ABSTRACT Csr (carbon storage regulation) of Escherichia coli is a global regulatory system that consists of CsrA, a homodimeric RNA binding protein, two noncoding small RNAs (sRNAs; CsrB and CsrC) that function as CsrA antagonists by sequestering this protein, and CsrD, a specificity factor that targets CsrB and CsrC for degradation by RNase E. CsrA inhibits translation initiation of glgC, cstA, and pgaA by binding to their leader transcripts and preventing ribosome binding. Translation inhibition is thought to contribute to the observed mRNA destabilization. Each of the previously known target transcripts contains multiple CsrA binding sites. A position-specific weight matrix search program was developed using known CsrA binding sites in mRNA. This search tool identified a potential CsrA binding site that overlaps the Shine-Dalgarno sequence of hfq, a gene that encodes an RNA chaperone that mediates sRNA-mRNA interactions. This putative CsrA binding site matched the SELEX-derived binding site consensus sequence in 8 out of 12 positions. Results from gel mobility shift and footprint assays demonstrated that CsrA binds specifically to this site in the hfq leader transcript. Toeprint and cell-free translation results indicated that bound CsrA inhibits Hfq synthesis by competitively blocking ribosome binding. Disruption of csrA caused elevated expression of an hfq′-′lacZ translational fusion, while overexpression of csrA inhibited expression of this fusion. We also found that hfq mRNA is stabilized upon entry into stationary-phase growth by a CsrA-independent mechanism. The interaction of CsrA with hfq mRNA is the first example of a CsrA-regulated gene that contains only one CsrA binding site.

ABSTRACT The csr regulatory system of Salmonella regulates the expression of the genes of Salmonella pathogenicity island 1 (SPI1) required for the invasion of epithelial cells. This system consists of the posttranscriptional regulator CsrA and an untranslated regulatory RNA, CsrB, that opposes the action of CsrA. Here we identify and characterize the role of a second regulatory RNA, CsrC, whose ortholog was discovered previously in Escherichia coli. We show that a mutant of csrC has only mild defects in invasion and the expression of SPI1 genes, as does a mutant of csrB, but that a double csrB csrC mutant is markedly deficient in these properties, suggesting that the two regulatory RNAs play redundant roles in the control of invasion. We further show that CsrC, like CsrB, is controlled by the BarA/SirA two-component regulator but that a csrB csrC mutant exhibits a loss of invasion equivalent to that of a barA or sirA mutant, indicating that much of the effect of BarA/SirA on invasion functions through its control of CsrB and CsrC. In addition to their control by BarA/SirA, each regulatory RNA is also controlled by other components of the csr system. The loss of csrB was found to increase the level of CsrC by sevenfold, while the loss of csrC increased CsrB by nearly twofold. Similarly, the overexpression of csrA increased CsrC by nearly 11-fold and CsrB by 3-fold and also significantly increased the stability of both RNAs.

