At present, two viruses affecting kiwifruit (Actinidia spp.), Actinidia virus A (AcVA) and Actinidia virus B (AcVB), both belonging to the genus Vitivirus in the family Betaflexiviridae, have been reported from New Zealand (2). The infected trees showed leaf vein chlorosis, flecking, and ringspots. China is the largest commercial kiwifruit producer. During field investigations in the growing season of 2013, symptoms of leaf chlorosis or ringspots, similar to those caused by AcVA and AcVB (1), were observed on some kiwifruit (Actinidia chinensis) plants in Hubei Province in the central China. Leaf samples were collected from three symptomatic and two symptomless plants of two A. chinensis cultivars. Total nucleic acids were extracted from the samples using a CTAB-based protocol described by Li et al. (3) and used as template in RT-PCR for the detection of AcVA and AcVB. Each virus was detected using two sets of primers reported by Blouin et al. (1). Primer sets AcVA 1F/1R and AcVA5F/5R were used for the AcVA detection, and AcVB1F/1R and AcVB5F/Viti3'R were used for the AcVB detection. AcVA was detected in three symptomatic plants (ID: Ac-HN-1, Ac-HN-3, and Ac-HN-5), and AcVB was detected in two symptomatic plants (ID: Ac-HN-1 and Ac-HN-3) and in one symptomless plant (ID: Ac-HN-2). Neither virus was detected in the second symptomless plant (ID: Ac-HN-4). Samples Ac-HN-1 and Ac-HN-3 had mixed infection of AcVA and AcVB, and sample Ac-HN-2 had the latent infection of AcVB. The sequenced 283-bp RT-PCR amplicons of the replicase-encoding gene from AcVA isolates AC-HN-3 and AC-HN-5 using AcVA1F/1R shared 90.8% nucleotide (nt) identity with the corresponding sequence of the New Zealand AcVA isolate (GenBank Accession No. JN427014.1). The 269-bp fragments of the RNA-binding protein-encoding gene obtained by using AcVA5F/5R shared 85.5 to 85.9% nt identities with the corresponding sequence of JN427014.1. The AcVB5F/Viti3'R products of 365 to 369 bp from three AcVB isolates shared 85.5 to 88.6% nt identities with the corresponding sequence of the New Zealand AcVB isolate. The representative sequences were submitted to GenBank with accession numbers KJ696776 and KJ696777 for the 269-bp fragments of AcVA-HN-1 and AcVA-HN-3, and KJ696778 and KJ696779 for the 365-bp and 369-bp fragments of AcVB-HN-1 and AcVB-HN-2, respectively. In addition, 12 and 14 out of 42 kiwi samples (excluding HN-1 to HN-5) collected randomly were positive for AcVA and AcVB as detected by RT-PCR. Meanwhile, the sample affected by AcVA-HN-5 was subjected to deep sequencing of the small RNAs (sRNAs) for complete survey of the infecting viruses. De novo assembly of sRNAs generated four sequence contigs, with lengths ranging from 161 to 285 nt, matching to ORFs 1 to 3 of the genome of the New Zealand AcVA isolate with significant nucleotide (91 to 95%) and amino acid (80 to 94%) similarities, and some other contigs from a new virus (unpublished). The result further confirmed AcVA infection in the kiwi plant. To our knowledge, this is the first report of both AcVA and AcVB outside of New Zealand. The Chinese isolates of the two viruses are distinct from those reported from New Zealand. Kiwifruit (Actinidia spp.) is an economically substantial fruit crop with China the main producer. China is the primary source of wild kiwifruit and the largest producer of kiwifruit in terms of both production and planting area, and Shaanxi province is the largest kiwifruit producer in China. Previous studies reported presence of kiwifruit viruses in Actinidia chinensis. In this study, six viruses were identified in kiwifruit ‘Xuxiang’ (A. deliciosa) in Shaanxi, China. The incidence, distribution, and genetic diversity of these viruses were studied. The results showed that Actinidia virus A (AcVA), Actinidia virus B (AcVB), Actinidia chlorotic ringspot-associated virus (AcCRaV), cucumber mosaic virus (CMV), apple stem grooving virus (ASGV), and potato virus X (PVX) were the main viruses infecting Xuxiang kiwifruit in Shaanxi, China. Incidence of the various viruses with both single and multiple infection varied with different kiwifruit-growing counties. For single virus infection, the highest and the lowest numbers of samples infected were about 22 for AcCRaV and 0 for AcVB in Meixian out of 170 samples, 12 for AcVA and 0 for CMV in Zhouzhi out of 120 samples, 10 for AcVA and 0 for AcVB, AcCRaV, ASGV, PVX, and CMV in Yangling out of 70 samples, and 8 for AcCRaV and CMV and 0 for AcVA, AcVB, ASGV, and PVX in Hanzhong out of 80 samples, respectively. Samples which were multiply infected with two or more viruses were also detected. Analysis of the phylogenetic tree of these viruses showed some genetic variability in the AcVA, AcVB, and AcCRaV isolates of Shaanxi kiwifruit. There was no obvious molecular variation in the coat protein genes of ASGV, CMV, and PVX virus isolates from Shaanxi kiwifruit. The present study is the first large-scale survey of kiwifruit viruses in Shaanxi, China. To our knowledge, this is the first report of PVX infecting kiwifruit and the first report of molecular variability of AcVA, AcVB, and AcCRaV. These results provide important data for studying the genetic evolution of AcVA, AcVB, AcCRaV, ASGV, CMV, and PVX.

