Aberrant production of reactive oxygen species (ROS) is a common feature of damaged retinal neurons in diabetic retinopathy, and antioxidants may exert both preventive and therapeutic action. To evaluate the beneficial and antioxidant properties of food supplementation with Lisosan G, a powder of bran and germ of grain (Triticum aestivum) obtained by fermentation with selected lactobacillus and natural yeast strains, we used an in vivo model of hyperglycemia-induced retinal damage, the fruit fly Drosophila melanogaster fed with high-sucrose diet. Lisosan G positively affected the visual system of hyperglycemic flies at structural/functional level, decreased apoptosis, and reactivated protective autophagy at the retina internal network. Also, in high sucrose-fed Drosophila, Lisosan G reduced the levels of brain ROS and retina peroxynitrite. The analysis of oxidative stress-related metabolites suggested 7,8-dihydrofolate, uric acid, dihydroorotate, γ-L-glutamyl-L-cysteine, allantoin, cysteinyl-glycine, and quinolate as key mediators of Lisosan G-induced inhibition of neuronal ROS, along with the upregulation of glutathione system. Of note, Lisosan G may impact oxidative stress and the ensuing retinal cell death, also independently from autophagy, although the autophagy-ROS cross-talk is critical. This study demonstrated that the continuous supplementation with the alimentary integrator Lisosan G exerts a robust and multifaceted antioxidant effect on retinal neurons, thus providing efficacious neuroprotection of hyperglycemic eye.

Lisosan G (LG), a fermented powder obtained from whole grains, is a recognized antioxidant compound that improves the bioactivity and survival of different cell types. The purpose of this study was to investigate whether LG ameliorates both the neural and the vascular damage characterizing early stages of diabetic retinopathy (DR). The effects of LG were studied in cultured explants of mouse retinas challenged with oxidative stress (OS) or in retinas of streptozotocin (STZ)-treated rats. Apoptosis, vascular endothelial growth factor (VEGF) expression, OS markers, blood-retinal barrier (BRB) integrity, and inflammation were assessed, while retinal function was evaluated with electroretinogram (ERG). LG extensively inhibited apoptosis, VEGF expression, and OS both in retinal explants and in STZ rats. In addition, STZ rats treated with LG displayed an almost total BRB integrity, reduced levels of inflammatory markers and a partially restored visual function as evaluated with ERG. In summary, we demonstrated that LG exhibits antioxidant and anti-inflammatory effects that exert powerful protective actions against neural and vascular defects characteristic of DR. Therefore, LG-containing foods or supplements may be considered to implement DR treatments.

Endogenous and exogenous signals derived by the gut microbiota such as lipopolysaccharides (LPS) orchestrate inflammatory responses contributing to development of the endothelial dysfunction associated with atherosclerosis in obesity, metabolic syndrome, and diabetes. Endothelial progenitor cells (EPCs), bone marrow derived stem cells, promote recovery of damaged endothelium playing a pivotal role in cardiovascular repair. Since healthy nutrition improves EPCs functions, we evaluated the effect of a fermented grain, Lisosan G (LG), on early EPCs exposed to LPS. The potential protective effect of LG against LPS-induced alterations was evaluated as cell viability, adhesiveness, ROS production, gene expression, and NF-kB signaling pathway activation. Our results showed that LPS treatment did not affect EPCs viability and adhesiveness but induced endothelial alterations via activation of NF-kB signaling. LG protects EPCs from inflammation as well as from LPS-induced oxidative and endoplasmic reticulum (ER) stress reducing ROS levels, downregulating proinflammatory and proapoptotic factors, and strengthening antioxidant defense. Moreover, LG pretreatment prevented NF-kB translocation from the cytoplasm into the nucleus caused by LPS exposure. In human EPCs, LPS increases ROS and upregulates proinflammatory tone, proapoptotic factors, and antioxidants. LG protects EPCs exposed to LPS reducing ROS, downregulating proinflammatory and proapoptotic factors, and strengthening antioxidant defenses possibly by inhibiting NF-κB nuclear translocation.

