Abstract A series of Cas9 variants have been developed to improve the editing fidelity or targeting range of CRISPR–Cas9. Here, we employ a high-throughput sequencing approach primer-extension-mediated sequencing to analyze the editing efficiency, specificity and protospacer adjacent motif (PAM) compatibility of a dozen of SpCas9 variants at multiple target sites in depth, and our findings validate the high fidelity or broad editing range of these SpCas9 variants. With regard to the PAM-flexible SpCas9 variants, we detect significantly increased levels of off-target activity and propose a trade-off between targeting range and editing specificity for them, especially for the near-PAM-less SpRY. Moreover, we use a deep learning model to verify the consistency and predictability of SpRY off-target sites. Furthermore, we combine high-fidelity SpCas9 variants with SpRY to generate three new SpCas9 variants with both high fidelity and broad editing range. Finally, we also find that the existing SpCas9 variants are not effective in suppressing genome instability elicited by CRISPR–Cas9 editing, raising an urgent issue to be addressed.

AbstractIn this paper, we propose to work in the 2.5D space of the scene to facilitate composition of new spherical panoramas. For adding depths to spherical panoramas, we extend an existing method that was designed to estimate relative depths from a single perspective image through user interaction. We analyze the difficulties to interactively provide such depth information for spherical panoramas, through three different types of presentation. Then, we propose a set of basic tools to interactively manage the relative depths of the panoramas in order to obtain a composition in a very simple way. We conclude that the relative depths obtained by the extended depth estimation method are enough for the purpose of compositing new photorealistic panoramas through a few elementary editing tools.

Beyond time frame editing in video data, object level video editing is a challenging task; such as object removal in a video or viewpoint changes. These tasks involve dynamic object segmentation, novel view video synthesis and background inpainting. Background inpainting is a task of the reconstruction of unseen regions presented by object removal or viewpoint change. In this paper, we propose a video editing method including foreground object removal background inpainting and novel view video synthesis under challenging conditions such as complex visual pattern, occlusion, overlaid clutter and variation of depth in a moving camera. Our proposed method calculates a weighted confidence score on the basis of normalized difference between observed depth and predicted distance in 3D space. A set of potential points from epipolar lines from neighbor frames are collected, refined, and weighted to select a few number of highly qualified observations to fill the desired region of interest area in the current frame from video. Based on the background inpainting method, novel view video synthesis is conducted with arbitrary viewpoint. Our method is evaluated with both a public dataset and our own video clips and compared with multiple state of the art methods showing a superior performance.

Анотація:
Purpose The purpose of this paper is to reveal the co-editing mechanism aiming at content creation, and an entry of online encyclopedia is taken as a case, for the purpose of promoting and enhancing the development of wiki-based digital humanities projects (WDHPs), specifically, the projects that focus on gathering contextual information in the culture heritage domain. Design/methodology/approach An exploratory study was conducted by three steps. A representative entry’s editorial records were reorganized to obtain a data set of discussion statements (n=608), based on which linked-structures were built, and PageRank algorithm was used to analyze the co-editing process. Skewness statistic was applied to measure the consensus of co-editing, and consensus evolution over time was explored. Linear or curve fitting was performed to analyze the correlation between consensus evolution and its influential factors. Findings In WDHPs, co-editing activity of content creation can be considered as a large-scale group discussion, consensus can evaluate the efficiency of co-editing, which evolves with time and is influenced by the number of statements, breadth and depth of argumentation structure. Taking “Mogao Grottoes” as an example, group discussions around 15 key issues dominate the content creating process, consensus is on a rise with time, finally reaches a relatively high level, and consensus evolution is more influenced by breadth than by depth of argumentation structure, which indicates that co-editing efficiency of “Mogao Grottoes” is fine and more argumentation in a depth manner should be guided. Practical implications For researchers of WDHPs, it is beneficial to apply online encyclopedia platform combining with consensus analysis to develop WDHPs. For designers of WDHPs, the elements related to argumentation structure can be absorbed into the design to promote co-editing in an effective manner. For DH researchers, the analytic procedure can be beneficial of revealing the interest of contributors in a specific DH field. Originality/value This research is novel in comprehensively understanding co-editing mechanism of content creation in WDHPs, resulting in a three-step analytic procedure of presenting co-editing process, evaluating and improving co-editing efficiency.

