Abstract:
User development of applications provides end users with an alternative to the traditional process of systems development by allowing them to solve job related problems by developing their own software applications. User developed applications (UDAs) support decision making and organisational processes in the majority of organisations, and the ability to develop small applications forms part of the job requirements for many positions. Despite its pervasiveness, there are many risks associated with user development of applications. These risks result primarily from decreases in application quality that arise when end users have had little training and do not follow system development methodologies. The primary aim of the research described in this thesis is to gain a better understanding of UDA success. In particular, the thesis considers the role of system quality in UDA success and the ability of end user developers to judge whether the applications they develop will have a positive impact on their performance of tasks. The research also investigates factors that might impact upon this ability. The research objectives were addressed through two empirical studies. Two possible models of UDA success provided the starting point for Study 1. The first model is DeLone and McLean's (1992) model of IS success, and the second model is a version of this model that was modified to address concerns about the DeLone and McLean model and to reflect current research about UDA success. The models were tested using data from a field study involving business people participating in a business policy simulation, where they developed spreadsheet applications to assist in decision making. Structural equation modelling was used to test the models. Neither of the models was well supported by the data. However, the analysis provided strong support for relationships between perceived system quality and user satisfaction, information quality and user satisfaction, user satisfaction and intended use, and user satisfaction and individual impact. It is notable that the model paths that were supported in Study 1 were primarily those that reflect user perceptions rather than objective measures. This study highlighted that user perceptions of information systems success play a significant role in the UDA domain. The results did, however, suggest that there might be a direct relationship between system quality and individual impact. Study 2 was a laboratory experiment and the participants were end users from a range of organisations. A revised research model was developed based on the findings of Study 1, and structural equation modelling was again used to test the model. The model paths that were supported suggest that for small to moderate applications, increases in spreadsheet development knowledge lead to increases in system quality and consequently the development of better quality spreadsheets. They also suggest that for these kinds of applications, end users have realistic perceptions of system quality and hence that user satisfaction may be an appropriate measure of UDA success. The results of Study 2 also provided insight into the role of user involvement in end user development, clarifying the process by which benefits are obtained. The study also provided insight into the importance of spreadsheet development knowledge for successful use (as well as development) of a spreadsheet application. The results described in this thesis have practical implications for the management of user development of applications. They highlight the need either to increase end user levels of development knowledge via training so that end users can cope with applications of greater complexity, or to provide other forms of support for development. The role of organisational standards and guidelines is also be considered in the thesis and it is suggested that there is a particular need for guidelines on what kinds of applications are suitable for end user development.

This thesis is an ethnographic study of a group of Chinese postgraduate students in a British university as they become adjusted to the culture of teaching and learning in the new learning environment during their first year of overseas study. It focuses on these Chinese students’ initial perceptions of British teaching and learning practices compared with their inherited culture of learning and how they make adjustments, emotionally, cognitively, and behaviourally, in order to make their learning successful, with the result of changes and developments in their conceptions and beliefs about knowing and learning. The present study seeks to draw together understanding from the fields of intercultural adaptation theories, tertiary students’ conceptions of learning research, and the interface of culture and learning, i.e. cultures of learning, to explore the impact of studying abroad on students’ intellectual development and personal growth so as to inform international and intercultural education.

Abstract:
The international relations literature on internationalism in foreign policy has not taken account of the internationalist methods and motives of countries of the developing world. This thesis aims to correct this absence through an analysis of Southern internationalism, as evidenced by the foreign policy approaches of South Africa and Brazil in the first decade of the 21st century. By utilising a neoclassical realist approach to the study of the emergence of new powers, the use of internationalism as a foreign policy tool is interrogated as a response both to domestic imperatives, such as perception and identity, and systemic constraints and opportunities. Central to the analysis is an examination of the role of governing parties in foreign policymaking, both as key actors in determining policy, and as the sources of ideational constructs, in this case ‘internationalism’, that have a bearing on foreign policy. Foreign policymakers are limited in their perceptions and responses to external threats and opportunities by the domestic institutional structure, as well as by external threats and opportunities. In South Africa, responses are often limited to rhetoric, owing to limited resource extraction capacity, in spite of the highly centralised foreign policymaking structure under Mbeki. In Brazil, constitutional checks and balances also limited the state’s responses to external stimuli under Lula; yet, these responses, when they are implemented, can be more forceful owing to greater resource capacity. The ‘new Southern internationalism’, propounded by both South Africa and Brazil, is a function of domestic politics and external pressures, as evidenced by the Haiti case. These findings make a contribution to advancing the analysis of emerging powers, their trajectory and intentions in international relations, as well as the extent to which governing parties can influence foreign policy outcomes, and under which conditions

Abstract:
This thesis is about parental experiences and practices in association with disabled children in South Korea. It is first and foremost about disablement. It aims to expose the inequality and exclusion experienced by disabled children and their families. It was intended to challenge the long-established tradition in Korea of regarding the problem of disability as a personal and family issue, and aims to purposes to enhance the social recognition of the rights of disabled children and their families. This thesis is also about the development of disability-parenthood. It explores the ways in which parents construct and reconstruct their notions of parenthood and disability through their continual experiences of parenting a disabled child. It intends to resituate parents as potentially active social agents who create their own knowledge about disability. It examines how parents are struggling against disabling forces and acting on behalf of their disabled children in their own lives. Another main purpose of this thesis is to explore how parents develop their politics of disability by working together. In order to research parents' group activities, this study engaged the participation of parents involved self-help or advocacy groups. The research employed both in-depth interviews and long-term participant observation.

Abstract:
Malaria continues to pose a significant global health and socio-economic burden on those regions where it is endemic. Despite substantial investment in the delivery of artemisinin-based combination therapies, causing a fall in malaria mortality, recent data suggest that this parasitic disease still imposes a significant impact. A major problem is the narrow drug discovery pipeline, made worse by reports of artemisinin resistance. In recent years, high-throughput screening of natural products derived from plants and marine organisms has led to the discovery of potent anti-malarial indole alkaloids (such as dihydrousambarensine), many of which contain an indoloisoquinoline core. Building on previously discovered methodology in our group, we have developed a series of novel, enantiomerically pure, synthetic indoloisoquinoline and their potential as anti-malarial leads was assessed. The structure-activity relationship of these compounds was investigated in several areas and a lead compound was generated with an activity close to that of a known anti-malarial natural product dihydrousambarensine.

Abstract:
This thesis examines the notion of misdirection and queer women of colour representation in contemporary fiction and graphic narratives. Its aim is to analyse the gaps in discursive debates that overlook or minimise queer women of colour subjectivities. In so doing, it develops new lines of critical enquiry that bring into dialogue recurring themes of race, gender, class, homophobia, sexual violence and trauma. The thesis discusses five texts published between 1982 and 2008, opening up critical pathways into the interrogation of intersecting oppressions that limit and/or completely eradicate the voices and realities of queer women of colour. Developing the notion of misdirection as both an analytical and performative tool that problematises ‘difference’, this thesis shows that through contemporary framings, queer women of colour representation in literary fiction and comics/graphic narratives can be liberated from the cultural hegemony of whitecentric paradigms. By adding the visual dimension of graphic narratives/comics to unpack the sometimes explicit, but more often than not, coded messages regarding the subaltern in literary fiction, this thesis places within a contemporary context trauma narratives that fail to adequately address the unique experiences of queer women of colour.

