Abstract We describe a new analytical approach--"capsule chemistry"--for high-speed, selective analysis of a wide variety of analytes. Sequential micro-aliquots of sample and reagents are encapsulated within an inert fluorocarbon liquid. The resulting "test capsule" is introduced into a single analytical flow path, composed of a solid fluorocarbon, Teflon, where the sample is incubated, mixed, reacted, and measured as a moving series of individual tests. The gas-phase proton affinities of several highly basic amino acids and diamines were determined using the extended kinetic method in an ESI-quadrupole ion trap instrument. The non-protein amino acid L-canavanine is structurally related to L-arginine with an oxygen substituted for the terminal methylene group of L-arginine and is highly toxic to humans. The proton affinity of L-arginine, a protein amino acid, was determined to be 1036 kJ mol -1, whereas the proton affinity of L-canavanine was determined to be 1005 kJ mol-1. The disulfide bond pattern in the galactose-specific lectin 24K from the egg jelly of the Chinook salmon Oncorhynchus tshawytscha was determined, and its previously reported amino acid sequence was confirmed by mass spectrometry. A combination of tryptic digestion, HPLC separation, and chemical, modifications was used to establish the location of seven disulfide bonds and one pair of free cysteines. Chemistry<br>Ph.D.<br>This dissertation details an investigation into the use of laser pulses from a titanium:sapphire regenerative amplifier laser system to vaporize analytes in ambient air for mass spectral analysis. The laser system was modified to operate in one of two distinct modes. In femtosecond (fs) mode the laser produced 2.5 mJ, ~60 fs laser pulses centered at 800 nm. In nanosecond (ns) mode the laser produced 2.4 mJ, ~10 ns laser pulses centered at 800 nm. The LMFBRs (Liquid Metal Cooled Fast Breading Reactors) is a key technology for the future electricity demand. Therefore, the establishment of safety of LMFBRs is deeply desired. To evaluate the safety of LMFBR steam generators, empirical studies have been performed for the ruptures of heat transfer tube caused by the overheat due to sodium-water reaction in case of a practical scale conditions. But few systematical experiments have been performed for the clarification of phenomena from the viewpoint of thermal hydraulics and physical chemistry.
