Abstract Glioblastoma multiforme (GBM) is the most common primary brain cancer and carries a poor prognosis. GBM cells exhibit extensive metabolic alterations that enhance survival and proliferation in the mixed normoxic-hypoxic tumor microenvironment. Lactate dehydrogenase (LDH) enzymes are critical mediators of the normoxic to hypoxic transition in cells. Two LDH genes (A/B) encode monomers that combine to form five isoenzymes (LDH1-5) with different properties for pyruvate to lactate interconversion. Hypoxic induction of LDHA in all cells appears to occur via HIF-1 mediated transcription. However, little is known about hypoxic regulation of LDHB in cancer. We report on hypoxic regulation of LDHA/B in T98G, a rare cell line that has both normal and neoplastic features. Human T98 GBM cell lines were cultured in a humidified incubator at 37° C and 5% CO2 and were grown in normoxia (21% O2) or hypoxia (95% N2, 5% C02) for 72 hours. Relative expression of LDH isoforms 1-5 was assessed using native gel electrophoresis. Expression of the LDHA and LDHB genes was measured using qRT-PCR. LDHA-dominant isoforms (4/5) were detected in T98G cells subjected to normoxia and hypoxia via gel electrophoresis, however, LDHB-dominant isoforms (1/2) were not. The LDHA/B-equimolar isoform (3) was decreased in T98G cells subjected to hypoxia. LDHA gene expression was over two-fold greater than LDHB in normoxia (p = .00256 by one-tailed Mann-Whitney U test), and over nine-fold greater in hypoxia (p = .00256). LDHA:LDHB expression in hypoxia compared to normoxia was significantly different (p = .00256). LDHA expression increased three-fold in hypoxia (p = .00256), while LDHB expression decreased 0.3-fold in hypoxia (p = .03288). We document LDHB dysregulation in T98G cells as the gene is minimally responsive to oxygen. Therapeutic strategies aimed at promoting LDHB expression may complement inhibition of LDHA and reduce GBM survival in hypoxia.

Background: Megaloblastc anemia corresponds to severe macrocytic anemia with hypersegmented neutrophils and very high serum Lactate Dehydrogenase (LDH). The present study was undertaken to evaluate the utility of serum LDH and chloroform inhibited serum LDH in the diagnosis of megaloblastic anemia and to observe if this can be used to differentiate megaloblastic anemia from iron deficiency anemia and hemolytic anemia.Methods: The present study was carried out on 75 patients of anemia categorised on bone marrow examination (into megaloblastic and non-megaloblastic anaemia) to evaluate the efficacy of total serum LDH levels and LDH isoenzyme pattern in the diagnosis of megaloblastic anemia. About 25 healthy adults were taken as controls.Results: In megaloblastic anemia, total serum LDH level was found to be increased to about nineteen folds and in hemolytic anemia it was found to increased four folds as compared to normal. On statistical analysis this increased total serum LDH level in megaloblastic anemia and hemolytic anemia as compared to control group was found to be significant.In the present study serum LDH level above 3000IU/L was associated with megaloblastic anemia and serum LDH level below 900IU/L was suggestive of iron deficiency anemia. The chloroform inhibition test was less than 25% in megaloblastic anemia and more than 25% in hemolytic anemia and these differences were found to be statistically significant (t=9.62, df=49, p<0.001).Conclusions: Total serum LDH levels more than 3000IU/L are diagnostic of megaloblastic anemia. Reversed LDH isoenzyme pattern (LDH1>LDH2) by chloroform inhibition test is an adjuvant in the diagnosis where total serum LDH levels are between 451-3000IU/L and can also differentiate megaloblastic anemia from hemolytic anemia.

220 Background: There is a lack of biomarkers to predict docetaxel response. Here we assess the potential of pre-treatment LDH, associated to the mutational background of the primary biopsy taken prior to 1st line therapy with ADT, to predict docetaxel response in the castrate resistant setting. Methods: Clinical and associated primary biopsy targeted next-generation sequencing data from matched training (n = 150) and test (n = 120) sets of retrospective progressive mCRPC patients (diagnosed 1996-2006) treated with docetaxel therapy were analyzed. LDH thresholds were determined and validated on the training and test sets respectively. Clinico-pathological associations with LDH were assessed using student t-test, Wilcoxon test and Chi-square test and predictive value of LDH determined by linear regression analysis for PSA and radiologic response. Time to event outcomes (PFS & OS) was evaluated by Kaplan-Meier. Meta-analysis of LDH association with DNA repair genes in the MSKCC and SU2C/PCF was conducted using cBioPortal Results: Using pre-docetaxel serum LDH of ≥450U/L as a prognostic threshold (AUC:0.75 (SD 0.054, 95% CI, 0.650–0.864, P < 0.001)) patients with LDH ≥450U/L were poorer PSA responders, with shorter PFS (213 vs 372 days, HR 1.876 (95% CI, 1.289-2.730)) and OS (362 vs 563 days, HR 1.630 (95% CI, 1.127-2.357)). LDH was also an independent surrogate marker for prediction of poor radiologic response and survival following docetaxel chemotherapy (P = 0.043). Genomic analysis showed that patients with LDH ≥450U/L (9/14 (64.3%)) have a higher tumour mutational burden (TMB) at the point of primary pre-ADT biopsy (mean, 46.2 vs 6.2 mutations/MB; p = 0.129) and that they were more likely to have DNA repair gene mutation(s) (BRCA1/2, ATM, CHEK2 and Fanconi’s anemia gene). Meta-analysis of the large MSKCC and the SU2C-PCF patient genomic databases confirmed the positive correlation between the LDH gene LDHA and PARP1 gene (r = 0.667, p < 0.01) expression and expression of other DNA repair genes. Conclusions: High pre-treatment serum LDH coupled with mutations in the DNA repair pathway in primary pre-ADT biopsy correlates with poor docetaxel response and overall survival in the castrate resistant state.

