Анотація:
Ground-penetrating radar (GPR), as a non-invasive instrument, has been widely used in civil engineering. In GPR B-scan images, there may exist random noise due to the influence of the environment and equipment hardware, which complicates the interpretability of the useful information. Many methods have been proposed to eliminate or suppress the random noise. However, the existing methods have an unsatisfactory denoising effect when the image is severely contaminated by random noise. This paper proposes a multi-scale convolutional autoencoder (MCAE) to denoise GPR data. At the same time, to solve the problem of training dataset insufficiency, we designed the data augmentation strategy, Wasserstein generative adversarial network (WGAN), to increase the training dataset of MCAE. Experimental results conducted on both simulated, generated, and field datasets demonstrated that the proposed scheme has promising performance for image denoising. In terms of three indexes: the peak signal-to-noise ratio (PSNR), the time cost, and the structural similarity index (SSIM), the proposed scheme can achieve better performance of random noise suppression compared with the state-of-the-art competing methods (e.g., CAE, BM3D, WNNM).

Elevation in the cytosolic free calcium is crucial for plant growth, development and adaptation. Calcium influx into plant cells is mediated by Ca2+ depolarisation-activated, hyperpolarisation-activated and voltage-independent Ca2+-permeable channels (DACCs, HACCs and VICCs respectively). These channels are encoded by the following gene families: (1) cyclic nucleotide-gated channels (CNGCs), (2) ionotropic glutamate receptors (GLRs), (3) annexins, (4) ‘mechanosensitive channels of small (MscS) conductance’-like channels (MSLs), (5) ‘mid1-complementing activity’ channels (MCAs), Piezo channels, and hyperosmolality-induced [Ca2+]cyt. channel 1 (OSCA1). Also, a ‘tandem-pore channel1’ (TPC1) catalyses Ca2+ efflux from the vacuole in response to the plasma membrane-mediated Ca2+ elevation. Recent experimental data demonstrated that Arabidopsis thaliana (L.) Heynh. CNGCs 2, 5–10, 14, 16 and 18, GLRs 1.2, 3.3, 3.4, 3.6 and 3.7, TPC1, ANNEXIN1, MSL9 and MSL10,MCA1 and MCA2, OSCA1, and some their homologues counterparts in other species, are responsible for Ca2+ currents and/or cytosolic Ca2+ elevation. Extrusion of Ca2+ from the cytosol is mediated by Ca2+-ATPases and Ca2+/H+ exchangers which were recently examined at the level of high resolution crystal structure. Calcium-activated NADPH oxidases and reactive oxygen species (ROS)-activated Ca2+ conductances form a self-amplifying ‘ROS-Ca2+hub’, enhancing and transducing Ca2+ and redox signals. The ROS-Ca2+ hub contributes to physiological reactions controlled by ROS and Ca2+, demonstrating synergism and unity of Ca2+ and ROS signalling mechanisms.

The transport of cationic amino acids across animal cell membranes is largely mediated by a small group of well-described transport system (y+, bo,+, Bo,+). Only recently have genes encoding transport proteins in some of these systems been isolated. Two genes, mCAT-1 and mCAT-2, encode related multiple membrane-spanning proteins that share substantial amino acid sequence identity and virtually superimposable hydrophilicity profiles. mCAT-1 and mCAT-2 proteins expressed in Xenopus oocytes are functionally indistinguishable and similar to transport system y+, but have distinct tissue distribution patterns. mCAT-1 expression is nearly ubiquitous and produces a single protein, while mCAT-2 is highly tissue-specific, has two distinct protein isoforms encoded by a single gene and is expressed in different tissues using at least two widely separated promoters. All three proteins facilitate the ion-independent transport of arginine, lysine and ornithine. Both mCAT-1 and mCAT-2 proteins have low amino acid sequence similarity but strikingly similar hydrophilicity profiles with amino acid antiporters, uniporters and symporters of yeast, fungi and eubacteria. Current work will elucidate whether any of the mCAT proteins interact with members of a newly identified family of single membrane-spanning proteins, such as rBAT, 4F2 and NAA-Tr, which are thought to modulate or activate y+L and/or bo,+ transport systems.

