Abstract:
Abstract: In this article we will carry out a quantitative, semantic and comparative analysis of the concepts ‘Spain’ and ‘Catalonia’ in its singular and plural forms of the Catalan, Spanish and Latin languages, contained in the institutional diaries of the Consell de Cent of Barcelona and the Deputation of the General of Catalonia. The chronology analysed will be the start of the 18th century, a period in which the death of the Spanish monarch Charles II of Austria (1700), the enthronement of the Bourbons (1701) and the consequent war of succession for the Monarchy of Spain (1702-1715) that was fought in the Principality of Catalonia between 1704 and 1714. The results of the study offer new perspectives on some aspects of the historiographical debate unleashed by the celebration of the Tricentenary of 1714, which revolves around the collective identity of Catalans at the beginning of the 18th century. Keywords: Conceptual history, Spain, Catalonia, Barcelona, War of Spanish Succession.

Abstract:
Introduction. The inventory of buildings and property are an accounting document. The largescale description of the lands and property of the spiritual patrimony of 1701–1705 carried out within the framework of the Church Reform of Peter I should be considered as a stage in the control of the state over the material welfare of the Church. Materials. Inventories of bishops houses and monasteries were found in the central and regional archives. A number of documents of the early 18th century were published. Analysis. Different census takers carried out descriptions of the patrimonies of the Vologda Bishops house in five regions of the country, which indicates applying the uyezd by uyezd principle of the description of the regions. The comparison of the texts shows that census books of the patrimonies of the Vologda Bishops house of 1701–1702 were primary in relation to the census of economy (statements) of the Vologda Bishops house in 1702–1703. An inventory of the Bishops Treasury was also made. Results. The comparison of three censuses (patrimony, economy, treasury) gave state documents on the basis of which it was possible to make a complete picture of the economy and property of the Vologda house of St. Sophia. In this regard, a complete secularization of Church possessions actually took place, however it was not fully formalized legally.

Abstract:
Amanuscript account by a Scots merchant, Robert Allen, of his life in New Spain from 1698 until 1707 has recently come to light. In 1708 Allen sent the three-page memorial to the British secretary of state for the south, Charles Spencer, Earl of Sunderland, outlining his experiences over the preceding decade with the view towards securing employment under the crown in the Americas. Allen began with the year 1698 when he left the Scottish colony at Darien to work for Jamaican merchants on trading voyages in the Spanish West Indies. In 1701 he returned to the coast of Darien in search of Nicaragua wood and, by chance, began six years of residence on the continent. Surviving one Indian massacre, he lived among friendly natives, joined the Jamaican expedition of 1702 which raided the Isthmus of Panama, escaped a second Indian and Spanish massacre, and was taken as a prisoner to Quito where in 1704 he secured appointment as secretary to the fiscal of the audiencia and surveyor general of the province of Quito. When this official was promoted to the Council of the Indies in 1707, Allen accompanied him on the journey to Spain. However, their ship was one of those attacked and sunk by Admiral Wager, who in turn persuaded Allen to return to England and approach the government.

Abstract:
In the newly revealed complete Dictionnaire des termes published in 1701 by Sébastien de Brossard there is a list of more than 60 “Auteurs qui ont écrit en François”, whose works he has personally “seen, read, and examined”. De Saint Lambert is named among them. This definitely proves that one of de Saint Lambert’s works had been published before 1701. This recently unveiled evidence and the diachronic examination of practically all possibly available historic and contemporary sources, showed that in some points the current view should be changed. In harpsichord teaching and performance, in thorough-bass accompaniment the treatises of de Saint Lambert, F. Couperin and J.-P. Rameau are the basic instruction editions. Only de Saint Lambert and his two treatises bring forth many bio-bibliographic questions, i.e. Les principes du clavecin (the dates of publication have been proved to be 1697 and 1702), Nouveau traité de l’accompagnement is Paris 1707; and the publication of the Traité de l’accompagnement (1680) is under question whether it was a “myth” or it indeed existed according to the historical evidence.

Abstract:
The article analyses two conventions of the Grand Duchy of Lithuania republicans, aristocracy and nobility that won the power in Lithuania at the end of 1700, which took place in Vilnius in 1701. First of the conventions in Vilnius took place 0n May 2–14, 1701. The second convention in Vilnius started meetings in same year, from July 23 through to August 12, 1701. The article discusses documents that were approved in these conventions, location of the gatherings and their significance in the sequence of republican conventions in 1698–1703. The analysis is focused on the influence of the conventions in establishing a new form of republican confederate governance in the Grand Duchy of Lithuania, also assessing the international and military context of the conventions. The conventions of the republicans of the Grand Duchy of Lithuania, having gathered for two meetings in Vilnius in 1701, were the most important political events in the life of Lithuanian state. In the course of these conventions the supreme Lithuanian state power of the time, the nature of which was quite special – close to confederate, decided on the most important issues, facing the new authority, established after the victory by the republicans against the Sapiehas, the aristocrats of the Grand Duchy of Lithuania who by the end of the seventeenth century had reached the status of hegemons. These conventions at the capital of Lithuania were extraordinary events that attracted key politicians of the time and some active nobility, which would not participate in great numbers but still were more actively than at other forums, such as inaugurations and sessions of the Lithuanian Tribunal. Republican conventions were initiated in 1698 and ended in 1703. Both 1701 conventions held their meetings in Vilnius alongside the Vilnius sessions of the Lithuanian Tribunal. First of the conventions took place at the eve of the Sejm of the Republic, and the second one soon after the Sejm, thus problems discussed in the conventions were closely related to the agenda of the Sejm of the Polish and Lithuanian state. Keywords: Grand Duchy of Lithuania, eighteenth century, Great Northern War, Lithuanian Civil War, the Sapiehas, August II, confederation, republicans, Vilnius.

