ABSTRACT Class AII and AIII lantibiotics and mersacidin are antibacterial peptides containing unusual residues obtained by posttranslational modifications of prepeptides, presumably catalyzed by LanM. LctM, the LanM for lacticin 481, is essential for the production of this class AII lantibiotic. Using the yeast two-hybrid system, we showed direct contact between the prelacticin 481 and LctM, supporting the proposed LctM function. Sixteen domains are conserved between the 10 known LanM proteins, whereas three additional domains were found only in class AII LanM proteins and in MrsM, the LanM for mersacidin. All the truncated LctM proteins that we tested presented impaired LctA-binding activity.

Анотація:
Protein merupakan senyawa yang banyak terkandung di dalam limbah cair tahu. Kadar protein yang tinggi dapat menyebabkan kenaikan BOD dan COD limbah cair tahu di lingkungan. Penelitian ini bertujuan untuk mengetahui kemampuan kitosan dalam mengadsorpsi protein limbah cair tahu dari Desa Tinalan, Kota Kediri. Kadar protein awal limbah cair tahu yang teramati menggunakan spektrofotometer UV-Vis yaitu 0,0904%. Pada saat adsorpsi, massa kitosan yang digunakan yaitu 0,25 g; 0,5 g; 0,75 g dan 1 g dengan kode sampel masing-masing kitosan-LCT1; kitosan-LCT2; kitosan-LCT3 dan kitosan-LCT4. Kitosan ditambahkan ke limbah cair tahu dan diaduk selama 8 jam. Hasil penelitian menunjukkan bahwa massa kitosan yang optimum dalam mengadsorpsi protein limbah cair tahu adalah 0,25 g (kitosan-LCT1). Hal ini ditunjukkan dengan penurunan kadar protein limbah cair tahu hingga 37,78%. Hasil SEM menegaskan bahwa telah terjadi perubahan morfologi permukaan pada kitosan-LCT1 yang berbentuk pipih dan guratan putih. Hal ini menunjukkan telah terjadi proses adsorpsi antara kitosan dan protein. Selain itu, adsorpsi juga menyebabkan peningkatan pH limbah cair tahu mencapai 6 – 7.Kata kunci : Protein, Limbah Cair Tahu, Kitosan, Adsorpsi

Abstract The article discusses methods of controlling the processes of mechanical processing based on electrochemical effects. The corresponding anodic polarization curves of 1X18H9T steel obtained in electrolyte solutions without and with stirring are presented. The article discusses methods of machining processes control based on electrochemical effects. Lubricating and cooling technological media (LCTM) used in machining are in most cases electrolytes, therefore, electrochemical processes and phenomena actively occur during contact dynamic machining. It is possible to control the processes of machining by acting on the system elements of the tool - LCTM- part, in particular by activating the LCTM and reducing the strength characteristics of the processed steel in the cutting zone. A reserve for increasing the efficiency of mechanical processing can be the composition selection of the applied LCTM, combined with the simultaneous electrochemical polarization of the treated surface of friction pair parts. It was found that when cutting, the efficiency of machining and the chip shapes are changed, which is explained by the influence of the current density on the strength of the processed steel. In the conditions of machining, complex dynamic processes occur due to the rotation of the work piece and/or tool, so it is necessary to take into account the hydrodynamic phenomena and processes that arise in this case. Electrode potentials are considered to be the most important characteristic of the metal cutting process. The potential of the system can regulate such processes and indicators as wear and surface micro hardness. Anodic polarization curves of the steel 1X18H9T obtained in various electrolyte solutions without stirring and with stirring on a rotating disk electrode are given. The study allowed determining the factors affecting the processes occurring in the cutting zone and to identify rational current densities due to simulating the conditions of real technological processes of the combined steel processing. The increase in the processing intensity of the steel 1X18H9T with the cutting zone polarization is associated with the action of factors activating the selective anodic dissolution of the processed alloy.

The article deals with the experimental evaluation (in the laboratory) of environmentally secure lubricating and cooling technological means’ (LCTM) influence on the force characteristics and tool wear in the turning process of structural materials with different chemical activity. The experiment results confirm that the use of vegetable and animal fats in combination with the minimal quantity lubrication technology (MQL) provides for a significant reduction of cutting forces and the contact area.

