Neste estudo foram utilizadas as técnicas de birrefringência magnética estática, espectroscopia fotoacústica e microscopia eletrônica de transmissão para investigar o efeito da diluição de amostras de fluido magnético biocompatível à base de maghemita recoberta com ácido dimercaptosuccinico. As amostras investigadas apresentam características semelhantes, pórem com diferentes graus de cobertura molecular. Os dados obtidos foram analisados comparando, entre si, as diferentes amostras usadas e analisando as características de cada uma em função do tempo. In this study static magnetic birefringence, photoacoustic spectroscopy and transmission electronic microscopy were used to investigate the dilution effect of dimercaptosuccinic acidcoated maghemite nanoparticles suspended as biocompatible magnetic fluids. The samples investigated have similar characteristics though presenting different surface grafting coefficient. The data were analyzed comparing different diluted samples and the time evolution of their properties. Повний текст джерелаThe Earths magnetic field presents some specific characteristics in the South American continent, including a region where a significant decrease in intensity is observed. This region is called the South Atlantic Magnetic Anomaly (SAMA), at which an approximation of the internal radiation belt results in enhanced precipitation of energetic particles into the atmosphere. This particle precipitation increases the electrical conductivity in much of the South American ionosphere, producing peculiar aeronomic effects. This work aimed to study the effects of SAMA on Pc3 and Pc4 magnetic pulsations recorded by fluxgate magnetometers operated by the Brazilian Studies and Monitoring of Space Weather (EMBRACE) and by the INTERMAGNET global network in different ground stations. The data were used to evaluate effects produced during magnetically quiet and disturbed periods in the year 2015. Except for an anomalous result observed for Pc3 during a quiet interval at the March equinox, possibly associated with the increase in Total Electronic Content, pulsation amplitudes showed a consistent amplification in SAMA, being the largest amplification factors observed near the central region of the anomaly. A generic model is suggested as the source of the pulsations, which includes the generation of surface waves by plasma instabilities at the dayside high-latitude magnetosphere boundary, the propagation of these waves through magnetic field lines until polar latitudes and the subsequent leakage of the currents toward lower latitudes by an Earthionosphere waveguide. The horizontal transmission of these polar fields through the SAMA is influenced by variations of ionospheric conductivity and generates ground geomagnetic pulsations with different amplitude as a function of the distance to the center of the anomaly.

Nessa tese foram investigadas as propriedades magnéticas e fotoacústicas de nanopartículas de maghemita dispersas em microesfera de BSA (bovine serum albumine), nanopartículas de ferrita de cobalto dispersas em microesfera de PLA (polylactic acid), nanopartículas de ferrita de cobalto peptizadas na forma de fluido magnético iônico e nanopartículas de magnetita coberta com dextrana e DMSA (dimercaptosuccinic acid) e peptizadas na forma de fluido magnético biocompatível. Foram realizadas medidas de microscopia eletrônica de transmissão (MET) e medidas de difração de raios-X para obtenção das micrografias, difratogramas, diâmetros médios e polidispersão das nanopartículas investigadas. Medidas Mössbauer permitiram a caracterização da fase magnética dos nanocompósitos (maghemita em BSA e ferrita de cobalto em PLA). Medidas de fotoacústica foram realizadas e analisadas em termos de três bandas (L, S e C). Medidas de susceptibilidade dinâmica foram realizadas na faixa de freqüência de 2 a 80 MHz, em temperatura ambiente e de nitrogênio líquido. Finalmente, a técnica de ressonância magnética foi empregada na investigação das propriedades magnéticas das amostras. As medidas de ressonância magnética foram realizadas em um espectrômetro comercial Bruker ESP-300 operando em banda-X (9,6 GHz), permitindo a investigação das amostras em função da temperatura (100 a 300K) e concentração das nanopartículas. As dependências do campo de ressonância e largura de linha de ressonância com a temperatura foram analisadas usando a aproximação proposta por Morais e colaboradores [98, 110, 112]. __________________________________________________________________________________________ ABSTRACT
In this thesis the magnetic and photoacoustic properties of maghemite nanoparticles dispersed in BSA (bovine serum albumine) microsphere, cobalt ferrite nanoparticles dispersed in PLA (polylactic acid) microsphere, cobalt ferrite nanoparticles peptized as ionic magnetic fluid and magnetite nanoparticles surface-coated with dextran and DMSA (dimercaptosuccinic acid) and peptized as biocompatible magnetic fluids were investigated. Transmission electron microscopy and X-ray diffraction measurements were performed in order to obtain the micrographs, diffractograms, average diameter and polydispersity of the investigated nanoparticles. Mössbauer measurements allowed characterization of the nanocomposites magnetic phase (maghemite in BSA and cobalt ferrite in PLA). Photoacoustic measurements were performed and analyzed in terms of three bands (L, S, and C). Dynamic susceptibility measurements were performed in the frequency range of 2 to 80 MHz, at room temperature and liquid nitrogen temperature. Finally, the magnetic resonance technique was employed in the investigation of the magnetic properties of the samples. Magnetic resonance measurements were carried out in a commercial Bruker ESP-300 spectrometer operating in the X-band (9.6 GHz), allowing investigation of the samples as a function of the temperature (100 to 300K) and nanoparticle concentration. The temperature dependence of both resonance field and resonance linewidth were analyzed within the approximation proposed by Morais and coworkers [98, 110, 112].

