Abstract:

The aim of this diploma thesis is to find proper use of object at Kapitulská 22 street in Trnava, Slovakia. Object is located in the close city centre, in its historical context. According to this I tried to keep current character of place and add new function which will use its potential. In line with current escalating problem of oncological diseases I decide to built there Maggie’s centre – community centre for people which suffer from an kind of it. Centre provides them with a platform where they can meet, change experiences and spend time during activities which is fitted to them. Peoples can find there professional psychological help and support from people with same destiny. In my design I use current object which is faced to street as service part with accommodation of clients. To this part I added community centre itself. Centre is separated into five modules which are connected by main corridor. Everywhere between this module if designed gardens and whole this area is roofed by transparent construction to keep it usable whole year.

Abstract:

This thesis deals with the issue of power flow analysis in a medium voltage (22kV) distribution network with a focus on the influence of distributed energy generation recources. At first the issue of operation of distributed energy resources and their reverse effects on the distribution system is presented in the thesis. Furtherly there are described their operational conditions from the legislation point of view. The part dealing with calculation compares two network configurations in terms of power quality and how the power quality is impacted by distributed energy resources. The attention is given to voltage conditions, short-circuit power, power conditions, losses and flicker. The first configuration corresponds to the real situation in the network, the other one is describing the changes after the new 110/22 kV substation implementation to the power supply area and the overal change of network configuration. The network model and run simulation is done using the E-Vlivy program.

Syftet med studien är en djupdykning i satsens grundstruktur: funktionsanalys, formanalys, motivanalys, sonatform samt en beskrivning av min instuderingsprocess. Jag guidar läsaren genom verket och kommenterar parallellt mina konstnärliga val. Studien påvisar att verket genomsyras av en kammarmusikalisk dominans såväl som det innehåller en genomgående violinistisk naturlighet som tydligt återspeglar Wieniawskis gedigna personliga violinkännedom.

The purpose of this MBA Project was to investigate and provide a comprehensive analysis of the supporting requirements for the V-22 Osprey from the perspective of the United States Navy and Marine Corps. The primary objective was to assist in determining the specifics that will be necessary to support the successful fielding of the weapons system. Various analysis techniques help identify and document specific requirements including training, spares, and the contracting options that will be necessary to support the overall weapons system from "tip to tail".. This report also includes potential outliers or issues related to the current plan. The benefit of choosing a commercial support plan in the form of Performance Based Logistics versus a traditional government supportability plan is measured in readiness. Impacts to cost over the lifecycle of the weapons system are measured in flying hours per dollar.

Abstract:

The diploma thesis processes project documentation in the stage of construction for modernization of an apartment house near the center on the street Vlhká 22 in Brno. The building is divided into four residential floors and nonresidential basement and attic. On the first floor there are two residential units and the other four residential units. The foundation structures are made of solid bricks. The vertical load-bearing and non-load-bearing structures are also made of solid bricks. Exceptionally drywall partitions. In the basement, the ceiling structures are made of brick barrel vaults, above the above-ground floors there are wooden beamed ceilings. The roof structure of the building is made of purlin system with standing stool and covering of ceramic roof tiles. Three additional housing units will be built in the attic. In the basement, the masonry will be undercut by a chain saw, partly by pressure grouting and partly by a new layer with a waterproofing layer. All wood-beamed ceilings will be replaced with prefabricated ceiling beams with aerated concrete inserts without overhead slab. The garden part of the building will be insulated with mineral wool. The roof structure will be constructed from the street side as a shed roof and in the garden part a flat roof with a gradient layer of expanded polystyrene thermal insulation and a waterproofing layer of thermoplastic poleolefin.