ABSTRACTCyclic AMP (cAMP) and the cAMP receptor protein (cAMP-CRP) and CsrA are the principal regulators of the catabolite repression and carbon storage global regulatory systems, respectively. cAMP-CRP controls the transcription of genes for carbohydrate metabolism and other processes in response to carbon nutritional status, while CsrA binds to diverse mRNAs and regulates translation, RNA stability, and/or transcription elongation. CsrA also binds to the regulatory small RNAs (sRNAs) CsrB and CsrC, which antagonize its activity. The BarA-UvrY two-component signal transduction system (TCS) directly activatescsrBandcsrC(csrB/C) transcription, while CsrA does so indirectly. We show that cAMP-CRP inhibitscsrB/Ctranscription without negatively regulating phosphorylated UvrY (P-UvrY) or CsrA levels. Acrpdeletion caused an elevation in CsrB/C levels in the stationary phase of growth and increased the expression ofcsrB-lacZandcsrC-lacZtranscriptional fusions, although modest stimulation of CsrB/C turnover by thecrpdeletion partially masked the former effects. DNase I footprinting and other studies demonstrated that cAMP-CRP bound specifically to three sites located upstream from thecsrCpromoter, two of which overlapped the P-UvrY binding site. These two proteins competed for binding at the overlapping sites.In vitrotranscription-translation experiments confirmed direct repression ofcsrC-lacZexpression by cAMP-CRP. In contrast, cAMP-CRP effects oncsrBtranscription may be mediated indirectly, as it bound nonspecifically tocsrBDNA. In the reciprocal direction, CsrA bound tocrpmRNA with high affinity and specificity and yet exhibited only modest, conditional effects on expression. Our findings are incorporated into an emerging model for the response of Csr circuitry to carbon nutritional status.IMPORTANCECsr (Rsm) noncoding small RNAs (sRNAs) CsrB and CsrC ofEscherichia coliuse molecular mimicry to sequester the RNA binding protein CsrA (RsmA) away from lower-affinity mRNA targets, thus eliciting major shifts in the bacterial lifestyle. CsrB/C transcription and turnover are activated by carbon metabolism products (e.g., formate and acetate) and by a preferred carbon source (glucose), respectively. We show that cAMP-CRP, a mediator of classical catabolite repression, inhibitscsrCtranscription by binding to the upstream region of this gene and also inhibitscsrBtranscription, apparently indirectly. We propose that glucose availability activates pathways for both synthesis and turnover of CsrB/C, thus shaping the dynamics of global signaling in response to the nutritional environment by poising CsrB/C sRNA levels for rapid response.

Escherichia coli C is a strong biofilm producer in comparison to E. coli K-12 laboratory strains due to higher expression of the pgaABCD operon encoding the enzymes for the biosynthesis of the extracellular polysaccharide poly-β-1,6-N-acetylglucosamine (PNAG). The pgaABCD operon is negatively regulated at the post-transcriptional level by two factors, namely CsrA, a conserved RNA-binding protein controlling multiple pathways, and the RNA exonuclease polynucleotide phosphorylase (PNPase). In this work, we investigated the molecular bases of different PNAG production in C-1a and MG1655 strains taken as representative of E. coli C and K-12 strains, respectively. We found that pgaABCD operon expression is significantly lower in MG1655 than in C-1a; consistently, CsrA protein levels were much higher in MG1655. In contrast, we show that the negative effect exerted by PNPase on pgaABCD expression is much stronger in C-1a than in MG1655. The amount of CsrA and of the small RNAs CsrB, CsrC, and McaS sRNAs regulating CsrA activity is dramatically different in the two strains, whereas PNPase level is similar. Finally, the compensatory regulation acting between CsrB and CsrC in MG1655 does not occur in E. coli C. Our results suggest that PNPase preserves CsrA-dependent regulation by indirectly modulating csrA expression.

Orthologues of the Salmonella enterica serovar Typhimurium (S. typhimurium) BarA/SirA two-component system are important for biofilm formation and virulence in many γ-Proteobacteria. In S. typhimurium, SirA activates the csrB and csrC carbon storage regulatory RNAs and the virulence gene regulators hilA and hilC. The regulatory RNAs antagonize the activity of the CsrA protein, allowing translation of those same virulence genes, and inhibiting the translation of flagellar genes. In this report, it was determined that SirA and the Csr system also control the fim operon that encodes type 1 fimbriae. sirA orthologues in other bacterial species, and the fim operon of S. typhimurium, are known to play a role in biofilm formation; therefore, all members of the S. typhimurium sirA regulon were tested for in vitro biofilm production. A sirA mutant, a csrB csrC double mutant, and a fimI mutant, were all defective in biofilm formation. Conversely, inactivation of flhDC increased biofilm formation. Therefore, SirA activates csrB, csrC and the fim operon to promote biofilm formation. In turn, csrB and csrC promote the translation of the fim operon, while at the same time inhibiting the translation of flagella, which are inhibitory to biofilm formation.

