Virus-induced gene silencing (VIGS) is an attractive tool for determining gene function in plants. The present study constitutes the first application of VIGS in S. pseudocapsicum, which has great ornamental and pharmaceutical value, using tobacco rattle virus (TRV) vectors. Two marker genes, PHYTOENE DESATURASE (PDS) and Mg-chelatase H subunit (ChlH), were used to test the VIGS system in S. pseudocapsicum. The photobleaching and yellow-leaf phenotypes of the silenced plants were shown to significantly correlate with the down-regulation of endogenous SpPDS and SpChlH, respectively (P ≤ 0.05).

During the summer of 1999, a sudden wilt of 60-day-old plants was observed in the Albenga region (Northern Italy), an area of intensive floriculture. Pathogenicity of the gram-negative plant pathogen Xanthomonas campestris pv. vesicatoria depends on a type III secretion system, which translocates bacterial effector proteins into the plant cell. In this study, we identified two novel type III effectors, XopE1 and XopE2 (Xanthomonas outer proteins), using the AvrBs3 effector domain as reporter. XopE1 and XopE2 belong to the HopX family and possess a conserved putative N-myristoylation motif that is also present in the effector XopJ from X. campestris pv. vesicatoria 85-10. In May 2005, the Plant Protection Service in the Netherlands received two tomato (Lycopersicon esculentum) plant specimens for diagnosis from a protected crop production facility of 2.5 ha near Kebili in Tunisia. Growth of the plants was reduced and leaves were chlorotic and brittle. Ripening of the fruits was delayed and their storage life was reduced from 3 weeks to 1 week. 1. The greenhouse experiment was performed to study the effect of gibberellic acid (GA3) on Jerusalem cherry. 2. Four levels of GA3 (zero, 100, 200, and 400 mg/lit) and three types of consumption (foliar, drip treatment, and foliar + drip treatment) were used in the experiment. 3. The results showed that the reciprocal effect of experimental factors on plant height, lateral shoot length, lateral shoot number, fruit number, root length, root wet weight, stem wet weight, chlorophyll a, chlorophyll b, total chlorophyll, pigment function, chlorophyll index, and resuscitate sugars were significant.
