Повний текст джерелаChronic obstructive pulmonary disease (COPD) - slowly progressive airway obstruction resulting from abnormal inflammatory response to lung damaging particles of inhaled air or gas. It is a major cause of high morbidity and moratlity worldwide. Patients with COPD has lung injury, but determined increased blood levels of inflammatory markers shows and systemic inflammation. Neutrophils are the key inflammatory cells both in local and systemic inflammatory process. COPD is heterogeneous disease and in order to understand a variety of pathogenic mechanisms it is necessary to evaluate local and systemic inflammation combination characteristics and principles. It is also important to investigate if airway mucus influence blood neutrophils activity. The aim of this study was to evaluate both chemotactic and phagocytic activity and reactive oxygen species (ROS) production in peripheral blood neutrophils during COPD. Blood neutrophils chemotaxis was measured by activation of the interleukin-8 (IL-8), induced sputum and induced sputum and IL-8 different concentration (10, 30 and 100 ng/ml) combination. Phagocytic activity and ROS generation of blood neutrophils was measured after neutrophils activation with serum osponized/non-osponized S. aureus bacteria, induced sputum and induced sputum combination with S. aureus bacteria. Повний текст джерелаThe Influence of the Substances Produced by Activated Neutrophils on the Migratory Activity of Mesenchymal Stem Cells Adult mesenchymal stem cells (MSCs) are multipotent stem cells which have the ability to differentiate into cells of multiple organs and systems such as bone, fat, cartilage, muscle, neurons, hepatocytes and insulin-producing cells. That is why they have generated a great deal of interest for their potential use in regenerative medicine and tissue engineering. MSCs are thought to be crucial for tissue regeneration, acting via migration into the site of injury, proliferating and differentiating into functional cells. The lesion of a tissue is always followed by the inflammatory reaction. It means that the MSCs migrate and access to the inflammatory environment. Neutrophil infiltration is a common pathological feature in acute inflammatory disorders. In the pathology sites they release oxygen species and their internal constituents, which are stored in different granules. The goal of this research work was to estimate the influence of the substances produced and released by activated neutrophils on the migratory activity of MSCs. Повний текст джерелаNeutrophils and macrophages play an important role in inflammatory process. Activated during chronic obstructive pulmonary disease (COPD), these cells synthesize a variety of biologically active compounds, which may further amplify both systemic and local airway inflammation. The features of biologically active compounds (cytokines, proteinases) released by neutrophils and macrophages during COPD and according to smoking status are unknown. Still, a direct influence of airway mucous from COPD patients on functional neutrophil features is unknown. The aim of this study was to evaluate the functional features of neutrophils and macrophages during chronic obstructive pulmonary disease and possible differences according to smoking status. An expression of matrix metalloproteinase-12 in airway mucous of COPD smokers and ex-smokers was evaluated. The functional features (chemotaxis, phagocytosis, production of reactive oxygen species) of biologically and chemically activated blood neutrophils and a modulator effect of airway mucous were also analysed. The neutrophils and macrophages of patients with chronic obstructive pulmonary disease were more activated than the cells of healthy individuals; smoking status has influenced some inflammatory features of neutrophils and macrophages in airway mucous and blood.

Повний текст джерелаThe purpose of the current study was to investigate the presence and localization of neutrophil granulocytes (Neutrophil Elastase+) and B-cells (CD20+) in patients with chronic periodontitis and/or peri-implantitis. These cells play an important role in the adaptive as well as the innate immune system, and can be considered as representative for responses of different character. Inflamed tissue consisting of epithelia and connective tissue was collected from twenty sites (thirteen with periodontitis and seven with peri-implantitis) on sixteen individuals undergoing surgical treatment. The biopsies were prepared immunohistochemically (Dako’s Two-Step Polymer Method) and analyzed in microscopes. On tissue-level a longer pocket epithelium and a greater inflammatory infiltrate in the apical portion was observed in peri-implantatis. The apical half of the peri-implant tissue generally indicated a more local inflammatory infiltrate in the contact area than in periodontitis, both concerning B-cells and neutrophil granulocytes. The level of vascularisation was also higher. B-cells were predominant in both conditions and had a general distribution in most cases. Neutrophil granulocytes occurred in a smaller extent than B-cells, above all in periodontitis.

Повний текст джерелаFor the first time, a complex comparative study was performed on the oxidative function of non-activated and activated neutrophils of peripheral blood from subject with differing functional activity of neutrophils (i.e. patients with periodontitis and healthy subjects). The study employed two very sensitive techniques – luminol-dependent and lucigenin-dependent chemiluminescence. For the first time, the study showed that the homeopathic medication Traumeel S had antioxidant effect in vitro, i.e. it significantly reduced superoxide anion levels generated by blood neutrophils in patients with periodontitis. The results of the study also complemented knowledge about the pathogenesis of periodontal inflammation. This finding may have a significant clinical implementation.