Abstract:
The circo criollo, or Argentine native circus, arose as one of the most important forms of popular expression during the late nineteenth century. This performance context can serve as a site for exploring old and new constructions of Argentine identity that encompassed the lower and middle classes and included native and immigrant groups, particularly in Buenos Aires. Although the native circus contributed greatly to the creation of such an identity, little is known about the musical practices of the circus itself, including what types of music or dance were performed, the manner in which they were interpreted, and how the audience responded. This thesis therefore aims to bring to light these previously obscure circus traditions. It discusses the negotiation and conflict of power relationships that informs Argentine identity construction within this popular expressive medium. By examining the circo criollo as a site of hegemonic power differentials, this study probes more deeply into the contradictions that underlie such a fragile yet persistent sense of incipient Argentine identity.

Abstract:
My dissertation challenges the longstanding dismissal of love in James Joyce's texts by examining the ethical and political implications of his love stories. Primarily using Martin Buber's works (but also including perspectives derived from bell hooks and Julia Kristeva), I define love as an affirmation of otherness and adopt a critical framework that promotes the love of others over the narcissistic devotion to oneself. In so doing, I highlight love as the ultimate challenge to authoritarian systems because the embrace of the other is necessary to transcend the boundaries that alienate individuals from each other and that justify imperialist and racist political structures. I thus offer a love ethic that not only compels meaningful individual interaction, but also establishes a model for effective social and civic participation, encouraging a climate of cooperation that embraces the solidarity and empathy needed for progressive politics. I also argue that analyzing Joyce's works provides a fruitful opportunity to recognize the individual and political viability of this love ethic. Focusing on Dubliners, Stephen Hero, A Portrait of the Artist as a Young Man, Exiles, and Ulysses, I examine the relationships between his characters' pursuits of love and their socio-political struggles, arguing that their love for others directly influences their acceptance of otherness within the colonialist discourses of Joyce's Dublin. For example, James Duffy's refusal of Emily Sinico in "A Painful Case" also rejects her advice to engage in the political cooperation that would promote his socialist ideas. Similarly, Stephen Dedalus's promotion of symbolic romance over real-world attachments focuses his aesthetics on ideal beauty instead of everyday Dublin, which alienates him from his audience and limits the practical success of his art. By contrast, Leopold Bloom's love for his wife Molly reflects a broader empathy for others that encourages social dialogue and counteracts what Joyce called "the old pap of racial hatred," an element in both British imperialism and Irish nationalism. My dissertation's afterword anticipates the amorous potential of Finnegans Wake, reading ALP's concluding soliloquy as a demonstration of her enduring affection for HCE that is reignited through each iteration of the text's cyclical narrative.

In the last decade, market pressures for greater variety of products forced a gradual shift from continuous manufacturing to batch manufacturing in various industries. Consequently batch scheduling problems have attracted the attention of researchers in production and operations management. This thesis addresses the scheduling of parallel non-identical batch processors in the presence of dynamic job arrivals, incompatible job-families and non-identical job sizes. This problem abstracts the scheduling of heat-treatment furnace operations of castings in a steel foundry. The problem is of considerable interest in this sector as a large proportion of the total production time is spent in heat treatment processing. This problem is also encountered in other industrial settings such as burn-in operation in the final testing stage of semiconductor manufacturing, and manufacturing of steel, ceramics, aircraft parts, footwear, etc. A detailed literature review and personal communications with experts revealed that this class of batch scheduling problems have not been addressed hitherto. A major concern in the management of foundries is to maximize throughput and reduce flow time and work-in-process inventories. Therefore we have chosen the primary scheduling objective to be the utilization of batch processors and as secondary objectives the minimization of overall flow time and weighted average waiting time per job. This formulation can be considered as an extension of problems studied by DOBSON AND NAMBINADOM (1992), UZSOY (1995), ZEE et a/. (1997) and MEHTA AND UZSOY (1998). Our effort to carefully catalogue the large number of variants of deterministic batch scheduling problems led us to the development of a taxonomy and notation. Not surprisingly, we are able to show that our problem is NP-hard and is therefore in the company of many scheduling problems that are difficult to solve. Initially two heuristic algorithms, one a mathematical programming based heuristic algorithm (MPHA) and the other a greedy heuristic algorithm were developed. Due to the computational overheads in the implementation of MPHA when compared with the greedy heuristic, we chose to focus on the latter as the primary scheduling methodology. Preliminary experimentation led us to the observation that the performance of greedy heuristics depends critically on the selection of job-families. So eight variants of the greedy heuristic that differ mainly in the decision on "job-family selection" were proposed. These eight heuristics are basically two sets {Al, A2, A3, A4} and the modified (MAI, MA2, MA3, MA4}, which differ only on how the "job-family" index, weighted shortest processing time, is computed. For evaluating the performance of the eight heuristics, computational experiments were carried out. The analysis of the experimental data is presented in two perspectives. The goal of the first perspective was to evaluate the absolute quality of the solutions obtained by the proposed heuristic algorithms when compared with estimated optimal solutions. The second perspective was to compare the relative performance of the proposed heuristics. The test problems generated were designed to reflect real-world scheduling problems that we have observed in the steel-casting industry. Three important problem parameters for the test set generation are the number of jobs [n], job-priority [P], and job-family [F]. We considered 5 different levels for n, 2 different levels for P and 2 different levels for F. The test set reflects (i) the size of the jobs vary uniformly (ii) there are two batch processors and (iii) five incompatible job-families with different processing times. 15 problem instances were generated for each level of (n, P, and F). Out of many procedures available in the literature for estimating optimal value for combinatorial optimization problems, we used the procedure based on Weibull distribution as discussed in Rardin and Uzsoy (2001). For each problem instance of the randomly generated 300 problem instances, 15 feasible solutions (i.e., the average utilization of batch processors (AUBP)) were obtained using "random decision rule for first two stages and using a "best-fit heuristic' for the last stage of the scheduling problem. These 15 feasible solutions were used to estimate the optimal value. The generated 15 feasible solutions are expected to provide the estimated optimal value of the problem instance with a very high probability. Both average performance and worst-case performance of the heuristics indicated that, the heuristic algorithms A3 and A4, on the average yielded better utilization than the estimated optimal value. This indicates that the Weibull-based technique may have yielded conservative estimates of the optimal value. Further, the other heuristic algorithms found inferior solutions when compared with the estimated optimal value. But the deviations were very small. From this, we may infer that all the proposed heuristic algorithms are acceptable. The relative evaluation of heuristics was in terms of both computational effort and the quality of the solution. For the heuristics, it was clear that the computational burden is low enough on the average to run all the proposed heuristics on each problem instance and select the best solution. Also, it is observed that any algorithm from the first set of {Al, A2, A3, and A4} takes more computational time than any one from the second set {MAI, MA2, MA3, and MA4}. Regarding solution quality, the following inferences were made: ٭ In general the heuristic algorithms are sensitive to the choice of problem factors with respect to all the scheduling objectives. ٭ The three algorithms A3, MA4 and MAI are observed to be superior with respect to the scheduling objectives: maximizing average utilization of batch processors (AUBP), minimization of overall flow time (OFT) and minimizing weighted average waiting time (WAWT) respectively. Further, the heuristic algorithm MAI turns out to be the best choice if we trade-off all three objectives AUBP, OFT and WAWT. Finally we carried out simple sensitivity analyses experiments in order to understand the influence of some parameters of the scheduling on the performance of the heuristic algorithms. These were related to one at a time changes in (1) job-size distribution, (2) capacities of batch processors and (3) processing time of job-families. From the analyses it appears that there is an influence of changes in these input parameters. The results of the sensitivity analyses can be used to guide the selection of a heuristic for a particular combination of input parameters. For example, if we have to pick a single heuristic algorithm, then MAI is the best choice when considering the performance and the robustness indicated by the sensitivity analysis. In summary, this thesis examined a problem arising in the scheduling of heat-treatment operations in the steel-casting industry. This problem was abstracted to a class of deterministic batch scheduling problems. We analyzed the computational complexity of this problem and showed that it is NP-hard and therefore unlikely to admit a scalable exact method. Eight variants of a fast greedy heuristic were designed to solve the scheduling problem of interest. Extensive computational experiments were carried out to compare the performance of the heuristics with estimated optimal values (using the Weibull technique) and also for relative effectiveness and this showed that the heuristics are capable of consistently obtaining near-estimated) optimal solutions with very low computational burden for the solution of large scale problems. Finally, a comprehensive sensitivity analysis was carried out to study the influence of a few parameters, by changing them one at a time, on the performance of the heuristic algorithms. This type of analysis gives users some confidence in the robustness of the proposed heuristics.

