Abstract Lipocalin-2 (Lcn-2) is an innate immune protein whose expression is upregulated by several orders of magnitude in response to inflammation/injury/infection by variety of cells including neutrophils. However, the role of Lcn-2 induction in IC-induced inflammation is largely unknown. Here in, we have investigated the role of Lcn-2 during IC-mediated inflammation using Lcn-2 knock out (Lcn-2KO) and wild type (WT) mice. IC-mediated inflammation upregulated systemic Lcn-2 in WT mice when compared to naïve mice. The neutrophil chemoattractant KC was significantly elevated in WT, but not in Lcn-2KO mice, in response to IC-induced inflammation. Consistently Lcn-2KO mice showed about 50% reduced IC-mediated inflammation when compared to WT mice. Histopathological analysis of skin biopsy showed reduced inflammatory cell infiltration at inflamed site in Lcn-2KO mice relative to WT mice. Administration of recLcn-2 into Lcn-2KO mice resulted in inflammation similar to that of WT mice in response to IC. Lastly, we observed that neutralization of Lcn-2 via neutralizing mAb significantly reduced inflammation in WT mice. These results suggest that engagement of FcγRs expressed on local inflammatory cells with ICs leads to the induction of Lcn-2. Such IC-induced Lcn-2 expression may be essential for infiltration inflammatory cells at the site of inflammation. Our studies show that therapeutic targeting of Lcn-2 may be useful approach for treating IC-mediated inflammatory/autoimmune diseases

Dectin-2 is a C-type lectin receptor that recognizes high mannose polysaccharides.Cryptococcus neoformans, a yeast-form fungal pathogen, is rich in polysaccharides in its cell wall and capsule. In the present study, we analyzed the role of Dectin-2 in the host defense againstC. neoformansinfection. In Dectin-2 gene-disrupted (knockout) (Dectin-2KO) mice, the clearance of this fungus and the inflammatory response, as shown by histological analysis and accumulation of leukocytes in infected lungs, were comparable to those in wild-type (WT) mice. The production of type 2 helper T (Th2) cytokines in lungs was higher in Dectin-2KO mice than in WT mice after infection, whereas there was no difference in the levels of production of Th1, Th17, and proinflammatory cytokines between these mice. Mucin production was significantly increased in Dectin-2KO mice, and this increase was reversed by administration of anti-interleukin 4 (IL-4) monoclonal antibody (MAb). The levels of expression of β1-defensin, cathelicidin, surfactant protein A (Sp-A), and Sp-D in infected lungs were comparable between these mice. Inin vitroexperiments, IL-12p40 and tumor necrosis factor alpha (TNF-α) production and expression of CD86 and major histocompatibility complex (MHC) class II by bone marrow-derived dendritic cells and alveolar macrophages were completely abrogated in Dectin-2KO mice. Finally, the disrupted lysates ofC. neoformans, but not of whole yeast cells, activated Dectin-2-triggered signaling in an assay with nuclear factor of activated T cells (NFAT)-green fluorescent protein (GFP) reporter cells expressing this receptor. These results suggest that Dectin-2 may oppose the Th2 response and IL-4-dependent mucin production in the lungs after infection withC. neoformans, and it may not be required for the production of Th1, Th17, and proinflammatory cytokines or for clearance of this fungal pathogen.

Abstract Streptococcus pneumoniae possesses a thick polysaccharide capsule. Host defense to this bacterium is mediated by neutrophilic inflammatory responses, which is critically regulated by innate immune mechanism. S. pneumoniae has been reported to be sensed by pattern-recognition receptors such as TLR2, TLR4 and SIGN-R1. In this study, we examined the role of Dectin-2 in the host defense to S. pneumoniae infection. When bone marrow-derived dendritic cells from wild-type (WT) and Dectin-2KO mice were stimulated with culture supernatants (Sup) of a strain of S. pneumoniae (URF918) or its ConA-bound fraction, IL-12p40 production was abrogated in Dectin-2KO mice. In a NFAT-GFP reporter assay, Sup provided a positive signal in Dectin-2 gene-transfected reporter cells. These results suggest that Dectin-2 senses mannose-containing polysaccharides in S. pneumoniae capsule. Dectin-2KO mice infected intratracheally with S. pneumoniae showed shorter survival and larger bacterial burden and lower IFN-γ production in lungs than WT mice. Although the neutrophilic infiltration in lungs was equivalent in both mice, the ratio of engulfed/un-engulfed neutrophils was lower in KO mice than in WT mice. The levels of anti-pneumococcal polysaccharide Ab in bronchoalveolar lavage fluid and serum were lower in KO mice than WT mice. These results suggest that Dectin-2 plays critical roles in the host defense to S. pneumoniae infection through promoting the Ab-mediated phagocytosis by neutrophils.

Abstract Introduction Osteoarthritis of the first carpometacarpal joint is a common degenerative disease and surgical treatment includes resection suspension interposition arthroplasty (RSIA) with or without temporary transfixation of the first metacarpal. One major drawback includes proximalization of the first metacarpal during the postoperative course. Specific data comparing different transfixation techniques in this context is sparse. Materials and methods In this retrospective study, we measured the trapezial space ratio (TSR) in 53 hands before and after RSIA to determine the proximalization of the first metacarpal depending on the type of Kirschner (K)-wire transfixation. We, therefore, compared transfixation of the first metacarpal to the scaphoid with one K-wire (1K) to transfixation of the first metacarpal with two K-wires (2K), either to the carpus (2Ka), or to the second metacarpal (2Kb), or to both second metacarpal and carpus (2Kc). Results While preoperative TSR did not differ between group 1K and 2K (p = 0.507), postoperative TSR was significantly higher in group 2K compared to 1K (p = 0.003). Comparing subgroups, postoperative TSR was significantly higher in group 2Kc than 1K (p = 0.046), while we found no significant difference comparing either group 2Ka or 2Kb to 1K (p = 0.098; p = 0.159). Neither did we find a significant difference within 2K subgroups, comparing group 2Ka and 2Kb (p = 0.834), 2Ka and 2Kc (p = 0.615), or 2Kb and 2Kc (p = 0.555). Conclusions The results of our study suggest that transfixation with two K-wires should be preferred to transfixation with one K-wire after RSIA. Specifically, transfixation from first to second metacarpal and from first metacarpal to carpus resulted in least proximalization of the first metacarpal postoperatively.

Biodiesel is a clean-burning, oxygenated monoalkyl-ester fuel manufactured from natural, renewable sources like new/used vegetable oils and animal fats. The injection time has a significant impact on engine performance, particularly pollutant emissions. The purpose of this research is to see if Chlorellea Emersonii methyl ester (CEME) can be used as a fuel alternative in a compression ignition (CI) engine. The CEME was synthesised using a transesterification technique, and the engine parameters (performance, emission, and combustion) were investigated using 20 (v/v%) biodiesel blends at retard, standard, and advanced injection timings (IT). In this study an alteration in injection timing were done with TRC and BF combination. Other than conventional injection timing of 23o bTDC three more injection timing included ie 24o bTDC (advance IT), 21o bTDC (retarded IT) and 22o bTDC (retarded IT). Outcome results showed 22o bTDC(retarded IT) with 2.12% improvement in BTE followed by 24.5%,35.15% and 90.2% reduction in HC,CO and smoke.

