Background: During total knee arthroplasty (TKA) for osteoarthritis, the sagittal gap imbalance (SGI) with a relatively large extension gap is an important surgical challenge. We determined the predictors of SGI with a relatively large extension gap and evaluated the surgical outcomes of knees with SGI. Methods: 551 consecutive cases of primary TKA for osteoarthritis were retrospectively reviewed. The cohort was divided into two groups according to the SGI and statistically matched according to baseline characteristics via the inverse probability of treatment weighting method. Multiple linear and logistic regression analyses were performed to determine the predictors of sagittal gap difference (SGD) and SGI. Intergroup differences in clinical and radiological outcomes were analyzed. Results: Of all the knees included, 8.5% (n = 45) presented with SGI with a relatively large extension gap and required femoral sagittal balancing to manage SGI. The hyperextension angle (HA), preoperative joint line convergence angle (JLCA), and the change in posterior tibial slope (PTS) significantly correlated to SGD and predicted SGI with a relatively large extension gap. SGI group showed significant changes in femoral posterior condylar offset and joint line height compared to those without SGI (1.48 vs −0.45, 1.37 vs −0.51, respectively). Postoperative ROM and knee society knee scores were lower in SGI group. Conclusion: Knees requiring sagittal balancing to manage SGI with a relatively large extension gap is not uncommon in TKA for osteoarthritic knees. The change in PTS is an independent and modifiable predictor of SGI.

Purpose: To evaluate and compare the dichotic listening abilities in non-hearing-impaired adults monolingual speakers of the Brazilian Portuguese language (CG) and simultaneous Brazilian Portuguese-German (GG) bilingual speakers or successive Brazilian Portuguese-Italian bilingual speakers (IG). Methods: This is about an observational, descriptive, transverse and quantitative research. The sample consisted of 87 subjects aged between 18 and 55 years, divided into: Control Group (CG), of 30 monolingual speakers of Brazilian Portuguese; Study Group A (SGA), of 31 simultaneous Brazilian Portuguese-German bilingual speakers; and Study Group I (SGI), of 26 successive Brazilian Portuguese-Italian speakers. The individuals were submitted to the Dichotic digits test (DDT) and to Staggered Spondaic Words (SSW). Results: The DTT results showed difference in right ear and total scores when comparing SGA to CG. Comparing the CG and the SGI, it was observed difference in right and left ears and total scores. Comparing the SGA and the SGI, no difference was observed between the groups. Results of SSW showed that both bilingual groups were significantly better in the right and left ears scores and even in total one when compared to CG. Comparing the SGA and the SGI, the SGI showed better significant scores in the right ear and total. Conclusion: Bilingual experiences seem to influence positively the ability of high predictability dichotic listening, evaluated by DDT, and the low predictability dichotic listening, evaluated by SSW test. The SSW results also showed statistically significantly better results for successive Brazilian Portuguese-Italian bilingual speakers when compared to simultaneous Brazilian Portuguese-German speakers.

Gas reservoirs are mainly produced by depletion with an aquifer rise; reservoir simulation requires two main SCAL inputs: the amount of trapped gas by the aquifer (residual gas saturation: Sgr) and the relative permeability to water due to aquifer flooding. As it is quasi impossible to predict aquifer strength, the primary SCAL input for reservoir simulation is the Sgr. The recovery factor is directly defined by initial and residual gas saturations. In fact, the residual gas saturation Sgr highly depends on the initial gas saturation Sgi and there is no universal petrophysical parameter governing the shape of this curve. This relationship can be described by several different models (Land, Aissaoui…). While Land’s model is widely used, the Aissaoui model better fits the experimental results (Suzanne et al. 2003), at least for homogeneous sandstones. For a given threshold of initial gas saturation Sg0, this relationship typically exhibits a plateau at high Sgi>Sg0 and an increasing linear trend at low Sgi<Sg0. The challenge here is to properly estimate the value of the Sg0 threshold. Classical laboratory method would require one experiment per point in the Sgr/Sgi plot, and therefore can be achieved in a matter of months. Here we propose a laboratory method allowing the acquisition of the Sgr/Sgi curve in a few days. The proposed method combines centrifugation and capillary rise under imaging. First, the centrifuge allows creating a saturation profile along a sample; measured by NMR. Then, capillary rise is used to capture Sgr under NMR monitoring. By adding NMR imaging, this technique allows combining the benefits of centrifugation to explore a wide range of Sgi; and the ease and cost effectiveness of capillary rise to measure the resulting Sgr. Therefore, at a timescale close to a traditional capillary rise, the proposed technique avoids Land extrapolation and provides a direct measurement of Sgr in a wide range of Sgi. As an additional benefit, the combination of NMR and centrifuge can provide at the same time a direct measurement of capillary pressure, providing information on the gas in place and potential imbibition process in the reservoir.