The development of a methodology to improve the parametric representation of the failure criteria used to characterize rock strength of a reservoir rock in the near-wellbore region is the focus of this study. We adopted a statistical method, so-called error-in-variables (EIV), to take into account experimental errors in all of the measured variables. The proposed methodology is employed to obtain the parameters of the failure envelope (2D criteria) from experimental data in the following cases: When the experimental data are used directly to determine the failure envelope in the Mohr plane. When a failure envelope is first obtained in the principal-stress plane and then transformed into the Mohr plane using Computer Algebra. When the presence of pore fluid requires the consideration of effective stresses. When the brittle-ductile transition requires special form of the envelope (cap models) Using generalization of previously developed methods, we employed EIV methods to obtain the parameters of a failure surface in the principal-stress space (3D criteria).The basic hypothesis of this work is that the EIV method provides a better representation of failure criteria than the previous methodology. The basic application of any failure criterion is to determine whether a certain stress state will or will not lead to failure. A parametric representation is considered better than another if the likelihood of obtaining the wrong answer is less than in the other case. Therefore, the best representation (within a certain family of envelopes or surfaces) is the one minimizing the objective function, which is nothing else but the likelihood of this wrong answer. To test the basic hypothesis of this work, I compared the objective function (likelihood of erroneous decision) calculated with parametric representations obtained by various methods. To achieve this I evaluated a well-documented, published set of experimental data, for which failure envelopes have been fitted by other methods. This work is limited to the processing of data obtained in experiments conducted in homogenous, isotropic rock at isothermal conditions. Sedimentary rocks such as sandstone are the focus of this study because of their importance in near-wellbore reservoir rock stability problems. Nevertheless, the methodology developed in this work is not limited to this type of rock.

Abstract:
As in years past, use of tobacco remains the leading cause of preventable death in this country. Smoking has been associated with elevated risks of 15 other forms of and has also been identified as a major cause of such chronic conditions as cardiovascular disease, cerebrovascular disease, bronchitis and emphysema. In 2008, an estimated 8.6 million Americans suffered from smoking-related chronic conditions (American Cancer Society, 2010). As 80% of tobacco use begins in adolescence (Villanti, Boulay & Juon, 2010), this age group has long been the focus of intervention efforts. Furthermore, animal studies suggest that the adolescent brain is at increased risk for developing an addiction to nicotine compared to an adult brain (Morrell, Song & Halpern-Felsher, 2011). Additional studies have demonstrated that the younger an adolescent begins smoking, the more likely he is to become a regular smoker and less likely to quit smoking (Brown et al., 2010). The public health opportunity for primary and secondary prevention intervention is clear. To track adolescent risk-taking in the state, The Georgia Department of Education administers the Georgia Student Health Survey II [GSHS II] throughout all school districts. The purpose of this thesis study was to examine known smoking risk factors using the GSHS data in order to assess associations using an adolescent sample. Findings demonstrated that age, gender, and urbanicity were associated with smoking. Findings from this study provide insights for programming that can be tailored to meet the needs of adolescent subgroups that may be vulnerable to smoking initiation.

The current study investigated the effects of dispositional mindfulness on well-being conceptualized in two ways: as hedonic well-being and as eudaimonic well-being. These represent feeling well and functioning well, respectively. Mindfulness, a type of consciousness marked by a nonjudgmental, present centered awareness, was expected to relate more strongly with eudaimonic well-being. Data from 112 participants were collected through an online survey. The mechanisms emotion regulation, low rumination, attentive awareness to one's emotional states, and clarity and understanding of one's emotional states were examined as mediating variables between dispositional mindfulness and the two conceptualizations of well-being. Correlational analysis demonstrated that dispositional mindfulness is significantly correlated with both hedonic well-being and eudaimonic well-being, though more strongly related to eudaimonic well-being. Mediational analysis demonstrated that low rumination significantly mediated the relationship between dispositional mindfulness and hedonic well-being. Additionally, both low rumination and clarity and understanding of one's emotional states significantly mediated the relationship between dispositional mindfulness and eudaimonic well-being. Implications, limitations, and future directions of research are discussed.

Abstract:
The abuse of power, dominance of certain shared perspectives, and reduction of personal values into so-called organizational core values are problems that plague traditional organizational direction setting. The purpose of this study was to explore how power intersects with the knowledge formation process, shared perspectives, and participatory processes within organizational direction setting. The study used an ethnographic case study approach to focus on a church that went through a yearlong direction setting process. The church had lost its senior pastor and wanted to determine its direction before hiring a new pastor. The researcher was a participant in that process. His field notes and other documentation provided one source of data. The researcher also interviewed 20 people who participated in the process. In terms of the intersection of power and knowledge formation, the study revealed that the discursive practices of the facilitator along with the voices of those in privileged groups, the outspoken, and those who had engendered trust in others carried considerable weight during the process. At the intersection of power with shared perspectives, there were two major perspectives representing subgroup cultures: a traditional perspective that resisted change and a progressive perspective that wanted change. The progressives dominated the church’s privileged groups and exerted extensive influence on the direction setting process. The organizational symbols of church staff and worship music style served to galvanize some people in the battle over which perspective would prevail. Transparency functioned as a bridge that brought some on either side of the conflict closer together. The research revealed two major types of power related to the intersection of power with participatory processes: the power of pain and intimidation. Both minimized the participation of some women, youth, and traditionalists. All three intersections featured deployments of power that influenced the construction of directional knowledge. This knowledge helped to inform the rules of “appropriate” conduct within the organization’s emerging truth regime. The study revealed that, in this case, robust directional knowledge would have accommodated personal, subgroup, and widely shared values in a state of dynamic equilibrium. The researcher concluded with a discussion of implications for organizational leaders.

Abstract:
There is considerable evidence that delays in diagnosing and treating breast cancer reduce long-term survival. The purpose of this study was to assess the waiting time between diagnosis and treatment for Alberta women with breast cancer and to examine the influence of age, cancer stage, Regional Health Authority (RHA), community size, and year of diagnosis on this time interval. The data were obtained from the Alberta Cancer Board. The information included approximately all Alberta women with breast cancer between 1997 and 2000. The overall median waiting time was 17 days. The mean and median delay increased by an average of two days each year. Only 43.8% of cases were treated within the recommended 14 days. The delay was significantly longer for women younger than 70, with stage 1 disease and from Northern RHAs. Efforts must be made to decrease delay and ensure that all women receive equal access to health services.

