Resumen

Este trabalho visou verificar a precisão de aplicativos de smartphone para medir indiretamente a altura de árvores e a distância entre o operador e a árvore. Como testemunha para medir altura, adotou-se o Hipsômetro Vertex IV (T0a) e, para distância, a variável medida com trena (T0b). Foram selecionadas 30 árvores isoladas do Campus da Universidade Federal do Paraná em Curitiba - PR e 30 árvores pertencentes à um plantio de Eucalyptus sp. na Fazenda Experimental Canguiri em Pinhais – PR. Foram 7 tratamentos testados para altura: Smart Measure (T1a), sMeasure (T2a), Distance meter (T3a), Hypsometer (T4a), 3D-Prumo (T5a), Height and Distance (T6a) e Tree-H (T7a). Foram 8 tratamentos para medir distância: Smart Measure (T1b), sMeasure (T2b), Distance meter (T3b), Hypsometer (T4b), 3D-Prumo (T5b), Height and Distance (T6b), Easy Measure (T7b) e Smart Distance (T8b). Avaliou-se a precisão dos aplicativos com o teste t de Student pareado com 5% de significância em relação às testemunhas e cálculo dos erros. Para altura em árvores isoladas, o tratamento T2a apresentou diferença estatística a probabilidade de 5%. O tratamento T6a obteve menor erro (2,48 m). No plantio de Eucalyptus sp., os tratamentos T1a, T6a e T7a apresentaram diferenças significativas e T3a obteve menor erro (2,99 m). Para a medição de distância em árvores isoladas, os tratamentos T4b e T6b não apresentaram diferença e T4b apresentou menor erro (2,28 m). No plantio, todos apresentaram diferença estatística. A incorporação de aplicativos para smartphones é alternativa para uso em inventários florestais para alturas. Para distâncias, deve-se ter cautela.

Resumen

Five compounds of mono para-hidroxy chalcon were synthesized (TC1, TC2, TC3, TC4, and TC5) and tested their cytotoxicity against HeLa cell and Raji cell. The difference in substituent of TC1 (R4 =H), TC2 (R4 = OCH3), and TC3 (R4 = F), showed the difference of their citotoxicity against HeLa cell. The citotoxicity of TC1 (LC50 = 16.08 µg/mL) ≈ TC3 (LC50 = 13.37 µg/mL), but the substituent difference of TC2 (LC50 = 147.43 µg/mL), decreasing it citotoxicity 10 times. Like wise their citotoxicity against Raji cell of TC1 (LC50 = 36.44 µg/mL) ≈ TC3 (LC50 = 30.46 µg/mL), but the substituent difference of TC2 (LC50 = 468.94 µg/mL), decreasing it citotoxicity activity 15 times. Nevertheless the strength of citotoxicity TC4 (LC50 = 98.74 µg/mL) and TC5 (LC50 = 110.97 µg/mL) against Raji cell are stronger than the citotoxicity of two of them against HeLa cell (LC50 of TC4 = none, LC50 of TC5 = 576.53 µg/mL). Resumen

The objective of this study is to isolation and agonistic test ability of Trichoderma spp. againts Sclerotium rolfsii Sacc. cause of wilting on pepper plants and has been conducted in Pathogenicity Laboratory Faculty of Agriculture Unpatti. The study use 5 treatment of isolate Trichoderma spp. (Tc3, Tc4, Tc5, Tc6 and Tc7) with 3 replications so that there are 15 experimental units. The results showed that the five isolates Trichoderma spp. has an antagonistic power to S. rolfsii with an average percentage of inhibition of S. rolfsii of 26,01%. Percentage of inhibition bolth of isolate ware not significantly different at 95% level test results between treatment. Average percentage inhibition of S. rolfsii by Trichoderma spp. each treatment was Tc6 = 27,31%, Tc3 = 26,63%, Tc5 = 26,05%, Tc7 = 25,69% and Tc4 = 24,37%. Keywords: antagonism, Trichoderma spp., Sclerotium rolfsii  Abstrak Penelitian ini bertujuan mengisolasi dan menguji kemampuan antagonis Trichoderma spp. terhadap Sclerotium rolfsii Sacc. penyebab layu pada tanaman cabai dan telah dilaksanakan di Laboratorium Patogenisitas Fakultas Pertanian Unpatti, dengan menggunakan 5 perlakuan isolat Trichoderma spp. (Tc3, Tc4, Tc5, Tc6 dan Tc7) dengan 3 ulangan sehingga terdapat 15 satuan percobaan. Hasil penelitian menunjukkan bahwa kelima isolat Trichoderma sp. mempunyai daya antagonis terhadap S. rolfsii dengan rata-rata persentase penghambatan S. rolfsii sebesar 26%. Hasil analisis varians pada taraf 95% menunjukkan tidak ada perbedaan nyata antara perlakuan. Rata-rata persentase penghambatan S. rolfsii oleh Trichoderma spp. masing-masing perlakuan berturut-turut adalah Tc6 = 27,31%, Tc3 = 26,63%, Tc5 = 26,05%, Tc7 = 25,69% dan Tc4 = 24,37%, dengan rata-rata 26,01%. Kata kunci: antagonisme, Trichoderma spp., Sclerotium rolfsii