The hybrid sensor kinase BarA and its cognate response regulator UvrY, members of the two-component signal transduction family, activate transcription of CsrB and CsrC noncoding RNAs. These two small RNAs act by sequestering the RNA binding protein CsrA, which posttranscriptionally regulates translation and/or stability of its target mRNAs. Here, we provide evidence that CsrA positively affects, although indirectly,uvrYexpression, at both the transcriptional and translational levels. We also demonstrate that CsrA is required for properly switching BarA from its phosphatase to its kinase activity. Thus, the existence of a feedback loop mechanism that involves the Csr and BarA/UvrY global regulatory systems is exposed.

ABSTRACT The global regulator CsrA (carbon storage regulator) ofEscherichia coli is a small RNA binding protein that represses various metabolic pathways and processes that are induced in the stationary phase of growth, while it activates certain exponential phase functions. Both repression and activation by CsrA involve posttranscriptional mechanisms, in which CsrA binding to mRNA leads to decreased or increased transcript stability, respectively. CsrA also binds to a small untranslated RNA, CsrB, forming a ribonucleoprotein complex, which antagonizes CsrA activity. We have further examined the regulatory interactions of CsrA and CsrB RNA. The 5′ end of the CsrB transcript was mapped, and acsrB::cam null mutant was constructed. CsrA protein and CsrB RNA levels were estimated throughout the growth curves of wild-type and isogenic csrA,csrB, rpoS, or csrA rpoSmutant strains. CsrA levels exhibited modest or negligible effects of these mutations. The intracellular concentration of CsrA exceeded the total CsrA-binding capacity of intracellular CsrB RNA. In contrast, CsrB levels were drastically decreased (∼10-fold) in thecsrA mutants. CsrB transcript stability was unaffected by csrA. The expression of a csrB-lacZtranscriptional fusion containing the region from −242 to +4 bp of thecsrB gene was decreased ∼20-fold by acsrA::kanR mutation in vivo but was unaffected by CsrA protein in vitro. These results reveal a significant, though most likely indirect, role for CsrA in regulatingcsrB transcription. Furthermore, our findings suggest that CsrA mediates an intriguing form of autoregulation, whereby its activity, but not its levels, is modulated through effects on an RNA antagonist, CsrB.

ABSTRACT The global regulator CsrA (carbon storage regulator) is an RNA binding protein that coordinates central carbon metabolism, activates flagellum biosynthesis and motility, and represses biofilm formation in Escherichia coli. CsrA activity is antagonized by the untranslated RNA CsrB, to which it binds and forms a globular ribonucleoprotein complex. CsrA indirectly activates csrB transcription, in an apparent autoregulatory mechanism. In the present study, we elucidate the intermediate regulatory circuitry of this system. Mutations affecting the BarA/UvrY two-component signal transduction system decreased csrB transcription but did not affect csrA′-′lacZ expression. The uvrY defect was severalfold more severe than that of barA. Both csrA and uvrY were required for optimal barA expression. The latter observation suggests an autoregulatory loop for UvrY. Ectopic expression of uvrY suppressed the csrB-lacZ expression defects caused by uvrY, csrA, or barA mutations; csrA suppressed csrA or barA defects; and barA complemented only the barA mutation. Purified UvrY protein stimulated csrB-lacZ expression approximately sixfold in S-30 transcription-translation reactions, revealing a direct effect of UvrY on csrB transcription. Disruption of sdiA, which encodes a LuxR homologue, decreased the expression of uvrY′-′lacZ and csrB-lacZ fusions but did not affect csrA′-′lacZ. The BarA/UvrY system activated biofilm formation. Ectopic expression of uvrY stimulated biofilm formation by a csrB-null mutant, indicative of a CsrB-independent role for UvrY in biofilm development. Collectively, these results demonstrate that uvrY resides downstream from csrA in a signaling pathway for csrB and that CsrA stimulates UvrY-dependent activation of csrB expression by BarA-dependent and -independent mechanisms.