ObjectivesLatent class trajectory modelling (LCTM) is a relatively new methodology in epidemiology to describe life-course exposures, which simplifies heterogeneous populations into homogeneous patterns or classes. However, for a given dataset, it is possible to derive scores of different models based on number of classes, model structure and trajectory property. Here, we rationalise a systematic framework to derive a ‘core’ favoured model.MethodsWe developed an eight-step framework: step 1: a scoping model; step 2: refining the number of classes; step 3: refining model structure (from fixed-effects through to a flexible random-effect specification); step 4: model adequacy assessment; step 5: graphical presentations; step 6: use of additional discrimination tools (‘degree of separation’; Elsensohn’s envelope of residual plots); step 7: clinical characterisation and plausibility; and step 8: sensitivity analysis. We illustrated these steps using data from the NIH-AARP cohort of repeated determinations of body mass index (BMI) at baseline (mean age: 62.5 years), and BMI derived by weight recall at ages 18, 35 and 50 years.ResultsFrom 288 993 participants, we derived a five-class model for each gender (men: 177 455; women: 111 538). From seven model structures, the favoured model was a proportional random quadratic structure (model F). Favourable properties were also noted for the unrestricted random quadratic structure (model G). However, class proportions varied considerably by model structure—concordance between models F and G were moderate (Cohen κ: men, 0.57; women, 0.65) but poor with other models. Model adequacy assessments, evaluations using discrimination tools, clinical plausibility and sensitivity analyses supported our model selection.ConclusionWe propose a framework to construct and select a ‘core’ LCTM, which will facilitate generalisability of results in future studies.

In this paper, we conducted numerical simulations to investigate single and two-phase flows around marine propellers in open-water conditions at different Reynolds number regimes. The simulations were carried out using a homogeneous compressible two-phase flow model with RANS and hybrid RANS/LES turbulence modeling approaches. Transition was accounted for in the model-scale simulations by employing an LCTM transition model. In model scale, also an anisotropic RANS model was utilized. We investigated two types of marine propellers: a conventional and a tip-loaded one. We compared the results of the simulations to experimental results in terms of global propeller performance and cavitation observations. The propeller cavitation, near-blade flow phenomena, and propeller wake flow characteristics were investigated in model- and full-scale conditions. A grid and time step sensitivity studies were carried out with respect to the propeller performance and cavitation characteristics. The model-scale propeller performance and the cavitation patterns were captured well with the numerical simulations, with little difference between the utilized turbulence models. The global propeller performance and the cavitation patterns were similar between the model- and full-scale simulations. A tendency of increased cavitation extent was observed as the Reynolds number increases. At the same time, greater dissipation of the cavitating tip vortex was noted in the full-scale conditions.

<b><i>Introduction:</i></b> Body mass index (BMI) is often elevated at type 2 diabetes (T2D) diagnosis. Using latent class trajectory modelling (LCTM) of BMI, we examined whether weight loss after diagnosis influenced cancer incidence and all-cause mortality. <b><i>Methods:</i></b> From 1995 to 2010, we identified 7,708 patients with T2D from the Salford Integrated Record database (UK) and linked to the cancer registry for information on obesity-related cancer (ORC), non-ORC; and all-cause mortality. Repeated BMIs were used to construct sex-specific latent class trajectories. Hazard ratios (HRs) and 95% confidence intervals (CIs) were estimated using Cox regression models. <b><i>Results:</i></b> Four sex-specific BMI classes were identified; stable-overweight, stable-obese, obese-slightly-decreasing, and obese-steeply-decreasing; comprising 41%, 45%, 13%, and 1% of women, and 45%, 37%, 17%, and 1% of men, respectively. In women, the stable-obese class had similar ORC risks as the obese-slightly-decreasing class, whereas the stable-overweight class had lower risks. In men, the obese-slightly-decreasing class had higher risks of ORC (HR = 1.86, 95% CI: 1.05–3.32) than the stable-obese class, while the stable-overweight class had similar risks No associations were observed for non-ORC. Compared to the stable-obese class, women (HR = 1.60, 95% CI: 0.99–2.58) and men (HR = 2.37, 95% CI: 1.66–3.39) in the obese-slightly-decreasing class had elevated mortality. No associations were observed for the stable-overweight classes. <b><i>Conclusion:</i></b> Patients who lost weight after T2D diagnosis had higher risks for ORC (in men) and higher all-cause mortality (both genders) than patients with stable obesity.

