Resumen
The intraerythrocytic malaria parasite constructs an intracellular haem crystal, called haemozoin, within an acidic digestive vacuole where haemoglobin is degraded. Haem crystallization is the target of the widely used antimalarial quinoline drugs. The intracellular mechanism of molecular initiation of haem crystallization, whether by proteins, polar membrane lipids or by neutral lipids, has not been fully substantiated. In the present study, we show neutral lipid predominant nanospheres, which envelop haemozoin inside Plasmodium falciparum digestive vacuoles. Subcellular fractionation of para

Lipins are phosphatidate phosphatases that generate diacylglycerol (DAG). In this study, we report that yeast lipin, Pah1p, controls the formation of cytosolic lipid droplets. Disruption of PAH1 resulted in a 63% decrease in droplet number, although total neutral lipid levels did not change. This was accompanied by an accumulation of neutral lipids in the endoplasmic reticulum (ER). The droplet biogenesis defect was not a result of alterations in neutral lipid ratios. No droplets were visible in the absence of both PAH1 and steryl acyltransferases when grown in glucose medium, even though the

Neutral lipids fulfill multiple specialized roles in cellular function. These roles include energy storage and utilization, the synthesis of complex lipids in cellular membranes, lipid second messengers for cellular signaling, and the modulation of membrane molecular dynamics. We have developed a novel mass spectrometric technology, now termed shotgun lipidomics, that can identify the types and amounts of thousands of lipids directly from extracts of biological samples. Shotgun lipidomics is well suited for the identification and measurement of the types and amounts of neutral lipid classes an

Since energy storage is a basic metabolic process, the synthesis of neutral lipids occurs in all kingdoms of life. The yeast, Saccharomyces cerevisiae, widely accepted as a model eukaryotic cell, contains two classes of neutral lipids, namely steryl esters and triacylglycerols. Triacylglycerols are synthesized through two pathways governed by the acyl-CoA diacylglycerol acyltransferase Dga1p and the phospholipid diacylglycerol acyltransferase Lro1p, respectively. Steryl esters are formed by the two steryl ester synthases Are1p and Are2p, two enzymes with overlapping function which also catalyz

The endoplasmic reticulum (ER) is a hub that coordinates neutral lipid synthesis, storage, and export. To fulfill this role, the ER maintains close contact with lipid droplets (LDs), which are evolutionarily conserved organelles for the storage of neutral lipids. Decades of biochemical evidence points to fatty acid modification and neutral lipid synthesis in the ER. Conceptually, lipid export into extracellular space or lipid retention intracellularly require the subsequent remodeling of an ER membrane leaflet that faces the lumen or cytoplasm, respectively. This is a particularly notable accomplishment due to the highly hydrophilic nature of the sulfate anion, and the associated challenges with partitioning such a species into a hydrophobic lipid membrane. Resumen
A method was developed for analyzing the high frequency dielectrophoresis (>20 MHz) response of microalgal cells with different lipid content on the basis of media conductivities. At these frequencies, the DEP response is expected to depend primarily on the dielectric properties of the cytoplasm. The medium used to perform DEP experiments has an important influence on their outcome since all properties are measured in relation to the medium, therefore its properties must be tightly controlled. This method is label free and only uses electric fields, and thus it is suitable for many other ap

Resumen
Bio-Synthetic Paraffinic Kerosene (Bio-SPK) is one of the most anticipated renewable energy to conventional Jet kerosene (CJK). Bio-SPK is plant lipid which is thermo-chemically converted to kerosene like compositions to serve as “Drop-in” biojet fuel. The environmental impact of Bio-SPK is to be understood to determine its potential as a carbon neutral / negative fuel. Assessment of Life Cycle Emissions of Bio-SPKs (ALCEmB) aims to deliver a quantitative, life cycle centered emissions (LCE) model, reporting the process related-carbon footprint of Bio-SPKs. This study also encompasses the key