Abstract:
Tall fescue, a cool-season grass native to Europe, central Asia, and northern Africa, has been widely distributed throughout the U.S. for use as turf and forage. Following its widespread planting, its ability to associate with a toxic fungal endophyte, Neotyphodium coenophialum, was discovered. Research has linked this fescue-endophyte association with increased biotic and abiotic stress resistance in endophyte-infected (E+) versus endophyte-free (E-) plants, and these differences may affect the ability of land managers to eradicate tall fescue and restore native grasslands. I conducted three studies to examine whether E+ tall fescue plants respond differently to management than E- plants, and whether the success of planted native species might be impacted via indirect soil effects. My overall hypotheses were that E+ plants would recover from restoration/eradication efforts better than E- plants, and that E+ fescue would reduce microbial symbionts in the soil needed by planted native species. I first conducted a field study of a tall fescue pasture consisting of four sub-units being restored with different combinations of prescribed burns and/or herbicide applications, as well as an unmanaged control. I found no evidence of E+ plants preferentially surviving restoration management; however this field had unusually low endophyte infection rates to begin with. The second study was a greenhouse experiment in which I measured growth of E+ and E- plants exposed to different watering regimes (wet, dry) and prescribed burn treatments (none, one, or two burns). Watering regime significantly affected all measured growth parameters (wet>dry), but few endophyte effects were found and when present were opposite the hypothesis (E->E+). All burned plants quickly re-grew tiller lengths comparable to the unburned control, with recovery occurring faster following the second burn compared to the first. My final study examined growth and arbuscular mycorrhizal colonization of native species planted by seed into soil from beneath E+ and E- tall fescue. I observed few differences in mycorrhizal colonization or biomass for seedlings between soil from E+ and E- tall fescue. Taken together, my results indicate endophyte status of tall fescue pastures being restored to native grassland species may not be important in governing restoration success.

Altered cerebral hemodynamics contributes to mechanisms of unexplained syncope. Wecompared dynamic interaction between respiration and cerebral autoregulation in two groups ofsubjects from 28 healthy adults. Based on development of tilt-induced presyncope, subjects wereclassified as Non-Presyncopals (n=23) and Presyncopals (n=5). Airflow, CO2, Doppler cerebralblood flow velocity (CBF), ECG and blood pressure (BP) were recorded. To determine whetherinfluences of mean BP (MBP) and systolic BP (SBP) on CBF were altered in Presyncopals, thecoherencies and transfer functions between these variables and mean and peak CBF (CBFm andCBFp) were estimated. To determine influence of end-tidal CO2 (ETCO2) on CBF, relative CO2reactivity was calculated. The two primary findings were, during tilt in Presyncopals: (1) Inrespiratory frequency region, coherence between SBP and CBFp (p=0.02) and transfer functiongain between BP and CBFm was higher (MBP, p=0.01, and SBP, p=0.01) than in Non-Presyncopals. (2) In the last 3 minutes prior to presyncope, Presyncopals had a reduced relativeCO2 reactivity (p=0.005). Thus the relationship of CBF with systemic BP was more pronouncedor cerebral autoregulation was less effective preceding presyncope. This decreasedautoregulation, secondary to decreased ETCO2, may contribute in the cascade of events leadingto unexplained syncope.

Abstract:
This study examines mRNAs of less than 5000 base pairs in size, to determine the effects of base composition on folding free energy. Statistical analysis between the native mRNA and its randomized sequences was conducted, and when comparing mRNAs in human, chimp, chicken, mouse, and several other transcriptomes, we found that the native mRNAs were more stable (greater negative free energy of folding). It has been found that when length and base composition are conserved, native mRNA sequences are more stable than random mRNA sequences. More stable folding conformations have greater negative free energy values. This negative bias in free energies can be statistically measured as a Z-score which normalizes for sequence length. In an effort to determine if sequence patterns correlate with secondary structure, a neural network (JavaNNS) was trained using three training sets (Negative-Z, Near Zero-Z, Positive-Z) separately to compare the effect of neural network learning from the folding characteristics of the gene sequences. The training sets were typically allowed to run for up to 100,000 generations, and the resulting sum square errors were periodically saved. We found that the negative Z-score training set gives lower neural network sum square errors than the positive Z-score training set, and the Z-scores near zero have the highest training error. This indicates that there are more detectable sequence patterns in genes with more secondary structure than in genes exhibiting more positive Z-scores.

Abstract:
The encompassing aim of this project was to gain a better understanding of the role of the cellular immune response to dengue virus (DV) infection. Dengue virus occurs as four distinct serotypes, called D1-D4. Symptomatic DV infection occurs as two forms of illness. The more severe form of DV infection, dengue hemorrhagic fever (DHF), is characterized by increased capillary permeability resulting in decreased plasma volume, which may be accompanied by hemorrhagic manifestations. At its most severe, DHF can result in circulatory shock and death. Epidemiological studies indicate that DHF is more likely to occur following a secondary infection with a serotype of DV other than that which caused the primary infection, and there is evidence of increased T cell activation in more severe disease. These data and others indicate that DHF may be of an immunopathological nature. The memory CD4+ T cell response of a D4-immune donor was analyzed. Bulk culture proliferative responses of peripheral blood mononuclear cells (PBMC) to noninfectious DV antigens showed the highest proliferation to D4V antigen, with lesser, crossreactive proliferation to D2V antigen. CD4+ cytotoxic T lymphocyte (CTL) clones were established by stimulation with D4 antigen using a limiting dilution method. Seven out of 15 clones recognized the D4V capsid protein. The clones showed heterogeneity in their usage of T cell receptor Vα and Vβ genes. Six of these CTL clones were crossreactive between 02 and 04, and one clone was specific for D4. Using synthetic peptides, the D4V-specific clone was found to recognize an epitope between amino acids (aa) 47-55 of the capsid protein, while the crossreactive CTL clones each recognized epitopes in a separate location, between aa 83 and 92, which is conserved between D2 and D4. These results showed that the DV capsid protein can be a target of the cellular immune response following DV infection. The bulk culture response of the donor's PBMC to the epitope peptide spanning aa 84-92 was also examined. Peptides containing this epitope induced proliferation of the donor's PBMC in bulk culture, but peptides not containing the entire epitope did not induce proliferation. Also, PBMC stimulated in bulk culture with noninfectious D4V antigen lysed autologous target cells pulsed with peptides containing aa 84-92. These results indicate that this donor exhibits memory CD4+ T cell responses directed against the DV capsid protein and suggest that the response to the capsid protein is dominant not only in vitro at the clonal level, but in bulk culture responses as well. Experiments were performed demonstrating that the CD4+ CTL clones were capable of mediating bystander lysis of non-antigen presenting target cells. Following activation on plate-bound anti-CD3 antibody or in the presence of unlabeled antigen-presenting target cells, these clones could lyse both Jurkat cells and HepG2 cells as bystander targets. Bystander lysis of neighboring, non-infected cells by activated CD4+ CTL clones might contribute to the pathology of DHF. The mechanisms of lysis employed by the T cell clones against both cognate and bystander target cells were assessed using chemical inhibitors of either the perforin- or Fas/FasL-mediated pathways. Three CD4+ CTL clones were demonstrated to lyse cognate, antigen-presenting target cells by a mechanism that primarily involves perforin, while bystander lysis occurred through Fas/FasL interactions. In contrast, one clone used a Fas/FasL mechanism to lyse both cognate and bystander targets. These experiments indicated that the perforin- and FasL-mediated mechanisms of target cell lysis are not mutually exclusive, in that a single clone can kill target cells using either mechanism. Additionally, the ability of CD4+ CTL clones to lyse target cells by the perforin pathway indicates that, like CD8+ CTL, these clones might play a role in viral clearance and recovery from infection through lysis of virus-infected cells. Cytokine production by the capsid-specific CTL clones was also examined. Six of six clones studied produced high quantities of IFN-γ in response to either D2V antigen or the epitope peptide. IFN-γ was also produced by PBMC in a bulk culture from this donor stimulated with D4V antigen. All of the clones produced both TNF-α and TNF-β following stimulation. Four of six clones produced low amounts of IL-2, and only three of six clones produced detectable amounts of IL-4. Production of cytokines by activated CD4+ T cell clones in vivo could contribute to both viral clearance and immunopathology. To better understand the role that cytokine production might play in vivo in response to DV infection, cytokine mRNA levels were examined by PCR in DV-infected Thai children. mRNA for the cytokines IFN-γ, TNF-β, TNF-α, IL-1β, and IL-6 were detectable in the PBMC of DV-infected children. Semi-quantitative PCR analysis indicated that TNF-α mRNA levels were elevated in Thai children with DHF compared to children with classical dengue fever, the less severe form of illness (p=.013). All other cytokines showed no statistically significant difference between children with DHF and those with DF, although IFN-γ showed a trend toward elevation in more severe disease (p=.l). Increased production of TNF-α and/or IFN-γ in vivo could potentially contribute to the immunopathology of severe dengue illness. Taken as a whole, the data presented in this thesis provide a better understanding of the role of the cellular immune response to dengue virus infection and its potential contribution to the immunopathology of dengue hemorrhagic fever.

