Pełny tekst źródłaIn this work new hybrid silica based materials containing organic fluorescent molecules, derivative from benzazoles were prepared. It was used the sol-gel method, making three different synthesis, with the following catalysts: fluoridric acid, sodium fluoride, and ammonium fluoride. It was performed the incorporation of two molecules. The dye 2-(5'- amine-2'-hydroxfenil)benzimidazole, which became disperse and physically adsorbed in the matrix; and the dye 2-(5'-N-(3-triethoxysilil)propylurea-2'-hydroxyfenil)benzimidazole, that, being a silylated dye, became chemically bonded to the matrix structure. These organic compounds are important due to its applications in chemistry, medicine and environmental sciences. Many of these applications are due to the fact that these fluorescent molecules present Stokes shift higher than 100 nm, caused by an intramolecular proton transfer in the excited state. These new fluorescent hybrid materials were obtained in the form of monolits and powders. The high-pressure processing of these powders (ca de 6,0 GPa), in a quasihydrostatic medium, enabled the preparation of dense, transparent and stable compacts. The optical and textural properties of these different materials were studied. The fluorescence excitation-emission spectra were performed, and using deconvolution, three emission bands were observed. Two of these bands presented Stokes shift higher than 100 nm, giving the warrant of the existence of intramolecular proton transfer in the excited state of these molecules, in this silica matrix. The materials catalyzed with fluoridric acid were always translucent and homogeneous; besides, the high-pressure processing of these materials resulted in compacts where the interesting optical properties were maintained, making them suitable for applications in optical devices. The materials catalyzed with sodium fluoride and ammonium fluoride became always opaque, but they present potential applications as hybrid pigments. The textural analysis was performed using the nitrogen adsorption/dessorption isotherms, where textural differences were observed ascribed to experimental parameters that influence the sol-gel process. The thermogravimetric curves were strongly influenced by silica surface characteristics, being evident the importance of synthesis conditions, already cited, for the preparation of these hybrid materials.

These substances present strong intramolecular hydrogen bonds and a large Stokes shift, wich is thought to be due a excited state intramolecular proton transfer (ESIPT) processo Reactions of those isothiocyanates with amines and proteins have been performed and the photophysical properties ofthe products have been studied in the aim to prove the feasibility oftheir use as tluorescent labels. It has been found that the synthesized isothiocyanates form tluorescent products with amines and proteins. having potential utility as tluorescent labels.

Diversas metodologias têm sido empregadas no estudo do processo de reparo alveolar. Em razão disso, o propósito deste trabalho é apresentar uma metodologia que possibilite a utilização dos fluorocromos como forma de análise do processo de reparo alveolar. Para tanto, 5 ratos tiveram o incisivo central superior direito extraído. Aos 6 dias foi injetada a calceína, aos 15 dias a alizarina e aos 21 dias a oxitetraciclina. Após 35 dias esses animais foram sacrificados. Obtidas as peças, foi analisado o terço médio do alvéolo. Observou-se predominância de calceína junto à parede lingual, grande quantidade de marcação com alizarina e a marcação da oxitetraciclina em menor quantidade, mostrando que houve menor taxa de mineralização óssea. Este trabalho foi capaz de mostrar a dinâmica do processo de reparo alveolar de forma clara e bastante ilustrativa, além de ser de fácil reprodução e apresentar um custo relativamente baixo.
Several methods have been employed in the study of the alveolar repair process. For this reason, the aim of this study is to present a methodology that allows the use of fluorochromes in the analysis of this process. Thus, 5 rats had their upper right central incisors extracted. At day 6 calcein was injected, at day 15 alizarin was injected, and at day 21, oxytetracycline was injected. After 35 days the animals were euthanized. After the samples were obtained, the middle third of the socket was analyzed. Predominance of calcein was observed next to the lingual wall. A great amount of alizarin staining and a smaller amount of oxytetracycline staining were also observed, showing that there was a smaller rate of bone mineralization. This study was able to show the dynamics of the socket repair process in a clear and illustrative manner, besides being easy to reproduce and relatively inexpensive.

Pełny tekst źródłaThe synthesis and efficient purification of the fluorescent dyes 2,5-bis(2'-benzoxazolil)hydroquinone and 2,5-bis(2'benzimidazol)hydroquinone in the quantities needed for laser work, are reported. Additionally, simple modifications of theses procedures are described for obtaining the corresponding monoheterocyclic compounds 4-(2'-benzoxazolyl)- and 4-(2'-benzoxazolyl)-2.5-dihidroxybenzoic acids. Methylation and/or acetylation of 2,5-bis(2'-benzoxazolyl)hydroquinone yielded the respective mono- and disubstituted derivatives, which are important model molecules for the photophysical study of the parent dye.