Abstract:

This Thesis is concerned with the concept of grounding of 110/22 kV transformer station. The aim of the Thesis is to create a ground network for grounding a specified outdoor substation. This 110 kV outdoor substation is formed according to H-scheme and encircles a wide area. In additon to this, an earth potential has to be distributed as equally as possible within the whole area of the substation. The most convenient alternative from the point of view of effective grounding and distribution of potential is a grid network. Therefore, the grid network is the means of grounding which I have chosen as a solution for this issue. In this solution, flat conductor network cables are used to cover the whole substation, with ground rods placed at the points of intersection. Parts of appliance can be then connected to this existing network, thus creating several parallel channels which provide grounding with low resistance. The network is made of common steel zinc-coated stip, size 30 x 4 mm. When proposing a concept of grounding and adjustment of ground electrodes, we have to take account of grounding requirements, stated according to ČSN standards. The proposed earthing system was created in appropriate size in order to meet all these grounding requirements. These requirements are also mentioned and described in a theoretical part of the Thesis. Second part of the Thesis consists of proposed concept of the grounding itself, together with calculated parameters. The grounding has to be carried out according to both safety regulations and electrical appliance requirements. The issue of grounding substations of power system is resolved by the ČSN standards 33 3201 (“Elektrické instalace nad AC 1kV”), 33 2000-4-41 (“Elektrická zařízení – Ochrana před úrazem elektrickým proudem”) and 33 2000-5-54 (“Elektrická zařízení – Výběr a stavba elektrických zařízení – Uzemnění a ochranné vodiče”) and by the PNE company standards 33 0000-4 (“Příklady výpočtů uzemňovacích soustav v distribuční a přenosové soustavě dodavatele elektřiny”) and 33 0000-1 (“Ochrana před úrazem elektrickým proudem v distribuční soustavě dodavatele elektřiny”). These standards apply to creating a concept of grounding and grounding of new, reconstructed or expanded electrical substations of power system.

Abstract:

L'interleukine 22 (IL-22) est une cytokine majoritairement sécrétée par des populations de lymphocytes T et innés. Bien qu'uniquement produite par le système immunitaire, elle agit presqu'exclusivement sur les cellules épithéliales par l'intermédiaire d'un récepteur membranaire dénommé IL-22R, principalement au niveau des tissus formant les barrières de l'organisme comme l'intestin, les poumons ou la peau. Par ses actions, elle permet de renforcer efficacement les propriétés de barrière et la régénération des épithéliums, exerçant ainsi des fonctions majeures dans le maintien de l'homéostasie de ses tissus cibles. Pour autant, de nombreux travaux ont également indiqué que des effets de l'IL-22 majorés ou prolongés pouvaient s'avérer délétères. Un rôle de l'IL-22 est ainsi clairement admis dans la physiopathologie du psoriasis et fortement suggéré dans le cancer colorectal. Ceci implique donc que les propriétés de l'IL-22 soient finement régulées et justifie l'existence d'un inhibiteur soluble, l'IL-22 binding protein (IL-22BP). Très peu de choses sont encore connues sur l'IL-22BP. Ce travail présente donc pour objectifs principaux de mieux en caractériser les sources cellulaires et la régulation, ainsi que de comprendre le rôle physiopathologique que son contrôle des actions de l'IL-22 pourrait exercer dans les maladies inflammatoires chroniques de l'intestin et le psoriasis. Une meilleure compréhension de l'impact de la régulation de l'IL-22BP sur l'axe IL-22/IL-22R est en effet importante afin de mieux définir la place de l'IL-22 dans la physiopathologie de ces maladies, et donc l'intérêt ou non d'en moduler les actions à des fins thérapeutiques
IL-22 is a cytokine mostly produced by T and innate lymphocytes. Despite a restricted production to the immune system, IL-22 almost exclusively acts on epithelial cells at barrier surfaces, through binding to its membrane receptor, namely IL-22R. IL-22 actions allow reinforcing the barrier function and regeneration of its target tissues like gut, lungs and skin, and are thus important in maintaining tissue homeostasis. However, several studies also indicate that exacerbated or prolonged actions of IL-22 may become deleterious, as illustrated in psoriasis or colorectal cancer. A strict control of IL-22 actions is consequently required and justifies the existence of IL-22 binding protein (IL-22BP), its soluble inhibitor. Almost nothing is currently known on IL-22BP. This work thus aimed at characterizing the cellular sources and regulation of IL-22BP, as well as understanding its pathophysiological role in controlling IL-22 effects during inflammatory bowel diseases and psoriasis. A better understanding of the IL-22BP-mediated control of IL-22/IL-22R axis is indeed required in order to better define the pathophysiological relevance of IL-22 production in these diseases, and thus the benefits that would be expected or not from its therapeutic modulation

