The phage-proximal part of the long tail fibres of bacteriophage T4 consists of a trimer of the 1289 amino-acid gene product 34 (gp34). Different carboxy-terminal parts of gp34 have been produced and crystallized. Crystals of gp34(726–1289) diffracting X-rays to 2.9 Å resolution, crystals of gp34(781–1289) diffracting to 1.9 Å resolution and crystals of gp34(894–1289) diffracting to 3.0 and 2.0 Å resolution and belonging to different crystal forms were obtained. Native data were collected for gp34(726–1289) and gp34(894–1289), while single-wavelength anomalous diffraction data were collected for selenomethionine-containing gp34(781–1289) and gp34(894–1289). For the latter, high-quality anomalous signal was obtained.

Abstract:
Six new hybrid rootstocks, ‘US-1279’, ‘US-1281’, ‘US-1282’, ‘US-1283’, ‘US-1284’, and ‘US-1516’, were released from the U.S. Department of Agriculture (USDA) citrus breeding program to provide improved tree tolerance to huanglongbing (HLB), the most destructive disease facing the citrus industry in the United States and many other parts of the world. Five of these new rootstocks were released based on field performance in trials with the rootstocks propagated by stem cuttings, rather than the traditional propagation using nucellar seedlings. In this study, we evaluated the fruit, seed, and seedling characteristics of these new rootstocks, along with four other USDA rootstocks of commercial importance. The study included a determination of the percentage of true-to-type and off-type seedlings by both plant morphology and simple sequence repeat (SSR) markers. All 10 rootstocks produced an acceptable number of seeds and good seedling emergence from those seeds. The rootstocks ‘Swingle’, ‘US-802’, ‘US-812’, ‘US-1283’, ‘US-1284’, and ‘US-1516’ had a high percentage of true-to-type seedlings and correspondingly good potential to be propagated by seeds. However, no true-to-type plants were observed among seedlings from the rootstocks ‘US-1279’, ‘US-1281’, and ‘US-1282’, indicating that economical seed propagation will be impossible for these cultivars. The 10 SSR marker sets used in this study were observed to easily differentiate the 10 rootstocks studied, and readily distinguished true-to-type and off-type seedlings among progeny from all 10 rootstock clones. This study presents information of significant value for commercial nurseries involved in propagation of citrus rootstocks, and those involved in citrus rootstock breeding and development around the world. We propose the use of these 10 SSR marker sets as readily applicable for accurate identification of most citrus rootstock cultivars and their true-to-type seedlings.

Després de presentar la figura de Ramon Llull com a escriptor de referència en l’època medieval i de situar el Llibre de les Bèsties dins de la producció literària de Llull, analitzem els paratextos extratextuals i textuals de les 6 adaptacions aquí estudiades. La presentació i anàlisi d’aquests paratextos ens permet determinar, d’una banda, a quin tipus de nivell lector va adreçada cada adaptació i quin treball didàctic en pot resultar; i, d’altra banda, com el missatge que Llull transmet en aquesta obra literària pot ser copsat pel públic infantil i juvenil del segle XXI.

Abstract:
Recently, numerous tumor-suppressive microRNAs (TS-miRs) have been identified in human malignancies. Here, we attempted to identify novel TS-miRs in oral squamous cell carcinoma (OSCC). First, we transfected human OSCC cells individually with 968 synthetic miRs mimicking human mature miRs individually, and the growth of these cells was evaluated using the WST-8 assay. Five miR mimics significantly reduced the cell growth rate by less than 30%, and the miR-1289 mimic had the most potent growth inhibitory effect among these miRs. Subsequently, we assessed the in vivo growth-inhibitory effects of miR-1289 using a mouse model. The administration of the miR-1289 mimic–atelocollagen complex significantly reduced the size of subcutaneously xenografted human OSCC tumors. Next, we investigated the expression of miR-1289 in OSCC tissues using reverse transcription–quantitative PCR. The expression level of miR-1289 was significantly lower in OSCC tissues than in the adjacent normal oral mucosa. Furthermore, 15 genes were identified as target genes of miR-1289 via microarray and Ingenuity Pathway Analysis (IPA) microRNA target filtering. Among these genes, the knockdown of magnesium transporter 1 (MAGT1) resulted in the most remarkable cell growth inhibition in human OSCC cells. These results suggested that miR-1289 functions as a novel TS-miR in OSCC and may be a useful therapeutic tool for patients with OSCC.