Resumen

Nine families of bread wheat (TC5, TC6, TC7, TC8, TC9, TC10, TC14, 5395-(243AA), and 5395) with resistance to barley yellow dwarf virus and containing putative translocations between wheat and a group 7 chromosome of Agropyron intermedium (L1 disomic addition line, 7Ai#1 chromosome) induced by homoeologous pairing or tissue culture were analyzed. C-banding, genomic in situ hybridization (GISH), and restriction fragment length polymorphism (RFLP) in combination with repetitive Agropyron-specific sequences and deletion mapping in wheat were used to determine the relative locations of the translocation breakpoints and the size of the transferred alien chromatin segments in hexaploid wheat–Agropyron translocation lines. All homoeologous compensating lines had complete 7Ai#1 or translocated 7Ai#1–7D chromosomes that substitute for chromosome 7D. Two complete 7Ai#1 (7D) substitution lines (5395-(243AA) and 5395), one T1BS–7Ai#1S∙7Ai#1L addition line (TC7), and two different translocation types, T7DS–7Ai#1S∙7Ai#1L (TC5, TC6, TC8, TC9, and TC10) and T7DS∙7DL–7Ai#1L (TC14), substituting for chromosome 7D were identified. The substitution line 5395-(243AA) had a reciprocal T1BS∙1BL–4BS/T1BL–4BS∙4BL translocation. TC14 has a 6G (6B) substitution. The RFLP data from deletion mapping studies in wheat using 37 group 7 clones provided 10 molecular tagged chromosome regions for homoeologous and syntenic group 7 wheat or Agropyron chromosomes. Together with GISH we identified three different sizes of the transferred Agropyron chromosome segments with approximate breakpoints at fraction length (FL) 0.33 in the short arm of chromosome T7DS–7Ai#1S∙7Ai#1L (TC5, TC6, TC8, TC9, and TC10) and another at FL 0.37 of the nonhomoeologous translocated chromosome T1BS–7Ai#1S∙7Ai#1L (TC7). One breakpoint was identified in the long arm of chromosome T7DS∙7DL–7Ai#1L (TC14) at FL 0.56. We detected some nonreciprocal translocations for the most proximal region of the chromosome arm of 7DL, which resulted in small duplications. Resumen

Abstract Six solid phases of [Mn(DMSO)6](ClO4)2 have been detected by differential scanning calorimetry. The phase transitions were found between the following solid phases: stable KIc ↔ stable KIb at TC5 = 225 K, metastable KIII ↔ metastable KII at TC4 = 322 K, stable KIb ↔ stable KIa at TC3 = 365 K, metastable KII↔overcooled K0 at TC2 = 376 K and stable KIa→stable K0 at TC1 = 379 K. The title compound melts at Tm = 488 K.

Twenty-seven L-shaped ortho-quinone analogs were designed and synthesized using a one pot double-radical synthetic strategy followed by removing methyl at C-3 of the furan ring and introducing a diverse side chain at C-2 of the furan ring. The synthetic derivatives were investigated for their cytotoxicity activities against human leukemia cells K562, prostate cancer cells PC3, and melanoma cells WM9. Compounds TB1, TB3, TB4, TB6, TC1, TC3, TC5, TC9, TC11, TC12, TC14, TC15, TC16, and TC17 exhibited a better broad-spectrum cytotoxicity on three cancer cells. TB7 and TC7 selectively displayed potent inhibitory activities on leukemia cells K562 and prostate cancer cells PC3, respectively. Further studies indicated that TB3, TC1, TC3, TC7, and TC17 could significantly induce the apoptosis of PC3 cells. TC1 and TC17 significantly induced apoptosis of K562 cells. TC1, TC11, and TC14 induced significant apoptosis of WM9 cells. The structure-activity relationships evaluation showed that removing methyl at C-3 of the furan ring and introducing diverse side chains at C-2 of the furan ring is an effective strategy for improving the anticancer activity of L-shaped ortho-quinone analogs.

Resumen

Objective. Trichloroacetic acid (TCAA), or trichloroethanoic acid, is a chemical analogue of acetic acid where three methyl group hydrogen atoms are replaced by chlorine. TCAAs are also abbreviated and referred to as TCAs, causing confusion with the psychiatric antidepressant drug class, especially among patients. TCAAs exist in dermatological treatments such as chemical peels or wart chemoablation medication. TCAA ingestion or overdose can cause gastric irritation symptoms including vomiting, diarrhea, or lassitude. This symptomatology is less severe than TCA overdose, where symptoms may include elevated body temperature, blurred vision, dilated pupils, sleepiness, confusion, seizures, rapid heart rate, and cardiac arrest. Owing to the vast difference in symptoms, the need for clinical intervention differs greatly. While overdose of either in a self-harm attempt can warrant psychiatric hospital admission, the risk of death in TCAA ingestion is far less. Case Report. A patient ingested TCAA in the form of a commercially available dermatological chemical peel as a self-harm attempt, thinking that it was a more injurious TCA. Conclusion. Awareness among physicians, particularly psychiatrists, regarding this relatively obscure chemical compound (TCAA) and its use by suicidal patients mistakenly believing it to be a substance that can be significantly more lethal (TCA), is imperative.

AbstractThe growth, gut ecology and immunocompetence of Oreochromis niloticus and the resistance to Aeromonas hydrophila were investigated after been fed with diets containing dietary Curcumin longa for 12 weeks. Diets were formulated to contain 30% crude protein with diet TC1, TC2, TC3, TC4 and TC5 having 0% (control), 0.25%, 0.5%, 0.75% and 1.0% turmeric powder, respectively. Diets were allotted to groups of O. niloticus (mean weight of 1.29± 0.15 g) and replicated thrice for 84 days. Results showed that the highest mean final weight (4.79±0.04 g) was obtained in TC3 and corresponded to the treatment with the highest feed intake. A significantly high (p<0.05) specific growth rate (SGR) was observed in TC3 (0.73±0.03 %day−1) while TC4 (0.57±0.02 %day−1) gave the lowest value. The highest microbial load in the gut was observed in TC1 groups and the least in TC4 groups. Red blood cell count, hemoglobin, packed cell volume did not show significant variation (p>0.05) across treatments. However, white blood cell (WBC) count was significantly higher in TC1 (control). There was an improved immunocompetence, as aspartate aminotransferase (AST) progressively reduces in fish fed supplements. Similarly, there was a better oxidative response in the treated groups with reduced hydrogen peroxidase, increased total protein and glutathione peroxidase. Mortality ranged from 25% in TC4 to 95% in TC1 after the challenge test with A. hydrophila. This study showed that C. longa inclusion at 0.5% is more beneficial when growth and health status of O. niloticus juveniles are considered.