Abstract:

The theoretical part of this thesis deals with distribution networks in terms of distribution according to different criteria, describes the main elements of electric distribution networks and their qualities. The following describes the methods of calculating the steady state of electrical networks. These methods are nowadays used as basics for computer programs. In the penultimate chapter of the theoretical part are summarized the principles for the development of present networks and construction of new electrical networks. The aim of practical part of the thesis is to assess the current condition of the network 22 kV in the city of Brno and assess the condition after installation of 110/22 kV transformation in Brno-Sever switching substation. After evaluating the results of steady state electrical network and identifying certain problems is the following aim a proposal of solving problems that emerged in the network. The result should be operational, functional and reliable electric distribution network. This thesis used to calculate the steady state electrical networks by software PAS DAISY Off-Line Bizon V4.

Abstract:

Environment friendly transports are one of the main topics in the industry and society today. Shell organizes an annual university competition called Shell Eco Marathon. The goal is to build the car that consumes the least fuel during a race. KTH participated for the first time in 2006. The engine for this competition, DoubleTwo, has been developed within the school. The principle is a two stroke counter piston HCCI engine with variable compression ratio. HCCI stands for homogenous charge compression ignition. The combustion is quick and simultaneous which leaves a high efficiency and naturally low emissions of NO xand soot. The compression ignition timing is instable which is the reason for using variable compression ratio. The mechanical system for changing compression ratio is now patent protected. So far the principle has been verified, but lots of tests remain. The indicated efficiency is 32,5 % at a 6500 rpm and I MEP(Indicated mean effective pressure) is 4 bar. The project will be continued by other students and therefore this report is focusing on the different systems around the engine and the function of the engine rather than engine tests.
Miljövänliga transporter är ett av de största samtalsämnena i vårt samhälle och i industrin idag. Shell organiserar en årlig tävlig som heter Shell Eco Marathon. Målet är att bygga bilen som förbrukar minst bränsle under en tävling. KTH deltog för första gången 2006. Motorn för den här tävlingen, tvåkvadratmotorn, har utvecklats på skolan. Principen är en tvåtakts motkolvs HCCI motor med variabelt kompressionsförhållande. HCCI står för: homogenous charge compression ignition. Förbränningen är snabb och simultan vilket ger en hög verkningsgrad och naturligt låga utsläpp av NO Xoch sot. Kompressionsantändningen är instabil vilket är motiveringen till det variabla kompressionsförhållandet. Det mekaniska systemet för att variera kompressionsförhållandet är nu patentskyddat. Hittills har principen verifierats, men de flesta planerade testerna återstår. Den indikerade verkningsgraden är 32,5% vid 6500 varv per minut och det indikerade medeltrycket är 4 bar. Projektet kommer drivas vidare av andra studenter och därför är fokus i denna rapport att visa motorns princip och kringsystem snarare än körresultat.