The purpose of this study is to investigate the properties of a bias reduction kernel estimator of a distribution function and to compare it with existing estimation techniques in the bootstrap. The procedure which is to be investigated, was proposed by Swanepoel and van Gram (2003). Monte Carlo simulation studies were performed to compare this procedure with existing procedures in the bootstrap methodology. The simulations involved constructing 90% and 95% two-sided percentile confidence intervals and upper bounds for the mean. The simulation study provided estimates for the coverage probabilities and expected lengths of the intervals. Findings and conclusions of these simulations are reported.

Abstract:
This thesis presents a comprehensive investigation of the formation of supported lipid membranes with vesicle hemifusion, their stability under detergents and organic solvents and their applications in molecular biology. In Chapter 3, we describe how isolated patches of DOPC bilayers supported on glass surfaces are dissolved by various detergents (decyl maltoside, dodecyl maltoside, CHAPS, CTAB, SDS, TritonX-100 and Tween20) at their CMC, as investigated by fluorescence video microscopy. In general, detergents partition into distal leafets of bilayers and lead to the expansion of the bilayers through a rolling motion of the distal over the proximal leaflets, in agreement with the first stage of the established 3-stage model of lipid vesicle solubilization by detergents. However, solubilization pathways are different for different detergents. For decyl/dodecyl maltoside, CHAPS and SDS, dissolution of bilayers starts only once the bilayers reach a critical lipid:detergent ratio in the bilayers, whereas for CTAB, TritonX and Tween20, dissolution starts once detergents partition into the bilayers. From the expansion dynamics of bilayers, we conclude that the energy loss due to the partitioning of detergents is balanced by the released heat and the increment of the planar bilayer's curvature energy. Moreover, in contrast to a previous view that detergents need to be present on both leaflets of bilayers to initiate solubilization, we find that detergents in the distal leaflets of bilayers are sufficient to induce micellization. Lastly, we estimate the energy barrier for detergents to partition into lipid bilayers and find it is on the order of 3 to 7 kT. Subsequently, we study the partitioning of organic solvents (methanol, ethanol, isopropanol, propanol, acetone and chloroform) into isolated bilayer patches on glass in Chapter 4 with fluorescence microscopy. The area expansion of bilayers due to the partitioning of organic solvents is measured. From the titration of organic solvents, we measured the rate of area expansion as a function of the volume fraction of organic solvents, which is proposed to be a measure of strength of interactions between solvents and membranes. From the same experiments, we also measure the maximum expansion of bilayers (or the maximum binding stoichiometry between organic solvents and lipids) before structural breakdown, which depends on the depth of penetration of solvents to the membranes. From the partitioning dynamics, it is observed that bilayers expand the area through the same rolling motion as described for detergent partitioning in Chapter 3. The energy loss due to the partitioning of organic solvents was balanced by the released heat and the increment of planar bilayer's curvature energy. Upon desorption of organic solvents, the bilayers shrink the area through the edges and by forming pores within the bilayers, concomitantly increase the contour length of bilayer edges, indicating self-healing is impossible for this system. In Chapter 5, we investigate the formation of sparsely-tethered bilayer lipid membranes (stBLMs) with vesicle hemifusion. In vesicle hemifusion, lipid vesicles in contact with a hydrophobic alkyl-terminated self-assembled monolayer (SAM) deposit a lipid monolayer to the SAM surface, thus completing the bilayer. Electrical Impedance Spectroscopy and Neutron reflectivity are used to probe the integrity of stBLMs in terms of their insulating and structural properties. Preparation conditions are screened for those that are optimal for stBLM formation. Concentrations of lipid vesicles, hydrophobicity of SAMs, the presence of calcium and high concentrations of salt are identified as the key parameters. We show that stBLMs can be formed with vesicles of different compositions. Vesicle hemifusion opens up a new route in preserving the chemical compositions of stBLMs and facilitating membrane proteins incorporation. In Chapter 6, we visualize the hemifusion pathway of giant unilamellar vesicles (GUVs) with planar hydrophobic surfaces at the single vesicle level with fluorescence video microscopy. When a GUV hemifuses to a surface, its outer leaflet breaks apart and remains connected to the surface presumably through a hemifusion diaphragm. Lipids from the outer leaflet are transferred to the surface as a lipid monolayer that expands radially outward from the hemifusion diaphragm, thereby forming the loosely packed outer hemifusion zone. The tension of the outer leaflet rises as lipids are transported to the outer hemifusion zone until it is large enough to rupture the GUV. Therefore, a pore forms near the hemifusion diaphragm through which the water encapsulated within the GUV is expelled. Additionally, lipids ipped from the inner to outer leaflet via the pore and are transferred to the surface. The hemifusion of lipids from both leaflets to the surface leads to the formation of the inner hemifusion zone densely packed with lipids. The inner and outer hemifusion zones expand radially as concentric discs with a rate of about 1000 m2=s, suggesting the expansion is driven by surface hydrophobicity. The spreading dynamics of the lipid monolayer is consistent with a model where the energy dissipated by friction between the monolayer and the surface is equal to the difference of surface energy when the surface is covered by a lipid monolayer. The mechanism revealed in this work provides insights into membrane reorganization and improves the understanding of vesicle-surface interactions. The tethering of membranes is a common process regulating membrane fusion throughout the secretory pathway. The Prof. Linstedt laboratory at the CMU Department of Biological Sciences has been focusing on the elucidation of the mechanism of Golgi membrane tethers, GRASP (Golgi Reassembly And Stacking Proteins), which are essential for the formation of the characteristic ribbon structure of the Golgi apparatus. Typically, such work involves in vivo expression of these proteins which tracks how specific mutations affect organelle morphology. However, it remains unclear in such investigations, whether a certain active protein is sufficient to trigger the formation of the characteristic structure or if other factors, for example auxiliary proteins, are involved. Therefore, in Chapter 7, we develop an in vitro assay employing stBLMs and lipid vesicles to examine the functionality of GRASP in membrane tethering. Membrane-bound GRASP on opposing membranes dimerizes and tethers fluorescently-labeled vesicles to stBLMs. The fluorescence intensity of images taken at stBLM surfaces is used to quantify the tethering activity. Both wild type and mutant proteins were studied to shed light on the molecular mechanism of tethering. We show that the GRASP domain is sufficient and necessary for membrane tethering. In addition, the tethering capability of GRASP is impaired when the internal ligands and the binding pockets participating in dimerization are deleted and mutated. Membrane anchors, sizes of vesicles and membrane compositions are explored for their influence on the outcomes of the assay. Furthermore, preliminary analysis from neutron reflectivity measurements shows that both the internal ligands and binding pockets are exposed instead of buried toward the membrane surface. In summary, we establish a functional assay for studying GRASP activity in vitro. This assay may also be used for studies of similar supramolecular structure formation processes in molecular biology.