Abstract Both of the sphingosine kinase (SK) subtypes SK-1 and SK-2 catalyze the production of the bioactive lipid molecule sphingosine 1-phosphate (S1P). However, the subtype-specific cellular functions are largely unknown. In this study, we investigated the cellular function of SK-2 in primary mouse renal mesangial cells (mMC) and embryonic fibroblasts (MEF) from wild-type C57BL/6 or SK-2 knockout (SK2ko) mice. We found that SK2ko cells displayed a significantly higher proliferative and migratory activity when compared to wild-type cells, with concomitant increased cellular activities of the classical extracellular signal regulated kinase (ERK) and PI3K/Akt cascades, and of the small G protein RhoA. Furthermore, we detected an upregulation of SK-1 protein and S1P3 receptor mRNA expression in SK-2ko cells. The MEK inhibitor U0126 and the S1P1/3 receptor antagonist VPC23019 blocked the increased migration of SK-2ko cells. Additionally, S1P3ko mesangial cells showed a reduced proliferative behavior and reduced migration rate upon S1P stimulation, suggesting a crucial involvement of the S1P3 receptor. In summary, our data demonstrate that SK-2 exerts suppressive effects on cell growth and migration in renal mesangial cells and fibroblasts, and that therapeutic targeting of SKs for treating proliferative diseases requires subtype-selective inhibitors.

Abstract Anti-pneumococcal polysaccharide vaccine (PPV) induces the production of serotype-specific Ab including IgG2 in spite of its feature of thymus-independent antigen. It remains to be clarified how PPV causes a class-switching from IgM to IgG. Recently, C-type lectin receptors (CLR) sensing polysaccharides have garnered much attention in the host defense to fungal infection. In our recent study, Dectin-2, a CLR involved in the recognition of mannose-rich polysaccharides, has been found to play an important role in the host defense to pneumococcal infection. Therefore, in this study, we addressed how Dectin-2 was involved in the vaccine effect of PPV using a mouse model. The serum levels of anti-serotype 3 pneumococcal capsular polysaccharide (PPS-3) two weeks after vaccination were significantly reduced in Dectin-2 knockout (KO) mice compared to wild-type (WT) mice. Weight loss and hypothermia of mice caused by infection with Streptococcus pneumoniae (URF918) were improved in WT mice transferred with spleen cells from vaccinated WT mice compared to those from vaccinated Dectin-2KO mice. Similar tendency was observed in the survival of infected mice. In in vitro experiments, IL-12p40, TNF-α and IL-6 production by bone marrow-derived dendritic cells (BM-DCs) stimulated with PPV was abrogated in Dectin-2KO. These results suggest that Dectin-2 is involved in the PPV-induced serotype-specific Ab production and host protection through sensing the pneumococcal polysaccharide antigens.

Abstract In DP thymocytes, the Tcra locus undergoes multiple rounds of V-J rearrangement in order to generate a functional TCRα chain. While Traj segment usage is strictly ordered from 5′ to 3′ as rearrangement proceeds, it is unclear to what degree Trav usage is also ordered. To test this, we assessed the Trav-Traj combinatorial repertoire in murine DP thymocytes using 5′ RACE followed by paired end sequencing. In 129Sv/J WT thymocytes, we found a clear bias toward proximal-proximal and distal-distal joins, but Trav-Traj pairings were highly diverse, even in primary V-J rearrangement. However, combinatorial diversity was highly constrained in HYα KI thymocytes, in which a fixed primary V-J rearrangement in the endogenous Tcra locus is the starting point for secondary rearrangements. Thus, secondary rearrangement is highly processive through the V and J arrays, but surprising diversity is introduced by primary rearrangement. Tcrd rearrangement precedes Tcra rearrangement in developing thymocytes. In INT1-2KO mice, CTCF site deletion biases Trdv usage toward the most proximal gene segments. These mice, like HYα KI, displayed a substantially constrained Tcra repertoire, with limited diversity of primary rearrangements. Thus, variable truncation of the Trav array by prior Tcrd rearrangement creates diversity in an otherwise highly processive Tcra rearrangement program by imparting combinatorial diversity to primary Tcra rearrangement. This diversification is functionally important, as Trav1-Traj33 pairing and MAIT cell abundance are substantially reduced in HYα and INT1-2KO mice.

A simplified, physically intuitive model of diffuse scattering of radio waves from a rough surface is used to present a self-contained derivation of first- and second-order cross sections, essentially in agreement with the standard expressions. Incoherent addition of the second-order contributions (electromagnetic and hydrodynamic) leads to a cross section which is slightly different from the more rigorously derived cross sections of Barrick (1972b) and Johnstone (1975). A surface current v has been incorporated in this model, with the main change to the cross section being a frequency shift of the entire spectrum by an amount Llw = -2ko . v.

Abstract Naïve T cells use peripheral node addressin (PNAd), normally found on lymph node (LN) high endothelial venules (HEV), to traffic into lymphoid organs. We previously reported that the vasculature of murine tumors expressed PNAd and CCL21, which supports infiltration and intratumoral activation of naïve T cells, in turn delaying tumor growth. PNAd expression on tumor vasculature depends on effector CD8+ T cells that secrete LTα3, which signals through TNFR on tumor-associated CD31+ endothelial cells. PNAd expression is low on tumor vasculature, suggesting that it might be regulated differently than PNAd on HEV. Using RT-qPCR, we found that most of the enzymes responsible for PNAd synthesis are expressed comparably in CD31+ cells from tumors and LN. However, the primary sulfotransferase responsible for the generation of the 6-sulfo sialyl Lewis X structure on PNAd HEV, GlcNAc6ST-2, is minimally expressed in tumors. Tumor CD31+ cells expressed equivalent levels of GlcNAc6ST-1, suggesting that it is responsible for low-level PNAd expression. Of the scaffolding proteins that make up PNAd, GlyCAM-1 was minimally expressed in tumor CD31+ cells, whereas all others were expressed comparably. To identify the components of PNAd biosynthesis in tumor CD31+ cells that are controlled by LTα3-TNFR signaling, we evaluated tumors from WT and TNFR1/2KO mice. GlyCAM-1 was the only assessed PNAd component that was ablated in TNFR1/2KO tumors. However, based on transcript levels, GlyCAM-1 accounts for only about 10% of the PNAd scaffolding proteins in tumor CD31+ cells. This suggests that TNFR signaling regulates additional PNAd biosynthetic components yet to be examined, or that it regulates one or more components at the level of protein translation.

An Einstein model for thermal diffuse scattering (TDS) has enabled quantitative calculation of the absorptive potential V'(r). This allows anomalous absorption to be accounted for in LACBED contrast. Fourier coefficients Vg-h of the absorptive component from each atom α are calculated from integrals of the formwhere fα is the scattering amplitude and M(Q) the Debye-Waller factor. Integration over the Ewald sphere (dΩ) requires the momentum transfer q to have values up to 2ko (the incident beam momentum). Dynamical ‘dechannelling’ is accounted for by the terms g ≠ h. The crystal absorptive potential is obtained by coherently summing over these atomic absorptive potentials within the unit cell. Unlike the elastic potential, the absorptive potential is a strong function of incident beam energy Eo, since the range of momentum transfer q and associated solid angles dΩ change with the Ewald sphere radius.Fig. 1 shows a LACBED pattern of the zeroth order beam from Si aligned along a <001> zone axis.