This study is aimed at analysing biochemical and genetic endpoints of toxic effects after administration of adrenaline. For this purpose, the study was carried out on Wistar rats and three doses of adrenaline were used: 0.75 mg/kg, 1.5 mg/kg, and 3 mg/kg body weight. To achieve these aims, we investigated the effects of adrenaline on catalase (CAT), Cu, Zn-superoxide dismutase (SOD), malondialdehyde (MDA), nitrite (NO2−), carbonyl groups (PCC), and nitrotyrosine (3-NT). Total activity of lactate dehydrogenase (LDH), its relative distribution (LDH1–LDH5) activity, level of acute phase proteins (APPs), and genotoxic effect were also evaluated. The obtained results revealed that all doses of adrenaline induced a significant rise in CAT activity, MDA level, PCC, NO2−, and 3-NT and a significant decrease in SOD activity compared to control. Adrenaline exerted an increase in total activity of LDH, LDH1, and LDH2 isoenzymes. Further study showed that adrenaline significantly decreased serum albumin level and albumin-globulin ratio, while the level of APPs (α1-acid glycoprotein and haptoglobulin) is increased. The micronucleus test revealed a genotoxic effect of adrenaline at higher concentrations (1.5 mg/kg and 3 mg/kg body weight) compared to untreated rats. It can be concluded that adrenaline exerts oxidative and nitrative stress in rats, increased damage to lipids and proteins, and damage of cardiomyocytes and cytogenetic damage. Obtained results may contribute to better understanding of the toxicity of adrenaline with aims to preventing its harmful effects.

Two kinds of layered double hydroxides (LDHs), MgAl-NO2 (N-LDH) and MgAl-CO3 (C-LDH), were incorporated to study the durability of mortar and concrete. The LDH contents of mortar were 1%, 2%, and 4% by mass and the LDH contents of concrete were 0.5%, 1%, 2%, 4%, respectively. The effect of LDHs on sulfate resistance of mortar was studied through dry-wetting cycle test, compressive strength test, and flexural strength test. In addition, the effects of LDHs on pore structure, chloride resistance, carbonation resistance, shrinkage, and creep of concrete were investigated by SEM, mercury injection test, XRD, chloride ion diffusion coefficient test, chloride salt corrosion depth test, carbonation depth test, shrinkage test, and creep test. The results showed that LDHs can improve the ability of resisting ion corrosion, carbonization, shrinkage, and creep, reduce the pore content, and optimize the pore structure of mortar and concrete to some extent. Moreover, 4% LDHs had a better effect on improving the durability of mortar and concrete compared to 0.5%, 1%, and 2% LDHs, and the effect of C-LDH was better than N-LDH.

Background: Myocardial Infarction is the irreversible necrosis of the heart muscle secondary to prolong lack of oxygen supply. Troponin T is a structurally bound protein found in striated muscle cells. They have rapidly attained central role in diagnosis, prognostication and planning of therapeutic strategies in MI patients. The objective of this study was to evaluate the status of Troponin T in MI patients and its role in diagnosis compare to normal subjects.Methods: The study was conducted at M. L .N. Medical College, Allahabad, Uttar Pradesh India. A total of 136 cases were included in our study. Out of these, 86 were patients of AMI and 50 were healthy controls. They were evaluated by measurement of various parameters including enzymatic markers such as CPK-MB, SGOT, LDH1, and LDH2 and non-enzymatic markers such as troponin-T and myoglobin. Apart from these, LDL, VLDL and HDL levels were also kept under evaluation.Results: Troponin-T test was better than CPK-MB or SGOT in diagnosing myocardial infarction. In our study, sensitivity (67.3%) and specificity (73.8%) of troponin-T test was higher than CPK-MB (56.2% and 45.7%) and SGOT (34.2% and 58.3%) respectively. Troponin-T test was better than CPK-MB or SGOT after 2 hours of onset of myocardial infarction. Troponin- T and I: both kind of evaluations are available and are well evaluated. However troponin T estimation is more standardized and thus more popular. The positivity of troponin-T test also varied with area of infarct.Conclusions: High LDL and VLDL were seen while at the same time HDL level was lowered. An Increase in the level of myoglobin (non-specific marker), Cardiac troponin I and T and among the enzymatic markers elevated levels of CPK-MB, LDH and SGOT were observed in patients of MI against the normal subjects. In case of LDH both LDH 1 and LDH 2 were observed and a flipped pattern was noted. Bedside troponin-T test is highly sensitive and specific in the diagnosis of acute myocardial infarction and can be used in emergency and ambulatory settings. Qualitative troponin-T test is reliable above serum level of ≥ 0.10 ng/ml.

Bovine serum albumin (BSA) is a familiar protein and often used as a model drug to test the adsorption efficiency of material, while the cytotoxicity of hybrid is overlooked. In this study, we synthesized hierarchical mesoporous flower-like Mg–Al layered double hydroxide (LDH) microspheres through anion surfactant mediated hydrothermal method. The scanning electron microscopic images demonstrated that 10S-LDH and 100S-LDH samples possessed a porous surface and the porosity structure were affected by the concentration of anionic surfactant, i.e., sodium dodecyl sulfonate (SDS). By being immersed in the BSA/phosphate buffer solution (PBS) solution, the 100S-LDH had a loading capacity about 44.6 mg/g. Compared to 1S-LDH and 10S-LDH, the SDS intercalated in 100S-LDH could form the hydrogen bond with BSA molecules and promote the adsorption efficiency. In the cytotoxicity test towards Raw264.7 cells, the hybrid of BSA and [Formula: see text]S-LDH showed a superior biocompatibility to the raw LDH materials. The SDS intercalated Mg–Al LDH is expected to be used in the bioactive molecules delivery.