Through analyses of six novels: 1) God Bless the Child, 2) Breath, Eyes, Memory, 3) The Hate U Give, 4) The Bluest Eye, 5) Daddy Was a Number Runner, and 6) The Poet X, this research presents rage as a canonical theme in Black women’s coming-of-age narratives and presents connections between rage, rights, and resistance. The connections, revealed through stimuli and adaptations associated with rage, frame an argument for North Americas as an arbiter of anger. The novels construct an “arc of anger” that places them in conversation about Black girl rage and presents a tradition of Black women crafting Black girl protagonists who are conduits for counternarratives of rage. This dissertation also examines how history, memory, and culture contribute to Black girls’ frustrations and knowledge bases. By looking to works published between the angry decade (the 1960s) and the age of rage (the 2010s), the research presents ways Black women novelists and their characters return to rage to combat social institutions and critique social constructions of Black girlhood and womanhood.

The term “at risk” is a label that is used to describe students who encompass a host of prominent socially and politically constructed titles that are intended to simplify student understanding and awareness and allow for clear reporting. The purpose of this study was to demythologize the concept of “at risk” by creating the conditions for student voice and critical dialogue to emerge, through the use of narrative inquiry. This research hoped to provide an outlet for young people to find and use their own voices, while finding their own place within their lived histories. The research also aimed to raise awareness of the reality of the contemporary U.S. educational system: we often create policies and programs without considering the perspectives of the young people whom these services are designed to serve. Through critical narrative inquiry, six former student’s engaged in unstructured interviews and a focus group. Through analysis of the data set, five themes emerged and include 1) relationships with bicultural adults who understand, 2) instrumentalizing pedagogy, 3) the impact of money-driven policy, 4) the awareness of limitations of opportunities, and 5) the overall theme of the transparency of hegemony. This research hoped to problematize the label in an effort to move toward an emancipatory understanding of how we speak about young people and make sense of the circumstances these young people must navigate through their education and their world.

The importance and the contribution of string matching algorithms to the modern society cannot be overstated. From basic search algorithms such as spell checking and data querying, to advanced algorithms such as DNA sequencing, trend analysis and signal processing, string matching algorithms form the foundation of many aspects in computing that have been pivotal in technological advancement. In general, string matching algorithms can be divided into the categories of exact string matching and approximate string matching. We study each area and examine some of the well known algorithms. We probe into one of the most intriguing data structure in string algorithms, the suffix tree. The lowest common ancestor extension of the suffix tree is the key to many advanced string matching algorithms. With these tools, we are able to solve string problems that were, until recently, thought intractable by many. Another interesting and relatively new data structure in string algorithms is the suffix array, which has significant breakthroughs in its linear time construction in recent years. Primarily, this thesis focuses on approximate string matching using dynamic programming and hybrid dynamic programming with suffix tree. We study both approaches in detail and see how the merger of exact string matching and approximate string matching algorithms can yield synergistic results in our experiments.

Platinum complexes (cisplatin, carboplatin and oxaliplatin) are effective anticancer agents. However the major drawbacks of platinum chemotherapy are toxic side effects and resistance. The affinity of platinum complexes to sulfur donor ligands of side chains of methionine and cysteine amino acids was assumed to be responsible for toxicity and resistance. Recently, it was found that the reaction of platinum complex with proteins containing sulfur donor ligands could actually favor its anticancer activity. Copper transporter 1 (Ctr 1), a protein involved in the transport of copper into the cell, also helps in the influx of cisplatin by binding to N-terminal domain of Ctr 1 which is rich in methionine and histidine residues. A better understanding of how the size and shape of amine ligand, and leaving groups affect the reaction of platinum (II) complexes with methionine could give new ways to optimize its anticancer activity. This preliminary research focuses to answer this by HPLC-UV-VIS analysis of bulky platinum complexes including [Pt(dien)Cl]Cl, Pt(Me4en)(NO3)2 and Pt(en)(NO3)2 with two methionine containing small peptides that serve as models for protein interactions.

Viver em sociedade é extremamente complexo, sendo necessário de tempos em tempos verificar como as instituições constituídas atuam para cumprirem seus deveres constitucionais e se a jurisdição consegue ter efetividade em estrita obediência aos Direitos Humanos. Necessário ressaltar que os direitos humanos não merecem uma visão estreita, no sentido de que são destinados à defesa daqueles que cometem crime – como muitas pessoas acreditam. Os direitos humanos visam a garantir a todos, indistintamente, o cumprimento dos preceitos constitucionais, principalmente o da dignidade da pessoa humana e os direitos fundamentais. Se as leis em vigência forem cumpridas, os direitos humanos a priori também serão respeitados. Para tanto, faz-se necessária a análise de dados levantados cientificamente a fim de se conhecer como o órgão estatal (Poder Judiciário), responsável pela prestação jurisdicional de resultado, está atendendo ao texto constitucional. Por estar investido de parcela do Poder Estatal, presume-se que está trabalhando conforme os ditames constitucionais, fazendo todo o necessário para que a paz social seja restaurada. Há de se verificar se o Poder Judiciário está cumprindo seu dever e prerrogativas e se o processo possui um rito que permite a sua duração em prazo razoável. E, no caso de procedência dos pedidos, o Poder Judiciário consegue entregar o bem da vida buscado, ou seja, dar eficácia e efetividade considerando a utilidade do processo? Houve ressarcimento da vítima proporcional ao dano sofrido e em tempo razoável para que possa minorar e compensar o seu sofrimento? A pesquisa deveria ser mais abrangente, ocorre que, em razão de dificuldades materiais, não há como agregar ao trabalho outros pesquisadores, ressaltando que os levantamentos foram realizados apenas por este mestrando no âmbito da Segunda Vara Cível da Comarca de Gurupi e junto à Polícia Militar do Estado do Tocantins (4º BPM). O Mapa da Violência de 2013, divulgado pelo Ministério da Saúde, mostra que, proporcionalmente, o Tocantins está em primeiro lugar no aumento do número de pessoas que morrem no trânsito. Na pesquisa, que traz números de 2001 a 2011, são 37,9 mortes para cada 100 mil habitantes no Estado. Em 2011, foram 531 mortes no trânsito, o que representa 64,9%, comparando a 2001, quando foram registradas 322 mortes. O comportamento irresponsável no trânsito, em especial o de dirigir sob efeito de álcool ou outras drogas, é uma preocupação constante das autoridades. Em 2011, a ONU proclamou a Década de Ação pela Segurança no Trânsito e lançou um desafio para os países: reduzir pela metade o número de mortes no trânsito em um prazo de 10 anos. Diante disso, investigam-se as sentenças proferidas em ações indenizatórias provenientes de acidente de trânsito, na Comarca de Gurupi, especificamente na Segunda Vara Cível, no período de setembro de 2012 (início do processo eletrônico na Comarca) até setembro de 2015, respondendo ao final qual foi o prazo da prestação jurisdicional efetiva, e qual seria o prazo razoável do processo para essas ações específicas. Ao final, constatando-se o não cumprimento do princípio, serão apontadas possíveis propostas de soluções à demora. De forma indutiva, foram analisados os dados levantados em processos judiciais, dados da Polícia Militar de Gurupi/TO, pesquisas disponíveis no CNJ e Segurador Líder (DPVAT) e pesquisa bibliográfica sobre o assunto.
Living in society is extremely complex, requiring from time to time checking how institutions established act to fulfill their constitutional duties and whether jurisdiction can be effective in strict adherence to human rights. It’s necessary to point out that human rights do not deserve a narrow view, in the sense that they are aimed at defending those who commit crime - as many people believe. The Human Rights are intended to guarantee to all, without distinction, compliance with the constitutional provisions, especially the human dignity and the fundamental rights. If the laws in effect are met, a priori human rights will also be respected. To this end, it is necessary the analysis of data collected scientifically to be known how the state agency (Judiciary) responsible for the jurisdictional service of results is considering the constitutional text. By being invested of part of the State power, it is assumed that it is working according to constitutional principles, doing everything necessary so that social peace is restored. It must be checked if the judiciary is doing its duty and prerogatives and if the process has a rite that allows its duration within a reasonable time. And in the case of origin of the requests, can the judiciary deliver the good of life sought, ie, give efficiency and effectiveness considering the process use? Was there victim compensation proportionate to the damage suffered and in reasonable time in order to minimize and compensate their suffering? Surely that the research should be more comprehensive, what happens is that, due to material difficulties, there is no way to add to the work other researchers, noting that the surveys were conducted only by this master student under the Second Civil Court of the Gurupi County together with the military police of Tocantins State (4th BPM). The Map of Violence of 2013, released by the Ministry of Health, shows that, proportionally, Tocantins is in first place in the rise in the number of people who die in traffic. In the survey, which brings numbers from 2001-2011 there are 37.9 deaths per 100,000 inhabitants in the state. In 2011, there were 531 traffic deaths, representing 64.9% compared to 2001 when 322 deaths were recorded. The irresponsible behavior in traffic, especially of driving under the influence of alcohol or other drugs, is a constant concern of the authorities. In 2011, the United Nations proclaimed the Decade of Action for Safety in Traffic and issued a challenge to the countries: halving the number of traffic deaths in a period of 10 years. Therefore, we investigate the judgments in suits for damages from traffic accident in Gurupi County, specifically in the Second Civil Court, from September 2012 (the beginning of the electronic process in the County) until September 2015, responding to end which was the deadline for the effective adjudication, and what would be a reasonable time of the procedure for these specific actions. At the end, confirming the failure to comply with the principle, it is pointed out possible proposals for solutions to the delay. Inductively, the data raised in legal proceedings were analized, the Military Police data from Gurupi / TO, research available in the CNJ and Seguradora Líder (DPVAT) and literature on the subject.

