ABSTRACTDuring experiments conducted within the vent pool of Medusa Geyser, Norris Geyser Basin, Yellowstone National Park, USA, amorphous opaline silica (opal-A) was deposited on/within plant tissues within 30 days of immersion. Initially, deposition created inter/intra-cellular films which lined cell walls plus intercellular colloid suspensions (sols) of opal-A nano/microspheres. By 330 days, opal-A deposition created a robust external and internal matrix that stabilised tissues against collapse and replicated plant structure. Opal-A films increased to micron-order thicknesses and intracellular sols were created. Systematic variation of opal-A fabric between tissues comprising living/dead cells at the time of deposition indicate that cell function, architecture and shape influence fabric development. Heterogeneity of opal-A fabric within adjacent cells of similar structure/function indicates spatially/temporally fluctuating physicochemical conditions and the presence of intraorganic microenvironments. Early deposition of opal-A films suggests a period of low silica supersaturation and slow opal-A deposition. In contrast, intracellular sols suggest high levels of supersaturation, and rapid opal-A deposition. Shell-like microsphere growth suggests cyclic variation of silica supersaturation, and alternations between rapid and slower opal-A deposition. Microsphere growth to the upper limit of colloidal stability and colloidal crystal structures indicate prolonged sol stability, whilst floc-like microsphere networks indicate localised sol instabi

AbstractThe Maricá restinga, located in the eastern part of the Rio de Janeiro State (Brazil), corresponds to one of the few remaining preserved areas of the state's coastal plain. This paper reports on a study of the Maricá restinga plant communities and also presents an identification of the main plant species present in each community, with the objective of establishing reference collections, by the methods of the proxies opal phytoliths and stable carbon isotopes, for paleoenvironmental reconstructions of this coastal area during the Quaternary. Six plant communities, distributed perpendicularly to the coast line over sandy barriers, lagoonal plain, lagoon margin and weathered basement were identified: halophile-psamophile, scrub, herbaceous swamp, slack, shrubby vegetation and dry forest. In general, the plant species analyzed in each community presented low productivity of opal phytoliths, as only the Poaceae, Cyperaceae and Arecaceae families produce a great amount and diversity of morphotypes of opal phytoliths. The results of the analysis of stable carbon isotopes in sediments indicated a predominance of C3 or a mixture of C3 and C4 plants, presenting a close correlation with the results found in plants collected in each community. In conclusion, it was verified that the carbon isotope analysis associated with that of the opal phytoliths are good proxies for the reconstruction of vegetation in the study area.

Unconventional fertilizers can act as elicitors to encourage the synthesis of phyto-pharmaceuticals in aromatic plants. In the present research, the effects of factorial combination between two red basil cultivars, ‘Opal’ and ‘De Buzau’, and four fertilization types, biosolids, organic, microorganisms and chemical, plus an unfertilized control, were assessed on fresh and dry yield, biometrical parameters, soil plant analysis development (SPAD) and antioxidant compounds and activity. Chemical fertilization increased fresh yield compared with the control, with no difference in organic and microorganism fertilization regarding dry weight. ‘De Buzau’ enhanced the number of lateral stems and plant height, the latter being better affected by chemical and microorganisms compared to the control. Chemical fertilization showed the highest leaf dry matter, nitrate content and SPAD, whereas the control showed the lowest. Compared to the unfertilized control, biosolids increased total phenolics in ‘Opal’; microorganisms, organic and biosolids enhanced total flavonoids in ‘Opal’, with the same effect under microorganisms and organic treatments in ‘De Buzau’. Total anthocyanins showed the highest content in ‘Opal’ under organic fertilization. The highest antioxidant activity in the basil extracts was detected under microorganisms and organic applications in ‘Opal’. The present investigation results demonstrate that unconventional fertilizers increase the synthesis of antioxidants and represent a sustainable alternative to chemical fertilization for growing red basil.