Анотація:
Learning during the Covid-19 pandemic caused learning activity to be online and causes student’s boredom in Geography. Therefore, teachers must integrating the technology in learning process, with mobile learning (M-Learning) based on mobile context aware systems (MCAS). The study purpose is to determine student’s opinions about implementation of MCAS based M-Learning during the pandemic. This research is a descriptive qualitative with a mix method approach used collection techniques field research and literature study. The results showed that students had a positive opinion regarding the implementation of MCAS based M-Learning during the pandemic, with an average score of 3.40-3.70 out of 4.00. Pembelajaran pada masa pandemi Covid-19 menyebabkan pembelajaran menjadi online sehingga menyebabkan terjadinya kejenuhan siswa dalam proses pembelajaran Geografi. Oleh karena itu, guru harus mampu mengintegrasikan teknologi dalam pembelajaran, melalui mobile learning (M-Learning) berbasis mobile context aware systems (MCAS). Penelitian ini bertujuan mengetahui opini siswa dalam penerapan M-Learning berbasis MCAS pada masa pandemi. Jenis penelitian ini termasuk kualitatif deskriptif dengan pendekatan mix method serta teknik pengumpulan data berupa penelitian lapangan serta studi kepustakaan. Hasil penelitian menunjukkan bahwa siswa memiliki opini positif terkait implementasi M-Learning berbasis MCAS pada masa pandemi, dengan perolehan skor rata-rata skor 3,40-3,70 dari 4,00.

Abstract MCAM (CD146) is a cell surface adhesion molecule that has been reported to promote cancer development, progression and metastasis and is considered as a potential tumor biomarker and therapeutic target. However, inconsistent reports exist, and its clinical value is yet to be confirmed. Here we took advantage of several large genomic data collections (Genotype-Tissue Expression, The Cancer Genome Atlas and Cancer Cell Line Encyclopedia) and comprehensively analyzed MCAM expression in thousands of normal and cancer samples and cell lines along with their clinical phenotypes and drug response information. Our results show that MCAM is very highly expressed in large vessel tissues while majority of tissues have low or minimal expression. Its expression is dramatically increased in a few tumors but significantly decreased in most other tumors relative to their pairing normal tissues. Increased MCAM expression is associated with a higher tumor stage and worse patient survival for some less common tumors but not for major ones. Higher MCAM expression in primary tumors may be complicated by tumor-associated or normal stromal blood vessels yet its significance may differ from the one from cancer cells. MCAM expression is weakly associated with the response to a few small molecular drugs and the association with targeted anti-BRAF agents suggests its involvement in that pathway which warrants further investigation.

The work summarizes the methods of production of composite frames and important parameters that must be respected when designing the frame. A Gravel frame was chosen for the design. The frame was designed for production using stainless steel printed lugs with glued composite tubes. The proposed solution was strength tested. The work also contains a design of a production jig for the construction of bicycle frames.

The late Archean Mount McRae Shale of the Hamersley Basin in Western Australia may record the presence of oxygen in the atmosphere before the Great Oxidation Event (2.4-2.3 Ga). Several prior studies (Anbar et al., 2007; Blum and Anbar, 2010; Duan et al., 2010; Kakegawa et al., 1998; McManus et al., 2006) have used isotopic systems to analyze the Mount McRae Shale and conclude that there was a presence of oxygen before the Great Oxidation Event. The purpose of this study is to determine if the U-Pb system can be used to see through later events to the initial conditions. The uranium-lead values of the Mt McRae Shale provide evidence of the mobilization of U and Pb gain. The geochemical disturbances have been linked to the tectonic activity (460 Ma) in the neighboring Canning basin, which could have possibly opened the geochemical system. In terms of the depositional environment the U-Pb data gathered here do not point to oxygenation of the atmosphere.

