This diploma thesis focuses on the optimization of bacterial cellulose production by Komagateibacter xylinus DSM 46604. The theoretical part of this thesis describes the properties of bacterial cellulose, its production and application possibilities. The experimental part aimed to assess the effect of different cultivation conditions on the production yields of bacterial cellulose. The effects of several cultivation strategies have been studied such as: (1) effect of acetate buffer used as a medium, (2) impact of oil added into the medium, (3) fed-batch cultivation, (4) variation of the volume of cultivation vessel and cultivation media and (5) aeration. In addition to the production of relatively thin samples, up to 100 µm was synthesized unique 3D structured bacterial cellulose in the form of cylinders, with a height up to 2 cm. The growth of bacterial cellulose in the form of cylinders was achieved by dynamically cultivating K. xylinus in combination with fed-batch approach.

In this thesis, optimization of production of lipolytic enzymes by submerzed cultivation of Bacillus subtilis (BS) was studied. Production of lipolytic enzymes was tested in three nutrient media, which differed mainly in main sources of carbon, respectively of nitrogen. The first medium contained mainly extract from calf brain and beef heart (BHIB), the second medium contained peptone and yeast extract (NB) and the third one contained peptone and yeast extract with the addition of 2% (w/v) glucose (NBG). The highest lipolytic activity (0.0784 Uml-1) was measured in NBG medium. Maximum of lipolytic activity was observed before the end of the exponential phase of BS growth in all the media. Temperature optimum in NBG medium was determined from 30 to 50 °C, pH optimum in the range of 5 to 11 and subsequently the temperature stability of lipolytic enzymes produced by the BS was estimated. The activity value was determined spectrometrically using p-nitrophenyllaurate as a substrate. Produced lipolytic enzymes showed maximum activity at 37 °C in the alkaline pH of 8.0. Measurement of temperature stability showed that lipolytic enzymes are relatively thermostable enzymes retaining 100 % of the activity even after 1 hour of cultivation at 30 - 50 °C. The presence of 1% (w/v) olive oil in medium NBG caused a decrease in lipolytic activity by 65 % as well as in pH from 6.5 to 5.4 after 14 days of cultivation. After substitution of glucose by fructose in medium NBG, lipolytic activity showed comparable values during the first week of cultivation. On the other hand, the decrease of lipolytic activity by 29 % in the medium with fructose was observed after 14 days of cultivation. A procedure for the identification of lipolytic enzymes of BS by peptide massfingerprinting was developed to understand the potential of synthetic polyester - poly(e-caprolactone) as a lipase inductor. Degradation study of commercial polyester poly(e-caprolactone) was carried out by submerged cultivation of Bacillus subtilis in NBG medium at initial pH 7.0 and 30 °C for 14 days. PCL (Mn = 10,000, Mw = 14 000) was studied in the form of films (1.0 x 1.0 cm), which were prepared by melt-pressing, rapid cooling of the melt to 4 °C and evaporation of the solvent from 2 % dichlormethane solution. The evaluation of the films shown occurrence of weight loss (7.8 - 17.0 wt.%) together with the formation of numerous holes and cracks in the sample surface in relation to the method of the films preparation. Lipolytic activity values increased by 9 - 17 % in the degradation media compared to control samples. Densitometric monitoring showed also higher increase in cell mass in the degradation medium compared with control samples. Based on the results obtained, the degradation process induced by BS could be suggested.

V této diplomové práci je popsána přechodná transfekce buněk 293 HEK adaptovaných na růst při suspenzní kultivaci bez přítomnosti séra za použití polyethyleniminů (PEI). Buňky byly transfekovány plasmidem pcDNA5/SEAP, který exprimuje sekretovanou formu lidské placentální alkalické fosfatázy. K porovnání účinnosti jednotlivých transfekcí byla měřena koncentrace exprimované fosfatázy v buněčném supernatantu. Cílem této práce bylo optimalizovat různé faktory ovlivňující účinnost transfekcí s důrazem na nalezení optimálního poměru DNA:PEI.

