I studied the existence of sentinels in Saltator atricollis in a 'campo-cerrado' (Brazilian savanna) during the breeding seasons of 1994-95 and 1995-96. Groups of this species consisted of two to seven birds (N = 25). Sentinels were present (mean ± sd) 62% ± 9% of the time, and most sentinel bouts lasted from less than one to six minutes (although in some occasions for more than 30 min.). Overlap of two or more sentinels occurred on average only 3.2% of the time with birds on guard. Sentinels commonly gave contact calls in the beginning and/or at the end of a sentinel bout. The group size had no effect on time with sentinels. However the encounter rate between raptors and groups significantly influenced the time guarded. The results of this study suggests that the ability of sentinels to detect predators may play a central role as an anti-predator device.

Bird mixed flocks including Cypsnagra hirundinacea and Neothraupis fasciata as species with sentinels were studied in "campo-cerrado" in order to investigate the possible relationship between alertness and the mixed flock leadership. This study was conducted from March to September 1996 and mixed flocks were observed on average for 2:30h. The time with sentinels were recorded for C. hirundinacea and N. fasciata. The sentinels of Cypsnagra hirundinacea performed most of the vigilance (time with sentinel was on average 42 ± 17%, 41 ± 17% of which by C. hirundinacea, whereas only 1.2 % by N. fasciata) and gave all the alarm calls recorded (54% of the encounters with raptors stimulated alarm calls). A relationship was verified between time with sentinel and the rate of encounters with raptor (ANOVA, F = 3.0, P < 0.05). The results of this study are an evidence for the anti-predatory function of mixed flocks, in campo-cerrado, and the alertness as a major feature of a leader species, since C. hirundinacea always led those flocks.

Beginning in 2015, athymic nude sentinel mice from conventional, medium-, and high-security facilities presented to the Comparative Pathology Laboratory (CPL) with weight loss, diarrhea, and/or rectal prolapse. Regardless of whether clinical signs were present or absent, the gross observation of ceco-colonic thickening corresponded histologically to pleocellular typhlocolitis with mucosal hyperplasia and lamina proprial multinucleated cells. A subset of affected sentinels exhibited granulomatous serositis and hepatosplenic necrosis with multinucleated cells. Initial suspicion of mouse hepatitis virus infection was excluded by polymerase chain reaction, electron microscopy, and serology. Multinucleated giant cells were confirmed as macrophages by positive immunoreactivity to Mac-3 and Iba-1 and negative immunoreactivity to pancytokeratin. From conventional and medium-security facilities, Helicobacter species were identified in 40 of 143 (27.9%) mice, with H. hepaticus accounting for 72.5% of identified Helicobacter species. Other agents included opportunistic bacterial infection (41/145, 28.3%), murine norovirus (16/106, 15.1%), and pinworms (2/146, 1.4%). From high-security facilities, only Enterobacter cloacae was identified (2/13, 15.4%), and no evidence of Helicobacter sp., murine norovirus, or pinworms was present. No potentially infectious disease agent(s) was identified in 71 of 146 (48.6%) affected nude sentinels from conventional and medium-security facilities and 11 of 13 (84.6%) affected nude sentinels from high-security facilities. No statistically significant differences in histologic lesion scores were identified between Helicobacter-positive and Helicobacter-negative mice. Thus, proliferative typhlocolitis with multinucleated giant cells was considered a nonspecific histologic pattern associated with a variety of primary and opportunistic pathogens in athymic nude mice.

Mountain plant communities are thought to be sensitive to climate change and, thus, able to reveal its effects sooner than others. The status as sentinels of two plant communities are reviewed. Alpine treeline ecotones and alpine vegetation have been observed to respond to climate change in recent decades. The treeline has moved upslope and alpine communities have had some species increase and others decrease. The response for both, however, has been inconsistent if taken as a whole. Problematic factors for this response are outlined for both: abiotic and biotic interactions partially decouple the plant communities from climate. Differences across spatial and temporal scales complicate interpretation. Partial decoupling leads to nonlinear responses and difficulties for prediction and for planning mitigation.

Earthworms and leeches are sentinel animals that represent the annelid phylum within terrestrial and freshwater ecosystems, respectively. One early stress signal in these organisms is related to innate immunity, but how nanomaterials affect it is poorly characterized. In this survey, we compare the latest literature on earthworm and leeches with examples of their molecular/cellular responses to inorganic (silver nanoparticles) and organic (carbon nanotubes) nanomaterials. A special focus is placed on the role of annelid immunocytes in the evolutionarily conserved antioxidant and immune mechanisms and protein corona formation and probable endocytosis pathways involved in nanomaterial uptake. Our summary helps to realize why these environmental sentinels are beneficial to study the potential detrimental effects of nanomaterials.

AbstractWrecks play an important role in enhancing marine biodiversity. SCUBA diving video-samplings were performed on eight wrecks, including seven shipwrecks and a sunken airplane, scattered over 180 km along the Ligurian coastline in the northwestern Mediterranean Sea, in water depths of 30‐65 m. Differences in composition of macrobenthic communities were found to be related to the bottom sedimentology and the composition and geometry of the investigated structures. The iron, flat, and even substrata of shipwrecks were dominated by Bivalvia (Ostrea edulis) and Anthozoa (Corynactis viridis), whereas the aluminum, cage-like structure of the airplane was dominated by massive sponges and bryozoans. Furthermore, abundances of some macrobenthic invertebrates were greater on the wrecks than those observed on natural, rocky substrates. Because of the increasing popularity of use of video and photo digital apparatuses among recreational divers, combined with wreck resistance to corrosion, artificial geometries and available surfaces that facilitate species’ settlement and growth, data from wrecks consisting of images and related metadata (depth, date, water temperature, etc.) could provide a new opportunity for monitoring Mediterranean marine biodiversity.