SUMMARYLegionella isolates recovered in 21 different Spanish provinces over 8 years from both clinical (67 isolates) and environmental (181) samples, mostly from case-associated buildings, are described: 92·5% of clinical isolates wereL. pneumophilaserogroup 1 (SGI), only five isolates belonging to other species or serogroups: twoL. pneumophilaSG6, two SG8 and oneL. bozemaniiSG1 not clearly related with clinical infection.L. pneumophilaSG1 accounted for 53·6% of isolates from the environment, followed by SG8 (27·6%). SG3 (9·4%) and SG6 (7·2%). Three isolates were labelled as SG8/10.Subtyping ofL. pneumophilaSG1 by the standardized panel of monoclonal antibodies revealed 90·3% of clinical and 78·3% of environmental isolates as belonging to Pontiac subgroup. Pontiac isolates were further divided into 55·3% Philadelphia 1 or Allentown 1, 21·9% Benidorm 030E and 20·4% Knoxville 1.Characterization of samples from four outbreaks in which both clinical and environmental isolates had been recovered permitted the recognition of three Philadelphia 1 or Allentown 1 and one Knoxville 1 strains as the aetiological agents.

Abstract Introduction: We have developed a promising leukemia vaccine in which patient derived AML cells are fused with autologous dendritic cells (DCs) effectively presenting a broad array of antigens that capture the heterogeneity of the leukemia cell population. A major challenge of developing effective immunotherapy is overcoming the immunosuppressive milieu that characterizes patients with AML. AML cells evade immune targeting, in part, through the limited presentation of antigen by primitive leukemia progenitors. In contrast, induction of differentiation through upregulation of reactive oxygen species may enhance antigen presentation and the capacity to target AML cell by a tumor vaccine. Hypomethylating agents have been shown to be effective therapeutic agents for patients with AML and MDS, in part due to immunomodulatory effects. In the current study, we have examined the immunomodulatory properties of the novel DNA hypomethylating agent SGI-110, a dinucleotide of decitabine (DAC) and deoxyguanosine, which is resistant to in-vivo inactivation by cytidine deaminase. We have explored the in-vitro effect of treatment with SGI-110 on ROS, AML immunogenicity and efficacy of the AML/DC fusion vaccine. Methods and results: AML blasts were isolated by ficoll density centrifugation of bone marrow mononuclear cells (BMMCs) from patients with AML at presentation. AML blasts were cultured for 4 days in the presence or absence of 1 uM SGI-110, added twice daily on days 1 and 2 of culture. ROS as detected by levels of H2O2 expression demonstrated a 40% increase after treatment with SG1-110 (n=2). We subsequently evaluated whether the increase in ROS levels correlated with enhanced targeting by immune effector cells. SGI-110 or control treated AML cells were assessed for their susceptibility to T cell mediated targeting, using a standardized flourochrome CTL assay. Remarkably, a 72% increase in autologous cytotoxic T lymphocyte-mediated lysis as measured by Granzyme B activity, was demonstrated following SGI-110 treatment of AML blasts (n=3). The expression of co-stimulatory molecules CD80 and CD86, were unchanged following treatment with SGI-110 (n=3). In contrast, exposure to SGI-110 resulted in increased expression of the antigen processing TAP proteins by immunocytochemical analysis. We have previously demonstrated that DC/AML fusion cells potently stimulate the expansion of leukemia specific T cells. The effect of SGI-110 on T cell response to DC/AML fusion vaccine stimulation in vitro was assessed. Autologous Dendritic cells (DCs) were generated by culture of adherent peripheral mononuclear cells obtained from AML patients following remission in the presence of GM-CSF, IL-4 and TNF-α. DCs were fused with AML blasts by co-culture at a 1:1 ratio in the presence of polyethylene glycol (PEG). DC/AML fusions were cultured at a 1:10 ratio with autologous T cells for 5-6 days in the presence and absence of 1uM of SGI-110. Exposure of fusion stimulated autologous T cells to SGI-110 resulted in an increase in T cell expression of IFN-γ, with mean fold increase of 2.2 and 2.3 for CD4 and CD8 T cells respectively (n=4). The percentage of CD4+CD25+FOXP3+ regulatory T cells (TRegs) and T cell expression of PD1 was not significantly changed in the presence of SGI-110. The effect of SGI-110 on CTL mediated killing by vaccine stimulated T cells was assessed. Following stimulation with the DC/AML fusion vaccine, T cells mediated killing of autologous AML blasts increased to 11% from 4% following co-culture with unstimulated T cells. Interestingly, exposure of the AML cells to SGI-110 led to a further increase in T cells mediated killing with mean levels of 20% tumor lysis (n=2). Conclusions: The results demonstrate that treatment with SGI-110 results in increased ROS levels and an associated enhanced susceptibility of AML blasts to immune mediated targeting. The addition of SGI-110 to T cell stimulation by an autologous DC/AML fusion vaccine results in an increase in interferon gamma and increased susceptibility of AML cells to T cell mediated killing. The immunomodulatory properties of SGI-110, combined with its favorable pharmacologic