This diploma thesis is focused on the study of bioactive wound dressings. During the thesis, hydrogel, lyophilized and nanofiber wound dressings were prepared. Hydrogel and lyophilized wound dressings were prepared on basis of two polysaccharides – alginate and chitosan. Nanofiber wound dressings were prepared by spinning polyhydroxybutyrate. All prepared wound dressings were enriched with bioactive substances, which represented analgesics (ibuprofen), antibiotics (ampicillin) and enzymes (collagenase). Into hydrogel and lyophilized wound dressings were all the mentioned active substances incorporated, whereas nanofiber wound dressings were only with ibuprofen and ampicillin prepared. The theoretical part deals with the anatomy and function of human skin. There was explained the process of wound healing and also there were introduced available modern wound dressings. The next chapter of the theoretical part deals with materials for preparing wound dressings (alginate, chitosan, polyhydroxybutyrate) and with active substances, which were used during the experimental part of this thesis. In the theoretical part, the methods of preparation of nanofiber wound dressings and also the methods of cytotoxicity testing used in this work were presented. The first part of the experimental part of this thesis was focused on preparing already mentioned wound dressings. Then, their morphological changes over time and also the gradual release of incorporated active substances into the model environment were monitored. The gradual release of ampicillin was monitored not only spectrophotometrically, but also by ultra-high-performance chromatography. In wound dressings, in which collagenase was incorporated, was also the final proteolytic activity of this enzyme monitored. The effect of the active substances was observed on three selected microorganisms: Escherichia coli, Staphylococcus epidermidis and Candida glabrata. The cytotoxic effect of the active substances on the human keratinocyte cell line was monitored by MTT test and LDH test. A test for monitoring the rate of wound healing – a scratch test – was also performed.

Individually administered intelligence tests are a routine component of psychological assessments of children who may meet criteria for Attention-Deficit/Hyperactivity Disorder (ADHD), learning disorders (LD), or emotional and behavioral disorders (EBD). In addition to providing potentially useful test scores, the individual administration of an intelligence test provides an ideal opportunity for observing a child’s behavior in a standardized setting, which may contribute clinically meaningful information to the assessment process. However, little is known about the associations between test scores and test session behavior of children with these disorders. This study examined patterns of test scores and test session observations in groups of children with ADHD, LD, EBD who were administered the Stanford Binet Intelligence Scales, Fifth Edition (SB5), as well as in control children from the SB5 standardization sample. Three hundred and twelve children receiving special education services for ADHD (n = 50), LD (n = 234), EBD (n = 28) and 100 children selected from the SB5 standardization sample were selected from a data set of children who were administered both the SB5 and the Test Observation Form (TOF; a standardized rating form for assessing behavior during cognitive or achievement testing of children). The groups were then compared on SB scores and TOF scores. Associations between test scores and TOF scores in children with ADHD, LD, and EBD and normal controls were also examined. The results of this investigation indicated that children with ADHD, LD, and EBD and normal control children differed on several SB5 and TOF scales. Control children scored higher on all of the SB5 scales than children with LD, and scored higher on many of the SB5 scales than children with ADHD and EBD. Children with EBD demonstrated the most problem behavior during testing, followed by children with ADHD. Children with LD were similar to control children with respect to test session behavior. In addition, several combinations of test scores and test session behavior were able to predict diagnostic group status. Overall, the results of this investigation suggest that test scores and behavioral observations during testing can and should be important components of multi-informant, multi-method assessment of children with ADHD, LD, and EBD.

Exploring massive amount of user generated data with topics posits a new way to find useful information. The topics are assumed to be “hidden” and must be “uncovered” by statistical methods such as topic modeling. However, the user generated data is typically short and messy e.g. informal chat conversations, heavy use of slang words and “noise” which could be URL’s or other forms of pseudo-text. This type of data is difficult to process for most natural language processing methods, including topic modeling. This thesis attempts to find the approach that objectively give the better topics from short and messy text in a comparative study. The compared approaches are latent Dirichlet allocation (LDA), Re-organized LDA (RO-LDA), Gaussian Mixture Model (GMM) with distributed representation of words, and a new approach based on previous work named Neural Probabilistic Topic Modeling (NPTM). It could only be concluded that NPTM have a tendency to achieve better topics on short and messy text than LDA and RO-LDA. GMM on the other hand could not produce any meaningful results at all. The results are less conclusive since NPTM suffers from long running times which prevented enough samples to be obtained for a statistical test.

De metoder som jämfördes var latent Dirichlet allocation (LDA), Re-organized LDA (RO-LDA), Gaussian Mixture Model (GMM) with distributed representation of words samt en egen metod med namnet Neural Probabilistic Topic Modeling (NPTM) baserat på tidigare arbeten. Den slutsats som kan dras är att NPTM har en tendens att ge bättre ämnen på kort och stökig text jämfört med LDA och RO-LDA. GMM lyckades inte ge några meningsfulla resultat alls. Resultaten är mindre bevisande eftersom NPTM har problem med långa körtider vilket innebär att tillräckligt många stickprov inte kunde erhållas för ett statistiskt test.