Abstract The comparative medicine approach, as applied to the study of laboratory animals for the betterment of human health, has resulted in important medical and scientific progress. Much of what is known about the human health risks of many toxic and infectious hazards present in the environment derives from experimental studies in animals and observational (epidemiological) studies of exposed human populations. Yet there is a third source of “in vivo” knowledge about host-environment interactions that may be underused and -explored: the study of diseases in naturally occurring animal populations that may signal potential human health threats. Just as canaries warned coal miners of the risk of toxic gases, other nonhuman animals, due to their greater susceptibility, environmental exposure, or shorter life span, may serve as “sentinels” for human environmental health hazards. Traditionally, communication between human and animal health professionals about cross-species sentinel events has been limited, but progress in comparative genomics, animal epidemiology, and bioinformatics can now provide an enhanced forum for such communication. The “One Health” concept involves moving toward a comparative clinical approach that considers “shared risks” between humans and animals and promotes greater cooperation and collaboration between human and animal health professionals to identify and reduce such risks. In doing so, it also creates new opportunities for the field of comparative medicine that can supplement traditional laboratory animal research.

SUMMARYSurveying and declaring disease freedom in wildlife is difficult because information on population size and spatial distribution is often inadequate. We describe and demonstrate a novel spatial model of wildlife disease-surveillance data for predicting the probability of freedom of bovine tuberculosis (caused by Mycobacterium bovis) in New Zealand, in which the introduced brushtail possum (Trichosurus vulpecula) is the primary wildlife reservoir. Using parameters governing home-range size, probability of capture, probability of infection and spatial relative risks of infection we employed survey data on reservoir hosts and spillover sentinels to make inference on the probability of eradication. Our analysis revealed high sensitivity of model predictions to parameter values, which demonstrated important differences in the information contained in survey data of host-reservoir and spillover-sentinel species. The modelling can increase cost efficiency by reducing the likelihood of prematurely declaring success due to insufficient control, and avoiding unnecessary costs due to excessive control and monitoring.

Invasive plant pests and pathogens pose a considerable threat to plant health worldwide. With increasing globalisation of trade in plants and plant material, and the effects of climate change, this threat is predicted to continue to rise. In recent years, there has been a sharp increase in the number of these harmful invasive organisms which cause large-scale environmental and economic damage. A significant issue in managing this threat is predicting which organisms will pose a threat in the future. Sentinel plants are individuals found outside their native ranges that can be surveyed for damage by organisms they would not otherwise encounter. Monitoring plant sentinels can build knowledge and understanding of pest/host relationships to support the development of management plans and risk assessments. Botanic gardens and arboreta, whose collections are estimated to include 30–40 per cent of all known plant species, many of which are exotic, are unique and under-utilised resources that can support sentinel research. The International Plant Sentinel Network (IPSN) consists of botanic gardens and arboreta, National Plant Protection Organisations (NPPOs) and plant health scientists who collaborate to provide an early-warning system for new and emerging plant pests and pathogens. Members provide scientific evidence to NPPOs to inform plant health activities and thus help safeguard susceptible plant species. In the UK, the IPSN conducts research activities prioritised by a Research and Development committee and preliminary findings of recent research activities are outlined in this paper. The IPSN also focuses on increasing knowledge and awareness, seeking best practice, developing standardisedapproaches and providing training materials and methodologies for monitoring and surveying to enable gardens to contribute to sentinel research. Through multi-disciplinary collaboration and information sharing the IPSN aims to reduce the risk that alien invasive pests and pathogens pose to global plant health.

Abstract In this study, the authors determined whether vvIBDV could be transmitted from chickens to pigeons and vice versa, and the relative severity of the lesions in the two species. Thirty 3-to 6-week-old pigeons and thirty 3-week-old chickens were grouped as follows: A (10 uninoculated pigeons), B (10 inoculated pigeons+10 sentinel chickens), C (10 inoculated chickens+10 sentinel pigeons) and D (10 uninoculated chickens). Inoculated birds were administered 0.20 mL of vvIBDV (titre of 109.76 CID/mL) followed by introduction of their respective sentinels post-inoculation. Post-inoculation/exposure (pi/ pe), dead birds were necropsied, organs grossly examined, weighed, and sections processed for histopathology. Results revealed mild, gross and histopathological lesions in pigeons at 7 and 14 dpi/dpe. In chickens, gross and histopathological lesions were severe at 3 and 4 dpi/dpe, moderate at 7 dpi/dpe and mild at 14 dpi/dpe. Carcass weight showed no statistical difference (P > 0.05) in all pigeons, but was statistically higher in uninoculated compared to inoculated and sentinel chickens. Relative weight (RW) of the liver was significantly lower at 14 dpi/ dpe in pigeons. In chickens, RW of the bursa of Fabricius (BF) was significantly higher in inoculated and sentinel at 3 and 4 dpi/dpe. In conclusion, there was transmission of vvIBDV from pigeons to chickens and pathological changes due to vvIBDV infection were less severe in pigeons than in chickens.

Climate change and other human activities are causing profound effects on marine ecosystem productivity. We show that the breeding success of seabirds is tracking hemispheric differences in ocean warming and human impacts, with the strongest effects on fish-eating, surface-foraging species in the north. Hemispheric asymmetry suggests the need for ocean management at hemispheric scales. For the north, tactical, climate-based recovery plans for forage fish resources are needed to recover seabird breeding productivity. In the south, lower-magnitude change in seabird productivity presents opportunities for strategic management approaches such as large marine protected areas to sustain food webs and maintain predator productivity. Global monitoring of seabird productivity enables the detection of ecosystem change in remote regions and contributes to our understanding of marine climate impacts on ecosystems.

SUMMARYPatagonia in southern South America is among the few world regions where direct human impact is still limited but progressively increasing, mainly represented by tourism, farming, fishing and mining activities. The sanitary condition of Patagonian wildlife is unknown, in spite of being critical for the assessment of anthropogenic effects there. The aim of this study was the characterization ofSalmonella entericastrains isolated from wild colonies of Magellanic penguins (Spheniscus magellanicus) located in Magdalena Island and Otway Sound, in Chilean Patagonia. Eight isolates ofSalmonellawere found, belonging to Agona and Enteritidis serotypes, with an infection rate of 0·38%. Resistance to ampicillin, cefotaxime, ceftiofur and tetracycline antimicrobials were detected, and some of these strains showed genotypic similarity withSalmonellastrains isolated from humans and gulls, suggesting inter-species transmission cycles and strengthening the role of penguins as sanitary sentinels in the Patagonian ecosystem.