Diploma thesis deals with use of Zymomonas mobilis for the production of bioethanol from waste paper. There were used three kinds of substrate (cardboard, drawing and office paper) to optimize of bioethanol production. Individual papers were subjected to the same pre treatment, namely a milling, a combination of microwave irradiation and NaOH, a combination of microwave irradiation and H2SO4 and combination microwave irradiation, H2SO4 and NaOH. The substrates were decomposed by enzymatic hydrolysis after pre treatment to evaluate the best pre-treatment. Simultaneous saccharification and fermentation was carried out for each substrate (with two of the best pre-treatment). The samples were taken during the hydrolysis and the simultaneous saccharification and fermentation, and were determined by HPLC. Growth curves of Zymomonas mobilis were constructed, as the most appropriate for SSF was chosen temperature of 40 ° C in which the exponential phase took place at the time of 6 15 hours. During hydrolysis was monitored glucose concentration in the solution. The maximum concentration of glucose was in the cardboard (microwaves + H2SO4 + NaOH) 16.46 gdm-3, a drawing (microwaves + H2SO4 + NaOH) 31.78 gdm-3, and office paper (microwaves + H2SO4) 25.04 gdm-3. The concentration of ethanol for SSF was highest in the same cases as in the hydrolysis. The cardboard was the maximum concentration of bio ethanol 9.5 gdm-3, for the drawing 16.1 gdm-3 and for the office paper 12.13 gdm-3.

In the diploma thesis are discussed the process of enzymatic hydrolysis of waste paper as a source for the production of bioethanol by bacteria Zymomonas mobilis. In the theoretical part summarize basic information about particular methods of hydrolysis, about paper used as a raw material for enzymatic hydrolysis, about possibilities of the fermentative production of bioethanol focusing on the method of simultaneous saccharification and fermentation comparison with enzymatic hydrolysis and fermentation. Suitable microorganisms for ethanolic fermentation and simultaneous saccharification and fermentation and their advantages and disadvantages, are further discussed in this part as well. The theoretical part ends with the suggestion of the technological process for production of bioetanol. It covers all necessary steps from the input of raw material to the separation of produced ethanol. In the experimental part various parameters of hydrolysis, fermentation and simultaneous saccharification and fermentation were optimized using enzymes from Novozymes® company and the Zymomonas mobilis CCM2770 and Zymomonas mobilis LMG457 bacterium. The conversion rate of paper cellulose to gluckose and production of ethanol were observed by HPLC/RI method. Type of buffer, quantity of cells, enzyme and substrate were optimized in order to maximize the efficiency of the process. All experiments were performed on paper containing high amount of cellulose and for comparison on standard medium which contains gluckose. The highest yields was achieved with the use of Novozymes® Cellulosic ethanol enzyme Kit. The strain Zymomonas mobilis LMG457 has demonstrated as a better producer.

Several yeast strains and microalgae were selected for this diploma thesis. -glucans, lipids, carotenoids, ergosterol and coenzyme Q were determined in selected producers, and the cultivation conditions for yeast strains were optimized to gain enhanced production of -glucans. Microalgae cultivations were carried out according to the instructions of the Collection of Autotrophic organisms (CCALA). Selected microalge strains include Desmodesmus acutus, Dunaliella salina, Arthrospira maxima and Cyanothece sp. Selected yeast species include Rhodotorula glutinis, Cystofilobasidium macerans and Sporidiobolus metaroseus. Edible yeast Saccharomyces cerevisiae was cultivated to compare with other yeast strains because of it's verified production of -glucans. -glucans were then determined by the enzymatic kit K-YBGL Megazyme, carotenoids, ergosterol and coenzyme Q were analyzed by HPLC/PDA and fatty acids were analyzed by GC/FID. The best producer of yeast -glucans was R. glutinis and S. metaroseus, and the best conditions for the production of -glucans and other metabolites was the C/N ratio of 70. Within the microalgae species, only -glucan production was observed, the best producer was D. acutus.