Adenine base editors (ABEs) enable precise and efficient conversion of target A•T base pairs to G•C base pairs in genomic DNA with a minimum of by-products. While ABEs have been reported to exhibit minimal off-target DNA editing, off-target editing of cellular RNA by ABEs has not been examined in depth. Here, we demonstrate that a current ABE generates low but detectable levels of widespread adenosine-to-inosine editing in cellular RNAs. Using structure-guided principles to design mutations in both deaminase domains, we developed new ABE variants that retain their ability to edit DNA efficiently but show greatly reduced RNA editing activity, as well as lower off-target DNA editing activity and reduced indel by-product formation, in three mammalian cell lines. By decoupling DNA and RNA editing activities, these ABE variants increase the precision of adenine base editing by minimizing both RNA and DNA off-target editing activity.

This article traces the rapid progression of policy pertaining to human genome germline modifications using genome editing. It provides an overview of how one fertility physician implemented and advertised experimental techniques as part of his fertility clinic services, examines US law and policy, and assesses the impact of rhetoric influencing global policy and interpretation of the law. This article provides an in-depth examination of the medical rationale driving the acceptance of genome editing human embryos in two contexts: to cure disease and treat infertility. It describes complexities in genomics and outlines risks currently associated with genome editing, asserting the available evidence fails to demonstrate genome editing constitutes a curative therapy.

This thesis introduces methods for adding user-guided depth-aware effects to images captured with a consumer-grade stereo camera with minimal user interaction. In particular, we present methods for highlighted depth-of-field, haze, depth-of-field, and image relighting. Unlike many prior methods for adding such effects, we do not assume prior scene models or require extensive user guidance to create such models, nor do we assume multiple input images. We also do not require specialized camera rigs or other equipment such as light-field camera arrays, active lighting, etc. Instead, we use only an easily portable and affordable consumer-grade stereo camera. The depth is calculated from a stereo image pair using an extended version of PatchMatch Stereo designed to compute not only image disparities but also normals for visible surfaces. We also introduce a pipeline for rendering multiple effects in the order they would occur physically. Each can be added, removed, or adjusted in the pipeline without having to reapply subsequent effects. Individually or in combination, these effects can be used to enhance the sense of depth or structure in images and provide increased artistic control. Our interface also allows editing the stereo pair together in a fashion that preserves stereo consistency, or the effects can be applied to a single image only, thus leveraging the advantages of stereo acquisition even to produce a single photograph.

L’affichage de données stéréoscopiques estaujourd’hui de bien meilleure qualité et le contenu ainsi que les écrans stéréoscopiques sont accessiblesà un large public. De plus, la stéréoscopie est un élément essentiel dans la compréhension d’un modèle3D, notamment dans le cadre de l’enseignement ou de la visualisation de modèles médicaux. Elle permetégalement d’appuyer le message véhiculé par une image. Utilisée à des fins artistiques, la stéréoscopiejoue un rôle émotionnel important.Cependant, la quantité de profondeur perçue dans une image stéréo est limitée par les technologiesd’affichages ainsi que par le système visuel humain. Il est alors toujours malaisé de créer du contenustéréoscopique assurant le confort du spectateur et utilisant de manière appropriée cette dimensionadditionnelle. Stereoscopy has developed since 1838 through photography, movies, virtual reality andscientific visualization. Stereoscopic displays have increased in quality since the first stereoscope andstereoscopic content is now widely available. Furthermore, the understanding of 3D models is greatlyeased by stereoscopy, in particular for educational purposes or medical data visualization. The messageconveyed by an image is also further held by stereoscopy. Used in an artistic way, stereoscopy can bean important emotional vector.However, the depth interval of a stereoscopic image is limited by the displays technologies as well asthe human visual system. It is thus still difficult to create stereoscopic content ensuring a comfortableexperience to the viewer while using stereoscopy appropriately. To overcome this limitation, severalmethods have been developed, in particular to ease stereo image acquisition, but a great amount ofmanual work still remains. Moreover, stereoscopic content manipulation requires specific tools, whichare still to define. Finally, many stereoscopic display methods are available, with some more appropriateto specific viewing conditions than others.This thesis was conducted in this context, with the following contributions : the definition of a stereoscopicuser interaction model designed for a desktop environment, a new depth editing method ensuringthe viewer’s comfort and a new way to create single-image stereo using chromo-stereoscopy