CsrA is a global post-transcriptional regulatory protein that has a great impact on many physiological pathways in a cell. It regulates central carbon metabolism, motility and biofilm formation as well as virulence, pathogenesis, quorum sensing and the oxidative stress response. However, CsrA is in turn also regulated by CsrB and CsrC sRNAs, among other factors. In this study, we explore its regulatory effect on three different genes/operons, cstA, glgCAP and flhDC. We performed beta-galactosidase assays on wild type (wt) and RNase P temperature sensitive (ts) cells to determine the dependence on RNaseP for the CsrA, CsrB and CsrC regulatory cascade on these three genes. Our results showed a clear decrease in cstA and glgCAP activity during CsrA activation, suggesting a regulatory cascade in which inhibition of RNase P leads to an inactivation of CsrB and CsrC. This in turn leads to an activation of CsrA and an inhibition of cstA and glgCAP. The effect on flhDC, however, was not as clear and needs further investigation.

Cette thèse s'intéresse à la simulation temps-réel de circuit audio nonlinéaires. Dans cette thèse, nous utilisons le formalisme des systèmes Hamiltoniens à ports (SHP) pour garantir le bilan de puissance et la passivité. De plus, nous adoptons un cadre fonctionnel à temps continu pour représenter des signaux "analogiques virtuels" et nous proposons d'approximer les solutions par projection sur des trames temporelles. En tant que résultat principal, nous établissons une condition suffisante sur les projecteurs de sorte à obtenir des trajectoires à bilan de puissance garanti. Notre but est double: premièrement, pour gérer l'expansion de bande-passante causée par les nonlinéarités, nous considérons des méthodes numériques traitant des signaux à bande non-limitée qui à la place ont un "taux d'innovation borné"; Deuxièmement, pour revenir dans le domaine des signaux à bande limitée, nous concevons des "convertisseurs analogique-numérique virtuels" Plusieurs méthodes numériques sont construites afin d'être à bilan de puissance garanti, avec une précision d'ordre élevé et un un ordre de régularité contrôlable. Leurs propriétés sont étudiées: existence et unicité, ordre de précision, dispersion, mais aussi, résolution fréquentielle au delà de la fréquence de Nyquist, rejet du repliement ainsi que noyaux reproduisants et noyaux de Peano. Cette approche révèle des ponts entre l'analyse numérique, le traitement du signal et la théorie de l'échantillonnage généralisé en mettant en relation la précision, la propriété de reproduction des polynômes, la bande passante ou les bancs de filtre de Legendre, etc. Nous exposons un cadre systématique pour transformer des schémas électronique en équations puis en simulations. Ce cadre est ensuite appliqué à des circuits audio représentatifs, contenant à la fois des équations différentielles ordinaires et des équation algebro-différentielles. Un travail spécifique est dédié à la modélisation SHP des amplificateurs opérationnels. Enfin, nous revisitons la modélisation des SHP dans le cadre de l'algèbre géométrique, ce qui ouvre des perspectives pour l'encodage de la structure géométrique des équations
This work addresses the real-time simulation of nonlinear audio circuits. In this thesis, we use the port-Hamiltonian (pH) formalism to guarantee power balance and passivity. Moreover, we adopt a continuous-time functional framework to represent "virtual analog" signals and propose to approximate solutions by projection over time frames. As a main result, we establish a sufficient condition on projectors to obtain time-continuous power-balanced trajectories. Our goal is twofold: first, to manage frequency-bandwidth expansion due to nonlinearities, we consider numerical engines processing signals that are not bandlimited but, instead, have a "finite rate of innovation"; second, to get back to the bandlimited domain, we design "virtual analog-to-digital converters". Several numerical methods are built to be power-balanced, high-order accurate, with a controllable regularity order. Their properties are studied: existence and uniqueness, accuracy order and dispersion, but also, frequency resolution beyond the Nyquist frequency, aliasing rejection, reproducing and Peano kernels. This approach reveals bridges between numerical analysis, signal processing and generalised sampling theory, by relating accuracy, polynomial reproduction, bandwidth, Legendre filterbanks, etc. A systematic framework to transform schematics into equations and simulations is detailed. It is applied to representative audio circuits (for the UVI company), featuring both ordinary and differential-algebraic equations. Special work is devoted to pH modelling of operational amplifiers. Анотація:
This study focuses on the topic of CSR and profitability and extends the current literature on the topic by examining the relationship between CSR and profitability from a game theory setting. Specifically, this study implements game modelling and develops two parts of analysis on the topic of CSR and profitability. The goal of part one analysis is to investigate competing CSR firms in comparison with competing non-CSR firms. The part two analysis is to study a CSR firm and a non-CSR firm that compete in the same market in two scenarios. In scenario 1, the CSR firm and its non-CSR counterpart compete for profitability. In scenario 2, the CSR firm is operating to optimize its defined welfare objective; whereas the non-CSR operates for a maximized profit level. The solutions to the models complemented with a detailed analysis reveal that the CSR could actually gain strategic and competitive advantages over its non-CSR counterpart in both defined scenarios. However, this study does not intend to advocate a positive relationship between CSR and profit performance. The positive relationship is qualified, and whether or not the relationship is positive, as this study shows, is dependent on the firm’s tactics.