Abstract:

Huvudsyftet med detta examensarbete är att visa de problem med köbildning som staden Söderköping har. Denna rapport består i stort av två delar. I den första analyseras dagens trafiksituation och det görs en beskrivning av de problem som uppstår i staden. I den andra delen analyseras dagens och möjliga framtida utvecklingar av trafiksituationen i staden med hjälp av en MatLab-modell. En röd tråd genom hela rapporten är att analysen i huvudsak görs med hjälp av stokastisk köanalys. Ett av målen med rapporten är att undersöka hur bra denna teknik är att beskriva uppkomsten och avvecklingen av köer i anknytning till signalljusreglerade korsningar, samt om den mer direkt går att använda till styrning av flöden genom exempelvis val av styrningsprogram i trafiksignalsanläggningarna. Den korsningen som valts för analysen är en av de mest problematiska när det gäller köbildning på genomfartsleden. Genom att använda den stokastiska köanalysen har även nya begrepp för beskrivning av kapaciteter tagits fram. Brytpunkten i flödesnivå för när en instabil köuppbyggnad uppstår är ett användbart mått i hantering av flöden i en korsning. Det resultaten visar är att det redan idag uppstår stora problem med köbildning under speciellt känsliga tidsperioder som vid exempelvis broöppningar. Att sedan trafikvolymerna under de kommande åren antas öka kommer att resultera i så stora problem att trafiksituationen under stora perioder kommer att vara helt oacceptabla med den infrastruktur som finns idag. Hur mycket olika kombinationer av trafikflödena kan öka och hur känsliga de olika ökningarna är presenteras även. Analysen visar att brytpunkten för hur stor ökningen kan bli går vid en allmän ökning av trafikflödet med 52,5% från den uppmätta maxtimmen i november. Det motsvaras av ett primärt flöde på 950 fordon/timme och ett sekundärt på 150 fordon/timme. En annan slutsats blir även att mått framtagna med hjälp av den stokastiska köanalysen är användbara för styrning av fordonsstyrda trafiksignalsreglerade korsningar.
The main purpose of this thesis is to confirm the suspected lack in traffic capacity for the thoroughfare in the town Soderkoping. This report is mainly made up by two parts. In the first the current situation with traffic problems in Soderkoping is analyzed and a presentation of the problems that occur due to the limitations of the infrastructure is given. In the second part, different possibilities of future development in the traffic flows are analyzed by implementing different scenarios with a Mat Lab-model. With the help of the model the sensibility of the different traffic flows are also established. A red thread through out the report is that of the use of stochastic queuing theory. One of the goals with the report is to investigate how capable the theory is on describing how queues develop and discharge in signalized intersections. To achieve this has new measurements to calculate the capacity in intersections been created. For instance, the calculation of a breaking-point in level of traffic flow that, when acceded, gives an instable queue growth. Furthermore the possibility of using the technique more direct, as in for example aiding the control of logical rules for shifting between signal phases is looked at. The results show that the infrastructure of today will not be enough in the future. The different scenarios and analyzes show that it is the primary traffic flows on the E22 that are more sensitive to an increase of flow levels and that the flows on the secondary streets can increase relatively much without any appreciably reduction of the accessibility of the junction. Another conclusion is that it is possibly with the aid of detectors to guide the prioritizing of phrases in traffic signal installations when the deployment of the detectors and the logic of the control system are established by using stochastic queuing theory on the different traffic streams. More traffic programs and better governing of the existing, with enhanced sensitivity and greater use of all programs, will increase the capacity of signalized intersections, in both total accessibility and the ability to prioritize the discharge of long queues.

Abstract:

A gene dosage study was conducted on a rare complete trisomy 22 human fibroblast cell line utilizing three lysosomal enzymes, ∝-iduronidase, ∝-galactosidase B, and arylsulfatase A, whose genes are located on chromosome 22 and two control enzymes, ,β-hexosaminidase A and -- fucosidase, with genes not on chromosome 22. A gene dosage effect was clearly demonstrated for an early passage number of the fibroblasts; however, later passage numbers gave inconclusive results. This study suggests that gene dosage studies must be carefully designed to be conducted only on early, matched passage number cells. ∝-fucosidase gave anomalous results most likely due to pleiotropic effects. The present gene dosage study confirmed the trisomic nature of the cell line studied and suggests that this type of study may be a useful diagnostic tool for small deletions, additions, or unbalanced translocations.

