A serious vine decline of cucurbits known as cucurbit yellow vine disease (CYVD) is caused by rod-shaped bacteria that colonize the phloem elements. Sequence analysis of a CYVD-specific polymerase chain reaction (PCR)-amplified 16S rDNA product showed the microbe to be a γ-proteobacterium related to the genus Serratia. To identify and characterize the bacteria, one strain each from watermelon and zucchini and several noncucurbit-derived reference strains were subjected to sequence analysis and biological function assays. Taxonomic and phylogenetic placement was investigated by analysis of the

The bacterium that causes cucurbit yellow vine disease (CYVD) has been placed in the species Serratia marcescens based on 16S rDNA and groE sequence analysis. However, phenotypic comparison of the organism with S. marcescens strains isolated from a variety of ecological niches showed significant heterogeneity. In this study, we compared the genomic DNA of S. marcescens strains from different niches as well as type strains of other Serratia spp. through repetitive elements-based polymerase chain reaction (rep-PCR) and DNA-DNA hybridization. With the former, CYVD strains showed identical banding

Background The red palm weevil (RPW), Rhynchophorus ferrugineus (Olivier), is an important quarantine pest, which has caused serious economic losses in various palm species, such as coconut, oil palm and date palm. Finding effective biocontrol resources is important for the control of this pest and the protection of palm crops. Methods A pathogenic strain HJ-01 was isolated from infected and dead pupa of Tenebrio molitor using tissue separation method. The HJ-01 strain was streak cultured and purified, and its morphological, physiological, biochemical characteristics, and 16S rDNA homology wer

Commercial plantings of summer squash in Charlemont, Franklin County, MA, were decimated in 1999 by 100% incidence of a yellowing disease resembling cucurbit yellow vine disease (CYVD) (1). Both plantings were established in the same field during the third week of May, one with transplants and the second by direct-seeding. Each planting consisted of four 30-m rows each of yellow zucchini (Cucurbita pepo cv. Gold Rush), summer squash (C. pepo cv. Seneca Prolific), and zucchini (C. pepo cv. Condor). Crops were produced organically and pyrethrum was used to control a high infestation of squash bu

Our study was conducted to detect virulence genes in Serratia marcescens. It has many virulence genes that cause nosocomal infections in immunocompromised persons and neonates. A total of 24/100 (24%) S. marcescens were obtained from neonates suffering from meningitis, and they were identified using culture characteristics biochemical- tests and confirmed by Polymerase chain reaction (PCR) technique, using the 16S rRNA gene. A strain of Acidithiobacillus ferrooxidans designated S t1 was isolated and identified from the acid mine drainage of Tianmashan Coal Mine, Tongling county, Anhui province. Its morphological, physiological and biochemical characters, growth conditions of pH value and temperature, 16S rDNA sequence and the phylogenetic tree was studied. Results showed that the bacteria strains from different sites have divergence at the curve of pH and Eh values during the enriching culture. The isolated strain S t1 grew in a pH rang from 2.0 to 3.0 and temperature 25 to 35°C with over 98%similarity toAcidithio

Rice bean (Vigna umbellata) is an underutilised legume with unexplored endophytic microbiome. Identification and characterisation of these endophytes are critical in understanding their roles in plant’s growth, health, and productivity. In this study, twelve morphologically and biochemically distinct bacterial endophytes were isolated from rice bean roots. Sequence analysis of the 16S rDNA of the isolates revealed that members of Proteobacteria, including Stenotrophomonas sp., Shinella sp., Roseomonas sp., Pantoea dispersa, and Serratia marcescens dominated the root tissues of rice bean. ABSTRACT This is the first report of filamentous actinobacteria isolated from surface-sterilized root tissues of healthy wheat plants (Triticum aestivum L.). Wheat roots from a range of sites across South Australia were used as the source material for the isolation of the endophytic actinobacteria. Roots were surface-sterilized by using ethanol and sodium hypochlorite prior to the isolation of the actinobacteria. Forty-nine of these isolates were identified by using 16S ribosomal DNA (rDNA) sequencing and found to belong to a small group of actinobacterial genera including Streptomyces, Microb

Symbiotic bacteria associated with the Medicago genus are separated into two closely related species named Sinorhizobium meliloti and Sinorhizobium medicae. To discriminate rapidly between these two bacterial species, two 15-base DNA probes, 16Smfs and 16Smed, were designed from the alignment of 16S rDNA sequences to differentiate S. meliloti from S. medicae. Their specificities were evaluated by dot-blot hybridization experiments on 25 reference strains representing 13 species of Rhizobium and Sinorhizobium, and by comparison with all 16S rDNA sequences available in the GenBank data base.