Abstract Engineered human induced pluripotent stem cells (hiPSC)-derived allogeneic Natural Killer (NK) cells are emerging as promising safe and effective off-the-shelf cell therapy drugs. Repeated dosing of NK cells can partly compensate the relatively shorter persistence of NK cells compared with chimeric antigen receptor T cells (CAR-T); However, with each repeat, the persistence and efficacy are increasingly challenged by activation of host innate and adaptive immunity. Thus, reducing immunogenicity of allogeneic NK cells holds promise for better efficacy. In this study, we report a combination of “hypoimmunity” gene edits including multiple transgene knock-ins and both class I and class II MHC knockouts (2KO). We show that hiPSC engineered by the hypoimmunity combo can be efficiently differentiated into NK cells, comparable to the non-engineered ones. This is contrary to the notion from mouse studies that NK cells devoid of class I molecules are hypo-responsive, these engineered NK cells can be expanded by aAPC (artifitial Antigen Presenting Cells, a K562 cell line with surface expression of mbIL-21 and CD137L) and are capable of killing tumor cells, Additionally, we show that the engineered NK cells can evade T cell immunity, as examined by in vitro T cell proliferation and cytotoxicity assays. A humanized mouse model with transplanted CD34-positive human hematopoietic stem cells confirmed that the engineered NK cells can persist longer than WT and 2KO NK cells. Hypoimmune-QN-019 NK cells are generated by incorporating hypoimmunity gene combo edits into our hiPSC-derived NK cell clinical candidate QN-019 which has 3 transgenes including membrane-bound IL-15, high affinity non-cleavable CD16 and CAR targeting CD19. Comparing to QN-019, hypoimmune-QN-019 displayed better efficacy in humanized CDX mouse model in clearing CD19-positive cancer cells. Taken together, we have engineered hiPSC-derived allogeneic NK cells with hypoimmunity features, which hold great promise in becoming an off-the-shelf drug with improved in vivo persistence and efficacy for repeated dosing. Penelitian ini dilakukan untuk mengetahui pengaruh sudut kemiringan (α) terhadap perpindahan panas pada penukar panas pipa konsentris dengan pengaturan forward insert strip louvered. Dalam penelitian ini, insert strip louvered divariasikan dengan α = 15o, 20o dan 25o. Cairan kerja di ban dalam adalah air panas dan anulus adalah air dingin, dengan arah aliran berlawanan. Pengujian dilakukan pada bilangan Reynolds (Re) 5300 - 17.500). Pada 5300 <Re <17500 nilai Nui pada pipa bagian dalam dengan penambahan insert strip louvered dengan kenaikan α = 15o, 20o dan 25o pada kisaran 19,04% - 22,86%, 48,09% - 54,83%, dan 67,43% - 77,02% dibandingkan dengan Nui tabung polos masing-masing. Pada Re 5300 <Re <17500 Nilai faktor gesekan pada pipa bagian dalam dengan penambahan insert strip louvered dengan α = 15o, 20o dan 25o adalah 1,52, 2,56 dan 3,35 kali lebih besar dari pada faktor gesekan tabung polos. Rasio penambahan perpindahan panas dengan penambahan insert strip louvered dengan α = 15o, 20o dan 25o masing-masing pada kisaran 1.00 - 1.06, 1.02 - 1.08 dan 1.03 - 1.12.

<p>Darb El-Arbeain area lies between long. 29o 00/ and 31o 00/ E and lat. 22o 00/ and 24o 30/ N. It is divided into three separate areas; The northern part extends 90 km to the south from Paris town and has an area of 90 km2. In this study four suggested scenarios of pumping rates have been explored to fit with the Egyptian ministry of irrigation using the three dimensional finite difference flow model (MODFLOW) to simulate the flow system. These scenarios include: first, model will run with abstraction from the aquifers equal 110 %, 180%, 280%, and 370% of calculated initial recharge. Results indicate that the second scenario has the most economic scenario on the area. The fourth scenario caused the highest increase of drawdown values which should be avoided<em>.</em></p>

An equation of motion phonon method, developed for even-even nuclear systems and extended to odd nuclei, is applied to 22O and to its odd neighbors 23O and 23F. A calculation using the chiral potential NNLOopt is carried out in a space encompassing up to two phonons. The computed dipole cross section in 22O and the spectra of 22O and 23O are in a satisfactory agreement with the experimental data. However, the calculation describes poorly the spectrum of 23F. This discrepancy originates from the strong coupling between the odd proton and the 22O phonons of neutron nature. This coupling pushes down in energy several states enhancing the level density at low energy. We suggest that a viable route for the solution of this problem could be the inclusion of the three-body interaction using the new chiral potential NNLOsat.

Rates have been calculated for electric dipole transitions in Ti II between the fine structure levels of the terms 4p z 4Go, 4 Fo, 2Fo, 2Do, 4Do, and 2Go to the levels of the groundterm 4s a 4F and comparisons are made with the most recent available observations as well as with the calculations of Kurucz. These are the only other calculations available for this ion. To include certain configurations with more than two open shells, new approximations are necessitated by the complexity of the algebra. The approximations used are described and the results of calculations are given to demonstrate their effectiveness and accuracy. It will be shown that the CPU time reductions are very substantial, sufficient to make the inclusion of potentially important configurations feasible where it would not be so otherwise.The loss in accuracy in the situations examined is negligible. Abstract Complement receptors (Cr)1 and 2 have been established to have important roles in control of complement activation in the immune microenvironment and B cell responses. Previous work investigating the effects of these receptors in vivo has relied on a double knockout mouse line in which each of these receptors was deleted. However, two drawbacks of this mouse must be acknowledged: 1) that both receptors are deleted and roles of each must be inferred, and 2) that the essential B cell signaling protein CD19 is expressed at a significantly higher level than WT cells. We have created a Cr1 deficient line of mice (expressing Cr2) by homologous gene targeting that expresses normal levels of Cr2 gene products on B cells. This animal has normal ratios of bone marrow, B2 cell, and B1 B cell populations. Cr2 negative marrow B cells express the same amount of surface CD19 as WT but mature Cr2-expressing B cells express less. These Cr2-only mice generate normal to slightly enhanced antibody titers to T-dependent and T-independent (both 1 and 2 antigens.) In conclusion Cr1 is not critical for development and maintenance of B cell populations nor for the generation of antibodies to naïve antigens. This mouse, along with the Cr1/2KO, will be a critical tool in delineating Cr1 specific immune response effects.

Abstract Transfusion-related acute lung injury (TRALI) remains a leading cause of transfusion-related deaths. In most cases, anti-leukocyte antibodies in the transfusion product trigger TRALI, but not all anti-leukocyte antibodies cause TRALI. It has been shown that the anti–major histocompatibility complex (MHC) class I antibody 34-1-2S (anti–H-2Kd) causes TRALI in BALB/c mice (MHC class I haplotype H-2Kd), whereas SF1.1.10 (anti–H-2Kd) does not. In C57BL/6 mice (MHC class I haplotype H-2Kb), TRALI only occurs when anti-MHC class I antibody AF6-88.5.5.3 (anti–H-2Kb) is administered together with a high dose of 34-1-2S. It remains unknown which specific antibody characteristics are responsible for eliciting TRALI. We therefore investigated several biological and structural features of 34-1-2S compared with other anti-MHC class I antibodies, which on their own do not cause TRALI: SF1.1.10 and AF6-88.5.5.3. No substantial differences were observed between the TRALI-causing 34-1-2S and the TRALI-resistant SF1.1.10 regarding binding affinity to H-2Kd. Regarding binding affinity to H-2Kb, only AF6-88.5.5.3 potently bound to H-2Kb, whereas 34-1-2S exhibited weak but significant cross-reactivity. Furthermore, the binding affinity to FcγRs as well as the Fc glycan composition seemed to be similar for all antibodies. Similar Fc glycosylation profiles were also observed for human TRALI-causing donor anti-HLA antibodies compared with human anti-HLA antibodies from control donors. 34-1-2S, however, displayed superior complement activation capacity, which was fully Fc dependent and not significantly dependent on Fc glycosylation. We conclude that TRALI induction is not correlated with Fab- and Fc-binding affinities for antigen and FcγRs, respectively, nor with the composition of Fc glycans; but increased Fc-mediated complement activation is correlated with TRALI induction.