Ce travail de thèse vise à évaluer la faisabilité des calculs de transition dans le cadre des codes industriels. Les points forts ainsi que les faiblesses des modèles de transition basés sur des corrélations locales, γ et γ-Re_θ, sont analysés sur des configurations bi et tridimensionnelles, en se concentrant sur la modélisation physique et les aspects numériques. L'objectif est d'analyser le potentiel de ces modèles RANS en tant qu'outils prédictifs, capables de gérer automatiquement et de manière autonome la transition d'un écoulement du régime laminaire au régime turbulent. Nous évaluons les performances des modèles de transition sur un grand nombre de cas de test, couvrant un large éventail de mécanismes de transition. Dans le cas des configurations 3D, un point crucial est la modélisation de la transition due aux modes transversaux et stationnaires, qui sont les principaux mécanismes de transition tridimensionnel dans un environnement à faible turbulence. A cet effet, nous présentons dans cette thèse un recalibrage original du critère transversal Tc1, proposé à l'origine par Menter et Smirnov en 2014 et basé sur une formulation locale du célèbre critère transversal C1 de Daniel Arnal. This research work aims at assessing transition calculations feasibility in the context of the industrial codes. We discuss the strengths and weaknesses of the Local Correlation Based Transition Models (LCTM) γ and γ-Re_θ for two- and three-dimensional configurations, focusing on both modeling and numerical aspects. The purpose is to analyze the potential of these RANS-based models as predictive tools, that can handle automatically and autonomously laminar-to-turbulence transition. We evaluate the transition models performance on an extensive number of test cases, covering a wide range of transition mechanisms. In the case of 3D configurations, a crucial point is the modeling of transition due to stationary crossflow modes, which are the main threedimensional transition mechanism in a low free-stream turbulence environment. For this purpose, we present in this dissertation an original re-calibration of the Tc1 crossflow criterion, proposed by Menter and Smirnov in 2014 and based on a local formulation of the renowned empirical C1 criterion of Daniel Arnal. This re-calibrated criterion notably extends the existing γ model for stationary crossflow transition prediction around threedimensional non-wing-like geometries

Composite flow simulation tools for LCM processing can be expensive and time-consuming but necessary to design a mold system with proper placement of resin inlets and vacuum outlets. Composites manufacturing engineers would benefit from data regarding the impact of mold curvature radius on resin flow. This could help determine whether or not a particular part and mold would require expensive simulation software designed to handle complex flow paths through curved fabric architectures exhibiting variable permeability over the curvature, or if simple flow modeling would provide accurate enough simulations for sound tooling setup decision making. Four molds, with double curvature having equal radii, were fabricated with radii ranging from 3.2 to 25.4 mm to characterize the permeability of two different fiber reinforcements 1) a carbon biaxial NCF and 2) a fiberglass CSM over the mold curvatures. Three infusions of each material type were conducted on each of the 4 molds for a total of 24 test infusions. Flow front position vs. time data was captured during each experimental infusion. The permeability in the bend regions, KB, was first estimated by the integrated form of Darcy's Law to evaluate the permeability for average flow across the entire bend region. This was done for both the convex and concave regions using a geometric estimate for the increased compaction in the bend regions. The permeability increases as the tool radius increases, and the rate of increase diminishes as the tool radius increases and the permeability approaches the flat region permeability. An estimate of KB for VI was then made by applying a ratio calculated from the resulting permeability from the rigid- and VI-based models in the flat regions. Generic power law fits are reported that could be used in LCM process simulation, to give a model to estimate the permeability for any bend in the reinforcement part geometry. The results suggest that any curve with a radius higher than 25 mm requires no adjustment to the flat permeability.

Les composants passifs, représentent actuellement une butée importante en terme de volume occupé, de pertes et de faisabilité de l'intégration. De nouvelles structures électromagnétiques proposent de marier les trois composants habituellement rencontrés dans les convertisseurs sous la forme d'un seul composant baptisé LCT, assurant simultanément trois fonctionnalités : inductance-condensateur-transformateur. Nous avons dimensionné deux structures intégrées de prototypes LCT (bobiné & planar), employant, tous deux, une nouvelle topologie de circuit magnétique. En nous basant sur ces réalisations de composants passifs intégrées ainsi que sur une réalisation discrète, diverses approches sont proposées pour mieux estimer les pertes dans ces composants afin de pouvoir réaliser une étude comparative sur chacun des dispositifs. En particulier, la caractérisation fine du LCT en vue d'obtenir les éléments d'un schéma équivalent constitue un moyen d'atteindre ce résultat.

