Com esta expressão latina, que significa “eu tento/eu ouso”, o Conde Everardo fundou no fim da Idade Média, em 11 de março de 1477, uma Universidade que leva o nome do Duque Carlos de Württemberg – a Universidade Everardo Carlos de Tubinga (Eberhard-Karls-Universität Tübingen), no Estado de Baden-Württemberg, na Alemanha. Sendo considerada uma das Universidades mais antigas da Europa, cujas primeiras atividades acadêmicas incluíram os cursos de Medicina, Teologia, Filosofia e Direito, a Universidade de Tubinga goza hoje de alta reputação acadêmica, despontando entre as 10 Universidades de excelência do país e com um total de 11 prêmios Nobel a ela associados. Aquilo que há 542 anos foi um ousado plano para iluminar o fim das trevas da Idade Média, estabeleceu-se como um projeto de grande brilho, cujos raios fulguram os dias de ciência do século XXI.[...]

Biodiversity is declining, and knowledge about biodiversity declines in a similar way. Previous studies have already addressed predictors of species knowledge. Here, we studied bird species knowledge related to demographics, but also to individual differences in affinity to nature, by including (i) birding specialization and (ii) bird-related activities/interest. Data were collected from July to October 2020 via an online questionnaire, containing demographic data, birding specialization, interest/activity, and images of 28 bird species native to Germany. Participants were adult students, lecturers and administrative staff of the Eberhard Karls University Tübingen. A total of 1967 questionnaires were returned in this study (35.3% male, 53.8% students, 69% had access to a garden). Mean identification score was 16.31 ± 6.38. Thus, participants were able to identify more than half of the species (total species n = 28). Men identified more species than women, garden owners had higher identification scores than non-owners, while hometown size was not significant. A distance to the next forest patch >10 km was related to lower identification scores. Employees scored higher than students. Correlation between species knowledge and birding specialization was high, as was the correlation with bird interest/activity. Higher scores were found in older people. In the linear univariate model, birding specialization and bird interest were the most influential predictors of species knowledge, followed by distance to next forest patch and occupation (student vs employees). Other variables were not significant. We suggest including such measures (interest, attitude, etc.) into further studies and move forward from the urban–rural narrative to more complex analyses of living circumstances.

Abstract. This paper presents the unmanned research aircraft Carolo P360 "ALADINA" (Application of Light-weight Aircraft for Detecting IN-situ Aerosol) for investigating the horizontal and vertical distribution of ultrafine particles in the atmospheric boundary layer (ABL). It has a wingspan of 3.6 m, a maximum take-off weight of 25 kg and is equipped with aerosol instrumentation and meteorological sensors. A first application of the system, together with the unmanned research aircraft MASC (Multi-Purpose Airborne Carrier) of the Eberhard-Karls University of Tübingen (EKUT), is described. As small payload for ALADINA, two condensation particle counters (CPC) and one optical particle counter (OPC) were miniaturized by re-arranging the vital parts and composing them in a space saving way in the front compartment of the airframe. The CPCs are improved concerning the lower detection threshold and the response time. Each system was characterized in the laboratory and calibrated with test aerosols. The CPCs are operated with two different lower detection threshold diameters of 6 and 18 nm. The amount of ultrafine particles, which is an indicator for new particle formation, is derived from the difference in number concentrations of the two CPCs. Turbulence and thermodynamic structure of the boundary layer are described by measurements of fast meteorological sensors that are mounted at the aircraft nose. A first demonstration of ALADINA and a feasibility study were conducted in Melpitz near Leipzig, Germany, at the Global Atmosphere Watch (GAW) station of the Leibniz Institute for Tropospheric Research (TROPOS) on two days in October 2013. There, various ground-based instruments are installed for long-term atmospheric monitoring. The ground-based infrastructure provides valuable additional background information to embed the flights in the continuous atmospheric context and is used for validation of the airborne results. The development of the boundary layer, derived from backscatter signals of a portable Raman lidar POLLYXT, allows a quick overview of the current vertical structure of atmospheric particles. Ground-based aerosol number concentrations are consistent with the results from flights in heights of a few meters. In addition, a direct comparison of ALADINA aerosol data and ground-based aerosol data, sampling the air at the same location, shows comparable values. MASC was operated simultaneously with complementary flight patterns. It is equipped with the same meteorological instruments that offer the possibility to determine turbulent fluxes. Therefore additional information about meteorological conditions was collected in the lowest part of the atmosphere. Vertical profiles up to 1000 m altitude indicate a high variability with distinct layers of aerosol especially for the small particles of a few nanometers in diameter. Particle bursts were observed on one day during the boundary layer development in the morning.

