Resumen
Endothelin (EDN) is a possible regulating factor of oviductal motility, which is important for the transport of gametes and embryo. To clarify the factors that control the secretion of EDN in the bovine oviduct, the expression of EDNs, EDN-converting enzymes (ECEs) and EDN receptors (EDNRs) were investigated. All isoforms of EDN (EDN1–3), ECE (ECE1 and ECE2) and EDNR (EDNRA and EDNRB) were immunolocalised in the epithelial cells of the ampulla and the isthmus. EDNRs were also immunolocalised in smooth-muscle cells. The mRNA expression of EDN2 and ECE2 was higher in cultured ampullary oviductal epithelial cells than in isthmic cells. The expression of EDN1, EDN2 and ECE2 in the ampullary tissue was highest on the day of ovulation. Oestradiol-17β increased EDN2 and ECE1 expression, while progesterone increased only ECE1 expression in cultured ampullary epithelial cells. These results indicate that EDNs are produced by epithelial cells and their target site is smooth-muscle and epithelial cells, and suggest that ovarian steroids are regulators of endothelin synthesis in ampullary oviductal epithelial cells.

Resumen
Environmental DNA (eDNA) analysis is a biological survey method that has drawn much attention in recent years. However, the results of eDNA analysis and capture surveys often do not completely match, and the validity of the eDNA analysis needs to be verified. Verification of eDNA metabarcoding was conducted in a river in Fukushima Prefecture, Japan, in comparison with capture survey data. Most of the captured species were detected, and 13 uncaptured lineages (two genera and 11 species) were detected in the eDNAs. Some rare species detected in the eDNAs were also identified, including exotic eels and an endangered yet out-of-range bitterling fish. To confirm the validity of the exotic Anguilla spp. detected in eDNAs, mitochondrial Cytb sequencing was performed on captured eel specimens. All eel specimens were sequenced as the native Anguilla japonica, indicating a small biomass of the exotic species difficult to capture. Our results clearly indicated the eDNA analysis as a powerful tool for detecting possible habitats of rare fish species without disturbance to the natural environment.

AbstractEndothelins (EDNs) participate in various physiological events including smooth muscle contraction, nitric oxide (NO) synthesis, and embryonic development. In this study, we investigated the regional roles of EDNs produced by bovine oviductal epithelial cells in NO synthesis and smooth muscle motility. Quantification of mRNA expressions indicated that expression of EDN receptor B (EDNRB) in the ampullary region was higher after ovulation than before ovulation, whereas expression ofEDNRAin the isthmic region was higher after ovulation than before ovulation. Immunohistochemistry revealed that the EDN receptors (EDNRA and EDNRB) were expressed in the epithelium, whereas smooth muscle showed positive staining only for EDNRA. The expressionsPlease suggest whether 'NOS2' can be treated as the updated symbol for 'iNOS' as per gene nomenclature. ofinducible NO synthase (iNOS) protein and its mRNA (NOS2)in cultured epithelial cells isolated from the ampulla were stimulated by EDN1, but not by EDN2 or EDN3, after 1h of incubation. In isthmic epithelial cells, none of the EDNs affected the expression ofNOS2. Isometric contraction tests indicated that spontaneous waves were strong in the isthmic region but weak in the ampullary region. EDN1 modulated smooth muscle motility in both the regions. The overall findings suggest that EDN1 plays region-specific roles in smooth muscle motility and epithelial NO synthesis, providing an optimal oviductal microenvironment for transport of gametes, fertilization, and development/transport of early embryo.

Resumen
Environmental DNA (eDNA) is an increasingly popular tool in biological and ecological studies. As a biproduct of its increasing use, large number of eDNA samples are being collected and stored, that potentially contain information of many non-target species. One potential use for these eDNA samples is a surveillance and early detection of pathogens and parasites that are otherwise difficult to detect. Echinococcus multilocularis is such a parasite with serious zoonotic concern, and whose range has been expanding. If eDNA samples from various studies can be repurposed in detecting the parasite, it can significantly reduce the costs and efforts in surveillance and early detection of the parasite. We designed and tested a new set of primer-probe for detecting E. multilocularis mitochondrial DNA in environmental medium. Using this primer-probe set, we conducted real-time PCR on repurposed eDNA samples collected from three streams flowing through an area of Japan endemic to the parasite. We detected the DNA of E. multilocularis in one of the 128 samples (0.78%). The discovery suggests that while detecting E. multilocularis using eDNA samples is possible, the rate of detection appear to be very low. However, given the naturally low prevalence of the parasite among wild hosts in endemic areas, the repurposed eDNAs may still be a valid option for surveillance in newly introduced areas with the reduced cost and efforts. Further work is needed to assess and improve the effectiveness of using eDNA for detection of E. multilocularis.