and pharmacokinetic features, identify SGI-110 as a useful agent to implement novel combined epigenetic–immunotherapeutic strategies in AML. Abstract Lysine specific histone methylation and deacetylation and DNA hypermethylation are involved in the epigenetic silencing of tumor suppressor genes (TSG), e.g., p15 and p16. DNA methyltransferase (DNMT) inhibitors 5-azacytidine and 5-aza-2’-deoxycytidine demethylate the CpG dinucleotide islands in or near gene promoters, leading to derepression of TSGs in AML. SGI-110 (S110) (Cancer Res.2007; 67:6400) and SGI-1036 (SuperGen, Inc.) are novel, DNMT inhibitors, which also deplete DNMT1 levels. SGI-110 is a dinucleotide containing 5-aza-2’-deoxycytidine and SGI-1036 is a non-nucleoside heterocycle. The multi-protein complex PRC (polycomb repressive complex) 2 that contains the three core proteins EZH2, SUZ12 and EED, has intrinsic histone methyltransferase (HMTase) activity. This is mediated by the SET domain of EZH2, which induces trimethylation of histone H3 on lysine (K)-27. We recently reported that treatment with the pan-HDAC inhibitor panobinostat (LBH589, Novartis Pharmaceutical Corp) acetylates and inhibits the ATP binding and chaperone function of hsp90, as well as depletes the levels of EZH2, Suz12 and EED in cultured and primary AML cells (Mol Cancer Ther.2006; 5:3096). Within the PRC2 complex, EZH2 was shown to interact with and modulate the DNA methyltransferases DNMT1, DNMT3a and DNMT3b, which affects their binding to the EZH2-targeted gene promoters. In the present studies we determined the effects of SGI-110 or SGI-1036 and LBH589 on the PRC2 proteins EZH2 and SUZ12, and DNMT1, in the cultured (HL-60, OCI-AML3 and K562) and primary AML cells. Treatment with SGI-110 (0.5 to 2.0 μM) or SGI-1036 (0.5 and 1.0 μM) for 24 hours depleted protein levels of DNMT1 and EZH2 in the cultured and primary AML cells. SGI-110 and SGI-1036 promoted proteasomal degradation of DNMT1 and EZH2 since co-treatment with bortezomib significantly restored DNMT1 and EZH2 levels in the AML cells. Following treatment with SGI-110 or SGI-1036, bisulfite modification and methylation specific PCR demonstrated increase in unmethylated promoter DNA of p15 and JunB. This was associated with induction of the mRNA and protein levels of p15 and JunB, as well as caused inhibition of cell cycle progression (% of cells increased in G1 and increased in S phase) and colony growth in the soft agar. Treatment with 1.0 μM of SGI-110 or SGI-1036 also induced PARP cleavage activity of caspases and induced morphologic evidence of apoptosis in the AML cells. Co-treatment with 10 to 50 nM panobinostat enhanced SGI-110 or SGI-1036 mediated depletion of DNMT1 and EZH2, with more de-repression of the p15 and JunB and significant increase in apoptosis of AML cells. Collectively, these findings indicate that, SGI-110 and SGI-1036 deplete DNMT1 and EZH2 levels, as well as exert potent anti-AML activity. Additionally, combined epigenetic therapy consisting of SGI-110 or SGI-1036 in combination with panobinostat may represent a promising novel treatment of AML.

Abstract. A new index for standardising groundwater level time series and characterising groundwater droughts, the Standardised Groundwater level Index (SGI), is described. The SGI is a modification of the Standardised Precipitation Index (SPI) that accounts for differences in the form and characteristics of precipitation and groundwater level time series. The SGI is estimated using a non-parametric normal scores transform of groundwater level data for each calendar month. These monthly estimates are then merged to form a continuous index. The SGI has been calculated for 14 relatively long, up to 103 yr, groundwater level hydrographs from a variety of aquifers and compared with SPI for the same sites. The SPI accumulation period which leads to the strongest correlation between SPI and SGI, qmax, varies between sites. There is a positive linear correlation between qmax and a measure of the range of significant autocorrelation in the SGI series, mmax. For each site the strongest correlation between SPI and SGI is in the range 0.7 to 0.87, and periods of low values of SGI coincide with previously independently documented droughts. Hence SGI is taken to be a robust and meaningful index of groundwater drought. The maximum length of groundwater droughts defined by SGI is an increasing function of mmax, meaning that relatively long groundwater droughts are generally more prevalent at sites where SGI has a relatively long autocorrelation range. Based on correlations between mmax, average unsaturated zone thickness and aquifer hydraulic diffusivity, the source of autocorrelation in SGI is inferred to be dependent on aquifer flow and storage characteristics. For fractured aquifers, such as the Cretaceous Chalk, autocorrelation in SGI is inferred to be primarily related to autocorrelation in the recharge time series, while in granular aquifers, such as the Permo-Triassic Sandstones, autocorrelation in SGI is inferred to be primarily a function of intrinsic aquifer characteristics. These results highlight the need to take into account the hydrogeological context of groundwater monitoring sites when designing and interpreting data from groundwater drought monitoring networks.