Повний текст джерелаIn the present work, concepts of statistical inference are used for application and comparison of different methods of signal analysis in functional magnetic resonance imaging. The central idea is based on obtaining the probability distribution of the random variable of interest, for each method studied under different values of signal-to-noise ratio (SNR). This purpose is achieved by means of numerical simulations of the hemodynamic response function (HRF) with gaussian noise. This procedure allows us to assess the sensitivity and specificity of the methods employed by the construction of the ROC curves (receiver operating characteristic) for different values of SNR. Under specific experimental conditions, we apply classical methods of analysis (Student\'s t test and correlation), information measures (distance of Kullback-Leibler and its generalized form) and a Bayesian method (independent pixel method). In particular, we show that the distance of Kullback-Leibler D (or relative entropy) and its generalized form are useful measures for analysis of signals within the information theory scenario. These entropies are used as measures of the \"distance\"between the probability functions p1 and p2 of the signal levels related to stimulus and non-stimulus. In order to avoid undesirable divergences of D, we introduced a small parameter d in the definitions of p1 and p2. We extend such analysis, by presenting an original study of the generalized Kullback-Leibler distance Dq (q is Tsallis parameter). In this case, the appropriate choice of range 0 < q < 1 ensures that Dq is finite. We obtain the probability densities f (D) and f (Dq) of the sample averages of the variables D and Dq, respectively, calculated over the N epochs of the entire experiment. For small values of N (N < 30), we show that f (D) and f (Dq) are well approximated by Gamma distributions (qui^2 < 0.0009). Afterward, we studied the independent pixel bayesian method, considering the probability a posteriori as a random variable, and obtaining its distribution for various SNR\'s and probabilities a priori. The results of simulations point to the fact that the correlation and the independent pixel method have better performance than the other methods used (for SNR> -20 dB). However, one should consider that the Student\'s t test and the entropic methods share the advantage of not using a model for HRF in real data analysis. Finally, we obtain the maps corresponding to real data series from an asymptomatic volunteer submitted to an event-related motor stimulus, which shows brain activation in the primary and secondary motor brain areas. We emphasize that the procedure adopted in this study may, in principle, be used in other methods and under different experimental conditions.

A área de estudo, localizada no município de Guatapará, possui 870 ha e é caracterizada pela transição geológica Basalto - Depósito Aluvionar - Depósito Colúvio Eluvionar, formas da paisagem côncavas e convexas e presença de Latossolos e Neossolo. Foram coletadas 371 amostras na profundidade de 0-0,25 m para a avaliação da SM, teor de argila e V%. A incorporação das informações secundárias nos modelos geoestatísticos foi realizada por meio da krigagem simples com médias locais variando. Para a SM as informações sobre a geologia e geomorfologia foram responsáveis por cerca de 45% e 44% da sua continuidade espacial. Para o teor de argila, estes resultados foram superiores indicando 54% (geologia) e 53% (geomorfologia). Para o V%, não foi observada relação entre as informações secundárias e sua continuidade espacial. As incorporações das informações geológicas e geomorfológicas aos dados de SM propiciaram, respectivamente, a redução em 44% e 37% no número de amostras requeridas para representar o padrão espacial do atributo. Para o teor de argila foi verificada a possibilidade de redução em 35% no número de amostras...