Resumen
The transport of eDNA is one of the key environmental behaviors for its spreading and dispersal. Microplastics (MPs) are widely present in the soil environment and directly affect the environmental behavior of co-coexisting soil pollutants. However, the effect of MPs on eDNA transport and its mechanism remain unclear. In this study, we systematically investigated the effect of MPs types and functional groups on eDNA transport. The results showed that different kinds of MPs promoted eDNA transport, but there was no significant difference between these two MPs types. MPs with two different functional groups inhibited eDNA transport, and the transport rate of eDNA decreased by 8.9% and 7.0%, respectively. PSNH2 inhibited eDNA transport by reducing electrostatic repulsion, enhancing electrostatic adsorption, and reducing porosity of porous media, enhancing the interaction between eDNA and MPs. In the presence of kaolin, the inhibition effect of MPs on eDNA transport increased with the proportion of kaolin increased. The positive charge of kaolin enhanced the electrostatic adsorption between MPs and eDNA, and inhibited the transport of eDNA. This study revealed the transport rule of eDNA in the presence of MPs, and provided a theoretical basis for a comprehensive assessment of the environmental and ecological risks of coexistence of MPs and eDNA.

Introduction. Pseudomonas aeruginosa grows in extracellular DNA (eDNA)-enriched biofilms and infection sites. eDNA is generally considered to be a structural biofilm polymer required for aggregation and biofilm maturation. In addition, eDNA can sequester divalent metal cations, acidify growth media and serve as a nutrient source. Aim. We wanted to determine the genome-wide influence on the transcriptome of planktonic P. aeruginosa PAO1 grown in the presence of eDNA. Methodology. RNA-seq analysis was performed to determine the genome-wide effects on gene expression of PAO1 grown with eDNA. Transcriptional lux fusions were used to confirm eDNA regulation and to validate phenotypes associated with growth in eDNA. Results. The transcriptome of eDNA-regulated genes included 89 induced and 76 repressed genes (FDR<0.05). A large number of eDNA-induced genes appear to be involved in utilizing DNA as a nutrient. Several eDNA-induced genes are also induced by acidic pH 5.5, and eDNA/acidic pH promoted an acid tolerance response in P. aeruginosa . The cyoABCDE terminal oxidase is induced by both eDNA and pH 5.5, and contributed to the acid tolerance phenotype. Quantitative metal analysis confirmed that DNA binds to diverse metals, which helps explain why many genes involved in a general uptake of metals were controlled by eDNA. Growth in the presence of eDNA also promoted intracellular bacterial survival and influenced virulence in the acute infection model of fruit flies. Conclusion. The diverse functions of the eDNA-regulated genes underscore the important role of this extracellular polymer in promoting antibiotic resistance, virulence, acid tolerance and nutrient utilization; phenotypes that contribute to long-term survival.

The focus of our research is on Edna Pontellier, the protagonist of The Awakening, a 19th century novel whose main character is a housewife who was in search of her own emancipation. Our theory and critical resources are supported by works written by Per Seyersted(1979) and Nadilza Moreira(2003) both references names for Chopin's scholarship.Besides, we used Beauvoir' (1970), Woolf' (2012) and Showalter's(1979) critical studies to help us to develop our understanding of the 19th century social-hisorical women's context in the USA. For the study of representation we used the concept fromAna Gabriela Macedo e Ana Luisa Amaral(2005). To support our analysis on the role of the narrative voice we used Chris Baldick(1991) Our conclusion is that: Edna Pontellier represents a multiplicity of images and they are presented to the reader as the narrative develops itself showing Edna's multiples “awakenings” as a woman in transition.
O foco de nossa pesquisa está voltado para Edna Pontellier, a protagonista da novela, O Despertar; Edna uma dona de casa do século XIX que lutou pela emancipação como sujeito. Nossa fundamentação teórica se apóia nas obras escritas por Per Seyersted (1979) e Nadilza Moreira (2003) ambos nomes de referências nos estudos sobre Chopin. Além disso, usamos Beauvoir '(1970), Woolf’ (2012) e os estudos críticos de Showalter para nos ajudar a desenvolver a compreensão do contexto sócio-histórico das mulheres do século XIX nos EUA. Para o estudo da representação utilizamos os estudos de Ana Gabriela Macedo e Ana Luisa Amaral (2005). Finalmente, para apoiar a nossa análise sobre o papel da voz narrativa na novela nos amparamos nos estudos de Chris Baldick (1991). Concluimos que: a representação da protagonista, Edna Pontellier, não é una, mas múltipla e caminha juntamente com os diversos “despertares” vivenciados pela personagem ao longo da trama narrativa.