Hypertension is considered a multi-factorial disease being an important worldwide public-health challenge due to its high frequency and concomitant risks of cardiovascular and other diseases. It has been identified as the leading risk factor for mortality, and is ranked as the third cause of disability-adjusted life-years[1]. The LASSBio-1289 compound was designed from the lead compound LASSBio-294 through the use of medicinal chemistry strategy of bioisosteric replacement of aromatic rings[2]. Many factors are involved in the task of describing the relationship between chemical structure and pharmacological activity of a prototype of a new drug. Among these factors the study of atomic-level structure can be included. The aim of this work is to carry out the structural characterization of LASSBio-1289, in solid-state. Since many NAH derivatives, as LASSBio-1289, do not form single crystals for proper structure determination, this task was overcome by using X-ray powder diffraction data and an ab initio simulated annealing procedure. This study can contribute to better understand the full pharmacodynamic profiles and physicochemical properties of this class of compounds. The LASSBio-1289 compound was synthesized following the procedure described in literature[3]. The sample was sofly hand-grinded in an agate mortar in order to get a fine powder suitable for X-ray powder diffraction analysis. The final refined values for the unit-cell parameters after the Rietveld fit were: a = 14.5119(3) Å, b = 12.1375(2) Å, c = 7.5498(1) Å, β = 91.113(1) Å, Z = 4, Z' = 1 and ρcalc = 1.44039(5) g.m-3. The goodness-of-fit indicator and R-factors were: χ2 = 1.290, RBragg = 1.749%, Rwp = 4.903%, Rexp = 3.802% and Rp = 3.661%. The crystal structure of LASSBio-1289 compound consists of four formula units per unit cell (Z = 4), accommodating one molecule in the asymmetric unit (Z' = 1). The sample was indexed as monoclinic space group (P21/c).

ABSTRACT Despite the fact that closely related bacteria can cause different levels of disease, the genetic changes that cause some isolates to be more pathogenic than others are generally not well understood. We use a combination of approaches to determine which factors contribute to the increased virulence of a Bordetella bronchiseptica lineage. A strain isolated from a host with B. bronchiseptica-induced disease, strain 1289, was 60-fold more virulent in mice than one isolated from an asymptomatically infected host, strain RB50. Transcriptome analysis and quantitative reverse transcription-PCR showed that the type III secretion system (TTSS) genes were more highly expressed by strain 1289 than strain RB50. Compared to strain RB50, strain 1289 exhibited greater TTSS-mediated cytotoxicity of a mammalian cell line. Additionally, we show that the increase in virulence of strain 1289 compared to that of RB50 was partially attributable to the TTSS. Using multilocus sequence typing, we identified another strain from the same lineage as strain 1289. Similar to strain 1289, we implicate the TTSS in the increased virulence of this strain. Together, our data suggest that the TTSS is involved in the increased virulence of a B. bronchiseptica lineage which appears to be disproportionately associated with disease. These data are consistent with the view that B. bronchiseptica lineages can have different levels of virulence, which may contribute to this species' ability to cause different severities of respiratory disease.

