ABSTRACT Background: A good bowel preparation for colonoscopy is the most important factor that has an impact on visualization and outcome of the procedure. Aim: The primary aim of the study was to assess the quality of bowel preparation as reported by the Boston Bowel Preparation Score (BBPS). The secondary aim of the study was to analyze the patient feedback on ease of administration, palatability, and side effects with the bowel preparation. Materials and Methods: Patients undergoing colonoscopy between March 2018 and September 2018 were enrolled in the study. Patients <18 years of age, those with the previous history of colorectal surgery, emergency procedures in an unprepared colon, and those not willing to participate were excluded from the study. Colon preparation of the patient was decided by senior consultants. A predesigned pro forma that included demography, indication for the procedure, preparation details, dietary recommendations the previous day if any, side effects, and patient’s comfort to preparation was completed by two-independent observers. The BBPS was used to assess the bowel preparation. A score of <5 was deemed inadequate. P < 0.05 was considered statistically significant. Results: The study cohort consisted of 141 patients, of which 78 were male (55.3%). Eighty (56.7%) patients received oral sulfate-based preparation and 61(43.4%) polyethylene glycol-based preparation. Nearly one third of cases reported the solution to be non palatable. 15.4% respondents reported nausea, vomiting and bloating as the major side effects of the preparation. The median duration of colonoscopy was 25 min (8–45 min). One hundred and eighteen patients (83.6%) had a BBPS score of ≥5. Sulfate preparation resulted in better bowel cleansing (P = 0.01). Age, gender, and dosing schedule of preparation, including bedtime dosing of stimulant laxative, did not alter the BBPS score. Conclusion: Sulfate- and polyethylene glycol-based preparations are commonly used for cleansing the colon. Bowel cleansing was adequate in most patients and sulfate-based yielded better bowel cleansing. Nearly 15.4% of patients reported side effects to these preparations.

In this study, cellulose and cellulose nanofibers (CNF) were extracted and prepared from cassava peels (CPs). The method of the cellulose extraction was performed by alkali treatment followed by a bleaching process. The CNF were prepared by mechanical disruption procedure (homogenization and ultrasonication), and the results were compared with a common acid hydrolysis procedure. The resulting cellulose and CNF from both procedures were then analyzed using FTIR, SEM, TEM, XRD, and TGA. The results show that cellulose and CNF were successfully prepared from both procedures. The physical properties of the produced CNF were different; however, they had similar chemical properties.

Abstract Residual lignin preparations were isolated from birch, aspen and Eucalyptus grandis kraft pulp by enzymatic hydrolysis of the pulps with cellulase:hemicellulase mixture. Residual lignin preparations were characterized by investigation of nitrogen content, carbohydrate composition and molecular mass distribution. The use of enzyme with high activity and optimization of enzyme charge resulted in significant decrease in protein contaminants in residual lignin preparations as compared to previously published results. A second order law correlation between enzyme mass charge and nitrogen content in birch residual lignin preparations indicates a strong effect of enzyme charge on the amount of protein contaminants. However, the enzyme charge in the range studied does not appreciably affect either the yields of the residual lignin preparations or percentage and composition of carbohydrates in these preparations. The optimal enzyme charge was highest for Eucalyptus grandis pulp and lowest for birch pulp. It has been suggested that a significant part of the hardwood residual lignin in pulps, especially in E. grandis pulp, consists of low molecular mass lignin fragments bonded to pulp carbohydrates. Higher amount of glucose in E. grandis residual lignin preparation compared to those from birch and aspen implied a higher frequency of lignin-cellulose bonds in eucalypt pulp. Different fractions of birch residual lignin have rather similar molecular mass distribution, which was not affected by the charge of the enzyme. Eucalypt residual lignin preparation had higher molecular mass than birch residual lignin.

Turnaround (TA) is a large-scale maintenance activity (extraordinary maintenance activities), which is carried out periodically (repeated large-scale maintenance), where the work referred to can only be carried out when the unit is stopped operating. The main scope of work is inspection (inspection), repair, cleaning equipment. The success or failure of turnaround activities is very influential from careful preparation, some of the failures of turnaround activities result from unplanned preparations. One factor is the readiness of the material. This paper discusses the direct involvement of the material procurement process in determining better TA preparation time. The historical data from the purchasing is analyses and then the result is combined with the new proposed procedure of the TA maintenance. The new procedure has three months less than thecurrent procedure.

Objectives: To study an individual's experience of either flexible sigmoidoscopy (FS) or colonoscopy in a colorectal cancer prevention programme. Methods: Consecutive individuals in a Bowel Cancer Prevention Programme, who had either an unsedated FS or a colonoscopy with sedation, participated in a prospective cross-sectional questionnaire-based study. Results: A total of 447 responses were obtained for 256 colonoscopies and 191 FSs (200 men [45%] and 247 women [55%]). The overall experience of colonoscopy was more comfortable than FS (75% versus 18%; P<0.001). Embarrassment was low for both procedures (8%). There was no pain associated with colonoscopy and most individuals had a pain score of less than 3 (11-point scale) for FS: 72% of men, 55% of women ( P<0.001). Most individuals did not have a gender preference for the endoscopist. For colonoscopy, the worst part of the procedure was the preparation (78%) and for FS the preparation and the procedure ranked equally worst (30%). Conclusions: We have shown that colonoscopy with sedation is a very comfortable procedure. FS is more uncomfortable than colonoscopy; however, for the majority it is a tolerable experience. Women found FS only slightly more painful than men. The worst part of either procedure was the preparation. Embarrassment with either procedure was minimal. Both procedures are well tolerated and suitable for colorectal cancer screening.