Syfte: Syftet med denna studie är att skapa ökad förståelse för medias roll i förmedlingen av begreppet CSR, corporate social responsibility. Med media menas i studien svensk papperstidning. Metod: Diskursanalys kring CSR i svensk tryckpress, inspirerad av Michael Foucault och Norman Fairclough, applicerad på nyhetsartiklar. I studien används ordet diskurs som en förklaring till att nyhetstexter påverkar den sociala verkligheten. CSR innebär företags sociala ansvar, kring frågor om bland annat miljö, som går utöver vad lagen kräver. Slutsatser: Analysresultatet visar att CSR i nästintill samtliga artiklar benämns synonymt med samhällsansvar, som alltså varit det mest dominerande området som behandlats i de artiklar som har analyserats. Kulturgeografiska institutionen, Arbetsrapportsserie, Uppsala Universitet     The purposes with this paper are firstly to find out how two chosen clothing companies within the high fashion era handle the important CSR (Cooperate Social Responsibility) challenges that today has become a natural part of the clothing industry, and secondly how these companies are communicating their CSR strategy, and then to note any possible differences between their way of doing that. The methods I have chosen to use are by qualitively interviews with the two Fashion companies: Sidenkompaniet and Filippa K. Results from my research shows that both companies have different approaches to the conception of CSR and also uses different strategies how to communicate the CSR message. I found out that Sidenkompaniet, a small company is using the CSR concept as such to communicate their message and strategy. Filippa K though (the larger company), have another approach to CSR. They are not mentioning CSR, instead they want their customer to sense that everything they do is connected to CSR. They want to project CSR as a way of living and that should be felt all trough their business idea from production to the customer without ever mentioning abbreviation CSR. These are the two major different approaches I found during my research.