The aim of the thesis is to describe the situation in transformation 110/22 kV in distribution area E.ON East. Next to describe the current practice in the replacement of 110/22 kV transformers and mention the possibilities of replacing the machines at the end of their lifecycle with respect to the development of the load of the individual transformer stations in the distribution area. An integral part of the thesis is also the economic evaluation of premature replacement of transformers from two different perspectives. The next point is to look at the current possibilities in the lifecycle lengthening of these devices and to suggest a method for monitoring the state of paper insulation to estimate the remaining lifetime of the transformer. The last point is the evaluation of the transition of a significant customer from a voltage level of 22 kV to a voltage level of 110 kV, including related technical and economic issues.

Interleukin 22 (IL-22) is а newly-described Т helper cytokine which has been implicated in tissue regenerative processes in the gut and skin. Allergic asthma results from exposure to otherwise harmless environmental inhaled allergens that trigger Th2 Т Iymphocyte-driven IgE-аssосiаtеd immune responses in atopic individuals that contribute to airway inflammation and reversible airflow obstruction. In the long-term, this aberrant immune response is believed to increase in complexity and contribute to the permanent changes in the airways, including tissue remodelling and associated irreversible airway obstruction. In view of the known effects of IL-22 in wound healing, the overall aim of this research was to establish whether IL-22 is present in allergy and allergic asthma and to understand the influence of IL-22 on structural cells of the airway, with particular emphasis оп tissue remodelling. We, therefore, explored the presence of IL-22 and its receptors during allergen-induced late responses in the bronchi and skin and measured IL-22 production and its regulation in peripheral blood mononuclear cells (PBMCs), Т cell lines and clones derived from asthmatic bronchial muсоsa. IL-22 was increased in bгonchoalveolaг lavage after allergen inhalation in asthmatics but not in control subjects. While we were unable to reliably immunostain IL-22, its receptors were present within the cutaneous connective tissue and co-Iocalised with HSP-47, а marker of early fibrobIasts. In PBMCs, IL-22 increased after allergen stimulation whereas there was nо difference between atopics and control subjects. In Т cell lines derived from asthmatic bronchial mucosa, IL-22 was mainly found in СD4+ Т helper cells of Th1 and Th22 origin. IL-22 receptors were expressed by cultured bronchial epithelial cells, smooth muscle cells and pulmonary fibroblasts and were increased in expression in the presence of interferon-γ (IFN-γ). IL-22 was able to accelerate wound closure of bronchial epithelial cells independent of effects on cellular proliferation. Thus, IL-22 appears to be а Th1-related cytokine that may have а dual role in airway inflammation and repair.

The F-22 flight test program used a traditional distributed data acquisition system and a non IRIG-106 Chapter 10 recording system for its flight test program. In addition, it required a separate and very large Harris DAU system to monitor and record avionic data buses carrying secure data. Due to the size, cost, and the obsolescence of the Harris DAU system and components, Lockheed evaluated replacement systems. TTC proposed to develop F-22 specific Fiber Optic avionics bus monitors and an avionics PCM Data Selector / Encoder as part of its distributed IRIG-106 Chapter 10 Multiplexer / Recorder system to replace the Harris DAU. This replacement system challenges the traditional system approach used in many flight test programs. This paper describes the evolutionary process to design two independent distributed data acquisition and recording systems handling data with different classification levels. The data separation is maintained by way of system wiring, proper hardware that holds no residual data once power is removed, different transmission channels, hardware-based message blocking, and a separate IRIG-106 Chapter 10 multiplexing / recording system.

22 is a series of six programmatic compositions. The extra-musical narrative deals with the fact that twenty-two veterans commit suicide each day, a low estimate by some accounts. The music is written not specifically for veterans but to bring attention to veterans. It is important for all Americans to know the plight of those who have served their country and now are marginalized by the same country they swore to defend with their lives. Many veterans’ lives have been horrifically changed by war and those battle scars are now internal. Music is the best way that I can explain the complex emotions and story of veterans to a non-veteran audience. This music is intended to inform society of veteran suicides, not make a moral judgment against veterans or civilian society but to bring to light the story as it is. The music is instrumental so the weight of words will not interfere with how the listener interprets the music.