Acute Helicobacter pylori infection of gastric epithelial cells induces CagA oncoprotein- and peptidoglycan (SLT)-dependent mobilization of NF-κB p50 homodimers that bind to H-K-ATPase α-subunit (HKα) promoter and repress HKα gene transcription. This process may facilitate gastric H. pylori colonization by induction of transient hypochlorhydria. We hypothesized that H. pylori also regulates HKα expression posttranscriptionally by miRNA interaction with HKα mRNA. In silico analysis of the HKα 3′ untranslated region (UTR) identified miR-1289 as a highly conserved putative HKα-regulatory miRNA. H. pylori infection of AGS cells transfected with HKα 3′ UTR- Luc reporter construct repressed luciferase activity by 70%, whereas Δ cagA or Δ slt H. pylori infections partially abrogated repression. Transfection of AGS cells expressing HKα 3′ UTR- Luc construct with an oligoribonucleotide mimetic of miR-1289 induced maximal repression (54%) of UTR activity within 30 min; UTR activity was unchanged by nontargeting siRNA transfection. Gastric biopsies from patients infected with cagA+ H. pylori showed a significant increase in miR-1289 expression compared with uninfected patients or those infected with cagA− H. pylori. Finally, miR-1289 expression was necessary and sufficient to attenuate biopsy HKα protein expression in the absence of infection. Taken together, these data indicate that miR-1289 is upregulated by H. pylori in a CagA- and SLT-dependent manner and targets HKα 3′ UTR, affecting HKα mRNA translation. The sensitivity of HKα mRNA 3′ UTR to binding of miR-1289 identifies a novel regulatory mechanism of gastric acid secretion and offers new insights into mechanisms underlying transient H. pylori-induced hypochlorhydria.

Tradicionalmente la historiografía ha considerado que las Cortes de Monzón de 1289 fueron las primeras Generales de la Corona de Aragón y que las ordenaciones allí aprobadas afectaron de manera común cuando menos a catalanes y aragoneses. En el presente artículo se presentan dos novedades con respecto a dicha interpretación. Por un lado, indicamos que con anterioridad, ya en época de Jaime I, hubo otras asambleas en que se congregaron miembros de los estamentos aragoneses, valencianos, catalanes y mallorquines, aunque ciertamente estas no poseían las características que posteriormente definieron las Cortes Generales de la Corona de Aragón. Por otro lado, mostramos que las constituciones aprobadas en Monzón en 1289 únicamente afectaron a Cataluña y Mallorca, mientras que las negociaciones político-fiscales con las comunidades de Aragón y Valencia se pospusieron para dos Cortes privativas posteriores, en Zaragoza y Valencia a lo largo de 1290. On the one hand, we indicate that previously, at the time of James I, there were other assemblies in which members of the Aragonese, Valencian, Catalan and Mallorcan estates gathered, although these meetings certainly did not possess the characteristics that later defined the General Corts of the Crown of Aragon. On the other hand, we show that the constitutions approved in Monzón in 1289 only affected Catalonia and Mallorca, while the political and fiscal negotiations with the communities of Aragon and Valencia were postponed for two subsequent privative Corts, in Zaragoza and Valencia throughout of 1290. In the spring of 1314, the three daughters-in-law of King Philip the Fair of France were seized as adulteresses, and two young knights, their alleged lovers, were brutally put to death at Pontoise, their property confiscated.1 The knights in question were brothers, Philippe and Gautier d’Aulnay, whose actions brought singular dishonor to their line and to their father Gautier, a faithful vassal and supporter of Count Charles of Valois, Philip the Fair’s brother and close confidant.2 Two of the king’s disgraced daughters-in-law were sent to the Norman fortress of Château-Gaillard. The oldest, Marguerite of ducal Burgundy (ca. 1289‐1315), the daughter of the late Duke Robert of Burgundy (1248‐1306) and of Saint Louis’s daughter Agnes of France († 1327), was married to Louis (1289‐1316, r. 1314‐1316), king of Navarre and heir to the throne of France. Taken with her was Blanche of Artois and comital Burgundy (1296/1297‐1325/1326), wife of the king’s third son Charles of La Marche (1294‐1328, r. 1322‐1328), and daughter of the late Count Othon of Burgundy († 1303) and of Mahaut († 1329), countess of Artois and Burgundy. Jeanne (1287/1288‐1330), Blanche’s elder sister and wife of Philip of Poitiers (1290/1291‐1322, r. 1316‐1322), enjoyed prestige and standing the other two lacked because of the great landed inheritance, the county of Burgundy, which she had brought to her marriage. Perhaps because of this, perhaps because her guilt seemed less clear than that of the others, she was treated differently and imprisoned near Paris, at Dourdan. After Philip the Fair died on 29 November 1314, Jeanne was released, around Christmastime, declared innocent after proceedings in the Parlement of Paris. News of the shocking and unprecedented scandal spread throughout the realm of France and beyond its borders. Marguerite and Blanche were generally considered guilty, even though there was wonderment at how the affair could have taken place.3