Denna uppsats berör och problematiserar kring ämnesområdet CSR, en förkortning av engelskans corporate social responsibility, eller som det heter på svenska; företagens sociala ansvar. Syftet med studien är att genom fem uppställda hypoteser undersöka om det finns något samband mellan omfattningen av företags CSR-kommunikation och variablerna: omsättning, segment (börsvärde), omsättning per anställd, branschtillhörighet, samt vilken typ utav CSR-kommunikation som används mest frekvent. För att besvara syftet genomfördes en kvantitativ undersökning där 254 företags årsredovisningar analyserades och poängsattes efter en framtagen skala rörande CSR-kommunikation. Den totala poäng de erhöll (företagens CSR-tal) jämfördes sedermera med ovan nämnda variabler genom regressionsanalys och medelvärdesberäkning. Teori kring ämnesområdet sammanställdes och användes senare som analysverktyg av de uppkomna resultaten. Genom undersökningen kan vi verifiera våra hypoteser gällande att omsättning och börsvärde skulle påverka företags CSR-tal samt att alla branscher arbetar mer med intern CSR-kommunikation än extern. Dock falsifierar vi hypotesen att företag med högre omsättning per anställd skulle kommunicera mindre kring sitt CSR-arbete. Slutligen fick vi delvis verifiera och delvis falsifiera hypotesen för vilka branscher som är mest benägna att kommunicera om sitt CSR-arbete.

- Hur tolkar företag begreppet CSR, hur arbetar de med det samt om det finns några skillnader/likheter mellan företagen?

- Hur kommunicerar företagen sitt CSR arbete via olika dokument, finns det några skillnader/likheter mellan företagen?

Syfte: Uppsatsen syftar till att beskriva, förklara och jämföra hur utvalda svenska företag inom konfektionsbranschen kommunicerar kring begreppet CSR mot sin marknad via olika dokument, hur företagen tolkar begreppet CSR samt hur de arbetar med det.

Metod: En fallstudie har genomförts på tre företag inom konfektionsbranschen på den svenska marknaden. Textanalys har genomförts på utvalda dokument såsom respektive företags hemsida, årsredovisningar från 2002 och framåt samt tillgängliga hållbarhetsredovisningar för att se hur företagen kommunicerar kring CSR. Även kvalitativa intervjuer med respektive företag har genomförts för att komplettera textanalysen och för att få ytterligare en synvinkel utöver kommunikation på hur företagen arbetar med CSR.

Teori: De teorier som använts är Carrolls CSR – pyramid samt företags olika ståndpunkter till CSR. Carrolls CSR pyramid tar hänsyn till olika ansvar som måste uppfyllas i CSR arbetet medan den andra teorin tar upp företags olika ståndpunkter till CSR samt hur ståndpunkten kan påverka i vilken utsträckning samt grad man arbetar med CSR.

Slutsats: När det gäller CSR arbetet finns det mer likheter än skillnader mellan företagen. Kommunikationen skiljer sig i omfattning mellan företagen.

Frågeställning: Vilka drivkrafter bidrar till avdelningarnas arbete med CSR? Hur genomför respektive avdelning sitt CSR-arbete samt vilka likheter och skillnader finns det? Hur samordnas CSR-arbete mellan avdelningarna? Slutsats: Att göra rätt saker i rätt avdelningen och att beroende på företagsavdelning göra olika CSR-aktiviteter är effektivt enligt uppsatsförfattarna. Samordningen däremellan behövs och för Stadium har kommunikation främst skett genom ett hållbarhetsråd. Uppsatsförfattarna ser också fler möjligheter, exempelvis dokumentation om CSR-strategi, resultat och nyckeltal. Om avdelningar tillsammans tar sitt ansvar, arbetar mot ett gemensamt mål samt stödjer varandra uppnås högre nytta och CSR är då ett strategi- och styrningsmedel som kan skapa nytta för företag och samhället. Problem discussion: The authors have not identified research on CSR strategy based on a departmental perspective. The knowledge gap that the study examines is therefore the business departments' driving forces to work with CSR, similarities and differences between departments' CSR work and how they are coordinated. Purpose: The purpose of the paper is to investigate how the departments' CSR work supports the integration of the company's overall CSR strategy. Research questions: What driving forces contribute to the departments' work with CSR? How do each department carry out its CSR work and what similarities and differences are there? How is CSR work coordinated between departments? Conclusion: To do the right things in the right department and to do different CSR activities according to the business department is effective according to the authors. Coordination between them is needed and for Stadium, communication has been done primarily through a Sustainability Council. The authors also see more opportunities, such as documentation about CSR strategy, results and key indicators. If departments take responsibility, work towards a common goal and support each other, greater benefit is achieved, and CSR is then a strategy and guidance that will benefit businesses and society. Science contributions: The study's knowledge contribution results in an increased understanding that coordination and integration between departments increases the chance of a total CSR strategy, with the desired strategic outcome of higher total value creation for companies and society.