A asma acomete cerca de trezentos milhões de pessoas em todo o mundo. A doença é caracterizada por falta de ar, chiado e compressão no peito, tosse como consequência da hiperresponsividade brônquica e limitação de fluxo aéreo, causadas pela inflamação pulmonar Th2 e eosinofílica. Porém, existem evidências de que a IL-22 e a IL-17 participam da patogênese da asma alérgica. Com intuito de compreender melhor o papel da IL-22 na asma alérgica, utilizamos camundongos deficientes em IL-22 (IL-22KO) comparando-os aos animais Wild Type (WT) expostos ao alérgeno. Animais WT e IL-22KO foram sensibilizados e desafiados com ovalbumina (OVA) e avaliados quanto à inflamação eosinofílica, produção de citocinas, produção de muco, e populações celulares no pulmão. Nossos resultados mostram que a ausência de IL-22 resultou em diminuição da eosinofilia, IL-5 e IL-13 no lavado broncoalveolar, de células CD4+IL-4+ nos linfonodos e diminuição na produção de muco nas vias aéreas. Além disso, os camundongos IL-22KO alérgicos apresentam diminuição na porcentagem e número de células dendríticas CD11c+CD11b+CD103- nos pulmões quando comparados aos respectivo grupo WT. A transferência de células Th17 geradas a partir de animais IL-22KO causou diminuição na eosinofilia em camundongos expostos ao alérgeno quando os mesmos foram comparados aos animais que receberam células Th17 geradas a partir de animais WT. Esse resultado atribui mais à IL-22 do que à IL- 17 papel patogênico na asma alérgica. Outro indício da participação patogênica da IL-22 na asma alérgica é o fato de que o tratamento alérgeno específico, combinado ou não com a terapia livre de alérgeno, induziu redução da eosinofilia, do infiltrado de células dendríticas e diminuição de IL-22 no lavado broncoalveolar. A provável ação da IL-22 é a manutenção da viabilidade e sobrevivência de eosinófilos nos pulmões, fazendo que estes leucócitos continuem auxiliando no recrutamento de células dendríticas responsáveis pela captura e apresentação do alérgeno nos linfonodos, onde haverá a diferenciação de linfócitos de padrão Th2. Essas células podem migrar para os pulmões, gerando aumento na inflamação no local. Em síntese, nosso estudo corrobora o papel proinflamatório da IL-22 na asma alérgica e mostra, de forma inédita, que a transferência de células Th17 produtoras de ambas as citocinas, IL-22 e IL-17, mas não a transferência de células produtoras apenas de IL-17, causa exacerbação da inflamação pulmonar, possivelmente relacionada com o papel da IL-22 em prevenir indiretamente a indução de apoptose nos eosinófilos.
Asthma affects approximately three hundred million people worldwide. The disease is characterized by shortness of breath, wheezing and chest compression, coughing as a result of bronchial hyperresponsiveness and airflow limitation caused by Th2 lung inflammation and eosinophilia. However, there is evidence that IL-22 and IL-17 participate in the pathogenesis of allergic asthma. With the intention to better understand the role of IL-22 in allergic asthma, we used IL-22 deficient mice (IL-22KO) comparing them to wild type animals (WT) exposed to the allergen. Animals WT and IL-22KO were sensitized and challenged with ovalbumin (OVA), and assessed for eosinophilic airway inflammation, cytokine production, mucus production, and cell populations in the lungs. Our results show that the absence of IL-22 resulted in decreased eosinophilia, IL-5 and IL-13 in the bronchoalveolar lavage, CD4 + IL-4 + cells in the lymph nodes and decrease in mucus production in the airways. In addition, allergic IL-22KO mice have decreased percentage and number of dendritic cells CD11c + CD11b + CD103- in lungs when compared to their corresponding WT group. The transfer of Th17 cells generated from IL-22KO animals caused a reduction in eosinophilia in mice exposed to the allergen when they were compared to animals that received Th17 cells generated from WT mice. This result assigns more IL-22 than IL-17 pathogenic role in allergic asthma. Another indication of the pathogenic involvement of IL-22 in allergic asthma is the fact that the specific allergen treatment, combined or not with allergen-free therapy induced a reduction of eosinophilia, the dendritic cell infiltration and decreased IL-22 in bronchoalveolar lavage. The possible action of IL-22 is maintaining the viability and survival of eosinophils in the lungs, making these leukocytes remain helping in the recruitment of dendritic cells responsible for capture and allergen presentation in lymph nodes, causing differentiation in lymphocytes Th2. These cells can migrate to the lungs, resulting in increased inflammation at the site. In summary, our study confirms the proinflammatory role of IL-22 in allergic asthma and shows, in an unprecedented manner, the transfer of producing Th17 cells of both cytokines IL-22 and IL-17, but not the transfer of cells producing only IL-17, cause exacerbation of pulmonary inflammation, possibly related to the role of IL-22 on indirectly prevent induction of apoptosis in eosinophils.