Thienoacenes represent an intriguing class of organic semiconducting molecules with potential applications in organic electronics. Some of thienoacenes have been reported with high charge carrier mobility in organic field-effect transistors (OFET). OFETs based on naphtho[2,1-b:3,4-b’]dithiophene (NDT) exhibited moderate device performance and low-band gap donor-acceptor copolymers based on NDT showed a promising solar power conversion efficiency. In this thesis, four novel series of thienoacenes based on naphtho[2,1-b:3,4-b’]dithiophene backbone were designed and synthesized for OFET applications. Firstly, a novel series of p-type semiconducting naphthodithieno[3,2-b]thiophene derivatives (NDTT-n) composed of six-fused aromatic rings were designed and synthesized (Figure 1). The OFETs based on NDTT-10, and NDTT-12 fabricated by vacuum deposition showed a hole mobility of 0.22 and 0.13 cm2/(Vs), respectively with Ion/Ioff above 107 after annealing at 80 oC. Secondly, the derivatives of NDT fused with benzene rings at the flanks of thiophene, namely NBBT-n (Figure 2) were also designed and synthesized. OFETs based on NBBTF-10 fabricated by vacuum deposition exhibited a hole mobility of 0.35 cm2/(Vs) with a current on/off ratio of 106 107 after annealing at 160 oC. Further extension of π-conjugation of NDTT by incorporating with fused thiophenes leading to a new NBTBT-n series was also developed (Figure 3). The OFETs fabricated by NBTBT-10 showed the hole mobility up to 0.25 cm2/(Vs) with a current on/off ratio of 105 106 after annealing at 220 oC. Lastly, two dimensionally π-extended, butterfly-shaped thienoacenes (Figure 4) were also synthesized. The OFETs based on SMB-10 fabricated by spin-coating showed the best performance in this series with an average mobility of 0.027 cm2/(Vs) for five devices and the highest mobility of 0.038 cm2/(Vs) with a current on/off ratio of 106 107 by from chloroform. Friction StirWelding (FSW) has been researched for a number of years since its inception in 1991. The work thus far has been based on understanding the material and thermal flow using the standard fixed pin tool. The keyhole resulting during tool extraction in a FSW weld, is a disadvantage and a current limiting factor. Eliminating this effect from a weld using a movable pin tools would make FSW more commercially viable. This dissertation focuses on the design of a novel retractable pin tool, and highlights the problems encountered during the welding of Aluminum plates, Al2024 and Al5083. Previously studied techniques of material and thermal flow were used, to investigate the effect of the tool during extraction in a FSW weld. A prototype retractable tool was designed using parametric and axiomatic design theory, and implementing a pneumatic muscle actuation system. The resulting problems in the calibration of the retractable pin tool and the resulting welds are presented, these results confirming previous studies. The movable pin produced discrepancies the heat generation around the shoulder during a FSW weld. The failure of this tool to produce a reasonable weld showed that previous ideas into the workings of a retractable pin tool requires further investigation, furthermore a fresh approach to the interpretation and understanding of the FSW weld process needs consideration.

In my dissertation, I argue that Marina Carr creates liminal spaces in her plays, exploring the tensions inherent in the issues of landscape, gender, and identity. She uses these liminal spaces to expose her audiences to more complex conceptions of Ireland in the twenty-first century. For example, Carr frequently challenges perceived notions of gender identity, drawing attention to gender as performance and creating female protagonists who resist their roles as wives and mothers. Landscape is also an important element of Carr's plays; most frequently she uses the landscape of the Irish Midlands as a space that is liminal both in terms of its geography in the center of the country and in terms of the bogs, which are neither land nor water. Finally, throughout her plays she combines elements of the Irish dramatic tradition with non-Irish elements as a way of expressing Ireland's complicated post-Celtic Tiger identity. I address Carr's plays chronologically in an attempt to trace her development of her use of liminality, which begins primarily with gender in Low in the Dark and expands to include landscape and identity through the Midlands plays. Most recently, plays like Woman and Scarecrow and The Cordelia Dream are set in the liminal moments between life and death and in the unconscious world of the characters' dreams, illustrating Carr's continuing exploration of new liminal spaces.

TITLE: A GIS APPROACH TO PRIORITIZE PRIVATE LANDHOLDINGS IN THE LOWER KASKASKIA RIVER CORRIDOR INCORPORATING MIGRATORY BIRD HABITAT CRITERIA MAJOR PROFESSOR: Dr. Eric Holzmueller Loss of forested area and corresponding increase in forest fragmentation has decreased Neotropical migrant bird habitat quality across the Midwestern United States. Typically, efforts to increase habitat quality by reforesting agricultural areas are done on a first come, first serve basis. In order to increase the efficiency of these restoration efforts, a prioritized ranking system is needed to obtain the greatest increase in habitat quality possible for the fewest amount of hectares restored to forest. This project examines the use of a GIS based multi-criteria approach to prioritize lands for reforestation in the Kaskaskia River Corridor (KRC), Illinois. We prioritized areas for reforestation based on nine landscape metrics: available agricultural land, forest cover gaps, edge density metric, proximity to river, 200 m corridor area, total forest core area metric, fringe core area, distance to primary core value, and primary core area. The multi-criteria analysis revealed that high priority areas for reforestation were most likely to be close to the riparian corridor and existing large blocks of forest. Analysis of simulated reforestation (0, 0.5, 1.0, 5.0 10.0, 25.0, and 50.0% of highest priority parcels reforested) revealed different responses for multiple landscape metrics used to quantify forest fragmentation following reforestation, but indicated that the KRC would get the greatest rate of return on reforestation efforts by reforesting 10.0% of the highest priority areas. This project demonstrates how GIS and a multi-criteria analysis approach can be used to increase the efficiency of restoration projects. This approach should be considered by land managers when attempting to identify the location and quantity of area for restoration within a landscape.