Plant genetic improvement<br />can be conducted through genetic engineering.<br />Parthenocarpic fruit production could increase fruit<br />production and its qulities. IAA genes were introduced into<br />three tomato cultivars Ratna, Opal and LV 6117 using two<br />constract genes DefH9-iaaM and DefH9-RI-iaaM. The iaaM<br />gene is able to increase auxin biosynthesis in transgenic<br />plant cells and organs because indol-eacetamide,<br />synthesized by the product of the iaaM gene, is converted<br />either chemically or enzimatically to indole-3-acetic acid<br />(IAA), while the promotor DefH9 enable IAA gene expressed<br />specifically in the ovules. The objectives of this experiment<br />was to identify gene introduction into plant genom of three<br />tomato cultivars. The factors tested were two constract of<br />IAA genes (DefH9-iaaM or DefH9-RI-iaaM), tomato cultivars<br />(Ratna, Opal, and LV 6117) and time of explant inoculation<br />(5, 15, 30 minute). The result showed that the best time<br />inoculation was 5 minute. Otherwise three tomato cultivars<br />response better to DefH9-RI-iaaM than DefH9-iaaM. The total<br />efficiency of regeneration and total efficiency of<br />transformation of both genes were 25.38% and 20.32%. PCR<br />analysis showed that 10 plant have positive PCR, were 1<br />plant carried (Opal) DefH9-iaaM gene and 9 plant (Ratna,<br />Opal, LV 6117) carried DefH9-RI-iaaM gene.</p>

The objective of this study was to evaluate growth regulators with purpose of reducing the size of heliconia ‘Red Opal’ potted plants. The experiment was carried out in randomized block design with five treatments (trinexapac-ethyl and paclobutrazol at rates of 37.5 and 75.0 mg of active ingredient per pot and control without growth regulator) and five replicates. The treatments were applied 40 days after planting the rhizomes in pots filled with soil. Thirty and 150 days after the growth regulator application, plant height, number of leaves and shoots, petioles length and leaf area were evaluated. One year after planting the rhizomes in pots the number of inflorescence and leaves (leaves, sheathing leaf bases and inflorescences) and rhizomes (rhizomes and roots) dry mass were determined. Trinexapac-ethyl had no differences compared to the control in any of the variables evaluated. Paclobutrazol proved effective in reducing plant height, leaf area and petiole length and increase in number of leaves and shoots but the effect was temporary. Also, it did not affect the inflorescences production and leaves and rhizomes dry mass. Paclobutrazol is efficient to promote height reduction and to increase the number of shoots in heliconia ‘Red Opal’ potted plants without affect the inflorescence formation but its effects is temporary.

Opal phytoliths in the leaves of 10 native species growing in and around a swamp were isolated and quantified, and shape and surface detail examined with a scanning electron microscope (SEM). The amount of plant opal in the leaves ranged from 0.10 to 2.45% by dry weight, and phytolith forms most commonly found were spheres, rods and sheets. Phytoliths in the silt size range of the swamp sediment were isolated by a simple fractionation technique. It was found that the forms with a small surface area to volume ratio survived in the sediment.

The objective of this work was to evalue the effect of bio-stimulant Stimulate applicated or not in differents development stages of cotton plant cv Delta Opal, cultivated in a vegetation house and submitted or not to hydric stress. There were applicated 25, 60 and 100% of total volume of pores (VTP) filled out with water and Stimulate (registrated mark) applicated on the seed and on the flower botton stage and without biostimulant application 69 days after sowing. The hydric stress period of time was 15, 30 and 45 days after hydric stress application. The experimental delineation was wholly randomized in a 3x3x3 factorial scheme, with four repetitions of five plants each one. The cotton plant cv Delta Opal did not support hydric eack of 25% VTP on the flower botton beginning, being observed the minor heights, lap diameters, number of flowers, and endangering the formation of reproductive frames. The bio-stimulant dose and application form did not offer improvement on plants development when submitted to eack of water, but on plants submitted to water excess, a lap diameter increase occurred
O objetivo do trabalho foi avaliar o efeito do bioestimulante Stimulate (marca registrada), aplicado ou não em diferentes fases do desenvolvimento do algodoeiro cv. Delta Opal, cultivado em casa de vegetação e submetido ou não ao estresse hídrico. Foram aplicados 25, 60 e 100% do volume total de poros (VTP) preenchidos com água.e o Stimulate® aplicado na semente, na semente e na fase de botão floral e sem a aplicação do bioestimulante, aos 69 dias após a semeadura. O período de duração do estresse hídrico foi de 15, 30 e 45 dias após a aplicação do estresse hídrico. O delineamento experimental empregado foi inteiramente casualizado em esquema fatorial 3 x 3 x 3, com quatro repetições de cinco plantas cada. O algodoeiro cv Delta Opal não tolerou deficiência hídrica de 25% VTP iniciando na fase de botão floral, sendo observadas as menores alturas, diâmetros de colo, número de folhas, e comprometendo também a formação de estruturas reprodutivas. O bioestimulante na dose e na forma aplicada não proporcionou melhoras no desenvolvimento das plantas quando submetidas à falta de água, já em plantas submetidas ao excesso de água, houve aumento do diâmetro do colo das plantas

