碩士<br>國立清華大學<br>生物資訊與結構生物研究所<br>98<br>Vascular patterning is a highly organized process in angiogenesis. It involves precise control of endothelial cell (EC) migration, branching, and connection. Based on previous studies, directed EC migration and branching depends on correct protrusions of ECs. However, the underlying mechanism of directional EC protrusions is still unclear. Here we propose a novel secreted protein, THSD7A, which is expressed in the zebrafish developing neuronal system, contributes to EC protrusion regulation. From our data, knockdown of THSD7A expression by injecting morpholino disrupted intersegmental vessel (ISV) sprouting of zebrafish at 30 hour-post-fertilization (hpf) stage. To acquire more information, we further examined the behavior and morphology of endothelial tip cells on the ISVs of Thsd7a-knockdown zebrafish by confocal microscopy. The data showed that morpholino knockdown of THSD7A slowed down ISV growth by misleading endothelia cells to stay longer in early stage of dynamic protrusion process, in which ECs extended numerous protrusions like a fan-shape. This phenotype implied malfunction of retracting excessive EC protrusions. Besides, ECs of ISVs formed ectopic EC branching and false connection, resulting in complex ISV pattern in 34hpf zebrafish. On the other hand, an in vitro system, 3D collagen angiogenesis assay, was constructed for investigating the specific role of THSD7A. The data from in vitro 3D collagen angiogenesis assay proved that THSD7A was related to angiogenic patterning, including EC protrusions and connections. Statistic analysis also implied that THSD7A played a dual role in angiogenesis depending on its concentration. Taken the in vivo and in vitro data together, this work suggests that THSD7A may mediate EC protrusion during angiogenic sprouting and plays an essential role in angiogenic patterning as a regular of EC branching and connection.

碩士<br>中華大學<br>電機工程學系<br>106<br>Cerebral microbleeds (CMBs) are products of local chronic blood on normal brain tissue, and the relation between CMBs and neurological diseases has been verified. The presence and quantity of CMBs in the brain are important for brain diagnosis. Physicians calculate the number and positions of micro bleeding points by means of diagnosing MRI images. The purpose of this research is to develop a computer aided detecting system which can precisely determine the CMB number and positions and reduce the time of diagnosis to assist physicians on patient observation and treatment. In the thesis we propose a method using image processing and texture analysis techniques to construct a computer-aided diagnosis system for detecting CMBs. First, the fully convolutional network (FCN) is applied to filter out all the CMB candidates from the MRI data. Second, choose an appropriate size of block for each CMB candidate. Third, calculate the 3-D discrete wavelet transform (DWT) for each CMB candidate block. Fourth, extract features form the lowest frequency band of the 3D DWT coefficients of each CMB candidate. Finally, the support vector machine (SVM) is used for training and testing. The MRI data of 20 patients were used in the research, and each subject has 150 512-by-512 MRI images. The total number of CMBs of the 20 patients is 71.

碩士<br>佛光大學<br>心理學系<br>101<br>In clinic, stroke hemorrhage has been demonstrated to cause central post-stroke pain (CPSP). However, whether the learning and memory would be affected remains inconsistent. The purposes of the present study investigated whether the ventral basal complex (VBC) of thalamus-lesion induced CPSP could impair the explicit and implicit memory. The conditioned place preference (CPP) and the water maze tests were determined respectively to test the implicit and explicit memory in the current study. All rats were microinjected collagenase into the left ventricle and destroyed the VBC of thalamus to cause the central post stroke pain symptom, and then measured the CPP effect and the spatial learning effect, respectively. The present results indicated that rats with the VBC lesion would induce the CPSP effect and reflected in stronger withdrawal responses, particularly in the right leg, in Experiment 1. Experiment 2 showed that the VBC lesions facilitated the CPP and produced a resistance effect to the CPP extinction learning, on the acquisition phase. However, the VBC lesion did not affect the spatial learning in water maze task. The present findings might imply that the VBC lesion-induced CPSP can affect the implicit memory but not the explicit memory. The data can provide some implications in clinic.