The lack of information on spatial variability of soil attributes is one of the limiting factors for the development of different areas of knowledge and productive sectors. This information and the knowledge of their uncertainties allow the correct interpretation of results and possible decision making. The study was conducted in two stages: the first stage aimed to use geological, geomorphological, and pedological information in order to reduce the sampling density needed for the spatial characterization of magnetic susceptibility (MS), clay content and base saturation (BS); the second stage aimed to use the spatial uncertainty of MS and clay content for the delineation of mapping units. The study area, located in the municipality of Guatapará, has 870 ha and is characterized by geological transition Basalt - Alluvial Deposit - Eluvial-Colluvial Deposit, concave and convex landforms and presence of Oxisols and Entisols. 371 samples were collected at a depth of 0-0.25 m for the evaluation of MS, clay content and BS. The incorporation of secondary information in geostatistical models was performed through the simple kriging with varying local means. For MS, information about geology and geomorphology were responsible for approximately 45% and 44% of its spatial continuity. For clay content, these results were higher, indicating 54% (geology) and 53% (geomorphology). For BS, there was no relation between secondary information and its spatial continuity. The incorporation of geological and geomorphology information to the MS data allowed a reduction of 44% and 37% in the number of samples required to represent the spatial pattern of the attribute. For clay content was verified the possibility of reducing by 35% the number of samples using both information. Повний текст джерелаThe aim of this work is to characterize a long-span absolute magnetic position transducer that presents a robust dynamic response. The transducer was conceived by the MSc studies of the author and is called named “Magnetic Ruler” by which its static response was studied. Unlike other magnetic transducers, the magnetic ruler is based on the principle of the division of magnetic flux between two distinct magnetic paths that depends on the linear position of its movable part along the span of the transducer. This present study examines the dynamic response of the transducer when its movable part or car is moving with constant speed. Under such conditions, there is a change in the signal output that is correlated to the position of that car. The study was conducted by means of two transducers of approximately 2-m long laminations of Si-Fe that were laser cut. The voltage signals induced through the search coils on the end yokes, current and magnetic flux in the excitation coil on car, and the position are acquired by a digital device and processed by a computer. That is done to obtain the static and the dynamic curves and, thus, to allow the comparison between the two behaviors. With this objective, a specially built and unique test bench was developed to test linear position transducers under static and dynamic cases. The difference between the two results is called dynamic difference. That test bench employs a resistive position transducer to provide position reference, a signal acquisition system for measuring the quantities involved in the process, and a controlled traction system based on a servo system to control the speed and the position of the moving part of the transducer under test. At the end this work the analytical and experimental results obtained as described above are compared.

Повний текст джерелаThis thesis presents Local Sparse Component Analysis (LSCA), a new method for analyzing resting state functional magnetic resonance imaging (fMRI) datasets. LSCA, a extension of Sparse Component Analysis (SCA), takes into account data spatial information to reconstruct temporal sources representing connected regions of significant activity. This study contains simulation data and real data. The simulated data were prepared to evaluate the LSCA in different scenarios. In the first scenario, the LSCA is compared with Principal Component Analysis (PCA) for detecting local sources under Gaussian white noise. Then, LSCA is compared with the expectation maximization algorithm (EM) for detecting the dynamics of local sources. Real data were collected for comparative and illustrative purposes. FMRI images from eleven healthy volunteers were acquired using a 3T MRI scanner during a resting state protocol. Images were preprocessed and analyzed using LSCA and Independent Components Analysis (ICA). LSCA components were compared with commonly reported ICA components. In addition, LSCA was applied for characterizing the dynamics of resting state networks. Simulated results have shown that LSCA is suitable for identifying local sparse sources.For real resting state FMRI data, LSCA is able to identify the same sources that are identified using ICA, allowing detailed functional connectivity analysis of the identified regions within and between components. This suggests that ICA resting state networks can be further decomposed into local sparse components that are not necessarily independent from each other. Moreover, LSCA sources better represent local FMRI signal oscillations than ISCA sources. Finally, brain connectivity analysis shows that LSCA can identify both instantaneous and causal relationships between resting state components. The main implication of this study is that independence and sparsity are equivalent assumptions in resting state FMRI. However, less restrictive criteria such as sparsity and source localization allow building much more compact and biologically plausible brain connectivity models.

