We consider the problem of encoding range minimum queries (RMQs): given an array A [1.. n ] of distinct totally ordered values, to pre-process A and create a data structure that can answer the query RMQ( i , j ), which returns the index containing the smallest element in A [ i .. j ], without access to the array A at query time. We give a data structure whose space usage is 2 n + o ( n ) bits, which is asymptotically optimal for worst-case data, and answers RMQs in O (1) worst-case time. This matches the previous result of Fischer and Heun, but is obtained in a more natural way. Furthermore, our result can encode the RMQs of a random array A in 1.919 n + o ( n ) bits in expectation, which is not known to hold for Fischer and Heun’s result. We then generalize our result to the encoding range top-2 query (RT2Q) problem, which is like the encoding RMQ problem except that the query RT2Q( i , j ) returns the indices of both the smallest and second smallest elements of A [ i .. j ]. We introduce a data structure using 3.272 n + o ( n ) bits that answers RT2Qs in constant time, and also give lower bounds on the effective entropy of the RT2Q problem.

In the last decades, Reconfigurable Manufacturing Systems (RMSs) rose as an emerging manufacturing strategy matching the modern industrial and market requirements asking for a wide variety of products in flexible batches. A traditional reconfigurable manufacturing environment consists of dynamic cells, called Reconfigurable Machine Cells (RMCs), including a set of machines called Reconfigurable Machine Tools (RMTs). Such machines are characterized by fixed elements, i.e., basic modules, and dynamic elements, i.e., auxiliary modules, allowing them to perform different operations. Despite their automation level, these systems require the intervention of the human operators in performing specific tasks, e.g., handling of the auxiliary modules from the warehouse to the RMTs and their assembly/disassembly to/from the RMTs. This issue rises relevant ergonomic and safety questions due to the human–machine collaboration. Following this stream, this paper proposes and applies a bi-objective optimization model for the design and management of RMSs. The technical objective function minimizes the reconfiguration time, i.e., the time needed to equip the RMTs with the required auxiliary modules, and the part and auxiliary module travel time among the RMCs. The ergonomic objective function minimizes the repetitive movements performed by the human operators during the working activities according to the ISO 11228-3 standard. Results show the existence of a good trade-off between the two objective functions, proving the possibility to improve the ergonomic conditions of the human operators without excessively increasing the total time needed for RMTs reconfiguration and for part and auxiliary module travelling.

Abstract Background: Assuring/demonstrating metrologic traceability of in vitro diagnostics necessitates the availability of measurand-specific reference measurement systems (RMSs) and the possibility for industry to work with competent reference measurement laboratories (RMLs). Here we report the results of a European project to investigate the feasibility of developing a RMS for serum total thyroxine. Methods: Four candidate RMLs (cRMLs) developed/implemented variants of a candidate reference measurement procedure (cRMP) based on isotope dilution–liquid chromatography–mass spectrometry. The sole constraint implemented was calibration with a common thyroxine primary calibrator. The RMPs were externally validated and assessed for comparability in round-robin trials using common samples, i.e., 5 lyophilized and 33 frozen native sera. At the same time, the performance of the cRMLs organized in a network was assessed. For uniform external quality assessment, common performance specifications were agreed on. Results: All cRMLs performed the cRMPs with fulfillment of the predefined specifications: total and between-laboratory CVs ≤2.0% and 2.5%, respectively, and a systematic deviation ≤0.9%, estimated with a target assigned from the mean of means obtained by the cRMLs. The mean expanded uncertainty for value assignment to the native sera was 2.1%. Conclusions: A network of cRMLs, with externally conformed competence to properly perform RMPs, has been established. Performance specifications were defined and will form the basis for admittance of new network members. A serum panel, successfully targeted during the validation process, is available for split-sample measurements with commercial routine measurement procedures. The model can now be used for other measurands for which traceability to the Système International d’Unités is needed.

The reconfigurable manufacturing system (RMS) is a new manufacturing technology and paradigm that resolves the contradictions regarding high efficiency, low cost and flexible production in the mass production of part families. Reconfigurable machine tools (RMTs) are the core components of RMSs. A new approach is proposed for the design of RMTs, which is closely related to the process planning of a given box-type part family. The concepts of the processing unit and the processing segment are presented; they are not only the basic elements of the processing plans of machined parts, but also closely related to the structural design of RMTs. Processing units created by processing features can be combined into various processing segments. All the processing units of one processing segment correspond to the machining operations performed by one RMT. By arranging the processing segments according to the processing sequence, a variety of feasible processing plans for a part can be obtained. Through analysis of the established similarity calculation model for processing plans, the most similar processing plans for the parts in a given part family can be determined and used for the structural design of RMTs. Therefore, the designed RMTs can achieve rapid conversion of processing functions with the least module replacement or adjustment to realize the production of the parts in the part family. Taking the production of a gearbox part family as an example, the validity of the presented method is verified.

