ABSTRACT During complementary chromatic adaptation (CCA), cyanobacterial light harvesting structures called phycobilisomes are restructured in response to ambient light quality shifts. Transcription of genes encoding components of the phycobilisome is differentially regulated during this process: red light activates cpcB2A2, whereas green light coordinately activates the cpeCDE and cpeBA operons. Three signal transduction components that regulate CCA have been isolated to date: a sensor-photoreceptor (RcaE) and two response regulators (RcaF and RcaC). Mutations in the genes encoding these components affect the accumulation of both cpcB2A2 and cpeBA gene products. We have isolated and characterized a new pigmentation mutant called Turquoise 1. We demonstrate that this mutant phenotype is due to a dramatic decrease in cpeBA transcript abundance and results from a lesion in the cpeR gene. However, in this mutant cpeCDE RNA levels remain near those found in wild-type cells. Our results show that the coordinate regulation of cpeBA and cpeCDE by green light can be uncoupled by the loss of CpeR, and we furnish the first genetic evidence that different regulatory mechanisms control these two operons. Sequence analysis of CpeR reveals that it shares limited sequence similarity to members of the PP2C class of protein serine/threonine phosphatases. We also demonstrate that cpeBA and cpeCDE retain light quality responsiveness in a mutant lacking the RcaE photoreceptor. This provides compelling evidence for the partial control of CCA through an as-yet-uncharacterized second light quality sensing system.

Photosynthetic organisms adapt to environmental fluctuations of light and nutrient availability. Iron is critical for photosynthetic organismal growth, as many cellular processes depend upon iron cofactors. Whereas low iron levels can have deleterious effects, excess iron can lead to damage, as iron is a reactive metal that can result in the production of damaging radicals. Therefore, organisms regulate cellular iron levels to maintain optimal iron homeostasis. In particular, iron is an essential factor for the function of photosystems associated with photosynthetic light-harvesting complexes. Photosynthetic organisms, including cyanobacteria, generally respond to iron deficiency by reduced growth, degradation of non-essential proteins and in some cases alterations of cellular morphology. In response to fluctuations in ambient light quality, the cyanobacterium Fremyella diplosiphon undergoes complementary chromatic adaptation (CCA). During CCA, phycobiliprotein composition of light-harvesting antennae is altered in response to green light (GL) and red light (RL) for efficient utilization of light energy for photosynthesis. We observed light-regulated responses to iron limitation in F. diplosiphon. RL-grown cells exhibited significant reductions in growth and pigment levels, and alterations in iron-associated proteins, which impact the accumulation of reactive oxygen species under iron-limiting conditions, whereas GL-grown cells exhibited partial resistance to iron limitation. We investigated the roles of known CCA regulators RcaE, RcaF and RcaC in this light-dependent iron-acclimation response. Through comparative analyses of wild-type and CCA mutant strains, we determined that photoreceptor RcaE has a central role in light-induced oxidative stress associated with iron limitation, and impacts light-regulated iron-acclimation responses, physiologically and morphologically.

This article addresses the Terezita Romo Papers, one of a handful of archival collections of the Royal Chicano Air Force—a large collective of young, mostly immigrant or first-generation Mexican American artists and activists who produced countless community events and art projects and programs in Sacramento, California during the second half of the twentieth century. While membership of the Royal Chicano Air Force (RCAF) and its activities are hard to calculate, its history has been shaped by a tendency towards iconization of the group’s male founders in archival description. Specifically, where collections are described to highlight the unique contributions of individuals, it is difficult to retain and promote the collective voice of action which made so many of these movements successful. Using the papers of Tere Romo, one member of the RCAF, this paper looks at how the archives of the RCAF have tended towards iconization—overshadowing the contributions of its female members—and explores ways in which archivists can reconsider the language of archives when processing and describing materials documenting collective action in American history. Penelitian ini dilakukan agar mengetahui ukuran kemajuan teknologi website LinkedIn yang merupakan salah satu jejaring sosial untuk kepentingan korporasi. LinkedIn merupakan tempat bagi sarjana untuk menemukan pekerjaan terbaik sesuai pendidikan yang telah ditekuni. Dalam penelitian ini digunakan teori dari Allan Albrecht yang tetap dikembangkan oleh International Function Point User Goup (IFPUG) dan akan menggunakan metode pengukuran terkenal yaitu Function Point Analysis (FPA) sebagai cara untuk menghitung serta memperkirakan hasil pengukuran dari website LinkedIn. Pengukuran ini dilakukan sebagai bentuk partisipasi pengembangan pada website LinkedIn yang merupakan sebuah website besar dan sangat banyak diminati oleh pengguna internet. Di hasil akhir penelitian diharapkan dapat mengetahui angka kemudahan akses pemakaian website LinkedIn terhadap pengguna dengan menggunakan rumus Crude Function Points (CFP), mengetahui bagaimana dan seberapa besar kompleksitas dari website LinkedIn dengan menggunakan rumus Relative Complexity Adjustment Factor (RCAF), yang kemudian hasil dari perhitungan rumus CFP dan RCAF akan langsung diimplementasikan pada rumus pengukuran FPA.