The objective of this thesis is to present the basics of production scheduling problems for workstations and to create an application of dynamic model. This application should help the UNIKOV company to produce similar schedules for their own operations. The theoretical part provides a brief description of terms and characteristics of the scheduling problems. Detailed characteristics are provided on the open shop problem - the task applied on the real world example. The paper then follows with a practical part, covering an explanation of the heuristics model written in VBA for MS Excel, allowing dynamic expansion and management of the production.

This diploma thesis deals with the risk associated with fish production. Selected risks are further taken into account. The risks are further described and addressed using the FMEA matrix. The thesis contains a mathematical model that solves the optimal population of the pond in order to maximize profits. The model includes fixed input parameters, which includes the price of hatchery material and the market price of fish. Furthermore, the model contains random input parameters that are limited by the intervals and includes the growth coefficient and the size of the population in the pond. The optimization model is based on simulated data, which are based on real data and statistically determined estimates. The used data are processed into tables in MS Excel for better visualization. GAMS software is used for software implementation of the model.

For that I will use suitable communication mix based on current experience since the launch of the app which was in September 2017. The communication of the application itself to all interest groups in managed by marketing agency in cooperation with marketing management of the certain shopping center. Though it seems on the first sight that it is mainly B-2-B market, main interest focus is on the final customer satisfaction and that is B-2-C market. There is a detailed analysis of the Prague shopping centers market with emphasis on another mobile applications of the same purpose made in the Thesis. Then there are the steps how to make the marketing communication of the application more effective a so make the awareness of the application better suggested from the results of the analytical part .

Veškeré investice do strukturovaných produktů jsou porovnávány s výnosem u indexu DJ Euro Stoxx 50 (Total Return).

This thesis focuses on the storage location assignment problem using modern meta-heuristic techniques combined with realistic simulation. A graphical tool implemented as part of this work is capable of warehouse model creation, generation of synthetic customer orders, optimization of product allocation using state of the art techniques, extensive warehouse simulation, and a pathfinder capable of finding the shortest path for orders going through the system. The work presents the comparison between different approaches based on many parameters to reach the most efficient allocation of products to warehouse slots. The author conducted tests on an experimental warehouse featuring almost twice the throughput -- 57%. The benefit of this work is a possibility to create model of an already built warehouse and its simulation and optimization, driving impact on the throughput of the warehouse, saving the user's resources, or helping him in planning and bottle-neck identification. Furthermore, this thesis introduces a new approach to warehouse optimization and new optimization criteria.

The purpose of this thesis is to highlight the importance of graphic design and its visual effects in day-to-day life, notice new methods, justify relevant investments to advertisement as well as mention continuously rising demands made on professional graphic designers. The theoretical part of the thesis is based on the research article, which subject is the extent of consumers' product understanding of products being sold online. The research concludes that it is possible to choose the appropriate presentation format according to task complexity (product features) in terms of graphical and multimedia means of online presentation, such as static pictures with text captions, videos with or without narration and Virtual Product Experiences (VPE ). This is followed by the definitions of communication process and relevant standards related to web design, concluding with the description and interpretation of the most interesting and useful theoretical foundations. The main body is about implementing the theoretical findings in the real world business case including advertising campaign in a project having the purpose to give a real economic/business value. All the project phases are described from finding the business partner to the launch of brand new web service and its propagation. The new web page and other propagation materials were made in order to attract selected companies.

The goal of this thesis is to optimize current marketing mixes in sports facilities. As such, this thesis will suggest appropriate optimization strategies for these mixes and recommend methods for improving marketing communication practices and service portfolios, in order that facilities may heighten their competitiveness, increase facility attendance, and enhance client satisfaction. The theoretical section of this thesis analyses marketing mixes, services, sports product, CRM, CEM, and facility environment. The practical component first presents the facility itself: a squash centre, where the analysis was done. An analysis and assessment of marketing activities of the squash centre was carried out afterwards. Quantitative forms of research, especially interviews, were used. Informal conversations were also held with the management of the squash centre. The thesis results in some recommendations for the squash centre which are applicable for sports facilities in general.