Abstract Mice deficient in T regulatory cells (T regs) die from systemic inflammation. Breeding these mice with IL-2KO mice clears inflammation in lungs and skin, suggesting that IL-2 has pro-inflammatory functions beyond promoting survival of T regs. We recently identified a dimeric form of IL2 mammalian tissues that is cytotoxic to cells expressing IL-2 receptor (IL-2R), which provides an explanation for the above. Exposure of IL-2R+ cells to dimeric IL-2 results in rapid release of LDH. We hypothesized that dimer-mediated cell death is consistent with the characteristics of necrosis. To test this hypothesis, we exposed Jurkat cells to dimeric IL-2 and processed them for electron microscopy at 5 minutes. EM revealed swollen nuclear membranes and mitochondria consistent with early necrosis. Exposure of vascular smooth muscle cells (VSMC) to dimeric IL-2 induced 50% loss of cellular ATP at 5 minutes and nearly complete loss at 20 minutes. Using the mitochondrial membrane potential indicator JC-1 in VSMC, nearly all mitochondria lost their MMP by 10 minutes. These data indicate that dimeric IL-2 kills cells by necrosis. Our finding that dimeric IL-2 is cytotoxic to cells expressing IL-2R, lymphoid or non-lymphoid, has implications beyond the immune system. These studies combined with prior studies showing presence of matrix-bound IL-2 in arteries, suggests dimeric IL-2 may contribute to vascular inflammation in processes such as atherosclerosis.

Abstract Regulatory T cell plays an important role in controlling immunity to self and foreign antigens. CD4+CD25+ require T cell receptor and IL-2 receptor activation for in vivo / in vitro expansion and function, but the source of IL-2 remains unknown. In identifying the important paracrine source of IL-2 for Treg cells, we concentrated on the dendritic cell as the paracrine source, because in vitro imaging studies demonstrated fine localization of CD25 at the membrane interface between RD6 and dendritic cells. We used ELISPOT assay to detect IL-2 production by the freshly isolated splenic and bone marrow dendritic cells Dendritic cells secrete a small fraction of IL-2 early in culture, CpG-B enhances the IL-2 production of DCs. Coculture with regulatory cells reduces the level of detectable IL-2.We examined the direct effect of dendritic cell IL-2 on Treg function, CD4+CD25+ cells suppressed CD4+CD25- effectors cells when stimulated with WT dendritic cells, but not when stimulated with IL-2KO DCs. Anti-CD25 abrogates the function of both Treg and RD6 co-cultured with WT DCs, suggesting that CD25 is required for delivery of IL-2 into the Tregs for processing and the addition of high doses of exogenous IL-2 can restore the inhibitory function of Treg and RD6 cells co-cultured with IL-2 -/- dendritic cells. These data suggest that DC IL-2 is essential for Treg function.

Abstract The two-pore channel (TPC) family is widely conserved in eukaryotes. Many vascular plants, including Arabidopsis and rice, possess a single TPC gene which functions as a slow vacuolar (SV) channel—voltage-dependent cation-permeable channel located in the vacuolar membrane (tonoplast). On the other hand, a liverwort Marchantia polymorpha genome encodes three TPC homologs: MpTPC1 is similar to TPCs in vascular plants (type 1 TPC), while MpTPC2 and MpTPC3 are classified into a distinctive group (type 2 TPC). Phylogenetic analysis suggested that the type 2 TPC emerged before the land colonization in plant evolution and was lost in vascular plants and hornworts. All of the three MpTPCs were shown to be localized at the tonoplast. We generated knockout mutants of tpc1, tpc2, tpc3 and tpc2 tpc3 double mutant by clustered regularly interspaced short palindromic repeats/Cas9 genome editing and performed patch-clamp analyses of isolated vacuoles. The SV channel activity was abolished in the Mptpc1 loss-of-function mutant (Mptpc1-1KO), while Mptpc2-1KO, Mptpc3-1KO and Mptpc2-2/tpc3-2KO double mutant exhibited similar activity to the wild type, indicating that MpTPC1 (type 1) is solely responsible for the SV channel activity. Activators of mammalian TPCs, phosphatidylinositol-3,5-bisphosphate and nicotinic acid adenine dinucleotide phosphate, did not affect the ion channel activity of any MpTPCs. These results indicate that the type 1 TPCs, which are well conserved in all land plant species, encode the SV channel, while the type 2 TPCs likely encode other tonoplast cation channel(s) distinct from the SV channel and animal TPCs.

Com a finalidade de estudar as glândulas paratireóides durante a fase de redução ponderal, 48 ratos Wistar, machos, foram distribuídos em dois grupos. Os animais do Grupo1, no total de 28, foram submetidos à derivação jejuno-cólica. Os animais do Grupo2, no total de 20, foram submetidos à operação simulada. Estudaram-se, à microscopia óptica, os aspectos histológicos, e à microscopia eletrônica, os aspectos ultraestruturais. A função das glândulas paratireóides foi estudada mediante determinações séricas do Hormônio da Paratireóide - Parato-hormônio (PTH), Cálcio, Fosfato, Albumina e Proteínas Totais. Procedeu-se à avaliação da evolução ponderal e variação percentual, pesando-se os animais nos dias 7, 14, 21 e 28. A determinação sérica do PTH, Cálcio, Fosfato, Albumina e Proteínas Totais foi realizada antes dos sacrifícios e os estudados da microscopia, após os mesmos, nos 7o, 14o, 21o e 28o dias. Os valores do peso, da variação percentual e das dosagens bioquímicas e hormonais foram submetidos à análise estatística separadamente para cada grupo. Compararam-se também os Grupos 1 e 2 em relação aos valores das variáveis estudadas nos 7o, 14o, 21o e 28o dias do sacrifício. Os animais submetidos à derivação jejuno-cólica apresentaram progressiva perda ponderal demonstrada a partir do sétimo dia e os animais de controle, crescente ganho de peso a partir do 14o dia. A média dos valores das determinações séricas do PTH dos ratos submetidos à derivação jejuno-cólica foi maior do que dos animais-controle já a partir do 7o dia, embora estatisticamente significantes. Os valores séricos do fosfato só foram estatisticamente significantes no 21o dia, o da albumina no 14o, 21o e 28o dias e os das proteínas totais em todos os períodos. A média dos valores do cálcio foi menor nos animais-controle dos que nos submetidos à derivação jejuno-cólica, não sendo esse valores estatisticamente significantes. O estudo da microscopia eletrônica e óptica nos animais submetidos à derivação jejuno-cólica diferenciou dos encontrados nos animais-controle pela predominância das células principais, sendo que as células escuras predominam no 21o dia e as células claras no 28º dia.