The number of Building Companies that searches for a certification of their Quality Management Systems (SGQ’s) has risen in recent years, focused mainly on ISO 9001 and PBQP-H. The same trend has been occurring with regard to Management Systems of Environmental, Safety and Health at work in view of the current global socio economic outlook. Although companies suffer a number of difficulties during the process of implementation and certification of SGQ’s and several works in the literature have already studied about that subject, not much is known upon those difficulties of SGI’s (Integrated Management System) implementation, as well as the rate and reasons for dropouts implementation and recertification of those systems, especially in construction. Therefore, the present Study has as objective to evaluate the level potential of implementation of Integrated Management System (SGI’s) covering Quality, Environment, Safety and Health at work at Building Companies that have ever had a Quality Management System (SGQ) implemented. As methodological proposal, it was used the available literature for the preparation of a compatibility matrix among reference standards (ISO 9001, SiAC (2012), ISO 14001 and OHSAS 18001) and the checklist containing the items (requirements and criteria) most relevant to the implementation of SGI’s. Also were developed quantitative indicators which that do references for the degree of implementation of SGI's. The research was conducted in the form of study case, as a way of method validation, from ten civil work of a medium size Building Company. As results, some hypotheses initially proposed was validated in the research that there are significant differences in the implantation potential of SGI's in civil work in the same organization, in addition, it was possible to find that distinct phases of execution of civil works influence about the potential of adherence into implementation to one SGI. The indicators established show as an important tool for the building companies to mark out to achieve the SGI. The elaborate method contributes to the advancement in construction with regard to implementation and maintenance of SGQ’s and, preliminary, of SGI’s. Besides trying to contribute to a better understanding of the evolution of quality, environment, safety and health at work in construction, this Study longed to highlight the benefits and critical issues, not only for the implementation of SGI's, but also for their maintenance and viability in the sector.
O número de empresas construtoras que buscam uma certificação de seus sistemas de gestão da qualidade (SGQ’s) tem crescido nos últimos anos, focadas principalmente na ISO 9001 e no PBQP-H. A mesma tendência vem ocorrendo quanto aos sistemas de gestão ambiental e de segurança e saúde no trabalho tendo em vista o atual panorama sócio-econômico global. Embora as empresas sofram uma série de dificuldades durante o processo de implantação e certificação de SGQ’s e, diversos trabalhos na literatura já tenham estudado sobre este assunto, pouco ainda se sabe sobre as dificuldades de implantação de SGI’s (Sistemas de Gestão Integrados) e sobre o índice e os motivos de desistências de implantação e de recertificação destes sistemas, principalmente, em construtoras. Deste modo, o presente trabalho tem como objetivo avaliar o potencial de grau de implantação de SGI’s abrangendo Qualidade, Meio Ambiente e Segurança e Saúde no Trabalho em empresa construtora que já fosse detentora de um SGQ. Como proposta metodológica empregou-se literatura disponível para a elaboração de uma matriz de compatibilidade entre as normas de referência (ISO 9001, SiAC (2012), ISO 14001 e OHSAS 18001) e de uma lista de verificação contendo os itens (requisitos e critérios) mais relevantes à implantação de SGI’s. Também foram desenvolvidos indicadores que fazem referências quantitativas ao grau de implantação de SGI’s. A pesquisa foi realizada, sob a forma de estudo de caso, como forma de validação do método, a partir de dez obras de uma construtora de porte médio. Como resultados, foram validadas as hipóteses inicialmente propostas na pesquisa de que existem diferenças significativas de potencial de implantação de SGI’s em obras de uma mesma organização. Ainda foi possível constatar que fases de execução distintas de obras influenciam quanto ao potencial de aderência de implantação a um SGI. Os indicadores estabelecidos mostraram-se importante ferramenta para as construtoras se balizarem para alcançarem o SGI. O método elaborado contribui para o avanço na construção civil no que diz respeito à implantação e manutenção de SGQ’s e, preliminarmente, de SGI’s. Além de tentar contribuir para melhor compreensão da evolução da qualidade, do meio ambiente e da segurança e saúde no trabalho na construção civil, este trabalho almejou ressaltar os benefícios e os aspectos críticos não só para a implantação de SGI’s, mas também para sua manutenção e viabilidade no setor.

