Introdução: A ativação do receptor Toll-símile 4 ou ?Toll-Like Receptor-4 (TLR4)??após lesão estéril leva à disfunção de órgãos em locais distantes. Tem sido demostrado anteriormente que a ruptura da barreira intestinal e alteração em proteínas das junções apertadas segue-se ao choque trauma/choque hemorrágico (T/HS), no entanto, o papel de TLR4, este processo permanece obscuro. Foi postulado que o aumento da permeabilidade e a ruptura da barreira intestinal após T/HS é um processo TLR4 dependente, portanto, silenciar o gene TLR4 teria um efeito protetor sobre a disfunção intestinal induzida por T/HS. Método: Camundongos machos C57BL/6J (TLR4+/+ do tipo selvagem ou “wild-type” [WT]) e C57BL/10ScN (TLR4-/- transgênico ou “knockout” [K0]) foram distribuídos aleatoriamente e submetidos a T/HS (laparotomia mediana + choque hemorrágico com pressão arterial média mantida em 35mmHg por 60 minutos) ou submetidos a operação simulada. Após 4h, a permeabilidade ao isotiocianato de fluoresceína (FITC)-dextrano administrado intraluminalmente foi avaliada por quantificação da fluorescência do plasma. Amostras intestinais foram analisadas para a presença das proteínas das juncoes apertadas, ocludina e ZO-1, por imunohistoquímica. Resultados: As concentrações séricas de FITC-dextran 4kDa estavam diminuídas no grupo TLR4-/-[K0] em comparação com camundongos TLR4+/+[WT] quando submetidos ao modelo de T/HS (38,96 vs 164,26 ?g/ml, respectivamente, p<0,05). Não foram observadas diferenças na permeabilidade intestinal entre os camundongos TLR4-/-[K0] e os animais do grupo simulação TLR4+/+[WT]. Os camundongos K0 tiveram preservação das concentrações de ocludina e ZO-1 após a lesão T/HS em ensaios de imunohistoquímica, mas as concentrações estavam reduzidas em animais WT. Conclusão: Camundongos com TLR4-/-[K0] demonstraram menor permeabilidade intestinal ao FITC-dextrano do que camundongos WT após lesão por T/HS resultantes de alterações nas proteínas das junções apertadas, ocludina e ZO-1. Estes achados sugerem que a maior permeabilidade e a ruptura da barreira intestinal após lesão por T/HS é um processo TLR4-dependente. ________________________________________________________________________________ ABSTRACT
Objective: Toll-like receptor 4 (TLR4) activation after sterile injury leads to organ dysfunction at distant sites. Has been shown previously that intestinal barrier breakdown and alteration of tight junction proteins follows trauma/hemorrhagic shock (T/HS); however, the role of TLR4 in this process remains unclear. Was postulated that increased intestinal permeability and barrier breakdown after T/HS is a TLR4 dependent process; hence, knocking down the TLR4 gene would have a protective effect on T/HSinduced intestinal dysfunction. Method: Male C57BL/6J (TLR4 wild type [WT]) and C57BL/10ScN (TLR4 knockout [K0]) mice were assigned randomly to either T/HS (femoral catheterization + laparotomy + hemorrhagic shock) or sham injury. At 4h, permeability to intraluminally administered fluorescein isothiocyanate (FITC)-dextran was assessed by measuring the fluorescence of the serum. Intestinal samples were analyzed for the presence of the tight junction proteins, occludin and ZO-1, by immunohistochemistry. Results: Serum concentrations of FITC-dextran were decreased in K0 mice compared with WT mice after T/HS injury (38,96 ?g/ml and 164,26 ?g /ml, respectively; p<0,05). After injury, no difference in intestinal permeability was observed between the K0 mice and the WT shamtreated mice. The K0 mice had preservation of occluding and ZO-1 concentrations after T/HS injury in immunohistochemistry assays, but concentrations were decreased in WT animals. Conclusions: Mice with TLR4K0 have less intestinal permeability to FITC-dextran than WT mice after T/HS injury as a result of alterations in the tight junction proteins occludin and ZO-1. These findings suggest that the greater intestinal permeability and barrier breakdown after T/HS injury is a TLR4-dependent process.

RU : Основной текст квалификационной работы магистра «Рецепция травмы в сборник малой прозы Наринэ Абгарян “Дальше жить”» содержит 45 страниц. Для выполнения работы изучено 43 источников. Объект исследования ‒ сборник малой прозы Наринэ Абгарян “Дальше жить”. Цель исследования ‒ проанализировать специфику художественной рецепции травмы в сборник малой прозы Наринэ Абгарян “Дальше жить”. В ходе исследования предполагается решить следующие задачи: 1) проанализировать подходы к проблеме травмы в современном гуманитарном дискурсе; 2) выявить специфику студий травмы в литературоведческом дискурсе; 3) охарактеризовать факторы, повлиявшие на характер рецепции травмы в анализируемом сборнике; 4) обозначить индивидуально - авторское видение травмы и охарактеризовать характер ее рецепции на разных структурных уровнях сборника. Источниками исследования послужили текст сборника малой прозы Наринэ Абгарян “Дальше жить”, научны работы, монографии и статьи, посещённые проблемам травмы и памяти, литературоведческие словари. Методы исследования ‒ проблема исследуется с помощью описательного метода. Практическая значимость ‒ материалы работы могут быть использованы при изучении проблемы травмы в художественной литературе, на практических занятиях по современной военной и исторической прозе и на занятиях, посвященных проблеме травмы в произведениях. Апробация работы ‒ результаты исследования были представлены на Всеукраинской научной конференции «Запорізькі філологічні читання», ЗНУ 11-12.11.2021 г.; тема доклада ‒ «След войны: особенности структурной организации сборника малой прозы Наринэ Абгарян “Дальше жить”».
EN : The main text of the master's qualification work "Reception of trauma in Narine Abgaryan’s collection of flash fictions “Дальше жить”" contains 45 pages. To carry out the work, 43 sources were studied. The object of the research is collection of flash fictions “Дальше жить” written by Narine Abgaryan. The purpose of the research is to analyze the specifics of artistic perception of trauma in Narine Abgaryan’s collection of flash fictions “Дальше жить”. In the course of the study, it is supposed to solve the following tasks: 1) analyze approaches to the problem of trauma in modern humanitarian discourse; 2) reveal the specifics of trauma studios in literary discourse; 3) describe the factors that influenced the nature of the reception of trauma in the collection of flash fictions “Дальше жить”; designate the individual author's vision of trauma and characterize the nature of its reception at different structural levels of the collection. The sources of the research were the text of the Narine Abgaryan’s collection of flash fictions “Дальше жить”, scientific works, monographs and articles on the problems of trauma and memory, literary dictionaries. Research methods – descriptive. Practical significance - materials of the research can be used in the study of the problem of trauma in fictions and literature, at practical classes on modern military and historical prose. Approbation of the work - the results of the study were presented at the All-Ukrainian scientific conference «Запорізькі філологічні читання», ZNU 11-12.11.2021 г.; the topic of the report is «“Trace of the War: Features of the Structural Organization of Narine Abgaryan’s Abgaryan’s collection of flash fictions “Дальше жить”».