Agrobacterium tumefaciens Mediated Genetic Transformation of acvB Gene in Potato (Solanum tuberosum L.). Genetic transformations are now routinely applied to plant mediated by Agrobacterium tumefaciens as the most convenient technique. This study aimed to prove the success of A. tumefaciens mediated genetic transformation in potato. A. tumefaciens LBA (pBI 121) and explant of potato shoot were used in this study. Explants were grown in vitro on Murashige and Skoog media. Transformation was implemented using smear technique by smearing A. tumefaciens to injured explant. Experimental groups consisted of two groups: control group which did not receive transformation treatment and treatment group receiving transformation treatment. Explant growth was observed through the presence of shoots, branches and the shoot height. Explants in the treatment group resulted in a higher number of shoots, branches, and shoot heights compared to control. Phenol compounds appear in explant epidermal tissue, indicating the wounds produced by A. tumefaciens infection, thus the gene predicted to be transformed. Identification by PCR is needed to prove the existence of the acvB gene in potato plants genome, using acvB specific PCR primer as the marker, such as (5?-CCCT CTAG AGAC CCGC GCCA AGGCG-3?) and (5?CGCG TCGA CCTT GTCG GAAAG -3?) with 540-bp in base pair size produced.

From July 1984 to September 1987, 737 patients with aggressive malignant lymphoma (ML) were treated by an intensive regimen (LNH-84) comprising three or four courses of doxorubicin, 75 mg/m2; cyclophosphamide, 1,200 mg/m2; vindesine, 2 mg/m2 x 2; bleomycin, 10 mg x 2; and prednisolone, 60 mg/m2 x 5 (ACVB), consolidation with high-dose methotrexate, ifosfamide, etoposide, asparaginase, and cytarabine, and a randomized late intensification with two courses of cytarabine, cyclophosphamide, teniposide, bleomycin, and prednisone (AraCVmB). Four hundred forty-two patients had intermediate-grade ML, 221 highgrade ML, and 74 unclassified ML. Most of the patients had advanced disease: stage IIE (23%), III (13%), or IV (47%); 38% disseminated nodes; 38% two or more extranodal sites; and 41% a tumoral mass greater than 10 cm. Five hundred fifty-three patients (75%) went into complete remission (CR), 63 (9%) into partial remission, 62 (8%) failed to respond, and 59 (8%) died during ACVB courses, 17 of them from progression of the disease. With a median follow-up of 23 months, the estimated 2-year overall survival time to failure (TTF), and time to relapse (TTR) survival are 67%, 56%, and 67%, respectively. Patients receiving a late intensification had the same relapse rate as the other patients. A persistent fibronecrotic mass was found in 150 patients (20%) and did not influence the relapse rate. Toxicity was mainly neutropenia and infection during the ACVB courses, with 40 patients (5%) dying from septic complications while responding to treatment. Fifty-three percent of the patients had a neutropenia less than 0.500 x 10(9)/L, 58% fever (6% grade 4), and 49% a documented infection (8% grade 4). These results obtained with the LNH-84 regimen demonstrate that this therapeutic scheme is an effective treatment for aggressive ML.