The design and mathematical model of a reflective type optical gas sensor is presented. Light source is radiated at an incident angle for 10 cm gas cell with an internal diameter of 0.4 cm. At an incident angle of 1o, optical path length obtained is 342.7886 cm, at 27o incident angle, optical path length is 10.4926 cm and at an incident angle of 28o, optical path length is 9.9631 cm. The model is most efficient at lower incident angles, precisely between (1o and 27o). Effects of variation in diameter and length of gas cell are also demonstrated.

Oil and oil products adversely affect both the biodiversity of the microorganisms and the soil function. In oil-contaminated soils, unique bacterial communities develop that are adapted to pollution. In this work, the bacterial structure and diversity of the microbial community have been studied in samples of oil-contaminated soils in Kazakhstan deposits using the Illumina MiSeq sequencer. The results of the study showed that the representatives of the following bacterial phyla dominated in the selected soil samples: Proteobacteria, prevailing in oil-contaminated soils (up to 48%), Actinobacteria (up to 29.33%), Firmicutes (up to 25.74%), Bacteroidetes (up to 33.28 %). The representatives of Planctomycetes, Verrucomicrobia, Chloroflexi (0.76%-4.62%) phyla were found in smaller amounts. All the uncontaminated soils were dominated by Micrococcaceae, Flexibacteraceae, Sphingomonadaceae, Planococcaceae, Flavobacteriaceae families, contaminated ones – by Halomonadaceae, Flavobacteriaceae, Alteromonadaceae, Dietziaceae, Pseudomonadaceae, Bacillaceae, Xanthomonadaceae, Anaerolinaceae, Mycobacteriaceae and Peptococcaceae families. At the genus level, samples of uncontaminated and contaminated soils also demonstrated significant diversity. The dominant bacterial genera in the samples of the uncontaminated soil were Hymenobacter, Arthrobacter, Gillisia. In contaminated soils of three deposits the microorganisms of the Halomonas, Marinobacter, Pseudomonas (mostly in 2KO soil sample), Bellilinea and Mycobacterium (mostly Md sample) genera were spread more widely; and a very large population of the microorganisms of the Halomonas genus was found in the contaminated soil sample from the Atyrau region. A comparison of the taxonomic structure of microbial communities of oil-contaminated soils indicates that the composition of the microbial population changes depending on the degree of oil pollution. Samples of uncontaminated background soils were characterized by higher bacterial diversity than samples of contaminated soils. The microorganisms belonging to the dominant phyla were mostly associated with the decomposition of oil hydrocarbons. The characterization of the bacterial communities living in the contaminated soils and the assessment of their ability to decompose oil can potentially be a guide for bioremediation of contaminated soils.

Dengan mengambil variabel sudut serang sebanyak 9 variabel yaitu 0o, 3o, 6o, 9o, 12o, 15o, 18o, 21o, dan 24o. Dari hasil Simulasi didapat Sudut Serang terbaik untuk tipe NACA 0013 adalah 21o dengan harga Gaya Lift sebesar 61,650 N.

Abstract Objective: To clarify Natural killer (NK) cell-mediated resistance under cytoreductive conditioning and T-cell depleted bone marrow transplantation (TCD-BMT), we investigated the effects of host NK cell depletion on engraftment and induction of stable mixed chimerism. Methods: BALB/c mice (H-2kd) were injected intraperitoneally with anti-asialoGM1 antibody (anti-NK Ab) on day -1. On day 0, they received TBI at a dose of 500 cGy, followed by intravenous infusion of 2 x 107 T-cell-depleted (TCD) bone marrow (BM) cells from C57BL/6 mice (H-2kb). Early engraftment and chimerism were determined by the relative ratio of peripheral blood (PB) lymphocytes expressing either H-2kd or H-2kb on day +21. Long-term engraftment and chimerism were evaluated on PB and spleen by multi-color flow cytometry. Results: Although no recipients treated with TBI alone showed engraftment, all the recipients conditioned with anti-NK Ab and TBI showed successful engraftment as well as a donor-dominant pattern of mixed chimerism in both PB and spleen. Spleen cells from recipients with mixed chimerism showed specific tolerance to both host and donor strains, but not to a third-party (C3H/He). None of the reconstituted mice showed signs of graft versus host disease, and all survived up to day +330. Conclusion: These observations indicate that host NK cell depletion may be used to reduce the intensity of conditioning regimens for engraftment of TCD grafts, and can contribute to establishment of stable mixed chimerism in MHC-mismatched NMT.

The cool room temperature is a requirement in the thermal comfort of a building. In earthquake prone areas, building structures should be designed with minimal material weight. Therefore, it is necessary to be aware of an alternative solution to ensure the room's thermal comfort with minimal weight. This research aims to analyze the influence of roof slope 15O, 20O, 25O, 30O, 35O, 40O, and 45O on the comfort of the room and the strength of the building in the house measuring 6 meters x 8 meters x 3 meters. Analysis is done with three software that is SketchUp as a design, CONTAM to analyze air pressure and TRNSYS to simulate all the data that has been created in SketchUp and CONTAM. Obtained the lowest temperature at a roof slope of 45O and the lowest weight is the roof with a 15O slope. Temperatur ruangan yang sejuk merupakan syarat dalam kenyamanan termal sebuah bangunan. Di kawasan rawan gempa, struktur bangunan harus dirancang dengan bobot material yang minimal. Oleh karena itu, perlu dicarikan solusi alternatif untuk menjamin kenyaman termal ruangan dengan bobot yang minimal. Penelitian ini bertujuan menganalisa pengaruh kemiringan atap 15o, 20o, 25o, 30o, 35o, 40o, dan 45o terhadap kenyamanan ruangan serta kekuatan bangunan pada rumah berukuran 6 meter x 8 meter x 3 meter. Analisa dilakukan dengan tiga software yaitu SketchUp sebagai perancangan, CONTAM untuk menganalisa tekanan udara serta TRNSYS untuk menyimulasikan semua data yang telah di buat pada SketchUp dan CONTAM. Didapatkan hasil suhu terendah berada pada kemiringan atap 45 o dan bobot terendah adalah atap dengan kemiringan 15o.

Well-constructed foundation must be able to resist forces that given by the structure. Foundation bearing capacity affected by some parameters and can be measured by various methods. Some methods that used in this study is Terzaghi, Meyerhof and finite element. The purpose of this study was to compare the bearing capacity value generated by these methods and the effect of parameters value variety against foundation bearing capacity. The parameters is foundation footing, friction angle and foundation depths. The highest value of foundation bearing capacity is generated by the finite element method. Bearing capacity that generated by finite element method experiences the biggest difference up to 41% compared to Terzaghi method and up to 26% biggest difference compared to Meyerhof method. Bearing capacity with friction angle 25o is 70% greater compared to 20o. Meanwhile with 30o increased to 220% compared to 20o. The value of bearing capacity differs according to the depths used. Within 1,5m depth the value of the bearing capacity is increased 35% compared to 1m depths with square footing, Meanwhile with rectangle footing only increased by 32%. The bearing capacity value also varies depends on the footing type. Rectangle footing was increased 9 – 14% compared to square footing. Metode yang digunakan penulis adalah Terzaghi, Meyerhof dan elemen hingga. Dalam penulisan ini penulis ingin membandingkan daya dukung yang dihasilkan oleh metode-metode tersebut dan pengaruh dari variasi nilai parameter terhadap daya dukung fondasi. Parameter tersebut adalah jenis tapak, sudut geser dan kedalaman fondasi. Daya dukung terbesar dihasilkan oleh metode elemen hingga. Daya dukung yang dihasilkan oleh metode elemen hingga mengalami perbedaan terbesar hingga 41% jika dibandingkan dengan metode Terzaghi dan 26% perbedaan terbesar jika dibandingkan metode Meyerhof. Daya dukung dengan sudut geser 25o lebih besar 70% jika dibandingkan dengan sudut geser 20o. Sedangkan pada sudut geser 30o daya dukung mengalami kenaikan sebesar 220% jika dibandingkan dengan sudut geser 20o. Daya dukung juga berbeda tiap kedalaman fondasi. Pada kedalaman fondasi 1,5m daya dukung mengalami kenaikan 35% jika dibandingkan pada kedalaman fondasi 1m pada fondasi bujur sangkar dan pada fondasi persegi panjang mengalami kenaikan 32%. Daya dukung juga berbeda tergantung jenis tapaknya. Pada jenis tapak fondasi persegi panjang, kapasitas daya dukung mengalami kenaikan 9 - 14% jika dibandingkan dengan jenis tapak fondasi bujur sangkar.