La mia tesi riguarderà il tema della tradizione di Robin Hood, in particolare la sua scomparsa. Sarà divisa in quattro capitoli. Il primo sarà l’introduzione nella quale descriverò le connessioni tra i diversi testi appartenenti alla tradizione. Nel secondo capitolo analizzerò i diversi testi scritti sulla morte di Robin sia per quanto riguarda la trama che i temi. Inoltre, verrà fornita un’edizione diplomatico-interpretativa di ogni testo nel terzo capitolo. L’ultimo capitolo sarà dedicato al confronto tra i testi utile per il raggiungimento dei miei obiettivi: trovare il testo più antico tra quelli studiati e le ragioni per le quali la storia della morte di Robin è cambiata così tanto nel tempo a partire dal Medioevo fino ai giorni nostri. Infatti, esistono nove diverse versioni sulla morte dell’eroe e l’unica caratteristica in comune ha a che fare con la morte di Robin dovuta all’inganno, probabilmente perché deve essere stata considerata come l’unica morte degna possibile. Il quarto capitolo cercherà di comprendere se i testi più recenti derivano dal testo più antico oppure derivano da diversi testi manoscritti ormai perduti. Infine, mostrerò le somiglianze e le differenze tra le versioni, per quanto riguarda la lingua, così come la trama e la descrizione di personaggi e luoghi.

The thesis presents an analysis of the death notice as a genre, which has been conducted by applying the research models of genre analysis designed by John Swales and Vijay K. Bhatia, and taxonomy of Polish death notices by Jacek Kolbuszewski. This in-depth structural analysis is based on a large corpus of texts (1843 texts consisting of 210,021 words), containing all death notices published in the online edition of The New York Times in a threemonth period (October 1st, 2012 – December 31st, 2012), and downloaded from Legacy.com (the leading global provider of online obituaries and death notices). The analysis involves identifying subgenres of the death notice and their communicative purposes, applying the Move and Steps analytical model to investigate the macrostructure of each subgenre of the death notice and its variants, and carrying out a register analysis, based on lexical and syntactic study with the aim of discovering patterns and lexemes characteristic of each move and/or step. Contrary to the well-researched staff-edited obituary, the genre of American death notice, written by non-professional authors (e.g. relatives, friends, employers or colleagues of the deceased) has not been thoroughly investigated; therefore, it is believed that the thesis will not only make a valuable contribution to the understanding of the genre in question, but it can be used as a reference manual helping prospective writers create a death notice in accordance with the American traditions and rules of the genre. The thesis consists of a theoretical part (Chapters One to Four) and a research part (Chapters Five to Eight). Chapter One revolves around the concepts of discourse, text and genre, and presents an overview of their theories. Chapter Two investigates the American discourse of death; it concentrates on the issue of death as a language taboo and various ways of coping with it, and provides a historical overview of numerous genres commemorating the dead. Chapter Three focuses on the both genres in question; it outlines their origin and evolution in the early British press, and summarizes contemporary research into them. Chapter Four introduces the research part as it discusses the corpus and principles of its division into subcorpora, the research model and applied methodology, and presents the discourse community and communicative purposes. Each of the four chapters constituting the research part deals with the Move and Step analysis of one of four subgenres of the death notice: informative (Chapter Five), farewell (Chapter Six), condolence (Chapter Seven), and anniversary (Chapter Eight); their lexico-structural analysis is illustrated with numerous excerpts from the respective sub-corpora. The Conclusion summarizes the research, and provides implications for future projects. The research has shown that the death notice is a highly conventionalized genre, deeply rooted in American culture and funeral tradition. While presenting biographies of the deceased (always in a positive way, according to the classical rule de mortuis nihil nisi bene), the American death notice emphasizes those specific periods and aspects of their lives (education, professional, political or military career, private life), accomplishments and traits that are valued and respected, and should be imitated by other members of the community. A notice usually contains a lengthy hierarchical list of relatives, both the predeceased and survivors. Each subgenre can be characterized by a specific set of communicative purposes, which are accomplished by a sequence of moves and steps. The commonest subgenre, the informative notice, continues the oldest traditions of the genre by informing the community about a person’s death (optionally its circumstances) and the date and place of the funeral and other services. The style and content of the farewell notice and the condolence notice depend on authorship: highly conventionalized formal institutional notices contrast with more original and intimate private ones. Their authors, whether representatives of an institution or relatives, friends, colleagues, etc., express their loss and grief, praise lives and deeds of the deceased, emphasize their importance for the authors or institution, and, in the case of the condolence notice, they offer their sympathy. The anniversary notice, the rarest subgenre, commemorates the anniversary of decedent’s birth or death, and frequently reminds the community about never-ending love and remembrance of its authors. A significant number of farewell and anniversary notices are addressed to the deceased themselves, the ‘virtual readers,’ which affects their structure and style. The register analysis displays a high level of intertextuality: non-professional obituarists tend to use conventional and stereotypical lexicon, phrases and structures, or even templates (they may copy or imitate other texts and study models provided in obituary manuals). There is no substantial evidence that the Internet has affected the genre: only few texts include hyperlinks that direct to the memorial sites at Legacy.com, where particular groups of the dead are commemorated (e.g. war veterans, university graduates, breast cancer victims).