Повний текст джерелаThe intervertebral disc is a structure whose function is to receive, absorb and transmit the impact loads imposed on the spine. Increasing age and the posture adopted by the individual can lead to degeneration of the intervertebral disc. Currently, Magnetic Resonance Imaging (MRI) is considered the best and most sensitive noninvasive method to imaging evaluation of the intervertebral disc. In this work were developed methods for quantitative computer-aided diagnosis of the intervertebral disc degeneration in MRI T2 weighted images of the lumbar column according to Pfirrmann scale, a semi-quantitative scale with five degrees of degeneration. The algorithms were tested on a dataset of 300 images obtained from 102 subjects with varying degrees of degeneration. Binary masks manually segmented of the discs were used to calculate their centroids, to create a reference point to enable extraction of attributes. A texture analysis was performed using the approach proposed by Haralick. For the shape characterization, invariant moments defined by Hu and central moments were also calculated for each disc. The rating of the degree of degeneration was performed using an artificial neural network and the set of extracted attributes of each disk. An average rate of correct classification of 87%, with standard error 6.59% and an average area under the ROC curve (Receiver Operating Characteristic) of 0.92 indicates the potential application of the algorithms developed as a diagnostic support tool to the degeneration of the intervertebral disc.

A elaboração e o refinamento de sistemas de liberação controlada têm sido extensivamente estudados a fim de se aprimorar as terapias com fármacos convencionais e aumentar a adesão do paciente ao tratamento. Para esses fins, vários polímeros biodegradáveis – como a quitosana – conjugados com outros materiais que trazem respostas estímulo-responsíveis ao compósito – como magnetismo – têm sido usados na pesquisa de transporte e liberação de fármacos. Nesse tipo de formulação, o comportamento físico-químico do biopolímero permite a incorporação do princípio ativo na sua estrutura e as estruturas magnéticas agem como sondas para conduzir magneticamente o compósito a uma região específica e/ou, por meio de suas propriedades físicas, causar uma hipertermia magnética que pode controlar a liberação de determinado fármaco pela aplicação de um campo magnético externo. Assim, nesse trabalho, nanoestruturas de quitosana de ~100 nm, incrustradas com até 25% de nanopartículas magnéticas de óxidos de ferro (magnetita/maguemita) de ~5 nm foram produzidas por precipitação em meio homogêneo, pela decomposição da ureia. Nessa mesma, e única etapa de síntese, o 5-hidroxitriptofano foi incorporado ao compósito, com uma eficiência de até 80 %. A caracterização química, morfológica e estrutural foi feita por XRD, TEM, FTIR, TGA, medidas de magnetização e de potencial zeta, enquanto a eficiência de incorporação e o perfil de liberação foram estudados por UV-vis. A cinética de liberação mostrou que trata-se de um sistema de liberação controlada pH-sensitivo governada pelo entumescimento e/ou dissolução da fase polimérica, sendo mais efetiva em meio ácido. O difratograma revela que há coexistência entre a fase polimérica e a magnética; por outro lado a espectroscopia no infravermelho confirma a interação entre quitosana e magnetita. O nanocompósito apresentou uma magnetização de saturação de 11 emu/g enquanto que a do material magnético foi de 50 emu/g; comparável à das ferritas; em função da análise termogravimétrica o nanocompósito apresentou um percentual de 24,6% de nanopartículas magnéticas. ________________________________________________________________________________ ABSTRACT
Design and tailoring of controlled release systems have been extensively studied in order to enhance drug therapy and increase patient compliance. For these purposes, various biodegradable polymers – such as chitosan – conjugated with another material which brings stimuli-responsive features – such as magnetism – to the composite have been used in drug delivery research. In this formulation, the physical-chemical behavior of biopolymer allows incorporation of pharmaceuticals in its structure and opens the possibility for applications in drug delivery. Here, the nanomagnets act as probes to magnetically drive the composite to a specific region and/or, through their magneto-thermal properties, can control a drug release with an external magnetic field. Thus, in this work, ~100 nm sized nanoparticles of chitosan embedded with 25 % (w/w) of iron oxide magnetic nanoparticles (magnetite/maghemite) and with a loading efficiency of about 80% for 5-hydroxytryptophan were synthesized, using homogeneous precipitation by urea decomposition, in an efficient one-step procedure. Characterization of morphology, structure and surface were performed by XRD, TEM, FTIR, TGA, magnetization and zeta potential measurements, while drug loading and drug releasing were investigated using UV-vis spectroscopy. Kinetic drug release experiments under different pH conditions revealed a pH-sensitive controlled-release system, ruled by polymer swelling and/or particle dissolution, and that drug release is more efficient in an acidic medium. he XRD pattern reveals that there is coexistence between the polymer phase and magnetic; On the other hand the infrared spectroscopy confirms the interaction between chitosan and magnetite. The nanocomposite showed a saturation magnetization of 11 emu / g while that of the magnetic material was 50 emu / g; comparable to ferrites; depending on the thermal analysis the nanocomposite showed a percentage of 24.6% of magnetic nanoparticles.