Syfte – Examensarbetets syfte är att skapa förståelse kring ett outsourcingbeslut genom att utveckla en beslutsmodell för outsourcing. För att uppnå syftet ska följande frågeställningar besvaras: Vilka faktorer bör påverka ett outsourcingbeslut? Vilka kostnader bör analyseras för att ett outsourcingbeslut ska bli väl avvägt? Vilka komponenter bör ingå i en beslutsmodell för outsourcing? Metod – Examensarbetet bygger på en analytisk konceptuell studie med ett deduktiv och induktiv förhållningssätt, där påverkansfaktorer togs fram genom litteraturstudier. Jämförelser och analyser av redan befintliga teorier gjordes för att identifiera påverkansfaktorer, utifrån dessa faktorer skapades en beslutsmodell för outsourcing. Resultat – Resultatet från studien visar att det finns ett flertal olika faktorer som påverkar ett outsourcingbeslut och kan sammanfattningsvis placeras som påverkansfaktorer, strategiska- och konkurrensfaktorer samt kostnadsfaktorer. Den utvecklade beslutsmodellen för outsourcing består av sju steg där varje steg innefattar olika analyser och beslut, de sju stegen baseras på de faktorer som bör påverkas vid outsourcing. Förslag till fortsatta studier – För att utveckla beslutsmodellen skulle fler teorier och faktorer som möjligen kan beröras i samband med outsourcing adderas. En ytterligare studie skulle kunna vara att utveckla ett verktyg där organisationen kan gradera hur organisationen förhåller sig till de faktorer som påverkas vid ett outsourcingbeslut och därmed utöka beslutsunderlaget. Praktiska konsekvenser – För att underlätta för beslutsfattaren och få en helhetsbild över faktorer och kostnader som påverkas vid outsourcing kan beslutsmodellen användas. Originalitet – Det finns enligt författarnas vetskap ingen fullständig beslutsmodell som belyser alla faktorer som kan påverkas vid ett outsourcingbeslut. Purpose – The purpose of the thesis is to create understanding of the outsourcing decision making by developing a decision model for outsourcing. To achieve this, the following questions are answered: What factors should influence the outsourcing decision? What costs should be analyzed so that an outsourcing decision should be balanced? What components should be included in a decision model for outsourcing? Method – The thesis is based on an analytical conceptual study with a deductive and inductive approach, where the influencing factors was generated through literature reviews. Comparison and analysis of existing theories were made to identify the influencing factors, based on the generated influencing factors a decision model for outsourcing was created. Findings – Results from the study shows that there are several factors that influence the outsourcing decision and can in summary be positioned as strategic and competitive factors and cost factors. The developed decision model for outsourcing consists of seven steps, where each step includes various analyzes and decisions, the seven steps are based on the factors that should be affected by outsourcing. Research limitations/implications – The results in this thesis are based only on theory. The model can still be generalized by being adaptable for different Organization. Practical implications – Decision model can be used to aid and support in an outsourcing decision. To facilitate the decision-maker and get an overview of the factors and costs that are affected by the outsourcing decision model can be used. With the decision model, the organization can analyze factors and costs to obtain a comprehensive decision-making. Originality – Since many organizations are making outsourcing decision wrongly without taking into account all the factors affected by the outsourcing decision. There is, in the authors' knowledge no complete decision model that highlights all the factors that may be affected by an outsourcing decision.

Resumen

A determinação precisa e rápida das configurações de equilíbrio Magnetohidrodinâmico (MHD) em tokamaks é de fundamental importância para o confinamento magnético do plasma. Através do conhecimento dos parâmetros que caracterizam este equilíbrio MHD é possível controlar o plasma durante a sua formação por processos de realimentação. Uma análise mais detalhada destes parâmetros é necessária, também, entre um disparo e outro, para a estruturação do experimento. Neste trabalho é investigada a reconstrução das configurações de equilíbrio MHD no tokamak TCA/BR a partir de medidas magnéticas externas, utilizando um método que permite uma rápida determinação dos parâmetros principais da descarga. A tese divide-se em duas partes. Na primeira, é apresentada a construção de um código de equilíbrio que resolve a equação de Grad-Shafranov para a configuração geométrica que caracteriza o tokamak TCA/BR. Na segunda, é descrito o processo de reconstrução do equilíbrio MHD através de medidas de campos e fluxos magnéticos externos ao plasma no TCA/BR, e utilizando o método de Função de Parametrização FP. Este método baseia-se no tratamento estatístico de um banco de dados simulados de configurações de equilíbrio, com o objetivo de obter uma expressão simples relacionando os parâmetros que caracterizam o equilíbrio e as medidas realizadas. Os resultados obtidos através da FP são comparados com os obtidos através de outros métodos convencionais. The accurate and rapid determination of the Magnetohydrodynamic (MHD) equilibrium configuration in tokamaks is a fundamental subject for the magnetic confinement of the plasma. With the knowledge of characteristic plasma MHD equilibrium parameters it is possible to control the plasma position during its formation using feed-back techniques. It is also necessary an on-line analysis between successive discharges to program external parameters for the subsequent discharges. In this work it is investigated the MHD equilibrium configuration reconstruction of the TCA/BR tokamak from external magnetic measurements, using a method that is able to determine fastly the main parameters of discharge. The thesis has two parts. Firstly it is presented the development of an equilibrium co de that solves de Grad-Shafranov equation for the TCA/BR tokamak geometry. Secondly it is presented the MHD equilibrium reconstruction process from external magnetic field and flux measurements using the Function Parametrization FP method. This method is based on the statistical analysis of a database of simulated equilibrium configurations, with the goal of obtaining a simple relationship between the parameters that characterize the equilibrium and the measurements. The results from FP are compared with conventional methods.