This dissertation contains a critical edition of the early Irish tale Aided Derbforgaill “the violent death of Derbforgaill”. It includes an introduction discussing the main thematic components of the tale as well as intertextuality, transmission and manuscript relationship. The edition is accompanied by transcripts from the three manuscript copies of the tale and textual notes. Aided Derbforgaill is an Ulster Cycle tale and belongs to a category of tales describing the death of prominent heroes, rarely heroines, in early Irish literature. Arriving in the shape of a bird to mate with the greatest of all heroes, Cú Chulainn, Derbforgaill is refused by Cú Chulainn on account of him having sucked her blood. Forced to enter a urination competition between women, and upon winning this, Derbforgaill is mutilated by the other competitors. The tale ends with two poems lamenting the death of Derbforgaill. This very short tale is complex, not only in its subject matter, but in the elliptical language of the poetry. Thematically the tale is a combination of very common motifs found elsewhere in early Irish literature, such as the Otherworld, metamorphosis and the love of someone unseen, and some rare motifs that are almost unique to this tale, such as blood sucking and the urination competition. The text also have clear sexual overtones.

Bien qu’arabophones, c’est en français que les enfants lisent principalement, langue qui est également, pour la majorité d’entre eux, celle de l’enseignement. Les éditeurs libanais francophones ne proposant qu’une production extrêmement réduite, les enfants vont devoir se tourner vers des romans français dont ils ne sont pas les destinataires premiers. Pour eux, se pose alors la question de la réception de ces romans où il leur sera difficile de se retrouver sachant que ces livres tournés vers la contemporanéité, donnent à voir la réalité sociale d’un monde qui n’est pas le leur.Aménagée en trois parties, la thèse apporte d’abord un éclairage sur certains particularismes culturels, linguistiques et éducatifs constitutifs de l’identité libanaise. Pour mieux comprendre le rapport des enfants avec la lecture, les productions des éditeurs francophones de jeunesse sont présentées ainsi que les structures publiques, privées ou associatives qui permettront la rencontre avec les livres. La seconde partie se concentre sur la réception par les enfants libanais du paratexte et de l’incipit d’un corpus de romans français destinés aux 8/11 ans. Cette analyse explicite les stratégies de séduction privilégiées par les éditeurs et les auteurs français. La troisième partie présente la production éditoriale des fictions francophones destinées au marché scolaire ainsi que les particularités de leur forme d’écriture. Le traitement fictionnel de certaines représentations sociales dans des titres français et libanais francophones est ensuite abordé comme les rôles parentaux, l’éducation, la mort et la guerre, dans les romans ayant le Liban comme thématique
Despite speaking Arabic as their mother tongue, it is French that children mostly read, obviously because French is - for the majority of them - the medium of instruction. However, Lebanese publishers of French-speaking books offer a very narrow selection, hence the children’s turning to French novels whom they are not the first recipients. But how do these children receive such novels? It would be difficult for them to identify with these modernity-oriented-books that portray the social reality of a world that is not theirs. This threefold-thesis first provides insight into a number of specific cultural, linguistic and educational constituents of the Lebanese identity. And to better understand the relationship between children and reading, the first part presents the productions of French-speaking publishers of youth novels, along with public, private and other community facilities that organize “book encounters”.The second part studies the Lebanese children’s reception of the paratext and the incipit of a corpus of French novels written for the 8/11 year olds. This analysis explains the strategies of appeal favored by French publishers and authors.The third part describes the editorial production and specific writing style of Lebanese French-speaking fiction intended for the school market. Over the last decade, the special effects movie is the most popular in the film industry. These special effects need to use the image combination to combine the avatars and the background. In order to get a perfect combination image, it has two key point: the illumination and the pixel intensity of the object and the object’s size. According to the different location, the object size also need to adjust. So that, it need the depth of the picture to measure the size. In our research, first, we use the stereo vision and the feature points to calculate the depth of the background. Second, to adjust the illumination and the pixel intensity of the object. Third, use the bilinear interpolation and the depth information to adjust the size. Finally, using the Possion algorithm to make the object melt in background naturally. In our research, we can use the depth and background information to adjust the object, and use the Possion editor to construct a complete video editing.