Abstract:

Abstract:
Johann Jacob QuandT’S ACADEMIC ACCOUNT: THE LITHUANIAN LANGUAGE SEMINAR AT THE KÖNIGSBERG UNIVERSITY IN 1724 S u m m a r y The article introduces a document found in the Secret State Archives Prussian Cultural Heritage Foundation (Germ. Geheimes Staatsarchiv Preußischer Kulturbesitz; GStA PK: I. HAGR, Rep. 7 Preußen, Nr. 187 [1716–1729]) in Berlin that sheds new light on the seminar of the Lithuanian language – the first centre for teaching Lithuanian – that was founded at the Faculty of Theology of the Königsberg University in late 1720s. It is an academic account by Johann Jacob Quandt (1686–1772), the chief preacher of the court and the then dean of the Faculty of Theology of the Königsberg University and the fourth professor of theology in ordinary, who ran the seminar of the Lithuanian language between 1723 and 1727. This account provides insights into the early activities of the seminar that have not been documented in much detail so far. Neither the account nor any of its three appendices – lists of students attending Quandt’s courses – are dated. Based on other documents in the same archive file and the Christian holidays to which the account refers, Quandt’s account has been dated between 28 December 1724 and 11 January 1725, and the data that it contains cover the first half of the 1724–1725 winter semester: October–December of 1724. Quandt’s account shows that during the winter semester of 1724–1725, the seminar of theLithuanian language at the Königsberg University was attended by thirty theological students. Theology and language was taught twice daily between 10 and 11 AM and between 3 and 4 PM . The seminar under Quandt’s management continued to apply the so-called collegium privatissimum, the teaching method of its first supervisor, Heinrich Lysius (1670–1731). The names of the seminar attendees from that period are documented in the second appendix to Quandt’s account titled ‘Beyl. B. Auditores Seminarii Lithvanici’: these were Peter Gottlieb Mielcke (1695–1753), who was in his second year as a teacher, Gottfried Boeckel (?–after 1724), Samuel Boeckel (?–after 1724), Alexander Deutschmann (?–after 1724), Michael Sigismund Engel (1700–1758), Carl Julius Fleischmann (1704–1778), Christophor Daniel Franck?–after 1724), Georg Friedrich Gehrke (?–after 1724), Heinrich Grabau (Grabovius, ?–after 1724), Friedrich Wilhelm Haack (1707–1754), Georg Ernst Klemm (1701–1774), Johann Friedrich Leo (1696–1759), Christophorus (Georg) Liebe (1705–1764), Joachim Friedrich Mey (?–after 1724), Johann Friedrich Mülner (?–after 1724), Jacob Friedrich Naugardt (1694–1751), Friedrich Gottlieb Perbandt (?–after 1724), Adam Heinrich Pilgrim (1702–1757), Heinrich Preuss (?–after 1728), Christoph Rabe (?–after 1724), Heinrich Ernst Rabe (1707/1708–1744), Gottlieb Richter (1707–1775), Johann Richter (1705–1754), Friedrich Rosenberg (?–1727), Adam Friedrich Schimmelpfennig (1699–1763), Ernst Gottfried Schimmelpfennig (1704–1768), Martin Schimmelpfennig (1706–1778), Gottfried Schumacher (1704–1786), Friedrich Sigismund Schuster (1703–after 1732), Johann Trentovius (Trentowski, 1700–1765). Seven of them attended the seminar back in the winter semester of 1723–1724 and were among the first attendees of the seminar of the Lithuanian language under Quandt after it had been reinstated in 1723. Peter Gottlieb Mielcke was the first teacher at the reinstated seminar. During the winter semester of 1724–1725, the age of the theological students attending the seminar of the Lithuanian language at the Königsberg University was between 17 and 30. Most of them were from Prussian Lithuania. After finishing their studies, at least 19 of the attendees were ordained priests and served in Lithuanian parishes. Out of the thirty students who signed the second appendix to Quandt’s account, at least one-half have not been known as attendees of the seminar of the Lithuanian language yet. Even though the Pietist Georg Friedrich Rogall was very critical towards the seminar of the Lithuanian language under the orthodox Lutheran Quandt in his 1725 letter to August Hermann Francke (1663–1727), professor of theology at the Halle University, it is beyond any dispute that the seminar had brought up a new generation of authors of Lithuanian writings. Six of the theological students who attended the seminar in the winter semester of 1724–1725 had become involved in Lithuanistic activity, albeit from the camps of two protestant movements – the orthodox Lutherans and the Pietists. Three of them – Peter Gottlieb Mielcke, Adam Heinrich Pilgrim, and Adam Friedrich Schimmelpfennig – were actively involved in Johann Jacob Quandt’s project that aimed to renew and enhance the repertoire of religious Lithuanian literature. Three others – Johann Richter, Friedrich Wilhelm Haack (by the way, he became involved in Lithuanistic activity with his proof-reading of the 1727 New Testament published by Quandt in Lithuanian), and Martin Schimmelpfennig – later went to Halle, the centre of Pietism, where they became teachers at the seminar of the Lithuanian language that was founded there in 1727 and drafted Lithuanian books. Quandt’s pupils made a significant contribution to the breakthrough in Lithuanian writings between the 1730s and 1760s.

Abstract:
The purpose of the article is to carry out a comparative analysis of the news contained in the registration books of «all sorts of things» of the Military Chancery of 1701-1708 and legislative acts published in the Complete Collection of Laws of the Russian Empire (CCL). This study aims to investigate the informativeness of the Registration books. The fact that these books contain many unique unknown documents is stressed. The article outlines some principles of the selection and publishing of legislations which were used by drafters of the Complete Collection of Laws. The author draws conclusions that drafters of the Complete Collection of Laws were completely unacquainted neither with the data from the original Registration books, nor with the extracts of these books which were made for the famous Russian-German historian G.-F. Müller. The study is carried out with the financial support of the Russian Foundation for Basic Research within the framework of the scientific project № 19-09-00464 «Registration books of the Military Chancery of 1701–1709: research and publication».