This PhD project explored the role of tricarboxylic acid (TCA) cycle enzymes in regulating Chinese hamster ovary (CHO) cell metabolism with respect to growth and recombinant protein expression. It was hypothesised that regulation of central metabolism can influence CHO cell productivity in terms of biomass and protein production. Thus the aim of these studies was to identify the key metabolic reactions of the TCA cycle associated with growth and protein expression in CHO cells. The gene expression of all TCA cycle genes was independently knocked-down using RNAi technology. The small interfering RNA (siRNA) mediated silencing of 11 TCA cycle genes significantly reduced cellular growth along with a decline in adenylate energy charges and an increase in catabolic reduction charges. The gene profiling of glucose and amino acid metabolism (not targeted by siRNA) suggested siRNA mediated knock-down of targeted TCA cycle genes led to cellular stress along with an enhanced rate of glycolysis leading to channelling of glucose for the generation of pyruvate. For the purpose of estimating intracellular metabolites, quenching and extraction method using ammonium bicarbonate and methanol was optimised to use with UCB CHO-K1 cell line and static transient siRNA transfections. A gas chromatography-mass spectrometry (GC-MS) analysis post-silencing of the aconitase gene, which catalyses the conversion of citrate to isocitrate in the TCA cycle, yielded higher MS peak intensities of at least four metabolites (gluconic acid, lysine, threonine and leucine) 72 h post-transfection in comparison to the controls. Transient knock-down of gene expression of seven TCA cycle genes in a recombinant stable cell line (expressing a rabbit monoclonal antibody) reduced cellular growth and altered the energy charges leading to a decline in antibody expression. Although silencing of the pyruvate dehydrogenase E1 gene, which is the component of the pyruvate dehydrogenase complex connecting glycolysis to the TCA cycle, did not affect cell viability, a reduction in antibody expression was recorded. Seven TCA cycle genes which demonstrated the most significant effect on cellular growth and energy charges were transiently over-expressed along with a monoclonal antibody in CHO-K1 cells with addition of their corresponding preceding intermediates. No differences in protein expression and cell specific productivity were observed compared to the control transfections. These results could be due to limitations of the effects of transient transfections for enhancing the metabolic activity of CHO cells. The aconitase gene demonstrated the most significant effect on CHO cell growth and proliferation in this study, therefore this gene was proposed as a novel selection marker for a metabolic selection system for the generation of recombinant therapeutics. This PhD project also established metabolite analysis tools and siRNA protocols for future metabolomic studies for investigating the intracellular CHO metabolism. The findings validated the hypothesis that TCA cycle plays an important role in CHO cell growth and recombinant protein production. The key metabolic genes affecting cellular growth and altering energy metabolism can be further explored for generation of an energy efficient CHO host-cell line (by over-expression of key TCA cycle genes) for enhanced recombinant protein production.

De 2007 a 2015, foram realizadas 112 apresentações de 104 espetáculos de várias linguagens artísticas. A pesquisa para esta dissertação foi realizada utilizando-se dos registros do Sistema TCA, relatórios institucionais, pesquisas acadêmicas e entrevistas com os atores envolvidos, desde os principais gestores da Secult, aos artistas, produtores e público em geral. O objetivo foi analisar o projeto enquanto instrumento de políticas públicas de cultura e verificar a sua importância para cena e as políticas culturais baianas. A dissertação foi estruturada em três capítulos, de forma que, no primeiro momento, sejam abordados temas conceituais da área da cultura, passando depois ao estudo do Teatro Castro Alves em si e do projeto Domingo no TCA. The dissertation Cultural policies, democratization and access to culture: Domingo no TCA has as object of study the Domingo no TCA (Sunday at TCA) project, which seeks to provide a greater democratization of access to the Castro Alves Theater (TCA) and its events. Created in a context of changes in federal and state public policies, the project is being carried out for almost ten years by the TCA, Bahia's largest theater, linked to the Secretariat of Culture (Secult) and the Bahia State Cultural Foundation (Funceb). The Domingo no TCA presentations take place at the theater's Main Hall, which can accommodate up to 1,554 spectators, with tickets costing a symbolic price of R$ 1.00 (full price). Monthly, on Sunday mornings, they have been attracting a large and eclectic audience consisting of adults, children and the elderly, from all kinds of neighborhoods of Salvador. From 2007 to 2015, there were 112 presentations of 104 different shows from several art forms. The research for this dissertation was carried out using the records of the TCA System, institutional reports, academic research and interviews with the people involved, from the main managers of Secult, to the artists, producers and the public in general. The objective was to analyze the project as an instrument of public policies of culture and to verify its importance for the cultural scene and policies of Bahia. The dissertation was structured in three chapters, such that in the first moment conceptual themes related to culture were approached, then going to the study of the Castro Alves Theater itself and the Domingo no TCA project. La disertación Políticas culturales, democratización y acceso a la cultura: El Domingo en TCA tiene como objeto de estudio el proyecto Domingo no TCA (Domingo en TCA), que busca una mayor democratización del acceso al Teatro Castro Alves (TCA) y su programación. Creado en un contexto de cambios en las políticas públicas federales y estatales, el proyecto es realizado desde hace casi diez años por el TCA, el teatro más grande de Bahía, vinculado a la Secretaría de Cultura (Secult) y la Fundación Cultural del Estado de Bahía (Funceb). Las presentaciones del Domingo no TCA tienen lugar en la sala principal del teatro, que tiene capacidad para 1554 espectadores, con entradas a un precio simbólico de R$ 1,00 (precio entero), y vienen atrayendo mensualmente, en las mañanas de domingo, un público numeroso y ecléctico, formado por adultos, niños y ancianos, oriundos de los más diversos barrios de Salvador. De 2007 a 2015, se llevaron a cabo 112 presentaciones de 104 espectáculos de diversos lenguajes artísticos. Genetic mapping of pDS15 revealed that this cosmid clone carries the Sinorhizobium meliloti TCA cycle genes mdh, sucCDAB, sdhAB and part of lpdA. Three genes (mdh, sucC , and sucA) were completely sequenced and submitted to GenBank. The nucleotide and amino acid sequences of the TCA cycle genes encoded on pDS15 were aligned and found to be highly homologous with other closely related rhizobial species. S. meliloti cells grown in LBmc express the mdh-sucCDAB operon as one transcript, based on RT-PCR results. Alternative sigma factor sigma54 was not found to have a role in mdh-sucCDAB expression. Despite considerable effort, we have not been able to isolate sucA mutants via random transposon Tn5tac1 mutagenesis to date. Homologous recombination between a plasmid-borne sucA::Tn5 and wild-type S. meliloti sucA failed to generate a bona fide mutant, as revealed by Southern blot analysis. Plasmid pDS15 was mutagenized with transposons Tn5, Tn5tac1, and Tn5-B20. Three Tn5-B20 insertions were mapped to mdh, sucD, and sucA respectively, and preliminary gene expression studies were done.