The potential for improving the efficiency of medical organizations that provide medical care to the population in conditions of limited resources largely depends on an objective and comprehensive assessment of their use. In this regard, the research of methodological and practical approaches to assessing the efficiency and rational use of resource potential, which are important for different levels of the organization of the health system, including the provision of medical care to citizens living in rural areas, is of particular relevance. The purpose of the study was to determine the relationship between indicators for assessing the quality of resource management and indicators of access to medical care of medical organizations of the state health system in Moscow that provide primary health care to adults in 2019. The quality of resource management was assessed using 27 indicators selected for the purpose of the study in 4 areas of resource management (financial management, procurement management, property management, personnel management), provided by the methodology of the resource management quality standard (RMQS). For each indicator, the calculated score based on the importance value (weight value) a normative criterion of evaluation and the degree of difficulty, and by summing up of scores obtained a composite score. The assessment of the availability of medical care is also calculated using the method of point estimates based on 7 indicators developed through the use of the methodology of performance audit. The article presents the results of a study based on data from 9 Moscow city polyclinics, which confirmed the existence of a correlation between the selected indicators of the quality of resource management and the availability of medical care. The total score for 2019 for the selected indicators of resource management quality varies from 9.62 points to 13.92 points, availability-from 5.54 to 11.63 points, and the correlation coefficient was 0.612

Purpose The purpose of this paper is to analyse whether hotels that use a revenue management system (RMS) outperform non-RMS-users in a context of decreasing demand. Design/methodology/approach A database of chain hotels with a rating of three or more stars was used to estimate MANOVA and ANOVA models to analyse the role of RMSs in hotel performance. Findings In a context of strong competition in prices and surplus capacity, the findings suggest that RMSs have been more effective in improving occupancy than in achieving higher rates. Also, the use of RMSs did not have a significant impact on hotel labour productivity. Research limitations/implications Managers may believe that they have adopted an RMS when, in fact, they have not fully done so. In addition, establishment-level unobserved heterogeneity, such as the quality of management or unobserved quality of service, cannot be fully controlled because of the nature of the data used. The main implication of this paper is that the potential of RMSs as revenue enhancer might be influenced by unstable market and economic conditions. However, the absence of significant effects on RevPAR performance might be also the result of firms’ adopting inadequate RM strategies. Further research could investigate whether the findings are context-specific or whether firms are failing to implement effective RMSs for other reasons. Originality/value The approach used in this paper is new to the literature, given that it uses statistical methods to analyse the impact of implementing an RMS on hotel performance under specific economic conditions and using alternative indicators.

Rhabdomyosarcomas (RMSs) are the most common soft tissue sarcomas of childhood and adolescence. To date, there are no effective treatments that target the genetic abnormalities in RMS, and current treatment options for high-risk groups are not adequate. Over the past two decades, research into the molecular mechanisms of RMS has identified key genes and signaling pathways involved in disease pathogenesis. In these studies, members of the receptor tyrosine kinase (RTK) family of cell surface receptors have been characterized as druggable targets for RMS. Through small molecule inhibitors, ligand-neutralizing agents, and monoclonal receptor-blocking antibodies, RTK activity can be manipulated to block oncogenic properties associated with RMS. Herein, we review the members of the RTK family that are implicated in RMS tumorigenesis and discuss both the problems and promise of targeting RTKs in RMS.

A 7 mo old Labrador retriever presented for evaluation of facial swelling associated with a 5 cm oral mass extending caudally from the upper third premolar on the left side. Cytology revealed an atypical population of round cells of undetermined origin. A diagnosis of alveolar rhabdomyosarcoma (RMS) was reached via histopathology and confirmed with positive immunohistochemical staining for desmin. In humans, RMSs have a well-described round cell cytologic appearance. Few descriptions of veterinary cases of RMS exist. This report describes the cytologic appearance of alveolar RMS in a young dog and both summarizes and compares findings throughout the veterinary and human literature.