Research on the "Third Dementia Management Comprehensive Management Plan (2016 - 2020)" that comprehensively summarizes the efforts of Korean dementia policy and reflects the needs that elderly people with dementia have in their lives and the situation of the efforts being implemented by the plan was examined by using the theory framework of theory evaluation. In order to revise the plan, I excluded the evaluation centered the conventional performance measurement and empirically clarified the validity of the program from the perspective of elderly people with dementia.

This thesis presents HELIOS, an in-house laboratory for time-resolved pump-probe spectroscopy with extreme-ultraviolet (XUV) probe radiation. A wide span of pump wavelengths can be generated using commercial laser equipment while XUV probe radiation is generated via a high harmonic generation process in a noble gas delivering probe photons with energies between 20 eV and 72 eV. The XUV beam path features a time-preserving monochromator and was constructed and built in-house. HELIOS features an overall time resolution of about 50 fs when using 800 nm pump and 41 eV probe photons. An energy resolution of 110 meV at 41 eV photon energy can be achieved. HELIOS features two beamlines. One µ-focus beamline with an XUV focal size of about 20 µm can be used with experiments that require such a small XUV focal size as well as with different end stations. The other beamline features a semi-permanently mounted end station for angle-resolved photoelectron spectroscopy under ultra-high vacuum conditions. Experiments demonstrating the usability of HELIOS and the two beamlines are presented. A pump-probe measurement on graphene demonstrates the capability of determining a large part of the k-space in only one measurement due to the use of an ARTOF angle-resolved time-of-flight electron spectrometer. A non-angle-resolved pump-probe measurement on the conducting polymer PCPDTBT demonstrates the high signal-to-noise ratio achievable at this beamline in non-angle-resolved photoelectron-spectroscopy pump-probe measurements. The usability of the µ-focus beamline is demonstrated with time-resolved measurements on magnetic samples employing an in-house-designed spectrometer. These experiments allow the retrieval of element-specific information on the magnetization within a sample employing the transversal magneto-optical Kerr effect (T-MOKE). Additionally, a Fourier transform spectrometer for the XUV is presented, the concept was tested at a synchrotron and it was used to determine the longitudinal coherence of the XUV radiation at HELIOS.

A previous study indicates OPEL is a novel elicitor, which induce cell death, reactive oxygen species (ROS) accumulation, callose deposition, PAMP-triggered immunity (PTI), and systemic acquired resistance (SAR) on Nicotiana tabacum. In this study, we focus on OPEL homologous genes to verify whether OPEL is a pathogen-associated molecular pattern (PAMP) or not. Phylogenetic analysis OPEL homologous genes by tblastn search on NCBI, JGI and Broad institute genome database has shown homologs of OPEL have gene duplication in oomycetes. Besides, oomycetes plant pathogens have the most copy number of OPEL homologous genes. Analysis domain conformation of OPEL homologs indicates thaumatin-like domain is a distinct domain in oomycetes plant pathogen. Phylogenetic analysis glycoside hydrolase family 16 (GH16) in Phytophthora parasitica demonstrated OPEL homologous genes are different from other GH16 genes. These study imply OPEL homologs are unique genes in oomycetes. To characterize the interaction between OPEL homologs and plants, we overexpress OPEL homologs of P. parasitica by using agroinfiltration. PPTG_17496 induces necrosis on N. benthamiana, and PPTG_16550, PPTG_17497, and PPTG_17498 induce small region cell death on N. tabacum. Both results indicate OPEL homologs are PAMPs. However, PPTG_17496 consume 10 days to establish necrosis symptoms on N. benthamiana, which may be caused by DAMP-triggered immunity (DTI) but PTI. We need further research to find out OPEL homologs induce PTI or DTI on plants.