A adopção da Comunicação Não Verbal como uma estratégia narrativa usada pela escritora irlandesa Edna O’Brien na trilogia The House of Splendid Isolation (1994), Down by the River (1996) e Wild Decembers (1999) constituiu o ponto de partida para a elaboração da presente dissertação. As áreas da Comunicação Não Verbal como a linguagem cinestésica ou a proxémica (a percepção do espaço e o uso que dele fazemos) revelaram-se pertinentes para a análise da referida trilogia. É, igualmente, objectivo deste trabalho problematizar algumas questões que se prendem com a expressão da voz e da experiência no feminino. O corpo, portador de significação, deve ser lido como um texto que oferece uma multiplicidade de leituras e de diferentes perspectivas. Neste contexto, a expressão da voz e da experiência feminina passa necessariamente pela linguagem corporal, assumindo-se como uma alternativa ao discurso patriarcal. A exploração das potencialidades que o corpo oferece como portador de significado e como elemento de ruptura do discurso dominante será também motivo de análise e de discussão neste trabalho. ABSTRACT: The main purpose of this work is to analyse the trilogy, The House of Splendid Isolation (1994), Down by the River (1996) and Wild Decembers (1999) by the Irish writer Edna O’Brien, highlighting the role of Non Verbal Communication as a narrative strategy which reveals women’s voice and experience. It will also be discussed how body language and proxemics are cunnily used by O’Brien as means to convey the idea that women have a discourse of their own which assumes itself as an alternative to the patriarchal one. The body portrays meaning and it should be read as a meaningful text. The multiplicity of readings and perspectives that are offered are quite interesting and enriching. This dissertation will basically offer personal readings of the above trilogy as well as an opportunity to discuss the potentialities of body language as a way of asserting women’s identity and self expression.

Abstract This paper presents the results of the application of eDNA analysis as a biodiversity survey technique in onshore O&G concessions, in comparison with conventional survey methods, with the aim of highlighting the advantages and disadvantages and making specific recommendations. Two case studies, in Italy and Albania, are presented. In Italy (Val d'Agri), Eni has been using eDNA since 2021 as part of a Biodiversity & Ecosystem Services Action Plan (BAP) to complement conventional monitoring methods of two freshwater species: yellow-bellied-toad (Bombina pachypus) and Eurasian otter (Lutra lutra). The results confirmed the presence of both species. In 2023 monitoring for yellow-bellied toad was repeated to confirm distribution. The species was not detected, but the analysis of mammal primers enabled the collection of additional valuable data for other species. In Albania, Eni used eDNA analysis in 2022 focusing on freshwater fish to complement the collection of baseline data using conventional methods prior to a seismic survey. Samples were obtained from rivers and lakes within the survey area and at a control site. Both eDNA metabarcoding and DNA barcoding were used. eDNA analysis resulted in the identification of 24 taxa of fish. Two threatened species and six restricted-range species were detected. Nine taxa recorded with eDNA analysis were not caught with the traditional fish survey which identified eight species not detected by eDNA. Furthermore, DNA barcoding enabled the addition of seven new species of fish to the global DNA database. The case studies presented indicate that while eDNA analysis can provide valuable data with reduced time and cost compared to conventional methods, it is their combined utilization that leads to better results and a more cost and time-effective approach.

Resumen
Environmental DNA (eDNA) analysis, or the detection of trace DNA shed by organisms into their environment, has the potential to transform Army capabilities for threatened and endangered species (TES) and invasive species management by providing a rapid, noninvasive, and cost-effective option for monitoring wildlife. Despite these benefits, eDNA analysis is underutilized on military installations as limited access to guidance materials, protocols, training opportunities, and support from eDNA scientists makes it difficult for installation biologists and military land managers to design and execute eDNA surveys, let alone identify management questions that may benefit from eDNA monitoring. Therefore, the aim of this resource is to increase awareness of the benefits and limitations of eDNA monitoring and provide eDNA study design guidelines and field sampling protocols for nonexperts to make this tool more accessible to installation biologists and land managers and help facilitate the adoption of eDNA-based approaches for wildlife management on military ranges.