The protein phosphatase calcineurin (Cn), together with its substrates, the transcription factors NFATc, play an essential role in several physiological processes of vertebrates, such as the immune response, angiogenesis, morphogenesis of the heart valves and neural and muscle development. Concretely in the immune system, the Cn-NFATc signaling pathway is crucial in the generation of T cell repertory in the thymus and in several processes of mature T lymphocytes, as T cell activation and survival. The members of the Regulators of Calcineurin (RCAN) family interact with Cn and compete with NFATc transcription factors for binding to Cn. RCANs can act either promoting or suppressing Cn activity towards its substrates. Due to their capacity to negatively modulate Cn- NFATc signaling, RCAN proteins are considered as potential immunosuppressant proteins. The aim of the present work was to deepen in the physiological processes that regulate the expression of RCAN and to evaluate the effect of their overexpression in the immune system. To this end, we have analyzed the evolution of RCAN genes and the genomic structure of the three human members of the RCAN gene family and, more specifically, the regulation of RCAN3 gene expression. Furthermore, we have evaluated the expression pattern of the different mouse Rcan genes in lymphoid tissues, and we have analyzed the in vivo relevance of RCAN1 and RCAN3 overexpression in T cell development and function. In this context, RCAN1 and RCAN3 overexpression enhances positive selection of thymocytes. Moreover, RCAN1 overexpression increases the generation of nTregs in thymus and seem to influence the effector/memory T cells proportion in homeostasis. Furthermore, RCAN proteins appear to modulate other TCR signaling pathways different than Cn-NFATc. Therefore, RCAN proteins are important players of in vivo T cell development, differentiation and activation.
La fosfatasa calcineurina (Cn), junto con sus substratos, los factores de transcripción NFATc, juega un papel esencial en varios procesos fisiológicos de vertebrados, como la respuesta inmune, la angiogénesis, la morfogénesis de las válvulas cardíacas o el desarrollo neuronal y muscular. Concretamente en el sistema inmune, la vía de señalización celular Cn-NFATc es crucial en la generación del repertorio de células T en el timo y en varios procesos de la función de los linfocitos T maduros, como la activación y la supervivencia de las células T. Los miembros de la familia de Reguladores de Calcineurina (RCAN) interaccionan con Cn y compiten con los factores de transcripción NFATc por su unión a Cn. Las RCAN pueden actuar promoviendo o inhibiendo la vía de Cn sobre sus sustratos. Con este fin, hemos analizado la evolución de los genes RCAN y la estructura genómica de los tres miembros RCAN en humanos y, más específicamente, la regulación de la expresión génica de RCAN3. Asimismo, hemos examinado el patrón de expresión de los diferentes genes Rcan de ratón en tejidos linfoides y hemos analizado la relevancia in vivo de la sobrexpresión de RCAN1 y RCAN3 en el desarrollo y la función de las células T. En este contexto, la sobrexpresión de RCAN1 y de RCAN3 induce la selección positiva de timocitos. Además, la sobrexpresión de RCAN1 aumenta la generación de células nTregs en timo y parece influir en la proporción de células T efectoras/memoria en homeostasis. Asimismo, las proteínas RCAN parecen modular otras vías de señalización dependientes del TCR diferentes a la vía Cn- NFATc. Por tanto, las proteínas RCAN son agentes importantes en el desarrollo, la diferenciación y la activación de células T.