Recently, stereoscopic 3D image editing has attracted a great deal of research interest in the computer graphics community. Different from conventional 2D image editing, it usually requires adjustment of the image content as well as the perceived depth. In this dissertation, we present a framework for structural stereoscopic 3D image editing. The framework utilizes the additional information in stereoscopic 3D image, depth structure, to manipulate the image content with respect to the user-specified editing goals. We first adapt the framework to achieve two editing operations: stereoscopic 3D image cloning and stereoscopic 3D image synthesis. The proposed stereoscopic image cloning technique performs both shape adjustment and color blending so that the stereoscopic composite is seamless in both the perceived depth and color appearance. Additionally, we introduce a versatile and robust stereoscopic image synthesis technique for synthesizing stereoscopic images with respect to editing constraints used in different editing applications (eg, image retargeting, image inpainting, texture synthesis). We demonstrate several challenging cases to show the success of our proposed techniques.

As a whole, no pervasive quality unites what we now deem Harlem Renaissance literature outside of the era in which it developed. The inconsistency and varied nature of the works therefore place an even greater emphasis on the editorial processes that produced this canon. While the popular period has received much scholarly attention, the significance of editors and editing in the Harlem Renaissance—aside from the formative efforts of W.E.B. Du Bois and Alain Locke—remains understudied. As a remedy, Editing the Harlem Renaissance foregrounds an in-depth approach to relevant editing and editorial issues, offering a variety of voices to become the first centralized authority on the subject. Rather than limiting the examination to a narrow understanding of editorial practices, this collection takes a broad and inclusive approach, exploring not only those figures of the Harlem Renaissance who edited in professional capacities but also those authors who employed editorial practices during the writing process, as well as those texts that have been discovered and/or edited by others in the decades following the Harlem Renaissance. To achieve this end, the collection comprises chapters in several areas, including professional editing, authorial editing, textual self-fashioning, textual editing, documentary editing, and bibliography.

Irving Thalberg was not just a critically important producer during Hollywood’s Golden age, but also an innovative theorist of studio-era filmmaking. Drawing on archival sources, this is the first book to explore Thalberg’s insights into casting, editing, story composition and the importance of the mass audience from a theoretical perspective. The book argues that Thalberg’s views represent a unified conceptual understanding of production – one that is still significant in the modern day. It examines Thalberg’s impact on film-historical turning points, including the transition from silent to sound cinema and the development of the Production Code, and features in-depth analyses of his productions at Metro-Goldwyn-Mayer from 1924 to 1936. Indeed, each chapter offers a reading of Thalberg’s films through his own theoretical lens, thus highlighting his insights into production and introducing new ways of considering his classic pictures, including The Big Parade (1925), The Broadway Melody (1929) and Romeo and Juliet (1936). The work concludes by assessing his resonance in popular culture, tracing the mythology of Thalberg as it evolved after his death in 1936.

AbstractCRISPR technology is revolutionizing genomic engineering by enabling scientists to precisely modify plant DNA, thus representing a powerful tool for plant breeding.This chapter provides a summary of the approaches and constraints of CRISPR-mediated genome editing in plants, with a focus on the critical prerequisite of efficient CRISPR reagents for successful gene editing in plants.While computational tools have tremendously improved our ability to design specific guides, their limitations make guide effectiveness prediction unreliable, especially for plants. Therefore, it is strongly recommended to validate CRISPR reagents before investing time and resources in the challenging process of plant transformation.A number of in vitro and in planta assays coupled with analytical methods have been proposed to assess the editing performances. Each approach has its own strengths and weaknesses, so the choice of the most suitable system depends on the specific plant species and the type and depth of the genotypic data required.In many cases, the hairy root assay can provide a good compromise between rapidity, reliability and cost-effectiveness for assessing editing performance in numerous plant species.