Cryptococcosis is typically a sporadic disease that affects a broad range of animal species globally. Disease is a consequence of infection with members of the Cryptococcus neoformans or Cryptococcus gattii species complexes. Although cryptococcosis in many domestic animals has been relatively well-characterized, free-living wildlife animal species are often neglected in the literature outside of occasional case reports. This review summarizes the clinical presentation, pathological findings and potential underlying causes of cryptococcosis in various other animals, including terrestrial wildlife species and marine mammals. The evaluation of the available literature supports the hypothesis that anatomy (particularly of the respiratory tract), behavior and environmental exposures of animals play vital roles in the outcome of host–pathogen–environment interactions resulting in different clinical scenarios. Key examples range from koalas, which exhibit primarily C. gattii species complex disease presumably due to their behavior and environmental exposure to eucalypts, to cetaceans, which show predominantly pulmonary lesions due to their unique respiratory anatomy. Understanding the factors at play in each clinical scenario is a powerful investigative tool, as wildlife species may act as disease sentinels.

Foraging in groups provides many benefits that are not necessarily experienced the same way by all individuals. I explore the possibility that foraging mode, the way individuals exploit resources, varies as a function of spatial position in the group, reflecting commonly occurring spatial differences in predation risk. I show that semipalmated sandpipers ( Calidris pusilla ), a social foraging avian species, tended to adopt a riskier foraging mode in the central, more protected areas of their groups. Central birds effectively used the more peripheral group members as sentinels, allowing them to exploit a wider range of resources within the same group at the same time. This finding provides a novel benefit of living in groups, which may have a broad relevance given that social foraging species often exploit a large array of resources.

The green turtle Chelonia mydas is a widely distributed, slowly maturing species with a complex life cycle, using both oceanic and coastal environments. The species is exposed to different threats and is considered an environmental sentinel that indicates variation among, and the severity of hazards to marine ecosystems. This study aimed to describe both anthropogenic impacts, and infectious and parasitic diseases in C. mydas - including cases along the Brazilian coast - and implications for conservation. Bycatch is reported as the main threat to the conservation of this species, followed by debris ingestion, collisions with boats, dredging, and chronic environmental contamination. All of these impacts may directly or indirectly cause death, by facilitating contact with pathological agents and by increasing vulnerability to secondary diseases. The pathological agents associated with lesions include viruses, bacteria, fungi, protozoa, and helminths. Fibropapillomatosis is an example of a chronic disease characterized by cutaneous and visceral tumors that affects mostly juvenile C. mydas worldwide and is associated with the Chelonid herpesvirus 5. The bacterias Vibrio alginolyticus, Aeromonas hydrophila, and Pseudomonas fluorescens are found in the aquatic environment and among C. mydas lesions in various organs. Trematode adults and eggs of the family Spirorchiidae are also frequent in systemic cardiovascular diseases of C. mydas. The direct impacts of anthropogenic activities and diseases are synergistic and may affect the specie’s health and conservation. Therefore, the monitoring and systematic diagnosing of diseases and causes of death - including necropsy, histopathology, and molecular exams - are necessary to assess a population’s health, to support appropriate decisions of coastal management and to target future research topics that optimize C. mydas conservation.

SUMMARYSentinel species are increasingly used by disease managers to detect and monitor the prevalence of zoonotic diseases in wildlife populations. Characterizing home-range movements of sentinel hosts is thus important for developing improved disease surveillance methods, especially in systems where multiple host species co-exist. We studied ranging activity of major hosts of bovine tuberculosis (TB) in an upland habitat of New Zealand: we compared home-range coverage by ferrets (Mustela furo), wild deer (Cervus elaphus), feral pigs (Sus scrofa), brushtail possums (Trichosurus vulpecula) and free-ranging farmed cattle (Bos taurus). We also report in detail the proportional utilization of a seasonal (4-monthly) range area for the latter four species. Possums covered the smallest home range (<30 ha), ferrets covered ∼100 ha, pigs ∼4 km2, deer and cattle both >30 km2. For any given weekly period, cattle, deer and pigs were shown to utilize 37–45% of their estimated 4-month range, while possums utilized 62% during any weekly period and 85% during any monthly period of their estimated 4-month range. We suggest that present means for estimating TB detection kernels, based on long-term range size estimates for possums and sentinel species, probably overstate the true local surveillance coverage per individual.

Marine mammals are sentinels for the marine ecosystem and threatened by numerous factors including infectious diseases. One of the most frequently isolated bacteria are beta-hemolytic streptococci. However, knowledge on ecology and epidemiology of streptococcal species in marine mammals is very limited. This review summarizes published reports on streptococcal species, which have been detected in marine mammals. Furthermore, we discuss streptococcal transmission between and adaptation to their marine mammalian hosts. We conclude that streptococci colonize and/or infect marine mammals very frequently, but in many cases, streptococci isolated from marine mammals have not been further identified. How these bacteria disseminate and adapt to their specific niches can only be speculated due to the lack of respective research. Considering the relevance of pathogenic streptococci for marine mammals as part of the marine ecosystem, it seems that they have been neglected and should receive scientific interest in the future.

Although the majority of publications on extant nonmarine ostracode species in North America are concerned with lacustrine settings, many species that are potentially valuable as indicators of water quality changes live in non-lacustrine settings. Ostracode distributions in 157 springs, wetlands and streams in the United States are examined here in order to assess 1) species richness, 2) association with physical and chemical parameters of their habitats and 3) the presence of potentially useful biomonitors and environmental sentinels. The 157 non-lacustrine sites are a subset of a large database (North American Non-marine Ostracode Database: NANODe version 1) consisting of 611 mostly lacustrine sites with ostracode species, presence-absence data, hydrochemistry and climate data (Forester et al., in review). Of the 89 species represented in NANODe version 1, 51 species are found in springs, 59 species are found in wetlands and only 15 species are found in streams. Many species are found in at least two of these habitats and some in all three. Principal Components Analysis of these 157 sites indicates that 71% of the variance is explained by salinity (total ionic concentration), alkalinity and temperature, a result consistent with previously published analyses of natural water. Cluster analysis shows that spring species are most strongly tied to temperature, whereas wetlands and streams are most strongly tied to ionic composition. Three species are found to be potentially valuable biomonitors: Cavernocypris wardi in springs, Fabaeformiscandona rawsoni in wetlands and Physocypria globula in streams.