Purpose. Rhabdomyosarcoma (RMS) is an embryonal tumor thought to arise from skeletal muscle cells that fail to differentiate terminally. The majority of RMSs express MyoD, a protein essential to the differentiation of skeletal muscle. It was recently shown that during myogenesis, MyoD activates the expression of the cyclin-dependent kinase inhibitor (CDKi), p21, which itself plays a critical role in normal muscle development. To investigate the integrity of the MyoD/p21 pathway in RMS, we analyzed p21 and its relationship to MyoD expression in RMS.Methods. A panel of RMS samples was assembled from primary biopsies and from cell lines. Integrity of p21 was analyzed by single-strand conformation polymorphism (SSCP) and sequencing. Expression of p21 and MyoD was determined by Northern blot analysis, and the ability of exogenous p21 to arrest the cell cycle of RMS cell line was determined by transfection studies.Results. Our analysis indicates that although p21 is wild type in RMS, there is an inverse correlation between the levels of p21 and MyoD in these tumors. Tumors that express significant amounts of MyoD fail to express p21. This does not appear to be the result of mutations within the potential CACGTG sites present in the p21 promoter region or in the coding region of p21. An additional group of RMSs express very high levels of p21 but express little, if any, MyoD. Furthermore, RD, a RMS cell line which expresses high levels of endogenous p21, undergoes withdrawal from the cell cycle following forced expression of p21, suggesting that the pathway which would lead to G1arrest from endogenous p21 activity is defective.Discussion. These data suggest that the interaction between p21 and MyoD is defective in RMS although the precise nature of the defect remains to be elucidated.

Abstract DNA methylation is a common epigenetic mark that influences transcriptional regulation, and therefore cellular phenotype, across all domains of life. In particular, both orphan methyltransferases and those from phasevariable restriction modification systems (RMSs) have been co-opted to regulate virulence epigenetically in many bacteria. We now show that three distinct non-phasevariable Type I RMSs in Escherichia coli have no measurable impact on gene expression, in vivo virulence, or any of 1190 in vitro growth phenotypes. We demonstrated this using both Type I RMS knockout mutants as well as heterologous installation of Type I RMSs into two E. coli strains. These data provide three clear and currently rare examples of restriction modification systems that have no impact on their host organism’s gene regulation. This leads to the possibility that other such nonregulatory methylation systems may exist, broadening our view of the potential role that RMSs may play in bacterial evolution.

Purpose The purpose of this study was to investigate the effects of home use of a remote microphone system (RMS) on the spoken language production of caregivers with young children who have hearing loss. Method Language Environment Analysis recorders were used with 10 families during 2 consecutive weekends (RMS weekend and No-RMS weekend). The amount of talk from a single caregiver that could be made accessible to children with hearing loss when using an RMS was estimated using Language Environment Analysis software. The total amount of caregiver talk (close and far talk) was also compared across both weekends. In addition, caregivers' perceptions of RMS use were gathered. Results Children, with the use of RMSs, could potentially have access to approximately 42% more words per day. In addition, although caregivers produced an equivalent number of words on both weekends, they tended to talk more from a distance when using the RMS than when not. Finally, caregivers reported positive perceived communication benefits of RMS use. Conclusions Findings from this investigation suggest that children with hearing loss have increased access to caregiver talk when using an RMS in the home environment. Clinical implications and future directions for research are discussed.

Multi-agent distributed systems are characterized by autonomous entities that interact with each other to provide, and/or request, different kinds of services. In several contexts, especially when a reward is offered according to the quality of service, individual agents (or coordinated groups) may act in a selfish way. To prevent such behaviours, distributed Reputation Management Systems (RMSs) provide every agent with the capability of computing the reputation of the others according to direct past interactions, as well as indirect opinions reported by their neighbourhood. This last point introduces a weakness on gossiped information that makes RMSs vulnerable to malicious agents’ intent on disseminating false reputation values. Given the variety of application scenarios in which RMSs can be adopted, as well as the multitude of behaviours that agents can implement, designers need RMS evaluation tools that allow them to predict the robustness of the system to security attacks, before its actual deployment. To this aim, we present a simulation software for the vulnerability evaluation of RMSs and illustrate three case studies in which this tool was effectively used to model and assess state-of-the-art RMSs.

