Nuclear entry of circadian oscillatory gene products is a key step for the generation of a 24-hr cycle of the biological clock. We have examined nuclear import of clock proteins of the mammalianperiod gene family and the effect of serum shock, which induces a synchronous clock in cultured cells. Previously, mCRY1 and mCRY2 have been found to complex with PER proteins leading to nuclear import. Here we report that nuclear translocation of mPER1 and mPER2 (1) involves physical interactions with mPER3, (2) is accelerated by serum treatment, and (3) still occurs in mCry1/mCry2double-deficient cells lacking a functional biological clock. Moreover, nuclear localization of endogenous mPER1 was observed in culturedmCry1/mCry2 double-deficient cells as well as in the liver and the suprachiasmatic nuclei (SCN) ofmCry1/mCry2 double-mutant mice. This indicates that nuclear translocation of at least mPER1 also can occur under physiological conditions (i.e., in the intact mouse) in the absence of any CRY protein. The mPER3 amino acid sequence predicts the presence of a cytoplasmic localization domain (CLD) and a nuclear localization signal (NLS). Deletion analysis suggests that the interplay of the CLD and NLS proposed to regulate nuclear entry of PER in Drosophilais conserved in mammals, but with the novel twist that mPER3 can act as the dimerizing partner.

Mammalian circadian rhythm is observed not only at the suprachiasmatic nucleus, a master pacemaker, but also throughout the peripheral tissues. Investigation of the regulation of clock gene expression has mainly focused on transcriptional and posttranslational modifications, and little is known about the posttranscriptional regulation of these genes. In the present study, we investigate the role of microRNAs (miRNAs) in the posttranscriptional regulation of the 3′-untranslated region (UTR) of the mouse Cryptochrome 1 (mCry1) gene. Knockdown of Drosha, Dicer, or Argonaute2 increased mCry1-3′UTR reporter activity. The presence of the miRNA recognition element of mCry1 that is important for miR-185 binding decreased mCRY1 protein, but not mRNA, level. Cytoplasmic miR-185 levels were nearly antiphase to mCRY1 protein levels. Furthermore, miR-185 knockdown elevated the amplitude of mCRY1 protein oscillation. Our results suggest that miR-185 plays a role in the fine-tuned regulation of mCRY1 protein expression by controlling the rhythmicity of mCry1 mRNA translation.

ABSTRACT The mammalian circadian rhythm is observed not only at the suprachiasmatic nucleus, a master pacemaker, but also throughout the peripheral tissues. Its conserved molecular basis has been thought to consist of intracellular transcriptional feedback loops of key clock genes. However, little is known about posttranscriptional regulation of these genes. In the present study, we investigated the role of the 3′-untranslated region (3′UTR) of the mouse cryptochrome 1 (mcry1) gene at the posttranscriptional level. Mature mcry1 mRNA has a 610-nucleotide 3′UTR and mediates its own degradation. The middle part of the 3′UTR contains a destabilizing cis-acting element. The deletion of this element led to a dramatic increase in mRNA stability, and heterogeneous nuclear ribonucleoprotein D (hnRNP D) was identified as an RNA binding protein responsible for this effect. Cytoplasmic hnRNP D levels displayed a pattern that was reciprocal to the mcry1 oscillation. Knockdown of hnRNP D stabilized mcry1 mRNA and resulted in enhancement of the oscillation amplitude and a slight delay of the phase. Our results suggest that hnRNP D plays a role as a fine regulator contributing to the mcry1 mRNA turnover rate and the modulation of circadian rhythm.

