Caudal Regression Syndrome (CRS) is a rare malformation syndrome associating to varying degrees agenesis of the coccygeal or lumbosacral vertebrae. This vertebral anomaly can therefore be reduced clinically to a simple coccygeal agenesis without any deficit or even lumbosacral agenesis accompanying a clinical picture with sphincter disorders associated or not with transit disorders and various deficits involving the lower limbs. This syndrome is accompanied by other orthopedic malformations such as shortening of the lower limbs, and / or gastrointestinal abnormalities, see also genitourinary as well as cardiovascular. Its incidence is 1 to 5 cases per 100,000 births. Its precise cause has not yet been identified, but its relationship to maternal diabetes is well established. We report in our study 5 patients with CRS from different clinics whose sphincter disorders were found in the foreground in all our patients and whose results vary according to the inaugural clinical picture. MRI made it possible to refine and confirm the diagnosis highlighting the congenital anomaly and the associated lesions. We have obtained 75% good results and 25% clinical stabilization; nor do we deplore any case of aggravation or death. The interest is to suspect the diagnosis of CRS; document it at the start of the prenatal period and determine its severity and associated abnormalities in order to present options for patient management; because once the diagnosis is made, surgical treatment becomes imperative due to the formidable neurological sequelae compromising the functional prognosis.

During embryogenesis, hematopoietic cells arise from the lateral plate mesoderm (LPM) following gastrulation. The transcriptional program required for this LPM to blood switch is not fully understood. Previous work on a zebrafish mutant with a deletion in the cdx4 gene demonstrated the importance of this caudal transcription factor in the LPM to blood transition. To explain how cdx4 regulates embryonic hematopoiesis, two main approaches were taken in this thesis. The first part of the thesis describes a chemical genetics screen that identified cdx4 interacting pathways. To find small molecules that could rescue the loss of red blood cells caused by the cdx4 deletion, cdx4 mutant embryos were incubated with 2640 compounds from the beginning of the gastrula stage to the 10-somite stage. Two related psoralen compounds, Bergapten (Ber) and 8-methoxypsoralen (8-MOP), rescued the erythroid progenitors in the cdx4 mutants. This rescue is closely linked to the compounds' effects on anteriorposterior patterning, reminiscent of retinoic acid pathway compounds. The second part of my thesis identifies a Cdx4-Sall4 transcriptional module in the LPM. Chromatin-immunoprecipitation coupled to sequencing (ChIP-seq) and microarray analysis revealed that Cdx4 directly regulates cdx4 and a zinc finger transcription factor spalt-like 4 (sall4) transcription. Sall4 ChIP-seq showed that Sall4 also binds to its own locus and to the cdx4 locus, suggesting an auto- and cross-regulation between two transcription factors. In addition, Cdx4 and Sall4 bind to common genomic regions proximal to mesodermal progenitor (tbx16 and mespa) and hematopoietic genes (scl, gata2a, and ldb1a), indicating Cdx4 and Sall4 co-regulate key genes that are required for LPM and blood specification. sall4 knockdown in the cdx4 mutants demonstrated that Sall4 synergizes with Cdx4 in regulating embryonic hematopoiesis. These findings suggest that auto- and cross-regulation of Cdx4 and Sall4 establish a stable circuit in the LPM that facilitates the activation of blood-specific program as development proceeds. How undifferentiated germ layers transition into various tissues is a key question in developmental biology. My thesis establishes a model based on LPM to blood transition, which is also applicable to other studies on germ layer specification.

_____________________________________________________________________________________ ABSTRACT
In this work we study the estimator proposed by Fan (2004) for the tail index of distributions in the domain of attraction of a _-stable law with 0 < _ < 2. This estimator has U-statistic structure and is robust and asymptotically unbiased. By using the classical tools theory of U-statistic, the consistency and the asymptotic normality of the estimator are proved.

Resumen
Congenital limb malformations rank behind congenital heart disease as the most common birth defects observed in infants. The term «sirenomelia» («mermaid syndrome») is derived from the physical similarity of the affected fetus to mythical creatures mermaids - charming women with the lower part of the body in the form of a fish tail, where there is a fusion of the lower extremities and partial or complete fusion of the feet.