PurposeThe purpose of this paper is to present an organized review of existing research on reconfigurable manufacturing system (RMS). The paper considers majority of the prominent research articles in the domain of RMS published ever since RMS was envisaged in 1997.Design/methodology/approachThe paper systematically reviews, classifies and analyses the published literature on postulations and design of RMSs. The general observations from the literature and research gaps recognized thereon are highlighted at the end of each section/sub-section.FindingsThe paper reveals important aspects related to RMS research since its inception. It also recognizes the areas of RMS research requiring more focus. The study also highlights open issues and future directions for further research.Practical implicationsThe literature in the domain of RMS has so far been narrow. This paper reviews the prominent research in this field and presents an overview of its conceptual developments and various mathematical models for the RMS design and its optimization so far developed by the researchers. Further, manufacturing advancements and future directions have also been proposed for the efficient execution of RMS paradigm in manufacturing industries.Originality/valueThe paper provides an organized listing of published research work in the field of RMS. This work will provide an insight to the researchers, practitioners and others related directly or indirectly to this field to develop and understand better strategies for supervising and controlling the smooth implementation of RMS.

Purpose This study examined the impact of home use of remote microphone systems (RMSs) on caregiver communication and child vocalizations in families of children with hearing loss. Method We drew on data from a prior study in which Language ENvironmental Analysis recorders were used with 9 families during 2 consecutive weekends—1 that involved using an RMS and 1 that did not. Audio samples from Language ENvironmental Analysis recorders were (a) manually coded to quantify the frequency of verbal repetitions and alert phrases caregivers utilized in communicating to children with hearing loss and (b) automatically analyzed to quantify children's vocalization rate, duration, complexity, and reciprocity when using and not using an RMS. Results When using an RMS at home, caregivers did not repeat or clarify their statements as often as when not using an RMS while communicating with their children with hearing loss. However, no between-condition differences were observed in children's vocal characteristics. Conclusions Results provide further support for home RMS use for children with hearing loss. Specifically, findings lend empirical support to prior parental reports suggesting that RMS use eases caregiver communication in the home setting. Studies exploring RMS use over a longer duration of time might provide further insight into potential long-term effects on children's vocal production.

A diverse set of mobile genetic elements (MGEs) transmit between Streptococcus pneumoniae cells, but many isolates remain uninfected. The best-characterised defences against horizontal transmission of MGEs are restriction-modification systems (RMSs), of which there are two phase-variable examples in S. pneumoniae. Additionally, the transformation machinery has been proposed to limit vertical transmission of chromosomally integrated MGEs. This work describes how these mechanisms can act in concert. Experimental data demonstrate RMS phase variation occurs at a sub-maximal rate. Simulations suggest this may be optimal if MGEs are sometimes vertically inherited, as it reduces the probability that an infected cell will switch between RMS variants while the MGE is invading the population, and thereby undermine the restriction barrier. Such vertically inherited MGEs can be deleted by transformation. The lack of between-strain transformation hotspots at known prophage att sites suggests transformation cannot remove an MGE from a strain in which it is fixed. However, simulations confirmed that transformation was nevertheless effective at preventing the spread of MGEs into a previously uninfected cell population, if a recombination barrier existed between co-colonising strains. Further simulations combining these effects of phase variable RMSs and transformation found they synergistically inhibited MGEs spreading, through limiting both vertical and horizontal transmission.

Rhabdomyosarcoma (RMS) is rare in adulthood, accounting for 2%–5% of adult soft tissue tumors, and less than 20% occur in genitourinary organs. Given its rarity, survival data on adult kidney, bladder, and prostate RMSs is limited. In this population-based analysis, we performed an analysis of all adult RMS cases reported in Surveillance, Epidemiology, and End Results (SEER) database to understand prognostic factors among kidney, bladder, and prostate RMS. A query of the SEER database was performed from 1973 to 2016 for patients >18 of age with RMS. The final cohort consisted of 14 kidney, 35 bladder, and 21 prostate RMS cases in the adult population. Demographic, treatment, and survival data were obtained. Analysis was performed using Fisher’s exact test, survival analysis, and model. The median (range) age of diagnosis for adult bladder RMS was 65 years old (19–84) compared to 52.5 (28–68) and 42 (19–87) for kidney and prostate ( p = 0.007). About 78.6% of patients underwent surgical intervention. Five-year overall survival (OS) for adult kidney, bladder, and prostate RMS are 17.1% (2.9–41.6%), 22.2% (9.4–38.4%), and 33.0 (12.8–55.0%), respectively. OS was not statistically associated with primary site ( p = 0.209). On multivariable analysis, compared to adult bladder RMS, kidney RMS had a higher incidence of mortality (HR: 2.16, 95% CI 1.03–4.53, p = 0.041). Incidence of mortality from prostate RMS was not significantly different from bladder RMS (HR: 0.70, 95% CI 0.30–1.65, p = 0.411). Extent of disease (HR: 5.17, 95% CI 2.09–12.79, p < 0.001) and older age (HR 1.03, 95% CI 1.01–1.04, p = 0.002) were adverse prognostic factors for OS. Overall survival at 5 years for adult kidney, bladder, and prostate RMS is poor. Localized disease and younger age are prognostic factors for improved outcomes in adult RMS. Hence, early diagnosis and intervention appear paramount to improved survival for this rare malignancy in adulthood.