To combat invasion by pathogens, plants have developed different defense mechanisms, including pattern-triggered immunity (PTI), which is elicited upon the perception of pathogen-associated molecular patterns (PAMPs) or danger/ damage-associated molecular patterns (DAMPs) by plant pattern recognition receptors (PRRs). Previously, a novel elicitor protein named OPEL was identified in the cultural fluid of Phytophthora parasitica, a notorious pathogen of a wide variety of economically important crop species. It is characterized by the presence of thaumatin-like domain, glycine-rich domain and glycoside hydrolase 16 (GH16) domain. Infiltration of OPEL recombinant protein into the leaves of tobacco (Nicotiana tabacum cv. Samsun-NN) resulted in PTI responses such as cell death, callose deposition, reactive oxygen species (ROS) production, and induced expression of salicylic acid-responsive defense genes, and the GH16 domain is indispensable for OPEL to induce PTI. Search of NCBI databases identified in total 25 GH16-containing genes in the genome of P. parasitica. Conserved sequence analysis showed that PPTG_16550, PPTG_17496, PPTG_17497, and PPTG_17498 are highly conserved with OPEL, thus they are very likely close homologs of OPEL. The purpose of this study is to investigate the role of OPEL homologs in plant basal defense response. Analysis by quantitative reverse transcriptase-PCR showed that all four genes showed different expression patterns. Especially, the expression of PPTG-17497 and PPTG_17498 was significantly induced at 12 hours post pathogen inoculation. Of the four putative OPEL homologs, PPTG_16550 is unique in that it lacks the thaumatin-like domain. To know its role in plant defense, PPTG_16550 recombinant protein was expressed in Escherichia coli, which caused cell death, reactive oxygen species (ROS) production, and callose deposition upon infiltration on the leaves of tobacco. Nevertheless, this protein shows no activity to induce systemic acquired resistance against Tobacco mosaic virus (TMV). These results indicate that PPTG-16550 is an elicitor, which elicits PTI response in tobacco. Its role in the process of plant infection by P. parasitica awaits further investigation.

O Plano Estratégico (“o Plano”) da Organização Pan-Americana da Saúde (OPAS) define a direção estratégica da Organização com base nas prioridades coletivas dos seus Estados Membros e na atenção voltada para os países, além de especificar os resultados em matéria de saúde pública a serem alcançados durante o período 2020-2025. O Plano estabelece o compromisso conjunto dos Estados Membros da OPAS e da Repartição Sanitária Pan-Americana (RSPA ou “a Repartição”) para os próximos seis anos. Os Estados Membros da OPAS deixaram claro que o Plano Estratégico é dos principais instrumentos para a implementação da Agenda de Saúde Sustentável para as Américas 2018-2030 (ASSA2030) e, por conseguinte, para a consecução dos Objetivos de Desenvolvimento Sustentável (ODS) relacionados à saúde na Região das Américas. Os 11 objetivos do ASSA2030 formam os objetivos no nível do impacto deste Plano. Com o tema A equidade, o coração da saúde, este Plano busca catalisar esforços nos Estados Membros para reduzir as iniquidades em saúde dentro dos países e territórios e entre eles, a fim de melhorar os resultados em termos de saúde. O Plano identifica ações específicas para enfrentar a desigualdade em saúde, como as recomendadas pela Comissão sobre Equidade e Desigualdades em Saúde nas Américas, com a orientação da Comissão de Alto Nível para a Saúde Universal. Quatro temas transversais são centrais para o enfoque deste Plano para abordar os determinantes da saúde: a equidade, o gênero, a etnia e os direitos humanos. Além de destacar um enfoque multissetorial integrado, este Plano aplica estratégias de saúde pública baseadas em evidências, como a promoção da saúde, o enfoque da atenção primária à saúde e a proteção social em saúde, para abordar os determinantes sociais. The contingency plan was carried out for the seven highest villages in Sleman Regency, Yogyakarta Special Region, as areas prone to eruption of Merapi Volcano. This contingency plan was prepared based on a scenario with a volcanic eruption index of 3, in the form of the collapse of a lava dome of 5 million cubic meters. The collapse of the lava dome formed a hot cloud that moved 6–9 km towards the Gendol River, Opak River, Kuning River, Boyong River and Krasak River. The number of exposed residents is 18,880. The emergency situation period is estimated to be 30 days, from the time the status is increased to Alert. This contingency plan was prepared in a participatory and collaborative manner, under the coordination of the Sleman Regency Regional Disaster Management Agency (RDMA). This process includes: (1) Updating the village level contingency plan as materials for the district level. (2) Formation of a substance team from local government, universities and non-governmental organizations, and (3) Conducting a series of workshops for related organizations to compile documents and equate perceptions on the adaptation of contingency plans with the Covid 19 health protocol. The preparation of this document also involves the participation of children, in order to express their opinions and needs in implementing emergency management.