Palabras clave: fortificación, ingeniería, Guerra de SucesiónTopónimos: GaliciaPeriodo: siglo XVIII ABSTRACTPhilip V’s ascent to the throne of Spain was accompanied by the arrival of individuals of French origin who occupied important positions of power within the new dynasty. They were joined by numerous individuals, following orders, who would carry out crucial fieldwork, essential in order to address the main problem facing the monarch during his first years, which was none other than guaranteeing the defence of the territory against the possibility of an armed conflict. Abstract:
In the late 17th and early 18th century, Ireland experienced a constitutional struggle in parliament, as well as the gradual development of a party system along the English partisan lines. Reflection of those events in the public sphere (primarily in the works of Molyneux and Swift) remains a popular research topic for Irish historians. This article attempts to look at the development of the Irish political system by examining poetic works in support of the chief governors of Ireland: lord lieutenants and lord justices of 1701–1714. Irish poems dedicated to governors were usually similar to English odes, which in turn were influenced by Abraham Cowley’s Pindarics. Irish odes to lord lieutenants of 1701–1711 had significant genre similarities, and most of them were also similar in general means of representing the chief governor. It was of utmost importance for the authors to show the brilliant ancestry of the ode’s hero; perhaps even more important for them was to show the similarity between the viceroy and the monarch, since the former was supposed to represent the latter. There were, however, significant differences between the odes, which were attributed to the shifting context of Irish politics. The odes of 1707 and 1711 are much more embedded in politics than the odes of 1701 and 1703: since at least 1707, the authors were more likely to include lord lieutenants in the context of Irish and British partisanship, while simultaneously emphasizing the loyalty of recipients to Queen Anne in her struggle against parties. The zenith of partisanship in Ireland coincides with the appearance of short poems with some features of an ode in 1710, which closely associate the figure of the lord lieutenant or lord justice with the Whigs or Tories.

Abstract:
The article presents historical research on the Church reform of Peter I, considering the Great Northern War (1700–1721) as one of the crucial transformational factors in church-state relations in Russia. The article introduces descriptive books of patriarchal estates in 17 counties of Russia. They were created during the audit of Church and monastic property in line with the decree of January 31, 1701. It considers features of the form and content of these documents and previously known descriptions of bishops' houses and monasteries, which were carried out according to the mandated memorials of the Monastic Order in 1701-1705. The article claims that the secularization of the patriarch's landholdings, in contrast to other church holdings, was complete and final already at the beginning of the 18th century. In this regard, there is an obvious need to reject historical parallels between the events of the early 18th century and the "monastic secularization" of 1764.Comparing the Alexei Kurbatov’s proposals to Peter I in a letter dated October 25, 1700, with the content of the personal decrees of January 24 and 31, 1701 on the restoration of the Monastic Order allows us to hypothesize that the defeat at Narva and preparation for the meeting with the Elector Augustus II that would result in the signing of the Biržai pact, significantly adjusted the plans of the Church reform. The novelty of the January decrees that presumably started the reform (in the then-common European sense of the concept) was the inclusion of the patriarch's estates in the list of the Church's possessions subject to the control of the state in the future.I conclude that the secularization of church-monastic possessions and property that followed at the beginning of the Great Northern War not only contradicted the centuries-old historical practice but was at that time organizationally overwhelming for the state, which was able to solve the problem of managing the escheat patriarchal lands but did not yet have the appropriate staffing and logistics capacities for control over all possessions of the Church.

Abstract:
The article is focused on the history of Koknese Fortress Front fortifications which were built from September 1700 to May 1701 in order to enhance the defence power of Koknese Fortress. The total length of the defence line exceeded 4 km. The line contained 25 redoubts. After the loss of Spilve battle close to Riga, the Saxon troops retreated from Koknese, on 25 July 1701 Koknese Fortress was blown up and after that was not used for military purposes anymore. The outer defence line of Koknese Fortress never faced military attacks and after 1701 was abandoned, partially levelled by agriculture work, destroyed by activities of the First and the Second World Wars, building of houses, roads and motorway, establishing a cemetery on one of the earthworks, flooding by Pļaviņas hydroelectric power station, etc. At present the front defence system of Koknese Fortress has partially survived, but until the last years the particular system of defence line had not been clearly identified. Now it has been done comparing an image of Koknese from 1701 with the results of aerial and traditional reconnaissance. Koknese front fortification line is a unique monument under circumstances of Latvia.

Abstract:
The purpose of this article is to identify the peculiarities of Western European military affairs as illustrated by a particular campaign of the War of Spanish Succession (1701-1714). The article is of interest in light of comparing military affairs of Western Europe and Russia at the beginning of the 18th century, military art during the War of Spanish Succession (1701-1714) and the Great Northern War (1700-1721). Scientific novelty: the study presents an episode of Western European military history not yet reflected in domestic historiography, combat operations between Franco-Bavarian troops and the allied army under the command of John Marlborough on the Danube theater of war in the summer of 1704. The study is carried out with wide use of memoir, epistolary and documentary sources. During the 1704 campaign on the Danube and in Bavaria, such typical features of the military art of the 18th century were manifested as excessive dependence on supplies and logistics, positional expectant nature of hostilities, their indecision and false methodism. As a result of the study, the author came to the conclusion that the Franco-Bavarian coalition was more inclined towards the canons of the military strategy that had developed by the beginning of the 18th century. It operated within the framework of positional maneuver tactics, enthusiasm for fighting on communications, demonstrating wait-and-see and passivity. French marshals were distinguished by their adherence to the template and therefore were predictable and expected. Unlike the enemy, John Marlborough and Eugene of Savoy managed to break out of the generally accepted tactical canons. During the 1704 campaign, they showed swiftness and offensive character, imposed a decisive aggressive struggle on the enemy, the culmination of which was a resounding victory at Blenheim on August 13, 1704.

Jean-Alphonse started his theological career as the pastor of the Italian congregation in Geneva in 1693; he was then named professor of church history at the Academy in 1697 and then rector in 1701 and finally professor of theology in 1705. Although his father was one of the principal architects of the Helvetic Formula Consensus (1675), Jean-Alphonse led the movement toward eliminating such credal religion through the abrogation of the Formula in 1706.