Nematodes of the genus Meloidogyne are known as “root-knot” nematodes due to the characteristic knots or galls found on the roots of infected plants. Root-knot nematodes attack over 2000 species of plants and cause over 80 billion dollars lost annually. Giant cells are feeding sites of root-knot nematodes and are enlarged multinuclear cells induced by the nematodes in susceptible host roots and that function to provide nutrients to the nematode. This thesis presents data on two techniques of studying the nuclear condition in giant cells. Colchicine was used to arrest mitosis in giant cells in a previous study. Here we test the effect of colchicine on nematode activity. The results showed that colchicine did not affect nematode egg hatch, juvenile activity, or hatch of eggs produced by treated juveniles. These results confirm that colchicine can be used to arrest mitosis in giant cells without affecting the nematode parasite. A major obstacle to the study of giant cells is collecting tissue samples that are specific to giant cells. Laser capture microdissection (LCM) is a technique that allows one to sample a single giant cell. A focused laser beam was used to collect samples of giant cell cytoplasm from fixed and sectioned tissues. RNA was then extracted from those isolated samples. Using three tomato genes as test samples, specific primers were designed to measure expression level of Rb7, LHA4, and HXK1 gene by Real-Time PCR. Expression of LHA4 and Rb7 increased with time after inoculation, and immature giant cells reached levels that were 3 and 6 times, respectively, that of cortical cells, but which were not different from root meristem cells. Expression of HXK1 did not change with time after inoculation and has the same level of that in root tip and cortical tissues. These data confirmed that the techniques of LCM coupled with RT-real-time PCR can be used to quantitate expression of genes at different stages of giant cell development without contamination from surrounding cells.

AbstractThe main goal of this paper is to present the history and actual situation in research on LCA and LCM in Poland. This task will be performed by reviewing the different activities and their results in this field, from the very beginning. The paper includes the review of the activities of LCA/LCM main research centres in Poznań (Poznań University of Technology (PUT), Poznań University of Economics and Business (PUEB)), Cracow (Polish Academy of Sciences, AGH University of Science and Technology, Cracow University of Economics), Zielona Góra (University of Zielona Góra), Bydgoszcz (UTP University of Science and Technology), Katowice-Gliwice (Silesian University of Technology), Częstochowa (Częstochowa University of Technology) and Szczecin (ZUT Western Pomeranian University of Technology). LCA/LCM researches are also performed in several smaller research groups in R&D centres. In the end of the paper, some conclusions referring to the actual situation of research on LCA/LCM, dealing with critical evaluation of the LCA/LCM centres in Poland location, issues and problems addressed, areas of the projects covered and the desired activities in the future, are presented.

The present work investigates the transitional flow around a smooth circular cylinder at Reynolds number Re = 140,000. The flow is resolved using the viscous-flow solver ReFRESCO, and distinct mathematical models are applied to assess their ability to handle transitional flows. The selected mathematical models are the Reynolds-Averaged Navier-Stokes equations (RANS), Scale-Adaptive Simulation (SAS), Delayed Detached-Eddy Simulation (DDES), eXtra Large-Eddy Simulation (XLES) and Partially-Averaged Navier-Stokes (PANS) equations. The RANS equations are supplemented with the k–ω Shear-Stress Transport (SST) with and without the Local Correlation Transition Model (LCTM). The numerical simulations are carried out using structured grids ranging from 9.32 × 104 to 2.24 × 107 cells, and a dimensionless time-step of 1.50 × 10−3. As expected, the outcome demonstrates that transition from laminar to turbulent regime is incorrectly predicted by the k–ω SST model. Transition occurs upstream of the flow separation, which is typical of the supercritical regime and so the flow physics is incorrectly modelled. Naturally, all Scale-Resolving Simulation (SRS) models that rely on RANS to solve the boundary-layer, called hybrid models, will exhibit a similar trend. On the other hand, mathematical models capable to resolve part of the turbulence field in the boundary layer (PANS) lead to a better agreement with the experimental data. Furthermore, the k–ω SST LCTM is also able to improve the modelling accuracy when compared to the k–ω SST. Therefore, it might be a valuable engineering tool if its computational demands are considered (in the RANS context). Therefore, the results confirm that the choice of the most appropriate mathematical model for the simulation of turbulent flows is not straightforward and it may depend on the details of the flow physics.