Os sistemas de gestão estão sendo cada vez mais usados e é possível constatar isso através do aumento do número de empresas certificadas em normas tais como ISO 9001, ISO 14001 e OHSAS 18001. Tem-se observado que ao mesmo tempo em que cresce o número de certificações, tem sido cada vez mais comum as organizações adotarem mais de uma norma e como consequência há a necessidade e uma tendência de se integrá-las. A integração traz diversos benefícios como melhora na tomada de decisão, diminuição do risco de se ter atividades redundantes e contraditórias, bem como melhor utilização de recursos, entre outros benefícios. Devido aos benefícios que a integração oferece, é importante haver um instrumento que avalie o grau de maturidade dos Sistemas de Gestão Integrados (SGI), para que através dessa informação se possa promover melhorias para atingir uma integração maior. Através de uma revisão na literatura, notou-se que não há trabalhos que realizem a avaliação da maturidade da integração de sistemas de gestão tomando como ponto de apoio o conceito de modelos de maturidade. Ao perceber essa lacuna, resolveu-se elaborar um instrumento que avalie o grau de maturidade do SGI na empresas. No desenvolvimento deste trabalho, foi feita uma revisão exploratória seguida de uma revisão bibliográfica sistemática sobre SGI, também foi realizada uma pesquisa bibliográfica sobre modelo de maturidade. Estas revisões, contribuíram para atingir o propósito principal desta dissertação que é a elaboração de uma ferramenta para avaliar o grau de maturidade utilizando o conceito de Modelos de Maturidade e tendo como ponto de apoio dois modelos já consolidados como CMMI - Capability Maturity Model Integration e o modelo de maturidade apresentado na ISO 9004, além das normas ISO 9001, ISO 14001, OHSAS 18001 e NBR 16001. A ferramenta desenvolvida recebeu a avaliação de especialistas acadêmicos e profissionais da indústria, e ela conta com 21 questões divididas em 4 áreas, sendo elas Política; Planejamento; Implementação/Execução; e Verificação/Ação. Como resultado alcançado pela dissertação, pode-se destacar a apresentação de um panorama geral sobre as pesquisas em SGI, bem como a ferramenta de avaliação do grau de maturidade da integração dos sistemas de gestão. Conclui-se que tem aumentado ao longo dos anos trabalhos relacionados a SGI e que o estudo sobre isso é importante tanto do ponto de vista do mercado como acadêmico.
Management systems have been largely used, which can be verified by the increase in the number of ISO 9001, ISO 14001 and OHSAS 18001 certifications. As the number of individual certifications increase, more companies have faced the challenge of integrating multiple management systems. Integration is important because it improves decision-making, reduces redundant and conflicting activities and promotes a better utilization of resources. Due to the benefits that integration offers, it is important to have an instrument to assess the degree of maturity of Integrated Management Systems (IMS). With such information it would be possible to plan improvements and reach a greater integration. By carrying out a literature review, it was possible to note that there is not a tool to evaluate the maturity degree of IMS that employs the concept of maturity models. This work presents a tool that assesses the maturity degree of an IMS. An exploratory review as well as a systematic literature review about IMS were performed. In addition, a literature review on maturity models was also conducted. These reviews helped achieve the main purpose of this dissertation that it is the development of a tool to assess the maturity level using the concept of Maturity Models and having as support two well-established models: CMMI - Capability Maturity Model Integration and the maturity model presented in ISO 9004, alongside ISO 9001, ISO 14001, OHSAS 18001 and NBR 16001 standards. The tool has 21 questions divided into 4 areas: (i) policy, (ii) planning, (iii) implementation / execution and (iv) verification / action. Afterwards, the tool was assessed by academic experts and industry professionals. An overview of the research into IMS, as well as the maturity models oriented towards evaluating the integration of management systems can be pointed out as relevant results of this research. Another important finding is that the number of IMS-related studies has increased over the past years and such studies are considered important from both market and academic standpoints.