The anomalous seismic activity prior to three mainshocks occurred in Arakan-yoma region, within the area bounded by 20o-28o N and 92o-98o E, has been studied considering spatial distribution and temporal patterns of earthquakes. The anomalous seismic activity has been investigated in this region using the seismicity data for the period 1963 to 2008 with events having magnitude mb ≥ 4.3. On a commercial eel farm, which uses a recirculation system, 400,000 glass eels were farmed for a period of 328 days at 20o – 23o C. The physicochemical parameters of the farm water were kept at normal conditions during the experiment. The NO 2 was kept between 1.0 and 3.0 mg/l. By the end of the experiment, 4,582 kg of fish feed were consumed and 2,939 kg of eels were produced (177,523 eels with mean final individual body weight of 16.6g and mean food conversion ratio of 1.625). The glass eels showed a high variability in their capacity to grow.

Weakly nonlinear stage of the ablative Rayleigh-Taylor instability has been studied by the perturbation theory. Mode coupling of linear growing waves with wave numbers kA and kB drives new excited waves with wave numbers k0 (= kA ± kB, 2kA, 2kB). We have investigated time evolution of the excited waves and found that the ablation effect plays an important role even in the nonlinear stage to reduce amplitude of the excited waves. Differences between an ablation surface and a classical contact surface have been discussed. Dependence of the excited wave amplitude on the wavenumber k0, the ablation velocity va, and the effective gravity g is also investigated.

Abstract Introduction: We have previously shown that in utero transplanted ES cells survive and integrate into the fetus development and have established a murine model for the study of the in vivo differentiation of ES cells. The goal of this study was to monitor the fetal immune responses post in utero transplantation of ES cells, bone marrow (BM) and fetal liver (FL) hematopoietic stem cells. Materials and Methods: Murine (MHC)-mismatched ES cells genetically engineered to express yellow fluorescent protein (YFP-ES cells) were cultured on mitomycin-treated feeder layers for four days prior to in utero transplantation (IUT) in medium containing leukemia inhibitory factor. A cell dose of 5x104 YFP-ES cells ((H-2kb) was injected intraperitoneally in the fetuses of Balb/c (H-2Kd) pregnant mice at E12- E14. BM and FL hematopoietic stem cells (H-2kb) were transplanted in utero at a dosage of 1×109 cells/kg fetal body weight into E12-E14 BALB/c fetuses (H- 2Kd). Fetal immune responses were monitored by in vitro mixed lymphocyte reaction and cytotoxicity assays performed with self (Balb/c), allogeneic ES cells, BM or FL hematopoietic cells, and 3rd party (C3H cells). Cytokine levels (IL-2, IFN-gamma, IL-4 and IL-10) were determined in the cell culture supernatants from cytotoxicity assays. Liver tissue sections were prepared from in utero transplanted fetuses and examined for the presence of lymphocytic infiltration. Results: In utero transplantation of YFP-ES-cells did not induce tolerance in the fetuses and was associated with increased cytokine production compared with BM and FL groups. Microscopic examination of liver sections of ES cell transplanted group revealed the presence of marked inflammatory infiltrate. Conclusions: Embryonic stem cells transplanted in utero induce fetal immune responses and increased cytokine production associated with (MHC) upregulation.

Abstract Introduction: Graft-versus-host disease (GvHD) is a severe complication arising in patients undergoing allogeneic hematopoietic stem cell transplantation (allo-HSCT). Potent and selective modulation of JAK1/STAT-mediated signaling is an attractive therapeutic strategy for the management of acute GvHD and is currently being evaluated in clinical trials (GRAVITAS-301: NCT03139604; GRAVITAS-119: NCT03320642). Methods: Acute GvHD was induced in BALB/c mice using the established MHC-mismatched mouse model. BALB/c (H-2Kd) recipients were given an intravenous injection of a combination of splenocytes and T cell depleted bone marrow cells from allogeneic cell transfer from donor C57BL/6 (H-2Kb) mice. Animals were dosed orally with vehicle or the selective JAK1 inhibitor, itacitinib (60 mg/kg or 120 mg/kg twice daily). Engraftment was analyzed for the proportion of donor and host leukocytes (CD45+, H-2Kb, and H-2Kd). GvHD clinical scores were assessed by standard methods and inflammatory cytokine profiles in blood and colon quantified by multiplex analysis. Colon samples were sectioned and stained with the following immunohistochemical (IHC) markers: CD4, CD8, phosphoSTAT3 and CD3+phosphoSTAT3 (dual staining) for pharmacodynamic assessment of JAK/STAT pathway activity in colon and infiltrating T-cells. Effects of itacitinib on preservation of Graft-versus-Leukemia (GVL) were evaluated by injecting BALB/c mice with A20 lymphoma cells that are of H-2Kd phenotype along with combination of splenocytes and T cell depleted bone marrow from C57BL/6 (H-2Kb) mice. Results: Itacitinib administration was highly effective in both prophylactic (from day −3) and therapeutic (from day 14) dosing regimens in ameliorating body weight loss and improving GvHD scores. Itacitinib did not significantly impact donor engraftment as determined by CD45+/H-2Kb quantification by flow cytometry. Similar efficacy was observed with 60 mg/kg versus 120 mg/kg twice daily dosing regimens. Oral itacitinib administration achieved JAK1 IC50 coverage for 4 h and 12 h at 60 mg/kg twice daily and 120 mg/kg twice daily, respectively. Associated with GvHD progression, maximal upregulation of inflammatory cytokines were observed in peripheral blood on day 17 (IFN-γ, TNF-α, IL-6, IL-13) and in colon on day 28 (IFN-γ, TNF-α, IL-1β). Itacitinib (120 mg/kg twice daily) treatment significantly reduced the inflammatory cytokine milieu at these disease stages. No differences were observed in absolute number of CD4+ T cells and CD8+ T cells in blood and spleen with itacitinib treatment, but significant reductions were detected in CD4+ T cells and CD8+ T cells in the inflamed colon tissue along with significant JAK1/STAT3 inhibition as measured by reductions in normalized pSTAT3 in T cells and colonic epithelial cells. Itacitinib treatment did not negatively impact GVL responses, as evidence by T cell mediated reduction of tumor burden. Furthermore, itacitinib treatment enhanced the survival of the recipient BALB/c mice in comparison to the vehicle treated animals. Conclusions: Itacitinib, a selective JAK1 inhibitor ameliorated GvHD severity when administered prophylactically or therapeutically and had no detrimental effects on engraftment and preservation of GVL. Furthermore, itacitinib inhibited JAK1/STAT3 activation in diseased colon tissue and infiltrating T-cells, and reduced disease burden and improved survival by modulating levels of inflammatory cytokines important in the pathophysiology of acute GvHD. Clinical benefits obtained from checkpoint blockade regimens demonstrate the importance of overcoming the immunosuppressive tumour microenvironment (TME) in cancer immunotherapy. Intravenous (i.v.) injection of B16 melanoma cells (H-2Kb) leads to lethal disseminated pulmonary metastasis in Balb/c recipients (H-2Kd). This lack of immune control is related to low major histocompatibility complex (MHC) expression on B16 cells which is associated with delayed and decreased anti-tumour adaptive immune responses (e.g., alloantibody formation) as: (i) other tumour types with normal H-2Kb expression are rejected with concomitant antibody production; (ii) preincubation of B16 with IFN-gamma to upregulate H-2Kb expression resulted in improved antibody production and anti-tumour activity. The delayed/decreased anti-tumour adaptive immune responses induced by B16 inoculation is not able to interrupt progression of primary metastases, while it is able to effectively eliminate secondary inoculated subcutaneously (s.c.) B16 cells from progression. This is due to the presence of an immunosuppressive TME within the primary metastases characterized by increased regulatory T cells (Tregs) and an increased T helper cells (Th) 2/1 profile. These tumour-induced immunosuppressive T cell populations are counteracted by improved adaptive immunity via active and passive immunization, resulting in effective elimination of the TME, destruction of the metastatic tumour and a reversal of Th2/1 profile in a time-sensitive manner. Thus, we here demonstrate that the TME is not irreversible and adaptive immunity is able to eradicate established solid tumour and its immunosuppressive TME. This study will help design treatments to overcome the immunosuppressive effect of the TME and improve efficacy of cancer immunotherapy.