Purpose Rhabdomyosarcoma (RMS) is classified into two main subgroups: the embryonal (ERMS) and the alveolar (ARMS) form. The majority of the ARMSs are associated with specific chromosomal translocations (pARMS). Because ARMS is much more aggressive than ERMS, RMS subclassification has clinical relevance. However, diagnosis of RMS subgroups on the basis of histology or molecular biology can be difficult, and supplementing diagnostic methods would be desirable. The aim of this study was to establish a panel of markers for RMS subgroup classification by immunohistochemistry. Materials and Methods Gene expression data were used for selection of subgroup-specific markers. Single sections of RMS with available expression data were used for establishment of the immunohistochemistry. Evaluation of the sensitivity and specificity of the markers was carried out using a tissue array representing 252 RMSs. Kaplan-Meier survival curves were calculated for determination of differences in overall survival of the different staining subgroups. Results AP2β and P-cadherin were selected as markers for pARMS, and epidermal growth factor receptor (EGFR) and fibrillin-2 as markers for ERMS. EGFR + fibrillin-2 detected ERMS with a specificity of 90% and with a sensitivity of 60%. AP2β + P-cadherin detected pARMS with a specificity of 98% and a sensitivity of 64%, and allowed the detection of several misclassified tumors. The EGFR + fibrillin-2–positive group is associated with a favorable outcome, and the AP2β + P-cadherin–positive group is associated with an unfavorable outcome. Conclusion The presented set of marker proteins detects RMS subgroups with high specificity and may be useful in routine subtype classification of RMS.

Distributed environments consist of a huge number of entities that cooperate to achieve complex goals. When interactions occur between unknown parties, intelligent techniques for estimating agent reputations are required. Reputation management systems (RMS's) allow agents to perform such estimation in a cooperative way. In particular, distributed RMS's exploit feedbacks provided after each interaction and allow prediction of future behaviors of agents. Such systems, in contrast to centralized RMSs, are sensitive to fake information injected by malicious users; thus, predicting the performance of a distributed RMS is a very challenging task. Although many existing works have addressed some challenges concerning the design and assessment of specific RMS's, there are no simulation environments that adopt a general approach that can be applied to different application scenarios. To overcome this lack, we present DRESS, an agent-based simulation framework that aims to support researchers in the evaluation of distributed RMSs under different security attacks.

One of the latest solutions in automated production systems is developing of Reconfigurable Manufacturing Systems (RMSs), which have an open structure that allows the integration of new technologies and modifying manufacturing for a new type of production based on adding, removing or updating the elements and units in the hardware and software level. To realize the automated processing RMS needs to have decentralized architecture, including hardware (module based) and software reconfiguration, which to enable distributed control, monitoring and diagnostics in real time and dynamic planning processes and resources in the system. One of the ways to realize this architecture is to development RMS control system on the base of IEC 61499 function blocks (FBs). Important part of the control system is a module for distributed planning and scheduling of processes, which consists all available information for technology of the products. This paper presents an approach for development of scheduling and planning modules in reconfiguration cutting work station on the base of IEC 61499 function blocks.

With the recent diversification of demands, manufacturing systems that can respond to multiple types of goods have become more important. In this circumstance, reconfigurable manufacturing systems (RMSs) that can provide flexible manufacturing with limited machine tools through reconfiguration have gained a lot of attention. As an RMS supports flexibility through layout reconfiguration, reconfiguration decision-making is very important and difficult. The development of IoT technology has made it possible to collect hidden information inside systems. This study focused on the reconfiguration decision-making system with the data acquisition system based on IoT technology. The decision-making system detected a reconfiguration situation and built a reconfiguration plan using the data collected by IoT sensors. The performance of the algorithm proposed in this study was verified in a simulation experiment. It was found that the algorithm had a stable performance under various reconfigurable situations. It is expected that the proposed system will help to improve the performance of RMS.