Purpose – Many organizations still struggle with sales force technology implementation because of low user adoption rates. The ubiquity of mobile computing devices, such as smartphones and tablets, and the proliferation of mobile customer relationship management (mCRM) applications, may lead to increased CRM adoption and higher returns on CRM technology investments. The purpose of this study is to attempt to extend the current literature by developing a model of mCRM antecedents and outcomes by incorporating the idiosyncratic mCRM characteristics that have not yet been examined in the sales technology literature. Design/methodology/approach – This research utilizes the technology acceptance model and the technology-to-performance chain as the foundation of a conceptual model of the drivers and outcomes of mCRM adoption. Findings – This conceptual study provides several contributions to both the sales technology literature and to practitioners within sales organizations. The proposed conceptual model outlines the benefits of providing mCRM capabilities to sales professionals. These benefits include increased productivity, sales activity and collaboration among both internal stakeholders (management and peers) and external stakeholders (prospects and customers). Originality/value – Despite the increased use of mobile applications in sales, research on this particular form of technology is limited, and sales researchers have yet to examine mCRM or its relationship to sales performance. Therefore, the purpose of this research is to forward a conceptual model that allows researchers to explore the drivers of mCRM use and how mCRM influences individual and organizational-level outcomes.

ABSTRACT Cryptochromes (CRYs) are composed of a core domain with structural similarity to photolyase and a distinguishing C-terminal extension. While plant and fly CRYs act as circadian photoreceptors, using the C terminus for light signaling, mammalian CRY1 and CRY2 are integral components of the circadian oscillator. However, the function of their C terminus remains to be resolved. Here, we show that the C-terminal extension of mCRY1 harbors a nuclear localization signal and a putative coiled-coil domain that drive nuclear localization via two independent mechanisms and shift the equilibrium of shuttling mammalian CRY1 (mCRY1)/mammalian PER2 (mPER2) complexes towards the nucleus. Importantly, deletion of the complete C terminus prevents mCRY1 from repressing CLOCK/BMAL1-mediated transcription, whereas a plant photolyase gains this key clock function upon fusion to the last 100 amino acids of the mCRY1 core and its C terminus. Thus, the acquirement of different (species-specific) C termini during evolution not only functionally separated cryptochromes from photolyase but also caused diversity within the cryptochrome family.

Water matrix certified reference material (MCRM) of volatile organic compounds (VOCs) is used to provide quality assurance and quality control (QA/QC) during the analysis of VOCs in water. In this research, a water MCRM of 28 VOCs was developed using a “reconstitution” approach by adding VOCs spiking, methanol solution into pure water immediately prior to analysis. The VOCs spiking solution was prepared gravimetrically by dividing 28 VOCs into seven groups, then based on ISO Guide 35, using gas chromatography-mass spectrometry (GC-MS) to investigate the homogeneity and long-term stability. The studies of homogeneity and long-term stability indicated that the batch of VOCs spiking solution was homogeneous and stable at room temperature for at least 15 months. Moreover, the water MCRM of 28 VOCs was certified by a network of nine competent laboratories, and the certified values and expanded uncertainties of 28 VOCs ranged from 6.2 to 17 μg/L and 0.5 to 5.3 μg/L, respectively.

3020 Background: Pharmacodynamic (PD) studies, using either surrogate or tumor tissues, are frequently incorporated in Phase I trials. However, it has been less common to base dose selection, the primary endpoint in Phase I trials, in PD effects. We conducted a PD-based dose selection study with rapamycin (Rap). Methods: We used the modified continuous reassessment method (mCRM), a computer-based dose escalation algorithm, and adapted the logit function from its classic toxicity-based input data to a PD-based input. We coupled this design to a Phase I trial of Rap with 2 parts: a dose estimation phase where PD endpoints are measured in normal tissues and a confirmation phase where tumor tissue is assessed. Patients (pts) had solid tumors refractory to standard therapy. Rap was given starting at 2 mg/day continuously in 3-pt cohorts. The PD endpoint was pP70S6K in skin and tumor. Biopsies were done on days 0 and 28 of cycle 1, and a PD effect was defined as ≥ 80% inhibition from baseline. The first 2 dose levels (2 and 3 mgs) were evaluated before implementing the mCRM. The data was then fed to the computer that based on the PD effect calculated the next dose level. The mCRM was set so escalation continued until a dose level elicited a PD effect and the mCRM assigned the same dose to 8 consecutive pts, at which point the effect of that dose will be confirmed in tumor biopsies. Other correlates were PET-CT and pharmacokinetics. Results: Ten pts were enrolled at doses of 2 mg (n = 4), 3 mg (n = 3) and 6 mg (n = 3). Toxicity was anemia (4 G1, 1 G2), leucopenia (1 G1, 2 G2), low ANC (2 G2), hyperglycemia (2 G1, 1 G2), hyperlipidemia (4 G1), and mucositis (1 G1, 1 G2). PD responses were seen in 2 and 1 pt at 2 and 3 mg dose levels. Input of data to the mCRM selected a dose of 6 mg for the third cohort, where PD effect was seen in 1 pt, and thus a fourth dose around 9 mg will be tested. No responses by RECIST occurred, but 2 pts had a response by PET. The PK was consistent with prior data (t1/2 24.6 ± 10.2 h, CL 31.4 ± 12.0 L/h, vol of distribution 235 ± 65 L), and exposure increased with dose. Steady-state concentration were in the 5–20 nM range. Conclusions: mCRM-based dose escalation based on real-time PD assessment is feasible and permits the exploitation of PD effects for dose selection in a rational manner. No significant financial relationships to disclose.

