Abstract Background AlloBMT is a potentially curative treatment for multiple myeloma (MM). However, its effectiveness has been compromised by high transplant related mortality (TRM) and acute and/or chronic graft-versus host disease (GvHD). Our development of PTCy has reduced the incidence of GvHD allowing safe and effective related haploidentical alloBMT. PTCy promotes tolerance in both alloreactive host and donor T cells, leading to suppression of both graft rejection and GVHD after alloBMT. Furthermore, it is not clear whether mortality rates have changed over the past 4 decades. Abstract Objective Graft vs. host disease (GVHD), where donor T cells recognize and destroy alloantigen-expressing host tissues, is a major complication after allogeneic bone marrow transplantation (alloBMT). To create space for donor stem cells and remove recipient immune cells, alloBMT protocols rely on conditioning regimens involving irradiation and/or chemotherapy. Interleukin (IL)-33 is augmented in recipient barrier tissues by conditioning before alloBMT and is a required signal to donor T cells for GVHD. In the current studies, we tested the hypothesis that the dominant role for IL-33 i