Así pues, aunque los RMSs pueden ser esenciales para la mejora continua, la globalización ha hecho difícil ganar ventaja (o aún paridad) competitiva a largo plazo con sólo implementar tales sistemas innovadores. Ello se debe en parte a la incesante e impredecible (y a veces predecible) evolución de las estructuras de mercado, de las demandas y de la tecnología de productos y de procesos. Por tanto, puede ser arriesgado el hacer predicciones de posibles rendimientos del impacto de futuros RMSs, desde mucha de la literatura teórica y empírica relacionada con el tema. Además, la evidencia de estudios empíricos en HPM muestra que la piedra angular de la competitividad industrial no es crear una nueva tendencia, sino más bien mejorar continuamente al seleccionar y adaptar, según sea necesario, sólo las practicas avanzadas de producción (PAPs) que se ajusten al contexto especifico de la fábrica, estableciendo vínculos entre las PAPs existentes y las nuevas por instalar. Así pues, tomando el contexto del HPM, se establecen las condiciones actuales para la implementación del RMS. Finalmente, este artículo hace sugerencias en relación alRMSe identifica posibles direcciones para futura investigación.

Technology options for children with limited hearing unilaterally that improve the signal-to-noise ratio are expected to improve speech recognition and also reduce listening effort in challenging listening situations, although previous studies have not confirmed this. Employing behavioral and subjective indices of listening effort, this study aimed to evaluate the effects of two intervention options, remote microphone system (RMS) and contralateral routing of signal (CROS) system, in school-aged children with limited hearing unilaterally. Nineteen children (aged 7–12 years) with limited hearing unilaterally completed a digit triplet recognition task in three loudspeaker conditions: midline, monaural direct, and monaural indirect with three intervention options: unaided, RMS, and CROS system. Verbal response times were interpreted as a behavioral measure of listening effort. Participants provided subjective ratings immediately following behavioral measures. The RMS significantly improved digit triplet recognition across loudspeaker conditions and reduced verbal response times in the midline and indirect conditions. The CROS system improved speech recognition and listening effort only in the indirect condition. Subjective ratings analyses revealed that significantly more participants indicated that the remote microphone made it easier for them to listen and to stay motivated. Behavioral and subjective indices of listening effort indicated that an RMS provided the most consistent benefit for speech recognition and listening effort for children with limited unilateral hearing. RMSs could therefore be a beneficial technology option in classrooms for children with limited hearing unilaterally.

Sclerosing RMS (SRMS) is a recently described subtype of RMS that has not yet been included in any of the classification systems for RMSs. We did pubmed search using keywords “sclerosing, and rhabdomyosarcomas” and included all pediatric cases (age ≤ 18 years) of SRMSs in this review. We also included our case of an eleven-year-old male child with skull base SRMS and discuss the clinical, histopathological, immunohistochemical, and genetic characteristics of these patients. Till now, only 20 pediatric cases of SRMSs have been described in the literature. Pediatric SRMS more commonly affects males at a mean age of 9 years. Extremeties and head/neck regions were most commonly affected. Follow-up details were available for 16 patients with mean follow-up of 25.3 months. Treatment failure rate was 43.75%. Overall amongst these 16 patients, 10 were alive without disease, 4 were alive with disease, and two died. Thus, overall and disease-free survival amongst these 16 patients were 87.5% and 62.5%, respectively. The literature regarding clinical behaviour and outcome of pediatric patients with SRMSs is patchy. Detailed molecular/genetic analysis and clinicopathological characterization with longer follow-ups of more cases may throw some light on this possibly new subtype of RMS.

Purpose The aim of this study is to investigate anaplastic lymphoma kinase (ALK) protein expression and underlying genetic aberrations in rhabdomyosarcoma (RMS), with special attention to clinical and prognostic implications. Patients and Methods A total of 189 paraffin-embedded RMS tumor specimens from 145 patients were collected on tissue microarray. ALK protein expression was evaluated by immunohistochemistry. ALK gene (2p23) copy number and translocations were determined by in situ hybridization. cDNA sequencing of the receptor tyrosine kinase domain of the ALK gene was assessed in 43 samples. Results Strong cytoplasmic ALK protein expression was more frequently observed in alveolar RMS (ARMS) than in embryonal RMS (ERMS) (81% v 32%, respectively; P < .001). ALK gene copy number gain was detected in the vast majority of ARMS (88%), compared with 52% of ERMS (P < .001). ALK copy number correlated with protein expression in primary tumors (n = 107). We identified one point mutation (2%) and seven tumors harboring whole exon deletions (16%). In ERMS, specific ALK gain in the primary tumor correlated with metastatic disease (100% in metastatic disease v 29% in nonmetastatic disease; P = .004) and poor disease-specific survival (5-year disease-specific survival: 62% v 82% for nonspecific or no gain; P = .046). Conclusion Because ALK aberrations on genomic and protein levels are frequently found in RMSs, in particular ARMS, and are associated with disease progression and outcome in ERMS, ALK may play a role in tumor biology and may provide a potential therapeutic target for these tumors. Future research should aim at the oncogenic role of ALK and the potential effect of ALK inhibitors in RMS.

