AbstractRanid frogs of the genus Amolops occur in Southeast Asia and are typically found near waterfalls. Their phylogenetic relationships have not been resolved. We include 2,213 aligned nucleotide sites of the 12S, 16S and tRNAval gene regions of the mitochondrial DNA genome from 43 individuals of Chinese and Vietnamese Amolops, Huia, Hylarana, Meristogenys, Odorrana and Rana. The outgroup species were from the genera Chaparana, Limnonectes, Nanorana, and Paa. The data were analyzed within the framework of a refutationist philosophy using maximum parsimony. Previous analyses of molecular and larval morphology have suggested that Meristogenys amoropalamus is composed of two cryptic species, but no diagnostic characters of their adult morphology have been reported. Here, we compared adult characters of these two species and found that they differed in iris colour (yellowish-green and sandy brown), tympanum size and relative limb length. Based on the results of analysis of DNA sequences of the type specimens and a discriminant analysis using 18 morphological variables, we conclude that the lineage with green irises is the true M. amoropalamus, and t

We identified three distinct sympatric lineages of frogs among specimens previously considered a single species (Meris- togenys amoropalamus Matsui), based on 909 bp of mitochondrial DNA (12S rRNA and cytochrome b). To seek evidence of reproductive isolation between these lineages, we first analyzed a 249-bp fragment of the nuclear proopiomelanocortin (POMC) gene and found five haplotypes, of which two were limited to lineage 1 and three belonged to lineages 3 and 4. In a subsequent phylogenetic analysis of a 1313-bp fragment of nuclear POMC, Rag-1, and rhodopsin, lineage 1 was again distinct,
