Animal behaviour is coordinated by biological rhythms over multiple timescales. In the temporal cortex, rhythmic oscillatory activity in the 5 to 12 Hz theta frequency range supports behaviour through temporal windows of changes in excitability and circuit computation. Subcortical inputs, including those from the medial septum are thought to generate the theta oscillatory activity. However, the rules of innervation of identified types of cortical cells in different cortical regions by a diverse population of septal neurons are largely unknown. Mutations in the leucine-rich repeat kinase 2 (LRRK2) gene are known to cause late-onset, familial forms of Parkinson’s Disease, with prevalence up to 40% in specific populations. These mutations are also linked to sporadic PD, highlighting the importance of the related protein in the development of the disease. LRRK2 is a large, complex, multi-domain protein, with two main enzymatic activities: a GTPase domain (bearing three pathogenic mutations) and a kinase domain, in which the most common mutation (G2019S) is found. 博士
國防醫學院
生命科學研究所
101
Neurodevelopmental disorders, including autism, schizophrenia and attention deficiency and hyperactivity disorder, are complex neuropsychiatric diseases. Both environmental factors and genetic deficits contribute to the pathogenesis of neurodevelopmental disorders. The most well-known environmental factor is prenatal immune challenge caused by infection. Robust cytokine production induced by inflammatory response is believe to affect neural development and leads to several psychiatric disorders. However, the detail mechanism is not clear yet. Sensory deprivation in one modality can enhance the development of the remaining modalities via mechanisms of synaptic plasticity. Mice of C3H strain suffers from RD1 retinal degeneration that leads to visual impairment at weaning age. Independently on the retinal degeneration there is also present olivocerebellar degeneration caused by Lurcher mutation. This neurodegenerative disorder causes motor deficits, increased CNS excitability as well as changes in synaptic plasticity. The aim of this study was to evaluate a role of whiskers in compensation of the visual deficit and to assess the influ

PART I Animals need to constantly evaluate their external environment in order to survive. In some cases the internal state of the animal changes to cope with it’s surrounding. In our study we wanted to investigate the role of amines in modulating internal states of Drosophila. We have designed a behavioral paradigm where the flies are fixed in space but can walk on a small styrofoam ball suspended by a gentle stream of air. The walking activity of flies was used as behavioral readout. PART I Animals need to constantly evaluate their external environment in order to survive. In some cases the

Interestingly, recent evidence suggests novel roles of Tau in synaptic structure and function. On the other hand, Tau hyperphosphorylation and subsequent malfunction is suggested to be a key parameter of synaptic dysfunction and atrophy and memory loss that characterized Alzheimer´s disease. Indeed, cumulative evidence from experimental studies demonstrated that absence of Tau blocked Aβ detrimental properties on synaptic dysfunction and loss

However, poorly managed pain during this period will result in long-term physiologic and behavioral consequences. Animal studies reveal that neonatal peripheral inflammatory pain results in long-standing changes on the development of nociceptive neuronal circuitry. Morphologically, fine primary afferent fibers termination area in spinal cord dorsal horn expands longitudinally and sprouts into deeper lamina. Electrophysiologic study shows increase in
