5-Hydroxytryptamine (serotonin, 5-HT) contracts the guinea pig trachea through stimulation of 5-HT2A receptors, an effect previously reported to rapidly desensitize. The present studies were designed to examine further the putative desensitization to serotonin. In vitro studies investigating functional desensitization of the guinea pig tracheal 5-HT2A receptor documented that 5-HT (3 × 10−7 M) significantly (50%) but incompletely reduced subsequent tracheal maximal contraction to 5-HT. We developed a new excised tracheal preparation to measure the epithelial permeability of large lipid-insoluble nonelectrolytes and macromolecules. Tracheae were suspended vertically in a Ringer solution bath, and a solution containing labeled test solutes was positioned in the center of the tracheal segment, away from damaged ends. In this study, capsaicin induced a long-lasting and marked contraction in the guinea pig trachea and nicotine-induced contraction was partially reduced in the capsaicin-treated muscle. Effects of baicalein on release of slow reacting substance of anaphylaxis (SRS-A) or leukotriene (L T) from the sensitized guinea pig lung after antigen challenge and tonus of guinea pig tracheal muscles were studied. Baicalein inhibited release of SRS-A from sensitized guinea pig lung after antigen challenge. To study the intact trachea from guinea pigs we developed an in vitro preparation that permits independent stimulation from either the inner epithelial surface or the outer serosal surface. Its IC50 was 13.98±2.54 (n=7), 14.56±0.92 (n=6) and 21.15 ± 2.92 (n=9) μM for the precontractions induced by histamine (30 μM), carbachol (0.2 μM) and isotonic KC1 (17.5 mM), respectively. There was no significant difference among them. It shows that 3-MQ has no special selectivity to these three contractile agents. Pretreatment of tracheahs with 3-MQ (30 μM) for 20 min non- competitively inhibited the contractions induced by cumulative histamine. Its calculated pD2' value was 5.29 ±0.

AbstractwIRA has been shown to reduce chlamydial infections in vitro and in vivo and might therefore offer an innovative therapeutic approach for fighting trachoma. Abstract Navigation algorithms are presented that can be used to guide autonomous vehicles along a right of way to inspect poles or towers for corrosion, or to inspect enclosed spaces such as power substations. These algorithms use the combined methods of sensor feature extraction, PID control, dynamic window obstacle avoidance (DWA), and genetic algorithm (GA) solutions to the traveling-salesman problem (TSP).