She explains that she alternated the two author’s words to describe the daily events of their joint fieldwork, using both voices a in such a way as to create a coherent narrative flow, sometimes using both authors’ accounts of the same event to create perspective and depth. She emphasizes how the two authors’ keeping of detailed, personalized field notes enabled her to create the experience for the reader of entering into the story and participating in the adventure.

Such cross-fertilization is considered as inviting the depth of this filmmaker’s discourse and the practical aesthetics through which this is articulated. It also points to the filmmaker’s self-awareness: of the tropes of the horror movie and of cinematic language, tone, camera, and editing. This thesis is based on the promotion of collaborative editing of large comprehensive reports, in-depth study of related technologies including the timeliness of collaborative editing, unified document format standards, unified and effective process control, unified document storage and version management from tools, processes and platforms, and proposes a construction plan to solve these existing problems, and finally provides a collaborative editing platform for each major. Construction and application of collaborative editing system for the nuclear power large comprehensive report realizes workflow and standardization, greatly improves editing efficiency and quality of large comprehensive reports, reduces manpower input, human errors, saves management costs of various professions and lately helps HPR1000 successfully pass qualification verification.

Abstract This paper describes a circuit editing procedure in which the authors used a gallium column Focused Ion Beam (FIB) tool to divide a merged 32nm multi-finger planar transistor into two separate operating components. Rather than rely on live imaging or the various endpoint detection techniques commonly used during an active mill, the authors opted for a ‘blind’ dose-driven technique. The paper explains how the authors made multiple attempts on practice material in order to determine the exact beam placement location and the depth of cut required to perform the operation with a minimum of lateral damage. The loss of a pair of poly gate fingers in the middle of the multi-gate structure seemed to have minimal impact on the final electrical parameters and the separate data paths worked per design specifications.

Modern biomedical technologies pose bioethical dilemmas for humanity. On the one hand, medical advances can make life much easier for people, but, on the other hand, the problem of interference in human nature actualizes the most fundamental questions regarding his ontology, the boundaries of permissible transformations, the responsibility of a scientist and a specialist who applies the latest technologies, for remote and unpredictable consequences, due to the integrity and interconnectedness of various aspects of human nature. In the scientific literature, there is a lot of information about the attitude of various denominations to genetic manipulation. This paper presents the experience of generalizing and systematizing the attitude of the main Christian confessions to the problem of editing the human embryo genome. The assessment of modern biomedical technologies from the standpoint of the Christian worldview differs, on the one hand, in the moral depth due to spiritual experience in relation to the higher divine principle, and, on the other hand, if we bear in mind the specificity of the Orthodox, Catholic and Protestant views on the problem of genetic manipulations, it is diversity interpretations in connection with historically arisen and existing to this day confessional and doctrinal differences.

Currently, the qualitative spectrum of methods in the philological sciences is being substantially expanded, with far-reaching implications, through the integration of the empirical, quantitative, and evaluative possibilities of the Digital Humanities. The example of the planning and establishment of „Kallimachos,“ the Center for Philology and Digitality (ZPD) at the University of Würzburg, demonstrates how a research center in the field of interplay between the humanities and cultural studies, digital humanities, and computer science can bring about a surge of change by providing in-depth insights into each other‘s subjects and ways of thinking. It not only brings with it a new view of the epistemological interests of philology, its questions, its canon, and its key concepts, but also makes computer science aware of the ‚recalcitrance‘ of humanities subjects and thus confronts it with new tasks. The ZPD is the result of a systematic reflection on the digital transformation of philology, with its traditional focus on editing and analyzing, in order to advance this development both in terms of content and methodology. For example, the formation of linguistic conventions in speaking and writing about music in 19th-century composers‘ texts and in music journals would be an ideal subject for the application of digital methods of analysis and the development of new research questions based on them. Research networks that jointly develop and rethink methods on the level of data structures across disciplines are likely to be a proven means of preserving our own discipline in the future, even if this may occasionally be a relationship borne more by reason than by love.