Ce travail concerne l'étude et la réalisation de stratégies d'auto-test intégrées pour VCO radiofréquence (RF). La complexité des circuits intégrés RF devient un obstacle pour la mesure des principaux blocs RF des chaines de transmission/réception. Certains nœuds ne sont pas accessibles, l'excursion en tension des signaux baisse et les signaux haute fréquence ne peuvent pas être amenés à l'extérieur de la puce sans une forte dégradation. Le s techniques habituelles de test deviennent très couteuses et lentes. Le test pour le wafer-sort est étudié en premier. La solution proposée est la mise en œuvre d'une stratégie d'auto-test intégrée (BIST) qui puisse discriminer entre circuits sans fautes et circuits avec fautes pendant le wafer-test. La méthodologie utilisée est le test structurel. La couverture des fautes est étudiée pour connaitre la quantité à capter au niveau intégré afin de maximiser la probabilité de trouver tous les défauts physiques dans le VCO. Le résultat de cette analyse montre que la couverture des fautes est maximisée quand la tension crête-crête en sortie du VCO est captée. La caractérisation complète (validation de la puce et process-monitoring) du VCO est étudiée dans la deuxième étape. Les informations à extraire de la puce sont: amplitude des signaux, consommation du VCO, fréquence d'oscillation, gain de conversion (Kvco) et une information à propos du bruit de phase. Un démonstrateur pour le test au niveau wafer est réalisé en technologie ST CMOS 65nm. This work deals with the study and the realization of Built-In Self-Tests (BIST) for RF VCOs (Voltage Controlled Oscillators) The increasing complexity of RF integrated circuits is creating an obstacle for the correct measurement of the main RF blocks of any transceiver. Some nodes are not accessible, the voltage excursion of the signals is getting lower and lower and high frequency signals cannot be driven off the die without a main degradation. The common test techniques become then very expensive and time consuming. The wafer sort is firstly approached. The proposed solution is the implementation of a BIST strategy able to discriminate between faulty and good circuits during the wafer test. The chosen methodology is the structural test (fault-oriented). A fault coverage campaign is carried out in order to find the quantity to monitor on-chip that maximizes the probability to find all possible physical defects in the VCO. The result of the analysis reveals that the fault coverage is maximized if the peak-to-peak output voltage is monitored. The complete on-chip characterization of the VCO is then addressed, for chip validation and process monitoring. The information that need to be extracted on-chip concern: amplitude of the signal, consumption of the VCO, frequency of oscillation, its conversion gain (voltage-to-frequency) and eventually some information on the phase noise. A silicon demonstrator for wafer sort purposes is implemented using the ST CMOS 65nm process. It includes a 3.5GHz VCO, an LDO, a temperature and supply-voltage independent voltage reference, a peak-to-peak voltage detector and a comparator. The Vpp detector outputs a DC-voltage that is compared to a predefined acceptance boundary. A logic pass/fail signal is output by the BIST. The attention is then turned to the study of the proposed architecture for an on-chip frequency-meter able to measure the RF frequency with high accuracy. Behavioral simulations using VHDL-AMS lead to the conclusion that a TDC (Time-to-Digital Converter) is the best solution for our goal. The road is then opened to the measure of long-time jitter making use of the same TDC

This study investigated the effect of instruction that included graphic organizers on LDS seminary students’ ability to understand scriptural text and their ability to identify doctrines in scriptural text, utilizing a repeated measures, quasi-experimental design involving 209 ninth-grade student participants. The participants were randomly assigned by class to one of two treatment groups. Participants in the treatment group received instruction using graphic organizers with the standard curriculum and participants in the comparison group received instruction using only the standard curriculum. Three different measures were employed to measure the effectiveness of the graphic organizers intervention: (a) a multiple-choice test of LDS doctrines and principles; (b) an identifying doctrines and principles in text test; and (c) a student perception survey. Results of the ANOVA for the multiple-choice test indicated no significant difference between instructional groups for ability to recall facts from the class instruction and the class text, F (1, 205) = 1.60, p = .21, partial ή² = .21. Results of the ANOVA for the identifying doctrines and principles in text test, measuring transferability of the skills learned while studying the Doctrine and Covenants to a different text containing some of the same doctrines and principles, also indicated no significant difference between groups, F (1, 196) = 1.93, p = .17. The results for the student perception survey were positive; most students felt confident about their ability to comprehend scriptural text, but were slightly less confident about their ability to identify doctrines and principles in the text. The participants in this study were generally positive in their willingness to learn about and use graphic organizers. Results of this study indicated that graphic organizers did not significantly impact students’ ability to identify doctrines and principles in scriptural text or to learn concepts from scriptural text.

In this work, we focus on the task of clustering businesses in the state of Kansas based on the content of their websites and their business listing information. Our goal is to cluster the businesses and overcome the challenges facing current approaches such as: data noise, low number of clustered businesses, and lack of evaluation approach. We propose an LSA-based approach to analyze the businesses’ data and cluster those businesses by using Bisecting K-Means algorithm. In this approach, we analyze the businesses’ data by using LSA and produce businesses’ representations in a reduced space. We then use the businesses’ representations to cluster the businesses by applying the Bisecting K-Means algorithm. We also apply an existing LDA-based approach to cluster the businesses and compare the results with our proposed LSA-based approach at the end. In this work, we evaluate the results by using a human-expert-based evaluation procedure. At the end, we visualize the clusters produced in this work by using Google Earth and Tableau. According to our evaluation procedure, the LDA-based approach performed slightly bet- ter then the LSA-based approach. However, with the LDA-based approach, there were some limitations which are: low number of clustered businesses, and not being able to produce a hierarchical tree for the clusters. With the LSA-based approach, we were able to cluster all the businesses and produce a hierarchical tree for the clusters.