Conversational semantic role labeling (CSRL) is a newly proposed task that uncovers the shallow semantic structures in a dialogue text. Unfortunately several important characteristics of the CSRL task have been overlooked by the existing works, such as the structural information integration, near-neighbor influence. In this work, we investigate the integration of a latent graph for CSRL. We propose to automatically induce a predicate-oriented latent graph (POLar) with a predicate-centered gaussian mechanism, by which the nearer and informative words to the predicate will be allocated with more attention. The POLar structure is then dynamically pruned and refined so as to best fit the task need. We additionally introduce an effective dialogue-level pre-trained language model, CoDiaBERT, for better supporting multiple utterance sentences and handling the speaker coreference issue in CSRL. Our system outperforms best-performing baselines on three benchmark CSRL datasets with big margins, especially achieving over 4% F1 score improvements on the cross-utterance argument detection. Further analyses are presented to better understand the effectiveness of our proposed methods.

The Harmonized Common Structural Rules (CSR-H) for Bulk Carriers and Oil Tankers has been issued on 1st Jan 2014, and will enter into force on 1st July 2015 to supersede the current two separate CSR versions, namely CSR-OT for Oil tankers and CSR-BC for bulk carriers. CSR-H aims at a consistent methodology harmonizing CSR-OT and CSR-BC, and establishes new criteria and requirements with the further aim of compliance with the IMO Goal Based Standard (GBS) where GBS functional requirements fall within Classification Rule scope. To maintain equivalent or higher safety level than current CSR, the rule developments within CSR-H are as a guideline aimed at scantling requirements that will be the same or higher for a given CSR vessel given the same ship design parameters and structural arrangement both globally and locally. The purpose of this study is to introduce the main changes from CSR to CSR-H, the new criteria set up for GBS compliance, and evaluate their impact on the structural design, especially on the scantlings. A carefully selected set of ten bulk carriers and eight oil tankers are investigated with the focus not only on the midship area but also on the foremost and aftmost cargo block areas as well. The typical critical areas affected by CSR-H due to fatigue or buckling criteria are identified, discussed and analyzed in detail. The impacts on scantlings and improvement suggestions for structural design based on CSR-H are further summarized and conclusions made.

The US Army Corps of Engineers, Galveston District, is executing the Sabine Pass to Galveston Bay Coastal Storm Risk Management (CSRM) project for Brazoria, Jefferson, and Orange Counties regions. The project is currently in the Pre-construction, Engineering, and Design phase. This report documents coastal storm water level and wave hazards for the Port Arthur CSRM structures. Coastal storm water level (SWL) and wave loading and overtopping are quantified using high-fidelity hydrodynamic modeling and stochastic simulations. The CSTORM coupled water level and wave modeling system simulated 195 synthetic tropical storms on three relative sea level change scenarios for with- and without-project meshes. Annual exceedance probability (AEP) mean values were reported for the range of 0.2 to 0.001 for peak SWL and wave height (Hm0) along with associated confidence limits. A response-based stochastic simulation approach is applied to compute AEP runup and overtopping for levees and overtopping, nappe geometry, and combined hydrostatic and hydrodynamic fluid pressures for floodwalls. CSRM structure crest design elevations are defined based on overtopping rates corresponding to incipient damage. Survivability and resilience are evaluated. A system-wide hazard level assessment was conducted to establish final recommended system-wide CSRM structure elevations.