An ambitious reduction in greenhouse gas emissions is set by the political initiative �The European Green Deal� and �Fit for 55� package. The municipalities, which carry out the strategic management of the district heating systems, will have to ensure the sustainable development of the territory in a way that includes a significant increase in energy efficiency both in the building stock and in the related district heating system. The aim of this research is to study and develop an easy-to-use methodology and an appropriate web-based application for sustainable management of the urban heating system, which would allow a quick and at the same time accurate and objective analysis of the potential and benefits of the different building renovation depths and corresponding heating system renovation variants in a given area during the pre-design stage, as well as compare them according to their efficiency. A tree data structure is chosen as the most suitable algorithm, allowing creation of hierarchies of the necessary depth and complexity. Urban (root) level combines many renovation projects to create a dynamic functional model of the specific heating system and for initial modelling the overall impact of various renovation variants � heat consumption and primary energy savings, reduction of CO2 emissions, changes in costs and investments to be performed. At the 2nd - project level � the development of concrete projects and their options takes place. Each project option is a possible heating solution; the attributes of the option define benefits of this project. At the 3rd � component level � heat balances of components (buildings and heat system (DH or local)) are created on the basis of key data of their main elements. The last, 4th level ensures entering and editing thermal, constructive, and financial data on the main elements of heating system components. Developed management methodology and tool based on the data from real buildings and heating systems shows achievable energy consumption and primary energy savings, reduction of CO2 emissions, takings into account heating energy tariff and renovations investments. Achieving the goal means the serious scientific novelty in the management science and evaluation of the climate policy implementation, as well as the new technological know-how and innovation in data processing.

The existing analogues analysis is carried out, due to which a list of advantages and disadvantages is formed and taken into account in developing the virtual laboratory. Root-knot nematodes, Meloidogynespp., are extremely destructive pathogens with a cosmopolitan distribution and a host range that affects most crops. Safety and environmental concerns related to the toxicity of nematicides along with a lack of natural resistance sources threaten most crops in Israel and the U.S. This emphasizes the need to identify genes and signal mechanisms that could provide novel nematode control tactics and resistance breeding targets. The sedentary root-knot nematode (RKN) Meloidogynespp. secrete effectors in a spatial and temporal manner to interfere with and mimic multiple physiological and morphological mechanisms, leading to modifications and reprogramming of the host cells' functions, resulted in construction and maintenance of nematodes' feeding sites. For successful parasitism, many effectors act as immunomodulators, aimed to manipulate and suppress immune defense signaling triggered upon nematode invasion. Plant development and defense rely mainly on hormone regulation. Herein, a metabolomic profiling of oxylipins and hormones composition of tomato roots were performed using LC-MS/MS, indicating a fluctuation in oxylipins profile in a compatible interaction. Moreover, further attention was given to uncover the implication of WRKYs transcription factors in regulating nematode development. In addition, in order to identify genes that might interact with the lipidomic defense pathway induced by oxylipins, a RNAseq was performed by exposing M. javanicasecond-stage juveniles to tomato protoplast, 9-HOT and 13-KOD oxylipins. This transcriptome generated a total of 4682 differentially expressed genes (DEGs). Being interested in effectors, we seek for DEGs carrying a predicted secretion signal peptide. Among the DEGs including signal peptide, several had homology with known effectors in other nematode species, other unknown potentially secreted proteins may have a role as root-knot nematodes' effectors which might interact with lipid signaling. The molecular interaction of LOX proteins with the Cyst nematode effectors illustrate the nematode strategy in manipulating plant lipid signals. The function of several other effectors in manipulating plant defense signals, as well as lipids signals, weakening cell walls, attenuating feeding site function and development are still being studied in depth for several novel effectors. As direct outcome of this project, the accumulating findings will be utilized to improve our understanding of the mechanisms governing critical life-cycle phases of the parasitic M. incognita RKN, thereby facilitating design of effective controls based on perturbation of nematode behavior—without producing harmful side effects. The knowledge from this study will promote genome editing strategies aimed at developing nematode resistance in tomato and other nematode-susceptible crop species in Israel and the United States.