Resumen

In the symbiotic association between S. meliloti and its plant host, the nutrient exchange from plant to bacteroid is in the form of C4-dicarboxylic acids such as malate. These compounds directly enter the TCA cycle and the derived energy is used for the nitrogen-fixation process. Malate dehydrogenase (mdh), the two subunits of succinyl-CoA synthetase (sucCD), and two of the three subunits of 2-oxoglutarate dehydrogenase (sucAB) are encoded as an operon with the order mdh-sucCDAB. The expression of this operon is controlled by a single promoter found directly upstream of mdh. The transcrptional start site was mapped (a guanine residue at postion -63) and RT-PCR demonstrated that expression is as one polycistronic message in cells grown in LBmc. Transcriptional lacZ-gene fusions to mdh, sucD and sucA demonstrated that the mdh promoter is under catabolite control as evidenced by the change in ß-galactosidase expression depending on the carbon-source. Expression was highest with acetate followed closely by arabinose and glutamate but lowest with pyruvate as sole carbon-source. MDH was purified, N-terminal sequenced and kinetic assays were performed to determine Km, Vmax. A pH 10 was found to be the optimal for MDH. 2-oxoglutarate exhibited competitive inhibition on MDH. The annotated genome of S. meliloti contains two alleles (sucB), one chromosomal and one on megaplasmid pSymB, which putatively encode the E2 subunit of 2-oxoglutarate dehydrogenase. Only the chromosomally-borne allele was found to be a functional sucB. Lastly, results of a study of the E3 subunits of pyruvate dehydrogenase, 2-oxoglutarate dehydrogenase and branched-chained keto-acid dehydrogenase were presented. Dans la relation symbiotique entre S. meliloti et la plante M. sativa, l'apport en nutriments de la plante vers les bacteroids se fait sous la forme d'acides C4-dicarboxiliques tel que le malate. Ces composés entrent directement dans le cycle de Krebs et l'énergie produite est utilisée pour la fixation d'azote. Malate déhydrogénase (mdh), les deux composantes de succinyl-CoA synthèse (sucCD), et deux des trois composantes de 2-oxoglutarate déhydrogénase (sucAB), sont inscrites en un opéron dans l'ordre qui suit ; mdh-sucCDAB. L'expression de cet opéron est contrôlée par un seul promoteur situé directement en amont de mdh. Le site de début de transcription a été identifié et RT-PCR a démontré la nature polycistronique de l'expression dans les cellules provenant de bactéries cultivées avec LBmc. Fusions transcriptionelles aux gene-lacZ de mdh, sucD et sucA ont été utilisées pour déterminer les niveaux relatifs d'expression dans des cultures provenant de médiums minimaux contenant des sources de carbone spécifiques. MDH a été purifié, le N-terminal séquencé, et des dosages cinétiques ont été performés pour déterminer Km, Vmax, le pH optimal, et l'effet des inhibiteurs allostériques. Le génome annoté de S. meliloti contient deux allèles qui encodent supposément les composantes E2 (sucB) de 2-oxoglutarate déhydrogénase. Un seul des allèles a démontré être une version fonctionnelle de sucB. Finalement, les résultats d'une étude sur les composantes E3 de pyruvate déhydrogénase, sur 2-oxoglutarate déhydrogénase et sur déhydrogénase de kéto-acide de chaines ramifiées seront présentés.