Rhizobium tropici CIAT899 is highly acid tolerant and a good competitor for Phaseolus vulgaris nodule occupancy at low pH values. Using Tn5 mutagenesis, we identified an operon required for acid tolerance and nodulation competitiveness. The insertion was mapped to the 5′ end of atvA, encoding a product with high sequence identity to the agrobacterial AcvB virulence protein. Complementation analyses indicated that atvA is an ortholog of acvB, both genes being required for acid tolerance. A Ser/Ala substitution in the LIPASE_SER motif of AtvA resulted in an acid sensitive Fix+ but very poorly competing strain, demonstrating that Ser-313 is essential for AtvA function. atvA is the second gene in an operon that is transcriptionally upregulated by acid shock. The acid-responsive promoter was mapped to a 469-bp intergenic region located upstream of lpiA, the first gene in the operon. lpiA-like genes are found in several α, β, and γ Proteobacteria that interact with eukaryotic host cells, and they are predicted to encode membrane proteins related to the FmtC/MprF family from low G+C Firmicutes. The latter proteins are involved in resistance to cationic antimicrobial peptides. A nonpolar deletion in lpiA caused a sevenfold decrease in relative nodulation competitiveness.

Com a crise energética que o Brasil vem enfrentando e a pressão internacional para a diminuição das emissões de CO2, nota-se o aumento, nos últimos anos, de pesquisas relacionadas à produção de edificações considerando estes dois aspectos ambientais. Na produção das vedações verticais de edificações habitacionais, algumas tecnologias têm sido utilizadas, entre elas o light steel framing (LSF), as quais precisam atender requisitos e critérios de desempenho do usuário, relacionadas à segurança, habitabilidade e sustentabilidade ambiental. Uma forma de medir o desempenho ambiental de um produto ou processo é a avaliação do ciclo de vida (ACV), que está sendo aplicada nas edificações. Muitos trabalhos de ACV tem tido como foco questões relacionadas ao consumo de energia e emissões de CO2 ao longo do seu ciclo de vida, o que resultou no surgimento dos termos Avaliação do Ciclo de Vida Energético (ACVE) e Avaliação do Ciclo de Vida de Emissões de CO2 (ACVCO2). Neste contexto, o presente trabalho teve como objetivo a realização da ACVE e ACVCO2 de uma edificação habitacional unifamiliar (EHU) localizada em Brasília – DF. Foram comparados dois diferentes sistemas de vedação vertical (externa e interna): light steel framing (LSF) e blocos cerâmicos de vedação convencional (VC), considerando o ciclo de vida completo da EHU, também chamado de berço ao túmulo, a partir das etapas de pré-uso, uso e pós-uso da edificação. A metodologia utilizada se baseou em estudos nacionais e internacionais, realizados por meio da pesquisa bibliográfica e da aplicação a um estudo de caso. Foram utilizados diferentes dados de inventário, obtidos da literatura, para valores mínimos, médios e máximos. As equações utilizadas foram aplicadas em planilhas eletrônicas e foi utilizado o software de simulação termoenergética, DesignBuilder, para obtenção do consumo de energia e emissões de CO2 na etapa operacional da edificação em estudo, avaliando a relação do desempenho térmico dos sistemas de vedação vertical. O sistema de VC apresentou melhor desempenho térmico quando comparado ao sistema de LSF, no entanto, esta diferença não resultou significativa no consumo de energia e emissões de CO2. A EHU de VC apresentou maior consumo de energia total quando comparada a EHU de LSF, para as situações de valores mínimos e médios. Em relação às emissões totais de CO2, a habitação de VC apresentou maior valor para as três situações de dados. A etapa de uso se mostrou a mais impactante ao longo do ciclo de vida das habitações. Com participação da fase operacional variando de 65 a 75% para energia e 58 a 70% para emissões de CO2, e de manutenção com 15 a 20% para energia e 11 a 18% para emissões, seguida da etapa de pré-uso, variando de 8 a 17% para energia e de 14 a 28% para as emissões. A etapa de pós-uso apresentou participação inferior a 1% tanto para energia como também para as emissões de carbono. Desta forma, foi possível concluir que além do aspecto do consumo de energia é interessante avaliar questões das emissões de CO2. Por fim, destaca-se que para a realidade de Brasília a partir dos dados adotados neste trabalho o LSF foi o sistema de vedação vertical mais vantajoso do ponto de vista energético e de emissões de carbono. ________________________________________________________________________________________________ ABSTRACT