Rapid anthropogenic production of CO2 has driven the carbonate chemistry of the sea, causing lowered pH in surface waters. Increasingly, scientists are called on to study ocean acidification and its effects. The ‘minor’ phylum Bryozoa shows considerable potential in understanding temperate southern hemisphere shelf carbonate dynamics, thus complementing tropical studies based mainly on corals. Lowered pH affects skeletons differently depending on their composition, but skeletons are even more strongly affected by morphology. Different bryozoans will manifest the effects of acidification at different times, thus some particularly vulnerable species may act as ‘canaries’ providing an early warning for some shelf communities, such as bryozoan-dominated thickets. A carbonate budget based on several studies of the bryozoan Adeonellopsis in Doubtful Sound, New Zealand, shows that increasing dissolution pressure in cool temperate environments dramatically reduces sediment accumulation rates. Bryozoan shelf carbonate sediments, which blanket the southern shelves of New Zealand and Australia, may serve as biological saturometers, monitoring the effects of acidification over shelf depths. Whether acting as canaries, models or sentinels, bryozoans have great potential to provide insight into the next global challenge: ocean acidification.

Monitoring infectious diseases is a crucial part of preventive veterinary medicine in zoological collections. This zoo environment contains a great variety of animal species that are in contact with wildlife species as a potential source of infectious diseases. Wild birds may be a source of West Nile virus (WNV) and Usutu (USUV) virus, which are both emerging pathogens of rising concern. The aim of this study was to use zoo animals as sentinels for the early detection of WNV and USUV in Slovenia. In total, 501 sera from 261 animals of 84 animal species (including birds, rodents, lagomorphs, carnivores, ungulates, reptiles, equids, and primates) collected for 17 years (2002–2018) were tested for antibodies to WNV and USUV. Antibodies to WNV were detected by indirect immunofluorescence tests in 16 (6.1%) of 261 animals representing 10 species, which were sampled prior to the first active cases of WNV described in 2018 in Slovenia in humans, a horse, and a hooded crow (Corvus cornix). Antibodies to USUV were detected in 14 out of 261 animals tested (5.4%) that were positive prior to the first positive cases of USUV infection in common blackbirds (Turdus merula) in Slovenia. The study illustrates the value of zoological collections as a predictor of future emerging diseases.

PRRs are sentinels of the innate immune system, with TLRs being the most important. Assays for TLR ligand interactions have been used to gain insights into their function and signaling pathways. As significant differences exist between species with regard to ligand recognition, it is necessary to adapt these tools for TLRs of other species. In the present work, we describe a species-specific cell-based assay adapted for the analysis of single PRRs. Human embryonic kidney 293T cells were stably transfected with the NF-κB-inducible reporter gene secreted embryonic alkaline phosphatase (SEAP) together with bovine TLR2. We compared the SEAP response with an existing luciferase NF-κB reporter assay for correlation with IL-8 production. A dose-dependent response was detected upon stimulation using both methods with good correlation to IL-8 secretion. Lower stimulant concentrations were detected by SEAP assay than IL-8 secretion. The luciferase assay produced high non-specific background for all ligand concentrations. Of all assays tested, we found the bovine-specific SEAP reporter assay to be the most convenient and delivered results in the shortest time. The developed reporter cell line would lend well to rapid, high-throughput TLR ligand screening for cattle.

Brucella abortus, B. canis, and pathogenic Leptospira are zoonotic pathogens that infect humans, as well as domestic and wild animals. In wild canids, they may affect their fertility and reproduction, threatening their conservation. Wild canids play a crucial role in the environment as meso- and top-predators and environmental sentinels for zoonotic pathogens. In Chile, three species of wild canids are present, and due to changes in land use and environmental dynamics, it is of utmost relevance to determine the role of these species in the epidemiology of brucellosis and leptospirosis. This study aimed to detect the exposure to B. abortus, B. canis, and pathogenic Leptospira by serologic, bacteriologic, and molecular techniques in native foxes from rehabilitation and exhibition centers in Central Chile. Forty-six blood samples were obtained from Lycalopex culpaeus and L. griseus, detecting 10.9% of seropositivity to B. canis and 7.7% to L. Javanica. No seropositivity was seen for B. abortus. Exposure was not registered by culture and qPCR in any of the sampled animals. Our findings are the first register of exposure to any Brucella species in wild canids in Chile and highlight the need to establish surveillance programs of these emerging pathogens.

AbstractThe polar cod (Boreogadus saida) and the Antarctic silverfish (Pleuragramma antarctica) are pelagic fish endemic to the Arctic and Antarctica sea, respectively. Both species are abundant and play a central role as midtrophic wasp-waist species in polar ecosystems. Due to their biological and ecological characteristics (small size, complex life histories, relatively short generation cycles, movement capability, planktivorous diet, and importance as prey), the polar cod and the Antarctic silverfish are potentially good sentinels of ecosystem change. Changes in polar zooplankton communities are well documented. How changes impact ecosystems as a whole largely depend on the degree of diet specialization and feeding flexibility of midtrophic species. Here, we provide the ecomorphological characterization of polar cod and Antarctic silverfish feeding performances. A comparative functional ecology approach, based on the analysis of morpho-anatomical traits, including calculation of suction index and mechanical advantage in jaw closing, was applied to profile the feeding modes and flexibility of the two species. Ecomorphological evidence supports differences in food acquisition: the polar cod appears able to alternate particulate ram-suction feeding to a pump filter feeding, and the Antarctic silverfish results be both a particulate ram and a tow-net filter feeder. Both species exhibit opportunistic feeding strategies and appear able to switch feeding mode according to the abundance and size of the available prey, which is a clue of potential resilience to a changing environment.