Resumo: A doença hepática crônica apresenta diversas causas, sendo caracterizada por diferentes graus de inflamação e necrose hepatocelular. A hepatite crônica pode ocorrer devido a vírus, reações autoimunes, drogas ou toxinas. Utilizando-se o modelo experimental de hepatopatia em coelhos induzido por CCU, o objetivo deste estudo foi investigar uma possível relação entre alguns parâmetros relacionados ao processo inflamatório (quimiotaxia de neutrófilos e análise total e diferencial dos leucócitos) e a lesão hepática. A extensão do dano hepático produzido pelo CCU foi investigada através da análise histológica dos fígados. Nossos resultados demonstraram que o tratamento com CCL4 induziu lesão hepática nos coelhos, conforme demonstrada pela análise histológica e pela elevação dos níveis séricos das enzimas ALT e AST. A análise histológica indicou a presença de hepatite após a 2a semana e de cirrose na 6a ou 8a semana de tratamento com CCL4. A resposta quimiotática in vitro dos neutrófilos verificada uma semana após a administração da droga, mostrou aumento entre a 2a e 6a semana de tratamento. Quando verificada 24 h após a droga, a quimiotaxia apresentou elevação a partir da 2ª até a 8a semana de tratamento. Neste mesmo período observou-se também aumento no número dos neutrófilos do sangue periférico. Levando-se em conta a verificação da presença de neutrófilos no fígado dos animais com cirrose, estes resultados 24 horas após a droga, sugerem que o aumento da migração in vitro dos neutrófilos e a elevação do número dos polimorfonucleares do sangue periférico podem estar relacionados à ativação e ao recrutamento destas células inflamatórias para o fígado, onde desempenham importante função no desenvolvimento do dano hepático
Abstract: The aim of this study was to investigate neutrophil chemotaxis during the induction of liver cirrhosis in rabbits. The study had three expérimentai arms: Group A (n=16) served as controls and received only normal rabbit chow. All animals were sacrificed at 16 weeks. Group B (n=19) was sacrificed 48 h after the chemotaxis assay, which was performed 24 h after CCL4 administration, at weeks 2, 4, 6 and 8. Group C (n=8) was followed by 16 weeks. The chemotaxis assay was performed 1 week after CCU administration. All animals were sacrificed at 16 weeks. The rabbits were treated with intragastric administration of CCU once weekly starting 14 days after addition of Phénobarbital to the drinking water (50mg/day). The CCU dose was adjusted weekly in order to obtain serun ALT and AST levels between 400 and 800 IU/I. Our results showed that increased ALT and AST levels were found in all of CCU-treated rabbits compared to the controls. In liver from animals sacrificed in 2, 4, 6 and 8 weeks of CCU treatment, hepatitis and fibrosis were already found at week two, and liver cirrhosis at week 6 and 8. Sixteen-week CCU treatment resulted in liver cirrhosis in 7 animals (87.5%). All animals with liver cirrhosis had an inflammatory infiltrate with neutrophils. Treatment with CCU did not interfered with total leukocyte count in blood from animals sacrificed in different periods, but a significant increase in polymorphonuclear cells between weeks 2 to 8 and a significant reduction in mononuclear leukocytes at week 4 were observed when compared with control animals. Neutrophil chemotaxis was also significantly increased from week 2 through week 8 compared with the control group. Animals that received CCU for 16 weeks present a significantly increased total leukocyte and polymorphonuclear number at week 10 when compared to controls. The mononuclear cell number was unchanged during the study period. A significant increase in neutrophil chemotaxis was observed from week 2 trough week 6. The presence of neutrophils in the liver of all animals with cirrhosis associated with an increase in polymorphonuclear cell chemotaxis during CCL4 administration in our experiment supports the view that this cell line has an important role in the development of toxic liver damage

Повний текст джерелаSyftet med detta arbete var att studera inflammatoriska effekter i mänskligt helblod och neutrofila granulocyter exponerade och stimulerade av nanopartiklar av gadoliniumoxid. Projektet utreder den toxiska potentialen hos nanopartiklar med olika kemiska och morfologiska egenskaper.
I experimenten undersöktes cellresponsen hos blodceller exponerade med ofunktionaliserade och funktionaliserade nanopartiklarna. Effekterna av funktionaliserade och ofunktionaliserade nanopartiklarna på aggregation och syreradikalproduktion i helblod och hos neutrofila granulocyter studerades med hjälp av lumi-aggregometri.
Studier har visat att varken ofunktionaliserade eller funktionaliserade nanopartiklarna ger aggregation i blodet. Syreradikalproduktionen ökar däremot. Resultaten av studier i helblod visar att stimulering med spädnings serier av funktionaliserade nanopartiklar ger mindre frisättning av syreradikaler än spädnings serier med ofunktionaliserade nanopartiklar. Detta bekräftas med studier av morfologiska skillnader i neutrofila granulocyter som var stimulerade med olika typer av nanopartiklar. Detta gjordes med hjälp av fluorescensmikroskopi. Resultaten från studierna tyder på att funktionaliserade nanopartiklar är mindre inflammatoriska än ofunktionaliserade nanopartiklar.