Atualmente, é imperioso que as empresas assegurem a sua competitividade e expansão, num contexto global e de mudança, através da melhoria contínua dos seus produtos e processos. A inovação é um fator de diferenciação à disposição das empresas, quer para melhorar os seus produtos e/ou serviços, quer para aumentar a sua eficácia operacional, o reforço da sua competitividade e consequentemente, aumentar a satisfação do cliente, indo ao encontro dos seus requisitos. O trabalho apresentado neste relatório, consistiu no estudo e elaboração dos procedimentos inerentes à implementação da norma NP 4457:2007, no sistema de gestão integrado (SGI) da empresa “C”, uma indústria vidreira nacional, produtora de vidro de embalagem. Neste trabalho são abordadas as normas NP EN ISO 9001:2008, NP EN ISO 14001 e OHSAS 18001, que fazem parte do sistema integrado de gestão da empresa, sendo que estas normas partilham de princípios e técnicas de gestão comuns e facilitadores da integração da nova norma (NP 4457:2007). A NP EN ISO 9001:2008 e a NP 4457:2007, serviram de referência a todo o trabalho desenvolvido, ao longo deste projeto. No sentido da melhoria contínua e da satisfação do cliente, a empresa “C”, apesar de já possuir outros sistemas de gestão, sentiu a necessidade de juntar a NP 4457:2007 ao seu SGI, de forma a dar suporte ao cumprimento das normas que dele fazem parte. Este processo mostrou-se bastante facilitado, pelo facto de a maior parte das atividades inerentes à integração já estarem feitas e fazerem parte das rotinas organizacionais. Apesar da dificuldade em inovar, neste setor, porque os projetos vêm já desenvolvidos pelos clientes ou pelo grupo industrial no qual a empresa se insere, a integração foi conseguida com sucesso, trazendo consigo alguns benefícios já identificados, como o fortalecimento das relações com o cliente e a diminuição de custos, devido à redução da duplicação de processos e procedimentos. No entanto, esta incapacidade em inovar, levou ao abandono dos procedimentos implementados, desperdiçando assim as boas práticas desta norma.
Currently, it is imperative that companies ensure their competitiveness and expansion, in a global context and change through continuous improvement of its products and processes. Innovation is a factor of differentiation at the disposal of companies, either to improve their products and/or services, or to increase its operational effectiveness, strengthen its competitiveness and consequently, increase customer satisfaction, in order to meet their requirements. The work present in this report, consisted in the study and elaboration of the procedures involved in the implementation of the standard NP 4457:2007, in the integrated management system (IMS) of company “C”, a national industry, glass producer packing. In this work the standards NP EN ISO 9001:2008, NP EN ISO 14001 and OHSAS 18001 are addressed, that are part of the integrated system of management of the company, and these standards share common management principles and techniques and facilitators of integration of the new standard (NP 4457:2007). NP EN ISO 9001:2008 and the NP 4457:2007, served as a reference to all the work developed, throughout this project. In order to a continuous improvement and customer satisfaction, company “C”, even though having already other management systems, felt the need to join the standard NP 4457:2007 to their IMS, in order to support the observance of which it is part of. This process was quite easy, by the fact that most of the activities inherent in the integration are already made and being part of organizational routines. Despite the difficulty in innovating in this sector, because the designs already come developed by customers or by the industrial group in which the company is inserted, integration was achieved successfully, bringing with it some benefits already identified, such as the strengthening of relations with the customer and the cost reduction, due to the reduction of duplication of processes and procedures. However, this inability to innovate, led to the abandonment of the implemented procedures, wasting the good practice of this standard.

Currently in the corporate world grows the focus for the speed and intensity with which the process changes, mainly driven by frequent technological innovations, the performance of the economy and strong competition established in its market. Given this challenging scenario, there is the continuing need to identify new techniques, methods and procedures, aimed at the main goal of the strategy, improve performance, competitiveness and leverage their organizational management. Thus, the relentless pursuit of continuous improvement in organizations, demand the implementation of integrated management systems, systemic and measurable as a facilitator in reducing vulnerability to the impact caused by competitive forces, which so conducive to the strengthening of the value chain business. Therefore, this work will be developed based on the activities of CVI Soft Drinks industry Ltda., Franchised The Coca-Cola Company and Heineken Brazil, with factory in Rio Grande Central Region South, with the main objective to propose a Model for Performance Measurement Implementation and Enforcement in an Integrated Management System SGI. Methodologically, a case study will be conducted in order to identify the adherence of the Integrated Management System - SGI in relation to the full implementation of the concepts of Strategic Management, Process Management and requirements of International Standards Management System Certification, to obtain the end a performance measurement system model that facilitates the measurement and analysis of organizational continuous improvement.
Atualmente no mundo corporativo cresce o foco pela rapidez e intensidade com que se processam as mudanças, impulsionadas principalmente pelas frequentes inovações tecnológicas, pelo desempenho da economia e pela forte concorrência estabelecida no seu mercado de atuação. Diante desse cenário desafiador, surge a contínua necessidade de identificar novas técnicas, métodos e procedimentos, visando como principal meta da estratégia, melhorar o desempenho, a competitividade e a alavancagem da sua gestão organizacional. Desta forma, a busca incessante pela melhoria contínua nas organizações, demanda a implementação de sistemas de gestão integrados, sistêmicos e mensuráveis como meio facilitador à redução da vulnerabilidade em relação ao impacto causado pelas forças competitivas, os quais propiciem assim o fortalecimento da cadeia de valor do negócio. Metodologicamente, será realizado um estudo de caso com a finalidade de identificar a aderência do Sistema de Gestão Integrada SGI em relação à aplicação integral dos conceitos de Gestão Estratégica, Gestão de Processos e Requisitos das Normas Internacionais de Certificação de Sistemas de Gestão, visando obter ao final um modelo de sistema de medição de desempenho que facilite a mensuração e a análise da melhoria contínua organizacional.