With the energy crisis that Brazil is facing and international pressure to reduce CO2 emissions, there is an increase in recent years in research related to the production of buildings considering these two environmental aspects. In the production of wall system of residential buildings, some technologies have been used, like the light steel framing (LSF), which must meet user requirements and performance criteria related to safety, livability and environmental sustainability. One way to measure the environmental performance of a product or process is the life cycle assessment (LCA), wich is being applied in buildings. Many LCA studies has been focused on issues related to energy consumption and CO2 emissions throughout its life cycle, resulting in the emergence of terms Life Cycle Energy Assessment (LCEA) and Life Cycle CO2 Emissions Assessment (LCCO2A). In this context, this study aimed to study the application of LCEA and LCCO2A of two single-family residential building located in Brasília - DF. It was compared different wall systems (external and internal): light steel framing (LSF) and brick masonry (BM), considering the full life cycle of the residential buildings, also called cradle to grave, including pre-use, use and post-use phases of the buildings. The methodology used was based on national and international studies, conducted through literature review and application to a case study. Different inventory data were used, obtained from the literature, for minimum, medium and maximum values. The equations used were applied in spreadsheets and was used a thermo-energetic simulation software, DesignBuilder to study the relation between the thermal performance of the wall systems and the energy and CO2 emissions due to the air conditioning. The BM system showed better thermal performance compared to the LSF system, however, this difference did not result in significant energy consumption and CO2 emissions in operational phase. The BM building had higher total energy consumption when compared to LSF building to situations of minimum and medium values. Related to the total CO2 emissions, BM building showed a higher value for the three situations. The use phase showed the most impressive throughout the life cycle of buildings, with contribution from operational stage ranging from 65 to 75% for energy, 58 to 70% for CO2 emissions, maintained between 15 and 20% for energy and 11 to 18% for carbon emissions, followed by the pre-use phase varying 8-17% for energy and 14-28% of CO2 emissions. The post-use stage had participation below 1% for energy and CO2 emissions. Thus, it was concluded that in addition to the aspect of energy consumption is interesting to evaluate issues of CO2 emissions. Finally, to the reality of Brasilia, from the data adopted in this study, the LSF was the more advantageous vertical system from the energetic and carbon point of view.

Resumen
Multidrug efflux pumps are membrane proteins that actively transport foreign objects out of cells. The active efflux of these pumps is a critical self-defense mechanism that enables the survival of bacteria under hostile environments. Efflux pump AcrB is a member of the Resistance-Nodulation-Division (RND) super family. In E. coli, it associates with periplasmic protein AcrA and outer membrane channel TolC to extrude a variety of noxious compounds out of cell from both the cytoplasm and the periplasm. My dissertation research focused on two aspects of this multidrug efflux pump: the oligomerization process during the biogenesis of AcrB and its degradation. Oligomerization is an important aspect of the structure and function for many proteins and has been the subject of many studies. However, most of such studies focused on soluble proteins. The oligomerization process of membrane proteins, including AcrB, is rarely explored. In chapter 2, the co-assembly of AcrB variants co-expressed in the same cell was used as a tool to investigate the assembly of AcrB trimers during its bio-production. By monitoring the portion of pure trimers containing only one type of subunit and hybrid trimers containing a mixture of the two kinds of subunits, it was found that the oligomerization of membrane proteins is not a random process as the formation of pure trimer is favored. In chapter 3, the GALLEX system was used to monitor AcrB oligomerization in cells under the native condition. Previously GALLEX has only been used to monitor the oligomerization of small transmembrane proteins. By constructing a series of fusion proteins with different linker length between LexA and AcrB, and optimizing inducer concentration, we finally developed a system that could be used to differentiate AcrB trimers of different oligomerization affinities. While chapters 2 and 3 focus on the trimerization of AcrB, a critical step of its biogenesis, chapters 4 and 5 focus on its life time and degradation. In chapter 4, the life time of AcrB was measured by incorporating non-natural amino acid azidohomoalanine (AHA) into protein translation. Using this method, it was determined that that the half-life of both AcrA and AcrB in E. coli were six days. The surprisingly long lifetime of these detoxification proteins might represent a strategy by the bacteria to conserve energy and maximize their competition niche for survival in a hostile environment. In chapter 5, the degradation process of ssra tagged AcrB was investigated. In-vivo degradation test showed that properly inserted AcrB can be digested after addition of ssra-tag to its C-terminus. It was found that cytoplasmic unfoldase-protease complex ClpXP and chaperone SspB are involved in the degradation. In vitro assay is still being optimized to quantitatively analyze the activity of ClpXP in the degradation of AcrB.