In January 1957 the military and colonial government accepted a master plan drawn up by the anthropologist Jean Servier to undertake Opération Pilote in the Chelif region, the biggest counterinsurgency (COIN) experiment of the Algerian War. The arrival of Salan and Indochina specialists in command accelerated the adoption of the doctrine of revolutionary warfare, that victory over the FLN could not be achieved by conventional ‘big’ force operations, but only through winning over the indigenous population. The catastrophic failure of counterinsurgency in Kabylia in late 1956 diverted attention to the Chelif where a dispersed population was seen as advantageous to COIN operations. Servier’s plan was linked to the revival of Lucien Paye’s communal reform of 1945 to 1948, seen as the key reform to retain Algérie française. By late 1956 the colonial government and military had rapidly lost control of the Dahra and Ouarsenis mountains, a collapse signalled by the evacuation of isolated colons, and the aim was to ‘reconquer’ the interior through driving the ALN and its OPA structures from each douar, and replacing it by ‘djemâa amie’ that could form the core of future rural municipalities in which peasants could take on board their own local government and auto-defense. Unable to guarantee open elections Opération Pilote aimed to secretly train future douar leaders in the psychological warfare centre at Arzew.

Since Assistant Director Leopold was not needed at the lab until July 1, he took the train with Starker and Luna to Burlington to visit his mother and brothers. (Marie had married and moved to Illinois with her husband.) Together again, the “boys” immediately began plans for a trip—this time to Carhart’s proposed wilderness area in the boundary waters of Minnesota for two weeks of canoeing, camping, and fishing. Prior to trip departure, though, Aldo had to travel to Madison to prepare the way for the family move. Starker and his uncles Carl and Frederic headed straight to Ely, Minnesota, to arrange for the canoes and equipment (Luna stayed with Opa Leopold to await the arrival of the rest of his family). Their missions accomplished, Aldo and the other adventurers met and set off into the wilds of the Superior National Forest and the Canadian Quetico. The Leopold brothers had at last reached their magical North. For two weeks, the voyageurs paddled past granite outcroppings and tall pines, fishing for trout and listening to the mournful wail of loons. Aldo wrote in his journal, “The number of adventures awaiting us in this blessed country seems with­out end.” Later he added, “How Dad would have loved it! I am reminded of Isaac [sic] Walton’s terse but loving tribute—‘an excellent angler, now with God.’” Leopold and Starker returned to Madison, an “awfully dolled up town.” He had written earlier to Estella: “Nobody has any backyard except in the slums—not even a woodshed or fence or chickens—the backyards are open lawns just as the fronts. We have a little garden room at this place I am thinking of—which is not usually the case, but nobody keeps chickens, so we would do so at our own peril!” The rest of the family arrived shortly, and the Leopolds moved into a rented home “not far from the country.” Within the month, they bought a stucco house about a mile from the lab—at 2222 Van Hise Avenue—and their energies went into transforming it into a real home.