Em função dessas mudanças as organizações devem buscar meios para melhorar o seu desempenho e garantir a sua sobrevivência, principalmente no que tange ao fator concorrência. Dessa forma, é necessário que as organizações otimizem o uso dos seus recursos para que possam tornar-se mais eficientes e competitivas. A implantação dos sistemas de gestão da qualidade, de gestão ambiental, e de gestão de segurança e saúde ocupacional em único Sistema de Gestão Integrado (SGI) tem contribuído para alcançar a melhoria da qualidade dos produtos e serviços, favorecendo a proteção ao meio ambiente e a avaliação dos perigos e riscos relacionados à segurança e saúde do trabalhador, que são requisitos importantes nos sistemas de gestão das organizações. Nesse contexto, procurou-se investigar e analisar os resultados obtidos com a implantação e implementação do Sistema de Gestão Integrado (SGI), relacionado à Gestão da Qualidade, Gestão Ambiental e Gestão de Segurança e Saúde Ocupacional, em uma Empresa Prestadora de Serviços que atua no ramo de Manutenção e Montagens Industriais, situada na região do Vale do Paraíba Paulista, Estado de São Paulo. Para tanto, foram utilizadas as pesquisas bibliográfica, exploratória e documental ao estudo de caso. Verificou-se que a implantação do sistema de gestão integrado foi uma estratégia de negócios para a Empresa que, além de conquistar os seus objetivos, adquiriu boas práticas de responsabilidade social e contribuiu para a sua sobrevivência. Além disso, houve melhoria no desempenho, na eficiência e um crescimento de forma organizada e ordenada, que favoreceu o aumento da sua participação no ambiente competitivo.
In the business competitive scenario changes happen fast and constant. Because of these changes, organizations must seek ways to improve their performance and ensure its survival, especially in regard to the competition factor. Thus, it is necessary for organizations to optimize the use of its resources to enable them to become more efficient and competitive. The deployment of quality management system, environmental management and occupational, health and safety management in a single Integrated Management System (IMS) has helped to achieve improved quality of products and services, encouraging environmental protection and hazard assessment and risks related to safety and health of workers, which are important requirements in the management of organizations. In this context, it was sought to investigate and analyze the results achieved with the implementation of the Integrated Management System regarded to Quality, Environmental and Occupational, Health and Safety Management in a service company, located in the Vale do Paraíba Paulista, Sao Paulo State. To this end, it was used the research literature, documentary and exploratory case study. It was found that the implementation of integrated management system was a business strategy for the company, which not only achieved their objectives, but also acquired good social responsibility practices and contributed to its survival. Besides that, there was improvement in performance, efficiency and growth in an organized and orderly way, which favored the increase of its participation in the competitive environment.

Resumo: No mundo dos negócios acontecem mudanças constantemente. Atrelada a estas mudanças está a sobrevivência organizacional, principalmente no que tange ao fator competitividade. Conseqüentemente, é fundamental que haja otimização do uso dos recursos disponíveis dentro de cada organização, tornando-o mais eficiente. Sendo assim, torna-se importante e necessária a integração máxima entre os diversos sistemas de gestão co-existentes dentro das organizações: gestão da qualidade, ambiental, da segurança e saúde ocupacional, financeira, de pessoas, estratégica, da responsabilidade social, entre outras. Com estes sistemas alinhados e integrados, as organizações tendem a maximizar sua eficiência com redução de custos, assegurando sua sobrevivência. Este trabalho, portanto, tem o objetivo de desenvolver e propor uma metodologia de avaliação da maturidade dos sistemas de gestão dentro das organizações, bem como do nível de integração entre eles, nas dimensões da qualidade: do produto, ambiental, da segurança e saúde ocupacional e ética e social. Com isto é possível identificar as oportunidades de melhorias na integração das diversas dimensões da gestão e de fato buscar a maximização da eficiência no uso dos recursos disponíveis
Abstract: In the business world changes happen constantly. Linked to these changes is the organization survival, mainly as for the competitiveness factor. Consequently, it is fundamental to have the otimization of the use of the resources available inside each organization, becoming it more efficient. So, it becomes important and necessary the maximum integration among the several management systems co-existant inside the organizations: quality management, environrnental, occupational health and safety, financial, people, strategic, social accountability, among others. With these systems aligned and integrated, the organizations tend to maximize their efficiency with cost reduction, assuring their survival. This paper, therefore, has the aim of developing and proposing a methodology for evaluation of the maturity of the management systems inside the organizations as well as the integration of them, in the dimensions of quality: of the product, environrnental, of the occupational health and safety and ethical and social. With this, it is possible to identify the opportunities for improvements in the integration of several management dimensions and, in fact, to search the maximization of the efficiency in the use of the available resources