This Master's thesis deals with the optimization of the material flow between two areas of the production plant of the selected company. At the beginning of the thesis there is shown theoretical framework which is needed for subsequent analysis and determination of the proposals to improvement. In this part there is pointed out to the understanding of the process and the various concepts related to the process management. Furthermore, for the purposes of the application part there are selected and analyzed specific tools of lean manufacturing. These include for example the spaghetti diagram, the product / process matrix and the value stream mapping as the essential tool of this thesis. All this is complemented by ABC analysis. After that these methods are also applied in the practical part for solving the current project in the selected company. At first in solving the project there is dealt with the determination of the most problematic material flow between two areas of production plant. Then there is suggested the optimization solutions and all of this is also complemented by the economic evaluation of the whole project.

The most difficult part of water treatment is to ensure that water will be delivered to consumer in unchanged form and hygienic unexceptionable. To hygienic safety is using doses of chlor, whiches are deadly for organisms and inhibite their proliferation in pipeline. Modern way how to clear water from organism is called chloramination. By the help of ammonium salts chlor is released in sequence. So large single-shot doses of chlor aren´t necessary. Due the big dose was THM and another dangerous compounds formed.

Thesis deals with design process optimization at Chute Corporation -- american startup that provides SaaS solutions for discovery, gathering, managing, publishing and analysis of multimedial user-generated content from social networks. The goal is to identify and analyze the processes, roles and tools of product design and propose an optimization. Thesis has two parts. First part describes the theoretical background for later application. Mainly a brief history of IT product design, design taxonomy and design workflow best practices are described. The other part provides a brief description of the company, especially its position on the market, business model, organizational structure, product portfolio, position of product team and also identifies the main processes, roles and tools that surround product design. The final part describes all optimized processes, roles and tools and summarizes all the facts as a set of recommendations. The subject of my thesis is the optimization of selected business processes in the specific company. These are processes, which optimization will lead to save time, money and will generate additional income and increase the current level of profit significantly. In my thesis, I proposed several indicators of metrics and describe new information systems that will be part of optimized processes. In the first part I present the theoretical aspects and the transition from functional to process management, modeling based on different methodologies, standards and measuring the performance of individual processes. In the next chapter I am dealing with the practical application of theoretical knowledge on a specific company. First I briefly introduce this company, define its position in the market, and going through complex of strategic analysis. Next I give solution how to find out the problematic aspects of the most critical processes and then I suggest optimization. An important part of this thesis are suggested metrics and indicators for these metrics, which are monitored and evaluate targets of each individual process and thus his successful implementation. After incorporating the proposed optimization, the company has significantly shifted not only in monitored metrics, but will also increase its value. These ideas I will summarize in the end of the work.

The use of banking products is an integral part of business. For this reason, the theoretical part of the work focuses on the general description. It also highlights the benefits of optimization in this sphere. It also indicates the differences between Czech and European banks, which are most obvious in the bank charges. They discussed the impact of financial crisis, affecting in particular the availability of credit. Practical part of the work focuses on the comparison of products, namely the current accounts of domestic and foreign bank -- ČSOB and Deutsche Bank. The result of this analysis is greater partiality for the establishment of current account at a bank in the Czech Republic. In addition, bank products are analyzed, leading joint-stock company CZ LOKO, which are mentioned experience with their use. Subsequently, the company CZ LOKO recommended a new current account, which is based on the comparison of bids for five lesser-known banks. From this analysis came to establish a new account at a branch of Austrian Oberbank AG, based in the Czech Republic. Finally, carry out a survey on the use of banking products, which demonstrated that all of today's entrepreneurs, to a lesser or greater extent, using bank's services.