Devido à grande concorrência do mercado o controle de custos passa a ser peça fundamental no auxílio à tomada de decisão quanto à redução dos custos, visando uma lucratividade maior por parte das empresas. Essa dissertação trata sobre um novo modelo de custos, o RCA – Resource Consumption Accounting, que ainda não foi adotado por nenhuma empresa. Esse novo método de custeio foi aplicado como modelo piloto em uma empresa Americana onde os resultados foram favoráveis a ele em relação aos modelos existentes. Muitos autores defendem o RCA como sendo um grande aliado aos gestores e ao setor financeiro por ser de fácil entendimento, apurar a capacidade ociosa e principalmente por apurar os custos com maior precisão em relação aos métodos mais utilizados atualmente. O RCA deve superar ao método ABC – Activity Based Costing, pois ele possui algumas vantagens em relação a este, por exemplo, como tratar separadamente os custos fixos dos variáveis, evitando assim rateios arbitrários e gerando uma apuração dos custos dos produtos e das atividades com maior precisão, além de apurar a capacidade ociosa, auxiliando assim os gestores na tomada de decisão. O RCA, assim como o ABC, é caro de ser implantado, pois precisa ser utilizado junto com sistemas integrados, sendo a sua principal desvantagem.
Due to market competition, the cost control becomes a critical tool to assist managers in taking decisions pursuing the cost reduction improving the companies profitability. This dissertation is about a new cost management system, the RCA - Resource Consumption Accounting, which has not been adopted yet by any company. This new cost management system was applied as a pilot model for an American company and the results were favorable to him. Many authors support RCA as a great ally to managers and financial managers because it is easy to understand, work with embedded systems, and determine the idle capacity mainly determine costs with greater accuracy compared with the methods currently used. The RCA must overcome to ABC - Activity Based Costing, because presents some advantages over this, for example, treat separately the fixed costs of the variable ones, avoiding arbitrary apportionments and generating a calculation of the costs of products and activities more accurately, also determine the idle capacity, thereby assisting managers in taking decision. The RCA, as ABC, is expensive to deploy because it must be used with integrated systems, being its main disadvantage.

In this study a new wind gust estimate (WGE) method proposed by Brasseur (2001) is implemented in a limited area climate model (RCA, Rossby Centre regional Atmospheric model). The WGE method assumes that wind gusts develop when air parcels higher up in the boundary layer deflect down to the surface by turbulent eddies. The method also gives an interval of possible gusts by estimating an upper and lower bound of a bounding interval. Two separate storms (December 3-4, 1999 and January 8-9, 2005) and a three month period (November 1, 2004 - January 31, 2005) are simulated with RCA. The results are compared to direct observations and to gridded analysis (MESAN). The result is highly dependent on how well the meteorological fields are represented in RCA. Since the storm of December 1999 was not well captured by RCA, the wind gusts were consequently not correctly estimated. The storm of January 2005 was well captured by the RCA and the wind gusts relatively well described. Both the storm of January 2005 and the simulation over a three month period give rather good estimated gusts over sea areas, while over land there is an obvious overestimation of the calculated gusts. A correction to the estimated gust is necessary in order to make the parameterisation useful. Such a correction is tested in this study. It shows significant improvement over most land areas and also gives a certain underestimation in other areas.
Sammanfattning av ”Beräkning av byiga vindar i RCA” En ny metod (WGE-metoden) för att bestämma byvindar har i den här studien implementerats i en regional klimatmodell (RCA, Rossby Centre regional Atmospheric model). WGE-metoden utgår från att vindbyar genereras när luftpaket högre upp i gränsskiktet förs ner till marken av stora turbulenta virvlar. Ett intervall av möjliga byvindar erhålls genom att en övre och undre gräns för detta intervall beräknas. Två stormar (3-4 december 1999 och 8-9 januari 2005) och en tremånaders period (1 november 2004 – 31 januari 2005) har simulerats, och resultaten har jämförts med mätdata och MESAN. Resultatet är till stor del beroende av hur väl de meteorologiska fälten representeras av RCA. Stormen i december 1999 simulerades dåligt av RCA, vilket medförde att byvinden inte heller simulerades korrekt. Både stormen januari 2005 och simuleringen över tre månader ger en tämligen korrekt byvind över hav, samtidigt som man över land får kraftiga överskattningar av den beräknade byvinden. För att byvind-parametriseringen ska vara användbar krävs korrigeringar för att komma till rätta med överskattningen över land. The formal Root Cause Analysis (RCA) process consists of a structured approach to identifying the underlying factors that result in the unwanted outcomes of one or more typically negative past events (chronic or sporadic). The process helps to identify what systems or behaviors need to be modified to prevent recurrence of similar outcomes. The fundamental driver of the RCA is to eliminate the causes that lead to the unwanted event rather than addressing the symptoms of the event. This generally leads to multiple corrective actions rather than a single solution. Based on various root causes analyses conducted by a department in Enbridge Pipelines since 2006, various benefits provided by RCA exercises are highlighted in this paper. The findings were mainly derived from completed RCAs as well as RCA team member surveys. Firstly, the cost effectiveness of the RCA process was identified as a primary driver for this exercise. For example, one RCA involving a multidisciplinary team of 8 members, costing approximately $35,000, led to $1M in recommended solutions generating $16M in potential savings. In other words, each of the 350 hours spent on the RCA produced $8,571 in savings underlining the value creation to the company. Secondly, the RCA process combined with the power of the team reflection led to the identification of new issues which were causing other revenue losses to the company. Because these issues were not part of the defined scope of the original RCA, their root causes were not be immediately assessed but nevertheless their impact could easily be quantified and forwarded to Management for review. The third benefit highlighted by RCAs is more of an educational benefit to team members and subsequently the organization. Through cross functional examination of the issues and events, the team members became aware and educated on issues that would not have otherwise been apparent. Additionally, by working through the RCA methodology, the team members enhanced their problem solving abilities which benefit their everyday work. In conclusion, other than identifying techniques to preclude unwanted outcomes and providing cost saving opportunities in a very cost effective manner, RCAs can be considered as an effective knowledge enhancement tool for an organization.