Aim. The aim of our work was to evaluate winter wheat samples on yielding and environmental plasticity for further use as a initial material to create a variety adapted to the Forest-steppe conditions. Methods. Working collection of 30 varieties of Ukrainian and foreign breeding on yield and economically valuable features was characterized in the field condition of Forest-Steppe. Regression analysis was used to determine the coefficient of ecological stability and plasticity. Results. The results of the study of breeding samples indicate the difference in yielding of the genotypes. Varieties that characterized by high environmental plasticity and yield and significantly outperformed the standard variety Podolyanka were Akteur (235 g/m2), Voloshkova (222 g/m2), Smuhlianka (241 g/m2), Lisova Pisnia (283 g/m2), Sarlota (215 g/m2), Perlyna Lisostepu (266 g/m2), Polisska-90 (237 g/m2), Stolychna (290 g/m2). Conclusions. The results obtained indicate that the tested varieties have high potential productivity and respond positively to the improvement of growing conditions, so they can be used as parent components in the creation of high-intensity varieties for Polissia and Forest-steppe. Urine Albumin Creatinine Ratio (UACR) merupakan metode untuk mengukur mikroalbuminuria, petanda awal dan paling banyakdigunakan untuk diagnosis nefropati diabetes. N-asetil-beta-d-glukosaminidase (NAG) air kemih adalah enzim lisosom dengan beratmolekul besar sehingga tidak difiltrasi oleh glomerulus. Ekskresi NAG di air kemih sangat mudah terganggu terutama oleh filtrasialbumin. Tujuan penelitian ini untuk mengetahui kenasaban NAG air kemih dengan UACR di pasien DM tipe 2. Penelitian ini merupakanpenelitian analitik dengan rancangan potong lintang terhadap 25 pasien DM tipe 2 yang berobat ke Poliklinik Endokrin Penyakit DalamRSUP Dr. M. Djamil Padang. Penelitian dilakukan mulai bulan Januari 2016–September 2016. Albumin air kemih diukur denganmetode imunoturbidimetri, kreatinin air kemih dengan metode Jaffe dan NAG dengan Enzyme-Linked Immune Sorbent Assay (ELISA).Kenasaban Spearman dilakukan untuk mengetahui hubungan aktivitas NAG air kemih dan UACR. Penelitian ini terdiri dari laki-laki68% dan perempuan 32%. Rerata umur 56,16±7,6 tahun dengan rentang 39–67 tahun. Kadar glukosa, ureum dan kreatinin serumberturut-turut, 148±49 mg/dL, 24±6,2 mg/dL dan 0,9±0,3 mg/dL. Normoalbuminuria sebanyak 80% dan mikroalbuminuria 20%.Median nilai UACR pasien DM tipe 2 adalah 6,02 (16,46) mg/g dengan rentang 1,53–119,41 mg/g dan rerata kadar NAG adalah51,01±31,88 ng/mL dengan rentang 9,45–144,38 ng/mL. Uji kenasaban Spearman menunjukkan kenasaban yang kuat antara aktivitasNAG air kemih dengan UACR dengan r=0,614 dan p<0,05. Terdapat kenasaban yang kuat antara aktivitas NAG air kemih denganUACR di pasien DM tipe 2.

Introduction: post-bariatric hypoglycemia (PBH) is considered a chronic complication after bariatric surgery that increase cardiovascular risk. Changes in gut microbiota and a dysregulated inflammatory response to a glucose load seems to be the conditions favoring the development of PBH. Lisosan G (LG) is a fermented powder obtained from whole grains (Triticum aestivum), rich in polyphenols and flavonoids with antioxidant, anti-inflammatory and prebiotic properties. Aims: The aims of the study were 1) to evaluate the effectiveness of using LG added to the diet in reducing PBH events in patients undergoing RYGB, 2) investigate the mechanism by which LG acts on the gut-pancreas axis. Methods: Twenty patients self-reporting symptoms/signs of PBH, who had undergone gastric bypass between 2015 and 2018, were enrolled. At baseline, patients underwent clinical examination, blood test and 4-hour oral glucose load test (OGTT). After that, the patients were kept on a free diet with 15 days of continuous glucose monitoring (CGM). The CGM was then repeated for another 15 days, adding LG to the same dietary regimen (5 g LG-powder, bid). At the end of the treatment period, patients repeated the 4-hours OGTT and blood test. Plasma insulin and C-peptide were measured by electrochemiluminescence on a Cobas e411 instrument. Plasma total glucose-like peptide-1 (GLP 1) concentrations were measured by ELISA (Millipore). Insulin sensitivity was assessed by the oral glucose insulin sensitivity index (OGIS). PBH was defined as a plasma glucose level of ≤ 60 mg/dl in presence of typical symptoms. Results: a marked reduction in PBH episodes recorded by CGM was observed after LG administration (6.5 [5-11] vs 2.5 [2-3], p= 0.009), as well as a reduction in the overall length of hypoglycemia (410 [129-633] vs 39 [20-89], minutes, p=0.003). During OGTT, a marked increase in the blood glucose nadir (44 ± 11 vs 56 ± 10, mg/dl, p= 0.038) was found after LG treatment. Conversely, no difference were observed in fasting glycemia, in the time-to-nadir, as well as in the blood glucose zenith nor the time-to-zenith. After treatment, the peak of GLP-1 was attenuated and total GLP-1 AUC significantly decrease (7.6 ± 4.1 vs 6.5 ± 3.8, nmol/L*min, p= 0.043), as well as potentiation factor ratio (1.5 ± 0.5 vs 0.8 ± 0.4, p= 0.038) and total insulin AUC (57±12 vs 49±9, nmol/m2 , p= 0.041). Conclusion: LG is effective on reducing frequency, overall duration and severity of PBH episodes. The main mechanism by which LG acts appears to be the attenuation of GLP-1 peak and, consequently, the second phase of insulin secretion in response to the glucose load. Further specific studies are necessary to evaluate the effects of LG on the microbiota and inflammation to better understand the mechanisms of action of this precious supplement.