Pond and intensive aquaculture of percids relies mainly on juveniles supplied by semi-artificial and artificial reproduction. Fish hatcheries do not always provide optimal conditions for percid reproduction. This project was undertaken to optimize reproduction methods of Eurasian perch (Perca fluviatilis L.) and pikeperch (Sander lucioperca L.). Reproduction parameters were compared in farmed and pond-reared perch broodstock without hormone treatment. Spawning was successful without hormone induction at 92.3% and 76.9% in farmed and pond-reared perch, respectively. Significant differences in morphological characteristics (gonadosomatic, hepatosomatic and viscerosomatic indices), fecundity, hatching, and post-spawning mortality were detected. All results, with the exception of spawning success and post-spawning mortality, were superior in pond reared broodstock. Further studies are required to optimize protocols for using better artificial food in farmed perch and decreased stress and post-spawning mortalities in pond reared perch broodstock. The possibilities of using clove oil, Propiscin, or 2-phenoxyethanol as an alternative to tricaine methane sulphonate (MS222) as anaesthetic, particularly with respect to reducing fish stress, were investigated. The haematological and biochemical profiles of pikeperch anaesthetized with clove oil (33 mg l?1), Propiscin (1.5 ml l?1), 2-phenoxyethanol (0.3 ml l?1), and MS222 (150 mg l?1) were determined. Each tested group was divided into two subgroups; the first subgroup was sampled 10 min after application and the second 24 h post-application. Clove oil was shown to be associated with the lowest effects in pikeperch, and therefore could be recommended as an alternative to MS222, while Propiscin and 2-phenoxyethanol were shown to be unsuitable for use with pikeperch. In Eurasian perch the use of semi-artificial reproduction for mass production of embryos was shown to be simpler and more effective. With mass incubation, 1 134 000 embryos (59% of total production) were obtained with a semi-artificial spawning regime and 796 000 embryos (41% of total production) with artificial spawning. Based on these results, we can recommend the semi-artificial spawning for production of perch embryos. Differing doses of two commonly used hormone preparations: human chorionic gonadotropin (hCG) and mammalian GnRHa (Supergestran) were tested for induction of ovulation in pikeperch. A broad range of effective doses is used in aquaculture of pikeperch. The minimal effective dose based on ovulation success is still missing. Hormone therapy applied to excess can result a detrimental impacts on broodstock fecundity and quality of eggs. Results indicated that a single injection of hCG or Supergestran could be used for successful ovulation, while no spawning was observed in a control group treated by saline solution. Among hormone treated groups, the best results were observed in groups treated with hCG at 500 and 750 IU kg-1 BW and in groups treated with mGnRHa at 25 ?g kg-1 BW. The sperm of pikeperch and Eurasian perch show similar morphology and fine structure, with the exception of differences in arrangement of midpiece and centrioles and the presence of a fin-like structure on the flagellum in pikeperch. Our results suggest use of an activation medium containing Ca2+ for activation of short-term stored sperm. Efficacy of five concentrations of Alcalase enzyme for elimination of egg stickiness compared to a milk/talc suspension was determined. Results indicated that 0.5-2.0 ml l-1 of Alcalase for 2 min may increase pikeperch egg hatching success (80-85%) compared to the milk-talc suspension. The highest hatching rate (85.4%) was observed with 1.5 ml l-1 of Alcalase. The application of Alcalase successfully eliminated pikeperch egg stickiness in 2 min compared to 60 min with traditional milk/clay/talc methods.

Prof. Veronika Nováková, Ph.D. Title of Thesis: Optimization of methods for determination of singlet oxygen production and fluorescence emission of azaphthalocyanine derivatives Photodynamic therapy (PDT) with a singlet oxygen as an essential agent is believed to be an alternative way of cancer treatment or treatment of some cutaneous diseases. The principle of PDT is based on excitation of a photosensitizer by light absorption, followed by transfer of energy to tissue oxygen (3 O2) forming cytotoxic singlet oxygen (1 O2). The efficiency by which photosensitizer transforms absorbed energy to singlet oxygen is characterized by singlet oxygen quantum yields (ΦΔ). The aim of this thesis was to develop and optimize absolute method for determination of ΦΔ. In comparison to a relative method, no reference is needed in this case, which enables accurate results with lower error. Verification of the new method was performed in N,N- dimethylformamide with a zinc phthalocyanine as a model photosensitizer because of its well-known ΦΔ and with 1,3-diphenylisobenzofuran as a chemical quencher of 1 O2. The aim of the thesis was to propose possible measures to optimize existing waste management system in Hermanuv Mestec based on the description and analysis of the current situation. In the theoretical sphere the thesis works with basic terms of the current Czech legislative norms and regulations. The geographical position, city planning solution and demographic evolution were characterized. The whole system ensuring waste management in the investigated area was described and its compositions were specified, including their production practices and quantification of economic costs. To analyze the issue of city's waste management system the SWOT analysis and the method of comparison were employed. Recommendations, based on the conducted analysis and findings, were presented for optimizing current system of the organizational sphere, personnel management and financial parts.