Abstract The reliability of the currently used quantitative assay for commercially available chlorophyllin copper complex is discussed. It was shown that optical measurements at 405 nm can overestimate the purity of the preparation by 16.4–49.5%. This conclusion is from comparative studies using spectrophotometry at 405 nm and elemental analysis (copper content) of chlorophyllin copper complex. Spectrophotometry at 630 nm resulted in closer agreement with data calculated from elemental analysis. These observations have to be taken into consideration by those involved in the manufacture and distribution of chlorophyllin copper complex as well as in the development of the United States Pharmacopeia compendial monograph for this preparation.

The preparation and characterisation of food-grade chitosan from housefly larvae are reported. A refinement procedure was developed to remove larval mouth hooks from the primary chitosan product, which greatly improved the quality of the final product and simplified the production procedures. Different factors affecting chitosan preparation were studied and an orthogonal experiment was designed to determine optimal preparation conditions. When prepared under optimal reaction conditions, the end product was snow-white in colour, had a high deacetylation percentage, good viscosity, and a low ash content. The end product was characterised by Fourier transform infrared spectral analysis, X-ray diffraction analysis, thermo-gravimetric analysis, and differential scanning calorimetry. Its physical and chemical properties and sanitary index were determined and compared to the relevant Chinese standards. The results show that the chitosan we produced under optimal conditions meets the Chinese Fishery Trade Standard SC/T3403-2004 for food-grade chitosan.

AbstractThe adrenal gland is important for many physiological and pathophysiological processes, but studies are often restricted by limited availability of sample material. Improved methods for sample preparation are needed to facilitate analyses of multiple classes of adrenal metabolites and macromolecules in a single sample. A procedure was developed for preparation of chromaffin cells, mouse adrenals, and human chromaffin tumors that allows for multi-omics analyses of different metabolites and preservation of native proteins. To evaluate the new procedure, aliquots of samples were also prepared using conventional procedures. Metabolites were analyzed by liquid-chromatography with mass spectrometry or electrochemical detection. Metabolite contents of chromaffin cells and tissues analyzed with the new procedure were similar or even higher than with conventional methods. Catecholamine contents were comparable between both procedures. The TCA cycle metabolites, cis-aconitate, isocitate, and α-ketoglutarate were detected at higher concentrations in cells, while in tumor tissue only isocitrate and potentially fumarate were measured at higher contents. In contrast, in a broad untargeted metabolomics approach, a methanol-based preparation procedure of adrenals led to a 1.3-fold higher number of detected metabolites. The established procedure also allows for simultaneous investigation of adrenal hormones and related enzyme activities as well as proteins within a single sample. This novel multi-omics approach not only minimizes the amount of sample required and overcomes problems associated with tissue heterogeneity, but also provides a more complete picture of adrenal function and intra-adrenal interactions than previously possible.

Objective To perform a single-centre, detailed analysis of the preparations for the introduction of the first pressurized intraperitoneal aerosol chemotherapy (PIPAC) programme in the eastern part of Central Europe. Methods The study analysed the 14-month preparation period prior to the performance of the first PIPAC procedure with respect to: (i) general preparations; (ii) patient referral and qualification; (iii) the first PIPAC procedure; (iv) the 2 weeks following PIPAC programme establishment; and (v) general problematic issues that arose. Results The length of time needed to prepare our institution for the first PIPAC procedure was extremely long compared with other European Union PIPAC centres: 14 months versus a standard 3–6 months of preparation. The longest amount of time (12 months) was required to prepare the required paperwork. Conclusions A new PIPAC programme was successfully established in the eastern part of Central Europe. The length of time to implement this method was significantly longer because of lengthy bureaucratic processes. These current findings should help new centres, especially in this part of Europe, to establish a PIPAC programme more quickly.

The new prototype device is applied to the Solvent Front Position Extraction (SFPE) sample preparation procedure. The mobile phase is deposited onto the chromatographic plate adsorbent layer by the pipette, which is moved, according to programmed movement path, by a 3D printer mechanism. The application of the prototype device to SFPE procedure leads to the increased repeatability of the results and significant reduction of the analysis time in comparison to the classical procedure of chromatogram development. Additionally, the new equipment allows use procedures that are not possible to run using the classic chromatogram development. In this paper, the results of manual and semi-automatic sample preparation with SFPE are compared and the possible application of this prototype device is discussed.

For a quantum system interacting with its environment, the role of state preparation is nontrivial. The reason is that before the state preparation procedure, the system and the environment are correlated. Consequently, the state preparation procedure (which acts on the system) indirectly influences the state of the environment depending on the state preparation. In this paper, we use an experimentally realizable model describing a collection of N two-level atoms coupled to a common environment to investigate the influence of the state preparation procedure. We show that the dynamical map describing the evolution of the open quantum system can depend appreciably on the state preparation procedure. Moreover, this effect can be enhanced by increasing N. Our results should be useful for quantum control and quantum tomography.

Wound is a common problem following burn, trauma or infection. There are various methods to limit the infection and to cover the raw area. Before definitive cover, the wound bed must be ready to accept the reconstructive procedure. Various methods help in wound bed preparation. Recently hemoglobin spray has been advocated to hasten the wound bed preparation. In this article, we share our experience of using innovative hemoglobin spray in wound bed preparation.

We study preparation of states for open quantum mechanics. For non-Markovian systems that are initially correlated with the environment, the effects of the preparation procedure are nontrivial. This is due to the indirect effects on the state of the environment induced via the correlations with the system and the act of preparation on the system. We give three concrete examples of preparation procedure to elucidate our claims.