Abstract:

Peripheral Myelin Protein-22 (PMP22) is a relatively major component of peripheral nerve myelin. Either missense mutations or duplication of the PMP22 gene can be the cause of Charcot-Marie-Tooth disease type 1 A (CMT1A). The control of PMP22 expression could be a basis for treatment. The aim of my research is to understand the regulation of the PMP22 gene in a transgenic mouse model. Initially, -8.5 kb 5' flanking sequence of the rat Pmp22 gene was used to drive reporter gene expressed in transgenic mice. This construct confers tissue specificity, weak developmental regulation, weakly detectable levels of expression in the dorsal root ganglia (DRG), but not the expected up-regulation during nerve regeneration. Secondly, I generated transgenic mice using -21 kb 5' Pmp22 flanking sequence. This construct confers robust reporter gene expression in the peripheral nervous system (PNS), and little to the DRGs in one line. The other lines show weak expression in the PNS, and stronger expression in the DRGs. Thirdly, I targeted a 21 kb Pmp22 promoter/reporter transgene to the hypoxanthine phosphorybosyl transferase (Hprt) locus in male embryonic stem cells. Using this methodology, I obtained a line carrying -21 kb of Pmp22 promoter driving beta-galactosidase gene in the hprt locus. This line confers expression similar to the four lines derived by pronuclear injection carrying the same construct. Fourthly, I introduced the LacZ reporter gene at the start codon of Pmp22 in a 100 kb bacterial artificial chromosome (BAC). Six transgenic lines were produced and analyzed, showing embryonic and postnatal developmental regulation, and high level of expression in the PNS similar to endogenous Pmp22 expression. From my results and the literature, I suggest that (1) the stretch of -10.5 to -21 kb could carry repressor(s), (2) there is (a) positive element(s) that lie(s) between -8.5 and -10.5 kb, and (3) 3' element(s) are important for Pmp22 expression.

Denna rapport behandlar mitt examensarbete som genomfördes i samarbete med Haldex

Brake AB. Haldex Brake AB håller på och utvecklare en ny generation broms åt lastbilar.

Uppdraget bestod av tre delar. Att ta fram lösningsförslag som säkerhetsställer beläggjärnets

placering i förhållande ett beläggjärn så kallad Cross-beam. Att säkerställa att bultar monteras

med rätt åtdragningsmoment på 230Nm. Den sista tredjedelen av arbetet bestod i att

presentera lösningar som helt eller delvis eliminerar oljud, när olika komponenter med spel,

slår i delar i okhuset.

I projektet har flera olika principkonstruktioner tagits fram och genom Freddy Olsson

utvärderingsmallar har sedan en primärkonstruktion utarbetats fram.

I dem andra två resterande projektdelarna belyses lösningarna väldigt generellt av den

anledningen att det kräver specialkomponenter.