Automotive sector is a driving sector for countries due to forward and backward linkages and employment and value added it creates. Starting from this point, this study aims to analyze the competitiveness of CIS and Turkey automotive sector in their market. The competitiveness of the countries should be assessed with their production potential. The countries subject to the study are thirteen Eurasian countries, Armenia, Azerbaijan, Belarus, Georgia, Kazakhstan, Kyrgyz Republic, Republic of Moldova, Russian Federation, Tajikistan, Turkey, Turkmenistan, Ukraine and Uzbekistan. The data from UNCTAD for 1995-2011 period for these countries and Liesner Index (RCAL), Balassa Index (RCA), Relative Trade Advantage Index (RTA) and Relative Competitiveness Index (RC) are used and the rank correlation between the outcomes are checked via Spearman Rank Correlation coefficient. According to the outcomes, Belarus, Turkey and Uzbekistan have higher comparative advantage compared to the other countries. The cooperation of these countries with Russia and Ukraine on production and trade of automotive sector will enhance their comparative advantage against third countries.

Dwindling supplies of natural concrete aggregates, the cost of landfilling construction waste, and interest in sustainable design have increased the demand for recycled concrete aggregates (RCA) in new portland cement concrete mixtures. RCA repurposes waste material to provide useful ingredients for new construction applications. However, RCA can reduce the performance of the concrete. This study investigated the effectiveness of ternary blended binders, mixtures containing portland cement and two different supplementary cementitious materials, at mitigating performance losses of concrete mixtures with RCA materials. Concrete mixtures with different ternary binder combinations were batched with four recycled concrete aggregate materials. For the materials used, the study found that a blend of portland cement, Class C fly ash, and blast furnace slag produced the highest strength of ternary binder. At 50% replacement of virgin aggregates and ternary blended binder, some specimens showed comparable mechanical performance to a control mix of only portland cement as a binder and no RCA substitution. This study demonstrates that even at 50% RCA replacement, using the appropriate ternary binder can create a concrete mixture that performs similarly to a plain portland cement concrete without RCA, with the added benefit of being environmentally beneficial.

This paper presents advancements in a surrogate-based, rotor blade design optimization framework for improved helicopter performance. The framework builds on previous successes by allowing multiple airfoil sections to designed simultaneously to minimize required rotor power in multiple flight conditions. Rotor power in hover and forward flight, at advance ratio 𝜇 = 0.3, are used as objective functions in a multi-objective genetic algorithm. The framework is constructed using Galaxy Simulation Builder with optimization provided through integration with Dakota. Three independent airfoil sections are morphed using ParFoil and aerodynamic coefficients for the updated airfoil shapes (i.e., lift, drag, moment) are calculated using linear interpolation from a database generated using C81Gen/ARC2D. Final rotor performance is then calculated using RCAS. Several demonstrative optimization case studies were conducted using the UH-60A main rotor. The degrees of freedom for this case are limited to the airfoil camber, camber crest position, thickness, and thickness crest position for each of the sections. The results of the three-segment case study show improvements in rotor power of 4.3% and 0.8% in forward flight and hover, respectively. This configuration also yields greater reductions in rotor power for high advance ratios, e.g., 6.0% reduction at 𝜇 = 0.35, and 8.8% reduction at 𝜇 = 0.4.