Floodplain and swamp forests are undergoing extensive changes due to altered flow regimes, invasive species, logging, and various land use changes. These changes often go unnoticed due to the absence of adequate baseline data and monitoring. Using a data set from 55 years ago, we resampled 50 lowland forest stands in southern Wisconsin to assess changes in forest overstory composition, structure, diversity, and dominant species abundances. We also applied univariate and multivariate analyses to determine whether these changes varied between dam-regulated and unregulated rivers, tree species with different flooding tolerances, the presence of logging, and variations in edaphic and hydrologic variables. Although these forests display various types of resilience, their forest canopies are substantially different from 55 years ago, reflecting shifts in hydrology and the impacts of disease. On average, these forests have retained the same local (alpha) diversity but have converged in species composition (declined in beta diversity). They are now composed of more and smaller trees. Along the unregulated rivers, colonizing species have declined while later successional flood-tolerant species have increased. In the aftermath of Dutch elm disease, Ulmus spp. have greatly declined in abundance and size. Species with less flooding tolerance have generally increased across sites, especially along dam-regulated rivers. Because they are subject to chronic disturbances that reset succession, floodplain forests may respond more readily to shifts in disturbances regimes. Such forests may therefore serve as sentinels for forecasting the types of change that we can expect to unfold more gradually in upland forests.

Brazilian spotted fever (BSF) is a vector-borne zoonosis caused by Rickettsia rickettsii bacteria. Dogs can be host sentinels for this bacterium. The aim of the study was to determine the presence of antibodies against Rickettsia spp. in dogs from the city of São José dos Pinhais, State of Paraná, Southern Brazil, where a human case of BSF was first reported in the state. Between February 2006 and July 2007, serum samples from 364 dogs were collected and tested at 1:64 dilutions by indirect immunofluorescence assay (IFA) against R. rickettsii and R. parkeri. All sera that reacted at least to one of Rickettsia species were tested against the six main Rickettsia species identified in Brazil: R. rickettsii, R. parkeri, R. bellii, R. rhipicephali, R. amblyommii and R. felis. Sixteen samples (4.4%) reacted to at least one Rickettsia species. Among positive animals, two dogs (15.5%) showed suggestive titers for R. bellii exposure. One sample had a homologous reaction to R. felis, a confirmed human pathogen. Although Rickettsia spp. circulation in dogs in the area studied may be considered at low prevalence, suggesting low risk of human infection, the present data demonstrate for the first time the exposure of dogs to R. bellii and R. felis in Southern Brazil.

SummaryBetween 1998 and 2011 we monitored the winter ranging behaviour of eight female Saker Falcons Falco cherrug fitted with satellite-received transmitters. Our tracking revealed that the winter home range area occupied by individual Saker Falcons varied greatly (median = 166 km2, range = 5-18,469 km2). A random forest model showed that Saker Falcons wintering on the Qinghai-Tibetan Plateau preferentially occupied areas with rich grassland (cover > 50%) on high altitude plateaus (4,000–5,000 m asl) with low levels of anthropogenic influence. Plant biomass in rich grasslands can support high winter densities of plateau pikas Ochotona curzoniae, which likely explains the preference exhibited by Saker Falcons for grassland cover > 50%. Factors influencing the abundance and distribution of this ‘keystone’ prey species are likely to have an effect on Saker Falcons and other predatory species. A key element of rangeland management on the Qinghai-Tibetan plateau has been the establishment of extensive protected areas as part of a strategy to balance economic and social development with the requirement of sustainably managing water resources, maintaining rangelands for pastoralists and conserving biological diversity. Wide ranging predatory species, such as the Saker Falcon, can be useful indicators of biodiversity in protected areas and act as ‘sentinels’ for anthropogenic changes that may impact many different taxa.

Skin-associated microorganisms have been shown to play a role in immune function and disease of humans, but are understudied in marine mammals, a diverse animal group that serve as sentinels of ocean health. We examined the microbiota associated with 75 epidermal samples opportunistically collected from nine species within four marine mammal families, including: Balaenopteridae (sei and fin whales), Phocidae (harbour seal), Physeteridae (sperm whales) and Delphinidae (bottlenose dolphins, pantropical spotted dolphins, rough-toothed dolphins, short-finned pilot whales and melon-headed whales). The skin was sampled from free-ranging animals in Hawai‘i (Pacific Ocean) and off the east coast of the United States (Atlantic Ocean), and the composition of the bacterial community was examined using the sequencing of partial small subunit (SSU) ribosomal RNA genes. Skin microbiotas were significantly different among host species and taxonomic families, and microbial community distance was positively correlated with mitochondrial-based host genetic divergence. The oceanic location could play a role in skin microbiota variation, but skin from species sampled in both locations is necessary to determine this influence. These data suggest that a phylosymbiotic relationship may exist between microbiota and their marine mammal hosts, potentially providing specific health and immune-related functions that contribute to the success of these animals in diverse ocean ecosystems.

High-mountain lakes are among the most comparable ecosystems globally and recognized sentinels of global change. The present study pursued to identify how the benthic macroinvertebrates (BMI) communities of two tropical, high mountain lakes, El Sol and La Luna, Central Mexico, have been affected by global/regional environmental pressures. We compared the environmental characteristics and the BMI communities between 2000–2001 and 2017–2018. We identified three principal environmental changes (the air and water temperature increased, the lakes’ water level declined, and the pH augmented and became more variable), and four principal ecological changes in the BMI communities [a species richness reduction (7 to 4), a composition change, and a dominant species replacement all of them in Lake El Sol, a species richness increase (2 to 4) in Lake La Luna, and a drastic reduction in density (38% and 90%) and biomass (92%) in both lakes]. The air and water temperature increased 0.5 °C, and lakes water level declined 1.5 m, all suggesting an outcome of climate change. Contrarily to the expected acidification associated with acid precipitation, both lakes deacidified, and the annual pH fluctuation augmented. The causes of the deacidification and the deleterious impacts on the BMI communities remained to be identified.