Resumen
The caudal fin of zebrafish (danio rerio) is often used to study regeneration thanks to its extraordinary regenerative ability, easy access, and relative simplicity in structure. Branching morphogenesis is observed in many organs, including lungs and salivary glands in mammals, as well as the fin rays in zebrafish and is thought to follow unifying principles. An important developmental gene, sonic hedgehog a (shha), has been shown in other studies to play an essential role in the branch formation. Previous studies in our lab have shown that the transient depletion of the shha-expressing cells following laser ablation of the shha-expressing cells in the regenerating caudal fin results in a delay of fin rays branch formation. In order to study the long-term effect of ablating the shha-expressing cells, I generated a new zebrafish transgenic line (Tg)(2.4shha:CFP-NTR-ABC) to perform a conditional cell ablation using the Metronidazole/Nitroreductase (Mtz/NTR) system. Preliminary data suggest that cell ablation using the Mtz/NTR system is successful in the Tg(2.4shha:CFP-NTR-ABC) embryos. In addition, short-term ablation of the shha-expressing cells through Mtz/NTR system delays branch formation during caudal fin regeneration of the Tg(2.4shha:CFP-NTR-ABC) adult fish. Further work will involve the analysis of the effects of the long-term ablation of the shha-expressing cells and the involvement of other signaling pathways in the ray branching formation during zebrafish caudal fin regeneration. This study can provide insights into understanding of the molecular mechanisms underlying branching morphogenesis in various organs. During the course of the above project, I have observed an organ-wide response to local injury in the zebrafish caudal fin. In this study, I have shown, for the first time, an immediate organ-wide response to partial fin amputation characterized by the damage of blood vessels, nerve fibers and the activation of inflammatory response in the non-amputated tissues. I established that the adult zebrafish caudal fin serves as an excellent model for the study of the organ-wide response to local injury, and such study may provide new insights into the field of regenerative medicine in which stimulating regeneration locally may trigger responses in unintended locations.

In this study, we investigate the modeling framework for a robotic fish actuated by a flexible caudal fin, which is filled with electrorheological (ER) fluid and thus enables tunable stiffness. This feature can be used in optimizing the robotic fish speed or maneuverability in different operating regimes. The robotic fish is assumed to be anchored and the flexible tail undergoes undulation activated by a servomotor at the base. Lighthill’s large-amplitude elongated-body theory is used to calculate the hydrodynamic force on the caudal fin, and Hamilton’s principle is used to derive the dynamic equations of motion of the caudal fin. The dynamic equations are then discritized using the finite element method, to obtain an approximate numerical solution. In particular, simulation is conducted to understand the influence of the applied electric field on the stiffness and thrust performance of the caudal fin.

Resumen
Three-dimensional numerical simulations are used to investigate the hydrodynamic performance and the wake patterns of a sunfish in steady swimming. Immersed boundary method for deformable attaching bodies (IBM-DAB) are used to handle complex moving boundaries of one solid body (fish body) attached with several membranes (fins). The effects of the vortices shed from both the dorsal and anal fins on the hydrodynamic performance of the caudal fin are analyzed by prescribing an undulatory swimming kinematics to a full body sunfish model. Results show that both the dorsal fin vortices and the anal fin vortices can increase the thrust and efficiency of the caudal fin comparing to caudal fin only case. This is because the dorsal/anal fin not only can feed vorticity into the caudal fin wake via vortex shedding, but also can modulate the flow in the downstream in a way of forming a jet with stronger backward component.

Resumen
Abstract This paper studies the effect of the the caudal fin superimposed chordwise deformation on the propulsion performance. The calculation results show that the appropriate chord deformation of the caudal fin can effectively increase the propulsion efficiency. It describes the shape change of dolphin tail by mathematical formula, and studies the propulsion performance of dolphin tail by numerical method combined with morphing mesh technology. The quasi-propulsive efficiency is used to evaluate the propulsive efficiency of self-propelled objects. The results show that the active deformation of the caudal fin can effectively increase the propulsive efficiency and provide a theoretical basis for the design of the flapping bionic propeller.