Abstract Decarbonising heavy-duty truck fleets is essential for meeting the European Union’s ambitious transport-related emission reduction targets. This paper presents the techno-economic and environmental modelling results of zero-or-low-emission truck fleets (ZLETs). ZLETs like battery electric and hydrogen fuel-cell electric trucks are compared against diesel truck fleets. Key parameters used to compare ZLETs against diesel trucks are total cost of ownership (TCO) and total carbon abatement (TCA) cost. TCO and TCA analysis are evaluated for three different electricity and hydrogen production scenarios: off-grid, on-grid, and hybrid (use of both dedicated renewables and grid electricity adhering to renewable energy directive-II). Results show TCO of both ZLETs is higher than diesel trucks for an off-grid scenario. Battery trucks show the lowest TCO for on-grid scenario due to the continuous availability of cheap grid electricity and no large-scale energy storage cost. In contrast, fuel-cell trucks are cheaper than battery trucks in a hybrid scenario due to the relatively low cost of large-scale hydrogen storage compared to electricity in expensive onsite batteries. Both ZLETs have minimum TCO for on-grid scenario. However, higher well-to-wheel emissions can be abated using a hybrid scenario. This shows only when cheap renewable electricity is available continuously without large-scale battery storage, battery trucks are more cost-effective and truly green choice over fuel-cell trucks.

AbstractTriclosan (TCA) is an antibacterial and antimicrobial compound that is incorporated into toothpaste, soap, and liquid dishwasher. Continuous TCA exposure may contribute to the emergence of antibiotic-resistant bacteria in the microbiome. Triclosan also reacts to form dioxins, which bioaccumulate and are toxic to aquatic organisms, impedes the thyroid hormone metabolism of the human body. Laccases are multi copper-containing enzymes that can degrade the aromatic compounds and thus reduce their toxicity. To effectively degrade the compound, it is essential to understand the molecular function of the enzyme. Hence, a molecular docking study of laccase enzymes with Triclosan was done. The Tramates versicolor laccase structure was retrieved from PDB and ligand structure was taken from Pubchem. The binding mode and interaction of TCA and laccase were studied using Auto dock Vina software and the stability of the docked complex had been explored via Molecular Dynamics (MD) simulation study using Schrodinger Desmonde. The binding affinity score was found to be −6.5kcal/mol. The majority of the residues in RMSF were within the 2.5Å limit. The radius of gyration remained within the range from 21.7 to 22.1Å for Laccase – TCA complex throughout the 50 ns simulation. MD simulation results show that the enzyme complex remains stable all through the catalytic action.

Abstract We have investigated the role of various structural fragments, viz. C=C, C=O and the aromatic benzene ring present in trans-cinnamaldehyde (TCA) on its inhibition performance using density functional theory (DFT). For this purpose, the adsorption behavior of 2-methylbutanal (2MB), benzene (BZ), crotonaldehyde (CA), and trans-cinnamaldehyde (TCA) on Fe (001) surface were investigated. The quantum chemical descriptors and computed adsorption energies indicate that the inhibition efficiencies vary in the following order: 2MB&lt; CA &lt;TCA. Additionally, a deeper analysis of adsorption energies of 2MB, BZ, CA and TCA reveals that the contribution of the C=O, C=C and benzene ring in the corrosion inhibition mechanism of TCA is nearly equal. The bond lengths of the optimized geometries, electron density difference plot and projected density of states revealed that 2MB, BZ, CA and TCA chemisorb on Fe (001) surface through Fe—C and Fe—O bonds.

Abstract Part of any well completion job is the installation of multiple casings to isolate the well from any other reservoirs such as water aquifers. The space between each two casings is called the annulus. There are two kinds of annuli; tubular-casing annulus (TCA) and casing-casing annulus (CCA). While CCA is maintained at zero pressure, TCA should be maintained at positive pressure and this is usually accomplished by displacing the TCA with inhibited diesel. This practice would enable the quick detection of any possible casing leaks or downhole failure due to corrosion leading to TCA communication; either surface or downhole during production. The composition of diesel is mainly C9+ with a very minimum amount of light ends C3 to C7. Arab Light crude oil on the other hand is rich in light ends mainly because of the associated gas. In this paper, we have studied the ratio between light ends to heavy ends in different mixtures of diesel with two grades of crude oil utilizing gas chromatography. This will help production engineers for the early detection of any possible downhole communication caused by corrosion to maintain the well integrity. This study was conducted to assist one of the giant Arab Light reservoirs in the world as well as two additional reservoirs.

Descripción precisa de los objetivos: El trastorno por consumo de sustancias (TCS) es una comorbilidad común en individuos con trastorno bipolar (TB) y se asocia a una peor evolución clínica del TB. El presente estudio tiene como objetivo adoptar un algoritmo de machine-learning para identificar qué características sociodemográficas o clínicas están más asociadas a la presencia de diferentes tipos de TCS en el TB. Material y métodos: Pacientes adultos con TB fueron reclutados en la Unidad de Trastornos Bipolares y Depresivos del Hospital Clínic de Barcelona. Se recogieron las características sociodemográficas y clínicas y se entrenaron cuatro Random Forests (RFs) para predecir la presencia de TCS en general, de trastorno por consumo de alcohol (TCA), o de TCA y al menos otro tipo de TCS (TCA+TCS) en la muestra total o en diferentes submuestras. Las variables más importantes seleccionadas por los RFs se consideraron como variables independientes en regresiones logísticas múltiples para predecir la presencia de TCSs. Resultados y conclusiones: Se incluieron a un total de 508 pacientes (276 mujeres) con TB-I (345) o TB-II (205); entre ellos 262 presentaron comorbilidad con TCS (108 con solo TCA, 106 con TCA+TCS). El TCS se predijo con una precisión del 65,3%, y se asoció con un comórbido trastorno de la personalidad de cluster B (OR=2,31; p=0,006) y tener una relación (OR=0,6; p=0,015). El TCA+TCS (precisión=75%) se asoció con un primer episodio hipomaniaco (OR=4,34; p=0,01), y hetero-agresividad (OR=3,15; p=0,003). El TCA+TCS en una submuestra que incluía solo a personas con TCA (precisión=62.5%) se asoció con un primer episodio afectivo depresivo (OR=0,41; p=0,011). Ninguna variable se asoció con el TCA (precisión=53.8%). Incluir sólo los factores sociodemográficos y clínicos en modelos de machine-learning limita su eficacia, y los futuros modelos deberían considerar también predictores biológicos, clínicos y de neuroimagen para explorar más esta asociación.

