Resumen
Produksi etanol (bioetanol) dari bahan baku biomassa merupakan salah satu cara mengurangi polusi lingkungan dan konsumsi terhadap minyak mentah. Salah satu bahan bakar yang dapat diperbaharui yakni bioethanol yang merupakan produk turunan dari singkong dan tetes tebu dengan menggunakan teknik proses multiple feedstock system. Industri bioethanol tidak hanya memproduksi produk utama berupa etanol, tetapi juga menghasilkan air limbah dalam jumlah besar. Air limbah bioethanol berbahan baku singkong disebut dengan thinslop, sedangkan air limbah bioethanol berbahan baku tetes tebu disebut dengan vinasse. Adanya proses perubahan substrat dari thinslop ke vinasse akan menyebabkan perubahan beban mikroorganisme dalam penguraian bahan organik . Tujuan dari penelitian yakni untuk mengukur potensi produksi biogas dari proses rekayasa aklimatisasi perubahan substrat. Penelitian menggunakan tiga bioreaktor CSTR dengan kapasitas 50 L. Perlakuan setiap bioreaktor yakni 50% thinslop dan 50% vinasse; 25% thinslop dan 75% Vinasse, dan 100% Vinasse, dengan laju pembebanan berbeda setiap minggu (0,5 g / L hari, 1,0 g / L hari, 1,5 g / L hari, dan 2,0 g / L hari). Hasil penelitian menunjukkan bahwa nilai potensial biogas dari bioreaktor pertama (50% thinslop: 50% vinasse), bioreaktor kedua (25% thinslop: 75% vinasse), dan bioreaktor ketiga (100% vinasse) berturut-turut yakni 39,39 L/hari, 38,97 L/hari 27,23 L/hari. Hasil penelitian juga menunjukkan bahwa kandungan gas metana secara berturut-turut dari bioreaktor pertama, bioreaktor kedua, dan bioreaktor ketiga (100% vinasse) sekitar 32,67-52,81%, 40,29-67,29%, 20,46-57,33%. Temperatur merupakan salah satu faktor yangmempengaruhi proses fermentasi pada bioreaktor. Dimanaproses fermentasi dipengaruhi oleh perubahan suhu.Pengendalian suhu berguna untuk menjaga suhu pada mediumsaat terjadi proses pengadukan. Sebab jika suhu pada mediummengalami perubahan suhu yang tidak beraturan, maka hasildari medium menjadi tidak maksimal. Oleh karena itudibutuhkan teknik kontrol otomatis untuk mengatur suhu padatabung bioreaktor. Pengujian dilakukan dengan memberigangguan pada tabung bioreaktor berupa air dingin yangdimasukkan pada tabung bioreaktor. Dengan turunnya suhumaka kontroler akan bekerja untuk mencapai suhu yangdiinginkan sesuai dengan setpoint yang diberikan. Kontrol on–off digunakan untuk mengontrol suhu pada bioreaktor denganmenggunakan sensor PT100 sebagai sensor suhu dan komporlistrik sebagai aktuatornya. Semua kontrol akan diproses padaArduino Mega 2560, hasil pengujian menunjukkan bahwaproses pengontrol suhu berhasil menjaga suhu tabungbioreaktor pada suhu 400C dari temperatur 250C dalam waktu840 detik dan didapatkan akurasi pengontrolan 98,9% denganerror 1,1%.

Resumen
The purpose of this project is to evaluate the potential of a bioreactor system to degrade DDT based upon co-metabolism of indigenous bacteria. The study was performed with soil samples spiked with four different concentrations of DDT. The prepared sludge was circulated at a steady rate of revolution per minute in bioreactors with added M8 solution, cabbage leaf extract and molasses. The experiment was carried out for 7 days and chemical analysis and toxicity testing was accomplished at the beginning and the end of the experiment. The chemical analysis was essential to support the conclusions of the ecotoxicology tests. Ecotoxicology test was performed for the assessment of the toxicity (in terms of bioavailable measures) of the sludge samples, and was carried out with the Ostracodtoxkit sediment toxicity test, with the freshwater benthic crustacean test species Heterocypris incongruens. As part of the project the potential of the bioremediation method phytoremediation have been studied. Brassica Juncea seeds have been cultivated in the soil spiked with four different concentrations of DDT for one month, under stable circumstances. Growth of the plants was measured at the end of the experiment, and a chemical analysis was carried out. A thorough literature review was carried out for both the bioreactor and the phytoremediation experiments in order to obtain information about methods and theoretical background. The ecotoxicology tests and the chemical analysis showed increased p,p’- DDT concentrations in the bioreactors I. and II. at the end of the 7 day experiment, the reasons of which are not known, and require further studies.