Resumen
We designed two novel environmental DNA (eDNA) assays for the detection of Ridgway’s Rail (Rallus obsoletus), and successfully validated each assay using eDNA samples collected from the species’ known distribution within the United States. These assays add to the suite of tools available for the monitoring of this rare and secretive marsh bird, and may help to further elucidate its movement patterns as well as identify important migration corridors. Observed sensitivity of the assays indicates exceptional performance, with limits of detection at ≤ 8 copies of the target eDNA fragment per reaction. Our publication adds to the growing body of literature supporting eDNA surveys as viable tools for bird monitoring endeavors.

Resumen
Aquatic environmental DNA (eDNA) surveys are sometimes impacted by polymerase chain reaction (PCR) inhibitors. We tested varying concentrations of different inhibitors (humic, phytic, and tannic acids; crude leaf extracts) for impacts on quantitative PCR (qPCR) assays designed for eDNA surveys of bighead and silver carp (Hypophthalmichthys nobilis and Hypophthalmichthys molitrix). We also tested for inhibition by high concentrations of exogenous DNA, hypothesizing that DNA from increasingly closely related species would be increasingly inhibitory. All tested inhibitors impacted qPCR, though only at very high concentrations — likely a function, in part, of having used an inhibitor-resistant qPCR solution. Closer phylogenetic relatedness resulted in inhibition at lower exogenous DNA concentrations, but not at relatively close phylogenetic scales. Inhibition was also influenced by the qPCR reporter dye used. Importantly, different qPCR assays responded differently to the same inhibitor concentrations. Implications of these results are that the inclusion of more than one assay for the same target taxa in an eDNA survey may be an important countermeasure against false negatives and that internal positive controls may not, in the absence of efforts to maximize inhibition compatibility, provide useful information about the inhibition of an eDNA assay.

Resumen
Os esforços para a construção de um banco de dados genético com códigos de barra de DNA das espécies da Flora de Carajás possibilitaram obter “impressões digitais” das espécies que ali ocorrem. A partir do banco de dados de referência criado pelo ITV, o ITVBiobase, outras tecnologias moleculares como a de DNA metabarcoding pode ser aplicada visando o monitoramento das espécies de forma rápida, automatizada e eficiente. A técnica de DNA metabarcoding possibilita identificar simultaneamente múltiplas espécies de plantas a partir dos rastros de DNA presente em uma amostra ambiental, descartando a necessidade de amostragem direta de espécimes. Nesse sentido, estudos para validação do uso da técnica de DNA metabarcoding para monitoramento da biodiversidade da flora em Carajás, foram realizados com o objetivo de criar um manual padronizado incluindo as etapas desde a coleta em campo até as análises laboratoriais e de bioinformática.

A ictiofauna amazônica é composta por uma enorme diverdidade de peixes sendo muitas espécies de ocorrência local. Programas de conservação e monitoramento são muitas vezes dificultados tanto pela falta de especialistas quanto pela dificuldade da captura dos espécimes. A criação de indicadores biológicos é de suma importância principalmente para mineração, que necessita realizar monitoramento periódico para verificar se suas atividades estão ou podem causar impactos sob a diversidade ao entorno do empreendimento. Na Serra dos Carajás, um dos maiores depósitos mineral do mundo, existem lagoas que demandam monitoramento devido suas peculariedades e possibilidades de impactos. Essas lagoas são formadas sob crostas lateríticas ricas em ferro, em platôs acima de 770 m acima do nível do mar, no qual o nível de água é mantido pelo do regime de chuvas da região. Pensando em estratégias de manejo mais eficientes e rápidos, este trabalho teve como objetivo validar uma nova abordagem de levantamento de biodiversidade da ictiofauna utilizando os rastros de DNA presentes no ambiente. Para isso, foi usado a técnica de DNA metabarcoding que permite avaliar a diversidade e riqueza de espécies sem necessidade de captura, e sim através da análise de pequenos fragmentos de DNA a partir de uma amostras ambiental, como água ou sedimentos dos rios e lagoas.