The ability to automatically analyze and segment news articles by their content is a growing research field. This thesis explores the unsupervised machine learning method topic modeling applied on Swedish news articles for generating topics to describe and segment articles. Specifically, the algorithms non-negative matrix factorization (NMF) and the latent Dirichlet allocation (LDA) are implemented and evaluated. Their usefulness in the news media industry is assessed by its ability to serve as a uniform categorization framework for news articles. This thesis fills a research gap by studying the application of topic modeling on Swedish news articles and contributes by showing that this can yield meaningful results. It is shown that Swedish text data requires extensive data preparation for successful topic models and that nouns exclusively and especially common nouns are the most suitable words to use. Furthermore, the results show that both NMF and LDA are valuable as content analysis tools and categorization frameworks, but they have different characteristics, hence optimal for different use cases. Lastly, the conclusion is that topic models have issues since they can generate unreliable topics that could be misleading for news consumers, but that they nonetheless can be powerful methods for analyzing and segmenting articles efficiently on a grand scale by organizations internally. The thesis project is a collaboration with one of Sweden’s largest media groups and its results have led to a topic modeling implementation for large-scale content analysis to gain insight into readers’ interests.

This chapter demonstrates how big data and computation can be used to identify and track recurrent themes as the products of external influence. It first considers the limitations of the Google Ngram Viewer as a tool for tracing thematic trends over time before turning to Douglas Biber's Corpus Linguistics: Investigating Language Structure and Use, a primer on various factors complicating word-focused text analysis and the subsequent conclusions one might draw regarding word meanings. It then discusses the results of the author's application of latent Dirichlet allocation (LDA) to a corpus of 3,346 nineteenth-century novels using the open-source MALLET (MAchine Learning for LanguagE Toolkit), a software package for topic modeling. It also explains the different types of analyses performed by the author, including text segmentation, word chunking, and author nationality, gender and time-themes relationship analyses. The thematic data from the LDA model reveal the degree to which author nationality, author gender, and date of publication could be predicted by the thematic signals expressed in the nineteenth-century novels corpus.

The patient who presents with microangiopathic haemolytic anaemia, thrombocytopenia, and evidence of acute kidney injury presents a diagnostic and management challenge. Haemolytic uraemic syndrome (HUS) and thrombotic thrombocytopenic purpura (TTP) are two of the conditions that frequently present with this triad. They are characterized by low platelet count with normal or near-normal coagulation tests, anaemia, and signs of intravascular red cell fragmentation on blood films, and high LDH levels.HUS associated with shiga-like toxins produced usually by E.coli (typically O157 strains) may occur in outbreaks or sporadically, with geographical variations in incidence. It is predominantly a disease of young children in which painful blood diarrhoea in a minority of infected patients is succeeded by microangiopathy and acute kidney injury. Management is supportive and recovery is usual, although permanent renal damage may lead to later deterioration. Older patients may be affected and tend to have worse outcomes. Neuraminidase-producing Streptococcus pneumoniae infections (usually pneumonia) very rarely cause a similar HUS.Atypical HUS occurs sporadically and is increasingly associated with defects in the regulation of the complement pathway, either genetic or autoimmune-caused. It may respond to plasma exchange for fresh frozen plasma. Recurrences are common, including after transplantation.TTP is associated with more neurological disease and less renal involvement, but HUS and TTP overlap substantially in their manifestations. The underlying problem is in von Willebrand factor (vWF) cleavage. The plasma metalloprotease ADAMTS13 is responsible for cleaving vWF multimers, a process that is important to prevent thrombosis in the microvasculature. Autoantibodies or rarely genetic deficiency may impair this process. Plasma exchange may remove antibodies and replenish the protease.

This article deals with the acquisition of lexical knowledge, instrumental in complementing the ambiguous process of NLP (natural language processing). Imprecise in nature, lexical representations are mostly simple and superficial. The thesaurus would be an apt example. Two primary tools for acquiring lexical knowledge are ‘corpora’ and ‘machine-readable dictionary’ (MRD). The former are mostly domain specific, monolingual, while the definitions in MRD are generally described by a ‘genus term’ followed by a set of differentiae. Auxiliary technical nuances of the acquisition process, find mention as well, such as ‘lexical collocation’ and ‘association’, referring to the deliberate co-occurrence of words that form a new meaning altogether and loses it whenever a synonym replaces either of the words. The first seminal work on collocation extraction from large text corpora, was compiled around the early 1990s, using inter-word mutual information to locate collocation. Abundant corpus data would be obtainable from the Linguistic Data Consortium (LDC).

Though more studies have been dedicated to the place of Kant in Agamben’s oeuvre, Hegel – that other major Enlightenment philosopher indispensable to modernity – holds an equally formative, if perhaps more subtle, place in his work. From the very earliest to the latest texts, Agamben’s work seeks to surpass the horizon of Western metaphysics through a philological engagement with the negative, formed in large part through a complex confrontation with Hegel. Agamben’s grappling with the dialectic in search of its idling is not merely strategic, but as he puts it, ‘one of the most urgent tasks today’ for a Marxist philosophy shored on its wreckage (IH 39). In ‘The Discreet Taste of the Dialectic’, Antonio Negri claims that the work of Agamben enables a ‘discreet dialectical rediscovery’ typifying left Hegelianism and the young Marx, resulting not in ‘the triumph of the Aufhebung‘, but in ‘the heroism of the negative’.1 Rather than valorising the negative, however, as Agamben painstakingly argues in his early text Language and Death, it is precisely the negative structure of the Voice, or, in Hegel’s terms, the ‘bad infinity’ predicted on division, that Agamben’s thought seeks to absolve (LD 100).