Abstract:
The article discusses the problem of establishing a permanent diplomatic mission of Peter I in the Sublime Porte. At the initiative of the envoy E. I. Ukraintsev, an article, enabling the tsar to send an ambassador for a permanent residence in Istanbul, was included in the Treaty of Constantinople (1700). After the envoy’s departure, only the Non-diplomatic ministers of the Ambassadorial prikaz (chancellery), namely translator S. F. Lavretsky, podyachy (clerk) Gr. Yudin (died in December 1700) and interpreter D. Petrov, stayed in the Ottoman capital. Translator S. Lavretsky became the head of the Russian diplomatic mission. In 1701, a messenger M. Larionov arrived in Istanbul with the tsar’s charter. According to it translator and podyachy had to stay in Istanbul until the arrival of the plenipotentiary ambassador with ratification. On the basis of the archival documents stored in the RGADA (Moscow), the author analyzes the activities of diplomatic missions in 1700-1701. The main task of the translator and the ambassador was to inform the Russian government about the political situation in the Ottoman Empire. Peter I sent the main forces of his state to the war with the Swedes, so he needed peace on the southern borders. The translator and podyachy maintained contact with ministers of the Sublime Porte, the Jerusalem Patriarch, agents, etc. They also monitored the preparation of the Sublime Porte for the arrival of the Russian plenipotentiary ambassador. Information collected from various sources regarding the situation in Istanbul, Crimea and other parts of the Ottoman Empire, as well as Western Europe, they sent to the Ambassadorial prikaz. The activities of Russian diplomats in 1700–1701 largely corresponded to the functions of the ambassadorial secretary (charge d’affaires).

The New England Company had the longest history and is the oldest Protestant missionary organization. The Society for the Propagation of the Gospel in Foreign Parts sent no fewer than three hundred missionaries to America between 1701 and 1783. While the NEC and the SPG have received scholarly attention, the Society in Scotland for Propagating Christian Knowledge has been virtually ignored.

Abstract:
AbstractFrancis Hauksbee (1660–1713) is well known for his double-barrelled air-pump. However, the origin of this pump, and Hauksbee's background, are often described as a mystery. This text seeks to dispel the riddle. It is argued that Hauksbee's competence as an exceptional maker of air-pumps was developed between 1699 and 1703 as a result of his experiences with the construction, manufacturing and sale of cupping-glasses. His cupping utensils embodied a new design, where syringes were used to evacuate the glasses, instead of the traditional way by fire or mouth suction. These syringes, which in fact were small air-pumps, were perfected between 1699 and 1701. A larger syringe, introduced in 1701, served as a transition from the cupping-syringe to his first air-pump for use in natural philosophy. This syringe was described as a ‘combined engine’, which could serve as an air-pump, a condensing engine and a syringe for injecting air, wax or mercury into pathological specimens. Hauksbee's first air-pump was a single-barrelled model introduced in 1702, based on the combined engine. Its various features, such as easy and convenient leak-tightening, exact pressure measurements by an in-built barometer and an air-inlet function for readmission of air into the receiver, are discussed. Finally, it is shown that these activities gave Hauksbee the reputation of being an outstanding instrument-maker, years before the double-barrelled air-pump was in sight.

Abstract:
Abstract RTA 1701 is an orally-bioavailable, selective RORγt inhibitor. RORγt orchestrates the differentiation of Th17 cells and contributes to autoimmune disease pathogenesis. Inhibition of RORγt suppresses Th17 cells and the associated secretion of the effector pro-inflammatory cytokine IL-17A. As such, RTA 1701 is highly efficacious in rodent models of rheumatoid arthritis and multiple sclerosis. This study was conducted to evaluate the pharmacokinetics of RTA 1701 and its effects on ex vivo stimulation of IL-17A production in whole blood after oral administration to naïve healthy cynomolgus monkeys. Monkeys received a single administration of RTA 1701 (0.3, 3, 30, or 300 mg/kg). After a one-week washout, monkeys received the same doses of RTA 1701 once daily for 14 consecutive days. Blood was collected after a single dose and after 14 days of dosing for analysis of plasma RTA 1701 concentrations through 72 hours post-dose and for evaluation of RTA 1701 effects on ex vivo stimulation of IL-17A at 24, 30, and 72 hours post-dose. RTA 1701 exhibited oral bioavailability in monkeys with dose-dependent increases in exposure over the wide range of doses evaluated. Further, oral administration produced systemic exposure to RTA 1701 at sufficient levels to produce significant and dose-dependent suppression of ex vivo stimulation of IL-17A secretion in whole blood at 24 and 30 hours after single and repeat dosing, with IL-17A levels returning to baseline within 72 hours after a single dose. Collectively, these data demonstrate that RTA 1701 is orally bioavailable in non-human primates and significantly suppresses the IL-17A response. These data also support the future clinical development of RTA 1701 for the potential treatment of autoimmune diseases.

Abstract:

Abstract:
7525 Background: TG-1701 is a selective, covalent BTK inhibitor administered once daily (QD). Both the “U2” combination (anti-CD20 mAb ublituximab + the PI3Kδ-CK1ε inhibitor umbralisib) and BTK inhibition are highly active in treatment-naïve (TN) and relapsed/refractory (R/R) CLL, each having previously demonstrated superiority over standard chemoimmunotherapy. Here we report the results of the dose escalation of TG-1701 monotherapy and TG-1701+U2. Methods: Pts with R/R CLL and lymphoma were enrolled in a Ph 1 study initially evaluating dose escalation (DE) of oral TG-1701 QD continuously administered in 28-day cycles (100, 200, 300, and 400 mg). After characterizing the safety profile of TG-1701 monotherapy, we implemented a parallel DE arm of TG-1701+U2. Select dose levels of TG-1701 monotherapy were expanded in CLL, MCL and Waldenström's (WM). All pts were treated until disease progression. The primary objectives are to characterize the safety profile and define the recommended Ph 2 doses for the drugs alone and in combination. Results: As of 03 February 2021, 123 pts were treated with TG-1701: 25 in the monotherapy DE arm, 61 in the 200 mg disease-specific cohorts (20 CLL [5 TN], 21 MCL [4 TN], 20 WM [8 TN]), 20 in the 300 mg CLL cohort (4 TN), and 17 in the 1701+U2 DE arm. The median # of prior therapies was 1 (range, 1 - 10). All pts were BTKi-naïve. All 123 pts were evaluable for safety. TG-1701 was well tolerated and the maximum tolerated dose (MTD) for monotherapy was not reached at 400 mg (demonstrating near 100% saturation of the BTK at all dose levels studied). Treatment emergent adverse events (TEAE) of clinical interest included atrial fibrillation (AF 4.0% of pts, G ≥3 in 1 case), G ≥3 hypertension (2.4%), and bleeding events (18.7%, all G1-2). No cases of ventricular tachyarrhythmia were reported. TEAEs leading to TG-1701 dose reduction occurred in 6.5% of pts. TEAEs leading to treatment discontinuation occurred in 1.6% of pts (AF, COVID-19). At the data cut-off, 119 pts were evaluable for response, including 40 in DE (Table). The median duration of response has not been reached among responders overall. The median follow-up (mFU range) was 15.9 mos (1.3 - 28.6+) in DE and 8.5 mos (1.4 -15.6+) in disease-specific cohorts. Conclusions: TG-1701 exhibits an encouraging safety and efficacy profile. The combination of 1701+U2 has been well tolerated and dose escalation continues. The combination shows enhanced depth of response over TG-1701 monotherapy. Recruitment to this study continues. Response per investigator review by treatment group. Abstract:
Abstract Background LSVT-1701 is an anti-staphylococcal phage lysin being developed for treatment of MRSA infections in combination with SoC antibiotics. The safety and PK of single ascending doses of LSVT-1701 0.1 to 10 mg/kg in healthy adult volunteers were previously described (Jun, et.al, AAC 2017;61:e02629-16). We further evaluated the safety and PK of multiple ascending doses of LSVT-1701 in healthy adult subjects. Methods Study ITB-101-1b was a Phase 1, randomized, double-blind, placebo-controlled, multiple ascending dose study. 8 subjects were randomized 3:1 to active:placebo in each cohort. LSVT-1701 was administered as a 6 mg/kg single dose and twice daily (BID) doses of 1.5, 3.0, and 4.5 mg/kg for 4 days (24h between Doses 1-2, 12h between Doses 2-6). Study drugs were administered as a 1-hour IV infusion. Serial serum samples were collected over 24 hours following the first and last doses for measurement of LSVT-1701 concentrations by a validated ELISA method. PK analysis of LSVT-1701 concentration-time data was done using noncompartmental methods. Safety was assessed by AEs, clinical labs, vital signs, and ECG. Results 30/32 (94%) subjects completed the study. No subjects withdrew due to AEs, and there were no severe AEs, no serious AEs, and no deaths. AEs were of mild (97%) to moderate (3%) intensity and were reported by all subjects in the LSVT-1701 6 mg/kg single dose group and 1-3 (17-50%) of subjects receiving 1.5 to 4.5 mg/kg BID or placebo. The most common AEs of headache, chills, rigors, and fever generally lasted for ≤2 days with or without acetaminophen treatment, and no clinically significant changes in blood pressure, heart rate, ECG, or clinical labs (other than transient increases in CRP) were observed. Infusion site reactions (erythema, pain, induration, warmth) were observed with BID administration of LSVT-1701, but not with the single 6 mg/kg dose or placebo. LSVT-1701 exposure increased greater than in proportion to dose and t1/2 was concentration-dependent, increasing with higher doses. No accumulation in LSVT-1701 exposure was observed. Summary of LSVT-1701 PK Parameters Summary of LSVT-1701 PK Parameters Conclusion The safety and PK profile of LSVT-1701 is favorable for evaluation in patients with S. aureus infections, including bacteremia and infective endocarditis, for which new treatments are needed. Abstract:

Abstract:
Abstract Introduction: We previously showed that derivatives of the Chinese traditional medicine indirubin promote survival in murine models of the brain tumor glioblastoma. However, poor drug solubility hampered further development of this approach. Here we introduce PPRX-1701, a 6’-bromoindirubin acetoxime (BiA) containing drug/polymer nanoparticle formulation which can be injected intravenously at relatively high concentrations. Here we examine the efficacy and mechanism of PPRX-1701 in murine glioblastoma models. Methods: In vitro glioblastoma cell proliferation and migration assays were performed using standard approaches. Effects of PPRX-1701 on interferon-γ (IFN γ) signaling were determined by Western blotting. Delivery to murine GBM was assessed using a luminescent reporter assay, and survival studies were performed in immunocompetent murine glioblastoma models. Results: PPRX-1701 effects on glioblastoma cell proliferation and migration were indistinguishable from BiA alone. Our data show that intravenous administration of PPRX-1701 is well-tolerated and can reach intracranial murine glioblastoma as assessed by luminescent reporter assays. PPRX-1701 administration leads to improved survival in the GL261 glioblastoma mouse model (median survival 30 days (control), 47 days (treated), p < 0.0001). Treatment with PPRX-1701 was associated with alterations in the tumor immune microenvironment, with reduced Tregs and pro-tumor macrophages, and increased CD8+ T cells. Further mechanistic studies have shown that PPRX-1701/BiA blocks the expression of multiple immunosuppressive molecules in GBM downstream of interferon-γ (IFNγ) including PD-L1 and indoleamine 2,3-dioxygenase 1 (IDO1) - a key enzyme in the tryptophan-kynurenine-aryl hydrocarbon receptor (Trp-Kyn-AhR) immunosuppressive pathway. BiA promoted more effective T-cell mediated tumor cell killing using patient derived glioblastoma ex vivo co-culture models. Conclusion: This data supports further development of PPRX-1701 as a candidate immunotherapeutic agent for glioblastoma treatment. Ongoing pre-clinical studies are investigating combination with other relevant therapies. Abstract:
The history of Russian female monasticism is one of the little-studied topics. One of the main sources that allows us to characterize the monasteries of the late XVII – early XVIII centuries are the census books that were compiled as part of the initial stage of the church reform of Peter I. The census books of 1701-1703 are a multifaceted source. A large place in them is occupied by information about monastic temples and their interior decoration, about the treasury, where monastic documents were kept, libraries, economic activities, land holdings, etc. The study showed that the census books for the study of nunneries are a very informative source. On their basis, it is possible to establish the number of women's monasteries. They contain information about the management of monasteries, about the social status of nuns, their daily life, as well as about the income and financing of monasteries by the state. The materials of the description to one degree or another reflect the church transformations of Peter I carried out in the country in 1701-1703, which had a significant impact on the life and way of life of the nuns. This makes the census books a very promising source for studying the history of women's monasteries in Russia at the turn of the XVII–XVIII centuries.