The model training algorithm is a critical component in the statistical pattern recognition approaches which are based on the Bayes decision theory. Conventional applications of the Bayes decision theory usually assume uniform error cost and result in a ubiquitous use of the maximum a posteriori (MAP) decision policy and the paradigm of distribution estimation as practice in the design of a statistical pattern recognition system. The minimum classification error (MCE) training method is proposed to overcome some substantial limitations for the conventional distribution estimation methods. In this thesis, three aspects of the MCE method are generalized. First, an optimal classifier/recognizer design framework is constructed, aiming at minimizing non-uniform error cost.A generalized training criterion named weighted MCE is proposed for pattern and speech recognition tasks with non-uniform error cost. Second, the MCE method for speech recognition tasks requires appropriate management of multiple recognition hypotheses for each data segment. A modified version of the MCE method with a new approach to selecting and organizing recognition hypotheses is proposed for continuous phoneme recognition. Third, the minimum verification error (MVE) method for detection-based automatic speech recognition (ASR) is studied. The MVE method can be viewed as a special version of the MCE method which aims at minimizing detection/verification errors. We present many experiments on pattern recognition and speech recognition tasks to justify the effectiveness of our generalizations.

Understanding the future of a Large Technical System (LTS) through a sociotechnical analysis is a complex notion. The authors will investigate the phenomenon by studying residential water consumption in Sweden. Although Sweden has great access to raw water, compared to many other countries, it does not justify the fact that consumption patterns have to be unsustainable. To perceive the future, one will need to have knowledge of history. Therefore, this thesis has identified 11 drivers that will affect future residential water consumption. From these 11 drivers, the authors have posed four future scenarios for residential water consumption. The top 3 identified drivers are connected to behaviour and awareness. Thus, implying individuals' responsibility and accountability will be of greater significance for the sustainable development of residential water consumption. For this research, the authors have adopted a framework based on the application of LTS, Multi-Level Perspective (MLP), Scenario building, PESTLE, and Multi-Criteria Analysis (MCA). The authors have conducted a qualitative study, gathering data through a literature study and semi-structured interviews. The 18 interviewees covered a broad spectrum of knowledge, experiences, and general interest in water. By understanding the dynamics of the system and drivers, the authors will be able to present the gathered knowledge to stakeholders for possible strategic decisions.
Att förstå framtiden för ett stort tekniskt system (LTS) genom en socioteknisk analys innebär stor komplexitet. Detta kommer att undersökas genom att studera hushållens vattenförbrukning i Sverige. Även om Sverige har stor tillgång till råvatten, jämfört med många andra länder, berättigar det inte förekommandet av ohållbara konsumtionsmönster. För att öka förståelsen av framtiden måste en ha kunskap om historien. Därför har denna avhandling identifierat 11 drivkrafter som kommer att påverka den framtida hushållens vattenförbrukning. Från dessa 11 drivkrafter har författarna formulerat fyra framtida scenarier för hushållens vattenförbrukning. De tre högst rankade identifierade drivkrafterna är kopplade till beteende och medvetenhet. Detta antyder på att individens enskilda ansvar och skyldigheter är av större betydelse för en hållbar utveckling av hushållens vattenförbrukning. För denna forskning har författarna använt sig av ett teoretiskt ramverk som bygger på tillämpningen av LTS, Multi-Level Perspective (MLP), Scenario building, PESTLE and Multi-Criteria Analysis (MCA). Författarna har genomfört en kvalitativ studie, där insamling av data härstammar från den genomförda litteraturstudien och 18 semi-strukturerade intervjuer. De 18 intervjukandidaterna täckte ett brett spektrum av kunskap, erfarenhet och intresse av vatten. Genom att skapa ett samband och förstå dynamiken i systemet och drivkrafterna, kommer författarna att kunna presentera den samlade kunskapen till intressenter för eventuella strategiska beslut.