Mobile technology has evolved drastically over the years, and so has the customer’s perception of and expectations on mobile services. Mobile phones have become the essential part of customer’s life style and they expect consumer services to be available on mobile phone. In this saturated market, providing better customer experience through the channel preferred by customers is as important as selling the actual product or services. In the quest to retain existing customers as well as attracting new, companies are developing innovative mobile customer relationship management (mCRM) strategies to strike a balance between its investments and fulfilling of customers need to generate maximum profit. However, not all mCRM strategies succeed leaving customers dissatisfied and switching to competitors providing better mobile services. The purpose of this study is to understand the dynamics between the customer acceptance, customer satisfaction and customer loyalty in the context of mCRM services. Thus, answering the following research questions: a) What are the factors behind customer acceptance? b) Do mCRM services have any effect on customer loyalty? Qualitative research method were utilized to closely study two different mCRM strategies from different industries with costumer in focus. Data for the study was gathered from a costumer survey as well as from interviews with informants within the company. In addition, external data from public domain was used to validate the findings of the study. Furthermore, a research model was developed by identifying the constructs adopted from literature study of technology acceptance model (TAM3) and customer loyalty. The research model was the basis for developing the customer feedback survey. In addition, the constructs helped in identifying the impact on customer acceptance and customer loyalty for the respective mCRM strategy. This study confirms previous findings and contributes to our understanding of technology acceptance as well as customer loyalty. The study reveals that mCRM services are mostly useful to urban customers with busy and on-the-go lifestyle. Furthermore, the study shows that through well practice use behavior customer develops a habit based on the prior evaluation of perceived usefulness and perceived value. In addition, the study indicates that customers build up a new trust towards the mCRM services through the usage of the service. Finally, the results of the study indicate that the mCRM services have an impact on company’s net customer loyalty.

Managing customer relations had become a necessity for companies who want to succeed in today’s customer oriented market. New systems are developed that allow firms to manage and govern their relationships to the customers. These applications are called Customer Relationship Management (CRM) Systems and have become one of the most widely adopted business solution in the private sector. In the mean time, mobile internet and mobile marketing have become two of the most popular subjects mentioned in business literature today.

A new system that combines CRM and mobile technologies has recently been developed: mobile CRM (mCRM). This type of system will allow companies to enhance their way of working with customer relations and enables new ways of offering value to customers. The market size of mCRM systems has grown from a $120 million industry in 2001 to a $1, 7 billion industries in 2005. The combination of the growing trend of customer focus, mobile technologies and mCRM systems will most likely lead to more companies investing in mCRM solutions. Still an interesting question of the reasons behind mCRM adoption remains unanswered. The main focus of this thesis is to find out why companies chose to adopt or not adopt mCRM systems.

Theories about CRM, mCRM and the spreading of technology (The Technology Acceptance Model) have been gathered and examined in order to support the research. After a gained insight of the subject matter, a methodology is discussed explaining research approach. A qualitative approach is used and interviews are conducted in order to find barriers and facilitators to mCRM adoption. Four companies from different industry sectors where interviewed, answering to questions concerning their level of IT adoption, CRM strategy and perception of mCRM.