Abstract:
The article studies the military campaign of 1703 in the Netherlands of the War of the Spanish Succession of 1701– 1714. The presented campaign, still undervalued by military historiography, is the most typical of the European military art of the early 18th century. The article shows the activities of the Anglo-Dutch and French command in the planning and organizing of military operations. Based on the correspondence of Louis XIV, Duke of Marlborough, French Marshals Villeroy and Bouffler, the article traces the course of the fighting in Flanders and Brabant in the summer and autumn of 1703. At the same time, the war is considered as a combination of careful maneuvering and rapid marches, false demonstrations and decisive strikes, methodical sieges of fortresses and unexpected breakthroughs of fortified lines. As a result, the little-known stage of the War of the Spanish Succession of 1701–1714 is studied in detail. On the example of the above-mentioned campaign, the article reveals such features of Western European military affairs as the limited operational goals and plans, positional nature of strategy, rejection of decisive forms of struggle, commitment to siege warfare and maneuver tactics, increasing role of engineering and fortification and rear services, communications and logistics.

Abstract:
Abstract Several HER2-targeting antibody-drug conjugates (ADC) have gained market approval for the treatment of HER2-expressing metastasis. However, these ADC drugs still face challenges of acquired resistance and/or severe adverse effects associate with their payload toxins. Eribulin, a market approval agent for the treatment of metastatic breast cancer and liposarcoma , is a new choice of ADC payload with distinct mechanism of action (MOA). Here we report preclinical studies of BB-1701, a novel HER2-tageting eribulin-containing ADC. BB-1701 exhibited potent and specific cytotoxicity activities to various types of cancer cells where HER2-exprssing levels vary in large ranges. BB-1701 induced tumor-growth suppression, even regression in in vivo models with a single dose of 1-5 mg/kg dosage. In comparison with other HER2-targeting ADCs with different toxin payloads (DM1, MMAE and Dxd), eribulin-containing ADC demonstrated higher potency to cancer cell lines with low HER2 expressing levels (with lower IC50). BB-1701 also showed effective tumor suppression in models insensitive to T-DM1 or T-Dxd. Mode of action studies revealed significant bystander cytotoxicity of BB-1701 to HER2-low cells when co-cultured with HER2-high cells. Additionally, BB-1701 treatment increased ATP releasing and Calreticulin expression, both were deemed immunogenic cell death (ICD) markers. Lastly, repeat dose pharmacokinetic and toxicology studies in nonhuman primates revealed favorable PK and safety profiles of BB-1701 in the clinical intend dosage route and schedule. In conclusion, these comprehensive preclinical data have strongly supported BB-1701 as a potent antitumor agent to cancers including HER2-low expressing and/or cancers insensitive to other HER2-targeting ADCs. A phase I clinical study of BB-1701 in those patients is ongoing. Abstract:
Abstract Background Anti-staphylococcal phage lysins, such as LSVT-1701, represent important candidate adjunctive agents against invasive MRSA infections because of both their microbicidal and anti-biofilm properties. We, thus, sought to examine the in vivo efficacy of LSVT-1701 combination with daptomycin, a standard-of-care anti-MRSA agent with proven efficacy against bacteremia and IE in humans. Methods We utilized the rabbit model of aortic valve infective endocarditis (using the prototype MRSA strain, MW2) to examine the combined efficacy of LSVT-1701 plus daptomycin. We examined microbiologic outcomes in distinct target tissues (cardiac vegetations, spleen and kidney) in this model, as well as the pharmacokinetic and pharmacodynamic drivers and target attainment values most predictive of treatment outcomes. LSVT-1701 was given at two dose-regimens (32.5 mg/kg and 50 mg/kg) with different dose-durations (single dose vs daily dose for 2 d vs daily dose for 4 d); daptomycin was administered in combination with daptomycin at a sub-lethal daily dose of 4 mg/kg for 4 d to maximize potential synergistic interaction outcomes. Results The Table below shows all LSVT-1701 regimens in combination with daptomycin significantly reduced MRSA burdens in all target tissue as compared to untreated controls. The reduction in MRSA counts was statistically significant in instances of both increasing LSVT-1701 dose level (i.e., single doses of 50 mg/kg vs 32.5 mg/kg iv), as well as increased numbers of lysin doses (i.e., four daily doses vs a single-dose or two daily-doses) in combination with daptomycin. Of note, both the LSVT-1701 50 mg/kg and 32.5 mg/kg daily dose-strategies given for four days in combination with daptomycin sterilized all target tissues (i.e., quantitative cultures ≤ the lower limit of detection of 1 log10 CFU/g. tissue). MRSA bio-burdens in blood, vegetations, kidneys and spleen in rabbit IE model Conclusion LSVT-1701 administered at 32.5 or 50 mg/kg in a 4 d daily-dose regimen in combination with daptomycin resulted in microbiologic sterilization of all target organs in this MRSA IE model. These data support further clinical development of LSVT-1701 for the treatment of MRSA endovascular infections including IE. Abstract:
Abstract Retinoic acid-related orphan receptor γt (RORγt) is a transcription factor that directs the differentiation of Th17 cells, a subset of CD4+ helper T cells involved in autoimmune disorders. Once differentiated, Th17 cells secrete interleukin-17A (IL-17A) and other key cytokines implicated in autoimmune and inflammatory disease. Antibodies that target Th17 cells are approved for several autoimmune disorders, validating the important role of Th17 cells in disease. RTA 1701 is a potent and selective RORγt inhibitor in preclinical development for autoimmune disorders. It inhibits the activity of RORγt, as assessed using ligand binding domain and full-length RORγt reporters. Moreover, RTA 1701 is selective for RORγt and does not inhibit the activity of two closely-related receptors, RORα and RORβ. By inhibiting RORγt, RTA 1701 blocks Th17 differentiation of naïve CD4+ T cells and IL-17A production by both Th17 polarized CD4+ T cells and activated peripheral blood mononuclear cells (PBMCs). The expression of other RORγt target genes and Th17 signature genes (eg, IL-17F, IL-21, CCL20) is also reduced following RTA 1701 treatment of Th17-polarized naïve CD4+ T cells, as assessed by qPCR. RTA 1701 is orally bioavailable with excellent pharmacokinetic properties in rodents and monkey. Furthermore, this compound is efficacious in animal models of autoimmune disease, including the collagen-induced arthritis (CIA) mouse model of rheumatoid arthritis and the experimental autoimmune encephalomyelitis (EAE) mouse model of multiple sclerosis. Together, these results suggest that RORγt inhibition by RTA 1701 may have therapeutic potential in Th17-associated autoimmune disorders.