Dissemination of antibiotic resistance (AR) in marine environments is a global concern with a propensity to affect public health and many ecosystems worldwide. We evaluated the use of sea turtles as sentinel species for monitoring AR in marine environments. In this field, antibiotic-resistant bacteria have been commonly identified by using standard culture and sensitivity tests, leading to an overrepresentation of specific, culturable bacterial classes in the available literature. AR was detected against all major antibiotic classes, but the highest cumulative global frequency of resistance in all represented geographical sites was against the beta-lactam class by a two-fold difference compared to all other antibiotics. Wastewater facilities and turtle rehabilitation centres were associated with higher incidences of multidrug-resistant bacteria (MDRB) accounting for an average of 58% and 49% of resistant isolates, respectively. Furthermore, a relatively similar prevalence of MDRB was seen in all studied locations. These data suggest that anthropogenically driven selection pressures for the development of AR in sea turtles and marine environments are relatively similar worldwide. There is a need, however, to establish direct demonstrable associations between AR in sea turtles in their respective marine environments with wastewater facilities and other anthropogenic activities worldwide.

ABSTRACT The usual agent of visceral leishmaniasis in the Old World is Leishmania donovani, which typically produces systemic diseases in humans and mice. L. donovani has developed efficient strategies to infect and persist in macrophages from spleen and liver. Dendritic cells (DC) are sentinels of the immune system. Following recognition of evolutionary conserved microbial products, DC undergo a maturation process and activate antigen-specific naïve T cells. In the present report we provide new insights into how DC detect Leishmania in vivo. We demonstrate that in both C57BL/6 and BALB/c mice, systemic injection of L. donovani induced the migration of splenic DC from marginal zones to T-cell areas. During migration, DC upregulated the expression of major histocompatibility complex II and costimulatory receptors (such as CD40, CD80, and CD86). Leishmania-induced maturation requires live parasites and is not restricted to L. donovani, as L. braziliensis, L. major, and L. mexicana induced a similar process. Using a green fluorescent protein-expressing parasite, we demonstrate that DC undergoing maturation in vivo display no parasite internalization. We also show that L. donovani-induced DC maturation was partially abolished in MyD88-deficient mice. Taken together, our data suggest that Leishmania-induced DC maturation results from direct recognition of Leishmania by DC, and not from DC infection, and that MyD88-dependent receptors are implicated in this process.

Toscana phlebovirus (TOSV) and Sicilian phlebovirus (SFSV) are endemic in the Mediterranean area where they are transmitted to humans by infected sandflies. Vertebrates of several species have been postulated to act as reservoirs of these viruses, but convincing evidence is still awaited. Among them, bats have been suggested, however documented evidence is lacking. Here we tested a total of 329 bats belonging to eight species collected from twelve localities in southern Spain for the presence of neutralizing antibodies specific to TOSV and SFSV. Positive sera were detected in Schreiber’s long-fingered bat (Miniopterus schreibersii), mouse-eared Myotis (Myotis myotis), European free-tailed bat (Tadarida teniotis), and common serotine (Eptesicus serotinus) with the latter showing the highest prevalence rates for SFSV (22.6%) and TOSV (10%). There was no difference between females and males. Results suggest that bats are not likely to play a major role in the natural cycle of these two sandfly-borne phleboviruses. However, large breeding colonies of bats can be used as sentinels for surveillance of the presence of such viruses in a given locality. In addition, capture–recapture studies should be initiated in order to understand better the dynamics of TOSV and SFSV in bat populations.

AbstractThe aim of this survey was to study two Hymenolepididae species in urban rodents, Rattus rattus and Rattus norvegicus, and to analyse factors that favour their presence in the environment and pose a sanitary risk. Hymenolepis diminuta and Rodentolepis nana were found in R. rattus and R. norvegicus in different proportions. Values of prevalence, mean abundance and mean intensity were recorded, and new morphometric characters were described, adding to previously known information. No significant differences were found. However, the results revealed that there is a tendency for these parasites to develop in areas with deficient socio-structural conditions and in water bodies. This study thereby shows that certain areas on the periphery of the Gran La Plata favour the existence of rodents, which act as sentinels of zoonotic diseases, and stresses the need to take action to minimize them in order to avoid putting human and animal health at risk.

Inter-annual variations in the diets of seabirds are often a reflection of resource availability, with population dynamics and community structure implications. We investigated the trophic niche of Magellanic penguins Spheniscus magellanicus during the pre-molt stage in 6 years (2009 and 2013-2017) at Martillo Island, Beagle Channel, Argentina, using carbon and nitrogen stable isotope analysis of feathers. We found higher values in 2009 compared to the other years for both isotopes and estimated different proportions of prey in the diet of Magellanic penguins throughout the years. In 2009, penguins consumed mainly a mixture of benthic and benthopelagic fish, whereas from 2013 to 2017, the proportion of the pelagic form of squat lobster Munida gregaria, considered a key species in the Beagle Channel, increased over time and nearly dominated diets in 2017. Our results confirm that Magellanic penguins act as sentinels, reflecting shifts in the marine community of the Beagle Channel via inter-annual variation in their trophic niche and diet composition.

Abstract Rickettsia spp. bacteria are responsible for tick-borne diseases worldwide, mostly maintained by rickettsial amplifiers capybaras in Brazilian endemic areas. The campus of the University of São Paulo, in southeastern Brazil, is an area endemic for Brazilian spotted fever (BSF), with high density of capybaras and Amblyomma spp., along with confirmed human cases. Besides capybaras, the university has also an in-campus high population of sheltered and free-roaming cats. Accordingly, the aim of this study was to determine the prevalence and characteristics associated with Rickettsia rickettsii, Rickettsia parkeri and Rickettsia felis exposure among cats in a BSF-endemic area. Out of 51 cats sampled, 23/35 shelter (65.7%) and 5/16 free-roaming (31.2%) were positive (titers ≥ 64) for at least one Rickettsia species. Ticks species were present in 3/16 free-roaming cats (18.8%), consisting of Amblyomma spp., nymphs of Amblyomma sculptum and adult Rhipicephalus sanguineus sensu lato. Despite sharing the capybaras environment, the seropositivity among the free-roaming and shelter cats was lower than owned cats in other endemic areas. Whether equally or less exposed to rickettsial infection, compared with owned cats in endemic areas, free-roaming and shelter cats may be used as environmental sentinels for human exposure to rickettsiae in such areas.