Herzinfarkt und Tod stellen häufige Folgen der koronaren Herzerkrankung dar, die durch eine rechtzeitige aortokoronare Bypassoperation vermieden werden können. Im Gegensatz zur klassischen offenen Operation bieten minimal invasive Verfahren entscheidende Vorteile. Die erschwerte Orientierung stellt jedoch eine große Herausforderung in der minimal invasiven Herzchirurgie dar, insbesondere bei der Verwendung telemanipulatorischer Systeme, wie bei der total endoskopischen Bypassoperation (TECAB). Die Entwicklung des „Surgical Cartographic Navigation System“ (SCNS) verspricht eine deutliche Verbesserung der Orientierung des Chirurgen mithilfe der Nutzung modernster Techniken der Augmentierten Realität. Hierbei wird auf der Basis von CT-Datensätzen ein virtuelles Herzmodell geschaffen, welches als Grundlage der assistierten Navigation dient. Im Speziellen wird bei der durch das SCNS unterstützten TECAB Operation die aufzufindende Koronararterie in das Sichtfeld des Endoskopes projiziert („augmentiert“). Ziel dieses Dissertationsvorhabens war die Evaluation der klinischen Anwendbarkeit des SCNS während einer Simulation einer Inzision mit dem da Vinci™-System auf fünf individuell angefertigten Herzphantomen. Es sollte überprüft werden, ob der Chirurg mit Hilfe der Unterstützung durch die SCNS Sicht mit eingeblendeter Koronararterie einen direkten Kontakt zur LAD (Left Anterior Descending), der häufigsten Zielarterie der TECAB, herstellen kann. In einem Studienkollektiv, bestehend aus zehn medizinisch unerfahrenen Personen und zehn Herzchirurgen, wurde die Treffergenauigkeit der SCNS-gestützten Auffindung der LAD in insgesamt 300 Testversuchen überprüft. Insgesamt konnte die Arterie in 58 % der Fälle korrekt identifiziert werden. Dabei lag kein signifikanter Unterschied zwischen den beiden Gruppen vor. Hiermit konnte die klinische Anwendbarkeit des SCNS für die TECAB erstmals gezeigt werden. Des Weiteren wurden zwei Faktoren als vordringliche Ziele für zukünftige Fortentwicklungen identifiziert: Die Optimierung von Landmarken für die Registrierung des Herzens sowie die verbesserte optische Darstellung der Augmentierten Realität im Endoskopiesichtfeld. Zusammengefasst konnte in diesem Dissertationsvorhaben in einer umfassenden Studie gezeigt werden, dass das SCNS einen erfolgversprechenden Lösungsansatz für die Behebung kritischer Orientierungsprobleme der minimal invasiven Herzchirurgie bei der TECAB bietet. Die hier gewonnenen Erkenntnisse stellen die Grundlage für weitergehende Studien zur Fortentwicklung des SCNS dar, die vor einem klinischen Ersteinsatz erfolgen müssen
In the therapy of coronary heart disease minimally invasive and endoscopic methods offer considerable benefits to the patient, while for the surgeon difficult orientation and missing haptic feedback are still the leading problems in Endoscopic Bypass Grafting with telemanipulative systems. To support the surgeon with improved vision, a three dimensional model of the coronary artery tree based on CT scans is integrated into the view of the endoscope. The “Surgical Cartographic Navigation System” (SCNS) is a tool which provides this feature called Augmented Reality (AR). Aim of this study was the first technical analysis of the SCNS during a simulation of an incision with the da Vinci™ surgical system on an electronic heart phantom. The hypotheses was that with the guidance of the SCNS augmented reality view, the surgeon can perform a direct contact to the Left Anterior Descending Coronary Artery (LAD). Five anatomically correct heart phantoms were created using the rapid prototyping technology. The heart models were covered with an electrical conducting layer for the detection of the contact with the coronary artery or with the surrounding tissue. A 3D model of the coronary artery tree based on a CT scan was registered to the heart phantom and overlaid into the video screen of the da Vinci™ robot master console. Ten inexperienced medical students and ten experienced heart surgeons used the SCNS in a surgery simulation with the goal of finding the LAD artery and contacting the LAD with robot instruments. In 300 test runs 58 % of both groups hit the LAD correctly. The overlaid information created with the SCNS enables the surgeon to correctly identify the coronary artery. The clinical applicability of the SCNS for the TECAB Operation is hereby demonstrated. Resumen