Abstract Background LSVT-1701, formerly SAL200, is a novel, recombinantly-produced, bacteriophage-encoded lysin that specifically targets staphylococci via cell wall enzymatic hydrolysis. We reported the in vitro activity of LSVT-1701 against clinical isolates of S. aureus and coagulase-negative staphylococci (CoNS) collected worldwide. Methods LSVT-1701 and comparators were tested against 415 S. aureus (n=315) and CoNS (n=100) clinical isolates expressing various resistance phenotypes. The isolates were collected in 2002-2019 from medical centers located in the United States (50 medical centers; 174 isolates; 41.9% overall), Europe (37 medical centers; 140 isolates; 33.7% overall), Asia-Pacific region (15 medical centers; 55 isolates; 13.3% overall), and Latin America (12 medical centers; 46 isolates; 11.1% overall). These isolates originated mostly from the year 2019 (n=323).The isolates were susceptibility tested by the CLSI broth microdilution method. MIC interpretations were based on CLSI and EUCAST criteria where available. Results LSVT-1701 was highly active against S. aureus and CoNS isolates with MIC90 values of 2 mg/L for all S. aureus, methicillin-susceptible S. aureus (MSSA), methicillin-resistant S. aureus (MRSA), and CoNS (Table). The highest LSVT-1701 MIC values were 4 and 8 mg/L among S. aureus and CoNS, respectively. LSVT-1701 retained potent activity against S. aureus isolates showing resistance or decreased susceptibility to oxacillin, vancomycin, teicoplanin, telavancin, linezolid, daptomycin, ceftaroline, or lefamulin; MIC50 values ranged from 0.5 to 1 mg/L and MIC90 values ranged from 1 to 4 mg/L among S. aureus resistant subsets. Summary of LSVT-1701 activity against S. aureus, CoNS and resistant subsets Conclusion LSVT-1701 demonstrated potent in vitro activity against contemporary clinical isolates of S. aureus and CoNS collected from medical centers worldwide and against resistant S. aureus isolates with uncommon resistance phenotypes. Abstract:
The article analyzes the plans of Peter the Great related to the consolidation of Russia on the shores of the Baltic Sea. It focuses on the emergence of the idea to gain access to the Baltic Sea and the stages of its implementation. During the Northern War of 17001721, Russia's peace conditions changed. The article tries to explain the basis for the evolution of Peter the Greate's ideas of achieving the desired peace. It shows that after the "embarrassment at Narva" in November 1700 and the first success of B.P. Sheremetev at Erestfer in December 1701 in 1702 1709, the conduct of hostilities and the diplomatic efforts of Russia were directed exclusively at keeping the outlet to the sea. The creation of St. Petersburg, the transfer of the capital to it, the construction of a defensive system at the mouth of the Neva all this was aimed at securing the mouth of the Neva for Russia. Russia agreed to the conclusion of a peace, according to which it received only Ingria and Karelia – the old Russian provinces. After the victory under Poltava in 1709, a new struggle for peace began. The annexation of Livonia, Estland, the temporary occupation of Finland – all these conquests were motivated by the idea for which Russia started the war: keeping and ensuring the safe outlet to the sea. Russia constantly showed its readiness to conclude peace, but Sweden rejected all such attempts because it could not get along with the idea of parting with imperial ambitions.Russia began ten-year-long harsh coercion of Sweden to peace, which resulted in the devastation of a part of the Swedish territories proper by the Russian army and the forced consent of the Swedes to peace. The Nishtad Peace of 1721 ended the war and became the starting point for the extraordinary development of the imperial imagination of Peter the Great. Russia's entry into the world arena as an empire, an autocratic state dangerous to its neighbors and actively participating in the incessant division of the world.