Abstract We generated nucleotide sequences for H-2Kk and H-2Dk from the C3H mouse, as well as for a genomic clone of H-2Db, in order to conduct an evolutionary analysis of the H-2 genes from three haplotypes, k, d, and b. H-2Kk from both the C3H and AKR strains, H-2Kd, H-2Kb, H-2Dk, H-2Ld, H-2Dd, H-2Db, and H-2Dp DNA sequences were aligned, and the alignments used to construct phylogenetic trees inferring the evolutionary relationships among the nine genes by two independent methods. Both approaches yielded trees with similar topologies. In addition, the sequence alignments revealed patterns of nucleotide substitutions which implicate both point mutation and recombination in the divergence of the H-2 genes. Future considerations for evolutionary analysis of class I genes are discussed.

Abstract Abstract 1889 Background: Graft-versus-host disease (GVHD) is a frequent complication of allogeneic hematopoietic cell transplantation (HCT). While cytolytic effector function of donor-derived CD8+ cytotoxic T lymphocytes (CTLs) is critical for the elimination of the underlying malignant disease, the goal is to achieve this graft-versus-leukemia (GVL) effect without inducing GVHD. We and others showed in murine models that administration of AAT in the peri-transplant period decreased GVHD incidence and mortality. Concurrently, levels of pro-inflammatory cytokines were suppressed compared to albumin-treated recipients. Since both GVHD and GVL effects are initiated by donor cells, we asked whether donor treatment with AAT would modify GVHD or GVL effects in recipients. Of concern was the observation that AAT enhanced expression of heme oxygenase-1 (HMOX-1) which, via generation of carbon monoxide, enables the transcription factor Nrf2 to bind to the promoters of IL10 and IL1Ra, which might interfere with GVL activity. Methods: We determined the effect of donor AAT treatment on GVHD and GVL in two murine transplant models: C3H/SW(H-2Kbc) into C57Bl/6(H-2Kb) (minor mismatch), and C57Bl/6(H-2Kb) into BALB/C (H-2Kd) (major mismatch). Four regimens of donor and recipient treatment were studied: 1) Donor albumin /recipient albumin; 2) donor albumin/recipient AAT; 3) donor AAT/recipient albumin; 4) donor AAT /recipient AAT. AAT and albumin were given to donors at 3 μg/ dose every 72 hours for two weeks before cell harvesting. Recipients received 3 μ/dose every 72 hours following total body irradiation with 800 cGy. On day 4, recipients were injected with 5 × 106 T cell-depleted bone marrow (TCD-BM) plus 5 × 105 CD4+/CD8+ splenic T cells to induce acute GVHD. To test our hypothesis that donor AAT treatment would interfere with the GVL effect, BALB/C (H-2Kd) recipients were injected i.v. with 20 × 103 A-20 lymphoma cells containing a luciferase reporter construct (A20-luc/yfp; H-2Kd), following TBI. The infusion of TCD-BM and T cells from C57Bl/6(H-2Kb) donors was delayed until day 4 to allow the tumor to home and get established. Results: RT-PCR analysis of donor BM and T cells prior to transplantation showed up-regulation of HMOX-1 and Nrf2 (10 and 15 log2 increase, respectively), and enhanced transcription of IL-10 and IL-1Ra (50 and 10 log2, respectively, compared to albumin treated donors). In contrast, TNFa, was down-regulated (7 log2 decrease in comparison to albumin-treated donors) in both unsorted bone marrow and in CD8+/CD4+spleen cells from AAT pre-treated donors. FACS analysis revealed a 4-fold increase in frequency of CD56+NK cells, and a 5-fold increase in CD4+/CD24+FoxP3+ Treg cells, associated with a 3 fold increase in CD8+/CD205+ APCs/ DC in comparison to albumin treated donors. Mice transplanted with cells from AAT-treated donors showed lower clinical GVHD scores and decreased mortality (Figure 1) in comparison to mice transplanted from albumin-treated donors. BALB/C (H-2Kd) animals transplanted from AAT-treated donors showed significantly lower A-20 tumor signal (luciferase) intensity (over the first 3 weeks post-HCT) than did mice transplanted from albumin-treated donors (n=6, mean 4.8 × 106 (AAT) versus,7 × 106 (albumin) photons/sec/mouse, P =.03). Summary and conclusions: These data suggest that pre-transplant in vivo AAT exposure of donor cells results in protection against acute GVHD. Tumor growth was not increased in mice transplanted from AAT treated donors and, in fact, appeared to be suppressed, presumably related to a significant increase in NK cells. Анотація:
The aim of this study is to find the influence of cutting parameters and tools on surface roughness and dimensional deviation for better production quality in machining 0Cr18Ni12Mo2Cu2 steel. The most important parameters like cutting velocities, depth of cut and feed rate are considered. The experiments designed four types of boring tool rake angle and two types of coolant, and measured surface roughness and radial runout of workpiece with three acclivitous holes. The results showed that the surface integrity of workpiece obtained by 0.16 mm/rev feed rate with rake angle of tool 25o were superior to that obtained by the previous boring processes. Radial runout precision was highest when feed rate and rake angle were 0.13 mm/rev and 23o, respectively. According to experimental results, the rake angle and structure of boring tool were improved.