In the presented work we study xing of term deposit margins on Czech deposit market. Financial crisis which in fluenced almost each financial system hit also Czech banks. It resulted in almost no trading on interbank market and banks started to hoard liquidity from primary deposits. At the beginning of the thesis we introduce origin and expansion of liquidity crisis. Next we introduce term deposits and their basic properties on Czech deposit market. Then work is focused on term deposits and on xing their margins in dependence on several factors. Next part introduce model for optimal margin in dependence especially on demand for term deposits and competitive margin. There is also description of crisis impacts on suggested model. Results are built on term deposit of one of the largest banks on Czech deposit market. Data cover period from 2006 to 2008.

Pulse (fluid) pressure modulator was constructed for comprehensive gas chromatography. Optimal conditions of its connection to the gas chromatograph with a flame ionization detector were found for the analysis of selected mixture of volatile solvents. Pressure interdependences in the system and relations between dimensions of modulator capillaries and columns have been evaluated with respect to the length of the modulation period and pulse duration. Two non-polar columns (15 m × 0.25 mm DB-5MS, 10 m × 0.15 mm CP-Sil 5 CB) of the same stationary phase were tested to compare the separation efficiency in the first dimension and the DB-5MS column was selected. The Supelcowax 10 columns of the lengths 1, 2 and 3 m were compared in the second dimension. It was found that using of 3 m length column lead to the extension of analysis time, however the increase in separation efficiency in the second dimension was very evident. The experimental measurements have shown that the selected parameters for the evaluation of the separation efficiency (the peak capacity and total resolution product) were inappropriate. Gasoline, kerosene and diesel fuel were analyzed as examples of real samples. Amount of added ethanol as a biofuel additive was determined in the gasoline sample. My thesis which deals with optimization of durable meat products processing. Research has found that during drying a durable meat product, there has been a gradual water loss and impairment of aw below 0.85. Fat and salt levels were rising. Measured values show that an optimum time to dry durable meat products is 42 days and it should be done gradually. Measurements have shown that the results are strongly influenced by random variations. Only the difference in the average proportion of fat between 7th and 14th day wasn´t proved at the 5% level of significance.

These methods were applied on samples taken from four varieties of hop (Saaz, Brewer, Premiant and Agnus) in four locations (Blšany, Běsno, Nesuchyně and Strojetice) of žatec hop-growing area in the period from 18. 8. 2012 to 4. 9. 2012 during the hop harvest. Furthermore, the influence of the drying procedures on the hops was monitored, it means, difference in the content and composition of bitter acids between green and dry hops and the comparison between the band and chamber drying rooms.

DF is a non-steroidal anti-inflammatory drug with analgesic, antipyretic and anti-inflammatory effects. The degradation product DPI has been produced in formulations after a long-term storage, especially after exposure to light or heat. The method development was based on a method created by the Pharmaceutical Faculty of Charles University for the determination of DF, its degradation product and preservatives in topical emulgel and the method was optimised for determination of DF and DPI in tablets. Ondřej Zemek Organophosphorus derivatives for "click-chemistry" In this diploma thesis two new macrocyclic ligands were synthesized. They have DO3A moiety where metal (lanthanide) ion can be coordinated and terminal acetylenic or azide group connected through the phosphinic acid. Both terminal acetylenic and azide group should serve for connecting to another molecules or partitions by mean of "click chemistry" i.e. [1,3] copperI catalyzed dipolar cycloaddition between azide and acetylene. In this thesis two ligands, its complexes with some lanthanides as well as its precursors were prepared and characterized. Gd3+ complexes will be further used and studied for potential MRI contrast agents applications.