Abstract In social species, conspecific outsiders present various threats to groups and their members. These out-group threats are predicted to affect subsequent within-group interactions (e.g., affiliation and aggression) and individual behavior (e.g., foraging and vigilance decisions). However, experimental investigations of such consequences are rare, especially in natural conditions. We used field-based call playbacks and fecal presentations on habituated wild dwarf mongooses (Helogale parvula)—a cooperatively breeding, territorial species—to examine postinteraction responses to the simulated threat of a rival group. Dwarf mongooses invested more in grooming of groupmates, foraged closer together, and more regularly acted as sentinels (a raised guard) after encountering indicators of rival-group presence compared to control conditions. These behavioral changes likely arise from greater anxiety and, in the case of increased vigilance, the need to seek additional information about the threat. The influence of an out-group threat lasted at least 1 h but individuals of different dominance status and sex responded similarly, potentially because all group members suffer costs if a contest with rivals is lost. Our results provide field-based experimental evidence from wild animals that out-group threats can influence within-group behavior and decision making, and suggest the need for greater consideration of the lasting impacts of social conflict.

ABSTRACT The rabbit model of tuberculosis is attractive because of its pathophysiologic resemblance to the disease in humans. Rabbits are naturally resistant to infection but may manifest cavitary lung lesions. We describe here a novel approach that utilizes presensitization and bronchoscopic inoculation to reliably produce cavities in the rabbit model. With a fixed inoculum of bacilli, we were able to reproducibly generate cavities by using Mycobacterium bovis Ravenel, M. bovis AF2122, M. bovis BCG, M. tuberculosis H37Rv, M. tuberculosis CDC1551, and the M. tuberculosis CDC1551 ΔsigC mutant. M. bovis infections generated cavitary CFU counts of 106 to 109 bacilli, while non-M. bovis species and BCG yielded CFU counts that ranged from 104 to 108 bacilli. Extrapulmonary dissemination was almost exclusively noted among rabbits infected with M. bovis Ravenel and AF2122. Though all of the species yielded secondary lesions at intrapulmonary sites, M. bovis infections led to the most apparent gross pathology. Using the M. tuberculosis icl and dosR gene expression patterns as molecular sentinels, we demonstrated that both the cavity wall and cavity lumen are microenvironments associated with hypoxia, upregulation of the bacterial dormancy program, and the use of host lipids for bacterial catabolism. This unique cavitary model provides a reliable animal model to study cavity pathogenesis and extrapulmonary dissemination.

Capybaras (Hydrochaeris hydrochaeris) are among the main hosts of Amblyomma spp. ticks, which is able to transmit Rickettsia species to human beings and animals. Since they are often infested with potential vector ticks, capybaras may be used as sentinels for rickettsiosis, such as the Brazilian Spotted Fever. The aim of the present study was to determine the prevalence of antibodies against Rickettsia spp. using the indirect immunofluorescence assay (IFA) in 21 free-ranging and 10 captive animals from the Zoological Park of the 'Bela Vista Biological Sanctuary' (BVBS), Itaipu Binational, Foz do Iguaçu, Southern Brazil. Antigens of six rickettsial species already identified in Brazil (Rickettsia rickettsii, R. parkeri, R. bellii, R. rhipicephali, R. amblyommii and R. felis) were used for IFA. Ticks from each capybara were collected for posterior taxonomic identification. A total of 19 (61.3%) samples reacted to at least one of tested species. Seropositivity was found in 14 (45.2%), 12 (38.7%), 5 (16.1%), 4 (12.9%), 3 (9.7%) and 3 (9.7%) animals for R. rickettsii, R. bellii, R. parkeri, R. amblyommii, R. felis and R. rhipicephali, respectively. Two captive capybaras presented suggestive titers of R. rickettsii infection and one sample showed homologous reaction to R. parkeri. Only one free-ranging capybara presented evidence R. bellii infection. Ticks collected on capybaras were identified as Amblyomma dubitatum e Amblyomma sp. Results evidenced the rickettsial circulation in the area, suggesting a potential role of capybaras on bacterial life cycle.

Abstract Genotoxic chemicals, through damage and alteration of the genetic material of wild organisms, pose significant threats to the persistence of wild animal populations. Their damaging effects can ultimately impair the health of the ecosystem and its provision of services to human society. Bird species are good candidates for the role of sentinels of the effects of genotox-ins, thanks to (i) the diversity of their ecological niches, (ii) their ubiquity across environments, (iii) their conspicuousness, abundance and approachability, together with (iv) their well-known life histories and the availability of historical data series. Avian diversity increases the likelihood that adequate model species be available for monitoring genotoxicants and assessing their impact. This paper reviews the methods utilized by genetic ecotoxicological studies of wild birds, highlighting their benefits and shortcomings. It also summarizes the genetic ecotoxicological studies so far conducted. In spite of a paucity of studies, several classes of genotoxicants have already been investigated across a variety of species and environments, thus supporting the versatility of birds as monitors of genotoxic contamination. Future technical advancements and applications are suggested, with particular reference to the analysis of mutational events, gene expression and methylation patterns. Finally, I argue that the development of avian genetic ecotoxicology will contribute to the understanding of natural variation in the underlying machinery for coping with DNA damage and oxidative stress, both of which are increasingly recognized as proximate factors in the evolution of life history adaptations.

SUMMARYRift Valley fever virus (RVFV) is an emerging pathogen of major concern throughout Africa and the Arabian Peninsula, affecting both livestock and humans. In the past recurrent epidemics were reported in Mauritania and studies focused on the analysis of samples from affected populations during acute outbreaks. To verify characteristics and presence of RVFV during non-epidemic periods we implemented a multi-stage serological and molecular analysis. Serum samples of small ruminants, cattle and camels were obtained from Mauritania during an inter-epidemic period in 2012–2013. This paper presents a comparative analysis of potential variations and shifts of antibody presence and the capability of inter-epidemic infections in Mauritanian livestock. We observed distinct serological differences between tested species (seroprevalence: small ruminants 3·8%, cattle 15·4%, camels 32·0%). In one single bovine from Nouakchott, a recent RVF infection could be identified by the simultaneous detection of IgM antibodies and viral RNA. This study indicates the occurrence of a low-level enzootic RVFV circulation in livestock in Mauritania. Moreover, results indicate that small ruminants can preferably act as sentinels for RVF surveillance.

