity and life; chapter 6 explores Leibniz’s distinction between artificial and natural machines; chapter 7 focuses on Leibniz’s image of a living mirror, contrasting it with Pascal’s image of a mite; chapter 8 argues that Leibniz understands creatures as infinite and limited, or as infinite in their own kind, in distinction from the absolute infinity of God; chapter 9 argues that Leibniz’s concept of a monad holds at every level of reality; chapter 10 compares Leibniz’s use of life and primitive force. The conclusion presents Leibniz’s program of infusing life into every aspect of nature as an attempt to re-enchant a view of nature left disenchanted by Descartes and Spinoza.

It has been reported recently that DDAVP might be an useful tool in the therapy and prevention of bleeding in patients with congenital afibrinogenemia (CA).To study the mechanism of its efficacy, changes in the platelet functions of a patient with CA were examined prior to, and one hour after, the infusion of DDAVP (0.4 μg/Kg). A patient with Glanzmann's thrombasthenia (GT) was also examined, to allow a study of the role of platelet membrane glycoprotein IIb/IIIa (GP IIb/IIIa), a deficient platelet in GT, in the resulting effects of the drug. When both patients were infused with DDAVP, the level of plasma von Willebrand factor (vWF) increased two- to fourfold, accompanied by an enhancement of ristocetin-induced platelet agglutination. The level of plasma fibrinogen was never changed.The prolonged bleeding time observed was markedly improved only in the CA patient, remaining unchanged in the GT patient, after the infusion of DDAVP. This indicates that DDAVP is effective in diminishing the bleeding tendency in CA, but not in GT. Among the platelet functions tested, only the platelet retention rate on glass beads, ADP-induced platelet aggregation and collagen-induced platelet aggregation improved in CA, each remaining unchanged in GT. In particular, collagen-induced platelet aggregation was markedly improved in the CA patient. However, the platelet adhesion to collagen (50 μg/ml)-Sepharose remained normal, both before and after the infusion of DDAVP in CA.These results suggest that an increase in the plasma vWF level and the existence of platelet membrane GPIIb/IIIa may be necessary for the improvement of primary hemostasis, after the infusion of DDAVP. The vWF-mediated platelet aggregation by collagen or ADP may produce this effect in the CA patient.

It has been suggested that a vWF High Molecular Weight Multi-mers (HMWM) decrease or a PAA were involved in the pathogenesis of HUS. We have studied 8 children (6 girls,_2 boys; 7 months-8_1/2 years old) with HUS : plasma creatinine /μmol/l; mean(range)/=306 (105-524), hemoglobin (g/100ml)-7(6.3-7.8), schistocytes (%)=8(1-18), platelets (x103/mm3)-57(10-115). The vWF was studied quantitatively (antigen ; vWF RAg assay) and qualitatively (multimeric pattern : immunoblotting and autoradiography). PAA studied by incubating the patient's platelet poor plasma (RPR) with washed normal platelets (aggregometer, % light transmission) and confirmed by Thromboxane B2 (TXB2) assay and [14C] Serotonine release study. The PAA was characterized by studying the in vitro effect of several platelet aggregation inhibitors, Immunoglobulins (Igs) and Fresh Frozen Plasma (FFP) on the platelet aggregation.An increase of vWF RAg (%) was observed in 6 cases : mean:330, and possibly related with renal failure. A vWF HMWM decrease was found in 3 patients : 2/3 with associated infection(E.Coli, Pneumococcus), 1/3 with severe hemolysis. Two of these 3 patients had a favourable renal outcome and 1 a severe course (chronic hemodialysis, Arterial Thrombotic MicroAngiopathy at renal histology).An important PAA was evidenced only in 1 patient : post bone-marrow graft HUS during neuroblastoma(NB),arterial hypertension and chronic renal failure. This PAA was Ca++, TXB2 and cAMP dependent; it was moderately inhibited in vitro by Igs and FFP, but persisted after 5 days of Igs infusion (0.3g/Kg/day). Treatment with aspirin and dipyridamole (10mg/Kg/day each) suppressed the patient platelet auto-aggregation although the PAA persisted (follow up:10months). The PAA did not seem to be related with the NB (absence of GD2 ganglioside, specific marker of NB); it could be related with anti platelet antibodies. The coexistence of the two abnormalities could not be demonstrated in our patients.In conclusion, a vWF HMWM decrease was found in 3 out of 8 children patients with HUS. Its presence was not correlated with the severity of the disease. We could demonstrate the presence of PAA during childhood HUS in only 1 post bone-marrow graft case. The PAA characterization is useful for therapeutic decisions and contributes to a better pathogenetic understanding.