Introdução: O melão Cucumis melo L. é um fruto de grande importância econômica, com amplo consumo no mercado nacional e internacional. No Brasil, a região Nordeste destaca-se como a principal produtora dessa fruta. No entanto, a espécie sofre com diversos ataques ocasionados por uma vasta gama de pragas que estão associadas a esses cultivos. Dentre elas, está o fitonematóide Rotylenchulus reniformis, responsável por reduzir a qualidade e quantidade das produções agrícolas. O estudo da variabilidade genética desse organismo é relevante, pois as variações gênicas entre o fitopatógeno podem estar relacionadas à diferentes níveis de patogenicidade. A seleção de marcadores de DNA informativos representa uma etapa significativa neste tipo de estudo, pois possibilitará a economia de recursos e tempo investidos. Objetivo: O presente trabalho teve como objetivo a identificação de marcadores RAPD polimórficos para futuros estudos de diversidade genética dessa espécie. Material e Métodos: Para isso, o DNA do fitonematóide foi inicialmente extraído com kit de extração de DNA NucleoSpinTissue (Macherey-Nagel). Posteriormente, 80 primers RAPD foram testados quanto ao seu nível de polimorfismo e qualidade de bandas amplificadas gerada nessa espécie. Resultados: Como decorrência da triagem dos 80 iniciadores RAPD, foi constatado que 71 desses não amplificaram, ou obtiveram bandas fracas e não reprodutíveis, sendo selecionados somente nove iniciadores. Desse modo, 70 bandas foram amplificadas, com média de 7,78 por iniciador, das quais 67 foram polimórficas, gerando 95,81% de polimorfismo. Dos nove iniciadores, seis apresentaram 100% de polimorfismo. O iniciador menos eficiente foi o OPA-03. Dois iniciadores apresentaram maior quantidade de bandas amplificadas (OPA-16 e OPAA-04) sendo o iniciador OPAA-04 mais informativo (100% de polimorfismo). Conclusão: A identificação desses marcadores polimórficos ajudará a compreender, em estudos posteriores, a correlação da diversidade genética com interações planta-patógeno desse fitonematóide.

Los avances científicos y la dinámica de la sociedad hacen un llamado constante a la generación de nuevo conocimiento. Es por esto que las instituciones educativas han fomentado la creación de comunidades de aprendizaje para que articulen el conocimiento que surge entre el currículo y la investigación, siendo los semilleros de investigación una estrategia para tal fin. En este sentido, el Grupo de Investigación en Optimización y Organización de Sistemas Productivos, Administrativos y Logísticos-OPALO de la Universidad Industrial de Santander, ha institucionalizado sus semilleros de investigación como estrategia para el fortalecimiento de la formación para la investigación. De ahí que, ha diseñado y establecido un Plan de Formación para que, con el apoyo de los estudiantes de maestría, profesores cátedra y planta se logre la formación y vinculación de los estudiantes de pregrado en ingeniería industrial. El Plan de Formación de los semilleros de investigación está compuesto de tres fases, en donde la Fase 1 denominada Seminario de Habilidades Investigativas tiene como objeto contextualizar a los estudiantes en el mundo de la investigación científica y desarrollar habilidades esenciales para iniciar con el proceso investigativo mediante una formación metodológica. La Fase 2, está dirigida a estudiantes que han cursado previamente la Fase 1 y que están interesados en continuar con el proceso de aprendizaje a través de las líneas de investigación del grupo. En esta fase los estudiantes desarrollan habilidades básicas de programación en herramientas computacionales de optimización y análisis de datos que contribuyen a la formación integral. Finalmente, la Fase 3 está dirigida para aquellos estudiantes interesados en el desarrollo de propuestas y proyectos de investigación para ser presentados en revistas científicas, eventos académicos y/o como modalidad de trabajo de grado. El impacto que ha tenido los semilleros de investigación en los últimos tres años ha sido la masificación de los mismos en donde, por ejemplo, en los semestres académicos del año 2019 participaron 10 estudiantes de maestría apoyando las sesiones teórico-prácticas y, se certificaron en la Fase 1 163 estudiantes, 39 estudiantes en la Fase 2 y 94 estudiantes de pregrado quienes estuvieron vinculados al grupo OPALO con proyecto de grado bajo la modalidad de trabajo de investigación.

A reduction of CO2 emissions of aero engines can only be achieved by a reduction in the fuel consumption. For turbofan engines a major key for this is an increase in the engine bypass ratio to enhance the propulsive efficiency. This leads to an increased mass flow in the bypass duct and hence higher contributions of the bypass duct system to the flow pressure losses and hence the engine fuel consumption. Additionally the turbofan core engine gets smaller in size and the overall diameter of the engine increases. This requires advanced engine mounting concepts, such as core mounted subsystems, which influence the arrangement and the mechanical and aerodynamic design of components in the bypass duct such as fan outlet guide vane, struts, fairings or bifurcations. This requires an optimized design of the turbofan bypass system with regards to structural, aerodynamic and acoustic criteria. The topology of the bypass duct as well as the position and the design of the individual components have a significant impact on the weight, the efficiency and the noise radiation of the engine and hence need to be investigated in a systematic approach. Such a design approach was developed in the R&T project OPAL, led by Rolls-Royce Deutschland and funded by the German Federal State of Brandenburg. The design approach covers the following steps: - Optimization of the bypass duct shape with regards to minimum pressure loss taking into consideration the mechanical requirements of an engine with core mounted subsystems. - Optimization of strut, fairing and bifurcation shapes with regards to minimum pressure loss, robustness against flow deviations and minimized upstream flow effects taking into consideration structural and mechanical requirements by engine loads and subsystem routings. - Optimization of fan outlet guide vane profile and plan form shape with regards to minimum pressure losses and maximum working range taking into consideration structural and acoustic constraints. - Optimization of fan-outlet guide vane, strut and bifurcation interaction with regards to minimum pressure losses and maximized aerodynamic stability of the fan and outlet guide vanes. The current paper will present the design approach, the optimization processes and the results of the optimization of the turbofan bypass duct system for the application on modern high-bypass ratio aero engines.

