Texto completoPeter’s anomaly (PA) is a congenital defect of the anterior chamber of the eye. The aim of this study is molecular characterization of a de novo balanced chromosome translocation [t(11;18)(q23.3;q11.2)] identified in a proband with syndromic form of Peter´s anomaly (ectopia lentis and mild CNS abnormalities). Chromosome breakpoints were identified at nucleotide resolution. The 11q23.3 breakpoint is at position 120,097,868 (genome assembly GRCh37/hg19) within intron 3 of out of first protein homolog gene (OAF) while, 18q11.2 breakpoint in the intergenic region between CTAGE1 and RBBP8 genes, at position 20,220,714. Although OAF is disrupted, its expression level is unchanged in proband´s lymphoblastoid cell line (LCL). Cell adhesion protein Nectin 1 or PVRL1, located 500 kb upstream from 11q23.3 breakpoint, reveal 3.5 fold increase in proband’s LCL. Expression levels of additional genes from breakpoint regions are not significantly different. Furthermore, RT-qPCR confirmed that expression level CYP1B1 from chromosome 2p22.2, and EDIL3 from chromosome 5q14 are significantly changed in proband´s LCL, 16.5 fold decrease and 126 fold increase, respectively. Alterations in CYP1B1 have been reported as PA causing mutations. Therefore, mutation screening of CYP1B1 was performed and no pathogenic mutation was identified in the proband, although, a disease-causing 13 bp, homozygous deletion was identified in a Portuguese patient with PA. In conclusion, we hypothesize that PVRL1 is a candidate gene for at least some of the observed clinical features, which is elicited in mouse models by association of its paralog PVRL3 to lens and other ocular defects involving the ciliary body. Furthermore, the involvement of the POU family domain containing transcription factor (POU2F3) from 11q23.3 and CYP1B1 or EDIL3, localized outside of the breakpoint regions, in the underlying molecular pathogenesis of syndromic PA remains to be determined.

Texto completoThe chicken egg is an aseptic and a self-sufficient chamber allowing the harmonious development of the chicken embryo in 21 days. The eggshell is the first physical barrier of the egg, which protects the egg from mechanical and microbial stresses. During eggshell biomineralization in the uterine fluid, the organic matrix, which is secreted by the uterus, ensures a crucial role in this process. To decipher the role of two major proteins of the organic matrix (EGF-like repeats and discoidin-like domains 3 : EDIL3 et Milk fat globule EGF-factor 8 : MFGE8), in silico and experimental approaches were performed. We firstly investigated the common evolution history of EDIL3 and MFGE8. We have estimated a divergent point between both paralogs 480 million years ago. The results also allowed us to highlight a vesicular transport of mineral (amorphous calcium carbonate) involved in eggshell biomineralization. EDIL3 and MFGE8 proteins would guide the vesicles to the mineralizing site and consequently ensure a critical role in this transport. This work also described additional molecular actors involved in vesicles formation, in calcium and bicarbonates supply and to the stabilization of amorphous calcium carbonate. We therefore proposed a global and coherent model of mineral transport during eggshell calcification. In this model, three alternative pathways could be involved (transcellular, vesicular and paracellular). Further investigations will be necessary to determine the respective roles of these pathways

Texto completoEating disorders (ED) affect a large population and are a serious public health issue. It is thus essential to deepen our knowledge of ED pathophysiology in order to open new therapeutic perspectives. The prevalence of ED according to sex, geographical origin or age of the study population and its increase over the last 15 years has been highlighted in a systematic review. The analysis of patients with ED from the Eating Disorder and Longitudinal Survey (EDILS) cohort underlined stress and / or diet as triggering factors and noted similarities between the comorbidities of the 3 broad categories of ED (Restrictive, bulimic and compulsive). These comorbidities were strongly associated with each other, and contributed in a vicious circle to the perpetuation of ED. The literature suggests during ED a dysregulation of neuropeptide signaling of food intake that may include some immunological mechanisms. Analyzes of plasma samples from the EDILS cohort allowed a better definition of the peptides and immunoglobulins profiles associated with the 3 broad categories of ED and to evaluate their utility for phenotyping patients. The collection of feces in the cohort offers the possibility of refining the specificity of the microbiota according to different type of ED. This thesis allows to better define the clinical characteristics of 3 broad categories of ED and disturbances of the microbiota-intestine-brain axis, particularly the associated biological dysregulations of neuropeptide signaling