Abstract The literature on formability and LDH (Limited Dome Height) testing is reviewed. The LDH tooling uses a 4.0 inch (100 mm) diameter punch, and various widths of specimens are used to give various strain ratios. The strains are then plotted on forming limit curves (FLD’s). In this paper, the development of a new mini-LDH test for thin sheet metal is described. The test involves a new geometry of specimens and a fixture which can be mounted in any universal testing machine with a low enough force range. The size effect is studied by conducting LDH tests with smaller diameter punches: 2.0 inch (50.8 mm), 0.50 inch (12.7 mm), and 0.25 inch (6.35 mm) diameters. Experimental results are presented for 0.010 inch (0.254 mm) thick 1100-H19 aluminum, and 5182-H19 aluminum. An analysis is also presented for the fully biaxial case and the plane strain case in order to model the experimental results.

In the present study, a basic comparison between the cold formability of a commercially-pure Titanium (CP Ti) sheet in the single-point incremental forming (SPIF) and stamping processes is presented. An attempt was made to evaluate the SPIF formability by employing two tests. In the first test, parts having continuously varying wall angles were formed. While in the second test, parts having fixed wall angles were formed. The stamping formability was determined by conducting the limiting dome height (LDH) test. It is concluded that the forming limit curve (FLC) in SPIF is located much higher than the stamping FLC, even higher than the fracture limit curve in stamping. Moreover, the SPIF formability shows dependence on the test employed.

Анотація:
Leukocyte poor platelet concentrates (PC), containing less than 10 leukocytes, prepared from buffycoats can be stored in normal PVC bags for 7 days at 22°C without deterioration of the pH. We assumed that a low number of leukocytes present in the PC, is a critical factor to maintain the pH. To test this hypothesis increasing amounts of leukocytes were added to four groups of three PC with comparable plasma volumes (mean 58.6 ± 0.8 (SD) ml) and platelet concentrations (1.01 ± 0.04×109 /ml). Group I had a leukocyte concentration of 0.14±0.048×106 /ml, group II 1.96±0.09×106 /ml, group III 5.53±0.98×106 /ml, and group IV 13.0±0.93×106 /ml. The PC were stored in normal PVC bags for 7 days at 22°C. Measurements in vitro were performed at day 0, 2, 5 and 7.The initial mean pH value was 7.12±0.02 (SD) for all PC and dropped to 6.89, 6.85, 6.77 and 6.61 for group I to IV respectively, at day 7. A significant correlation (Spearman rank test) between low pH values and high leukocytes was found. The same significant positive correlation was observed between high leukocyte concentrations and high glucose consumption and high lactate production and LDH release during storage.These results show that the amount of leukocytes in PC has a significant contribution to the detrimental effect on pH during platelet storage. It is therefore important to prepare PC with a leukocyte count lower than 10 . Moreover the risk of alloimmunisation against HLA antigens will be diminished.

Lipid droplets (LDs) are small mobile organelles conserved in all eukaryotic cells. We wanted to test if the LD movement can be muffled by an incomplete inhibition of mitochondrial respiration, induced by treating hyphae of filamentous fungus Phycomyces blakesleeanus with 0.5 mM sodium azide. Nitrogen starved hyphae were used, in order to obtain LDs in larger sizes and numbers. The data obtained unequivocally showed: 1. Sodium azide treatment dramatically reduces the LD velocity and the distances LDs travel; 2. LDs in both controls and in azide-treated hyphae oscillate in a small confined space instead of travelling through the cell; 3. Azide-treated LDs oscillate less frequently and in smaller confinement than controls.

Context: Ataxic syndromes in the emergency department have a vast differential diagnosis, including immune, nutritional, and other etiologies. Case report: Man, 67yo, alcoholic, but previously healthy, came to ED due to complaints of falls and progressive dysesthesia in lower limbs which started 7 days from admission. He denied other toxic exposures or infectious history, except covid-19 one month before the current symptoms. Physical examination revealed mild hoarseness, left hemipalate paresis, tactile and position sense hypoesthesia, no sensory level, absence of profound reflexes; right hypotonia, global ataxia, and he was unable to walk. Neuroaxis MRI was normal. CSF obtained at D10 had 2 lymphocytes/mm3, 96mg/dL of proteins and no other abnormalities. Hb=13g/ dL, VCM=103fL, B12=424pg/mL, LDH and homocysteine were high. Negative anti-HTLV and -transglutaminase; covid-19 rapid test was positive, but rt-PCR was negative. ENMG revealed moderate sensory-motor axonal polyneuropathy. We hypothesized that the main differential diagnoses were subacute combined degeneration of spinal cord (although high level of protein in CSF) and Miller-Fisher syndrome (although ophthalmoparesis was absent). Considering that there was no motor nor respiratory symptoms, we did not offer pulse therapy and opted for B12 vitamin replacement. The patient had a favorable response to therapies and was able to walk at two weeks of treatment. Conclusions: We did not define the etiology of this case, however, it is important to keep B12 deficiency in mind (even when it is into the lab reference range) and to pay attention to post-viral atypical manifestations, mostly in the context of the current covid-19 pandemic.

Pipelines in remote and ecologically sensitive regions pose special challenges for pipeline integrity monitoring. These challenges include difficulties of access, reliability issues of communication and instrumentation that may impact the leak detection technology applied in these regions. The selection, application and continuous testing of an appropriate technology to detect possible leaks are important to pipeline integrity monitoring. The paper reports theoretical assessment and extensive testing on an Enbridge subarctic liquid pipeline. It also reports the comprehensive cost-benefit analysis to guide the leak detection instrument design/configuration and the evaluation of the capabilities of alternative computational pipeline monitoring (CPM) technologies for leak detection. The selected test pipeline is an 869 kilometer (540 mile), NPS 12 inch pipeline that transports sweet crude through environmentally sensitive areas. This pipeline currently uses a real-time transient model (RTTM) style CPM as the leak detection system (LDS). The pipeline LDS is tested annually by a number of industry-recognized methodologies. These include fluid withdrawal tests, simulated leak tests and an API-1130 instrument adjustment approach. This pipeline is also assessed by API-1149 for its theoretical CPM leak detection sensitivity. A pilot project invited commercial CPM-style vendors to participate in an LDS test using data from fluid withdrawal and simulated leak tests. Five vendors responded and were included in the test suite. The paper describes the design and implementation of the test process. The results of the commercial systems are presented in aggregated form and the participating vendors remain anonymous. Performance assessment focuses on the LDS evaluation factors of sensitivity and accuracy. The paper concludes with a “lessons learned” review of issues associated with test design.