Abstract:
Introduction Agents targeting BTK have demonstrated activity in a variety of B-cell malignancies, however not all patients respond to therapy, and amongst those that do respond, complete remissions are rare. BTK-based combination regimens have the potential to increase depth of response and permit time-limited therapy, however innovative approaches to accelerate the development of combination regimens are needed. TG-1701 is a novel, orally available and covalently-bound BTK inhibitor that exhibits superior selectivity for BTK compared with ibrutinib in in vitro whole kinome screening (Abstr 3973, EHA 2018). Herein we report the first results of a unique Phase 1 parallel dose-escalation study of TG-1701 monotherapy and TG-1701 in combination with umbralisib, a novel PI3K-δ and casein kinase-1ε inhibitor, and ublituximab, a glycoengineered anti-CD20 mAb (1701 + U2). Methods The primary objectives of the study are to characterize the safety profile and to determine the recommended Phase 2 dose of TG-1701 as a single agent and in combination with U2. Other objectives include assessment of pharmacokinetics (PK), preliminary antitumor activity, and pharmacodynamics (PD [BTK occupancy]). Eligible patients must have B-cell malignancy relapsed or refractory (R/R) to one or more prior standard therapy. Treatment consists of escalating doses of oral TG-1701 once daily (QD), continuously administered in 28-day (D) cycles (C). Patients in the 1701 + U2 combination arm receive escalating TG-1701 oral QD + umbralisib 800 mg oral QD + ublituximab 900 mg IV on D1, 8, 15 of C1, and D1 of C2-6. All patients are treated until disease progression, unacceptable toxicity, or investigator/patient decision to withdraw study consent. Results As of July 15, 2019, 19 patients (WM = 6, CLL = 4, FL = 4, MZL = 2, DLBCL = 2, MCL = 1) have been treated with TG-1701: 3 patients at 100 mg QD, 9 patients at 200 mg QD (expansion before opening combination), 3 patients at 300 mg QD single agent arm, and 4 patients at 100 mg QD combination arm. Patients had a median of 2 prior systemic therapies (range, 1 - 5), and all of them received previous anti-CD20 therapy. Four patients were refractory to their last prior therapy, 8 had extranodal disease, and 8 had bulky disease. To date, patients have received a median of 4 (range, 2 - 10) cycles of TG-1701. No dose-limiting toxicity (DLT) have been observed to date. The most common treatment-related adverse events (TRAE) are: neutropenia (21%), diarrhea (16%), nausea (16%), bruising (16%), infection (16%), ALT/AST elevation (11%), rash (11%), abdominal pain (11%), and fatigue (11%). Grade 3 TRAE are: asymptomatic and isolated lipase elevation (N=1), nausea (N=1), rash (N=1), and acute respiratory tract infection (N=1). There have been no grade 4 TRAE nor treatment discontinuations due to adverse events. Linear kinetics are apparent, evidenced by approximately dose proportional increase in AUC on C1D1 and C1D8. High systemic clearance (CL) has been observed with a mean CL/F of 55.4 L/hr and half-life of 2.24 hours. Tmax is observed between 1 to 4 hours post dose. PK-PD (BTK occupancy) relationship at 200 mg on C1D1 is presented in Figure 1. Decrease from baseline in tumor burden is presented in Figure 2. Two patients at the lowest dose (100 mg QD) have achieved a partial response (PR): MCL = ↓78% and WM = ↓88%. All 3 evaluable patients treated with 100 mg 1701 + U2 have achieved a response at the first response assessment: 1 CR (FL) and 2 PR (FL ↓88%, and MZL ↓65%). All patients remain on study treatment, except for both patients with DLBCL that discontinued due to disease progression. All patients with CLL have shown lymphocytosis during the first 2 cycles of therapy. Near complete BTK occupancy has been achieved in all patients at all dose levels (Figure 1). Conclusions TG-1701, a once daily BTK inhibitor has an encouraging preliminary safety profile, with clinical and pharmacodynamic activity at all dose levels evaluated. This study (NCT03671590) continues enrollment in TG-1701 single-agent and 1701 + U2 combination arms. Abstract:
TPS1122 Background: Treatment options for patients (pts) with unresectable or metastatic BC have improved with the development of novel targeted therapies including trastuzumab deruxtecan (T-DXd), an ADC directed against HER2 with a cytotoxic topoisomerase I inhibitor as a payload. Currently, there are limited treatment options for pts with HER2+ or HER2-low metastatic BC that has progressed on T-DXd. BB-1701 is an ADC consisting of a monoclonal antibody targeting HER2 with eribulin (microtubule inhibitor) as a payload that is capable of exerting a bystander effect. A first-in-human dose-finding Study 101 of BB-1701 is ongoing. Here we describe Study 205, an open label, multicenter, phase 2 study of BB-1701 in pts with HER2+ or HER2-low unresectable or metastatic BC who have disease progression after treatment with T-DXd. Methods: Study 205 (NCT06188559) includes dose-optimization and dose-expansion parts for BB-1701. Eligible pts must have histologically confirmed HER2+ or HER2-low, unresectable or metastatic BC, with measurable disease per RECIST v1.1. Prior treatment must include 1–3 prior chemotherapy-based regimens for unresectable or metastatic BC, including T-DXd. The primary objectives of the dose-optimization part include determining the recommended dose (RD) and evaluating safety and tolerability of BB-1701. The RD will be assessed by randomly assigning ~50 pts into 1 of 3 dosing cohorts (1.6 mg/kg on day 1 Q3W, 0.8 mg/kg on days 1 and 8 Q3W, and 1.2 mg/kg on day 1 Q3W; IV) in a 2:2:1 ratio. Randomization will be stratified by HER2 status (HER2+ vs. HER2-low) documented prior to T-DXd treatment. The dose-expansion part will enroll ~85 pts at RD; the primary objective includes evaluating the efficacy of BB-1701 at the RD. In both study parts, secondary objectives include assessment of additional efficacy measures (i.e., duration of response [DOR], progression free survival [PFS], overall survival [OS], disease control rate [DCR], clinical benefit rate [CBR], and time to response [TTR]), and pharmacokinetics. Efficacy endpoints will be assessed per RECIST v1.1 by the investigator during the dose-optimization part or by Blinded Independent Central Review during the dose-expansion part. Safety assessments will consist of recording, monitoring, and grading of adverse events (AEs) based on CTCAE v5.0, AEs of special interest, and serious AEs. In both study parts, exploratory objectives will include evaluation of biomarkers of response to BB-1701 and BB-1701 immunogenicity; other efficacy measures (i.e., PFS on the next line of therapy) and population pharmacokinetics will be analyzed in the dose-expansion part only. Pt enrollment is currently open. Clinical trial information: NCT06188559 .