It is generally considered that economic feasibility of a reconfigurable manufacturing system (RMS) is only attainable if the system is defined to be reconfigurable at the outset of its design. In this work, we consider the potential exception to this perception, in the context of a common industrial scenario where a specialized and expensive manufacturing machine or system will otherwise be rendered useless due to the loss of business of the particular product being manufactured. Specific guidelines to convert from a fixed to a reconfigurable system are proposed, and evaluated through a case study. It is shown that under certain conditions, RMSs may be economically feasible even if they are developed through the modification of pre-existing dedicated systems.

Rhabdomyosarcomas (RMS) are group of soft tissue malignant tumours predominantly childhood. Alveolar rhabdomyosarcoma (aRMS) is the second most common variant of RMS. The majority of aRMSs display a translocations of FOXO1 gene. Such tumours are aggressive, metastasize early and are associated with a worse prognosis for the patient. However, some aRMS cases are rhabdomyosarcomas without classic chromosomal rearrangements. These tumors also have alveolar morphology, but neoplastic cells lack the FOXO1 gene translocation. Such fusion-negative round-cell RMSs behave clinically differently and have a better prognosis. The aim of the present study was to assess the prevalence of FOXO1 gene rearrangements in the group of round cell rhabdomyosarcomas with alveolar morphology. This study is supported by the Independent Ethics Committee and approved by the Academic Council of the Dmitry Rogachev National Medical Research Center of Pediatric Hematology, Oncology and Immunology. The study group consisted of 250 formalin-fixed paraffin-embedded samples from patients with RMS. The cytogenetic study was performed by fluorescence in situ hybridization with a locus-specific identifier (LSI) for FOXO1 (13q14). The PAX3-FOXO1 (COSF247) and PAX7-FOXO1 (COSF287) fusion transcripts was detected by RT-PCR. In the study group 1 (аRMS), the rearrangement of PAX3/7-FOXO1 was detected in 44% of cases, in 32% of cases was detected a combined rearrangement with amplification of the 3' FOXO1. In one case, the amplification of the 3' end of the FOXO1 gene was detected without rearrangement; this sample was sent for additional PCR study, as a result of which the chimeric PAX3-FOXO1 transcript was detected. In 22% cases cytogenetic abnormalities were not found. has not been identified. In group 2 (embryonal RMS) we did not detect translocation. The group of round-cell PMCs is heterogeneous and is represented by three variants of genetic events that determine the disease prognosis. At the same time, FOXO1 gene abnormalities are not found in the RMS group with non-alveolar morphology.

The International Standardization Organization and several countries have published standards that provide guidance on evaluation of human response to vibration in buildings, e.g. ISO 2631-2:1989, BS 6472: 1984, and ANSI S3.29: 1983. This paper demonstrates the application of these standards for the case of vibrations induced in residential homes by transit buses. Difficulties and ambiguities in applying the standards are revealed. Several vibration evaluation methods were applied and compared including rms, rmq, peak, and vibration-dose-value methods. Actual vibration levels measured in 6 residential homes were used to assess the various methods. Two evaluation methods were identified as the most appropriate.

mTOR activation has been observed in rhabdomyosarcoma (RMS); however, mTOR complex (mTORC) 1 inhibition has had limited success thus far. mTOR activation alters the metabolic pathways, which is linked to survival and metastasis. These pathways have not been thoroughly analyzed in RMSs. We performed immunohistochemistry on 65 samples to analyze the expression of mTOR complexes (pmTOR, pS6, Rictor), and several metabolic enzymes (phosphofructokinase, lactate dehydrogenase-A, β-F1-ATPase, glucose-6-phosphate dehydrogenase, glutaminase). RICTOR amplification, as a potential mechanism of Rictor overexpression, was analyzed by FISH and digital droplet PCR. In total, 64% of the studied primary samples showed mTOR activity with an mTORC2 dominance (82%). Chemotherapy did not cause any relevant change in mTOR activity. Elevated mTOR activity was associated with a worse prognosis in relapsed cases. RICTOR amplification was not confirmed in any of the cases. Our findings suggest the importance of the Warburg effect and the pentose-phosphate pathway beside a glutamine demand in RMS cells. The expression pattern of the studied mTOR markers can explain the inefficacy of mTORC1 inhibitor therapy. Therefore, we suggest performing a detailed investigation of the mTOR profile before administering mTORC1 inhibitor therapy. Furthermore, our findings highlight that targeting the metabolic plasticity could be an alternative therapeutic approach.