SUMMARYThe role that animals play in the transmission of Schistosoma japonicum to humans in the Philippines remains uncertain and prior studies have not included several species, adjustment for misclassification error and clustering, or used a cohort design. A cohort study of 2468 people providing stool samples at 12 months following praziquantel treatment in 50 villages of Western Samar, the Philippines, was conducted. Stool samples from dogs, cats, rats, and water buffaloes were collected at baseline (2003–2004) and follow-up (2005). Latent-class hierarchical Bayesian log-binomial models adjusting for misclassification errors in diagnostic tests were used. The village-level baseline and follow-up prevalences of cat, dog, and rat S. japonicum infection were associated with the 12-month cumulative incidence of human S. japonicum infection, with similar magnitude and precision of effect, but correlation between infection levels made it difficult to divide their respective effects. The cumulative incidence ratios associated with a 1% increase in the prevalence of infection in dogs at baseline and in rats at follow-up were 1·04 [95% Bayesian credible interval (BCI) 1·02–1·07] and 1·02 (95% BCI 1·01–1·04), respectively, when both species were entered in the model. Dogs appear to play a role in human schistosomiasis infection while rats could be used as schistosomiasis sentinels.

ABSTRACT The potential impact of food animals in the production environment on the bacterial population as a result of antimicrobial drug use for growth enhancement continues to be a cause for concern. Enterococci from 82 farms within a poultry production region on the eastern seaboard were isolated to establish a baseline of susceptibility profiles for a number of antimicrobials used in production as well as clinical environments. Of the 541 isolates recovered, Enterococcus faecalis (53%) and E. faecium (31%) were the predominant species, while multiresistant antimicrobial phenotypes were observed among all species. The prevalence of resistance among isolates of E. faecalis was comparatively higher among lincosamide, macrolide, and tetracycline antimicrobials, while isolates of E. faecium were observed to be more frequently resistant to fluoroquinolones and penicillins. Notably, 63% of the E. faecium isolates were resistant to the streptogramin quinupristin-dalfopristin, while high-level gentamicin resistance was observed only among the E. faecalis population, of which 7% of the isolates were resistant. The primary observations are that enterococci can be frequently isolated from the poultry production environment and can be multiresistant to antimicrobials used in human medicine. The high frequency with which resistant enterococci are isolated from this environment suggests that these organisms might be useful as sentinels to monitor the development of resistance resulting from the usage of antimicrobial agents in animal production.

The monitoring of marine species as sentinels for ecosystem health has long been a valuable tool worldwide, providing insight into how both anthropogenic pollution and naturally occurring phenomena (i.e., harmful algal blooms) may lead to human and animal dietary concerns. The marine environments contain many contaminants of anthropogenic origin that have sufficient similarities to steroid and thyroid hormones, to potentially disrupt normal endocrine physiology in humans, fish, and other animals. An appropriate understanding of the effects of these endocrine disrupting chemicals (EDCs) on forage fish (e.g., sardine, anchovy, mackerel) can lead to significant insight into how these contaminants may affect local ecosystems in addition to their potential impacts on human health. With advancements in molecular tools (e.g., high-throughput sequencing, HTS), a genomics approach offers a robust toolkit to discover putative genetic biomarkers in fish exposed to these chemicals. However, the lack of available sequence information for non-model species has limited the development of these genomic toolkits. Using HTS and de novo assembly technology, the present study aimed to establish, for the first time for Sardinops sagax (Pacific sardine), Scomber japonicas (Pacific chub mackerel) and Pleuronichthys verticalis (hornyhead turbot), a de novo global transcriptome database of the liver, the primary organ involved in detoxification. The assembled transcriptomes provide a foundation for further downstream validation, comparative genomic analysis and biomarker development for future applications in ecotoxicogenomic studies, as well as environmental evaluation (e.g., climate change) and public health safety (e.g., dietary screening).

The aim of the present study was to macroscopically and microscopically describe the main pathological findings occurring in this invasive species, in order to better understand the real risks for naïve animals and humans. The present study was conducted on Callosciurusfinlaysonii squirrels (n = 165), captured in the Maratea area and euthanatized with CO2 according to a population control of invasive species of the Italian Agriculture Ministry project (ex CIPE project) and conferred to the Department of Veterinary Medicine of Bari (Italy). Macroscopic analysis demonstrated heart, lung, and liver congestion, probably due to the euthanasia method, and variable lesions of bowel, liver, and kidney. The microscopically examination showed the presence of lymphocytic infiltration in the lower layers of the bowel, suggesting enteritis. To the best of our knowledge, this is the first systemic report of gross and microscopical anatomopathological lesions in C.finlaysonii, in South Italy. The results could be useful to fill a gap of knowledge of this species in Italy.

Neospora caninum is a protozoan parasite which affects dogs as definitive hosts and several mammalian species as intermediate hosts mainly causing abortions and central nervous system disorders. The reemerging population of cart horses for carrying recycling material in urban areas of major cities in Brazil may have an impact on disease spreading, and these animals may be used as sentinels for environmental surveillance. Thus, the present study investigated the frequency of Neospora sp. antibodies in cart horses from Curitiba and surrounding areas, Paraná State, Southern Brazil. IgG antibodies against Neospora sp. were detected using indirect fluorescence antibody test (IFAT), and titers equal to or higher than 1:50 were considered reactive. Of all samples, 14/97 (14.4%) were positive: 2/29 (6.9%) were younger than 5; 5/26 (19.2%) between 6 and 9; and 6/31 (19.4%) older than 10 years of age. One of the 11 animals with unknown age was positive (9.1%). Cart horses are likely to be more exposed to dog feces and to Neospora sp. oocyst contamination in urban settings and a lower frequency of disease in dogs may have a negative impact on horse infection risk in these areas.