Resumen

OBJETIVOS: Demostrar la relación que existe entre el alcohol y el TCA. MATERIAL Y METODOS: Revisión sistemática y exposición de un caso clínico. RESULTADOS : Desde hace décadas se han realizado investigaciones en la línea de buscar una comorbilidad entre los TCA y el consumo de sustancias. Según la evidencia científica se ha demostrado que existe una elevada prevalencia de abuso de sustancias entre pacientes con TCA, mucho más acentuada con el alcoholismo. La bulimia nerviosa es la enfermedad en la que se han descrito mayor número de trastornos por abuso de sustancias, siendo la prevalencia en torno al 2,6 mayor que en anoréxicas purgativas y a su vez que en las restrictivas. Por otro lado, se ha encontrado que el 35-40% de alcohólicos tienen historia de TCA. Se ha establecido como factor común en ambas patologías el déficit en la regulación del control de impulsos y el afecto. Así como una elevada prevalencia de antecedentes familiares de alcoholismo (73,3%) y antecedentes de malos tratos (43,3%), que podría indicar que estas pacientes presentan una marcada vulnerabilidad para el desarrollo de alcoholismo y cierto grado de indefensión. CONCLUSIONES:Se ha observado que el TCA aparece previo a la dependencia alcohólica en un 86,6% de los casos, mayoritariamente en la adolescencia, con mayor frecuencia de conductas de atracones y purgativas. Describiéndose además la utilización del alcohol con fin anorexígeno, lo que nos sugiere que el alcohol cumpliría una función dentro del TCA y que a su vez una abstinencia mantenida durante 6 m se asociaría a una buena evolución de aproximadamente el 50% de los TCA. El abordaje precoz del riesgo de dependencia alcohólica en pacientes diagnosticadas de TCA disminuiría la comorbilidad de ambos trastornos y mejoraría el TCA. Esto supondría beneficio clínico para el paciente y la disminución de costes sociosanitarios.

INTRODUCCION Y OBJETIVOS A pesar de la alta tasa de trastornos alimentarios (TCA) y trastornos por uso de sustancias (TUS) concurrentes en las mujeres, existe una falta de tratamiento integrado. Los trastornos por uso de sustancias (TUS) y los trastornos de conducta alimentaria (TCA) ocurren con frecuencia en mujeres. Las opciones de tratamiento para esta población siguen siendo limitadas. A menudo ambos trastornos son abordados secuencialmente, abordando primero el trastorno más grave, o de manera simultánea, pero en programas separados. Los TCA y TUS concurrentes se asocian con recaídas frecuentes y malos resultados del tratamiento. Las investigaciones llevadas a cabo sobre programas de tratamiento integrados son escasas. El objetivo de este trabajo es revisar la situación actual respecto de los programas de tratamiento ya existentes, para personas con TCA y TUS concurrentes. así como una propuesta de tratamiento por parte de la UTCA Santa Cristina. MATERIAL Y METODOS Para la elaboración de este trabajo se ha realizado una exhaustiva revisión bibliográfica de más de 30 artículos de carácter científico, tanto de revisión como experimentales pertenecientes al subtipo de ensayos clínicos acerca de los programas integrados de tratamiento para TCA y TUS existentes hasta la actualidad a través de los buscadores Pub Med y Google académico. Desde la UTCA del HU Santa Cristina llevamos a cabo un programa de tratamiento en aquellas pacientes con TCA y TUS lo más integrador posible en dos vertientes. Se expondrán los tipos de intervenciones y la adaptación del tratamiento, integrador, a este perfil de pacientes. CONCLUSIONES Las mujeres con TCA y TUS concurrentes que ingresan al tratamiento de la UTCA son una población especial donde el TUS a menudo recibe un tratamiento insuficiente. Abordar el uso de sustancias dentro del contexto del tratamiento del TCA puede derribar barreras actuales, demostrando su eficacia para esta población.

Resumen

Objetivos La relación clínica entre trastornos adictivos (TA) y trastornos de la conducta alimentaria (TCA) tuvo un fundamental apoyo con los criterios diagnósticos de Goodman, que permitieron clasificar conductas de estirpe impulsiva como son el juego, la compra compulsiva y, posteriormente (finales de los noventa) bulimia nerviosa y anorexia nerviosa. Si bien las semejanzas sintomatológicas son destacadas por los clínicos que trabajan en ambos grupos, es preciso realizar una comparativa entre características fisiopatológicas, anatomoestructurales, bioquímicas y genéticas para alcanzar el objetivo del presente trabajo, desarrollar un marco teórico de similitud entre TA y TCA, que pudiese acercar maniobras terapéuticas en ambos grupos de patologías. Métodos Se realiza una revisión narrativa con los últimos hallazgos característicos de patologías TA y TCA, a nivel fisiopatológico, estructural (neuroimagen) y molecular (bioquímico y genético), expuestos a modo de espejo, para una mayor clarificación. Resultados y conclusiones En los estudios de neuroimagen en las adicciones, así como en las investigaciones neuropsicológicas, además del sistema dopaminérgico, se presta especial atención a algunas regiones del cerebro que han surgido repetidamente en dichos estudios. Las regiones más consistentemente implicadas son partes de la corteza frontal (corteza orbitofrontal y corteza prefrontal dorsolateral), el cíngulo rostral, la amígdala y la ínsula. Los estudios de análisis GWAS ha conseguido relacionar TCA y TA de manera específica, no en su conjunto, lo que ha permitido inferir la diferencia incluso entre los propios TCA, desde un punto de vista genético. Desde un punto de vista bioquímico, los niveles anómalos de receptores dopaminérgicos D2 en circuitos de recompensa entre trastornos por atracón y adicción por sustancias, acerca los grupos de TCA y TA, pero diferencia también de manera intragrupal las enfermedades. La dificultad para tratar de manera farmacológica los TCA, podría verse mitigada si se consiguiese establecer una correlación entre ambos grupos de enfermedades.