Анотація:
This paper described the evaluation of rigid sail performances by using the wind tunnel test. The rigid sail models were developed in the three variations of shape geometry which was having the same value of aspect ratio. The tests were performed to investigate the performances of sail models in terms of lift, drag, resultant, driving and heeling coefficient. The three sail models were tested at the variation of angle of attack such as 15o, 20o and 25o respectively, and it was fluided by a uniform flow of air with three different speeds. The comparisons of test results were evaluated to look for which sail models had the best performance. Based on the test results, Model 2 which is a triangle shape had generated a maximal efficiency and thrust force compare to the other models.Keywords: rigid sail, wind tunnel, sailing ship, renewable energy

The district along with its surrounding areas basically the south central parts are very suitable for the cultivation of banana one of the important fruit crop of the district. The district is not only rich in the cultivation and production of banana but also it has some big collecting centers mainly at Daranggiri, Dhupdhara, Rongjuli, Dudhnoi, etc. The century-old Daranggiri banana market in Goalpara district has now reached the peak of selling with rank first as the largest banana market in Asia. Bananas from Daraggiri are being supplied to Bangladesh, Nepal, Bhutan, Bihar, West Bengal, Uttar Pradesh, and many other places of India and abroad. Abstract In utero hematopoietic stem cell transplantation (IUHSCTx) can achieve mixed hematopoietic chimerism and donor-specific tolerance without cytoreductive conditioning or immunosuppression. The primary limitation to the clinical application of IUHSCTx has been minimal donor cell engraftment, well below therapeutic levels for most target diseases. Donor lymphocyte infusion (DLI) has been used in postnatal circumstances of mixed chimerism as targeted immunotherapy to achieve a graft-versus-hematopoietic effect and to increase levels of donor cell engraftment. In this report we demonstrate in the murine model that a combined approach of IUHSCTx followed by postnatal DLI can convert low-level, mixed hematopoietic chimerism to complete donor chimerism across full major histocompatibility complex barriers with minimal risk for graft-versus-host disease (GVHD). Time-dated embryonic day 14 (E14) to E15 Balb/c (H-2Kd, CD45.2) fetuses underwent intraperitoneal injection of 5 × 106T-cell–depleted B6 (H-2Kb, CD45.2) bone marrow cells. Chimeric recipients then received transplants at either 4 or 8 weeks of age with 1 of 3 doses (5, 15, or 30 × 106cells) of donor congenic splenocytes (B6-Ly5.2/Cr, H-2Kb, CD45.1). The response to DLI was dose dependent, with conversion to complete donor peripheral blood chimerism in 100% of animals that received high-dose (30 × 106 cells) DLI. Only 1 of 56 animals receiving this dose succumbed to GVHD. This study directly supports the potential therapeutic strategy of prenatal tolerance induction to facilitate nontoxic postnatal cellular therapy and organ transplantation, and it has broad implications for the potential treatment of prenatally diagnosed genetic disorders.

Abstract NK cells reject non-self hematopoietic bone marrow (BM) grafts via Ly49 receptor-mediated MHC class I-specific recognition and calibration of receptor expression levels. In this paper we investigated how Ly49+ subset frequencies were regulated dependent on MHC class I expression. The development of donor and host Ly49A+ (recognizes H-2Dd and H-2Dk ligands) and Ly49C/I+ (Ly49CBALB/c recognizes H-2Kb, H-2Kd, and H-2Dd, and Ly49CB6 recognizes only H-2Kb) NK cell frequencies were monitored for 120 days in murine-mixed allogeneic BM chimeras. C57BL/6 (H-2b) BM was transplanted into BALB/c (H-2d) mice and vice versa. Peripheral NK cell populations were examined every 5 days. Chimerism was found to be stable with 80–90% donor NK cells. In contrast to syngeneic controls reexpressing pretransplant patterns, donor and host NK cells revealed new and mainly reduced subset frequencies 55 days after allogeneic transplantation. Recipient NK cells acquired these later than donor NK cells. In H-2d → H-2b chimeras Ly49A+, Ly49C/I+, and Ly49A+/Ly49C/I+ proportions were mainly diminished upon interaction with cognate ligands. Also in H-2b → H-2d chimeras, Ly49A+ and Ly49A+/Ly49C/I+ subsets were reduced, but there was a transient normalization of Ly49C/I+ proportions in the noncognate host. After 120 days all subsets were reduced. Therefore, down-regulation of developing Ly49A+ and Ly49C/I+ chimeric NK cell frequencies by cognate ligands within 7–8 wk after BM transplantation may be important for successful engraftment.

The abundance of clay in the Merauke district has not been utilized optimally. This is due to a lack of research and information about these clays. Clay is one of the natural minerals from the silica group whose particle size is less than two micrometers and has many uses, including basic materials for the manufacture of ceramics, paper-making materials, adsorbents, assisting drilling, and bleaching in CPO. In this study, characterization has been carried out including functional group characterization using an FTIR spectrophotometer, crystallinity using XRD, and morphological analysis by SEM. The results obtained that the mineral content of the clay from the sloping soil district was proposed to contain Illite and Quartz minerals with the main peaks of the diffractogram appearing at an angle of 2θ about 21o; 26o; 36o; and 42o; at angles d (Å) 3.34 and d (Å) 4.25. XRD results were also tested using search-match analysis, and the sample intensity was obtained containing quartz minerals (~ 98%). In the development of this material, it is necessary to obtain XRD data to determine the exact components of clay.

Rhynchophorus phoenicis, found in the tropical regions of Africa where it is regarded as a pest was used to synthesize chitosan and alumina-chitosan nano-composite. The synthesized Chitosan and alumina-chitosan nano-composite were characterized using instrumental methods of analysis which include Fourier Transform Infrared Spectroscopy (FTIR), X-ray Diffraction (XRD), Scanning Electron Microscope (SEM). FTIR reveals the existence of OH stretching vibration –NH stretching, NH bonding, vibration of methylene C-H bonding, O=C=O stretching, C-N bonding stretching and CO stretching vibration at 3842.33 cm-1, 3402.54 cm-1, 3286.81 cm-1, 2939.61 cm-1, 1527.67 cm-1, 1303.92 cm-1 and 1041.60 cm-1 respectively. The scanning electron micrograph proves that the chitosan has large particles that are regular with paltry pores while alumina-chitosan nano-composite has fairly small particles and porous surface with significant pores. X-ray diffractogram showed a distinct stretch at 22o; 16o and 25o; 17o for chitosan and alumina-chitosan nano-composite. The characterization of the products confirmed that the synthesis of chitosan and alumina-chitosan nano-composiote was effective.

Анотація:
This study examines the formation of Boron carbide with a wide homogeneity range B8C18 and B25C, from boric acid-glucose and boric acid-active carbon as precursor materials. The samples was analysed by means of X-ray diffraction and Fourier Transform Infrared spectrometer. X-ray diffraction pattern was analysed by GSAS software. X ray diffraction pattern of B8C18 shows peaks at 27o, 39o, 45o, 65o and 79o, orthorhombic crystal system and lattice parameters a = 13.640 Ǻ, b = 7.8500 Ǻ and c = 12.910 Ǻ and space group P212121, whereas for B25C sample show peaks at 2θ angle 28o and 40o, tetragonal crystal system, and the lattice parameters a = b = 8.753 Ǻ and c = 5.093 Ǻ and space group P-4 2 m. FTIR results show that for B8C18 have absorption bands with B-C bond at 1196.5 cm-1, 0-H 3216.7 cm-1 and B-O at 1477.1 cm-1, whereas the formation B25C have absorption bands with B-C bond at 1195.1 cm-1, O-H 3216.7 cm-1 and B-O 1460.8 cm-1.