Therefore, additional information about meteorological conditions was collected in the lowest part of the atmosphere. Vertical profiles up to 1000 m in altitude indicate a high variability with distinct layers of aerosol, especially for the small particles of a few nanometres in diameter on 1 particular day. The stratification was almost neutral and two significant aerosol layers were detected with total aerosol number concentrations up to 17 000 ± 3400 cm−3 between 180 and 220 m altitude and 14 000 ± 2800 cm−3 between 550 and 650 m. Apart from those layers, the aerosol distribution was well mixed and reached the total number concentration of less than 8000 ± 1600 cm−3. During another day, the distribution of the small particles in the lowermost ABL was related to the stratification, with continuously decreasing number concentrations from 16 000 ± 3200 cm−3 to a minimum of 4000 ± 800 cm−3 at the top of the inversion at 320 m. Above this, the total number concentration was rather constant. In the region of 500 to 600 m altitude, a significant difference of both CPCs was observed. This event occurred during the boundary layer development in the morning and represents a particle burst within the ABL.

Basement membrane loss is associated with reduced islet function and viability. Previous studies demonstrated that individual extracellular matrix proteins (ECMPs) can increase islet survival pre- and post-transplant. In the present study, we tested our hypothesis, that the combination of different ECMPs, particularly those forming suprastructures, are more efficient than individual ECMPs to protect human islets from hypoxia-induced damage. In contrast to previous studies, we dissolved ECMPs in the media rather than to coat culture surfaces. Methods Islets, isolated from pancreases of 11 human DBD donors (50±2 years, 29.3±1.2 BMI, 5.7±0.3 hours CIT), were cultured for 3–4 days in 2% oxygen and suspended in CMRL 1066 (2% FCS) supplemented with either 40 µg/mL of dissolved collagen-IV, 10 µg/mL laminin-521 or 12.5 µg/mL nidogen-1 used individually or as combination. Sham-treated islets (STIs) cultured without ECMPs served as controls. Post-culture characterisation included IEQ yield or islet number (IN), viability (FDA-PI), early plus late apoptosis (annexin V-PI), glucose stimulation index (SI: 2 vs 20 mM) and reactive oxygen species production. Parameters were normalised to IEQ, related to pre-culture data if appropriate and presented as mean ± SEM. Statistical analysis was performed by Friedman test followed by Dunn’s multiple comparison. Results Compared with STI (41±7%), post-culture recovery was higher when hypoxic islets were treated with collagen-IV (64±7%, p<0.001), laminin-521 (57±6%, p<0.01) or nidogen-1 (65±6%, p<0.001) used individually or combined (61±7% p<0.001). This correlated with islet fragmentation (IN/IEQ ratio) that was lower when collagen-IV (116±13%, p<0.001), laminin-521 (114±12%, p<0.01), nidogen-1 (121±12%, p<0.01) or combined ECMPs (119±13%, p<0.001) were compared with STIs (155±16%). Reactive oxygen species production in STIs was substantially reduced by 71±6% (NS), 73±6% (p<0.05), 90±2% (p<0.001), and 87±4% (p<0.001) in presence of collagen-IV, laminin-521, nidogen-1 or combined ECMPs, respectively. This resulted in improved viability (83±7% [p<0.01], 79±9% [p<0.01], 84±7% [p<0.001], 83±8% [p<0.001]) compared with STIs (63±7%). While individual ECMPs stabilised or reduced pre-culture apoptosis (94±17% [p<0.05], 117±16% [p<0.05], 68±13% [p<0.001]), combined ECMPs (171±18%, NS) were equal to STIs (196±28%). STIs did not adequately secrete insulin after glucose challenge (SI 0.97±0.13) in contrast to the physiological insulin response after treatment with collagen-IV (1.76±1.18 [p<0.01]), laminin-521 (1.53±0.25 [NS]), nidogen-1 (2.27±0.67 [p<0.01]) or combined ECMPs (1.95±0.25 [p<0.05]). Conclusion Among the three individual ECMPs tested, nidogen-1 appears to be most effective to protect human islets from hypoxia-induced damage. As its protective efficiency partially exceeds that of combined ECMPs, we have to reject our hypothesis. Further studies are required to clarify whether collagen-IV, laminin-521 and nidogen-1 spontaneously assemble to suprastructures in vitro.
