It was demonstrated that 2-month old protocorms were better than leaves and root tips for callus inducing. More calli were formed on 1/2 MS supplemented with 0.1-1.0 mg/l TDZ, and 10 mg/l 2,4-D was not suitable for callus induction. There were roots, white and yellow buds appeared while inducing callus.

After four weeks, the results showed that callus could be induced by 0.5–2 mg/L of 2,4-D + 0.1–0.5 mg/L of TDZ, but the root explants in PGR–free medium became browning.