Resumen
This thesis focuses on the study of the influence of selected parameters on the course of microbial cultivation and evaluation of bioprocess. It is divided into two parts. The first part deals with the production of mutant forms of the protein cryptogein yeast Pichia pastoris. The theoretical part summarizes the findings of the yeast P. pastoris and its expression. It also deals with cryptogein that induces defense reactions in plants. In the experimental part was produced mutant cryptogein X24, in which the concentration of each fraction and the ability to transfer sterols. The second part of this thesis is focused on aerobic and anaerobic oxidation of elemental sulfur by Acidithiobacillus ferrooxidans. In the theoretical section, our knowledge on A. ferrooxidans, its metabolism and the importance of ATP in cell metabolism was summarized. In the experimental part, the above bioprocess was monitored using pH, biomass concentration, the rate of oxidation of elemental sulfur the cellular ATP content.

Resumen
Nitrogen and phosphorus are involved in many processes on the Planet Earth. Especially in agricultural areas water is contaminated by nutrients, which can cause the eutrophication of surface waters, and other problems. The solution could be use of denitrifying bioreactors, which are used for the reduction of high nitrate concentrations in shallow groundwaters. The subject of the thesis was the study of phosphorus removal in the denitrification bioreactor by steel turnings, which are constituent part of the organic load of the bioreactor. Steel turnings release Fe, which causes the precipitation and adsorption of P. Eight bioreactors were filled with poplar woodchips. To these columns just above the surface were added model water enriched with nitrate nitrogen, phosphate phosphate was added to 4 columns, where two of them were enhanced by the addition of steel turnings upstream of the wood medium. Sampling and the analyses of the samples were determined weekly, determination of the phosphorus, iron and other substances necessary for the detection of processes in the bioreactor was performed. The dependence of phosphorus removal on the bioreactor operating conditions was evaluated based on measured data, and the effect of iron on the biological denitrification process was also assessed. Steel turnings have been found to be effective in removing TP, but it is necessary to solve iron leaching in the future. The concentration of phosphorus was reduced up to 0 mg/l on the effluent from the denitrification bioreactors, efficiency of phosphorus removal reached 100 %. The presence of steel chips had no effect on denitrification speed. The denitrification process was also successful in the phosphorus removal columns. From the point of view of leaching of substances and iron, the removal of N and P seems to be preferable in dry period during stoppage with no water fillings. Shutdown of bioreactors with flooded filling caused high concentrations of leached iron up to 149 mg/l

Resumen
Control of cultivation conditions in the~live cell imaging extends the possibilities of biological experiments and makes the experimental results more reliable. In order to change the~cultivation conditions in a controlled manner and increase the reproducibility of the experiments, it is necessary to reduce the amount of manual operations and replace them with automated procedures. Therefore, the concept of a new automated culture device (bioreactor) was created. This device controls the exchange of medium in the observation chamber, ensures the circulation and exchange of the atmosphere and controls its composition. The bioreactor is intended for use in the Laboratory of Experimental Biophotonics. This laboratory is equipped with coherence-controlled holographic microscope (CCHM), which uses quantitative phase imaging (QPI) method. Thus, the bioreactor is adapted to the current requirements of this laboratory and optical elements of the bioreactor meet the requirements of the QPI method. This text specifies the cultivation conditions of the living cells and summarizes, how the conditions could be controlled in the live cell microscopy. Next some commercially available culture devices are described and assessed, whether they are convenient for the~use in Laboratory of Experimental Biophotonics. The crucial part of the thesis is the~design, construction and testing of the new bioreactor.

Resumen
This diploma thesis deals with the bioreactor system and its use for the production of heat and biogas. The bioreactor uses the composting and metanogation process of fermentation that humanity has known for hundreds of years, but the combination of these processes is a relatively unexplored area. The theoretical part of the thesis analyzes the existing possibilities of utilization of the heat generated in the compost, and also describes the used technology of small biogas stations and other small systems for biogas production. It follows the practical part of the work, when it was the task to design a bioreactor working at low temperatures during the winter. Part of the solution is the initial design and testing of the basic test structure of the bioreactor, followed by the implementation of a more advanced and more complex system, including a control and measuring center realized by a microcontroller.