INTRODUCCION: La asociación entre los trastornos de conducta alimentaria (TCA) y los trastornos por uso de sustancias (TUS) pueden complicar el tratamiento y pronóstico de ambas patologías, siendo esta una comorbilidad frecuente. El efecto anorexígeno de la anfetamina puede relacionarse con el desarrollo de un TCA. CASO CLINICO: Presentamos el caso clínico de una mujer de 20 años, con diagnóstico en la adolescencia de TDAH y tratamiento con derivados anfetamínicos en aquel momento, que ha desarrollado un TCA tipo bulimia nerviosa, por el cual se encuentra en la unidad de tratamiento intensivo de TCA (UTCA). En la actualidad mantiene consumo de diversas sustancias que puedan alterar el apetito, siendo los derivados anfetamínicos los más utilizados por ella para este fin. Presenta rasgos de personalidad obsesivos y dependientes, e ideas devaluadoras permanentes sobre sí misma. DISCUSION: El carácter anorexigénico de los derivados anfetamínicos parece jugar un papel relevante a la hora de elegir su consumo, que puede llegar a ser buscado en mercados ilegales, en pacientes con vulnerabilidad para desarrollar un TCA. Ambos trastornos tendrían una influencia bidireccional, dificultando una buena evolución de la paciente, si no se tratan a la par. CONCLUSION: Siempre será necesario explorar un posible trastorno por uso de sustancias en pacientes con sintomatología alimentaria o diagnóstico de TCA. Además se recomienda, siempre que sea posible, evitar el uso de fármacos con características anorexigénicas en pacientes con perfil diana para el desarrollo de un trastorno de la conducta alimentaria, clásicamente, mujeres jóvenes de áreas urbanas.

INTRODUCCIÓN Y OBJETIVOS Los Trastornos de la Conducta Alimentaria (TCA) son trastornos mentales que se caracterizan por un comportamiento patológico frente a la ingesta alimentaria y el control del peso. Se ha observado que los TCA y los Trastornos por Uso de Sustancias (TUS) aparecen relacionados en muchas ocasiones e influyen en la mortalidad y morbilidad de estos. El objetivo principal de este trabajo es conocer la evidencia existente sobre la relación entre los TUS y los subtipos de TCA. METODOLOGÍA Se ha efectuado una búsqueda bibliográfica en la base de datos Pubmed, utilizando como palabras clave "eating disorder", "drug abuse", "substance use disorder", restringiendo la búsqueda a artículos en inglés, en los últimos 15 años y que contengan estas palabras clave en el título o abstract. RESULTADOS Y CONCLUSIONES Hay estudios que evidencian la concordancia entre los TUS y los diferentes tipos de TCA, observándose una mayor comorbilidad en los pacientes con bulimia o trastornos por atracón, que en los diagnosticados de anorexia nerviosa de tipo restrictivo. Estos trastornos tienen una relación directa con comportamientos impulsivos y buscan el alivio a los problemas de ansiedad, depresión y otros subyacentes. Por otra parte, se ha catalogado un nuevo trastorno alimentario, la "drunkorexia", que es común en la adolescencia y que se caracteriza por una restricción de comida en los días que se planea un consumo excesivo de alcohol, para no renunciar al consumo del mismo y, de esta forma, evitar un aumento de peso. Como conclusión, los TCA se asocian de forma significativa con los TUS. Es importante reconocer el abuso de alcohol y otras drogas en pacientes con TCA para facilitar su seguimiento y recuperación, monitorizándolos y desarrollando un tratamiento multidisciplinar, individualizado y adaptado a las diferentes patologías coexistentes.

Resumen

Aspergillus terreus was reported as the promising fungal strain for itaconic acid; however, the commercial production suffers from the low yield. Low production yield was claimed as the result of completing TCA cycle towards biomass synthesis while under limiting phosphate and nitrogen, TCA cycle was somewhat shunted and consequently the metabolite fluxes move towards itaconic acid production route. By regulating enzymes in TCA cycle, it is believed that itaconic acid production can be improved. One of the key responsible enzymes involved in itaconic acid production was triggered in this study. Pyruvate carboxylase was allosterically inhibited by L-aspartate. The presence of 10 mM L-aspartate in the production medium directly repressed PC expression in the living A. terreus whilst the limited malate flux regulated the malate/citrate antiporters resulting in the increasing cis-aconitate decarboxylase activity to simultaneously convert cis-aconitate, citrate isomer, into itaconic acid. The transport of cis-aconitate via the antiporters induced citrate synthase and 6-phosphofructo-1-kinase activities in response to balance the fluxes of TCA intermediates. Successively, itaconic acid production yield and final concentration could be improved by 8.33% and 60.32%, respectively compared to those obtained from the control fermentation with the shortened lag time to produce itaconic acid during the production phase.
