While forage plays an important role in equine nutrition, little research has been conducted evaluating fiber utilization by young horses. Therefore, studies were conducted to compare in vivo digestibility and digesta passage in weanlings and mature horses (Exp 1) and yearlings and mature horses (Exp 2). The objective of this thesis was to develop new methodologies to assess upper gastro-intestinal function in health and disease. Several different technologies were studied in a range of upper gastro-intestinal diseases and adapted to try and provide more meaningful insights. The thesis has three main sections. This work describes a biochemical, transcriptomic and proteomic analysis of the midgut and midgut glands (MMG) and digestive juice (DJ) from Nephilengys cruentata and Tityus serrulatus MMG. Cathepsin L, B, D and F, legumain, trypsin, astacin, carbohydrases and lipases were identified by these approaches. The project evaluated potential of soluble cellulose as a cheap feed ingredient for major farmed Australian freshwater crayfish species testing their growth performance, digestive enzyme activity and digestive enzyme gene expression patterns. Test animals showed an innate capacity to utilise a range of carbohydrate sources including complex structural polysaccharides. Results suggest that more plant-derived ingredient can be incorporated in formulated low-cost feeds for the culture industry.

The first chapter of this thesis describes the study of intestinal stem cells (ISCs) and their relationship to colon cancer. We describe the transcriptional landscape of intestinal epithelial populations through the use of the surface marker EphB2 which allowed us to build an ISC-specific gene signature. Elevated levels of this signature characterize CRC patients with high risk of disease relapse after therapy.