Original objectives: To create BAC contigs covering two QTL containing chromosomal regions (QTLR) and obtain BAC end sequence information as a platform for SNP identification. Use the SNPs to search for marker-QTL linkage disequilibrium (LD) in the test populations (US and Israel Holstein cattle). Identify candidate genes, test for association with dairy cattle production and functional traits, and confirm any associations in a secondary test population. Revisions in the course of the project: The selective recombinant genotyping (SRG) methodology which we implemented to provide moderate resolution QTL mapping turned out to be less effective than expected, due to problems introduced by incomplete marker informativity. This required a no-cost one-year extension of the project. Aside from this, the project was implemented essentially as envisaged, but only with respect to a single QTLR and single population association-test. Background to the topic. Dairy cattle breeders are looking to marker-assisted selection (MAS) as a means of identifying genetically superior sires and dams. MAS based on population-wide LD can be many times more effective than MAS based on within-family linkage mapping. In this proposal we developed a protocol leading from family based QTL mapping to population-wide LD between markers and the QTL Major conclusions, solutions, achievements. The critical importance of marker informativity for application of the SRG design in outcrossing random mating populations was identified, and an alternative Fractioned Pool Design (FPD) based on selective DNA pooling was developed. We demonstrated the feasibility of constructing a BAC contig across a targeted chromosomal region flanking the marker RM188 on bovine chromosome BTA4, which was shown in previous work to contain a QTL affecting milk production traits. BAC end sequences were obtained and successfully screened for SNPs. LD studies of these SNPs in the Israel population, and of an independent set of SNPs taken across the entire proximal region of BTA4 in the USA population, showed a much lower degree of LD than previously reported in the literature. Only at distances in the sub-cM level did an appreciable fraction of SNP marker-pairs show levels of LD useful for MAS. In contrast, studies in the Israel population using microsatellite markers, presented an equivalent degree of LD at a 1-5 separation distance. SNP LD appeared to reflect historical population size of Bostaurus (Ne=5000- 10,000), while microsatellite LD appeared to be in proportion to more recent effective population size of the Holstein breed (Ne=50-100). An appreciable fraction of the observed LD was due to Family admixture structure of the Holstein population. The SNPs MEOX2/IF2G (found within the gene SETMAR at 23,000 bp from RM188) and SNP23 were significantly associated with PTA protein, Cheese dollars and Net Merit Protein in the Davis bull resource population, and were also associated with protein and casein percentages in the Davis cow resource population. Implications. These studies document a major difference in degree of LD presented by SNPs as compared to microsatellites, and raise questions as to the source of this difference and its implications for QTL mapping and MAS. The study lends significant support to the targeted approach to fine map a previously identified QTL. Using high density genotyping with SNP discovered in flanking genes to the QTL, we have identified important markers associated with milk protein percentage that can be tested in markers assisted selection programs.

Review question / Objective: Precocious puberty is defined as the onset of secondary sexual characteristics before the age of 8 years in girls and 9 years in boys. It can be differentiated into central precocious puberty (CPP) and peripheral precocious puberty, and it is more common in girls than in boys. CPP may result in a decreased final adult height, an early age at menarche, and psychological and health problems in adulthood. Gonadotropin-releasing hormone (GnRH) GnRH stimulation test has been indispensable in the diagnosis of CPP. GnRH stimulation test is not only invasive, time-consuming and expensive, but also sometimes difficult to have patients cooperate. Nocturnal urinary LH and FSH can represent gonadotropin excretion in children with normal and early puberty. And urinary sample collection and evaluation are more convenient, more acceptable, cheaper, and noninvasive. This meta-analysis aims to assess the value of first-voided urinary luteinizing hormone (LH) and the ratio of urinary luteinizing hormone and follicle-stimulating hormone (FSH) in the diagnosis of female CPP and to compare the accuracy between urinary gonadotropins and serum GnRH-stimulated gonadotropins.

Reproductive failure associated with heat stress is a well known phenomenon in avian species. Increased prolactin (PRL) levels in response to heat stress have been suggested as a mechanism involved in this reproductive malfunction. To test this hypothesis, laying female turkeys were subjected to 40°C for 12 h during the photo-phase daily or maintained at 24–26°C. Birds in each group received oral treatment with parachlorophenyalanine (PCPA; 50 mg/kg BW/day for 3 days), an inhibitor of serotonin (5-HT) biosynthesis; or immunized against vasoactive intestinal peptide (VIP). Both treatments are known to reduce circulating PRL levels. Non treated birds were included as controls. In the control group, high ambient temperature terminated egg laying, induced ovarian regression, reduced plasma luteinizing hormone (LH) and ovarian steroids (progesterone, testosterone, estradiol) levels, and increased plasma PRL levels and the incidence of incubation behavior. Pretreatment with PCPA reduced (P< 0.05) heat stress-induced decline in egg production, increase in PRL levels, and expression of incubation behavior. Plasma LH and ovarian steroid levels of heat stressed birds were restored to that of controls by PCPA treatment. As in PCPA-treated birds, VIP immunoneutralization of heat-stressed turkeys reduced (P< 0.05) circulating PRL levels and prevented the expression of incubation behavior. But it did not restore the decline in LH, ovarian steroids, and egg production (P> 0.05). The present findings indicate that the detrimental effect of high temperature on reproductive performance may not be related to the elevated PRL levels in heat-stressed birds but to mechanism(s) that involve 5-HT neurotransmission and the induction of hyperthermia.