Pełny tekst źródłaThe synthesis and characterization of seven new heterocycles named 2-[5'-(N-viny1ene)- 2'-hydroxyphenil]benzazoles were accomplished in this work. These compounds belongs to the heterocycles class type 2'-hydroxyphenylbenzazoles wich present a strong emission of fluorescence due to excited-state intramolecular proton transfer -ESIPT- when they are excited by UV light. The seven heterocycles synthesized possess a strong intramolecular hydrogen bond between the nitrogen azole and the phenolic group and also a high Stokes shift (200 nm) which is typical of these compounds. The synthesized 2'-hydroxyphenylbenzazoles heterocycles present a structural fragment constituted by alkenes with several functional groups such as ester, cyan and alkylthio. Thus, the compounds formed in this work are unpublished in the literature and they call attention and the interest in order to synthesize fluorescent polymers, which are largely studied nowadays. Besides, these compounds can suffer intramolecular cyclization originating derivatives from the 4(1H)quinoline systems, which present chemotherapic interest to treat virial diseases. Preliminary teste of polymerization and cyclization were realized with some synthesized derivatives and we found extremely good results.

Pełny tekst źródłaNanocarriers systems of organic molecules have emerged as promising systems for application in therapy and diagnosis. In the present study polymeric nanocapsules containing fluorescent probes in their different domains, capable of providing image when analyzed in confocal fluorescence microscope, showing the potential of the system as an agent for optical imaging. An important feature of proposed molecules as fluorescent probes, there is no overlap of the spectra of absorption and fluorescence emission. Thus, a group of synthetic dye benzoxazólicos (HBO-C8) to mark the lipophilic core, and PCL and chitosan polymers were covalently linked to dye rhodamine B and acridine orange , respectively. The formulations developed and characterized with two or three markers showed nanotechnological character and transmission photomicrographs showed that nanoparticles containing PCL-Rho and HBO-C8 (without chitosan) preserved morphology of the nanoparticle. Additionally, the HBO-C8 was was quantified on formulation contained HBO-C8 and PCL-Rho, where it was found that 98% of the dye is internalized in the nanoparticle thus reaching the purpose proposed to use as a probe for marking the lipophilic core . By images of confocal fluorescence microscopy were obtained the colocalized of fluorescent markers to formulations containing two or three fluorescent domains, demonstrating that these nanocapsules can be proposed as a promising platform to be used as probes for optical image.

Plasmons, oscilações coletivas de elétrons de condução, podem ter grande influência sobre as propriedades ópticas de micro e nanoestruturas metálicas, e são de grande interesse no desenvolvimento de dispositivos fotônicos. Neste trabalho, produzimos filmes finos metálicos pelo método de evaporação à baixa pressão e, em seguida, fizemos a sua caracterização por medidas de refletividade óptica. A partir daí, estudamos o comportamento da emissão espontânea (fluorescência) induzida por plasmons superficiais em interfaces metal-dielétrico. O sistema estudado era constituído por um corante dissolvido em uma matriz polimérica pura ou contendo nanopartículas de prata ou de rutila. A excitação de plasmons foi obtida pelo uso da técnica de reflexão total atenuada (Configuração de Kretschmann-Raether), e foram utilizadas medidas de luminescência na detecção de sinal de fluorescência induzida por plasmons superficiais. A intensidade da fluorescência e sua largura de linha foram analisadas em função da intensidade de luz incidente na amostra. Foi observado que, para altas intensidades, a forma de linha da luminescência era modificada, indicando a ocorrência de emissão estimulada, mas com baixa eficiência devido à baixa potência do laser utilizado

Pełny tekst źródłaBiofilms provide an environment capable of protecting microbial cells from damage by chemical and physical factors of the immune system, and hinder the penetration of various antimicrobial agents, promoting nosocomial infections related to catheters, increasing mortality and morbidity of patients. Therefore, it is important to ensure proper hygiene to prevent contamination and infections in hospital environments. For this purpose, this study explored the identification of a substance that both detects and have biocide activity against fungal biofilms on stainless steel surfaces. Both nine fluorescent benzimidazole substances, coded NB1 to NB9 and eight imidazolium salts of azo dyes, named denominados C4MImErioCr, C10MImMO, C16MImMO, (C10)2MImMO, C4MImMO, C10MImORANGEII, C16MImORANGEII e (C10)2MImORANGEII were tested as candidates. These substances were tested applying a methodology developed to determine if a substance is able detecting and have biocide activity against fungal biofilms. Overall, this study involved the following methods: (1) Minimum Inhibitory concentration test following the CLSI protocol (M27-A3; the substances were tested against nine fungal strains, including C. tropicalis, C. albicans and C. parapsilosis.), (2) microscopy to determine the marker capacity, (3) spraying tests of the substances on surfaces (stainless steel) with fungal biofilms, (4) tests to verify the capability of the substances to both stain and were biocide against fungal biofilms, applying microscopic techniques and (5) cytotoxicity tests Within the set of seventeen substances, benzimidazole derivative NB7 was identified with the desired capabilities, staining and biocide activity against fungal biofilms at the same time. All tested fungal strains were sensible to this substance. A biocide activity was identified on the biofilms of ATCC 18804 (C. albicans), ATCC 22019 (C. parapsilosis) and ATCC 750 (C .tropicalis), grown on stainless steel 304, when exposed fifteen seconds to substance NB7. Although this substance showed being cytotoxic, it represents a promising candidate for sanitization purposes, including medical tools.