Rhabdomyosarcoma (RMS) is a common malignancy of soft tissue, subclassified as alveolar (ARMS), pleomorphic (PRMS), spindle cell/sclerosing (SRMS), and embryonal (ERMS) types. The Yes-associated protein (YAP) is a member of the Hippo pathway and a transcriptional regulator that controls cell proliferation. We have studied the immunohistochemical expression of YAP in different RMSs, arranged in tissue microarray (TMA) and whole slide formats. Pertinent clinical data including patient age, gender, tumor location, and clinical stage were collected. Out of 96 TMA cases, 30 cases (31%) were pleomorphic, 27 (28%) were embryonal, 24 (25%) alveolar, and 15 (16%) spindle cell. Positive nuclear YAP staining was seen in the PRMS (17/30, 56.7%), SRMS (7/15, 46.7%), ERMS (19/27 or 70%), and less in ARMS (37.5%). YAP nuclear staining was significantly more prevalent in ERMS than ARMS ( p=0.02). Of the 41 whole slide cases, nuclear staining was detected in all ARMS but was restricted in distribution to <30% of the cells, in contrast to ERMS and SRMS, which had diffuse or >30% staining. These results highlight the role of YAP in RMS tumorigenesis, a fact that can be useful in engineering targeted therapy. Restricted nuclear YAP staining (<30% of cells) may be of value in the diagnosis of ARMS.

Abstract Introduction/Objective Rhabdomyosarcoma (RMS) encompasses a heterogeneous group of tumors with striated muscle morphology and/or immunophenotype. Molecular analysis of rhabdomyosarcoma has identified a number of aberrations useful for classification. Initially, fusion associated RMS was restricted to tumors with FOXO1 rearrangements, confirming the diagnosis of alveolar RMS, and identifying patients with poor-outcome. Spindle cell RMS, have been described with VGLL2 fusions, EWSR1/FUS-TFCP2 rearrangements, and myoD1 mutations. In addition, NCOA2-MEIS1 fusion gene was recently described in two primary intraosseous RMSs sharing primitive histology. Herein we report two cases of spindle cell RMS, harboring different novel fusion genes, one presenting EP300-VGLL3, and the second with NCOA2-MEIS1 and CAV1-MET. Methods Two cases of RMS were retrieved from the pathology files at the University of Miami Hospital (UMH) and Jackson Memorial Hospital (JMH). Morphology and immunohistochemistry were reviewed. A commercial genomic profiling test for both cases was also analyzed. The study was approved by the Institutional Review Board at UMH. Results Case #1 is a 36-year-old man presented with a 1.5 cm submucosal mass in the ventral left tongue. The biopsy revealed a malignant spindle cell neoplasm and immunohistochemistry showed tumor cells positive for desmin, SMA, myogenin and MYOD-1. A partial glossectomy was performed, achieving negative resection margins. Next- generation sequencing (NGS) identified an EP300-VGLL3 fusion. The patient is currently being followed and has been disease-free for 6 months. Case #2 is a 19-years-old male with no medical history admitted with lower extremities weakness and a large expansile pelvic bone lesion and multiple spinal metastases. Biopsy from the pelvic lesion revealed malignant primitive round cells and short spindle cells arranged in fascicles which were positive for desmin, MYOD-1, TLE-1 and CD99 and focally positive for myogenin and WT-1. NGS identified two fusion genes, NCOA2- MEIS1 and CAV1-MET. Conclusion Different types of rhabdomyosarcoma are associated with a variety of different gene fusions and some may be of prognostic significance. Our two cases are the first reported with EP300-VGLL3 and CAV1-MET fusion genes. CAV1-MET oncogenic potential has been described only in lung cancer. The prognostic significance of rhabdomyosarcoma with novel gene fusions requires longer follow-up and the identification of additional cases.