The neoclassical growth model with two sectors in production is employed in this paper in order to investigate how a change in the tax structure affects informality and welfare. We calibrate and simulate the model and find that welfare always increases when we reduce the tax rate on the demand for labor and adjust the tax rate on the value added so that the government revenue remains constant.

Luego el agente de percepción entregará a la Sunat el importe de las percepciones efectuadas. Otro de los temas a tratar es la liquidez, la cual se define como la capacidad de pago con la que cuenta una empresa, para afrontar sus obligaciones conforme estas se vayan venciendo, cumpliendo así con la cancelación de sus pasivos. La liquidez es de vital importancia para cualquier empresa, ya que significa que la misma se encuentra en situación de equilibrio financiero. El último tema que se tratará es la importación, que es el régimen aduanero que permite el ingreso legal de mercancías del exterior destinadas al consumo. Este régimen es muy beneficioso para el país importador, ya que permite el mejoramiento de la tecnología y la tecnificación de dicho país, contribuye a que las empresas puedan mantenerse con los estándares de competitividad, genera el incremento de la recaudación tributaria, entre otros. Del estudio realizado de las variables antes mencionadas, podemos concluir que el régimen de percepciones del Impuesto General a la Ventas impacta negativamente en la liquidez de la empresa importadora Motor Import SAC; ya que la empresa al tener que desembolsar un pago adelantado por un bien importado, el cual no podrá ser recuperado sino hasta que dicho bien sea vendido, originaran que al mismo tiempo la empresa no pueda reinvertir su capital de trabajo, originando que la empresa solicite productos financieros, incrementando así los gastos financieros, y disminuyendo la rentabilidad de la empresa.

H-matrix techniques use a data-sparse tree structure to represent a dense or a sparse matrix. The leaves of the tree store matrix sub-blocks that are represented in full-matrix format or Rk-matrix (low rank matrix) format. H-matrix arithmetic is defined over the H-matrix representation, which includes operations such as addition, multiplication, inversion, and LU factorization. These H-matrix operations approximate results with almost optimal computational complexity. Based on the properties of H-matrices, the H-matrix preconditioner technique has been introduced. It uses H-matrix operations to construct preconditioners, which are used in iterative methods to speed up the solution of large systems of linear equations (Ax = b). To apply the H-matrix preconditioner technique, the first step is to represent a problem in H-matrix format. The approaches to construct an H-matrix can be divided into two categories: geometric approaches and algebraic approaches. In this thesis, we present our contributions to algebraic H-matrix construction approaches and H-matrix preconditioner technique. We have developed a new algebraic H-matrix construction approach based on matrix graphs and multilevel graph clustering approaches. Based on the new construction approach, we have also developed a scheme to build algebraic H-matrix preconditioners for systems of saddle point type. To verify the effectiveness of our new construction approach and H-matrix preconditioner scheme, we have applied them to solve various systems of linear equations arising from finite element methods and meshfree methods. The experimental results show that our preconditioners are competitive to other H-matrix preconditioners based on domain decomposition and existing preconditioners such as JOR and AMG preconditioners. Our H-matrix construction approach and preconditioner technique provide an alternative effective way to solve large systems of linear equations.

This thesis investigated the restoration of mauri (life-force) to Ōkahu Bay, Auckland New Zealand. Ōkahu Bay is part of the land and waters of Ngāti Whātua Ōrākei, a Māori hapū (sub-tribe). Ngāti Whātua Ōrākei has been driving the restoration, restoring Ōkahu Bay based on their worldview, visions, and concerns. This vision and control of the restoration process allows them to bring in the hapū in sustainable engagement and have the long-term vision and commitment necessary for self-determination. However, while there has been progress with projects and improved decision-making authority, hapū members are still not seeing their whānau (family) swimming in and caring for Ōkahu as much as they would like. Interviewees wanted to see an explicit focus on encouraging hapū members to use the bay, such as more educational programs and water-based activities, and continued efforts to improve water quality. Shellfish populations have also not recovered after a decade of monitoring due to structural aspects such as existing stormwater pipes. Changing these requires Auckland City Council to make stronger commitments to supporting Ngāti Whātua’s restoration. Overall, this investigation showed that in this restoration, a clean environment is essential to build community and a community is essential to build a clean environment. Abstract:
Cyberlink:Brainfingers is a human computer user interface that supports hands free computer access. This is especially valuable technology for disabled individuals who lack the motor skills necessary to use a keyboard and mouse easily. Over the course of two months, four students with cerebral palsy, a motor disorder, used Brainfingers to play a variety of spatial software. The primary aims were to see if use of Brainfingers with spatial software could improve the spatial motor abilities of students with cerebral palsy, as well as to assess any apparent behavioral changes. Two of the students showed a large increase in spatial abilities when assessed, and all students displayed improvement or no change in positive behavioral attributes. All students improved their control of Brainfingers over time, and expressed their desire to use Brainfingers in the future over other methods of accessing a computer. The implications of these results are discussed, as well as issues for further study.

Abstract:
The research in this dissertation explored the sources and chemistry of organic nitrogen aerosols in the atmosphere. Two approaches were employed: field measurements and laboratory experiments. In order to characterize atmospheric aerosol, two ambient studies were conducted in Cache Valley in Northern Utah during strong winter inversions of 2004 and 2005. The economy of this region is heavily dependent on agriculture. There is also a fast growing urban population. Urban and agricultural emissions, aided by the valley geography and meteorology, led to high concentrations of fine particles that often exceeded the national ambient air quality standards. Aerosol composition was dominated by ammonium nitrate and organic species. Mass spectra from an aerosol mass spectrometer revealed that the organic ion peaks were consistent with reduced organic nitrogen compounds, typically associated with animal husbandry practices. Although no direct source characterization studies have been undertaken in Cache Valley with an aerosol mass spectrometer, spectra from a study at a swine facility in Ames, Iowa, did not show any evidence of reduced organic nitrogen species. This, combined with temporal and diurnal characteristics of organic aerosol peaks, was a pointer that the organic nitrogen species in Cache Valley likely formed from secondary chemistry. Application of multivariate statistical analyses to the organic aerosol spectra further supported this hypothesis. To quantify organic nitrogen signals observed in ambient studies as well as understand formation chemistry, three categories of laboratory experiments were performed. These were calibration experiments, smog chamber studies, and an analytical method development. Laboratory calibration experiments using standard calibrants indicated that quantifying the signals from organic nitrogen species was dependent on whether they formed through acid-base chemistry or via secondary organic aerosol pathway. Results from smog chamber reactions of amines with ozone, nitrogen oxides, nitrate radical, and nitric acid showed that the secondary organic aerosol pathway was more plausible than acid-base chemistry, thus making the contribution of the organic nitrogen species to the total aerosol mass in Cache Valley significant. Gas phase and aerosol products formed from the smog chamber reactions were identified and used to devise reaction mechanisms. Finally, an ion chromatographic method for detecting and quantifying some key organic nitrogen species in aerosol was developed and tested.