The coronavirus pandemic has swept across a world unprepared to fight it, because countries had failed to choose policies to fight inequality. Only one in six countries assessed for the CRI Index 2020 were spending enough on health, only a third of the global workforce had adequate social protection, and in more than 100 countries at least one in three workers had no labour protection such as sick pay. As a result, many have faced death and destitution, and inequality is increasing dramatically. This third edition of the CRI Index report recommends that all governments adopt strong anti-inequality policies on public services, tax and labour rights, to radically reduce the gap between rich and poor. The international community must support them with Special Drawing Rights, debt relief and global solidarity taxes. See also the CRI Index website at www.inequalityindex.org

Countries around the world have endured over a year of extreme uncertainty in the context of the COVID-19 crisis, and economies in the Caribbean have suffered more than most. But with the increasing availability of vaccines and prospects for a resumption of international travel, light is emerging at the end of the Pandemic tunnel. With this in mind, The Inter-American Development Bank Caribbean1 Departments most recent Quarterly Bulletin reviews the latest available information regarding the crisis impacts on citizens, their economies, and key factors that will determine the speed and depth of recovery. As also discussed in previous editions, prospects for tourism-dependent economies will depend heavily on vaccine penetration and border normalization in source countries particularly the United States and Western Europe, while commodity-intensive economies could benefit from upward revisions to global demand growth estimates. All countries in the region can do much to support a rapid recovery through forward-looking policies aimed at ensuring they are well positioned to take advantage of post-Pandemic preferences with respect to travel and tourism, services trade, and investment. Our latest report considers these issues, what may lie ahead, and how counties can best position themselves for a recovery in 2021 and beyond.

Pepper exhibits large natural variation in chlorophyll content in the immature fruit. To dissect the genetic and molecular basis of this variation, we conducted QTL mapping for chlorophyll content in a cross between light and dark green-fruited parents, PI 152225 and 1154. Two major QTLs, pc1 and pc10, that control chlorophyll content by modulation of chloroplast compartment size in a fruit-specific manner were detected in chromosomes 1 and 10, respectively. The pepper homolog of GOLDEN2- LIKE transcription factor (CaGLK2) was found as underlying pc10, similar to its effect on tomato fruit chloroplast development. A candidate gene for pc1was found as controlling chlorophyll content in pepper by the modulation of chloroplast size and number. Fine mapping of pc1 aided by bulked DNA and RNA-seq analyses enabled the identification of a zinc finger transcription factor LOL1 (LSD-One-Like 1) as a candidate gene underlying pc1. LOL1 is a positive regulator of oxidative stress- induced cell death in Arabidopsis. However, over expression of the rice ortholog resulted in an increase of chlorophyll content. Interestingly, CaAPRR2 that is linked to the QTL and was found to affect immature pepper fruit color in a previous study, did not have a significant effect on chlorophyll content in the present study. Verification of the candidate's function was done by generating CRISPR/Cas9 knockout mutants of the orthologues tomato gene, while its knockout experiment in pepper by genome editing is under progress. Phenotypic similarity as a consequence of disrupting the transcription factor in both pepper and tomato indicated its functional conservation in controlling chlorophyll content in the Solanaceae. A limited sequence diversity study indicated that null mutations in CaLOL1 and its putative interactorCaMIP1 are present in C. chinensebut not in C. annuum. Combinations of mutations in CaLOL1, CaMIP1, CaGLK2 and CaAPRR2 are required for the creation of the extreme variation in chlorophyll content in Capsicum.

Despite marked progress in improving nutrition and health globally, micronutrient deficiencies remain prevalent in low and middle income countries (LMIC), especially among pregnant women with far-reaching consequences for mother and baby including preterm birth, stillbirth or neonatal death, impaired fetal growth. Multiple micronutrient supplements (MMS) taken during pregnancy provide 15 vitamins and minerals to fill the dietary gaps often present in LMIC and the increased nutrient needs of pregnancy. Crucially, MMS provide benefits far beyond those of traditional iron and folic acid supplements (IFA) – prompting the World Health Organization to add the UNIMMAP (United Nations International Multiple Micronutrient Antenatal Preparation) MMS formulation to its Essential Medicines List (EML) in 2021. The Sight and Life Special Report, Focusing on Multiple Micronutrient Supplements in Pregnancy: Second Edition puts forth a compelling case that maintaining the status quo of IFA supplementation is no longer possible when so many stand to benefit from MMS, and demonstrates the benefits and process of switching to MMS. The report compiles the latest evidence, country case studies, and resources in a single place to support implementers and country governments in introducing and scaling-up MMS.
