The filamentous fungus Aspergillus niger is an industrially exploited protein expression platform, well known for its capacity to secrete high levels of proteins. To study the process of protein secretion in A. niger, we established a GFP-v-SNARE reporter strain in which the trafficking and dynamics of secretory vesicles can be followed in vivo. The biological role of putative A. niger orthologues of seven secretion-specific genes, known to function in key aspects of the protein secretion machinery in Saccharomyces cerevisiae, was analysed by constructing respective gene deletion mutants in th

The Niger Delta Region, although the back bone of Nigerian economy, has been neglected by successive governments over time. There are no good schools, no health facilities, no water and other facilities within this area. The problem of the area is compounded by lack of good transportation system. This has been a hindrance both for security agencies and the people in the restive area. A good transportation system should ensure that people, goods and services move speedily, conveniently, safely and cost effectively. A nystatin-resistant mutant of Aspergillus niger is described that has quantitative changes in phospholipid composition compared with the wild type. The proportion of linoleic acid in total phospholipids was elevated in the mutant from 38 to 56% and the proportion of oleic acid was lowered from 28 to 12%. Amino acid transport is also altered, the Vmax values for the uptake of leucine, lysine, and glutamic acid being generally higher in the mutant; for glutamic acid the increase was from 15 to 35 nmol∙mg−1∙min−1. Excretion of organic acids, e.g. citrate, by anamorphic fungi is a frequent phenomenon in natural habitats and in laboratory cultures. In biotechnological processes for citrate production with Aspergillus niger extracellular citrate concentrations up to 1 mol l−1 are achieved. Intracellular citrate concentrations are in the millimolar range. Therefore the question arises whether citrate excretion depends on active transport. In this article thermodynamic calculations are presented for citrate excretion by A. niger at an extracellular pH of 3 and by Penicillium simplicissimum at an extracellula

ABSTRACT Oil spills in the Niger Delta region of Nigeria has occurred frequently since oil extraction started in the 1950's. The oil spills originates from facilities and pipelines, leaks from ageing and abandoned infrastructure and from spills during transport and artisanal refining of stolen oil under primitive conditions. It is estimated that spills in Nigeria amount to 100 000 to 200 000 tons per year and have been doing so for almost 60 years. While a number of reports have been written about the Niger Delta and the civil unrest in this area during the last decades, very few scientific re

In Aspergillus oryzae, maltose utilization requires a functional MAL locus, composed of three genes: MALR encoding a regulatory protein, MALT encoding maltose permease and MALS encoding maltase. Through a comparative genome and transcriptome analysis we show that the MAL regulon system is active in A. oryzae while it is not present in Aspergillus niger.

Abstract. A three-dimensional cloud-resolving model is used to investigate the vertical transport from the lower to the upper troposphere in a mesoscale convective system (MCS) that occurred over Niger on 15 August 2004. The redistribution of five passive tracers initially confined in horizontally homogeneous layers is analyzed. The monsoon layer tracer (0–1.5 km) is the most efficiently transported in the upper troposphere with concentrations 3 to 4 times higher than the other tracers in the anvil. On the contrary the African Easterly Jet tracer (~3 km) has the lowest contribution above 5 km.