Abstract Mahakam Block has been in operation for nearly half a century with cumulative production of approximately 20 trillion cubic feet of gas and 1.5 billion barrels of oil. Mature field challenges have become more evident as portrayed by declining production, more complex surface constraints, more challenging profitability of new projects and decreasing resources of new wells, which are also exacerbated by external factors such as volatility of oil and gas prices. Despite the aforementioned challenges and complexity in terms of operating numerous fields with different characteristics, Mahakam is currently still one of the biggest producing blocks in Indonesia. The success of sustaining production and prolonging the life of Mahakam is the result of continuous innovations, improvements and optimizations on various aspects over the years. Subsurface innovative ideas by restudying and redefining geological concepts has led Pertamina Hulu Mahakam (PHM) to drill step-out wells in Handil, Tunu, South Mahakam and Sisi Nubi fields that deliver positive results and open new opportunities. In the non-subsurface aspect, Indonesia's first Plan of Development that combines higher and lower value projects across fields called OPLL (Optimasi Pengembangan Lapangan-Lapangan) was initiated in order to develop fields with marginal value and to achieve economy of scale. Moreover, Capital Expenditure (CAPEX) optimization through evolution of platform design, well architecture and sand control method is crucial for exploitation of targets with lower resources over time. PHM has also launched CLEOPATRA (Cost Effectiveness and Lean Operations in Mature Asset), later renamed to LOCOMOTIVE-8 (Low Operations Cost of Mahakam to Achieve Effectiveness and Efficiencies), to achieve Operating Expenditure (OPEX) efficiency through various initiatives driven by each entity. Due to cost of money, budget accuracy is as important as expenditures reduction meaning that more detailed and deterministic budget estimation is necessary. In addition to optimizing cost structure, PHM strives to carry out gas commercialization efforts to improve revenue streams. In this rapidly changing era, especially for Mahakam, paradigm shift becomes highly critical. Changes in the structure and size of organization is essential to adjust with business dynamics. Adaptive organization structure is performed through digitalization and competency improvement to reduce repetitive tasks and increase productivity per capita. Cooperation between neighboring companies brings mutual benefit by sharing rig, transportation means, and pipeline network systems. Mutual benefit opportunity is also available between the company and Indonesian government by amendment of fiscal terms with the aim to enable the execution of sub-economic projects. Ultimately, one effort alone may be insignificant, but the combination of all of the efforts will be the key to the continuation of Mahakam story.

Hybridization of Back cross is one method to get varieties that are resistant to downy mildew. The purpose of this study was to obtain information on inheritance characteristics of downy mildew resistance. This research was conducted at the experiment center of Agro-Technology Study Program of Agriculture Faculty, University of Trunojoyo Madura. Research of Assessment of resistance to Downy Mildew used a randomized block design with 18 treatments (P1, P2, F1, F2, BC1P1 and BC1P2 in three sets of crosses, namely LGL x Mdr-3, T12 x Mdr-1 and E02 x Mdr-2) and three replications so there were 54 experimental units. Identification of polymorphic RAPD markers for endurance to downy mildew through Bulk Segregant Analysis (BSA) was done by amplifying the DNA in the resistant pool and susceptible pool. The random primers used were 120 primers from 6 operon groups, namely OPA, OPB, OPC, OPD, OPF and OPG. OPC-07 was a marker that was linkage close to the resistance to downy mildew with a genetic distance of 1.9 cM.