The research resulted in several reasons for mCRM acceptance and rejection. The primary reason for investing mCRM, or considering investing in mCRM, was found to be the need to reach customers and sales force anytime and anywhere. Also, current IT adoption and IT dependency, as well as industry sector, are found to influence attitude towards mCRM. Factors that are found not to affect mCRM adoption are size of the firm and external pressure from the market. The overall perception of mCRM is positive among the respondents and all believe that the mobility aspect of CRM will become increasingly popular.

Tendo em vista que a China domina o mercado das terras-raras e impõe restrições na produção e exportação sempre que surge a oportunidade, torna-se cada vez mais desafiador o desenvolvimento de empreendimentos nesse setor. No entanto, várias iniciativas foram tomadas nos últimos cinco anos no sentido de prospectar novos recursos e desenvolver a cadeia produtiva, inclusive a mineração, desses bens minerais ao redor do mundo. Porém, alguns fatores de incerteza já influenciaram ou vêm influenciando o futuro de alguns projetos em desenvolvimento, tais como o potencial de remuneração do depósito, a infraestrutura local existente, as expectativas de margens operacionais do empreendimento, as implicações dos elementos radioativos presentes nos depósitos, as expectativas potenciais de mercado com os elementos de terras-raras (ETR) produzidos e os fatores políticoeconômicos, conjunturais e de logística. Então, este trabalho introduz uma nova metodologia dinâmica de análise de competitividade (o modelo de competitividade e risco na mineração de terras-raras – MCRM-TR), na qual se consideram as variáveis/os fatores tidos como principais influências na performance de um empreendimento de mineração destinado ao aproveitamento dos ETR. Para o desenvolvimento do modelo, foram realizadas pesquisas nos âmbitos nacional e internacional, consultando empresas e instituições de pesquisa relacionadas com o setor de terras-raras. Por fim, com os dados obtidos nas pesquisas para cada fator dos respectivos depósitos, foram realizadas simulações por meio da geração de números aleatórios, utilizando-se para isso o método de Monte Carlo. A combinação dos principais resultados gerados produziu um índice de competitividade e risco na mineração de terras-raras (ICRM-TR). Com a aplicação dessa nova metodologia, foi possível constatar algumas realidades, como por exemplo o fato de o Projeto Mount Weld CLD – (AUS) da Lynas ter apresentado bons resultados no modelo e, no mundo real, ainda se manter resiliente diante das incertezas do setor de terras-raras, ao mesmo tempo em que o projeto da Molycorp apresentou resultados não tão satisfatórios e, no mundo real, passa por grandes dificuldades financeiras (em recuperação judicial). Também foi possível constatar que o Projeto Araxá da CBMM no Brasil está entre os mais competitivos do país.
It is known that China dominates the rare earth market and imposes constraints on production and export whenever the opportunity arises, so that it always becomes more and more challenging to develop enterprises in the sector. However, several initiatives have been taken in the last five years to explore new resources and develop the production chain, including mining, of these mineral assets around the world. However, some factors of uncertainty have already influenced or are still influencing the future of some projects under development, such as reservoir remuneration potential, existing local infrastructure, expectations of operating margins of the enterprise, implications of the radioactive elements present in the deposits, the potential market expectations with the rare earth elements (REE) produced and the political-economic, conjuncture and logistics factors. So, this work introduces a new dynamic methodology for competitiveness analysis (the competitiveness and risk model in rare earth mining ‒ MCRM-TR or CRM-REM), in which the variables/factors considered as main influences in the performance of a mining for the use of REEs are taken into account. In the development of the model, research was carried out at both national and international levels, consulting companies and research institutions related to the rare earth sector. Finally, with the data obtained in the surveys for each factor of the respective deposits, simulations were performed through the generation of random numbers, using the Monte Carlo method. The combination of the main results generated produced an index of competitiveness and risk in the rare earth mining (ICRM-TR or ICM-TRM). With the application of this new methodology, it was possible to verify some realities, such as the fact that the Mount Weld CLD Project (AUS) from Lynas had good results in the model and, in the real world, still remain resilient in the face of the uncertainties of the sector at the same time the Molycorp project presented less than satisfactory results and, in the real world, undergoing major financial difficulties (in judicial recovery). It was also possible to verify that the Araxá Project of CBMM in Brazil is among the most competitive in the country.