The general research objective was to increase herd pregnancy rates by enhancing corpus luteum (CL) function and optimizing follicle development, in order to increase conception rate and embryo survival. The specific objectives were: to determine the effect of the duration of the preovulatory LH surge on CL function; to determine the function of LH during the postovulatory period on CL development; to optimize CL differentiation and follicle development by means of a biodegradable GnRH implant; to test whether optimization of CL development and follicle dynamics in timed- insemination protocols would improve fertility in high-yielding dairy cows. Low fertility in cattle results in losses of hundreds of millions of dollars in the USA and Israel. Two major causes of low fertility are formation of a functionally impaired CL, and subsequent enhanced ovarian follicle development. A functionally impaired CL may result from suboptimal LH secretion. The two major causes of low fertility in dairy cattle in US and Israel are negative energy status and summer heat stress; in both situations, low fertility is associated with reductions in LH secretion and impaired development of the ovulatory follicle and of the CL. In Florida, the use of 450-mg deslorelin (GnRH analogue) implants to induce ovulation, under the Ovsynch protocol resulted in a higher pregnancy rates than use of 750-mg implants, and pregnancy losses tended to decrease compared to controls, due probably to decrease in follicular development and estradiol secretion at the time of conceptus signaling to maintain the CL. An alternative strategy to enhance progesterone concentrations involved induction of an accessory CL by injection of hCG on day 5 after the cows were inseminated. Treatment with hCG resulted in 86% of the cows having two CLs, compared with 23% of the control cows. Conception rates were higher among the hCG-treated cows than among the controls. Another approach was to replace the second injection of GnRH analogue, in a timed-insemination protocol, with estradiol cypionate (ECP) injected 24 h after the injection of PGF₂ₐ Pregnancy rates were comparable with those obtained under the regular Ovsynch (timed- AI) program. Use of ECP induced estrus, and cows inseminated at detected estrus are indeed more fertile than those not in estrus at the time of insemination. Collectively, the BARD-supported programs at the University of Florida have improved timed insemination programs. In Ohio, the importance of the frequency of LH episodes during the early stages of the estrous cycle of cattle, when the corpus luteum is developing, was studied in an in vivo experiment in which cows were subjected to various episodic exposures to exogenous bovine LH. Results indicate that the frequent LH episodes immediately following the time of ovulation are important in development of the corpus luteum, from the points of view of both size and functionality. In another study, rates of cell proliferation and numbers of endothelial cells were examined in vitro in CLs collected from cows that received post-ovulation pulsatile LH treatment at various frequencies. The results indicate that the corpora lutea growth that results from luteal cell proliferation is enhanced by the episodes of LH release that occur immediately after the time of ovulation in cattle. The results also show that luteal endothelial cell numbers did not differ among cows treated with different LH doses. In Israel. a longer duration of the preovulatory LH surge stimulated the steroidogenic capacity of granulosa-derived luteal cells, and might, thereby, contribute to a higher progesterone output from the bovine corpus luteum. In an in vivo study, a subgroup of high-yielding dairy cows with extended estrus to ovulation interval was identified. Associated with this extended interval were: low plasma progesterone and estradiol concentrations and a low preovulatory LH surge prior to ovulation, as well as low post- ovulation progesterone concentration. In experiments based on the above results, we found that injection of GnRH at the onset of estrus increased the LHpeak, prevented late ovulation, decreased the variability between cows and elicited high and uniform progesterone levels after ovulation. GnRH at estrus onset increased conception rates, especially in the summer, and among primiparous cows and those with low body condition. Another study compared ovarian functions in multiparous lactating cows with those in nulliparous non-lactating heifers. The results revealed differences in ovarian follicular dynamics, and in plasma concentrations of steroids and gonadotropins that may account for the differences in fertility between heifers and cows.

Hilly terrain poses challenges to truck platoons using fixed set speed cruise control. Driving the front truck efficiently on hilly terrain improves both trucks fuel economies and improves gap maintenance between the trucks. An experimentally-validated simulation model was used to show fuel savings for the platoon of 12.3% when the front truck uses long horizon predictive cruise control (LH-PCC), 8.7% when the front truck uses flexible set speed cruise control, and only 1.2% when the front truck uses fixed set speed cruise control. Purdue, Peloton, and Cummins have jointly configured two Peterbilt 579 trucks for relevant combinations of: (1) coordinated shifting, (2) constant or variable platoon gap controls, (3) flexible or constant speed setpoint cruise control of the front trucks, and (4) long-horizon predictive cruise control (LHPCC) of the front truck. Confirmation of this functionality during platooning was demonstrated at the Continental Test track in Uvalde, Texas. In Indiana, on-road experiments were limited to single truck operation with long-horizon predictive cruise control, flexible set speed cruise control, and constant setpoint cruise control. Data from all of the above was used to improve the fidelity of simulations used to arrive at the fuel savings and gap control findings for hilly terrain per what is summarized in the findings section. Additionally, in early summer 2020, Purdue submitted to, and received improvement from, INDOT for a safe truck platoon testing protocol (located in this report’s appendix), which could not be implemented in Indiana before the end of the project because of COVID-19. Presentations of the subject matter at COMVEC, MAASTO, Purdue Road School, and the Work Truck Show are listed in the appendix.