Acoustic modeling in state-of-the-art speech recognition systems is commonly based on discriminative criteria. Different from the paradigm of the conventional distribution estimation such as maximum a posteriori (MAP) and maximum likelihood (ML), the most popular discriminative criteria such as MCE and MPE aim at direct minimization of the empirical error rate. As recent ASR applications become diverse, it has been increasingly recognized that realistic applications often require a model that can be optimized for a task-specific goal or a particular scenario beyond the general purposes of the current discriminative criteria. These specific requirements cannot be directly handled by the current discriminative criteria since the objective of the criteria is to minimize the overall empirical error rate. In this thesis, we propose novel objective-driven discriminative training and adaptation frameworks, which are generalized from the minimum classification error (MCE) criterion, for various tasks and scenarios of speech recognition and detection. The proposed frameworks are constructed to formulate new discriminative criteria which satisfy various requirements of the recent ASR applications. In this thesis, each objective required by an application or a developer is directly embedded into the learning criterion. Then, the objective-driven discriminative criterion is used to optimize an acoustic model in order to achieve the required objective. Three task-specific requirements that the recent ASR applications often require in practice are mainly taken into account in developing the objective-driven discriminative criteria. First, an issue of individual error minimization of speech recognition is addressed and we propose a direct minimization algorithm for each error type of speech recognition. Second, a rapid adaptation scenario is embedded into formulating discriminative linear transforms under the MCE criterion. A regularized MCE criterion is proposed to efficiently improve the generalization capability of the MCE estimate in a rapid adaptation scenario. Finally, the particular operating scenario that requires a system model optimized at a given specific operating point is discussed over the conventional receiver operating characteristic (ROC) optimization. A constrained discriminative training algorithm which can directly optimize a system model for any particular operating need is proposed. For each of the developed algorithms, we provide an analytical solution and an appropriate optimization procedure.

One of the major challenges that the energy system is facing is how 100% renewable electricity generation should be designed and controlled. So far, utility-scale solar photovoltaic (PV) parks, have dominated the international market and is expected to grow in Sweden. In order for this expansion to thrive it needs to be done in a resource and system-efficient way. In this study, a methodology for a utility-scale solar guide (from Swedish 'Solbruksplan') is developed. This is done through electrical grid simulations and geographical analysis in order to find strategic locations for PV parks, without grid reinforcements, using Herrljunga municipality as a case study. The electrical grid is analyzed through power flow simulations and the geographical assessment is done using multi-criteria analysis (MCA) with a Boolean approach. Three different sizes of PV parks, 1, 3, and 5 MW, were simulated and assessed. The results show that 8.6% of the total area in the municipality, or 78,500 square meters per substation, is suitable for locating 1 MW PV parks. The majority of the grid’s substations (about 90%) also have enough capacity for a PV park of 1 MW. Furthermore, parts of the grid can host PV parks of 3 and 5 MW, but limited to the proximity of the urban areas. However, 3 and 5 MW PV parks are not suitable due to limited land availability in urban areas. The results highlight that a utility-scale solar guide can be used to effectively identify possible areas for PV parks, considering geography and grid capacity. Hence, it can function as a tool for utility companies, municipalities, PV companies and land-owners to find resource and system-efficient locations for PV parks.

Анотація:
This report describes activities performed, results obtained, and conclusions made from an independent technical review of past levee inspections and the proposed remediation plan for the Santa Margarita Levee that surrounds the U.S. Marine Corps Air Station (MCAS) Camp Pendleton. In support of the technical review, ERDC personnel performed a supplemental levee inspection on 19 and 20 November 2019 with MCAS personnel. Previous levee inspections had rated the levee system as Unacceptable due to unwanted vegetation encroaching on the levee right-of-way, which prevents full inspection during flooding. Concerns were raised by the U.S. Fish and Wildlife (USFW) about environmental impacts of the proposed remediation measures and the necessity of such actions. USFW personnel requested an engineering review from an independent party, and ERDC was tasked with performing the independent technical review. The following special report describes the tasks performed and results obtained from the independent technical review.