The Marine Corps Systems Command is evaluating alternative propulsion systems to achieve high water speed for the future USMC Amphibious Combat Vehicle (ACV). A gas turbine engine is one of the propulsion systems evaluated. The primary risk of operating a gas turbine engine in the ACV is power loss due to the ingestion of marine contaminants such as saltwater mist in the air, saltwater spray generated from the vehicle operation, green water wash caused by the operation of the vehicle in the surf zone or in rough seas, and hard particles such as sand present in the marine environment. The Auxiliary Ships and New Acquisition Support Branch (Code 425) of the Naval Surface Warfare Center, Philadelphia Division conducted a study to assist the Marine Corps Systems Command in assessing the feasibility of using a gas turbine engine as a propulsion system on future USMC ACVs. The study was focused on developing and testing a gas turbine intake solution for the ACV that can remove saltwater from the intake airstream of the notional 3,000 horsepower ACV engine. Code 425 developed a two-part solution for the intake of the ACV. The first part of the solution is the Combustion Air Protection Shroud (CAPS) located at the entrance of the engine intake and designed to protect the ACV engine from green water wash by elevating the intake above the ACV deck. The second part of the solution is a gas turbine intake filtration system located downstream of the intake shroud and designed to remove marine contaminants that enter the intake shroud. A reduced-scale model of the CAPS was designed by Code 425 in conjunction with Gibbs & Cox and tested at the Davidson Laboratory High Speed Test Basin at the Stevens Institute of Technology to determine the optimum extension height of the CAPS to protect the engine intake. This paper covers the design and testing of the CAPS. The results showed that a 2.67 ft. tall CAPS with selectively closeable air intake louvers is sufficient to keep out saltwater from the ACV gas turbine engine.

The goal of this research is to numerically model flexible finger seals of an air cushion vehicle (ACV) to more accurately predict resistance for these craft. Advancements in computational tools for fluid-structure interactions have set the stage for this effort. Prior work for this application has focused on planer type seals and not the finger type seals typical of modern ACVs. While this research effort remains underway and limited results are available, it is the authors’ desire to propose a pathway forward. Following the motivation and a brief review of prior work, verification of Abaqus/Explicit’s fluid solver is conducted using the analytical solution to Couette flow. Next, Abaqus/Standard (implicit scheme) is used to verify the finite element options for the structural portion. Various hyper elastic material models are also compared. Lastly, a FSI validation problem is conducted using Abaqus/Explicit for both linear elastic and hyper elastic materials.

Resumen
Sentence similarity modeling lies at the core of many natural language processing applications, and thus has received much attention. Owing to the success of word embeddings, recently, popular neural network methods have achieved sentence embedding, obtaining attractive performance. Nevertheless, most of them focused on learning semantic information and modeling it as a continuous vector, while the syntactic information of sentences has not been fully exploited. On the other hand, prior works have shown the benefits of structured trees that include syntactic information, while few methods in this branch utilized the advantages of word embeddings and another powerful technique ? attention weight mechanism. This paper makes the first attempt to absorb their advantages by merging these techniques in a unified structure, dubbed as ACV-tree. Meanwhile, this paper develops a new tree kernel, known as ACVT kernel, that is tailored for sentence similarity measure based on the proposed structure. The experimental results, based on 19 widely-used datasets, demonstrate that our model is effective and competitive, compared against state-of-the-art models.