The widespread use of Computational Fluid Dynamics (CFD) tools for engineering applications is certainly positive. However, users must also be aware of the physics of the problems being modeled, as well as the shortcomings of turbulence models in use. New state-of-the-art turbulence models are currently being developed with the aim of enhancing the turbulent flow predictions but the laminar-turbulent transition is still out of the scope of most the models. Bearing upon those ideas, this paper investigates the performance of the Local Correlation Transition Model (LCTM) for the cylinder flow with Solution Verification and Validation at high Reynolds numbers. Furthermore, attention is paid to characteristics of the setup, numerics and physical conditions and we study how these features alter the results. We also bring recommendations on the use of the transition model regarding grid, setup and physical conditions. The results show much better comparison of numerical and experimental results regarding drag coefficients than seen with the SST turbulence model, even with the two-dimensional calculations done herein.

In the event of sodium leakage from heat transfer circuits of fast breeder reactors (FBR), liquid sodium catches fire in ambient air leading to production of flame, smoke and heat. One of the passive fire protection methods involves immediate collection of the leaking sodium to a sodium hold-up vessel (SHV) covered with a sloping cover tray (SCT) having a few drain pipes and one vent pipe (as in Fig. 1). As soon as the liquid sodium falls on the sloping cover tray, gravity guides the sodium through drain pipes into the bottom tray in which self-extinction occurs due to oxygen starvation. This sodium fire protection equipment called leak collection tray (LCT) works without the intervention of an operator and external power source. A large number of LCTs are strategically arranged under the sodium circulating pipe lines in the FBR plants to serve as passive suppression devices. In order to test the efficacy of the LCT, four tests were conducted. Two tests were with LCT having three drain pipes and rest with one. In each experiment, nearly 40 kg of hot liquid sodium at 550 °C was discharged on the LCT in the open air. Continuous on-line monitoring of temperature at strategic locations (∼ 28 points) were carried out. Colour videography was employed for taking motion pictures of various time-dependent events like sodium dumping, appearance of flame and release of smoke through vent pipes. After self-extinction of sodium fire, the LCT was allowed to cool overnight in an argon atmosphere. Solid samples of sodium debris in the SCT and SHV were collected by manual core drilling machine. The samples were subjected to chemical analysis for determination of unburnt and burnt sodium. The results of the four tests revealed an interesting feature: LCT with three drain pipes showed far lower sodium collection efficiency and much higher sodium combustion than that with just one drain pipe. Thermal fluctuations in temperature sensor located near the tip of the drain pipe have indicated that transient freezing and remelting processes are responsible for this phenomenon. Moreover comparison of test results between present and earlier experiments has revealed that the LCT with funnel shaped SCT is superior to that with boat shaped SCT.

The Tl isotopic and trace element composition of K-feldspar, mica, pollucite and pyrite from 13 niobium-yttrium-fluorine (NYF)-type and 14 lithium-cesium-tantalum (LCT)-type rare-element pegmatites was investigated. In general, the epsilon-205Tl values for K-feldspar in NYF- and LCT-type pegmatites increases with increasing magmatic fractionation. Both NYF and LCT pegmatites display a wide range in epsilon-205Tl (-4.25 to 9.41), which complicates attempts to characterize source reservoirs. We suggest 205Tl-enrichment during pegmatite crystallization occurs as Tl partitions between the residual melt and a coexisting aqueous fluid or flux-rich silicate liquid. Preferential association of 205Tl with Cl in the immiscible aqueous fluid may influence the isotopic character of the growing pegmatite minerals. Subsolidus alteration of K-feldspar by aqueous fluids, as indicated by the redistribution of Cs in K-feldspar, resulted in epsilon-205Tl values below the crustal average (-2.0 epsilon-205Tl). Such low epsilon-205Tl values in K-feldspar is attributed to preferential removal and transport of 205Tl by Cl-bearing fluids during dissolution and reprecipitation. The combination of thallium isotope and trace element data may be used to examine late-stage processes related to rare-element mineralization in some pegmatites. High epsilon-205Tl and Ga in late-stage muscovite appears to be a favorable indicator of rare-element enrichment LCT pegmatites and may be a useful exploration vector.