Resumen
The dolphins’ cruising, generally, with an extremely high thrust efficiency and low drag, which attracted many researchers’ wide attention. It is hoped that we can improve the hydrodynamic performance of underwater vehicle by studying the thrust characteristics of dolphin’s kick and the relationship between the formation of vortex and the thrust generation. However, previous work is mostly focused on investigation of hydrodynamic performance of dolphin fluke motion with a rigid tail which means that the locomotion of caudal fin is defined only by the oscillating motion, without the chordwise deformation. In this paper, the dolphin’s fluke motion is realized by a flexible caudal fin which is defined by a combination of oscillating motion and chordwise deformation. The simulation of the dolphin fluke motion is achieved by STRA-CCM, and dynamic moving mesh is implemented for different stroke functions. This paper primarily analyzed the thrust characteristics and the formation of vortex of dolphin fluke motion, then compared with the available data from previous work with rigid tail. It can be found that the structure of the vortex generated by the dolphin fluke motion with flexible caudal fin is different from a rigid one. Finally, by analyzing the instantaneous flow condition behind the dolphin caudal fin, it can get the reason why the thrust generated by the flexible caudal fin is larger than rigid one.

Resumen
Presented in this study is a mathematical model and preliminary experimental results of a ribbed caudal fin to be used in an aquatic robot. The ribbed caudal fin is comprised of two thin beams separated by ribbed sectionals as it tapers towards the fin. By oscillating the ribbed caudal fin, the aquatic robot can achieve forward propulsion and maneuver around its environment. The fully enclosed system allows for the aquatic robot to have very little effect on marine life and fully blend into its respective environment. Because of these advantages, there are many applications including surveillance, sensing, and detection. Because the caudal fin actuator has very thin side walls, Kirchhoff-Love’s large deformation beam theory is applicable for the large deformation of the fish-fin actuator. In the model, it is critical to accurately model the curvature of beams. To this end, C1 beam elements for thin beams are developed by specializing the shear-deformable beam elements, developed by the authors, based upon Reissner’s shear-deformable nonlinear beam model. Furthermore, preliminary experiments on the ribbed fin are presented to supplement the FE model.

Resumen
Swimming fishes exhibit remarkable diversities of the caudal fin geometries. In this work, a computational study is conducted to investigate the effects of the caudal fin shape on the hydrodynamic performance and wake patterns in flapping propulsion. We construct the propulsor models in different shapes by digitizing the real caudal fins of fish across a wide range of species spanning homocercal tails with low aspect ratio (square shape used by bluegill sunfish, rainbow trout, etc.) or high aspect ratio (lunate shape adopted by tuna, swordfish, etc.), and even heterocercal caudal fin adopted by sharks. Those fin models perform the same flapping motion in a uniform flow to mimic fish’s forward swimming. We then simulate the flow around the flapping fins by an in-house immersed-boundary-method based flow solver. According to the analysis of the hydrodynamic performance, we have found that the lunate shape model (high aspect-ratio) always generates a larger thrust compared to other models. The comparison of the propulsive efficiency shows that the large aspect ratio fins (tuna and shark) have a higher efficiency when the Strouhal number (St) is in the range of steady swimming (0.2<St<0.4), while the lower aspect ratio caudal fins (catfish, trout, etc.) are more efficient when St>0.4, in which the fish is accelerating or maneuvering. Finally, the 3D wake patterns of those propulsors are analyzed in detail.

Resumen
Review question / Objective: Epidural injection (EI) has been used to manage lower back or radicular leg pain from herniation of lumbar disc (HLD). Three types of EI techniques, including transforaminal (TFEI) interlaminar (ILEI), and caudal epidural injections (CEI), are being applied. We aimed to evaluate the comparative effect of TFESI, ILEI, and CEI for reducing pain or improving function in patients with HLD. Condition being studied: For controlling inflammation by the HLD, various oral medications and procedures are used. Among these therapeutic methods, EI of the drugs is frequently used in clinical practice. Its positive HLD-induced pain reducing effect was reported in several previous studies. Three types of techniques, including TFEI, ILEI, and CEI, have been utilized in clinical practice. conflicting outcomes as to which technique is superior were reported in previous studies. So far, some meta-analysis studies for comparing the effects of different EI techniques on HLD were conducted. However, these previous studies conducted comparison between two procedures among TFEI, ILEI, and CEI. In the current study, using network meta-analysis, we synthesize and compare the effects of TFEI, ILEI, and CEI on pain from HLD, together.