Antagonism of the activin receptor signaling pathway represents a promising potential therapy for the muscular dystrophies and other muscle wasting disorders (i.e., cachexia or sarcopenia). Previous research has shown that antagonism of activin signaling promotes muscle growth, attenuates muscle wasting, and restores function in both wild type and diseased animals. Our laboratory has recently developed a novel small molecule (SGI-1252) that inhibits activin downstream (i.e., Smad2/3 phosphorylation) signaling. Purpose: In this study we determined how eight weeks of orally administered SGI-1252 affected TGF-ß signaling, whole body mass, individual limb muscle mass, and muscle fiber cross sectional area (CSA). Methods: Wild-type (WT) mice were treated with SGI-1252 or a vehicle control (VC) via oral gavage (400 mg/kg 3 times per week) for 8 weeks. Body mass was measured twice per week during the 8-week treatment period. At the end of the treatment period, gastrocnemius and tibialis anterior (TA) muscles were excised, weighed, and prepared for histological and biochemical analyses. Results: Following 8 weeks of treatment, there was no difference in weight gain between SGI-1252 (24.8 ± 1.8g) and VC treated mice (23.2 ± 1.5g) (p = 0.06). Gastrocnemius whole muscle mass was significantly greater in the SGI-1252 treated group relative to the VC treated mice (139.6 ± 12.8 mg vs 128.8 ± 14.9 mg) (p = 0.04), although when normalized with body mass there was no difference in gastrocnemius mass. For the TA muscle, there were no significant differences in whole muscle mass between SGI-1252 and VC groups, yet TA muscles in the SGI-1252 treated group had a reduced muscle fiber CSA compared to controls (621 ± 44 µm2 vs 749 ± 36 µm2) (p = 0.0005). There was a statistical trend of decreasing Smad2 phosphorylation in the SGI-1252 treated TA muscles (mean SGI-1252 = 0.668 vs VC = 0.848) (p = 0.06), and no significant differences in Smad2 phosphorylation in the gastrocnemius. Conclusions: Contrary to our hypothesis, 8 weeks of orally administered SGI-1252 was not effective in promoting increases in whole body mass, limb whole muscle mass, or myofiber cross sectional area. This may be due to the inability of SGI-1252, at the administered dose, to effectively decrease signaling downstream of the activin receptor. Clearly, studies using a wider range of doses and delivery methods will be needed to ascertain the efficacy of SGI-1252 as a potential therapeutic.

Resumen
ADI (Austempered Ductile Iron) - a bainitic ductile iron in which the resulting structure is obtained by the isothermal decay of austenite in the bainite region is currently a very promising material, which has a very wide application in various industries. It is characterized by a particularly favorable combination of strength and ductility, which exceeds the properties of heat-treated steels in the strength range of 800 - 1500 MPa. Its production is problematic because the ferritic or ferritic-pearlitic SGI (spheroidal graphite iron), which is a precursor in the preparation of ADI cast iron, has to meet demanding metallurgical properties, especially as many regular graphite globules evenly distributed in the matrix. The aim of this article is to present a method of metallurgical processing of SGI in such a way as to ensure the quality of the melt required for the preparation of ADI cast iron with respect to elimination of environmental impacts, i.e. inmold modification, as traditional SGI production causes exothermic reaction. work environment. Another possibility to influence the metallurgical quality of SGI cast iron is the action a magnetic field acting on the solidifying melt of SGI, which leads to improvement the metallurgical as well as mechanical properties of SGI.

Resumen
The aim of this project was to develop an impact framework for the Science Gallery Network (SGN). This work was commissioned by the Science Gallery International (SGI). The SGN has eight member organisations across four continents: Dublin, London, Melbourne, Bengaluru, Detroit, Rotterdam, Atlanta and Berlin. Whilst the network consistently sees unprecedented levels of accomplishment by its members, a testimony to their capacity, innovation and vision, the SGN does not have a systematic way to measure and monitor this impact. An impact framework that can assist with understanding and reporting the value of this impact will provide important recognition that the SGN has achieved what it sets out to do— bringing science, art, technology and design together to deliver world-class educational and cultural experiences for young people. This report details the robust consultation approach that was undertaken by the University of Melbourne’s project team—one that included a desktop review, focus group discussions, surveys and interviews—to ensure multiple perspectives were gathered on what could be considered a multi-faceted concept. The desktop review provided a thorough review and an environmental scan of the impact literature and its measurement. In addition, the focus group discussions and interviews provided a rich understanding of what ‘good impact’ means for the SGN and the implications of this to the measurement of impact outcomes. Five key recommendations are provided and summarised. Note that these key recommendations should be taken as a point of departure for further in-depth consultation throughout the wider SGN.