Abstract This study examines communication through the mobile medium in customer relationship management (hereafter mCRM). Although the academic research on mCRM is gradually evolving, there is still a lack of empirical research on this topical issue. Therefore, the purpose of this study is to begin to fill this gap by enhancing the theoretical and practical understanding of mCRM. The empirical part of the study is conducted as a qualitative case study. Seven Finnish companies utilizing the mobile medium as a communication channel in CRM were selected for the empirical analysis. Interviews, observations and personal experiences gained within those companies form the main sources of data. The main theoretical contributions of the present study to existing academic literature are as follows. First, the study proposes the conceptualization of the focal phenomenon of the study, mCRM, and further elicits the salient characteristics of it. Second, following the conceptualization, the study focuses on communication as the core of the interaction between the company and the customer via the mobile medium. Accordingly, the study sheds light on the key elements of the communication process within mCRM and its various phases. Third, the study founds a theoretical framework for mCRM; identifying the customer relationship management, mobile commerce and communication literature streams that define mCRM theoretically. Fourth, the study elaborates on the specific role of the mobile medium as a channel within CRM and highlights the ways in which it differs from all other channels within the multichannel CRM environment. Although mCRM communication has an inherent potential to benefit both company and customer, it has been utilized quite cautiously to date. This is rather surprising given that, compared to other channels, the mobile medium has an exceptional ability to reach people. In addition, greater utilization of the mobile medium could save time and money. This is because many face-to-face encounters could easily be replaced by communication through the mobile medium. The study offers guidelines on aspects ranging from the initiation of mCRM to actual communication through mCRM. Hopefully, the findings of this study will encourage managers, regardless of industry, to consider whether their company could provide added-value of any kind through the use of the mobile medium to benefit their customer relationships.

Mon projet de thèse s'inscrit dans un vaste projet de caractérisation de protéines nouvellement identifiées dont l'expression est sélective à certaines régions du cerveau. Cette expression sélective pouvant être liée aux phénomènes de dégénérescence neuronale qui caractérisent les maladies neurodégénératives, ces protéines constituent donc des biomarqueurs potentiels. Une étude structurale et physico-chimique a été effectuée sur une dizaine de protéines, dont la protéine CRYM murine (mCRYM) qui fait parti de la famille des Cytosolic- 3,3',5-triiodo-L-thyronine(T3)-Binding Protein car elle régule la concentration en hormone thyroïdienne T3 libre dans la cellule. MCRYM appartient également à la famille des µ-crystallines et à la superfamille des µ-crystallines/Ornithines Cyclodésaminases. Les protéines présentant des homologies pour ces trois familles sont la plupart différentes par leur fonction (enzymatique ou structurale), leur localisation tissulaire et leurs caractéristiques physico-chimiques. Cette diversité est due au recrutement de gènes de la superfamille des crystallines pour diverses fonctions métaboliques tout en conservant le taxon spécifique des crystallines. Je suis parvenue à résoudre sa structure cristallographique complexée au NADP(H) et à l'hormone thyroïdienne T3 à une résolution de 1,75 Å. La protéine mCRYM est un exemple intéressant d'évolution par son appartenance à différentes familles de protéines et, à ce jour, aucune activité enzymatique n'a été identifiée. Sa caractérisation structurale et thermodynamique a donc permis de mettre en évidence les différences et les similitudes avec ses homologues enzymatiques et d'émettre des hypothèses quant à son évolution moléculaire. Ces résultats soulèvent de nouvelles questions concernant son rôle physiologique : mCRYM est-elle une enzyme ou une protéine structurale ? Comment intervient le couple redox NADPH/NADP+ pour réguler l'action génomique et/ou non génomique de l'hormone T3 ? L'hormone T3 est-il le seul ligand physiologique de CRYM dans le cerveau ?
My Ph. D. Work takes part of a larger project dedicated to the characterization of proteins newly involved into selective expression of certain mouse brain regions. This selective expression being potentially linked to neuronal degeneration associated with neurodegenerative diseases, the corresponding proteins are therefore potential biomarkers. A structural and physico-chemical study has been performed on about ten proteins including CRYM of mouse (mCRYM), which belongs to the Cytosolic-3,3',5-triiodo-L-thyronine(T3)-Binding Protein family since it regulates the concentration of free thyroid hormone, T3, in the cell. MCRYM also belongs to the µ-crystallin family and to the µ-crystallins/Ornithin Cyclodesaminases superfamily. Proteins displaying sequence homologies to these three families of proteins have generally different functions (enzymatic or structural), different tissue localisation and different physico-chemical properties. This diversity is due to the recruitment of genes of the crystalline superfamily to carry different metabolic functions while preserving the taxon-specific crystallins. I have managed to resolve the crystallographic structure of mCRYM in complex with NADP(H) and the thyroid hormone, T3, to 1. 75 Å resolution. MCRYM is a very interesting evolution specimen as it belongs to a different family of proteins. However, no enzymatic function has ever been demonstrated for mCRYM. Its structural and thermodynamical characterization has revealed similitudes and divergences with the enzymatic homologues of CRYM and has allowed us to make hypothesis relative to its molecular evolution. These results raise new questions concerning the physiological role of mammalian CRYM: is CRYM an enzyme or a structural protein? How does the NADPH/NADP+ redox couple regulates the genomic and/or non genomic action of the T3 hormone? Is the T3 hormone the only physiological ligand of CRYM in the brain?

This chapter reviews the microwave complex ratio measuring (MCRM) circuits which are used for complex reflection coefficient measurement. This MCRM circuit is relatively simple and cost-effective. There are various structures for the MCRM circuit, such as multi-probe transmission line circuits, five-port ring circuits, six-port hybrid coupler-based circuits, switched-reflector circuits, dual-generator circuits, and Wheatstone bridge-based circuits. Each structure of the circuits has its own advantages and disadvantages. In this chapter, the MCRM circuit calibration process has been described in detail.

The Internet has served as an effective channel for companies to build and manage relationships with customers. The mobile channel, emerging from the convergence of wireless communications and the mobile Web, promises to deliver additional support to meet consumer needs. This chapter examines features of customer relationship management (CRM) as implemented on the Internet (eCRM) and the mobile channel (mCRM) from the customer’s perspective. It further explores how companies can better coordinate their CRM strategies between these two channels to support e-commerce customers. We propose an analytical framework to examine the current eCRM and mCRM practice in terms of customer loyalty, branding, customer satisfaction, customization, and trust. These five factors affect customer acquisition, sales and services, and customer retention. A checklist was developed to guide the evaluation of CRM practice for e-commerce sites. Several examples and research directions are discussed in the chapter.

This research is focused on the way Customer Relationship Management (CRM) systems are used, on a European level, by recruiting companies to assist candidates in finding a satisfactory job. A framework is presented to identify how CRM 2.0 and mCRM (mobile CRM) can help candidates to find jobs in a personalized way. A set of four hypotheses have been defined. To gain a better understanding of these CRM systems, the methodology used in the exploratory study was quantitative, employing a non-probabilistic sampling technique, with 35 recruiting agencies being studied. Results showed that the use of software in recruiting agencies is quite common and that CRM 2.0 is present in the vast majority of the studied companies. When it comes to mobile CRM, there's still much to be explored in this channel, as agencies focus their resources on Web 2.0, leaving this channel's great potential of mobile CRM unused.

Dimensionar o total populacional em redes cuja estrutura é composta por uma população rara e agrupada não é uma tarefa trivial. Neste cenário, é usual sobrepor uma grade à região na qual a população está contida e selecionar células dessa grade. Um framework chamado de Método 2-Camadas e 2-Estimadores - M2C2E foi proposto e ele implementa o Método de Captura e Recaptura Múltipla - MCRM na camada 1 para gerar as estimativas dentro da célula a serem usadas como entrada para a Amostragem Adaptativa por Conglomerados - AAC que é a camada 2 na qual estima-se o total populacional na grade. Os estudos com dados sintéticos e com dados reais revelam que o M2C2E fornece estimativas relevantes em relação à AAC e ao MCRM.
